Summary Fluorescence in situ hybridization revealed the presence of ribosomal RNA genes in paracentromeric regions of all A chromosomes and in the distal half of B chromosomes in embryonic cells from Moroccan specimens of the grasshopperEyprepocnemis plorans. The expression of these genes was monitored by the presence of nucleoli attached to each chromosome bivalent in diplotene cells from males collected from two different Moroccan populations and was compared to previous data of Spanish populations. Whereas only the nucleolus organizer regions (NORs) on S9–S11 and X chromosomes were active in the Spanish specimens. Moroccan individuals showed NOR activity in all chromosomes. The rRNA genes on the B chromosome were inactive in both populations. The S9 and S10 NORs were less active in Moroccan specimens than in Spanish specimen, which might be partly explained by the negative interdependence for expression of the S10 NOR with respect to those on L2 and X chromosomes. On the other hand, the X NOR was more active in Moroccan specimens than in Spanish specimens, and this might be partly due to the positive effect that the presence of B chromosomes has on the expression of this NOR. Sea lamprey Petromyzon marinus from the River Miño and Sella, Spain, showed an almost identical frequency for three observed haplotypes. None of these haplotypes were found among three North American populations. These results indicate an absence of exchange among sea lamprey populations spawning in the west and south‐east Atlantic coasts. The similarity between the collections from the two Spanish rivers suggests that homing does not occur in anadromous sea lamprey.  相似文献

Recent advances in molecular biology have made it possible to use the trace amounts of DNA in faeces to non-invasively sample endangered species for genetic studies. Here we use faeces as a source of DNA and mtDNA sequence data to elucidate the relationship among Spanish and Moroccan populations of great bustards. 834 bp of combined control region and cytochrome-b mtDNA fragments revealed four variable sites that defined seven closely related haplotypes in 54 individuals. Morocco was fixed for a single mtDNA haplotype that occurs at moderate frequency (28%) in Spain. We could not differentiate among the sampled Spanish populations of Cáceres and Andalucía but these combined populations were differentiated from the Moroccan population. Estimates of gene flow (Nm = 0.82)are consistent with extensive observations on the southern Iberian peninsular indicating that few individuals fly across the Strait of Gibraltar. We demonstrate that both this sea barrier and mountain barriers in Spain limit dispersal among adjacent great bustard populations to a similar extent. The Moroccan population is of high ornithological significance as it holds the only population of great bustards in Africa. This population is critically small and genetic and observational data indicate that it is unlikely to be recolonised via immigration from Spain should it be extirpated. To test phylogenetic relationships within the genus Testudo (Testudines: Testudinidae), we have sequenced a fragment of the mitochondrial (mt) 12S rRNA gene of 98 tortoise specimens belonging to the genera Testudo, Indotestudo, and Geochelone. Maximum likelihood and neighbor-joining methods identify two main clades of Mediterranean tortoises, one composed of the species Testudo graeca, Testudo marginata, and Testudo kleinmanni and a second of Testudo hermanni, Testudo horsfieldii, and Indotestudo elongata. The first clade, but not the second, was also supported by maximum parsimony analysis. Together with the genus Geochelone, a star-like radiation of these clades was suggested, as a sister-group relationship between the two Testudo clades could not be confirmed. The intraspecies genetic variation was examined by sequencing the mt 12S rRNA fragment from 28 specimens of T. graeca and 49 specimens of T. hermanni from various geographic locations. Haplotype diversity was found to be significantly larger in T. graeca compared with T. hermanni, suggestive of reduced genetic diversity in the latter species, perhaps due to Pleistocene glaciations affecting northern and middle Europe or other sources of lineage reduction. No ancient mt 12S rRNA gene haplotypes were identified in T. graeca and/or T. hermanni originating from islands in the Mediterranean Sea, suggesting that these islands harbor tortoise populations introduced from the European and African mainland.  相似文献

