Paleolimnological techniques were employed to document the limnological histories of the aurora trout lakes, located in the Sudbury region of Ontario. Two of these lakes are of special interest to fisheries managers, as they represent the only known native habitats of a rare strain of brook trout: the aurora trout. These lakes were limed as part of restoration efforts. Stratigraphic changes in diatom and chrysophyte assemblages from dated lake sediment cores indicate that all the lakes have been impacted by anthropogenic acidification, although the timing and the magnitude of acidification were different amongst the lakes. For example, Whirligig Lake was likely the most naturally acidic lake in the past, but it had further acidified since about 1960. This lake was limed in 1989 and then again in 1993. In Whitepine Lake, acidification started 1940; however, in the most recent sediments (1992), some recovery in lakewater acidity has occurred. In Little Whitepine Lake (a reference lake), acidification started earlier (1920) and the lakewater pH continued to decline until about 1990. This lake was limed in 1989. The chrysophyte paleoindicators suggest a recent recovery in this lake. The successful re-introduction of aurora trout in Whirligig and Whitepine lakes is undoubtedly related to the improved water quality through liming but, based on our paleolimnological indicators, the lakes' limnological characteristics (e.g. pH and metal concentrations) are still different from those present before atmospheric deposition of strong acids from the Sudbury smelters.  相似文献

Summary Freeze-fracture studies were conducted on the membranes of normal cockroach hemocytes. The plasmalemma is asymmetric with the A fracture face containing 80–100 Å membrane intercalated particles at a concentration of 2500/2. The B fracture face contains 120–150 Å particles with a relatively low density (800/2). The nuclear envelope displays an asymmetry with the A fracture face containing 1500 particles/2 and the B face containing 300/ 2. No significant particle size differences were observed in nuclear envelope fracture faces. Two types of symmetric membranes were also found in these cells. Both A and B fracture faces of the membrane surrounding the numerous cytoplasmic inclusion bodies contain particle sizes and concentrations similar to the B face of the plasmalemma. A second type of symmetry was observed in cells apparently engaged in exocytosis. Vesicles (0.1 D) from this process were completely particle free on both fracture faces. The methane cycle in constructed wetlands without plants and withPhragmites australis (reed) and Scirpus lacustris (bulrush) wasinvestigated. Variations in CH4production largely determined variations in CH4 emission among the systems, rather than variations inCH4 storage and oxidation. Twofoldlower CH4 production rates in theScirpus system (5.6–13 mmol m-2 d-1) relative to the control (16.7–17.6 mmolm-2 d-1) were accompanied by a lower contribution ofmethanogenesis to organic carbon metabolism (20% for Scirpus vs.80% for control). Sedimentary iron(II) reservoirs were smallerin the Scirpus than control sediment (300 vs. 485 mmol.m-2) and a shuttle role for iron asan intermediate between root O2release and carbon oxidation, attenuating the availability of substrate formethanogens, is suggested. Differences in CH4 production among the Phragmites and Scirpus systemswere controlled by the interspecific variation in sediment oxidationcapacities of both plant species. Comparatively, in the Phragmites sediment,dissolved iron reservoirs were larger (340 mmol.m-2) and methanogenesis was a more importantpathway (80%). Methane transport was mainly plant mediated inthe Phragmites and Scirpus systems, but ebullition dominated in thenon-vegetated control systems as well as in the vegetated systems when plantbiomass was low.  相似文献

The exudation of certain organic anions and protons by roots which may affect solubility of metals and P and uptake by plants, is affected by nitrogen form and pH. The objective of this work was to study exudation of carboxylates and H+/OH by tomato plants in response to NH4/NO3 ratio and pH in nutrient solution. Four NH4/(NH4+NO3) ratios (R= 0, 0.33, 0.67 and 1) and constant vs. variable solution pH treatments were investigated. The sum of the exudation rates of all carboxylates tended to decline with increasing R, particularly tri- and dicarboxylates. The molar fraction of the exuded tri- and dicarboxylates, averaged over all treatments and plant ages, increased in the order tartarate 2%), malate (6%), succinate (15%), citrate (26%) and fumarate (46%). At R=1 the solution pH dropped from 5.2 to 3 and at R=0 increased to 8. The R corresponding to the pH stat of tomato plant was 0.3. For the constant solution pH treatment, the effect of solution pH on carboxylate exudation rate was small as compared to the effect of R. The exudation of citrate and H+ efflux which were initiated when NO3 and NH4 uptake rates per plant exceeded certain threshold values, increased with plant age.  相似文献

