以盐敏感型黄瓜品种‘津春2号’为材料,研究了丛枝菌根真菌(AMF)对盐胁迫下黄瓜幼苗生长及叶片、根系中渗透调节物质含量和抗氧化酶活性的影响.结果表明:(1)在盐胁迫条件下,黄瓜幼苗生长受到明显抑制,其株高、地上部、地下部干鲜重均明显减小,同时体内可溶性蛋白、可溶性糖、脯氨酸和MDA含量,以及O2(÷)产生速率和SOD、POD、CAT活性均比对照显著升高.(2)盐胁迫下接种AMF可显著促进黄瓜植株的生长,进一步提高黄瓜幼苗体内可溶性蛋白、可溶性糖和脯氨酸含量及SOD、POD、CAT活性,而显著降低MDA含量和O2(÷)产生速率.研究表明,AMF可通过显著促进盐胁迫下黄瓜幼苗体内渗透调节物质积累和抗氧化酶活性提高,有效降低体内膜脂过氧化水平,从而缓解盐胁迫对植株的伤害,增强黄瓜幼苗对盐胁迫的耐性.  相似文献

以一年生灰胡杨幼苗为试验材料,利用田间控盐控水的方法,进行干旱和盐胁迫试验,通过测定生长和生理生化指标探讨幼苗在盐旱交叉胁迫下的生长发育及适应规律,旨在阐明干旱及盐交叉胁迫下植物抗旱抗盐机理。研究结果表明:在盐、旱及交叉胁迫下,灰胡杨幼苗抗氧化酶活性、MDA和脯氨酸含量与对照存在显著差异(P0.05)。(1)在8、11 g/L和15 g/L盐处理下,灰胡杨幼苗相对高生长、相对枝长和冠幅增量均受到抑制,且差异显著(P0.05),而干旱胁迫和盐旱交互胁迫下差异不显著。(2)在盐胁迫、盐旱交叉胁迫下,随着胁迫程度的加重,抗氧化酶SOD、POD、CAT活性表现出先增加后降低的趋势,三者协调一致;仅干旱胁迫时,抗氧化酶SOD、POD、CAT活性显著增加;(3)在盐、旱及其盐旱交叉胁迫下,脯氨酸含量呈上升趋势,MDA含量则表现出先降低后升高趋势,这与抗氧化酶活性先升高后降低的趋势相对应。因此,抗氧化酶活性对缓解脂膜过氧化的伤害具有一定限度,MDA含量与抗氧化酶活性呈负相关,灰叶胡杨幼苗在盐旱交叉胁迫下表现出一定的耐性。  相似文献

外源GSH对盐胁迫下番茄幼苗生长及抗逆生理指标的影响   总被引:5,自引:0,他引:5  
采用营养液栽培法,研究外源谷胱甘肽(GSH)对NaCl胁迫下番茄幼苗生长、根系活力、电解质渗透率和丙二醛(MDA)、脯氨酸(Pro)、可溶性糖含量以及超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性的影响,为利用外源物质减轻盐胁迫伤害提供理论依据。结果显示:(1)NaCl胁迫显著抑制了番茄幼苗的生长、根系活力和SOD、POD、CAT活性,提高了电解质渗透率及MDA、Pro、可溶性糖含量;(2)外源喷施GSH能够诱导NaCl胁迫下番茄幼苗叶片抗氧化酶SOD、POD、CAT活性上调,电解质渗透率及MDA含量下降,Pro和可溶性糖含量恢复至对照水平;(3)外源喷施还原型谷胱甘肽抑制剂(BSO)使NaCl胁迫下番茄幼苗的根系活力以及抗氧化酶SOD、POD、CAT活性下降,脯氨酸含量提高;(4)喷施GSH可诱导BSO和NaCl共处理番茄植株的根系活力、SOD、POD、CAT活性提高,MDA和Pro含量降低。研究表明,外源GSH可通过提高促进盐胁迫下番茄幼苗植株渗透调节能力及清除活性氧的酶促系统的防御能力、降低细胞膜脂过氧化程度、保护膜结构的完整性,从而有效缓解NaCl胁迫对番茄幼苗生长的抑制,提高其耐盐性。  相似文献

