黑色素是黑木耳的主要活性成分之一,在黑木耳的药理活性上发挥着重要作用。为了提高黑木耳黑色素的提取得率,实验采用正交法和响应面法对纤维素酶-超声波协同提取黑木耳黑色素的提取工艺进行了优化,并对最优条件下提取的黑木耳黑色素体外抗氧化活性进行了分析。实验结果表明,纤维素酶-超声波协同提取黑木耳黑色素的最优条件为:酶添加量12mg,酶解温度40℃,酶解pH 5.0,酶解时间120min,NaOH浓度1.27mol/L,料液比1:40,超声功率300W,超声时间52min,超声温度60℃。在最优条件下,黑木耳黑色素提取得率可达到10.48%,相比于实验设置的未添加纤维素酶的超声波组黑色素提取得率提高了12.93%。抗氧化结果表明,采用纤维素酶-超声波协同提取的黑色素相比于未加纤维素酶提取的黑色素清除ABTS、DPPH和羟基自由基的能力更强。研究结果为黑木耳黑色素的高效提取及其产品的应用开发奠定了基础。  相似文献

The beneficial effects of oyster extract against various disorders and diseases induced by oxidative stress have aroused great interest. In this article, ultrasonic-assisted enzymolysis was employed to produce polysaccharides of Crassostrea hongkongensis (CHP) and their antioxidant activity was investigated. A single-factor experiment and then a four-factor, three-level Box–Behnken design were first used to optimize ultrasonic extraction for polysaccharides. On the basis of ridge analysis, the optimum conditions are obtained as ultrasonic treatment time of 24 min, power of 876 W, temperature of 49°C, and material–solvent ratio of 1:6 (w/v). It is found that ultrasound pretreatment before protease hydrolysis was a great help to improve CHP yield and purity, especially more favorable with flavorzyme, neutrase, alcalase, and pepsin. Furthermore, the polysaccharide fraction, which was obtained by ultrasonic pretreatment and then alcalase hydrolysis at the conditions of 3000 U/g, 55°C, pH 8.0, for 4 hr, exhibited an obvious scavenging effect on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical (98.48 ± 0.55% and 99.20 ± 0.12%, respectively) and a lenoleic acid peroxidation inhibition effect (85.48 ± 0.65%) at a concentration of 5.0 mg/mL. These results reveal the potential application of CHP in functional food and nutraceuticals.  相似文献

The study was designed to investigate the use of ultrasound-assisted extraction (UAE) of rapamycin (sirolimus) from bacterial strain of Streptomyces rapamycinicus NRRL 5491. To achieve the maximum extraction yield, various parameters were optimized which include S. rapamycinicus (10?g) of biomass in toluene (50?mL), temperature (20°C), acoustic intensity (35.67?W/cm2), and duty cycle (40%) for 4?min extraction time with probe tip length of 0.5?cm dipped into extraction solvent from the surface. The maximum extraction yield 60.15?±?0.01?mg/L was attained under the mentioned optimum parameters. The use of ultrasound for the extraction of rapamycin shows about twofold increase in the yield as compared to the conventional solid–liquid extraction (29.7?±?0.2?mg/L). The study provides the effective UAE technique to produce potential value-added products.  相似文献

