There is currently great interest shown in understanding the process of embryogenesis and, due to the relative inaccessibility
of these structures in planta, extended studies are carried out in various in vitro systems. The culture of isolated zygotes
in particular provides an excellent platform to study the process of in planta embryogenesis. However, very few comparisons
have been made between zygotic embryos grown entirely in cultures and those grown in vivo. The present study analyses the
differences and similarities between the in vitro and in vivo development of wheat zygotic embryos at the level of morphology
and histology. The study was possible thanks to an efficient culture system and an appropriate method of preparing isolated
wheat zygotes for microscopy. The in vitro embryos were fixed, embedded and sectioned in the two-celled, globular, club-shaped
and fully differentiated stages. Embryos developing in vitro closely followed the morphology of their in planta counterparts
and their cell types and tissues were also similar, demonstrating the applicability of the present culture system for studying
the process of zygotic embryogenesis. However, some important differences were also detected in the case of in vitro development:
the disturbance of or lack of initial polarity led to changes in the division symmetry of the zygotes and subsequently to
the formation of uniform cells in the globular structures. Presumably, differences between the in vitro and in planta environments
resulted in a lower level of differentiation and maturation in in vitro embryos and in abundant starch and protein accumulation

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle. 相似文献

A meta-analysis was carried out in order to study the association of mycotoxins with performance and organ weights in growing pigs. A total of 85 articles published between 1968 and 2010 were used, totaling 1012 treatments and 13 196 animals. The meta-analysis followed three sequential analyses: graphical, correlation and variance–covariance. The presence of mycotoxins in diets was seen to reduce the feed intake by 18% and the weight gain in 21% compared with the control group. Deoxynivalenol and aflatoxins were the mycotoxins with the greatest impact on the feed intake and growth of pigs, reducing by 26% and 16% in the feed intake and by 26% and 22% in the weight gain. The mycotoxin concentration in diets and the animal age at challenge were the variables that more improved the coefficient of determination in equations for estimating the effect of mycotoxins on weight gain. The mycotoxin effect on growth proved to be greater in younger animals. In addition, the residual analysis showed that the greater part of the variation in weight gain was explained by the variation in feed intake (87%). The protein and methionine levels in diets could influence the feed intake and the weight gain in challenged animals. The weight gain in challenged pigs showed a positive correlation with the methionine level in diets (0.68). The mycotoxin effect on growth was greater in males compared with the effect on females. The reduction in weight gain was of 15% in the female group and 19% in the male group. Mycotoxin presence in pig diets has interfered in the relative weight of the liver, the kidneys and the heart. Mycotoxins have an influence on performance and organ weight in pigs. However, the magnitude of the effects varies with the type and concentration of mycotoxin, sex and the animal age, as well as nutritional factors. 相似文献

The effects of alternating magnetic field (AMF) with the frequency of 50 Hz on the dynamics of unfolding of cotyledon leaves, the composition and level of polar and neutral lipids and their component fatty acids (FA) were studied in 5-day-old radish seedlings (Raphanus sativus L. var. radicula D.L., cv. Rozovo-krasnyi s belym konchikom) grown in the light and in the dark. AMF weakened the inhibitory effect of light on unfolding of cotyledon leaves. In the light, the total content of lipids, as well as the level of polar and neutral lipids, in the seedlings in AMF was greater than in control material. In polar lipids, the total amount of glyco-and phospholipids increased; in neutral lipids, the level of triacylglycerols rose. The ratio between phospholipids and sterols (PhL/S) increased. In the dark, the total content of lipids and the level of neutral lipids in the seedlings in AMF were lower than in control material, and the ratio PhL/S decreased. In control material, there were no differences in the relative total content of unsaturated FA in the light and in the dark, whereas the level of linolenic acid was higher in the light than in the dark. AMF induced a decrease in the content of linolenic acid in the light and a rise in the dark; the level of erucic acid in the light decreased. The ratio between unsaturated and saturated FA decreased both in the light and in the dark. It was concluded that AMF with the frequency of 50 Hz was an adjusting agent considerably changing the content of lipids in the radish seedlings in the light and in the dark. 相似文献

