Scanning electron microscopy of sage (Salvia officinalis L.) leaves confirmed the presence of two basic types of glandular trichomes consisting of a capitate stalked form containing a multicellular stalk and surmounted by a unicellular secretory head, and a capitate sessile form containing a unicellular stalk and unicellular, or multicellular, secretory head. In the latter type, secretory activity and filling of the subcuticular cavity may begin at virtually any stage of the division cycle affording fully developed glands containing from one to twelve cells in the secretory head. Gas liquid chromatographic analysis of the oil content of the most numerous gland species (capitate stalked, capitate sessile with one and with eight secretory cells) indicated only minor quantitative differences in essential oil composition. Thus, each gland type is capable of producing the four major monoterpene families (p-menthanes, pinanes, bornanes and thujanes) characteristic of sage.  相似文献

Despite a number of recent molecular phylogenetic studies on Phlomoides, in terms of trichome morphology the genus is still among the most poorly studied taxa in the family Lamiaceae. In order to test the utility of trichome characters for delimitation of sections, subsection and species of Phlomoides, we examined trichomes of 64 species representing all recognized sections and subsections using stereomicroscopy and scaning electron microscopy. Two basic types of trichomes could be identified: non‐glandular and glandular. Both trichome types can be simple or branched. The glandular trichomes were sessile, short stalked or long stalked. Different kinds of branched trichomes were observed in most species of P. sect. Phlomoides, i.e. symmetrically stellate, stellate with a central long branch, bi‐ or trifurcate. The species of P. sect. Filipendula were mostly covered by simple trichomes. Moreover, variation in trichome characters appears to have particular value, not only for the classification at sectional or subsectional rank, but also for delimitation of species within each section. For example, all studied species of P. subsect. Fulgentes are characterized by various kinds of stellate trichomes, while the trichome variability in P. subsect. Tetragonae was sufficiently high for species discrimination. An ancestral character state reconstruction was performed in order to investigate the evolution of trichome types and it revealed the following evolutionary trends in trichome characters of Phlomoides: 1) branched trichomes are primitive in Phlomoides as compared to simple ones, 2) long simple non‐glandular trichomes are derived as compared to short simple ones and 3) the presence of stalked glandular trichomes is advanced as compared to subsessile or sessile ones.  相似文献

Development of peltate glandular trichomes of peppermint   总被引:1,自引:0,他引:1       下载免费PDF全文
Cryofixation and conventional chemical fixation methods were employed to examine the ultrastructure of developing peltate glandular trichomes of peppermint (Mentha x piperita). Our results are discussed in relation to monoterpene production and the mechanism of essential oil secretion. Peltate glands arise as epidermal protuberances (initials) that divide asymmetrically to produce a vacuolate basal cell, a stalk cell, and a cytoplasmically dense apical cell. Further divisions of the apical cell produce a peltate trichome with one basal cell, one stalk cell, and eight glandular (secretory) disc cells. Presecretory gland cells resemble meristematic cells because they contain proplastids, small vacuoles, and large nuclei. The secretory phase coincides with the separation and filling of the sub-cuticular oil storage space, the maturation of glandular disc cell leucoplasts in which monoterpene biosynthesis is known to be initiated, and the formation of extensive smooth endoplasmic reticulum at which hydroxylation steps of the monoterpene biosynthetic pathway occur. The smooth endoplasmic reticulum of the secretory cells appears to form associations with both the leucoplasts and the plasma membrane bordering the sub-cuticular oil storage cavity, often contains densely staining material, and may be involved with the transport of the monoterpene-rich secretion product. Associated changes in the ultrastructure of the secretory stage stalk cell are also described, as is the ultrastructure of the fragile post-secretory gland for which cryofixation methods are particularly well suited for the preservation of organizational integrity.  相似文献

This paper reports the results of a study of the morphology and development of glandular trichomes in leaves ofCalceolaria adscendensLidl. using light and electron microscopy. Secretory trichomes started as outgrowths of epidermal cells; subsequent divisions gave rise to trichomes made up of a basal epidermal cell, a stalk cell and a two-celled secretory head. Ultrastructural characteristics of trichome cells were typical of terpene-producing structures. Previous phytochemical studies had revealed thatC. adscendensproduces diterpenes. Comparison withC. volckmanni, which produces triterpenes, and has trichomes with eight-celled secretory heads, suggests that there could be a relationship between the type of glandular trichome and the class of terpene produced. Further work is needed to test the hypothesis and to develop trichome characters as taxonomic tools.  相似文献

