Cellulolytic bacteria were enumerated and cellulase activity was determined over a 98-day period from fecal samples of gestating swine fed various levels and sources of fiber. The diets, each fed to five pigs, were a corn-soybean control, 20% corn cobs, and 40 and 96% alfalfa meal. Fecal samples were collected from all pigs on days 0, 5, 14, 21, 35, 49, 70, and 98. Overall, the most probable number of cellulolytic bacteria from pigs fed the control, 20% corn cobs, and 40 and 96% alfalfa meal was 23.3 X 10(8), 15.2 X 10(8), 45.1 X 10(8), and 52.5 X 10(8) per g (dry weight) of fecal sample, respectively. Enumeration of cellulolytic bacteria by counting zones of clearing in roll tubes, as compared with the most probable number procedure, accounted for only 1.1 and 17.0% of the cellulolytic bacteria, respectively, from pigs fed the control or 96% alfalfa meal diet. Cellulolytic bacteria (most probable number) on days 70 and 98 accounted for 4.1 and 10.0% of the viable count for the pigs fed the control and 96% alfalfa meal diets, respectively. The viable count was not different between pigs fed the control and 96% alfalfa meal diets. The overall mean cellulolytic activity (milligrams of glucose released from carboxymethyl cellulose per gram [dry weight] fecal sample per 30 min was 17.0, 19.9, 23.8, and 20.6, respectively, for the control, 20% corn cobs, and 40 and 96% alfalfa meal diets. The results indicate that the cellulolytic flora can be increased by prolonged feeding of high-fiber diets and may represent 10% of the culturable flora.  相似文献

Xylanolytic and cellulolytic bacteria were enumerated over an 86-day period from fecal samples of 10 8-month-old gilts that were fed either a control or a 40% alfalfa meal (high-fiber) diet. Fecal samples were collected from all pigs on days 0, 3, 5, 12, 25, 37, 58, and 86. Overall, the numbers of xylanolytic bacteria producing greater than 5-mm-diameter zones of clearing on 0.24% xylan roll tube medium after 24 to 36 h of incubation were 1.6 X 10(8) and 4.2 X 10(8)/g (dry weight) of feces for the control pigs and those fed the high-fiber diet, respectively. After 1 week of incubation, a large number of smaller zones of clearing (1 to 2 mm) appeared. Besides Bacteroides succinogenes and Ruminococcus flavefaciens, which produced faint zones of clearing in xylan roll tubes, three strains which closely resembled B. ruminicola hydrolyzed and used xylan for growth. The overall numbers of cellulolytic bacteria producing zones of clearing in 0.5% agar roll tube medium were 0.36 X 10(8) and 4.1 X 10(8)/g for the control pigs and those fed the high-fiber diet, respectively. B. succinogenes was the predominant cellulolytic isolate from both groups of pigs, and R. flavefaciens was found in a ratio of approximately 1 to 15 with B. succinogenes. Degradation of xylan and cellulose, measured by in vitro dry matter disappearance after inoculation with fecal samples, was significantly greater for pigs fed the high-fiber diet than that for the controls. These data suggest that the number of fibrolytic microorganisms and their activity in the large intestine of the adult pig can be increased by feeding pigs high-alfalfa-fiber diets and that these organisms are similar to those found in the rumen.  相似文献

Bacterial populations from gastrointestinal tracts of genetically lean and obese pigs fed a low- or high-fiber diet (0 or 50% alfalfa meal, respectively) were enumerated with rumen fluid media and specific energy sources. Total culture counts in rectal samples declined 56 (P greater than 0.05) and 63% (P less than 0.05) in lean and obese animals, respectively, 3 weeks after feeding the high-fiber diet. After 8 weeks, culture counts had risen and were similar to those obtained before alfalfa was fed (0 week). At slaughter, 12 to 17 weeks after feeding the high-fiber diet, total counts from rectal samples of lean pigs continued to rise and were 13% greater than the 0-week counts, whereas counts from obese animals declined 37% (P greater than 0.05). The number of cellulolytic bacteria in rectal samples of lean-genotype pigs fed the high-fiber diet increased 80 and 71% from 0 to 3 weeks and 3 to 8 weeks, respectively. This overall increases from 0 to 8 weeks in lean pigs was significant (P less than 0.05); however, these increases were not seen in obese pigs. These data suggest that the microflora is initially suppressed when exposed to a high-fiber diet and that later some adaptation takes place, apparently more so in lean than in obese pigs. When specific energy sources were used to delineate the distribution of different bacterial populations in the cecum, colon, and rectum, trends could be detected between high- and low-fiber diets. These data also support the concept that bacteria populations from different sites in the large bowel differ.  相似文献