Colchicum autumnale L. also known as the autumn crocus, contains colchicine alkaloid having antifungal properties. The tuber of this plant is rich in terms of colchicine. In this research an ultrasound-assisted extraction (UAE) method was optimized for the extraction of colchicine from Colchicum autumnale L. bulbs before high-performance liquid chromatography with UV detection (HPLC-UV). Optimization of various extraction parameters was performed using response surface methodology (RSM) to evaluate the maximum colchicine yield from Colchicum autumnale L. bulbs. The Box-Behnken design (BBD) and RSM were used to investigate the effect of three key parameters (extraction time (20–60?min), extraction temperature (40–80?°C) and ultrasound power (500–700?W) on extraction efficiency. The variance analysis suggested that the dependent response variable of yield of colchicine may be expressed by a quadratic polynomial model. The optimal theoretical extraction conditions were found to be an ultrasonication power of 602.4?W, an extraction time of 42?min and a temperature of 64?°C. Under these conditions, the optimum foreseen yield was 0.237%. The experimental colchicine yield obtained by following the optimized conditions was found to be 0.238%. Psidium guajava leaves are rich in health-promoting flavonoids compounds. For better utilization of the resource, the ultrasound-assisted aqueous extraction was investigated using Box-Behnken design under response surface methodology. A high coefficient of determination (R2?=?97.8%) indicated good agreement between the experimental and predicted values of flavonoids yield. The optimal extraction parameters to obtain the highest total flavonoids yield were ultrasonic power of 407.41?W, extraction time of 35.15?min, and extraction temperature of 72.69?°C. The average extraction rate of flavonoids could reach 5.12% under the optimum conditions. Besides, HPLC analysis and field emission scanning electron microscopy indicated that the ultrasonic treatment did not change the main component of flavonoids during extraction process and the higher flavonoids content was attributed by the disruption of the cell walls of guava particles. Thus, the extraction method could be applied successfully for large-scale extraction of total flavonoids from guava leaves.  相似文献

Fucose-containing sulfated polysaccharides can be extracted from the brown seaweed, Sargassum sp. It has been reported that fucose-rich sulfated polysaccharides from brown seaweeds exert different beneficial biological activities including anti-inflammatory, anticoagulant, and anti-viral effects. Classical extraction of fucose-containing sulfated polysaccharides from brown seaweed species typically involves extended, multiple-step, hot acid, or CaCl2 treatments, each step lasting several hours. In this work, we systematically examined the influence of acid concentration (HCl), time, and temperature on the yield of fucose-containing sulfated polysaccharides (FCSPs) in statistically designed two-step and single-step multifactorial extraction experiments. All extraction factors had significant effects on the fucose-containing sulfated polysaccharides yield, with the temperature and time exerting positive effects, and the acid concentration having a negative effect. The model defined an optimized single-step FCSPs extraction procedure for Sargassum sp. (a brown seaweed). A maximal fucose-containing sulfated polysaccharides yield of ~7% of the Sargassum sp. dry matter was achieved by the optimal extraction procedure of: 0.03?M HCl, 90°C, 4?h. HPAEC-PAD analysis confirmed that fucose, galactose, and glucuronic acid were the major constituents of the polysaccharides obtained by the optimized method. Lower polysaccharide yield, but relatively higher fucose content was obtained with shorter extraction time. The data also revealed that classical multi-step extraction with acid ≥0.2?M HCl at elevated temperature and extended time had a detrimental effect on the FCSPs yield as this treatment apparently disrupted the structural integrity of the polymer and evidently caused degradation of the carbohydrate chains built up of fucose residues.  相似文献

Polysaccharides production from Poria cocos was carried out using aqueous NaOH with the assistance of ultrasonic. Experimental design was used to investigate the effect of three parameters (extraction time, extraction concentration of NaOH, and ratio of aqueous NaOH to raw material) on polysaccharides yields. The ranges of the factors investigated were 1–3 min for extraction time (X1), 0.5–1.0 mol/L for extraction concentration of NaOH (X2), and 30–50 for ratio of aqueous NaOH to raw material (X3). The statistical analysis of the experiment indicated that extraction concentration of NaOH had significant effect on P. cocos polysaccharides yields. The central composite design showed that polynomial regression models were in good agreement with the experimental results with the coefficients of determination of 0.9935 for P. cocos polysaccharides yield. The optimal condition for P. cocos polysaccharides yield within the experimental range of the variables studied was at 2.44 min, 0.789 mol/L, and 53.0. At this condition, the predicted yield of polysaccharides extracted was 82.3%.  相似文献