Mitochondrial DNA variation in Moroccan and Spanish honey bee populations   总被引:4,自引:0,他引:4  
The mitochondrial DNAs of 192 Moroccan and 173 Spanish honey bee colonies were characterized by a rapid test involving the restriction by DraI of a PCR-fragment of the COI-COII region. In Morocco, we found eight haplotypes, all characteristic of the African (A) lineage, suggesting that most if not all the maternal lineages of the colonies repeatedly imported from Europe over the last 150 years have not contributed mitochondrial genomes to the local population. Using two new genetic distances analogous to the shared allele distance defined for nuclear genes, we showed that Morocco was most probably colonized by two sublineages, one from the north-east and the other one from the south of the country and that the contact zone between them extends along both sides of the Atlas range. In Spain, we found eight haplotypes characteristic of lineage A (six in common with Morocco) and four of lineage M (the West European lineage). The distribution of haplotypes of both lineages forms a gradient with c. 10% of lineage M in the south of Spain (Seville) and up to 100% in the north (San Sebastian). Three hypotheses are presented to explain the large differences of haplotype frequencies between Moroccan and lineage A Spanish colonies: a non-Moroccan origin of lineage A in Spain, an ancient Moroccan origin or a recent Moroccan origin with a rapid shift of haplotype frequencies due to a founder effect.  相似文献

The brown frog Rana graeca was believed to be present in two areas, the Balkan Peninsula and the Italian Apennines. We have characterised the S1 satellite DNA family from Rana graeca graeca and compared it with that of Rana graeca italica. On Southern blots, the patterns of S1 satellite DNA bands are very different between Italian and Greek specimens, but homogeneous among various populations of the same taxon. The satellite DNA from the Greek taxon contains two repetitive units (S1a (494 bp) and S1b (363 bp)) that could be sequenced after amplification from genomic DNA to directly yield their consensus sequences in each genome. These consensus sequences were very similar among the Greek populations, but differed either in sequence (in S1a) or in both size and sequence (in S1b) from the corresponding repeats of the Italian taxon. A mechanism of concerted evolution is likely responsible for the high homogeneity of S1a and S1b repeat sequences within each genome and species. The genomic content of S1 satellite DNA was lower in the Greek than in the Italian populations (0.5 vs. 1.9%) and fluorescence in situ hybridization (FISH) analysis showed the S1 satellite on only 4 chromosome pairs in the Greek taxon and on all 13 chromosome pairs in the Italian taxon. The completely different structure and genomic organization of the S1 satellite DNA indicate that the Greek and Italian taxa are distinct species: R. graeca and R. italica.  相似文献

油松(Pinus tabulaeformis)是我国特有广布树种。秦岭是我国亚热带和暖温带气候的重要分界线,贺兰山是我国西北干旱区与半干旱区的分界线。旨在以分布于秦岭、贺兰山和晋陕两省的10个天然油松种群为研究对象,通过对其线粒体DNA(nad1和matR内含子)的序列,探讨地理阻隔对油松遗传结构的影响。结果表明,10个天然油松种群的100个个体中共检测到27个单倍型,多态位点172个,简约信息位点35个,单一多态性位点137个。其中1个单倍型为全部种群共有,4个单倍型为2-5个种群共有,其余22个单倍型为各个种群所独有。在这10个种群中,有8个种群分别具有1-4个独有单倍型。尽管秦岭南侧种群和北侧种群分别具有两个和一个独有单倍型; 贺兰山东侧种群和西侧种群均具有3个独有单倍型;但是与晋陕种群(即油松分布区中心种群)相比,其种群独有单倍型平均数目则明显较少。与此同时,不仅秦岭南北两侧或贺兰山东西两侧种群的独有单倍型的进化关系往往较近,而且秦岭北侧或贺兰山东侧种群的独有单倍型与油松分布区中心种群的某些独有单倍型的关系亦较近。从而导致秦岭南北两侧或贺兰山东西两侧种群与油松分布区中心种群之间表现出比较复杂的聚类关系。由此可见,油松的遗传结构与山脉屏障的存在没有明显关系。  相似文献

