Abstract

A field experiment documented the development of fouling communities on two shell regions, the lip and hinge, of the pearl oyster species Pinctada fucata, Pteria penguin and Pteria chinensis. Fouling communities on the three species were not distinct throughout the experiment. However, when each species was analysed separately, fouling communities on the lip and hinge of P. penguin and P. chinensis were significantly different during the whole sampling period and after 12 weeks, respectively, whereas no significant differences could be detected for P. fucata. There was no significant difference in total fouling cover between shell regions of P. fucata and P. chinensis after 16 weeks; however, the hinge of P. penguin was significantly more fouled than the lip. The most common fouling species (the hydroid Obelia bidentata, the bryozoan Parasmittina parsevalii, the bivalve Saccostrea glomerata and the ascidian Didemnum sp.) showed species-specific fouling patterns with differential fouling between shell regions for each species. The role of the periostracum in determining the community development of fouling species was investigated by measuring the presence and structure of the periostracum at the lip and hinge of the three pearl oyster species. The periostracum was mainly present at the lip of the pearl oysters, while the periostracum at the hinge was absent and the underlying prismatic layer eroded. The periostracum of P. fucata lacked regular features, whereas the periostracum of P. penguin and P. chinensis consisted of a regular strand-like structure with mean amplitudes of 0.84 μm and 0.65 μm, respectively. Although the nature and distribution of fouling species on the pearl oysters was related to the presence of the periostracum, the periostracum does not offer a fouling-resistant surface for these pearl oyster species.  相似文献

Results are presented from a three-year investigation of the relationship between accumulations of marine fouling organisms and growth of the pearl oyster Pinctada fucata (Gould). Estimates are provided of the diversity of the foulers, and data on certain hydrological features of the experimental site are also given.There was an inverse correlation between growth of the pearl oyster and diversity of the biofouling assemblages, whatever depth. Growth curves did not follow seasonal variations in the environmental factors very exactly. It was also observed that the polychaete, Polydora vulgaris Mohammad, preferred the oysters planted on the bottom to those suspended near the surface. Bottom oysters suffered the highest mortality, but definite evidence of a relationship between mortality and infestation by the polychaete was not apparent. Percentages of both infestation and mortality were higher among the fouled oysters than those cleaned periodically.  相似文献

For pearl culture, nucleus and mantle grafts are implanted into the gonad of the host oyster. The epithelial cells of the implanted mantle graft elongate and surround the nucleus, and a pearl sac is formed. Shell matrix proteins secreted by the pearl sac play an important role in pearl formation. We studied the gene expression patterns of six shell matrix proteins (msi60, n16, nacrein, msi31, prismalin-14, and aspein) in the epithelial cells associated with pearl sac formation. There were differences in the expression patterns of the six genes in the epithelial cells, and the relative expression levels for msi60 and aspein differed between the mantle graft and pearl sac (48 days after implantation). Therefore, the gene expression patterns of the epithelial cells were genetically undetermined, and changed between before and after pearl sac formation. The gene expression patterns of the epithelial cells of the pearl sac may be regulated by the host oysters.  相似文献

Pinctada margaritifera is French Polynesia's most economically important aquaculture species. This pearl oyster has the specific ability to produce cultured pearls with a very wide range of colours, depending on the colour phenotypes of donor oysters used. Its aquaculture is still based on natural spat collection from wild stocks. We investigated three rare colour variants of P. margaritifera – orange flesh, and red and white shell colour phenotypes – in comparison with the wild‐type black flesh and shell commonly found in this species. The study aimed to assess the geographic distribution and genetic basis of these colour variants. Colour frequencies were evaluated during transfer and graft processes of pearl oyster seed captured at collector stations. Among the collection locations studied, Mangareva Island showed the highest rate of the orange flesh phenotype, whereas Takaroa and Takume atolls had relatively high rates of red and white shell phenotypes respectively. Broodstocks were made of these rare colour variants, and crosses were performed to produce first‐ and second‐generation progenies to investigate segregation. The results were consistent with Mendelian ratios and suggest a distinct model with no co‐dominance: (i) a two‐allele model for flesh trait, whereby the orange allele is recessive to the black fleshed type, and (ii) a three‐allele model for shell trait, whereby the black wild‐type allele is dominant to the red coloration, which is dominant to the white shell. Furthermore, the proposed model provides the basis for producing selected donor pearl oyster lines through hatchery propagation.  相似文献