Summary Parenchymal cells of oat (Avena sativa) coleoptiles had an osmotic concentration of 410 mM (determined by plasmolysis); of this only 22 mM was K+ and 1 mM Na+ (flame photometry). Cells were impaled with micropipette electrodes. Iontophoretic injection of the dye Niagara sky-blue from the micropipette showed that the tip of the electrode penetrated the vacuole. When sections of tissue were immersed in a solution of 22 mM KCl, 1 mM CaCl2, and 50 mM glucose, average membrane potential was found to be 38.5 mV inside negative specific membrane resistance was 510 cm2, and specific membrane capacitance, 2 f cm-2. The cell membranes showed <25% retification and no electrical excitability. Electrotonic coupling of adjacent cells could not be demonstrated.  相似文献

Concentrations of Al, Fe, Mn, Cd, Cu, Pb, and Zn were measured in thereference and treatment basins of Little Rock Lake (Vilas County, Wisconsin), alow-alkalinity, seepage system (pH 6.1, alkalinity25eq/L) during six years of a whole-basinacidificationand the first four years of the lake's recovery. The treatment basin wasacidified with H2SO4 in three two-year steps to pH5.6, 5.1, and 4.7. By the end of year 4 of recovery, treatmentbasin pH increased to 5.3 as a result of internal alkalinity generation.During acidification, dissolved Mn and Fe (0.4mpore-size filters) increased at pH 5.6; dissolved Al, Cd, and Zn becameelevated at pH 5.1; and dissolved Pb at pH 4.7. Dissolved Cu remainedsimilar in both basins to pH 4.7. Al, Fe and Mn levels declinedsignificantly during the recovery period, approaching values at pH 5.3intermediate between the concentrations at pH 5.6 and 5.1 during acidification.Dissolved Al and Fe in the reference basin were near the equilibrium levels forsolubility of gibbsite (Al(OH)3) and amorphousFe(OH)3(s).The acidified basin was undersaturated relative to gibbsite, and dissolved Alwas limited by pH disequilibrium between the water column and sediments andpossibly by Al-DOC precipitation. Dissolved Fe apparently was controlled bysolubility of amorphous Fe(OH)3(s) and Fe-DOC precipitation.Dissolved Mn levels in both basins were consistent with manganite[-MnOOH(s)] solubility. Elevated levels of Cd, Pb, and Zn in thetreatment basin during acidification probably resulted from less efficientscavenging of atmospherically-deposited Cd, Pb, and Zn by settling particles.  相似文献

Combined paleolimnological investigations of a 1.8 m sediment core from the deepest north-western part of Lake Ladoga show stratigraphic changes in granulometric and chemical composition, organic matter content, diatom species composition and chlorophyll a concentration. The sediment accumulation rate was calculated and 5 stages of lake history over the last 4 000 years were described. Changes in lake environment were mainly caused by changes in climate and lake water balance. Human impact on the lake was also traced.  相似文献

