The presence of prostaglandins (PGs) was determined in gastric juice obtained from 3 conscious dogs, provided with a chronic gastric fistula. Outputs of acid (mequiv min?1) and PGs (pg min?1) were measured in gastric secretions stimulated by pentagastrin (100 or 200 ng kg?1min?1). Prostaglandin activity was estimated, after extraction and thin layer chromatography, by radioimmuno-assay of the PGB formed by treatment with alkali. Tritiated PGs were added to gastric juice for the purpose of correcting for PGs recovery. Using this method, the minimum mass of PGB which could be satisfactorily distinguished from zero was 25 pg. Prostaglandins A2 and E2 were present in pentagastrin-activated gastric secretions and averaged (mean ± SE, n = 8) 200.7 ± 18.1 and 260.1 ± 18.0 pg min?1 respectively. The identity of PGA2 and PGE2 was confirmed by gas liquid chromatography combined with mass spectrometry. The amount of PGE2 converted to PGA2 during extraction, separation and conversion procedures was estimated from the amount of [3H] PGA2 found when only [3H] PGE2 had been added to a sample of gastric juice and averaged 14.5% ± 2.0. Our preliminary results support the possibility that PGE2 and PGA2 may be of physiological importance in the regulation of canine gastric secretions.  相似文献

Three clinically efficacious vasodilatory drugs were found to be selective inhibitors of thromboxane A2 biosynthesis. Hydralazine, dipyridamole, and diazoxide inhibited platelet aggregation at 1 × 10?4 M, 1.75 × 10?4 M, and 2 × 10?3 M respectively. Their relative inhibitory potencies on thromboxane B2 production in human platelet microsomes were examined and found to be similar to that observed for their inhibition on human platelet aggregation. At 10?3 M, hydralazine, dipyridamole, and diazoxide inhibited thromboxane B2 formation by 65 percent, 27 percent and 18 percent respectively. These compounds were examined in the sheep vesicular gland system, and they were shown not to be inhibitors of the cyclooxygenase enzyme. Thus, the inhibition of thromboxane A2 biosynthesis by these three drugs in human platelet microsomes appeared to be specific at the thromboxane synthetase level.  相似文献

Ventilation was measured directly in the hagfish, Myxine glutinosa L., by means of an electro-magnetic blood flowmeter. Ventilatory flow and frequency increased from 0.86 ± 0.27 ml·min?, and 18.2 ± 5.1·min?, respectively, at 7°C to 1.70 ± 0.20 ml·min?, and 70.1 ± 9.5·min? at 15 ·C.Standard oxygen consumption,V?O2, was measured in non-buried hagfish. V?O2 was 0.57 ± 0.17μl O2·g?1·min?1 at 7°C, and 0.85 ± 0.12μl O2·g?1·min?1 at 15°C.  相似文献

The production of vasodilatory, antiaggregatory prostacyclin (PGI2) and vasoconstrictory, proaggregatory thromboxane A2 (TxA2) by the placenta was studied in the cases of hypertensive pregnancy complications by superfusing pieces from maternal and fetal sides of placentae of 9 pre-eclamptic, 6 hypertensive and 11 healthy women and measuring the release of 6-keto-prostaglandin F (6-keto-PGF) and thromboxane B2 (TxB2), the breakdown products of PGI2 and TxA2 respectively, from the superfusate. Both sides of the placentae from the controls produced 6-keto-PGF (maternal side 0.5±0.1 ng/g/min dry weight of tissue, mean±SEM; fetal side 0.7±0.2 ng/g/min) and TxB2 (maternal side 2.5±0.4 ng/g/min; fetal side 2.7±0.5 ng/g/min with no correlation between the two. The 6-keto-PGF production was normal in hypertensive complications whereas the TxB2 production was increased on the fetal side of the placentae obtained from the pre-eclamptic (3.7±0.3 ng/g/min: p<0.05) and hypertensive women (4.1±0.4 ng/g/min; p<0.025). This may explain the occurrence of microthrombi and infarctions in placentae of hypertensive women.  相似文献

Platelet rich plasma transforms exogenous prostaglandin endoperoxide H2 into thromboxane A2 immediately prior to the initiation of irreversible aggregation. Selective thromboxane synthetase inhibitors block thromboxane A2 formation and aggregation. Thromboxane A2 formation appears to be essential during arachidonate mediated aggregation. The results presented reconcile the previously accepted paradoxical behavior of thromboxane synthetase in platelet rich plasma toward the prostaglandin endoperoxide H2 substrate.  相似文献