Color is one of the most important factors determining the commercial value of pearls. Pinctada fucata is a well-known pearl oyster producing high-quality Akoya pearls. Phenotypic variation in amount of yellow pigmentation produces white and yellowish pearls. It has been reported that polymorphism of yellow pigmentation of Akoya pearls is genetically regulated, but the responsible gene(s) has remained unknown. Here, we prepared pearl sac pairs formed in the same recipient oyster but coming from donor oysters that differ in their color. These two pearl sacs produced pearls with different yellowness even in the same recipient oyster. Yellow tone of produced pearls was consistent with shell nacre color of donor oysters from which mantle grafts were prepared, indicating that donor oysters strongly contribute to the yellow coloration of Akoya pearls. We also conducted comparative RNA-seq analysis and retrieved several candidate genes involved in the pearl coloration. Whole gene expression patterns of pair sacs were not grouped by pearl color they produced, but grouped by recipient oysters in which they were grown, suggesting that the number of genes involved in the yellow coloration is quite small, and that recipient oyster affects gene expression of the majority of genes in the pearl sac.  相似文献

Crassostrea madrasensis is an important fouling oyster in tropical industrial cooling water systems. C. madrasensis individuals attach to surfaces by cementing one of their two valves to the substratum. Therefore, oyster fouling creates more problems than mussel fouling in the cooling conduits of power stations, because unlike the latter, the shell of the former remains attached to the substratum even after the death of the animal. However, there are no published reports on the tolerance of this species to chlorination and heat treatment. The mortality pattern and physiological behaviour (oxygen consumption and filtration rate) of three size groups (13 mm, 44 mm and 64 mm mean shell length) of C. madrasensis were studied at different residual chlorine concentrations (0.25, 0.5, 0.75, 1, 2, 3 to 5 mg lm 1) and temperatures (30°C to 45°C). The effect of shell size (=age) on C. madrasensis mortality in the presence of chlorine and taking into account temperature was significant, with the largest size group oysters showing highest resistance. At 1 mg lm 1 residual chlorine, the 13 mm and 64 mm size group oysters took 504 h (21 d) and 744 h (31 d), respectively to reach 100% mortality. At 39°C, the 13 mm size group oysters took 218 min to reach 100% mortality, whereas the 64 mm size group oysters took 325 min. The oxygen consumption and filtration rate of C. madrasensis showed progressive reduction with increasing residual chlorine concentrations. However, the filtration rate and oxygen consumption responses of C. madrasensis were not significantly different between 30°C (control) and 37.5°C. There was a sharp decrease in the filtration rate and oxygen consumption at 38.5°C. A comparison of the present mortality data with previous reports on other bivalves suggests that the chlorine tolerance of C. madrasensis lies in between that of Perna viridis and Perna perna, while its temperature tolerance is significantly higher than that of the other two bivalve species. However, in power station heat exchangers, where simultaneous chlorine and thermal stresses are existent, C. madrasensis may have an edge over other common foulants, because of its high temperature tolerance.  相似文献

The endemic mulberry whelk (Tenguella marginalba) is a common predator on Australian intertidal rocky shores. The introduced Pacific oyster (Crassostrea gigas), found within the natural range of T. marginalba, is potential prey for the whelk. In experiments designed to increase our understanding of predatory behaviour by the whelk on oysters, we found that adult T. marginalba detected C. gigas and increased movement in the presence of oyster prey. Tenguella marginalba showed a preference for smaller C. gigas, but consumed oysters up to 60?mm in shell height. To access oyster flesh, whelks used their radula to drill holes in the oyster’s shell. These holes were on average 0.68?±?0.09?mm in diameter, most frequently located central to the pericardial cavity on the right (upper) valve. Predation was greatest when predator and prey were both submerged, but was unaffected by a diurnal light cycle. When offered a choice among the native Sydney rock oysters (Saccostrea glomerata), mussels (Trichomya hirsuta) or the invasive C. gigas, whelks displayed no preference among prey. We conclude that the invasive oyster C. gigas represents a viable food source for T. marginalba, which may help to slow the spread of this invasive oyster throughout eastern Australia.  相似文献