With no acceptable method for collecting fresh rumen fluid from zoo ruminants, it was proposed that fecal bacterial concentrations may be correlated with rumen bacteria. If so, fecal bacterial concentrations could be used to study both the effects of diet on rumen bacteria as well as rumen abnormalities. Total and cellulolytic bacterial concentrations were determined in whole rumen contents and feces of sheep using a most‐probable‐number (MPN) assay. In a Latin square design, four crossbred ewes were fed diets of 100% long or chopped orchardgrass hay (OH) and 60% ground or whole shelled corn plus 40% chopped OH. In a second trial, the sheep were fed a pelleted complete feed at varying levels of intake i.e., control at 2.0% of body weight and at 1.8, 1.6, and 1.2% of body weight. Higher total rumen bacterial concentrations (P<0.01) were found on the high concentrate diets as compared with the high forage diets. Grinding the corn also increased total bacterial concentrations (P<0.05). Fecal concentrations of total bacteria were higher (P<0.01) with the high concentrate diets. Chopping the forage decreased the concentration of fecal cellulolytic bacteria (P<0.05) but had no effect on their concentration in the rumen. An inverse linear relationship (P<0.01) was observed between total bacterial concentrations in the feces and diet intake. Although relationships were observed between the rumen and feces for total and cellulolytic bacterial concentrations, they were dependent on diet, particle size, and level of intake. An experimental, microbially enhanced soybean product (MEPRO) was evaluated as a replacement for fishmeal (FM). Assessment of feedstuffs should include estimation of digestibility as well as pig performance and in combination with dietary additives. Digestibility values determined in growing pigs may not apply to nursery pigs; thus, standardized ileal digestibility (SID) of amino acids (AA) in MEPRO and FM were determined using 30±1.6 kg BW ileal-cannulated barrows (n=6) and 9.8±1.2 kg BW barrows (n=37; serial slaughter). Experimental diets included MEPRO, FM and nitrogen free where FM and MEPRO were included as the sole protein source. The SID of AAs was 3% to 5% lower in MEPRO than FM when fed to 30 kg pigs. The SID of arginine and methionine was greater (P<0.05) in MEPRO than FM when fed to 10 kg pigs. The SID of AAs was 12% to 20% lower in FM when fed to 10 v. 30 kg pigs but only 3% to 9% lower in MEPRO. A total of 336 barrows and gilts were weaned at 21 days of age (initial BW=6.1±0.8 kg) and used in a performance trial. Pens of pigs were assigned to one of the six experimental diets (8 pens/diet in two blocks). Treatment diets were fed in Phase I (7 days) and Phase II (14 days) with all pigs fed a common Phase III diet (14 days). Experimental diets included (1) negative control (NEG) containing corn, soybean meal and whey, (2) NEG+acidifier, (3) NEG+FM (POS), (4) POS+acidifier (POS A+), (5) NEG+MEPRO (MEPRO) and (6) MEPRO+acidifier. The FM and MEPRO were included at 7.5% and 5.0% in Phase I and II diets, respectively. Diets were formulated to meet the standard nutrient requirements for weaned pigs. Pig BW and feed disappearance was measured weekly and fecal scores were measured daily for the first 14 days post-weaning as an indicator of post-weaning diarrhea syndrome (PWDS). Performance (BW, daily gain, feed intake and gain : feed) was not significantly different among treatments. Treatment for PWDS occurred on different days in each block. Analysis of fecal score was completed separately by block. Pigs fed the NEG diets had higher (P=0.02) fecal scores than pigs fed the POS diets on days 2 and 3 (block 1) and higher (P<0.05) than pigs fed MEPRO or POS diets and diets with dietary acidifier on days 6 and 3 (block 2). The MEPRO holds promise as an alternative to FM in nursery pig diets.  相似文献