研究了大米草幼苗在不同培养盐度(0、20、30、50、100mmol/LNaCl)下,MDA、游离脯氨酸、可溶性糖、可溶性蛋白质含量以及保护酶(SOD、POD、CAT)活性等生理指标的变化情况。结果表明:保护酶(SOD、POD、CAT)活性在盐胁迫40d前逐渐上升且达显著差异。随着胁迫时间延长,MDA含量与CK相比逐渐降低。随着盐分胁迫浓度的增加及盐胁迫时间延长,大米草叶片中游离脯氨酸、可溶性糖、可溶性蛋白质含量呈上升趋势。在盐胁迫下,渗透调节物质的积累作用是大米草对盐胁迫的主要响应过程,其体内的抗氧化保护酶在此过程中也发挥了重要的作用。  相似文献

为明确外源一氧化氮(NO)对甜瓜幼苗耐盐性的影响,该研究以甜瓜品种‘农大甜10号’为试验材料,在300 mmol·L^(-1)NaCl胁迫条件下,叶面喷施不同浓度(50、100、150、200、250μmol·L^(-1))外源NO供体硝普钠(SNP),分析甜瓜幼苗生长、光合色素含量、细胞膜透性、抗氧化酶活性、渗透调节物质含量的变化。结果表明:(1)盐胁迫显著抑制甜瓜幼苗的生长,同时显著降低叶片光合色素含量、细胞膜透性、抗氧化酶活性、渗透调节物质含量。(2)叶面喷施150μmol·L^(-1)SNP能够显著提高盐胁迫下甜瓜幼苗的株高、茎粗、干鲜重、壮苗指数,并显著提高盐胁迫下甜瓜幼苗的光合色素含量,从而提高甜瓜的光合作用。(3)喷施150μmol·L^(-1)SNP可显著提高盐胁迫下甜瓜幼苗超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)、谷胱甘肽还原酶(GR)、单脱氢抗坏血酸还原酶(MDHAR)及脱氢抗坏血酸还原酶(DHAR)的活性,显著降低盐胁迫下甜瓜幼苗的丙二醛(MDA)、过氧化氢(H_(2)O_(2))含量及超氧阴离子(O^(-)·_(2))产生速率。(4)喷施150μmol·L^(-1)SNP可显著提高盐胁迫下甜瓜幼苗叶片脯氨酸(Pro)和可溶性蛋白的含量。研究认为,在盐胁迫环境下,适宜浓度的外源SNP(150μmol·L^(-1))可通过提高甜瓜幼苗的抗氧化酶活性以及光合色素和小分子可溶性有机化合物含量来增强活性氧的清除能力,降低膜脂过氧化作用,有效减轻盐胁迫对幼苗的伤害,从而增强其耐盐性,促进幼苗生长。  相似文献

以甜瓜品种‘西域一号’幼苗为材料,采用营养液水培方法,设置正常通气(对照)、正常通气+GABA(5mmol.L-1)、低氧胁迫、低氧胁迫+GABA(5mmol.L-1)4个处理,研究了外源γ-氨基丁酸(GABA)对正常通气和低氧胁迫下甜瓜幼苗活性氧代谢的影响。结果表明:与正常通气处理相比,低氧胁迫处理导致甜瓜幼苗体内O2.-产生速率和H2O2、MDA含量显著增加,同时SOD、POD、CAT、APX、GR等抗氧化酶活性和抗氧化物质AsA、GSH含量显著提高。低氧胁迫下外源GABA能显著提高甜瓜幼苗叶片SOD、CAT、APX、GR等酶活性和AsA、GSH含量,降低了植株体内O2.-产生速率和H2O2、MDA含量;而正常通气条件下添加外源GABA处理对甜瓜幼苗活性氧代谢的影响较小,仅CAT、GR活性和AsA、GSH含量显著提高,而H2O2、MDA含量显著降低。结果证明,添加外源GABA可以通过显著提高低氧胁迫下抗氧化酶活性和抗氧化物质含量来降低甜瓜幼苗活性氧积累,维持其细胞膜结构稳定性,从而有效减轻低氧胁迫对甜瓜幼苗的伤害。  相似文献