Ministik Lake (longitude 113°01; latitude 53°21), a well-mixed, shallow (mean depth 1.83 m), eutrophic lake is characterized by eutrophic chlorococcalean and cyanophycean phytoplankton associations, and little change in standing crop size with increasing depth. Standing crops and primary productivity are low during the winter but pronounced spring and late summer/early autumn maxima occur. Mean yearly areal standing crop (B) and primary productivity (A) were 199.2 mg m–2 chlorophyll a and 319.5 mg C hr–1 m–2 respectively. Annual productivity was estimated at 1399,6 g C m–2yr–1. The mean increase in the extinction coefficient () per unit increase in standing crop (B) was 0.03 In units m–1. High non-algal light attenuation (q) occurred averaging 46% which prevented the ratio B from attaining more than 74.2% of the theoretical maximum except once when selfshading occurred. Insignificant relationships existed between B (mg m–3 chlorophyll a) and Amax (mg C hr–1 m–3), A and B and A and B. Close correlations existed between A and Amax/ and A and I0 (W m–2). The depth of the euphotic zone (Zeu) varied between 0.6 and 2.0 m; the average relationship between zeu and was zeu = 3.78/te, and the mean standing crop in the euphotic zone represented 58.3% of the theoritical maximum. The high q values made the model of Talling (1957) inapplicable to this lake. The Q10 value for the lake was 1.20. max (mg C chlorophyll a –1 hr–1) was closely related to both temperature and irradiance, and depressed by high pH values.Growth of the cyanophycean algae was correlated with temperature, chlorophycean algae with phosphate-phosphorus and temperature and the diatoms with dissolved silica and phosphate-phosphorus, but only in the case of Chaetoceros elmorii did any nutrient appear limiting. Indirect evidence that free CO2 limited photosynthetic rates is provided by the max:pH relationship.  相似文献

It has been suggested that very large impact events ( 500 km diameter impactors) sterilized the surface of the young Earth by producing enough rock vapor to boil the oceans. Here, we consider surface heating due to smaller impactors, and demonstrate that surface temperatures conducive to organic synthesis resulted. In particular, we focus on the synthesis of thermal peptides. Previously, laboratory experiments have demonstrated that dry heating a mixture of amino acids containing excess Asp, Glu, or Lys to temperatures 170 °C for 2 hours yields polypeptides. It has been argued that such temperature conditions would not have been available on the early Earth. Here we demonstrate, by analogy with the K/T impact, that the requisite temperatures are achieved on sand surfaces during the atmospheric reentry of fine ejecta particles produced by impacts of bolides 10–20 km in diameter, assuming 1 – 100 PAL CO2. Impactors of this size struck the Earth with a frequency of 1 per 104 – 105 y at 4.2 Ga. Smaller bolides produced negligible global surface heating, whereas bolides > 30 km in diameter yielded solid surface temperatures > 1000 K , high enough to pyrolyze amino acids and other organic compounds. Thus, peptide formation would have occurred globally for a relatively narrow range of bolide sizes.  相似文献

Of 104 yeasts tested for the hydrolysis of rac-linalyl acetate and rac--terpinyl acetate twelve hydrolysed rac-linalyl acetate. Five of these belonged to five different species of Geotrichum. The most promising strain, Geotrichum capitatum CBS 0572.82, yielded (S)-linalool in 56% enantiomeric excess after 6h (E value 5, c = 32%). The cellular resistance of representatives from four euryhaline poikilosmotic bivalves caught in Büsum (North Sea, 30 S), Kieler Förde (Belt Sea, 15 S), and Tvärminne (Gulf of Finland, 6 S) has been tested. All experiments were performed in October.2. According to their cellular salinity rangesMacoma baltica andMytilus edulis are more euryhaline thanMya arenaria andCardium edule.3. The cellular heat resistance decreases in specimens from the North Sea in the following order:Cardium, Mya, Macoma, Mytilus. Among the representatives collected near Tvärminne,Macoma has the highest heat resistance.4. The cellular freezing resistance in specimens from the North Sea is highest inMytilus. The bivalves of the Gulf of Finland practically lack resistance to freezing.5. In localities of decreased salinity the cellular resistance of these bivalves to a dilution of the external medium is somewhat increased, but simultaneously the cellular resistance to other environmental factors, such as heat, freezing and high salinity, decreases unspecifically.