The effect of polysaccharidases (κ-carrageenase, β-agarase, xylanase, cellulase) on the protein extraction from three rhodophytes has been studied. The kinetic parameters (apparent V m, apparent K m) and the optimum activity conditions (pH, temperature) of each enzyme were determined by using pure substrates. All the tested enzymes possess Michaelis Menten mechanism with estimated substrate saturating concentrations of 8 000 mg l−1(carrageenan) for κ-carrageenase, 8 000 mg l−1 (agar) for β-agarase, 5000 mg l−1 (xylane) for β-xylanase and 6 000 mg l−1 (carboxymethylcellulose) for cellulase. The optimum activity conditions are pH 6.5–6.8 at 45°C for carrageenase, pH 6–6.5 at 55°C for agarase, pH 5 at 55°C for xylanase and pH 3.8 at 50°C for cellulose. Different alga/enzymes couples (κ-carrageenase/Chondrus crispus, β-agarase/Gracilaria verrucosa, β-xylanase/Palmaria palmata) were tested under the optimum activity conditions. Alga/cellulase + specific enzyme (e.g. Chondrus crispus/carrageenase + cellulase) systems were also studied at the optimum activity conditions of a specific enzyme (e.g. carageenase). The use of the only cellulose was also tested on each alga. Except for Palmaria palmata, the highest protein yields were observed with the procedures using cellulase coupled with carrageenase or agarase for an incubation period limited to 2 h. The Chondrus crispus/carrageenase + cellulose and Gracilaria verrucosa/agarase + cellulase systems gave ten-fold and three-fold improvements, respectively, in protein extraction yield as compared to the enzyme-free blank procedure. The combined action of xylanase and cellulose on protein extraction from Palmaria palmata does not significantly improve protein yield. The best overall protein yield for P. palmata is for P. palmata/xylanase with a 14-h incubation time. This study shows the interest in the use of a polysaccharidase mixture for improving protein extractibility from certain rhodophytes. This biotechnology approach, adapted from procedures for protoplast production or enzymatic liquefaction of higher plants, could be tested as an alternative method to obtain proteins from seaweeds of nutritional interest.  相似文献

The effect of ultrasonic extraction on extraction yields, cytotoxicity, and anticancer activity of Spirulina maxima was investigated in this study. Optimal extraction conditions were determined as 60 kHz frequency at 60°C for 30 min with 120 W intensity, which resulted in 19.3% of extraction yields and 19.1% of cytotoxicity on normal human cells. Yields from conventional water and ethanol extraction were 15.8% at 100°C and 8.3% at 80°C, respectively. It was found that the extracts obtained by ultrasonic extraction process selectively inhibited the digestive-related cancer cell lines, such as human stomach cancer cells, having 89% of the highest inhibition ratio and 4.5 of the highest selectivity. In adding 0.5 mg/mL of the extract, human promyelocytic leukemia cells' cell differentiation was increased 1.72 times over that of the control. Expression level of B cell lymphoma-2 from Hep3B cell was also effectively suppressed by the extract obtained at 60 kHz and 60°C, leading to the inhibition of the early step of carcinogenesis. This work suggests that anticancer activity of the extracts is due to water-soluble polysaccharides rather than proteins and is further supported by the result that the ultrasonification extraction process can efficiently extract relatively intact polysaccharides rather than digesting the proteins in S. maxima by matrix assisted laser desorption ionization-time of flight and high performance size exclusion chromatography chromatogram analyses. Therefore, ultrasonic extraction increases both extraction yield and the biological activity of S. maxima extracts, which might be useful as an alternative natural anticancer agent in the medical and food industries.  相似文献