采用砂培方式,研究了外源5-氨基乙酰丙酸(ALA)对盐胁迫下菘蓝种子的萌发、幼苗叶片的可溶性糖含量、丙二醛(MDA)含量及其抗氧化酶活性的影响,探讨ALA缓解菘蓝受盐胁迫伤害的响应机制。结果显示:(1)菘蓝种子萌发及幼苗生长在100 mmol·L-1 NaCl胁迫下受到明显的抑制,种子发芽率、发芽势、发芽指数、活力指数与自然含水量均显著降低,丙二醛含量、可溶性糖含量以及超氧化物歧化酶(SOD)、过氧化物酶(POD)活性显著升高。(2)盐胁迫下适宜浓度的ALA处理显著提高了种子萌发率、自然含水量及SOD、POD和CAT活性,降低了可溶性糖和丙二醛的含量,并以16.7 mg·L-1 ALA处理盐胁迫下菘蓝种子的发芽率、发芽势最大,其幼苗的SOD、POD、CAT活性最强。研究表明,盐胁迫显著抑制菘蓝种子的萌发及幼苗生长,适宜浓度的ALA能够有效缓解盐胁迫对菘蓝种子萌发及幼苗生长的伤害,提高植株的抗盐性,并以16.7 mg·L-1 ALA处理效果最佳。  相似文献

作者于塑料连栋大棚栽培条件下,研究了接种丛枝菌根真菌(AMF)Glomus mossea-2对低温下黄瓜(Cucumis sativus L.)幼苗同化产物积累、光合生理和过氧化氢酶(CAT)、过氧化物酶(POD)、超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)的活性及基因表达的影响。结果表明:低温显著抑制了AMF对黄瓜根系的侵染能力和菌根相对依赖性。接菌后30d—45d AMF为快速侵染期。接种AMF植株的鲜重根冠比、总干重、总鲜重均显著大于未接菌处理。低温胁迫下,接种AMF延缓了光合速率、根系活力、羧化效率和叶绿素、可溶性蛋白含量的下降,并且使丙二醛(MDA) 的含量保持相对较低的水平,诱导了抗氧化酶基因的表达及活性提高。接种AMF可以使叶片维持较高的抗氧化酶水平和光合能力,增强了对低温胁迫的抗性。 关键词:黄瓜;丛枝菌根真菌;低温;抗氧化酶  相似文献

选用4个不同产地披针叶茴香(Illicium lanceolatum)(浙江临安(LA)、浙江开化(KH)、江西武宁(WN)和福建南平(NP))1年生盆栽幼苗为试验材料,设置3个光照处理(Ⅰ:25μmol·m-2·s-1;Ⅱ:50μmol·m-2·s-1;Ⅲ:75μmol·m-2·s-1)和2个水分处理(正常供水(CK)和自然干旱2d),探讨弱光环境下短期干旱对披针叶茴香幼苗生理特性及适应性的影响。结果表明:两种水分处理下,披针叶茴香幼苗抗氧化酶活性和渗透调节物质含量对弱光的敏感性存在明显差异。在光处理Ⅱ和Ⅲ条件下,WN产地幼苗经2d水分胁迫后,过氧化氢酶(CAT)活性比正常水分条件明显增加,过氧化物酶(POD)活性在光处理Ⅲ下比正常水分条件的显著增加,各光处理下超氧化物歧化酶(SOD)活性比正常水分的显著下降,丙二醛(MDA)含量相反。水分胁迫后,各光处理下LA产地幼苗的SOD和POD酶活性低于正常水分条件的,可溶性蛋白和MDA含量均较正常水分处理的增加。与正常水分条件相比,水分胁迫后的KH产地幼苗SOD、POD和CAT酶活性在光处理Ⅰ下均显著增加,可溶性蛋白和MDA含量有所下降。NP产地幼苗各光处理的酶活性均表现出水分胁迫后的比正常水分下的有所提高。可知,短期水分胁迫对提高KH、WN和NP产地幼苗对弱光环境的适应性具有显著促进作用。研究揭示短期水分胁迫可通过增加抗氧化酶活性和渗透调节物质含量来提高幼苗对弱光的适应性。  相似文献