Animals use sensory stimuli either to assess and select habitats, mates or food, as well as for communication. The present study aimed to understand the behavioural processes enabling several Chaetodon species (butterflyfishes) to locate one of their food sources (epibionts present on pearl oyster shells) at Rangiroa atoll (French Polynesia). Among the five species tested, our 2-channel choice flume chamber experiments identified three species that were attracted to their food source by chemical stimuli. HPLC experiments showed that pearl oysters and epibionts have specific and unique chemical fingerprints, either one or nine specific peaks, respectively. Overall, chemical stimuli are emitted by both epibionts (used directly by Chaetodon auriga, C. lunula and C. citrinellus) and live pearl oysters (used indirectly by C. auriga and C. lunula) to locate their food source. Biosynthesis of these chemical stimuli could be used to artificially attract butterflyfishes to pearl oyster rearing stations in order to increase the natural cleaning of pearl oyster shells and thus reduce one large cost for this aquaculture.  相似文献

软体动物engrailed蛋白和骨形成相关蛋白对胚胎贝壳区域边界形成可能具有重要作用,engrailed还被推测为调节基质蛋白在外套膜组织区域化表达的重要调控因子.因此,弄清调控engrailed在软体动物中特征表达的分子机制有着重要的研究意义.但是,由于贝类基因组测序尚不完整,目前也没有建立获得贝类细胞系,以致于许多预测可能参与调控的基因需要通过克隆来鉴定,而且经典的研究细胞信号通路的方法也很难得到应用.目前,在中国南海广泛养殖的合浦珠母贝中,已获知其BMP2和Smad3的cDNA全长,以该贝的基因组为模板,PCR扩增获得了一段engrailed编码区片段.经软件分析,该片段含有EH4结构域,且与其他物种engrailed蛋白具有很高的同源性.研究的贝中,特别是外套膜组织中,engrailed、BMP2和Smad3三者表达之间的相关性,将有助于我们理解贝壳形成的分子机制.贝壳缺刻后半定量PCR试验结果表明,三者均参与贝壳修复,且在贝壳缺刻后的修复过程中,engrailed和Smad3的mRNA表达变化规律非常相似,提示它们之间可能存在相互影响的联系.用地塞米松(DXM)和过氧化氢(H2O2)分别处理原代培养的贝外套膜组织迁出细胞,实时相对定量PCR检测engrailed、BMP2和Smad3的mRNA表达水平,统计分析结果表明,三者具有显著的相关性.上述所有结果为进一步研究贝类生物矿化的发育和信号转导机制提供了新的思路和基础.  相似文献

After being ecologically extinct for almost a century, the discovery of a shellfish reef with native European flat oysters (Ostrea edulis) in the Dutch coastal area of the North Sea by the authors of this study called for an extensive survey to better understand some of the key requirements for the return of the native oyster in coastal waters. We assessed habitat conditions, its potential for increasing biodiversity, and the role of substrate provision by other bivalves such as the invasive alien Pacific oyster (Crassostrea gigas). Using underwater visual census, O. edulis size-frequency distributions and attachment substrate was investigated, as well as the composition of the epibenthic community and substrata types inside quadrats that were distributed across the reef. This reef was found to be composed of native European flat oysters, invasive alien Pacific oysters and blue mussels (Mytilus edulis), alternated with sandy patches. The O. edulis population (6.8?±?0.6 oysters m?2) consisted of individuals of different size classes. In quadrats with native and non-native oysters the number of epibenthic species was 60% higher compared to adjacent sand patches within the reef. Notably, our results showed that the native oyster predominantly used shell (fragments) of the invasive Pacific oyster as settlement substrate (81% of individuals). Our results optimistically show that conditions for native oyster restoration can be suitable at a local scale in the coastal North Sea area and suggest that the return of native oysters may be facilitated by novel substrate provided by invasive oysters at sites where their distribution overlap.  相似文献

