目的:建立快速、灵敏测定大鼠血浆中5-羟基-7-(4-羟基-3-甲氧基苯基)-1-苯基-3-庚酮(DPHA)的LCMS/MS方法。方法:血浆样品经正己烷萃取后进行分析,采用Kinetex XB-C18色谱柱(2.10 mm×50 mm,2.6μm),柱温40℃,以水(含0.01%甲酸)-甲醇(含0.01%甲酸)为流动相梯度洗脱,流速0.30 m L/min,ESI离子源,多反应离子监测(MRM),用于定量分析的离子对为m/z 329.2→163.0,内标化合物益智酮甲为313.1→137.0。结果:DPHA的线性范围为1.0~2000.0 ng/mL(r=0.9996),最低定量限为1.0 ng/m L;提取回收率为73.53%~85.77%,基质效应为99.63%~110.50%;日内和日间精密度RSD均低于15%,重复性好。用该法测定静脉注射给药DPHA(1.0 mg/kg)后0.5 h大鼠血药浓度为36.2±5.1 ng/m L(n=4,RSD=14.0%)。结论:本法经方法学验证,适用于大鼠血浆中DPHA浓度的测定,适合药代动力学研究。  相似文献

采用HPLC法建立测定藜豆中左旋多巴含量的方法。色谱柱为岛津C-18甲基硅烷柱(250 min×4.6mm,5μm),流动相为甲醇:0.1 mol/L醋酸溶液(25:75,v/v),以硫唑嘌呤为内标物,检测波长280 nm,流速1.0 mL/min,柱温为室温,供试品的前处理方法为微波辅助提取法。结果表明,该方法准确度高,线性关系好,重现性好,平均加样回收率为97.34%,RSD 1.01%(n=5),结果满意。  相似文献

目的:建立快速、高效、灵敏的HPLC法测定大鼠血浆中的EGCG血药浓度。方法:以睾丸酮为内标物,血浆经盐酸和乙酸乙酯等去除蛋白。采用Agilent 20RBAX SB-C18色谱柱,乙腈-0.3%乙酸为流动相,流速1.0ml/min,检测波长为280nm,柱温40℃。结果:EGCG和内标的出峰时间分别为12.21min和15.4min。EGCG在0.05μg/ml-100μg/ml范围内线性关系良好(R2=0.9920),高、中、低3种浓度下的日内、日间精密度RSD均低于15%,相对回收率与绝对回收率均在100±15(%)范围内,稳定性好。结论:本方法灵敏、准确、高效,适用于大鼠血浆中EGCG的血药浓度检测。  相似文献

目的:建立高效液相色谱法测定大鼠血浆中来那度胺的浓度。方法:色谱柱采用VenusilASBC18column(4.6 mm×250mm,5μm)购自于博纳艾杰尔科技有限公司。样品采用梯度洗脱,流动相为乙腈和0.05%甲酸溶液,流速为1.0 mL/min,检测波长为254 nm,以沙利度胺为内标。结果:来那度胺血药浓度的线性范围为0.1-5μg/mL,最低检测限为0.1μg/mL,三个浓度的QC样品(0.2,1 and 5μg/mL)的提取回收率分别为78.8±3.8,80.1±3.2 and 79.1±7.6%。结论:此方法准确、简便、灵敏度高,对来那度胺的血药浓度测定和药物代谢动力学研究极具价值。  相似文献

采用气相色谱法测定抗毒素中的间甲酚含量。样品用苯甲醇作内标,经高氯酸沉淀蛋白、氯仿抽提后,通过HP-INNO-WAX柱,以氢火焰检测器测定间甲酚含量。内标峰与样品峰的分离度大于1.5,平均回收率及变异系数分别为100.067%和1.93%,测定线性范围0.0202~1.7588mg/ml,最小检测限为0.01272mg/ml,该方法准确快速,比化学方法好。  相似文献

目的:建立HPLC法测定赖氨葡锌颗粒中盐酸赖氨酸含量。方法:色谱柱为μBondpark C18柱,以甲醇:0.05mol/L醋酸钠和醋酸溶液(65:35)作为流动相,流速为1.0ml.min-1,检测波长为360nm。结果:盐酸赖氨酸在2.4~24.6μg/ml的浓度范围内呈良好的线性关系,平均回收率为99.28%,RSD=1.0%(n=6)。结论:建立的方法简便、快速、重现性好、结果准确,可用于赖氨酸的含量测定。  相似文献

建立了气相色谱法测定银杏内酯衍生物中甲醇、丙酮、乙腈、乙酸乙酯等有机溶剂残留量的方法.以苯为内标,样品经溶解直接进样测定,色谱柱为CP-SIL 24CB 毛细管柱(30 m×0.25 mm×0.25 μm).有机溶剂添加回收率在96.9%~99.6%之间,有机溶剂的最小检出限在0.29~0.48 μg /mL之间.该方法快速、准确、操作简便,能满足银杏内酯酯衍生物中甲醇等有机溶剂残留的定量检测.  相似文献

建立了同时测定石榴皮多酚中绿原酸、表儿茶素、鞣花酸和槲皮素含量的反相高效液相色谱方法。采用色谱柱Hypersil ODS2(250 mm×4.6 mm,5μm),其流动相为乙腈-0.4%磷酸溶液(体积比17∶83),流速为1.0m L/min,检测波长310 nm,柱温为30℃。建立安石榴甙测量方法,色谱柱为Hypersil ODS2(250 mm×4.6 mm,5μm);流动相为甲醇-2%冰醋酸(体积比7∶93);体积流量为1.0 m L/min;检测波长为232 nm;柱温25℃。结果表明:绿原酸、表儿茶素、鞣花酸、槲皮素和安石榴甙在一定浓度范围内与峰面积呈良好的线性关系,其平均回收率分别为100.66%(RSD=2.06%)、100.05%(RSD=0.58%)、100.05%(RSD=0.51%)、99.51%(RSD=1.22%)和99.79%(RSD=0.52%)。此结果说明反相高效液相色谱可用于测定石榴皮多酚类物质。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献