利用盆栽的方式研究了干旱胁迫下接种兰科菌根真菌(OM)对铁皮石斛生长的影响,并分析了铁皮石斛叶片相对含水量、游离脯氨酸含量、电解质渗透率、丙二醛(MDA)含量、活性氧成分、抗氧化酶活性变化,用定量PCR技术分析了相关抗氧化酶基因的表达特性,以探讨菌根真菌对铁皮石斛干旱胁迫的缓解作用及其机制。结果表明:(1)与正常水分条件相比,干旱胁迫显著降低了铁皮石斛幼苗的生物量和叶片相对含水量,提高了叶片电解质渗透率、脯氨酸含量、MDA含量、O-·2产生速率和H2O2水平。(2)菌根真菌能显著提高干旱胁迫下铁皮石斛叶片相对含水量,降低叶片电解质渗透率、脯氨酸含量、MDA含量、O-·2产生速率和H2O2水平;在不同水分条件下,菌根真菌均能有效促进铁皮石斛幼苗生长,其株高、根重、茎叶重和总生物量均大于未接种组。(3)菌根真菌可诱导干旱胁迫下铁皮石斛超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)基因的表达,提高SOD、POD和CAT的活性,有效缓解干旱胁迫对质膜的过氧化伤害。研究认为,菌根真菌能提高干旱胁迫下铁皮石斛的抗氧化酶活性及其相关基因表达水平,增强铁皮石斛抗氧化防御能力,有效缓解干旱胁迫对铁皮石斛幼苗生长的抑制。  相似文献

Genetic engineering has improved the product yield of a variety of compounds by overexpressing, inactivating, or introducing new genes in microbial systems. The production of flavor-enhancing ester compounds is an emerging area of heterologous gene expression for desired product yield in Escherichia coli. Isoamyl acetate, butyl acetate, ethyl acetate, and butyl butyrate are reported here to be produced by expressing Saccharomyces cerevisiae genes ATF1 or ATF2 and the strawberry gene SAAT in E. coli when the appropriate substrates are provided. Increasing the concentration of alcohol added to the reaction generally resulted in increased ester production. ATF1 expression was found to produce more isoamyl acetate and butyl acetate than ATF2 expression or SAAT expression in the strains and culture conditions examined. Additionally, SAAT expression resulted in greater isoamyl acetate and butyl acetate production than ATF2 expression. Butyl butyrate is produced by cell-free extracts of E. coli harboring SAAT but not ATF1 or ATF2.  相似文献

Data from microscopic morphology, single-spore cultures, and DNA analyses of teleomorphs and anamorphs support the recognition of five species of Prosthecium with Stegonsporium anamorphs on Acer: P. acerinum sp. nov., the teleomorph of S. acerinum; P. acerophilum comb. nov., formerly known as Dictyoporthe acerophila; P. galeatum comb. nov., originally described as Massaria galeata; P. opalus sp. nov.; and P. pyriforme sp. nov., the teleomorph of S. pyriforme s. str. The morphology of both type specimens and freshly collected material was investigated. The teleomorphs have brown ellipsoidal ascospores with five distosepta and often a longitudinal distoseptum. The anamorphs of all species described here belong to Stegonsporium; their connection to the Prosthecium teleomorphs was demonstrated by morphology and DNA sequences of single spore cultures derived from both ascospores and conidia. The anamorphs and teleomorphs of all five Prosthecium species are described and illustrated by LM images, and a key to these species is provided. As perceived from this work, S. pyriforme is restricted to Europe and does not occur in North America, whereas S. acerinum is restricted to North America, not found in Europe. The host associations given in the literature are revised and evidence is provided that only A. opalus, A. pseudoplatanus, and A. saccharum are confirmed hosts of Prosthecium with Stegonsporium anamorphs. Molecular phylogenetic analyses of tef1, ITS rDNA, and partial nuLSU rDNA sequences confirm that the species with Stegonsporium anamorphs are closely related to P. ellipsosporum, the generic type species. Stilbospora macrosperma is confirmed as the anamorph of P. ellipsosporum by DNA data of single spore isolates obtained from both ascospores and conidia.  相似文献