Kurzfassung An vier euryhalinen poikilosmotischen Muschelarten aus Büsum (Nord-seeküste, 30 S), der Kieler Förde Beltsee, 15 S) und Tvärminne (Finnischer Meerbusen, 6 S), wurden zur gleichen Jahreszeit (Herbst) vergleichende Untersuchungen über die zellulären Resistenzgrenzen durchgeführt. Auf Grund ihrer zellulären Salzgehaltsbereiche sindMacoma baltica undMytilus edulis euryhaliner alsMya arenaria undCardium edule. Die zelluläre Hitzeresistenz nimmt bei den Individuen aus der Nordsee in der Reihenfolge:Cardium, Mya, Macoma, Mytilus ab. Bei Exemplaren aus Tvärminne ist die zelluläre Hitzeresistenz vonMacoma am größten. Die zelluläre Gefrierresistenz ist unter den Muscheln aus der Nordsee am größten, fehlt dagegen praktisch bei den Tieren aus dem Finnischen Meerbusen. Bei verringertem Salzgehalt des Fundortwassers ist die zelluläre Resistenz der untersuchten Muscheln gegenüber Verdünnung des Außenmediums jeweils etwas erhöht, gleichzeitig nimmt die zelluläre Resistenz gegenüuber anderen Außenfaktoren wie Hitze, Frost und hohen Salzkonzentrationen unspezifisch ab.

Summary Two deciduous tree species (yellow poplar and red maple) on Walker Branch Watershed (WBW), near Oak Ridge, Tennessee, were radiolabeled with 35S (87 day halflife) to study internal cycling, storage, and biogenic emission of sulfur (S). One tree of each species was girdled before radiolabeling to prevent phloem translocation to the roots, and the aboveground biomass was harvested prior to autumn leaf fall. Aboveground biomass, leaf fall, throughfall, and stemflow were sampled over a 13 to 24 week period. Sulfur-35 concentrations in tree leaves reached nearly asymptotic levels within 1 to 2 weeks after radiolabeling. Foliar leaching of 35S and leaf fall represented relatively unimportant return pathways to the forest soil. The final distribution of 35S in the nongirdled trees indicated little aboveground storage of S in biomass and appreciable (>60%) capacity to cycle S either to the belowground system by means of translocation or to the atmosphere by means of biogenic S emissions. Losses of volatile 35S were estimated from the amount of isotope missing (33%) in final inventories of the girdled trees. Estimated 35S emission rates from the girdled trees were 10-6 to 10-7 Ci cm-2 leaf d-1, and corresponded to an estimated gaseous S emission of approximately 0.1 to 1 g S cm-2 leaf d-1. Translocation to roots was a significant sink for 35S in the red maple tree (40% of the injected amount). Research on forest biogeochemical S cycles should further explore biogenic S emissions from trees as a potential process of S flux from forest ecosystems.  相似文献

Summary The electrical transients produced by chloroplast bilayer lipid membranes (Chl-BLM) from flash excitation are seen to result from three photocurrents and a discharge current. Each of the three photo-initiated charge transports in Chl-BLM (designated as Components A, B and C) exhibits an action spectrum similar to chlorophyll absorption spectra. The fast components (A and B), which are induced by electron acceptors such as Fe+3, have rise-times of 3 sec and 20 msec, and occur in TLM (thin lipid membranes, i.e., colored membranes up to 1 thick) as well as in BLM. Component C is induced by a transmembrane pH difference or applied voltage, has a rise-time of 1 sec, and occurs only in BLM. Component C is associated with exciton dissociation and proton transport. The mobility of the Component A current carriers in TLM is estimated to be about 1×10–2 cm2/volt sec, and are, hence,electronic. The photovoltage waveforms are described by equations developed, which consider Component A as being caused by a direct charge separation proportional to the illumination intensity (within 0.5 sec), and Components B and C being caused by two types of exciton processes which cause charge transport after the illumination period.  相似文献

The effect of salicylic acid (SA) on peroxidase activity in wheat (Triticum aestivum L.) calli cocultured with the bunt pathogen Tilletia caries was studied. Fungal infection was shown to activate cytoplasmic peroxidase. SA suppressed total peroxidase activity but did not inhibit the peroxidase with pI 9.8. A novel chitin-specific peroxidase with pI 3.5 appeared after the SA treatment. The infection of SA-treated cells with Tilletia caries activated the isoenzymes with pI 3.5, 4.8, and 7.5 and stimulated their secretion into the culture medium. The ability of SA to control wheat peroxidase activity during pathogenesis is discussed. The important role of this control in plant defense responses to the bunt pathogen is emphasized.  相似文献