The effects of CO2 enrichment on photosynthesis and ribulose‐1,5‐bisphosphate carboxylase/oxygenase (rubisco) were studied in current year and 1‐year‐old needles of the same branch of field‐grown Pinus radiata D. Don trees. All measurements were made in the fourth year of growth in large, open‐top chambers continuously maintained at ambient (36 Pa) or elevated (65 Pa) CO2 partial pressures. Photosynthetic rates of the 1‐year‐old needles made at the growth CO2 partial pressure averaged 10·5 ± 0·5 μmol m?2 s?1 in the 36 Pa grown trees and 11·8 ± 0·4 μmol m?2 s?1 in the 65 Pa grown trees, and were not significantly different from each other. The photosynthetic capacity of 1‐year‐old needles was reduced by 25% from 23·0 ± 1·8 μmol m?2 s?1 in the 36 Pa CO2 grown trees to 17·3 ± 0·7 μmol m?2 s?1 in the 65 Pa grown trees. Growth in elevated CO2 also resulted in a 25% reduction in Vcmax (maximum carboxylation rate), a 23% reduction in Jmax (RuBP regeneration capacity mediated by maximum electron transport rate) and a 30% reduction in Rubisco activity and content. Total non‐structural carbohydrates (TNC) as a fraction of total dry mass increased from 12·8 ± 0·4% in 1‐year‐old needles from the 36 Pa grown trees to 14·2 ± 0·7% in 1‐year‐old needles from the 65 Pa grown trees and leaf nitrogen content decreased from 1·30 ± 0·02 to 1·09 ± 0·10 g m?2. The current‐year needles were not of sufficient size for gas exchange measurements, but none of the biochemical parameters measured (Rubisco, leaf chlorophyll, TNC and N), were effected by growth in elevated CO2. These results demonstrate that photosynthetic acclimation, which was not found in the first 2 years of this experiment, can develop over time in field‐grown trees and may be regulated by source‐sink balance, sugar feedback mechanisms and nitrogen allocation.  相似文献

A radioimmunoassay for thromboxane B2 (TxB2), a stable metabolite of thromboxane A2, is described. The method consists of extraction of TxB2 into ethyl acetate from acidified plasma or serum samples and saturation analysis using specific antibodies produced in rabbits against TxB2-BSA conjugate. The 50 % displacement level of the standard curve was 19.1 ± 2.9 pg/tube (mean ± S.D., n = 19). The method blank was 3.4 ± 3.1 pg/ml (n = 15) and the assay sensitivity thus 9.6 pg/ml (mean blank + 2 S.D.). When 100 to 200 pg of TxB2 were added to plasma, 96.2–103.6 % were recovered. The intra-assay coefficient of variation varied from 6.7 to 9.7 %, and the inter-assay coefficient of variation was 18.6 % (n = 10). The TxB2 concentration in the plasma of 14 healthy subjects varied from 29.3 to 120.8 pg/ml with a mean ± S.D. of 70.1 ± 26.1 pg/ml, when the blood was collected into tubes containing acetylsalicylic acid (ASA), whereas significantly higher (p < 0.001) TxB2 concentrations of 68.3 – 285.3 pg/ml with a mean ± S.D. of 151.8 ± 66.6 pg/ml were obtained from the same subjects in the plasma of blood which was collected into tubes containing no ASA. When blood samples from 10 subjects were allowed to clot at 0, +24 or +37°C for 60 min., the TxB2 concentrations in the sera were 2053 ± 870 pg/ml, 4001 ± 1370 pg/ml and 178557 ± 54000 pg/ml, respectively. The TxB2 levels in sera which were separated from blood samples incubated at +37°C, correlated significantly (p < 0.001) with the TxB2 productions in platelet-rich-plasma (PRP) after an induced aggregation. Our results indicate 1) when TxB2 is measured in plasma, the use of prostaglandin synthesis inhibitor in the collection tubes is necessary and 2) the measurement of TxB2 in serum of blood which has been kept at +37°C for a strictly standardized period of time could replace the use of PRP in TxB2 studies.  相似文献