The spined soldier bug, Podisus maculiventris, is a generalist predator of insects and has been used in biological control. However, information on the digestion of food in this insect is lacking. Therefore, we have studied the digestive system in P. maculiventris, and further characterized carbohydrases in the digestive tract. The midgut of all developmental stages was composed of anterior, median, and posterior regions. The volumes of the anterior midgut decreased and the median midgut increased in older instars and adults, suggesting a more important role of the median midgut in food digestion. However, carbohydrase activities were predominant in the anterior midgut. In comparing the specific activity of carbohydrases, α‐amylase activity was more in the salivary glands (with two distinct activity bands in zymograms), and glucosidase and galactosidase activities were more in the midgut. Salivary α‐amylases were detected in the prey hemolymph, demonstrating the role of these enzymes in extra‐oral digestion. However, the catalytic efficiency of midgut α‐amylase activity was approximately twofold more than that of the salivary gland enzymes, and was more efficient in digesting soluble starch than glycogen. Midgut α‐amylases were developmentally regulated, as one isoform was found in first instar compared to three isoforms in fifth instar nymphs. Starvation significantly affected carbohydrase activities in the midgut, and acarbose inhibited α‐amylases from both the salivary glands and midgut in vitro and in vivo. The structural diversity and developmental regulation of carbohydrases in the digestive system of P. maculiventris demonstrate the importance of these enzymes in extra‐oral and intra‐tract digestion, and may explain the capability of the hemipteran to utilize diverse food sources. 相似文献

Aquaporins (AQP) are integral membrane proteins that serve as channels in the transfer of water, and in some cases, small solutes across the membrane. They are conserved in bacteria, plants, and animals. Structural analyses of the molecules have revealed the presence of a pore in the center of each aquaporin molecule. In mammalian cells, more than 10 isoforms (AQP0-AQP10) have been identified so far. They are differentially expressed in many types of cells and tissues in the body. AQP0 is abundant in the lens. AQP1 is found in the blood vessels, kidney proximal tubules, eye, and ear. AQP2 is expressed in the kidney collecting ducts, where it shuttles between the intracellular storage sites and the plasma membrane under the control of antidiuretic hormone (ADH). Mutations of AQP2 result in diabetes insipidus. AQP3 is present in the kidney collecting ducts, epidermis, urinary, respiratory, and digestive tracts. AQP3 in organs other than the kidney may be involved in the supply of water to them. AQP4 is present in the brain astrocytes, eye, ear, skeletal muscle, stomach parietal cells, and kidney collecting ducts. AQP5 is in the secretory cells such as salivary, lacrimal, and sweat glands. AQP5 is also expressed in the ear and eye. AQP6 is localized intracellular vesicles in the kidney collecting duct cells. AQP7 is expressed in the adipocytes, testis, and kidney. AQP8 is expressed in the kidney, testis, and liver. AQP9 is present in the liver and leukocytes. AQP10 is expressed in the intestine. The diverse and characteristic distribution of aquaporins in the body suggests their important and specific roles in each organ. 相似文献

Phytoplankton, submerged macrophytes, zoobenthos and zooplankton communities were studied concurrently in three lochs which progressively increased in trophic state. The phytoplankton showed a change in dominance from Bacillariophyceae (Pennales) and Desmidaceae in the mesotrophic Loch of the Lowes to Bacillariophyceae (Pennales) and Cyanobacteria in eutrophic Balgavies Loch and to Bacillariophyceae (Centrales) and Chlorophyceae in hypertrophic Forfar Loch. The Nygaard Compound phytoplankton quotient rose from 1.2 in the Lowes to 12 in Forfar. There was a decrease in number of species of submerged macrophytes, and in depth of colonisation from 5 m in the Lowes to 2 m in Forfar which was proportional to the decrease in mean transparency. Macrophyte biomass, however, increased in proportion to the concentration of total phosphorus in the lochs. There was a decrease in species of littoral zoobenthos, but an increase in density of profundal benthos from the Lowes to Forfar. Zooplankton also showed an increase in density with enrichment, accompanied by changes in dominance. Calanoida were dominant in the Lowes; Cladocera in Balgavies and Cyclopoda in Forfar. The importance of these differences in understanding the effects of progressive enrichment and to the management of the lochs is discussed. 相似文献