Crassostrea virginica, the eastern oyster, is a native foundational species that inhabits coastal and estuarine ecosystems along the western Atlantic seaboard. Introduction of C. virginica into estuarine areas with limited or no extant populations is gaining popularity as a pro‐active approach for improving estuarine water quality and creating natural wave breaks for shoreline stabilization. Adult oysters, grown by 113 community members under their private docks, were collected and deployed at three county‐owned sites along the Indian River Lagoon within Brevard County, Florida. In this shallow, warm‐water estuary, replicate treatments deployed at each site included bagged adult oysters collected from gardeners in fall 2014, bagged adult oysters from spring 2015 gardeners, bagged blank (clean) shell, and empty plots (control). Prior to deployment, morphometric data (shell length, weight) were collected on all gardened oysters. Morphometric data were then collected quarterly for all surviving and recruited oysters for 18 months. Our monitoring timeframe was sufficient for assessing survival of gardened oysters, but likely not sufficient to understand recruitment patterns. In areas with no recruitment and limited gardened oyster survival, regular enhancement with live oysters would be needed for long‐term success. In areas with natural recruitment, the blank shell treatment was most successful. Lessons learned from this study include: (1) need for better tracking of abiotic variables (e.g. salinity) where gardening occurred, (2) role of seasonality in initial post‐deployment survival, even in a warm‐water estuary, and (3) importance of pilot studies prior to large‐scale gardened oyster deployments.  相似文献

Plankton samples and gut contents of P. margaritifera were analysed monthly from April 1972 to March 1973. Coscinodiscus sp. was the most ingested food by pearl oysters of all ages. However, food variety increased as the oyster grew older. Experiments confirmed the absence of food selectivity in P. margaritifera. Unlike the young ones, the adult showed reduced growth during summer (July–September), which coincides with its spawning season.  相似文献

In order to study the relative growth of the pearl oyster Pinctada radiata in Tunisia, a total of 330 individuals of this species were collected from six sites along the Tunisian coastline. Quantitative measurements of collected oysters were conducted for shell height, shell length, shell width, hinge length, height and width of the nacreous part and wet weight. The size structure of the sampled populations was described and the relative growth between different morphometric characteristics was estimated as allometric growth lines for the six P. radiata samples. It appeared that the majority of examined samples were dominated by large individuals that exceed a shell height of 42 mm. The maximum size (100.5 mm), recorded in Bizerta lagoon, is bigger than that recorded elsewhere in particular in the Red Sea. Size distribution analysis also showed that the majority of P. radiata samples were dominated by two or more size groups. Differences of allometric regression were found between the examined samples for the tested relationships. Moreover, the Factorial Discriminant Analysis, coupled with Ascending Hierarchic Classification, classified the sub-populations according to geographic locations.  相似文献

The spawning cycle of the winged pearl oyster, Pteria penguin, was studied at Orpheus Island, north-eastern Queensland to observe seasonal changes in gonad development, mean oocyte diameter and sex ratio over time. A total of 201 oysters were collected over 10 sampling periods, approximately every 5–6 weeks (mean?=?20 oysters/sample). Histological analysis of gonads showed that spawning activity occurred mainly through the austral mid-spring to late summer (November to March), when mean monthly water temperature ranged between 27.5°C and 29.4°C. There was a primary spawning peak at the beginning of the spawning season in December, followed by a secondary peak in March. Mean oocyte diameter (µm) was highest during the warmer months, ranging from 31.2?±?2.4 in November to 36.2?±?3.2 in March, and sex ratio analysis showed the percentage of females in the population also peaked between 30% and 50% during this period.  相似文献

研究利用5个高度多态性的微卫星标记,对500个企鹅珍珠贝(Pteria penguin)的4个生长性状进行了关联分析。结果显示, QEB-D15和CL-232两个微卫星标记与企鹅珍珠贝的壳宽呈极显著相关(P<0.01);位点QEBD15基因型是239/263的个体壳宽为最大值,基因型是239/273的个体壳宽为最小值,推测263 bp等位基因与壳宽之间存在正相关关系,而273 bp等位基因与壳宽之间存在负相关关系;位点CL-232基因型为157/174的个体壳长、壳宽、总重的均值较同一位点的其他基因型均为最大值,该基因型推测为优势基因型;而基因型为177/192的个体壳长、壳宽、壳高和总重的均值较同一位点的其他基因型均为最小值,推测该基因型为劣势基因型,上述结果可为企鹅珍珠贝分子标记辅助选择育种提供理论依据和参考。  相似文献