We assess the genetic history and population structure of Cicada barbara in Morocco and the Iberian Peninsula, based on analysis of the mitochondrial cytochrome b gene. The divergence between Morocco and the Iberian Peninsula populations was strongly corroborated by the molecular data, suggesting genetically isolated populations with a low level of gene flow. The Ceuta population from Spanish North Africa was more similar to the Iberian populations than the surrounding Moroccan populations, suggesting that the Strait of Gibraltar has not been acting as a strict barrier to dispersal while the Rif Mountains have. The Iberian Peninsula specimens showed a signature of demographic expansion before that which occurred in Morocco, but some of the assumptions related to the demographic parameters should be considered with caution due to the small genetic variation found. The high haplotype diversity found in Morocco implies higher demographic stability than in the Iberian Peninsula populations. These results do not, however, suggest a Moroccan origin for Iberian cicadas; but the most northwest region in Africa, such as Ceuta, might have acted as a southern refuge for Iberian cicadas during the most severe climatic conditions, from where they could expand north when climate improved. The separation of two subspecies within C. barbara (C. barbara lusitanica and C. barbara barbara) finds support with these results.  相似文献

Potamopyrgus antipodarum from 13 locations in central and western Japan were investigated for mitochondrial DNA haplotypes. Two haplotypes (JA and JB) were found based on an analysis of a 343-bp sequence of the cytochrome c oxidase subunit I gene. The JB type comprised two subhaplotypes (JB-I and JB-II), which were distinguishable by an additional sequence of 289 bp. Eleven populations were likely monomorphic in the mtDNA haplotype, while two populations, which were supposed to have been established recently, were polymorphic, encompassing JA and JB haplotypes. This result suggests that different haplotypes invaded Japan via different routes and that polymorphic populations were formed by colonization from more than one source population.  相似文献

The biogeographical distribution of brown trout mitochondrial DNA haplotypes throughout the Iberian Peninsula was established by polymerase chain reaction-restriction fragment polymorphism analysis. The study of 507 specimens from 58 localities representing eight widely separated Atlantic-slope (north and west Iberian coasts) and six Mediterranean drainage systems served to identify five main groups of mitochondrial haplotypes: (i) haplotypes corresponding to non-native, hatchery-reared brown trout that were widely distributed but also found in wild populations of northern Spain (Cantabrian slope); (ii) a widespread Atlantic haplotype group; (iii) a haplotype restricted to the Duero Basin; (iv) a haplotype shown by southern Iberian populations; and (v) a Mediterranean haplotype. The Iberian distribution of these haplotypes reflects both the current fishery management policy of introducing non-native brown trout, and Messinian palaeobiogeography. Our findings complement and extend previous allozyme studies on Iberian brown trout and improve present knowledge of glacial refugia and postglacial movement of brown trout lineages.  相似文献

In recent years there has been growing interest in analyzing the geographical variations between populations of different Phlebotomus spp. by comparing the sequences of various genes. However, little is known about the genetic structure of Phlebotomus ariasi. In this study, we were able to sequence a fragment of the mitochondrial Cyt b gene in 133 sandflies morphologically identified as P. ariasi and proceeding from a wide geographical range covering 35 locations in 11 different regions from five countries. The intra-specific diversity of P. ariasi is high, with 45 haplotypes differing from each other by one to 26 bases and they are distributed in two mitochondrial lineages, one limited geographically to Algeria and the other widely dispersed across Mediterranean countries. The Algerian lineage is characterized by having 13 fixed polymorphisms and is made up of one sole haplotype. The European/Moroccan P. ariasi lineage is characterized by being made up of a great diversity of haplotypes (44) which display some geographical structuring. This could be one of the multiple factors involved in the epidemiological heterogeneity of the foci of leishmaniasis. Phlebotomus chadlii is the sister group of European/Moroccan P. ariasi. The separation of the Algerian haplotype, H45, from the rest of the specimens, European/Moroccan P. ariasi and P. chadlii, is well supported by the bootstrap analysis.  相似文献

Sixty‐four samples from 46 salmon populations totalling 2369 specimens were used for polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP) analysis of the mitochondrial ND1 region. The final analyses included 3095 specimens from 60 populations in Northern Europe. A subsample was analysed by RFLP of ND3/4/5/6. Representative RFLP haplotypes from different parts of the distribution area were sequenced and the phylogeny of European haplotypes and their relations to the North American lineage was described. The four common European haplotypes derive from the ancestral ND1‐BBBA (rooting the European clade to the North American) by one‐step substitutions: AAAA < AABA < BBBA > BBBB. The Swedish west‐coast populations differ from the geographically close southern Baltic, indicating absence of inward and limited outward gene flow through the Danish straits during the last 8000 years. Within the Baltic Sea, only three ND1 haplotypes were detected and there was no variation for ND3/4/5/6. In the whole southern Baltic and in lakes Vänern, Ladoga and Onega the haplotype AABA dominated. Proposed postglacial colonization routes to the Baltic Sea are discussed in relation to the haplotype distribution pattern.  相似文献