The natural products that accumulate in or exude from plant glandular trichomes are biosynthesized by secretory cells located at the apex of the trichome. To investigate the formation of glandular trichome constituents in several species of mints (Lamiaceae), a new procedure was developed for isolating large numbers of highly purified secretory cells. In this method, the leaf surface is gently abraded with glass beads in a way that fragments the glandular trichomes and yields clusters of intact secretory cells. The isolated, intact secretory cells and cell-free preparations derived from them are very active in monoterpene biosynthesis and provide useful starting materials for the purification of several key enzymes of monoterpene metabolism. The procedure described is adaptable to a broad range of plant species and should find wide application in the preparation of whole cell and cell-free systems for biosynthetic studies of plant natural products found in glandular trichomes.  相似文献

BACKGROUND AND AIMS: In the present study, the differences between glandular and non-glandular trichomes, the secretory process and the method of secretion were studied. Previous studies on leaves of Styrian oil pumpkin (Cucurbia pepo var. styriaca) plants have shown that four morphologically and ontogenetically independent glandular and non-glandular trichome types and one bristle hair type can be distinguished. The four types of trichomes can be categorized into three glandular trichome types: type I, a short-stalked trichome with four head cells including a 'middle-cell', two stalk cells and one basal cell; type II, a long-stalked trichome with two head cells, a 'neck-cell' region and a long stalk area; type IV, a 'stipitate-capitate' trichome with a mesophyll cell basement, a short stalk and a multicellular head; type III, a non-glandular 'columnar-digit' trichome, which consists of two head cells continuous with three-celled stalk, and the basal cell. METHODS: The histochemical studies (the main classes of metabolite in secreted material of glandular trichomes) were conducted in fresh and fixed hand sections, using the following tests: Sudan black B, Nile blue A, osmium tetroxide, neutral red, Naturstoffreagent A, FSA (fuchsin-safranin-astra blue), NADI (naphthol + dimethylparaphenylenediamine) and ruthenium red. Each suggested differences in the intercalations during the ontogenetical development of each trichome during the development stage. KEY RESULTS: The histochemical reactions revealed the main components of the materials secreted by all types of trichomes, which include lipids, flavones and terpenes and the different cell wall compositions. Glandular secretions were observed during environmental scanning electron microscopy (ESEM) and the trichomes compared with those seen by conventional scanning electron microscopy (CSEM). CONCLUSIONS: Scanning electron microscopy and histochemical analysis demonstrated that each of the trichomes studied produced and released secretory products in a characteristic way.  相似文献

We have investigated the trichome characteristics in representative species of Salvia and Pleudia in order to evaluate this source of morphological evidence for addressing problems regarding generic delimitation and subgeneric classification. Trichomes of 46 Salvia spp., representing three subgenera in Iran, were investigated using light microscopy and scanning electron microscopy. General trichome characteristics were constant among different populations of a certain species, but showed a degree of variability useful in the delimitation of taxa, specifically at lower taxonomic levels. Trichome characters of taxonomic interest are as follows: types of glandular hair; number of composing cells (uni‐, bi‐ or multicellular); size and thickness; branching pattern; and presence of papillae on the surface. Non‐glandular trichomes can be simple and branched. Glandular trichomes can be stalked, subsessile or sessile. Our investigation reveals the usefulness of such characters in providing fundamental taxonomic criteria for taxon delimitation in these genera at various levels, especially at the specific rank. Furthermore, the data presented here indicate the potential applicability of such characters in the determination of evolutionary trends in Salvia and allies. © 2015 The Linnean Society of London, Botanical Journal of the Linnean Society, 2016, 180 , 241–257.

The development and morphology of secretory trichomes of Calceolaria volckmanni was examined with light and electron microscopy. The formation and development of the glandular trichomes began with the outgrowth of a single epidermal cell which progressively increased in height and evolved into a pear-shaped cell. Subsequent divisions generated a mature trichome formed by a basal cell, a stalk "endodermal" cell and an 8-celled glandular head. Histochemical tests revealed the lipophilic nature of the secretion, the presence of terpenes and flavonoids, and displayed a particular cutinization of the walls of the stalk cell. The observed ultrastructural features of the lipophilic glandular hairs suggested the function of plastids in the secretory process.  相似文献