The concentration of free amino acids and total nitrogen was studied in needles, stems and roots of seedlings of Pinus sylvestris L. for five weeks during the second growth period ("summer"). In one group of seedlings the source/sink relation was disturbed through removal of the terminal buds. The seedlings were cultivated in artificial year-cycles in a climate chamber.
Total nitrogen increased in needles and sterns of intact seedlings in the beginning of the "summer" and decreased during shoot growth. In seedlings, from which the buds had been removed, nitrogen remained at high levels in the primary needles and accumulated in steins and roots. The results are consistent with utilization of nitrogen in older needles and in the stem during shoot elongation.
The pool of free amino acids increased in the beginning of the "summer" and decreased after bud break in primary needles, stems and roots. Arginine and glutamine, in the roots also asparagine, were the dominating amino acids (amides included). Together, these compounds (plus glutamate and aspartate) contributed about 90% of the nitrogen in the amino acid pool in all organs. In primary needles and in the stem, arginine predominated at the end of hardening (75–85% of the amino acid nitrogen). Free amino acids contributed at most ca 10% of the total nitrogen in primary needles, where the ratio of free amino acid nitrogen: total nitrogen was highest at the end of dormancy and in the early "summer". Free amino acids accumulated after bud removal in primary needles and especially in stems and roots. Glutamine became relatively more dominant than arginine in the different organs.
The observations are consistent with the role of arginine and glutamine for storage and transport of nitrogen in conifers. Because of the low concentrations of amino acid nitrogen in the primary needles, arginine is not considered a major nitrogen reserve in needles of Scots pine seedlings. 相似文献

To examine whether the calcium accumulation in aged arteries is related to the way of walking, the mineral contents were determined
in the arteries of Japanese monkeys of quadrupedal walk by inductively coupled plasma-atomic emission spectrometry. Sixteen
Japanese monkeys consisting of 7 males and 9 females ranging in age from 2 to 33 yr were studied.
The accumulation of calcium, phosphorus, and magnesium occurred progressively in most, but not all, of the arteries with aging.
It was found that independent of the upper and lower limbs, a higher accumulation of calcium, phosphorus, and magnesium occurred
in the arteries of the proximal regions with aging, compared with the arteries of the distal regions.
In a comparison between the arteries of anatomically corresponding regions of the upper and lower limbs, the accumulation
of calcium and magnesium was 20–60% higher in the external iliac and femoral arteries of the lower limb than in the axillary
and brachial arteries of the upper limb. Regarding phosphorus, the accumulation was 20–120% higher in the external iliac and
femoral arteries than in the axillary and brachial arteries. It was known that in humans, the accumulation of calcium, phosphorus,
and magnesium was three to seven times higher in the arteries of the lower limb than in the arteries of the upper limb. It
is clear that there is a very significant difference in the accumulation of calcium and magnesium in the arteries of the lower
limbs between Japanese monkeys and humans. The present study suggests that the accumulation of calcium and magnesium in the
arteries of the lower limb with aging is affected by the way of walking. 相似文献