The aim of this research is to statistically optimize enzymatic hydrolysis parameters for the production of R-phycoerythrin (RPE) from red algae Gracilaria verrucosa. Six independent variables, incubation temperature, incubation time, ratio of buffer to raw material, cellulase loading, xylanase loading, and pH, were selected for response surface methodology studies. A central composite design was employed to maximize RPE production. A mathematical model with high determination coefficient (R 2?=?0.86) was developed and could be employed to optimize RPE extraction. The optimal extraction conditions of RPE were determined as follows: incubation temperature (48°C), incubation time (6?h), ratio of buffer to raw material (20 w/v), cellulase loading (15%), xylanase loading (5%), and pH (6.5). Under this optimal condition, the experimental yield of RPE was 6.25?mg?g?1. Based on the result of response surface methodology and desirability function approach study, total sugar, the main by-product in RPE extraction was considered as another response. A new optimal condition was predicted as follows: incubation temperature (30°C), incubation time (12?h), ratio of buffer to raw material (20, w/v), cellulase loading (15%), xylanase loading (5%), and pH (6). Under this condition, similar RPE levels were obtained while the concentration of total sugar decreased by 40%.  相似文献

研究适合于以牡丹种皮为原料提取总黄酮的方法,对其提取方法工艺条件进行优化。考察了酶种类、浓度、酶解孵育温度、孵育时间和pH值等因素对提取工艺的影响,并在单因素实验基础上,利用正交实验法进行工艺优化。考察结果表明在果胶酶0.05 mg·mL-1、酶解孵育温度45℃、pH4.5条件下进行酶解孵育180 min后,总黄酮得率最高可达74.839 mg·g-1,相对于乙醇提取法牡丹种皮总黄酮得率显著提高。  相似文献

The present study optimized the extraction characterization and antioxidant activities of water-soluble compound polysaccharides (CPs) from hawthorn, lotus leaf, Fagopyrum tataricum, semen cassiae, Lycium barbarum, and Poria cocos Chinese herbal medicines that have mass ratios of 4 : 2 : 2 : 1.5 : 1 : 1. The CPs yield equation was predicted using quantitative theory, to which a maximum CPs yield of 7.18±0.24 % under the following optimal extraction conditions: a water-to-raw material ratio of 30 mL/g, an extraction temperature of 65 °C, an extraction time of 45 min, and extraction mode ultrasonic-assistant extraction. CPs were consisted of Ara, Gal, Glc, Xyl, Man, GalA and GlcA in a molar ratio of 3.1 : 2.6 : 50.6 : 1.7 : 20.4 : 17.2 : 4.2. The HPGPC profiles and FT-IR spectra implied that CPs were heterogeneous acidic polysaccharides and possessed the β-d -pyranose configuration. Congo red test, CD spectrum and SEM revealed that CPs with three helix conformation showed a flocculent, granulous or sheet-like appearance. Furthermore, the relationships between antioxidant activity and concentration of CPs displayed significant positive correlation, and the scavenging abilities for DPPH, hydroxyl radical, ABTS, superoxide-anion radical and reducing power of CPs were 93.56±2.51 %, 84.03±1.69 %, 83.29±1.93 %, 37.49±1.93 % and 0.467±0.006 at a concentration of 4.0 mg/mL. Therefore, CPs could be applied as a potential natural antioxidant in pharmaceutical or functional food fields.  相似文献

The main carotenoid in Chlorella vulgaris is lutein. The ultrasound alone or together with enzymatic pretreatment for the extraction of lutein from C. vulgaris was optimized using response surface methodology (RSM) to improve the extraction process. The optimal ultrasound extraction condition was: ultrasound frequency, 35 kHz; ultrasound intensity, 56.58 W/cm2; extraction temperature, 37.7°C; extraction time, 5 h; and ratio of solvent to solid, 31 mL/g, where the lutein recovery was 3.16 ± 0.03 mg/g wet C. vulgaris. The optimal enzymatic pretreatment was: reaction time, 2 h; enzyme concentration, 1.23% (v/w); pH, 4.5, and temperature 50°C. The optimal ultrasound extraction with enzymatic pretreatment was: ultrasound frequency, 35 kHz; ultrasound intensity, 56.58 W/cm2; extraction temperature, 37.7°C; extraction time, 162 min; and ratio of solvent to solid, 35.6 mL/g wet C. vulgaris, where the extraction yield of lutein was 3.36 ± 0.10 mg/g wet C. vulgaris. This was much higher than for ultrasound treatment alone. The surface areas of microalga cells treated by ultrasound with/without enzymatic pretreatment increased significantly, which might contribute to the increase in lutein yield. There were no significant differences in structure, color, and antioxidant activity of lutein between the ultrasound and conventional methods. The highest cost of the crude and lutein was obtained by the ultrasound with enzymatic pretreatment due to the complex process and liquid waste in the enzymatic pretreatment process, but the ultrasound treatment alone was the lowest. Therefore, ultrasound extraction is the most economical method for the extraction of microalgal lutein.  相似文献