A radioimmunoassay was developed for a mono-O-methyl derivative of thromboxane B2. The antibodies showed high specificity for this compound and cross reacted only 1.2% with thromboxane B2 and less than 0.1% with prostaglandins and prostaglandin metabolites. The method had a sensitivity of 7 picog. The radioimmunoassay was employed in studies where thromboxane A2 was generated in human platelets and immediately converted into mono-O-methyl thromboxane B2 by treatment of the sample with a large volume of methanol. In some of the experiments, thromboxane B2 was simultaneously measured by a separate radioimmunoassay. Using these two assays it was demonstrated that thromboxane A2 could be detected only during the earlier stages of the platelet aggregation, whereas thromboxane B2 rapidly reached a constant level. In a separate experiment, the half-life of thromboxane A2 in buffer was found to be 32.5±2.5 (S.D.) sec at 37°C; the compound was more stable at lower temperatures. The for thromboxane A2 was also considerably longer in plasma.  相似文献

依据涡度相关系统连续观测的2005年CO2通量数据,对青藏高原东北隅的高寒湿地生态系统源/汇功能及其部分环境影响因素进行了分析.结果表明,高寒湿地生态系统为明显的碳源,在植物生长季(5~9月份)吸收230.16 gCO2·m-2,非生长季(1~4月份及10~12月份)释放546.18 gCO2·m-2,其中净排放最高在5月份,为181.49 gCO2·m-2,净吸收最高在8月份,为189.69 gCO2·m-2,年释放量为316.02 gCO2·m-2.在平均日变化中,最大吸收值出现在7月份12:00,为(0.45±0.0012) mgCO2·m-2·s-1,最大排放速率出现在8月份0:00,为(0.22±0.0090) mgCO2·m-2·s-1.生长季中6~9月份表现为明显的单峰型日变化,非生长季的变化幅度较小.净生态系统交换量(NEE)和生态系统总初级生产力(GPP)与气温、空气水气饱和亏和地表反射率等环境因素呈现相似的相关性,与地上生物量和群落叶面积指数则为线性负相关,生态系统呼吸(Res)则与上述因子的相关性呈现相反的趋势.  相似文献

The prostaglandin endoperoxide PGH2, HHT, HETE, thromboxane A2, and thromboxane B2, which are all products of arachidonic acid metabolites of human platelets, were tested for their ability to modulate platelet cyclic nucleotide levels. None of the compounds tested altered the basal level of cAMP or cGMP, and only PGH2 and thromboxane A2 inhibited PGE1-stimulated cAMP accumulation. Thromboxane A2 was found to be a more potent inhibitor of PGE1-stimulated cAMP accumulation and inducer of platelet aggregation thatn PHG2.  相似文献

Dog platelets challenged with arachidonic acid fail to aggregate but synthesize a substance which aggregates rabbit and human platelets, this aggregation being suppressed by dibutyryl cyclic AMP. The aggregating substance contracts strips of rabbit aorta and of coeliac and mesenteric arteries, is soluble in diethyl ether, has a half-life of about 40 seconds at 37°C and of 100 seconds at 22°C. Its generation is blocked by various inhibitors of prostaglandin biosynthesis. The thromboxane A2 synthetase inhibitor imidazole and its analogue benzimidazolamine also suppress generation of vessel contracting activity in incubates of dog platelets and prostaglandin H2. Since dog platelets also transform prostaglandin H2 into thromboxane A2 their failure to aggregate, when stimulated by arachidonic acid or by prostaglandin H2, is not due to lack of thromboxane synthesizing ability.  相似文献

Homogenates of phagocytosing polymorphonuclear leukocytes obtained from rabbit peritoneum were incubated with the prostaglandin endoperoxides PGG2 or PGH2. After 2 min at 0°C, incubation mixtures contained an increased rabbit aorta contracting activity. Ether extracts of incubation mixtures contained a substance which contracted the superfused strips of rabbit aorta and coeliac artery and had a half life which was similar to thromboxane A2. The generation of thromboxane A2-like activity from PG endoperoxides was prevented by boiling the homogenate prior to incubation, or by pretreatment with benzydamine, a drug which blocks thromboxane formation in platelets. Production of thromboxane A2-like material by leukocyte homogenates was compared with platelet microsomal thromboxane synthetase.  相似文献