Ips typographus populations were analysed by enzyme electrophoresis and by sequence analysis in order to quantify the degree of population differentiation. Enzyme electrophoresis showed a high gene flow among all European populations. Analysis of single loci showed that aspartate aminotransferase-2 ( Aat-2 ) clustered the Scandinavian populations apart from the other populations whereas other enzyme loci showed no significant pattern. Analysis of mitochondrial DNA revealed eight haplotypes. The populations from Italy, Croatia and Belgium were the most polymorphic. Two haplotypes (I and II) were found in the majority of Central European populations. In the northern populations only one haplotype (I) was found. The Russian and Lithuanian populations had a particular haplotype (IV) that was not found in any of the other populations. In view of these data it is suggested that the population structure of I. typographus has been influenced by events which took place during and after the last ice age. Populations were forced into refugial areas in the south and in the area north of Moscow along with the host tree, Picea abies . P. abies had four refugial areas: the Apennine Alps, the Dinaric Alps, the Carpathian Alps and Kostroma, an area north of Moscow. With amelioration of the temperature, beetles spread after P. abies . According to the data presented here, I. typographus migrated from the Apennine and/or Dinaric Alps to the north. Whilst there is evidence for high gene flow among populations, bottleneck effects can still be detected in the north and also in the east. Beetles from the refugial area north of Moscow did not spread to Scandinavia or to the west like the host tree.  相似文献

A south central Moroccan sample was analyzed for 20 nuclear DNA polymorphisms (restriction fragment length polymorphisms). The population was chosen on the basis of available information on its history, making it suitable for comparisons with data from other European populations. The markers analyzed have been studied previously in several human groups from different continents, and data on African and European samples have been compared to evaluate the genetic affinity of the studied sample with other populations, especially with two Spanish groups: Basques and Andalusians. Heterozygosity levels showed intermediate values between the African and European groups and higher than those found so far in an African group for the studied markers. Genetic distances closely matched geographical relationships through neighbor-joining tree and correspondence analysis, the Moroccans being closer to the European groups than the sub-Saharan Africans included in the analysis. Allele distributions revealed specific population associations with large weight of several alleles in the differentiation of some groups. Gene flow from sub-Saharan Africa appears to be relevant in understanding the differentiation of present Moroccan populations.  相似文献

Susceptibility/resistance to scrapie in sheep and goats is influenced by host prion protein gene ( PRNP ) genotype. In this study, we report the analysis of prion protein gene polymorphisms in 137 goats of two Moroccan populations: D'man and Chaouni. We found seven previously described amino acid polymorphisms at codons 37, 127, 137, 142, 154, 222 and 240, as well as three known silent mutations. In addition, we identified three new allelic variants: 101R and 139S in D'man goats and 145D in D'man and Chaouni individuals. The high frequency of the resistant allele 154H could offer genetic protection against the disease to the analysed animals. A total of 12 haplotypes and 28 genotypes were found, the distribution of which shows significant differences between both groups. Moreover, haplotype frequencies were compared with bibliographic data showing that the haplotype distribution of PRNP in Moroccan populations is genetically similar to Southern Italian and Greek goats.  相似文献