Medical fungi polysaccharides belong to a very important species of biological macromolecules, which are the basic substances that effectively maintain and ensure the normal operation of biological life activities. However, research on extraction and biological activity of Inonotus cuticularis polysaccharides has never been reported. In this study, the optimum yield of Inonotus cuticularis polysaccharides was determined by the orthogonal experimental design. The highest yield of 3.10±0.06 % was obtained with extraction temperature of 80 °C, extraction time of 150 min, and water to raw material ratio of 30 mL/g and repeated twice. After deproteinization for 5 times, the protein removal rate reached 70.10±1.75 %, and the content of polysaccharides and protein were 46.64 and 0.42 %. Infrared spectrometer indicated that Inonotus cuticularis polysaccharides are typical β‐pyranose with characteristic peaks of polysaccharides. Subsequently, the activities of scavenging free radicals for the deproteinated polysaccharides were studied. When the concentration of Inonotus cuticularis polysaccharides was 0.3 mg/mL, the scavenging activities of the sample on DPPH., .OH, ABTS.+ and O2.? reached 83.67±0.27, 65.21±4.82, 43.45±1.36 and 80.28±2.30 %, respectively, and the reducing power reached 0.46±0.01. The IC50 values scavenging DPPH., .OH, ABTS.+ and O2.? were 0.139±0.13, 0.162±0.14, 0.317±0.30 and 0.121±0.10 mg/mL, respectively. Results showed that Inonotus cuticularis polysaccharides present potential stronger antioxidant activities, especially .OH scavenging activity and reducing power. Experimental results could provide research basis of Inonotus cuticularis polysaccharides for further exploitation and utilization.  相似文献

In this work, the subcritical water extraction technology was used to extract alkaloids from Macleaya cordata, and the effects of extraction temperature and time on its yield were investigated to find the best extraction conditions. Moreover, the antioxidant capacity and antibacterial activity of Macleaya cordata extract were studied. Furthermore, through the single-factor method, it was found that properly increasing the extraction temperature and prolonging the extraction time was conducive to increasing alkaloid yield. Still, a considerable amount of alkaloids might be decomposed by heat, resulting in a decrease in their yield. The results showed that the optimal extraction temperature of alkaloids from Macleaya cordata with subcritical water is 190 °C, the time is 45 min, and the corresponding maximum yield is 35.19±0.12 mg/g (sanguinarine equivalent in raw materials). In addition, the antioxidation and bacteriostasis abilities of subcritical water extract are better than those of traditional hot water extract, indicating that it is a feasible method to extract alkaloids from Macleaya cordata with subcritical water.  相似文献