The aim of the study was to determine the prostacyclin (PGI2) and thromboxane A2 (TXA2) synthetase activities of myocardial tissue and their variation during ischemia and reperfusion. Regional ischemia was induced by 10 min occlusion of the left anterior descending coronary artery in isolated Langendorff rabbit hearts. Biosynthesis of PGI2 and TXA2 were carried out by using arachidonic acid as substrate and left ventricle microsomes (LVM) from ischemic and non-ischemic areas as sources of PGI2 and TXA2 synthetase. 6-keto-PGF and TXB2, stable metabolites of PGI2 and TXA2 respectively, were determined by radioimmunoassay. Experiments carried out under the adopted conditions showed that LVM were able to synthetise PGI2 as well as TXA2 from arachidonic acid. On the other hand, ischemia depressed both PGI2 and TXA2 synthetase activities of cardiac tissue: the depression was more pronounced on TXA2 synthetase than on PGI2 synthetase with no significant difference between ischemic and non-ischemic regions. Moreover, ischemia increased the ratio indicating therefore that it can facilitate the formation of PGI2. The post ischemic reperfusion of the heart counteracted the decrease in PGI2 synthetase induced by ischemia which returned to the normal level: reperfusion also slightly reversed the decrease in TXA2 synthetase. However, the diminution in TXA2 synthetase of non-ischemic myocardium was attenuated but it remained lower than the normal level. These results suggested that the whole left ventricle is affected by regional ischemia. Furthermore it appears that myocardial TXA2 synthetase is more vulnerable than PGI2 synthetase to a lack of oxygen and nutrients.  相似文献

The influence of hyperventilation on the spontaneous generation of prostacyclin and thromboxane A2 by isolated rat lungs was studied. Both prostacyclin and thromboxane A2, as measured by RIA of their stable end-products, 6-oxo-PGF and TXB2 respectively, were continuously released into the perfusate. However, the concentration of prostacyclin in the perfusate was higher than thromboxane A2. Under normal ventilation at a rate 40–50 breaths/min, the ratio between these two compounds was 5:1. Increasing the rate of respiration to 100 breaths/min preferentially stimulated the release of prostacyclin. During hyperventilation, the ratio between 6-oxo-PGF and TXB2 was 12:1. Aspirin and indomethacin suppressed both basal and hyperventilation-stimulated release of prostacyclin and thromboxane A2. Hydroperoxy-fatty acids and tranylcypromine inhibited only the release of prostacyclin but did not affect the generation of thromboxane A2. Our findings confirm that the lung generates prostacyclin predominantly, and provide direct evidence that respiratory movements are involved in generation of pulmonary prostacyclin and thromboxane A2.  相似文献

In this study, the response of N2 fixation to elevated CO2 was measured in Scirpus olneyi, a C3 sedge, and Spartina patens, a C4 grass, using acetylene reduction assay and 15N2 gas feeding. Field plants grown in PVC tubes (25 cm long, 10 cm internal diameter) were used. Exposure to elevated CO2 significantly (P < 0·05) caused a 35% increase in nitrogenase activity and 73% increase in 15N incorporated by Scirpus olneyi. In Spartina patens, elevated CO2 (660 ± 1 μ mol mol 1) increased nitrogenase activity and 15N incorporation by 13 and 23%, respectively. Estimates showed that the rate of N2 fixation in Scirpus olneyi under elevated CO2 was 611 ± 75 ng 15N fixed plant 1 h 1 compared with 367 ± 46 ng 15N fixed plant 1 h 1 in ambient CO2 plants. In Spartina patens, however, the rate of N2 fixation was 12·5 ± 1·1 versus 9·8 ± 1·3 ng 15N fixed plant 1 h 1 for elevated and ambient CO2, respectively. Heterotrophic non-symbiotic N2 fixation in plant-free marsh sediment also increased significantly (P < 0·05) with elevated CO2. The proportional increase in 15N2 fixation correlated with the relative stimulation of photosynthesis, in that N2 fixation was high in the C3 plant in which photosynthesis was also high, and lower in the C4 plant in which photosynthesis was relatively less stimulated by growth in elevated CO2. These results are consistent with the hypothesis that carbon fixation in C3 species, stimulated by rising CO2, is likely to provide additional carbon to endophytic and below-ground microbial processes.  相似文献