1,3-Fucosyltransferase solubilized from human liver has been purified 40 000-fold to apparent homogeneity by a multistage process involving cation exchange chromatography on CM-Sephadex, hydrophobic interaction chromatography on Phenyl Sepharose, affinity chromatography on GDP-hexanolamine Sepharose and HPLC gel exclusion chromatography. The final step gave a major protein peak that co-chromatographed with 1,3-fucosyltransferase activity and had a specific activity of 5–6 µmol min–1 mg–1 and anM r 44 000 deduced from SDS-PAGE and HPLC analysis. The purified enzyme readily utilized Gal1-4GlcNAc, NeuAc2-3Gal1-4GlcNAc and Fuc1-2Gal1-4GlcNAc, with a preference for sialylated and fucosylated Type 2 acceptors. Fuc1-2Gal1-4Glc and the Type 1 compound Gal1-3GlcNAc were very poor acceptors and no incorporation was observed with NeuAc2-6Gal1-4GlcNAc. A polyclonal antibody raised against the liver preparation reacted with the homologous enzyme and also with the blood group Lewis gene-associated 1,3/1,4-fucosyltransferase purified from the human A431 epidermoid carcinoma cell line. No cross reactivity was found with 1,3-fucosyltransferase(s) isolated from myeloid cells. Examination by Northern blot analysis of mRNA from normal liver and from the HepG2 cell line, together with a comparison of the specificity pattern of the purified enzyme with that reported for the enzyme expressed in mammalian cells transfected with theFuc-TVI cDNA, suggests a provisional identification ofFuc-TVI as the major 1,3-fucosyltransferase gene expressed in human liver.Died June, 1991  相似文献

The summer stratification phosphorus budget for eutrophic Edinboro Lake in northwestern Pennsylvania was determined. Phosphorus loading from internal sources contributed 141 kg, (79%) and 55 kg, (68%) of the mass phosphorus increase in the lake in 1981 and 1982, respectively. Calculated anaerobic sediment release rates of total phosphorus were 9.9 and 3.7 mg m–2 day–1 for these two years. The observed summer maximum chlorophyll a concentration was 1.5–3 times greater than that predicted by existing models. Year-to-year variability in the internal phosphorus load for this lake and others is discussed. Without a data base that will permit the comparison of lakes and with and without a significant supply of internal phosphorus, prediction of the relative importance of internal loading in a particular lake will be difficult.  相似文献

Understanding the factors regulating the concentrations of basic cations in soils and surface waters is critical if rates of recovery are to be predicted in response to decreases in acidic deposition. Using a dynamic simulation model (PnET-BGC), we evaluated the extent to which atmospheric deposition of strong acids and associated leaching by strong anions, atmospheric deposition of basic cations through changes in emissions of particulate matter, and historical forest cutting have influenced soil pools of exchangeable basic cations and the acid-base status of stream water at the Hubbard Brook Experimental Forest (HBEF) in New Hampshire. Historical deposition of basic cations was reconstructed from regression relationships with particulate matter emissions. Simulation results indicate that the combination of these factors has resulted in changes in the percent soil base saturation, and stream pH and acid neutralizing capacity (ANC) from pre-industrial estimates of 20%, 6.3 and 45 eq L–1, respectively, to current values of 10%, 5.0 and –5 eq L–1, respectively. These current values fall within the critical thresholds at which forest vegetation and aquatic biotic are at risk from soil and surface water acidification due to acidic deposition. While the deposition of strong acid anions had the largest impact on the acid-base status of soil and stream water, the reduction in deposition of basic cations associated with reductions in particulate emissions was estimated to have contributed about 27% of the depletion in soil Ca2+ exchange pool and 15% of the decreases in stream water concentrations of basic cations. Decline in stream water concentrations of basic cation occurred under both increasing and decreasing exchangeable pools, depending on the process controlling the acid base status of the ecosystem. Model calculations suggest that historical forest cutting has resulted in only slight decreases in soil pools of exchangeable basic cations, and has had a limited effect on stream ANC over the long-term.  相似文献