Abstract. Locusts show density-dependent continuous phase polymorphism; they appear in two forms or phases, gregarious and solitary, and there is a continuous range of intermediates between the extreme phases. Although earlier studies showed that there are no major phase-dependent differences in the titres of ecdysteroid in the haemolymph of desert locusts, Schistocerca gregaria , recent studies showed some minor differences in the timing of the main peak of ecdysteroids. In crowded penultimate- and last-instar hoppers, peak titres were lower but longer-lasting, whereas in isolated hoppers they were higher but of shorter duration. The major component of the haemolymph peak of ecdysteroid was 20-hydroxyecdysone in both isolated and crowded hoppers, but differences were found in the relative amounts of two minor components (makisterone A-like compound and highly polar products). In S. gregaria adults, the regression of the prothoracic glands was irregular and subject to high individual variations, but phase-dependent differences in the rate of regression were significant, and the adult glands did not produce physiologically significant amounts of ecdysteroids. Peak titres of ecdysteroid in the haemolymph were higher in isolated than in crowded adults. Similar to larvae, adults of the solitary phase contain more ecdysone in the haemolymph than those of the gregarious phase. Moreover, the phase characteristic titres of ecdysteroid in the adult stage can be shifted from one phase to another phase in response to appropriate changes in density. In contrast, the maximum amount of ecdysteroids in both ovaries and eggs was significantly higher in the gregarious than in the solitary phase. The amounts, and to some extent the types of ecdysteroids, were the only difference between ovaries and eggs from solitary and gregarious locusts. In addition, in newly hatched larvae, the amount of ecdysteroid was more than five times higher in gregarious than in solitary phase. 相似文献

By using a high-performance capillary electrophoresis method, the pharmacokinetics of the enantiomers of trans-tramadol (trans-T) and its active metabolite, trans-O-demethyltramadol (M1), was studied in healthy male and female Chinese volunteers after oral administration of 100 mg trans-T hydrochloride. The values of Cmax for the enantiomers of trans-T and M1, and AUC0- infinity for (-)-trans T, (+)-M1, and (-)-M1 were higher in females than in males. The values of V(d)/F for the enantiomers of trans-T and CLr for (+)-M1 were lower in females than in males. The value of t(1/2) for (-)-M1 was longer in females than in males. There were significant differences in pharmacokinetic parameters of the two enantiomers of trans-T or M1 both in males and in females. The (+)/(-)-enantiomeric ratios of t(max), V(d)/F for trans-T in males were significantly different from those in females and the (+)/(-)-enantiomeric ratios of pharmacokinetic parameters for M1 in males were similar to those in females. There are gender-related differences in the pharmacokinetics of the enantiomers of trans-T and M1 which may be due to the greater body weights for men and/or the higher CYP2D6 activity in women. The pharmacokinetics of trans-T and M1 is stereoselective in men and women. There is a gender-related difference in the stereoselectivity in pharmacokinetics of trans-T in human and the stereoselectivity in pharmacokinetics of M1 in men is similar to that in women. Changes in transpiration and stomatal conductance and other characteristics of water relations, growth rate, and ABA content
have been followed in short- and long-term experiments in two barley cultivars (cv. Michaelovsky and cv. Prairie) with contrasting
drought resistance characteristics. The aim of this work was to reveal the importance of stomatal behavior in salt tolerance
and also the involvement of ABA in its control. Salinity stress brought about a reduction in stomatal conductance in both
cultivars, but the effect was initially more pronounced in the drought-tolerant cv. Prairie than in the drought-sensitive
cv. Michaelovsky. The difference between the two cultivars changed with time, and later on transpiration and stomatal conductance
became higher in Prairie than in Michaelovsky. In both the short and the long term, the extent of stomatal closure due to
salinity correlated with the level of ABA accumulation in the leaves of the plants. Fast stomatal closure was likely to be
responsible for growth resumption after an initial arrest by salt treatment and for the maintenance of extension growth later
on, thus enabling its higher rate in Prairie than in Michaelovsky plants. Leaves of Prairie accumulated less toxic chloride
ions, which may be the result of a lower transpiration rate observed during the first phase of salt treatment. A subsequent
increase in stomatal conductance observed in Prairie is likely to ameliorate their gas exchange and maintain photosynthesis
and growth. Thus, differences between the cultivars in the stomatal response to salinity changed with time, which may be why
there are discrepancies in the attempts to relate stomatal conductance to salt tolerance observed in literature. 相似文献