The total number of viable bacteria (2.16 x 10(9) to 5.44 x 10(9)/ml of ruminal fluid) and the number of cellulolytic bacteria (3.74 x 10(7) to 10.9 x 10(7)/ml) were not different among groups of animals fed each diet. The genera of protozoa in all of the animal groups were similar; however, when either the 100:0 or 50:50 diet was used the percentage of Entodinium sp. was lower and the percentage of Diplodiniinae was higher (P less than 0.05) in bison than in bison hybrids or cattle. Bacteroides succinogenes made up the largest number of cellulolytic isolates from bison (58 and 36%, respectively, on the 100:0 and 75:25 diets), which were more numerous (P less than 0.05) than those from bison hybrids (36 and 12%) and cattle (33 and 18%). This was offset by a lower number of cellulolytic Butyrivibrio isolates. The numbers of Ruminococcus albus and R. flavefaciens isolates, in general, were similar among the bovid species, although R. flavefaciens generally made up less than 10% of the cellulolytic isolates. In vitro digestibility coefficients were greater (P less than 0.05) for the bison when the 75:25 diet was used and similar for the other two diets. Cellulolytic bacteria became established 12 days after birth in the caecum and colon of conventionally-reared mice fed a diet containing 5 p. 100 crude cellulose (Weende). Their population reached a level between 10(6) and 10(7) bacteria per gram of digestive contents in 25-day-old animals. However, variations between animals were very large; 20 to 50% of the individuals were free of cellulolytic bacteria. A low cellulolytic population was observed in adult mice fed a cellulose-free diet. The amount of cellulose in the diet and its nature (crude or pure cellulose) affected the number of cellulolytic bacteria: the higher the percentage of cellulose in the diet, the higher the number of cellulolytic bacteria, in particular with crude cellulose-containing diet.  相似文献

Total and cellulolytic bacterial and fungal numbers were determined in ruminal and cecal contents of 20 blue duikers (Cephalophus monticola). The animals were equally divided by sex and fed two diets, either high roughage or high concentrate. The mean concentration for total bacterial numbers in the rumen was 26.0 × 108/g of contents, with values ranging from 2 × 108/g to 93 × 108/g. Cellulolytic numbers averaged 6.0 × 108/g with a range of 1.5 × 108/g to 24.0 × 108/g. No differences related to sex or diet were found. In contrast, total bacterial numbers in the cecum differed between diets (P < 0.02), i.e., 1,046 × 106 bacteria per g for animals fed the high-forage diet compared with 166 × 106/g for those fed the high-concentrate diet. Cellulolytic bacterial counts in the cecal contents averaged 3.1 and 7.0% of the total counts for the high-forage and high-concentrate diets, respectively. Low concentrations of fungi were found in both ruminal and cecal contents of some, but not all, animals. Unexpectedly, concentrations of bacteria and fungi in the rumen and cecum were highly correlated with their total numbers (concentration multiplied by total weight of contents).  相似文献

Three growth trials and a metabolism study were conducted to evaluate corn gluten meal—urea (CGM—urea) combinations compared to a urea control as sources of supplementary nitrogen for growing calves and lambs fed on diets based on corn cobs.In the first growth study cattle were fed individually twice daily while in the sheep growth study and the second cattle growth study, animals were fed in groups. The average daily gain and feed conversion were similar for animals given the soya bean meal (SBM) and CGM—urea supplements. Feed intakes were similar for all treatments except for growth study 1. Nitrogen retention and dry matter and protein digestibility did not differ significantly between the SBM and CGM—urea supplemented groups. Urea consistently gave inferior results in all the experiments. These results suggest that CGM—urea combination is essentially equal to SBM in supporting growth of calves and lambs.  相似文献