Samples of Kochia (K. scoparia), Atriplex (A. dimorphostegia), Suaeda (S. arcuata) and Gamanthus (G. gamacarpus) were collected and analyzed for chemical composition including crude protein (CP), ether extract (EE), ash, neutral detergent fiber (NDFom), acid detergent fiber (ADFom), non-protein N (NPN), Ca, P, Na, K, Cl, Mg, Fe, Cu and Se. In addition, in situ ruminal degradability and post-ruminal disappearance of dry matter (DM) and CP of the samples using a mobile bag technique were determined. Results indicate that the chemical composition of Kochia and Atriplex was notably different from those of Suaeda and Gamanthus. All of these halophytic plants had high concentrations of Na, K, Cl, Cu and Se, and low levels of Ca, P and Mg. The rapidly degradable fractions of DM and CP (g/g) of Kochia (0.31 and 0.35, respectively) and Atriplex (0.39 and 0.50, respectively) were lower than for Suaeda (0.53 and 0.55, respectively) and Gamanthus (0.56 and 0.66, respectively). Ruminal DM and CP disappearance of Kochia (444 and 517 g/kg, respectively) and Atriplex (472 and 529 g/kg, respectively) were lower (P<0.05) than those of Suaeda (553 and 577 g/kg, respectively) and Gamanthus (663 and 677 g/kg, respectively) (P<0.05) using the mobile bag technique. Suaeda had the lowest (P<0.05) NDFom and ADFom disappearance (214 and 232 g/kg, respectively) in the rumen. Kochia scoparia and Atriplex dimorphostegia have more beneficial chemical nutritive components and digestible values versus Suaeda arcuata and Gamanthus gamacarpus.  相似文献

Seven bean rhizobial strains EBRI 2, 3, 21, 24, 26, 27 and 29 identified as Rhizobium etli, and EBRI 32 identified as Rhizobium gallicum, isolated from Egyptian soils and which nodulated Phaseolus vulgaris efficiently, were subjected to hybridization with a nifH probe in order to estimate the copy number of this gene. Seven strains (EBRI 2, 3, 21, 24, 26, 27 and 29) which were only able to nodulate Phaseolus vulgaris, contained three copies of the nifH gene, consistent with their identification as Rhizobium etli bv. phaseoli. Only one strain (EBRI 32) which nodulated both Phaseolus vulgaris and Leucaena leucocephala, had one copy of nifH gene. This confirmed the classification of this strain as Rhizobium gallicum bv. gallicum.  相似文献

In order to dissect the genetic regulation of leafblade morphogenesis, 16 genotypes of pea, constructed by combining the wild-type and mutant alleles of MFP, AF, TL and UNI genes, were quantitatively phenotyped. The morphological features of the three domains of leafblades of four genotypes, unknown earlier, were described. All the genotypes were found to differ in leafblade morphology. It was evident that MFP and TL functions acted as repressor of pinna ramification, in the distal domain. These functions, with and without interaction with UNI, also repressed the ramification of proximal pinnae in the absence of AF function. The expression of MFP and TL required UNI function. AF function was found to control leafblade architecture multifariously. The earlier identified role of AF as a repressor of UNI in the proximal domain was confirmed. Negative control of AF on the UNI-dependent pinna ramification in the distal domain was revealed. It was found that AF establishes a boundary between proximal and distal domains and activates formation of leaflet pinnae in the proximal domain.  相似文献