It is noteworthy that in the rat the early postnatal life is marked by an activation of both the corticostimulating function
of the adenohypophysis in neonates of both sexes and of the gonadostimulating function mainly in males. In order to specify
if such neuroendocrine variations are temporally correlated with changes in the hypothalamic metabolism of neurotransmitters,
the hypothalamic metabolism of serotonin (5 HT), norepinephrine (NE), and dopamine (DA) and the hypothalamic content of neuropeptide
Y (NPY) have been investigated in newborn rats of both sexes, delivered at term by cesarean section, as well as changes in
the activity of both the hypothalamo-pituitary adrenal axis (HPA) and the hypothalamo-pituitary gonadal axis (HPG). Experimental
data suggested that 1) in males a rise in hypothalamic metabolism of 5 HT, NE and DA occurs during the first two hours after
delivery, whereas in females, only the metabolism of NE increases. Moreover, the postnatal metabolism of NE was higher in
females than in littermate males; 2) NPY content of the hypothalamus, which was at birth significantly higher in males than
in females, dropped in the former but not in the latter; 3) in newborn males, an early surge of plasma testosterone occurs,
suggesting postnatal activation of the HPG axis; on the other hand, in females, a late and slight increase in plasma estradiol
is observed; 4) in early postnatal life, a sex-independent rise in plasma ACTH and adrenal and plasma corticosterone levels
suggest a comparable activation of the HPA axis in newborns of both sexes. In conclusion, the early postnatal activation of
the corticostimulating function in neonates of both sexes and that of the gonadostimulating function, mainly in males, could
be temporally correlated with a rise in the hypothalamic metabolism of two neurotransmitters, 5 HT and NE, and of NPY content.
According to our data, a sex-dependent metabolsim of neurotransmitters in the hypothalamus is already apparent in early postnatal

The composition of lipids and fatty acids was determined at various developmental stages of gametophytes and sporophytes in the fern Adiantum capillus-veneris L. When the lipid content was expressed per g fresh weight, triacylglycerol was a major reserve lipid in spores and remained the dominant lipid in the gametophytes up to the 15th day after germination, but it was a minor one in the pinnae of sporophytes. In contrast, triterpenoids were abundant in sporophytes but not detected in gametophytes. When the content of fatty acids was expressed in percentage of the total fatty acids, the content of arachidonic acid in phosphatidylcholine and linolenic acid in diacyglyceryltrimethylhomoserine and phosphatidylcholine was significantly higher in both pinnae and petioles of sporophtes than in gametophytes. Significant differences in fatty acid composition of phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol and triacylglycerol were also noted between gametophytes and pinnae of sporophytes. All these differences were attributed to the difference of generation rather than tissue differences or age.
The content of monogalactosyldiacylglycerol, hexadecatrienoic acid in monogalactosyldiacylglycerol, and 3- trans hexadecenoic acid in phosphatidylglycerol was higher in the pinnae of sporophytes than in gametophytes or in young leaves of sporophytes. Electron microscopy of the ultrastructure of chloroplasts indicated that the high content of these components was consistent with the development of chloroplasts in pinnae. 相似文献

细胞周期蛋白cyclin G1(CCNG1)是一个重要的细胞周期调控因子,参与哺乳动物颗粒细胞增殖、卵母细胞成熟等繁殖生物学过程,但其在绵羊中鲜有报道。为了研究CCNG1基因对绵羊发情调控以及季节性繁殖的影响,文章对绵羊CCNG1基因进行了克隆和组织表达谱分析,利用Real-time PCR对该基因在多浪羊(常年发情)与中国美利奴羊(季节性繁殖)发情周期不同阶段性腺轴组织的表达变化进行了实时检测。获得了绵羊CCNG1基因部分cDNA序列,其中编码区全长885 bp,编码294个氨基酸。CCNG1蛋白结构经预测存在多个磷酸化位点和蛋白激酶C磷酸化位点。CCNG1基因在所检测绵羊各组织中均有表达,但在卵巢与肾脏中为高丰度表达;CCNG1在不同绵羊品种发情周期不同阶段性腺轴组织的表达变化规律基本一致,卵巢、子宫、松果体、垂体均是在发情期达到峰值。但是在发情期和发情后期卵巢CCNG1的表达量存在显著的品种间差异(P<0.01)。研究结果表明,CCNG1可能通过参与卵泡的生长发育继而达到对绵羊发情和季节性繁殖的调控。 相似文献