Four rumen-fistulated steers, randomly assigned to two groups (control and salinomycin fed) were used to monitor the changes in rumen microbial populations and volatile fatty acids (VFA) concentrations associated with feeding salinomycin (0.22 mg X kg-1 X day-1). Steers were adapted to an alfalfa hay and grain (80:20) diet before supplementing the diet with salinomycin, and then the diet was changed to 50:50 and 20:80 ratios of alfalfa hay to grain at 2-week intervals. Rumen samples for total and selective enumeration of anaerobic bacteria. VFA analysis, and enumeration of protozoa were collected during the 80:20 alfalfa hay-to-grain diet before salinomycin feeding, and during the 80:20, 50:50, and 20:80 hay-to-grain diets with salinomycin. At each sampling period, rumen samples were collected at 3 h after feeding on three consecutive days. Salinomycin feeding had no effect on rumen pH and total VFA concentration. The acetate-to-propionate ratio was significantly lower in salinomycin-fed steers than in the control. The molar proportion of butyrate increased in both control and salinomycin-fed steers. Total anaerobic bacterial counts were lower in salinomycin-fed steers than in the control steers after 8 weeks of salinomycin feeding. Salinomycin-resistant bacteria increased from 7.6 to 15.6% in salinomycin-fed steers but remained unchanged in control steers. Salinomycin had no effect on cellulolytic and lactate-utilizing bacteria, but the proportion of amylolytic bacteria was higher in salinomycin-fed steers than in control steers. The total number of protozoa decreased initially in salinomycin-fed steers. The initial reduction was due to reduced numbers of Entodinium species. Wet corn gluten feed (WCGF) is a high moisture feed containing rapidly digestible, non-forage fiber and protein. The objective of this study was to investigate the effect of substituting WCGF and corn stover for alfalfa hay in total mixed ration (TMR) silage on lactation performance and nitrogen balance in dairy cows. Nine multiparous Holstein dairy cows (BW = 532 ± 28.9 kg and day in milk = 136 ± 5.6 d; mean ± SD) were used in a replicated 3 × 3 Latin square design with 21-d periods (14 d of diet adaption and 7 d of sample collection). Groups were balanced for parity, day in milk, and milk production and consumed one of three treatment diets during each period. The treatment diets were fed as TMR and contained similar concentrate mixtures and corn silage but different proportions of roughage and WCGF. The three treatments were: (1) 0% WCGF, 0% corn stover, and 22.1% alfalfa hay (0% WCGF); (2) 6.9% WCGF, 3.4% corn stover, and 11.8% alfalfa hay (7% WCGF); and (3) 13.3% WCGF, 4.9% corn stover, and 3.9% alfalfa hay (13.3% WCGF). Compared to the 0% WCGF diet, the cows fed the 7% and 13.3% WCGF diets had a higher milk yield and concentration of milk fat, protein, lactose, and total solids. Effective degradability of DM was higher in the cows fed the 7% and 13.3% WCGF diets than it was with the 0% WCGF diet. Cows fed the 13.3% WCGF had a higher CP effective degradability and a lower rumen undegraded protein than cows fed the 0% WCGF diet. The concentration of ruminal volatile fatty acids and ammonia-N was higher in cows fed the 7% and 13.3% WCGF diets than cows fed the 0% WCGF diet. The fecal N was lower in cows fed the 7% and 13.3% WCGF diets than it was in cows fed the 0% WCGF diet. Milk N secretion and milk N as a percent of N intake were higher in cows fed the 13.3% WCGF diet than cows fed the 0% and 7% WCGF diets. In conclusion, it appears that feeding a TMR silage containing WCGF and corn stover in combination, replacing a portion of alfalfa hay, may improve lactation performance and nitrogen utilization for lactating dairy cows.  相似文献

Fifty-six weaner and growing-finishing Large White × Landrace pigs were used in two growth studies and a digestibility study to determine the value of undecorticated cottonseed meal (UCSM) as a protein supplement replacing soya bean meal (SBM). In a 35-day feeding experiment with pigs averaging 18.8 kg, four lots of six pigs were individually fed on isonitrogenous diets containing 0, 10, 20 and 30% UCSM. Daily feed intake (FI) did not differ significantly between treatments. Daily gain and feed/gain did not differ significantly between the control and the 10% UCSM diets but the control group gained significantly more than the 20 or 30% UCSM diets (P < 0.05).

In a second growth experiment lasting 35 days, 24 cross-bred pigs averaging 53.1 kg were fed on diets containing 0, 20, 30 and 41.1% UCSM (without SBM) in a randomized design involving six individually fed pigs per treatment. There were no differences in FI across the treatments. Weight gain was significantly depressed with only 20% UCSM and both gain and feed/gain declined linearly with increasing proportion of UCSM. The growth studies showed that 10% was the optimum level of inclusion of UCSM in the diets. These were supported by the results of the metabolic studies.  相似文献

为研究苜蓿(Medicago sativa L.)草粉对草鱼(Ctenopharyngodon idella)生长性能、肌肉品质和血清抗氧化指标的影响, 在基础饲料(粗蛋白: 32.0%, 粗脂肪: 4.12%)中分别添加0(对照组)、5%、10%、15%和20%的苜蓿草粉, 饲喂初始体重为(50.04±0.04) g的草鱼, 每个处理3个重复, 进行为期61d的养殖实验。结果表明: (1)与对照组相比, 20%苜蓿组的特定生长率显著降低(P<0.05), 15%苜蓿组的饲料系数和摄食率显著升高(P<0.05)。除5%苜蓿组外, 其余苜蓿添加组草鱼的肥满度显著降低(P<0.05); (2)饲料中添加苜蓿草粉显著降低草鱼肌肉中硫代巴比妥酸值和乳酸含量(P<0.05)。10%和15%苜蓿组草鱼肌肉羟脯氨酸含量显著高于对照组(P<0.05)。此外, 在添加苜蓿草粉后, 草鱼肌肉失水率显著升高(P<0.05)。除20%苜蓿组外, 其余苜蓿添加组草鱼肌肉黏附性显著升高(P<0.05), 草鱼肌肉硬度在10%苜蓿组显著高于对照组(P<0.05); (3)20%苜蓿组草鱼血清过氧化氢酶(CAT)活力显著高于对照组(P<0.05)。与对照组相比, 15%、20%苜蓿组草鱼血清谷胱甘肽过氧化物酶(GSH-PX)活力显著升高, 丙二醛(MDA)含量显著降低(P<0.05)。综上所述, 当苜蓿草粉的添加量不超过15%时, 对草鱼生长性能没有显著性影响, 且能改善草鱼肌肉品质, 提高血清抗氧化能力。因此, 草鱼饲料中苜蓿草粉的添加量不应超过15%。  相似文献