For pearl culture, the pearl oyster is forced open and a nucleus is implanted into the gonad with a mantle graft. The outer mantle epithelial cells of the implanted mantle graft elongate and surrounding the nucleus a pearl sac is formed. Shell matrix proteins secreted by the pearl sac play an important role in the regulation of pearl formation. Recently, seven shell matrix proteins were identified from the pearl oyster Pinctada fucata. However, there is a paucity of information on the function of these proteins and their gene expression patterns. Our study aims to elucidate the relationship between pearl type, quality, and gene expression patterns of six shell matrix proteins (msi60, n16, nacrein, msi31, prismalin-14, and aspein) in the pearl sac based on real-time PCR analysis. After culturing for about 2 months, the pearl sac tissues were collected from 22 individuals: 12 with high quality (HP), nine with low quality (LP), and one with organic (ORG) pearl formation. The surface of each of the 12 HP pearls was composed only of a nacreous layer; in contrast, that of the nine LP pearls was composed of nacreous and prismatic layers. The six target gene expressions were detected in all individuals. However, delta threshold cycle (ΔC T) for msi31 was significantly higher in the HP than in the LP individuals (Mann–Whitney’s U test, p = 0.02). This means that the relative expression level of msi31, which constitutes the framework of the prismatic layer, was higher in the LP than in the HP individuals.  相似文献

Magnesium is widely used to control calcium carbonate deposition in the shell of pearl oysters. Matrix proteins in the shell are responsible for nucleation and growth of calcium carbonate crystals. However, there is no direct evidence supporting a connection between matrix proteins and magnesium. Here, we identified a novel acidic matrix protein named PfN44 that affected aragonite formation in the shell of the pearl oyster Pinctada fucata. Using immunogold labeling assays, we found PfN44 in both the nacreous and prismatic layers. In shell repair, PfN44 was repressed, whereas other matrix proteins were up-regulated. Disturbing the function of PfN44 by RNAi led to the deposition of porous nacreous tablets with overgrowth of crystals in the nacreous layer. By in vitro circular dichroism spectra and fluorescence quenching, we found that PfN44 bound to both calcium and magnesium with a stronger affinity for magnesium. During in vitro calcium carbonate crystallization and calcification of amorphous calcium carbonate, PfN44 regulated the magnesium content of crystalline carbonate polymorphs and stabilized magnesium calcite to inhibit aragonite deposition. Taken together, our results suggested that by stabilizing magnesium calcite to inhibit aragonite deposition, PfN44 participated in P. fucata shell formation. These observations extend our understanding of the connections between matrix proteins and magnesium.  相似文献

目的:研究合浦珠母贝转录因子SOX9对Prismalin-14的转录调控机制。方法:应用在线预测软件PROMO分析Prismalin-14的启动子序列,以预测Prismalin-14启动子上可能的转录因子与其结合位点;运用细胞共转染实验和双荧光素酶报告系统以检测SOX9对Prismalin-14启动子的激活作用;构建Prismalin-14启动子截短体的荧光素酶报告载体,并和SOX9的真核载体共转到HEK-293T细胞中,再进行双荧光素酶报告系统检测Prismalin-14启动子的活性;构建SOX9截短体的真核表达载体,并与Prismalin-14启动子的荧光素酶报告载体共转到HEK-293T细胞中,再进行双荧光素酶报告分析Prismalin-14启动子的活性。结果:SOX9能激活Prismalin-14的启动子的活性,并具有剂量效应;对Prismalin-14启动子进行截短后,不包含结合位点的Prismalin-14启动子的活性是野生型Prismalin-14启动子活性的49%,推测Prismalin-14启动子上的-415bp到-405bp区域是SOX9激活作用的关键区域;对SOX9的SRY-related HMG结构域进行截短后,其对Prismalin-14启动子的激活作用显著减少,因此SOX9结构的完整对Prismalin-14启动子活性的激活作用是必须的。结论:Prismalin-14的转录可能受SOX9调控,为进一步研究合浦珠母贝的转录调控机制提供基础,将有助于从分子水平上理解贝壳形成的上游调控机理。  相似文献