The copper complex of indomethacin (1-(p-chlorobenzoyl)-5-methoxy-2-methyl-indole acetate), a common anti-inflammatory drug, was prepared and characterized. Crystal structure determination revealed the dimeric form of the 1:2 complex, namely Cu2(indomethacin)4 · L2, in the unit cell. Suprisingly, the copper-copper distance (263 pm) was very close to metallic copper (256 pm). The two coordination sites in the copper-copper axis can be readily replaced by superoxide. An intriguing similarity to Cu2(acetate)4 was seen.Due to the lipophilic nature of the indomethacin ligand, this copper complex reacted with superoxide in aprotic solvents. The superoxide dismutating activity was successfully demonstrated in Me2SO/water and acetonitrile/water mixtures using the nitro-blue tetrazolium assay and pulse radiolysis. The second-order rate constant of 6 · 109 M?1 · s?1 in strictly aqueous systems dropped only slightly to 1.1 · 109 M?1 · s?1 when aprotic solvents were used. This is the fastest rate constant ever observed for a copper-dependent dismutation of superoxide. The KO2-induced lipid peroxidation in both erythrocytes and liver microsomes was suppressed by 70% in the presence of 1 · 10?10 mol · ml?1 of Cu2(indomethacin)4. The inhibitory action dropped to 25% when Cu2Zn2superoxide dismutase was employed. The formation of copper · indomethacin in rat serum after administration of indomethacin was shown in vitro and in vivo.  相似文献

The biliary excretion of {3H}-25-hydroxyvitamin D3 was studied in rats fed a diet containing 36% of total calories as ethanol and in pair-fed controls. {3H}-25-hydroxyvitamin D3 was injected i.v. and the biliary appearance of {3H}-compounds studied over a period of 80 minutes. Choleresis was not affected by chronic ethanol feeding averaging 98.7±10.3 μl min?1 kg?1 in Controls and 101.8±8.7 μl min?1 kg?1 in ethanol-fed animals. The total biliary excretion of {3H} was found to be 29% higher in ethanol-treated than control rats and represented 6.3±0.4% and 4.9±0.4% (p<0.05) of the injected dose at the end of the experiment. The biliary excretion rate as well as the biliary concentration of {3H}-25-hydroxyvitamin D3 congeners were significantly increased by the ethanol treatment from the 25th to the 80th minute after the i.v. injection of {3H}-25-hydroxyvitamin D3. The plasma and liver {3H} concentrations were not significantly affected by the ethanol treatment. These results suggest that, in the rat, chronic ethanol ingestion promotes the biliary loss of {3H}-25-hydroxyvitamin D3. The higher concentration of {3H}-compounds into the bile of ethanol treated rats also suggests the presence of excretory pathway(s) which could be, at least in part, independent of bile flow. This increased loss of {3H}-25-hydroxyvitamin D3 into the bile may be a contributing factor in the impaired vitamin D status in alcoholics.  相似文献

Ventilation frequency (FV) in motionless common sole Solea solea was measured before and after a startling stimulus in normoxia and in hypoxia (15% air saturation). Startling reduced FV in normoxia (from mean ±s.e. 41 ± 3·3 beats min?1 to near zero, i.e. 2·0 ± 1·8 beats min?1) and in hypoxia (from mean ±s.e. 80 ± 4·4 to 58·8 ± 12·9 beats min?1). It is suggested that the maintenance of high FV in hypoxia may increase the probability of detection by predators compared to normoxia.  相似文献

The half-life of thromboxane A2 is increased in plasma as well as in a buffer containing serumalbumin. Moreover, the half-life of thromboxane A2 depends on the amount of unesterified fatty acids bound to albumin. When more fatty acids are bound, the half-life is shortened. The role of cAMP dependent regulation in thromboxane A2, prostacyclin and PGF synthesis (measured by radioimmunoassay) was investigated in isolated mouse hepatocytes and in microsomal membranes prepared from these cells. In isolated hepatocytes N6,O2-dibutyryl cAMP inhibited the formation of all the three derivatives, while calcium ionophore A 23187 stimulated their synthesis. Addition of the dissociated catalytic subunit of cAMP dependent protein kinase and ATP to microsomal membranes inhibited the production of TXA2, PGI2 and PGF by about 50% and this inhibition was counteracted by the combined addition of heat stable inhibitor protein of cAMP dependent protein kinase. It is concluded that in parenchylmal liver cells cAMP dependent phosphorylation is directly involved in the inhibition of prostanoid synthesis.  相似文献