The addition of 10%, 20%, 30%, and 40% sun-cured alfalfa meal to a high-energy, low-fiber, basal diet for weanling rabbits gave increased average daily gains as compared to gain on the basal diet. Deaths from enteritis complex occurred only at the 0% and 10% alfalfa levels. When 2.5%, 5%, 7.5%, and 10% alfalfa meal were used, the average daily gains were numerically higher for all alfalfa-fed groups than for the basal diet. Maximum gains were achieved at the 20% alfalfa level. Alfalfa extracted with 95% ethanol retained its growth-stimulating effect when added to the basal diet. Feed preference trials gave conflicting results. In one trial, the basal diet was preferred over all alfalfa diets in a two-choice feed preference test, while in a second trial, all alfalfa diets were preferred over the basal diet. It was concluded that the growth of weaning rabbits fed a high energy diet was improved by the addition of alfalfa meal.  相似文献

Eight isonitrogenous (35% crude protein approximately) and isocaloric (4.2 kcal g(-1) approximately) diets were formulated including raw and fermented duckweed (Lemna polyrhiza) leaf meal at 10%, 20%, 30% and 40% levels. A particular bacterial strain (Bacillus sp.) isolated from carp (Cyprinus carpio) intestine and having extracellular amylolytic, cellulolytic, proteolytic and lipolytic activities was used for leaf meal fermentation for 15 days at 37 degrees C. The fibre content of leaf meal reduced from 11.0% to 7.5% and the antinutritional factors, tannin and phytic acid, were reduced from 1.0% to 0.02% and 1.23% to 0.09%, respectively after fermentation. However, the available reducing sugars, free amino acids and fatty acids increased in the fermented leaf meal. The response of rohu, Labeo rohita, fingerlings fed the experimental diets for 80 days was compared with fish fed a fish meal based reference diet. On the basis of growth response, food conversion ratio and protein efficiency ratio, 30% fermented Lemna leaf meal incorporated in the diet resulted in the best performance of rohu fingerlings. In general, growth and feed utilization efficiencies of fish fed fermented leaf meal containing diets were superior to those fed diets containing raw leaf meal. The apparent protein digestibility (APD) decreased with increasing levels of leaf meal irrespective of treatment. The APD for raw leaf meal was lower at all levels of inclusion in comparison to those for the fermented meals. The highest carcass protein and lipid deposition was recorded in fish fed the diet containing 30% fermented leaf meal. The results showed that fermented Lemna leaf meal can be incorporated into carp diets up to 30% level compared to 10% level of raw meal.  相似文献

The fibrolytic microbiota of the human large intestine was examined to determine the numbers and types of cellulolytic and hemicellulolytic bacteria present. Fecal samples from each of five individuals contained bacteria capable of degrading the hydrated cellulose in spinach and in wheat straw pretreated with alkaline hydrogen peroxide (AHP-WS), whereas degradation of the relatively crystalline cellulose in Whatman no. 1 filter paper (PMC) was detected for only one of the five samples. The mean concentration of cellulolytic bacteria, estimated with AHP-WS as a substrate, was 1.2 X 10(8)/ml of feces. Pure cultures of bacteria isolated on AHP-WS were able to degrade PMC, indicating that interactions with other microbes were primarily responsible for previous low success rates in detecting fecal cellulolytic bacteria with PMC as a substrate. The cellulolytic bacteria included Ruminococcus spp., Clostridium sp., and two unidentified strains. The mean concentration of hemicellulolytic bacteria, estimated with larchwood xylan as a substrate, was 1.8 X 10(10)/ml of feces. The hemicellulose-degrading bacteria included Butyrivibrio sp., Clostridium sp., Bacteroides sp., and two unidentified strains, as well as four of the five cellulolytic strains. This work demonstrates that many humans harbor intestinal cellulolytic bacteria and that a hydrated cellulose source such as AHP-WS is necessary for their consistent detection and isolation.  相似文献