During a combined research project at several stations along the Lower Saxony coast (German North Sea) antifouling biocides
were analysed in water, sediment and biota. Pathological alterations in blue mussel, Pacific oyster and periwinkle found in
the harbour of Norderney and a reference station are presented here and discussed on the background of chemical analyses.
The molluscan species from the reference station Borkum East flat did not show any pathological effects in central organs,
except those provoked by an infestation in the gastro-intestinal tract by the copepod Mytilicola
intestinalis and trematode larvae. In most animals, the metacercaria were found in the interstitial tissue without any inflammatory reaction.
In a minor number of specimens, an inflammatory reaction in the mucosa and sub-mucosa of the intestine occurred in association
with Mytilicola infestation. These reactions may be evoked through mechanical irritation of the gut epithelium, metabolic products of the
parasites or invading bacteria. In contrast to the observed pathological changes of mussels, oysters and periwinkles in Norderney
harbour were not found to be associated with parasitic infestation. The most prominent pathological alterations were observed
in the digestive system and in the gonad. In the gastro-intestinal tract inflammatory reactions, atrophy and necrosis of tubules
in the mid gut gland were most pronounced in spring at the beginning of the pleasure boat season in the Pacific oyster and
to a minor degree in the blue mussel and the periwinkle. The latter displayed additional inflammatory and necrotic processes
in the gills. Especially in the gonad, an elevated resorption rate of gametes was present in the Pacific oyster and in the
periwinkle. In addition, impact of organotin compounds was reflected in an intersex index of up to 1.4 in Littorina littorea in coincidence with masculinization of the reproductive organs. 相似文献

为了解长江口新生湿地大型底栖动物时空变化格局,2004年11月—2005年10月对九段沙湿地的3个沙洲以及各主要生境进行了调查。结果表明:九段沙湿地大型底栖动物密度和生物量均呈春季最高,夏、秋季下降,秋季末和冬季回升的趋势,整体生物多样性水平较低,分布不均匀。夏季上沙和中沙底栖动物密度存在显著差异,四季生物密度和生物量空间梯度为下沙>上沙>中沙。由物种组成分析可知,九段沙湿地大型底栖动物群落时空变化与优势种的消长有关。各季海三棱藨草带和藨草带底栖动物密度和生物量最高,芦苇和互花米草次之,光滩区域最低,且群落季节波动大。外来物种互花米草生境下底栖动物群落特征与土著种芦苇生境下无显著差异。 相似文献

Abstract. The similarity in species composition between seed bank and vegetation was analysed in Mediterranean grasslands in relation to altitude, topography and grazing. Soil samples were collected in permanent plots in autumn at the end of the summer drought period and in spring, before the new seed fall and after the natural winter seed stratification. The seed bank composition was determined by greenhouse germination over a nine-month period. Presence/absence of species in the standing vegetation throughout the complete annual cycle, and the percentage area of bare ground in October, were recorded in the same plots. The species composition of the standing vegetation is clearly determined by altitude, topography and grazing, while the floristic composition of the seed banks is only related to altitude and topography in the case of autumn seed bank and with any of the three factors in the spring seed bank. Relative abundances of grasses, legumes and forbs also show different patterns in vegetation and seed bank data. Sørensen similarity between the autumn seed bank and the vegetation declines as altitude rises, but there are no significant differences for topography and grazing. This similarity decreases in the case of the spring seed bank and does not show any significant relationship with any of the factors. The perennial/ annual ratio and the proportion of bare soil in October are proposed as explanatory variables in a predictive model of similarity between the seed composition of the seed bank and vegetation. 相似文献