The present study evaluates the growth response of two strains of filamentous fungi; a Fusarium sp. and Alternaria tenuis, grown on both solid and liquid Czapek Dox medium amended with different concentrations of CdCl2. Colony extension and the mycelial dry weight of both fungi were significantly inhibited by high concentrations of cadmium. Extended lag phases and low growth rates resulted from cadmium administration. Cadmium drastically affected fungal morphogenesis by the production of stunted sterile thick mycelial filaments of the Fusarium sp. and chains of uncharacterized swellings instead of conidia in A. tenuis. Experiments showed that cadmium accumulation by the Fusarium sp. grown in liquid medium was a concentration dependent, and over the incubation time it displayed a plateau pattern. The cells grown on medium containing 0.25 mmol l–1 CdCl2 accumulated up to 89 ± 12 mol Cd (gm dw)–1 after two days, falling to 29 ± 10 mol Cd (gm dw)–1 after five days. At 0.5 mmol l–1 CdCl2 treatment the maximum cellular cadmium content was 132 ± 14 mol (gm dw)–1, attained after 3 days, and decreased to 98 ± 9 mol (gm dw)–1 at the end of the incubation time. There was a simultaneous marked drop in cadmium content and pH of the growth medium during the first few days. The presence of cadmium markedly altered the cellular essential cations; K+ and Mg2+ being decreased while Na+ increased during the growth period. Such findings resulted a reverse pattern of cellular Na+/K+ ratio for cells grown on cadmium-containing medium in respect to the control treatment. The results are discussed in relation to a further dimension of cadmium effects that might reflect its toxicity, as well as the implication of cadmium extrusion for tolerance during fungal growth.  相似文献

Stable isotopes (13C, D) and radiocarbon weremeasured in methane bubbles emitted from rice paddies and swamps in southernThailand. Methane emitted from the Thai rice paddies was enriched in13C (mean 13C; –51.5 ±7.1 and–56.5 ± 4.6 for mineral soil and peat soil paddies,respectively)relative to the reported mean value of methane from temperate rice paddies(– 63 ± 5). Large seasonal variation was observed in13C(32) in the rice paddies, whereas variationinD was much more smaller (20), indicating that variation in13C is due mainly to changes in methane production pathways.Values of 13C were lower in swamps (–66.1 ±5.1)than in rice paddies. The calculated contribution of acetate fermentation from13C value was greater in rice paddies (mineral soils:62–81%, peat soils: 57–73%) than in swamps (27–42%). Din methane from Thai rice paddies (–324± 7 (n=46)) isrelativelyhigher than those from 14 stations in Japanese rice paddies ranging from–362 ± 5 (Mito: n=2) to –322 ± 8(Okinawa: n=3), due tohigher D in floodwaters. 14C content in methane produced fromThai rice paddies (127±1 pMC) show higher 14Cactivity compared with previous work in paddy fields and those from Thai swamps(110±2 pMC).  相似文献

Domain structures of class I and class II MHC products were analyzed from a viewpoint of amino acid and nucleotide sequence homologies. Alignment statistics revealed that class I (transplantation) antigen H chains consist of four mutually homologous domains, and that class II (HLA-DR) antigen and chains are both composed of three mutually homologous ones. The N-terminal three and two domains of class I and class II (both and ) gene products, respectively, all of which being 90 residues long, were concluded to be homologous to 2-microglobulin (2M). The membraneembedded C-terminal shorter domains of these MHC products were also found to be homologous to one another and to the third domain of class I H chains. Class I H chains were found to be more closely related to class II chains than to class II chains. Based on these findings, an exon duplication history from a common ancestral gene encoding a 2M-like primodial protein of one-domain-length up to the contemporary MHC products was proposed.  相似文献