Trichomes of 37 taxa of the genus Stachys and one species of Sideritis (S. montana) were examined using light and scanning electron microscopy. The indumentum shows considerable variability among different species, but is constant among different populations of one species, and therefore, affords valuable characters in delimitation of sections and species. The characters of taxonomic interest were presence of glandular and non-glandular trichomes, thickness of the cell walls, number of cells (unicellular or multi-cellular), presence of branched (dendroid) trichomes, presence of vermiform trichomes, orientation of trichomes in relation to the epidermal surface, curviness of trichomes, and presence of papillae on trichome surface. Two basic types of trichomes can be distinguished: glandular and non-glandular trichomes. The glandular trichomes can in turn be subdivided into subtypes: stalked, subsessile, or sessile. The stalks of the glandular trichomes can be uni- or multi-cellular. Simple unbranched and branched trichomes constitute two subtypes of non-glandular trichomes. Our data do not provide any support for separation of Sideritis from Stachys. The following evolutionary trends are suggested here for Stachys: vermiform trichomes with stellate base are primitive against vermiform trichomes with tuberculate base, long vermiform trichomes are primitive against the short simple trichomes, appressed trichomes are advanced against spreading ones, and loss of glandular trichomes is advanced against their presence. Overall, trichome micromorphology is more useful in separation of species within sections rather than characterizing large natural groups known as sections, except for few cases.  相似文献

Electron microscopy confirms previous light microscope observations that tobacco leaf trichomes are glandular and that there are two different types. Both the tall trichome (multicellular stalk, unicellular or multicellular head) and the short trichome (unicellular stalk; multicellular head) exhibit characteristics common to gland cells—a dense cytoplasm, numerous mitochondria, and little vacuolation. The tall trichome contains structurally well developed chloroplasts and an elaborate network of endoplasmic reticulum. The short trichome contains undifferentiated plastids and endoplasmic reticulum which parallels the nucleus and plasmalemma. Few dictyosomes are seen either in the short trichome or in the tall trichome. The short trichome appears to undergo structural changes concurrently with the appearance of secretory product within the cells. The most noticeable change is the formation of the extraplasmic space between the cell wall and the plasmalemma. Electron dense secretory product is observed between the plasmalemma and the cell wall and within the intercellular spaces.  相似文献

The anatomy and ultrastructure of glandular trichomes at differentdevelopmental stages were investigated inPhillyrea latifoliaL.leaves by transmission electron microscopy and histochemicaltechniques. The trichome consisted of a multicellular secretoryhead, a unicellular stalk and a collecting cell surrounded byepidermal cells and spongy mesophyll cells. There were numerousplasmodesmata across the cell walls of trichome cells, and especiallybetween the stalk cell and the collecting cell. The collectingcell and stalk cell contained few chloroplasts. Mitochondria, elements of the endoplasmic reticulum and small vacuoles wereabundant in the secretory cells. Crystals were present in thesecretory cells and the collecting cell, especially at the matureand senescent stages of trichome development. As the cuticle, which covered the secretory cells, did not show pores or perforations,it is proposed that secretion occurred by accumulation of productsin subcuticular spaces followed by diffusion through the cuticle.Callose accumulation was observed between the stalk cell andthe collecting cell of senescent trichomes, especially in salt-treatedplants. Trichome ontogeny was accelerated in salt-treated plants.Copyright1998 Annals of Botany Company Cuticle;Phillyrea latifolia; secretion; transmission electron microscopy; trichome development.  相似文献

This study was initiated to characterize the distribution, morphology, secretion mode, histochemistry and ultrastructure of the glandular trichomes of Ceratotheca triloba using light and electron microscopy. Its leaves bear two morphologically distinct glandular trichomes. The first type has long trichome with 8-12 basal cells of pedestal, 3-14 stalk cells, a neck cell and a head of four cells in one layer. The second type has short trichome comprising one or two basal epidermal cells, a unicellular or bicellular stalk and a multicellular head of two to eight cells. There is a marked circular area in the upper part of each head cell of the long trichome. This area is provided with micropores to exudate directly the secretory product onto the leaf surface by an eccrine pathway. The secretory product has copious amount of dark microbodies arising from plastids which are positive to Sudan tests and osmium tetroxide for unsaturated lipids. The secretion mode of short trichomes is granulocrine and involves two morphologically and histochemically distinct vesicle types: small Golgi-derived vesicles which are positive to Ruthenium Red test for mucilaginous polysaccharides; the second type is dark large microbodies similar to that of long trichomes with low quantity. These two types are stored in numerous peripheral vacuoles and discharge their contents accompanied by the formation of irregular invaginations of the plasmalemma inside the vacuoles via reverse pinocytosis. These two secretion modes of long and short trichomes are reported for the first time in the family Pedaliaceae. The long trichomes have more unsaturated lipids, while the short trichomes contain more mucilaginous polysaccharides.  相似文献