In the present study, two cellulose-degrading bacteria (CDB-5 and CDB-12) were isolated from mangrove soils of Mahanadi river delta, based on halo zone formation in Congo red agar medium and evaluation for cellulase production in CMC broth medium. Based on morphological, biochemical and 16S rRNA gene sequencing, the two strains, CDB-5 and CDB-12, were identified as Brucella sp. and Bacillus licheniformis, respectively. The gene bank accession number of the strains CDB-5 and CDB-12 are KR632646 and KR632645, respectively. The strain Brucella sp. and B. licheniformis showed an enzyme activity of 96.37?U/ml and 98.25?U/ml, respectively, after 72?h of incubation period. Enzyme production was optimized under different growth conditions such as pH, temperature, agitation rate, carbon source, sodium chloride (NaCl), and nitrogen sources. Maximum cellulase production by both the strains was obtained in the same parameter condition such as pH (7.0), rpm (150), and NaCl (2%, w/v) which varies for other parameters. The strain, CDB-5, produced maximum cellulase at 35?°C temperature, maltose as a carbon source, and yeast extract as a nitrogen source where as the strain CDB-12 produces maximum cellulase at 45?°C temperature, carboxyl methyl cellulose (CMC) as carbon source and trypton as a nitrogen source. The bacterial crude enzyme was purified by ammonium sulfate precipitation followed by overnight dialysis. SDS-PAGE analysis of the partially purified cellulase enzyme exhibited band sizes of approximately 55 and 72?kDa.  相似文献

研究了超声波协同复合酶法提取姬松茸多糖的最佳工艺条件。采用均匀设计法分别考察不同时间、pH值、温度、酶浓度、固液比对纤维素酶、果胶酶以及木瓜蛋白酶酶解反应的影响,并研究三种酶联合使用时的加酶方式以及超声波协同提取时的最佳条件。结果表明,超声波协同复合酶法可显著提高姬松茸多糖的提取率,其最佳提取条件为:超声波作用20min,分步加酶法(先加果胶酶:pH值3.8、温度50℃、时间90min、加酶量7000U/g、固液比1:45;然后加纤维素酶:pH值3.6、温度75℃、时间120min、加酶量150U/g、固液比1:45;最后加木瓜蛋白酶:pH值3.6、温度75℃、时间120min、加酶量20000U/g、固液比1:45),多糖提取率达到14.51%。  相似文献

Introduction – Silybin, a standardised extract of flavanolignans from the seeds of Silybum marianum, has been used for centuries as a natural remedy in the treatment of hepatitis and cirrhosis. The higher yield of silybin by using more efficient extraction technique is of particular interest in the herbal products manufacture. Objective – To systematically investigate the important factors of enzyme‐assisted extraction of flavanolignans from the seeds of Silybum marianum to enhance the extraction yield of silybin. Methodology – The important factors of enzyme‐assisted extraction were optimised by employing Box–Behnken design with the aid of the orthogonal array design (OAD) OA8 (27). The effects of enzyme incubation temperature (EIT), the pH of enzyme solution (PES) and the size of seeds (SS) on the yield of silybin were visualised as three‐dimensional response surface and contour plots. Results – The predictive yield was 24.6 mg/g defatted seeds under the optimum enzymolysis conditions (EIT 40°C, PES 4.5 and SS 7003 μm). The coefficient of the model was r2 > 0.97 (n = 15). The actual yield of silybin was 24.81 ± 1.93 mg/g defatted seeds, higher by 138 and 123.6% than that from ethanol extraction in this study and in the previous literature, respectively. IR spectra and HPLC of the extracts by EAE were in agreement with those from ethanol extraction. SEM and TEM pictures of defatted seeds by variant extractions demonstrate that the extraction of silybin depends on the destruction of cell walls. Conclusion – The results suggest that EAE is a promising alternative for the extraction of silybin by the use of traditional ethanol extraction. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

 The thermotolerant yeast strain, Kluyveromyces marxianus IMB3, was found to be capable of ethanol production during growth at 45°C on media containing milled paper and exogenously added commercial cellulase. At maximum achievable cellulose concentrations in shake-flask cultures, ethanol production increased to 6.6 g/l at 45°C, representing an overall level of conversion of 21% of the maximum theoretical yield. Subsequent studies involving variations in added cellulase concentrations to the batch systems demonstrated that ethanol yields could be increased to 10 g/l at 45°C, which represented 39% of the maximum theoretical yield. As a result of ethanol production at 45°C in the systems examined, we suggest that the thermotolerant ethanol-producing yeast strain K. marxianus represents a novel candidate for use in simultaneous saccharification and conversion of the resulting substrates to ethanol.