Here, we examine the genetic diversity in the agricultural pest Nezara viridula (Linnaeus, 1758 ) from populations of Argentina using mitochondrial cytochrome c oxidase subunit I (COI) gene sequences. The DNA sequence comparisons of 718 base pairs of the COI gene revealed seven haplotypes. The observed total haplotype diversity (Hd) value was of 0.138, and the nucleotide diversity was of 0.00039 and 0.00135 according to π and θW, respectively. Eight out of the 10 populations analysed, mostly from soya bean crops, only presented the more frequent haplotype, while 2 haplotypes were found in a mixed culture and 6 haplotypes in a peanut culture. Factors such as differential insecticide applications, as well as the surrounding habitat, and the host plant preference could be related to the genetic diversity differences observed among samples of N. viridula. The analysis of genetic diversity in samples collected in crops treated and non‐treated with insecticides, as well as in samples collected from different seasons, could help to clarify the role of the factors that led to the pattern of genetic diversity detected in this study. The result of a comparative analysis of COI gene sequences among populations from South America, Africa, Asia and Europe was consistent with the hypothesis of an African origin of N. viridula. On the other hand, the haplotypes of Europe were clustered with haplotypes from South America. In addition, specimens from Madeira (west of Europe) shared ancestry with South America and Europe. It has been suggested that a probable route of colonization of America could have been from Western Europe towards the eastern coasts of South America.  相似文献

To help conservation programs of the endangered spur-thighed tortoise and to gain better insight into its systematics, genetic variation and evolution in the tortoise species Testudo graeca (Testudines: Testudinidae) was investigated by sequence analysis of a 394-nucleotide fragment of the mitochondrial 12S rRNA gene for 158 tortoise specimens belonging to the subspecies Testudo graeca graeca, Testudo graeca ibera, Testudo graeca terrestris, and a newly recognized subspecies Testudo graeca whitei. A 411-nucleotide fragment of the mitochondrial D-loop was additionally sequenced for a subset of 22 T. graeca, chosen because of their 12S gene haplotype and/or geographical origin.  相似文献

Phlebotomus papatasi (Scopoli) (Diptera: Psychodidae) is the main vector of Leishmania major Yakimoff & Schokhor; which is the cause of self-limiting cutaneous leishmaniasis in the Old World. This sandfly is found in houses, animal shelters, caves and rodent burrows. It has a large geographical range, which includes the Middle East and the Mediterranean regions. A population analysis of colony and field specimens of P. papatasi was conducted on 25 populations originating from 10 countries. The distribution of haplotypes of the maternally inherited mitochondrial cytochrome b gene were analysed to assess the population differentiation of P. papatasi. Alignment of a 442-basepair region at the 3' end of the gene identified 21 haplotypes and 33 segregating sites from 131 sandflies. The pattern of sequence variations did not support the existence of a species complex. The median-joining network method was used to describe both the origin of the haplotypes and the population structure; haplotypes tended to cluster by geographical location, suggesting some level of genetic differentiation between populations. Our findings indicate the presence of significant population differentiation for populations derived from Syria, Turkey, Palestine, Israel, Jordan and Egypt. Knowledge of population differentiation among P. papatasi populations is important for understanding patterns of dispersal in this species and for planning appropriate control measures.  相似文献

The Japanese regional population of the Oriental white stork (Ciconia boyciana) became extinct in 1986. Mitochondrial DNA (mtDNA) D-loop region from 20 mounted specimens preserved at public facilities in Toyooka City, Hyogo Prefecture, Japan and its vicinity (n = 17), the area inhabited by the last of the Japanese population, and samples originating from China (n = 3) which were kept at a zoo was analyzed. After extracting DNA from small pieces of skin from mounted specimens, a 1210-bp region of the mtDNA D-loop region was analyzed. The haplotypes among 11 specimens of storks captured or found dead at Toyooka City just before the population became extinct were completely identical. Four haplotypes observed among the mounted specimens preserved in the vicinity of Toyooka City were differentiated from those of captive storks originating from China and Russia in a previous study. Therefore, the last Japanese population might have been a genetically unique group. However, phylogenetic analysis using the maximum likelihood method showed that haplotypes found in the Japanese regional population were closely related to the Chinese and Russian lineages (sequence difference = 2.1%). One mounted specimen collected in 1935 at Izushi village, in the vicinity of Toyooka City, showed the same haplotype as the captive storks from China, suggesting that genetic flow may have historically occurred between the populations of Japan and the continent. Recently, reintroduction for the Oriental white stork has been planned in Toyooka City. The planning for the recovery of extinct populations should not only involve translocation of species to the range from which it disappeared, but also reconstruction of regional populations while considering the genetic lineage between the extinct and introduced populations.  相似文献

The Moroccan distribution of Testudo graeca graeca L. is defined by locality records (109) and related to data from nearby or bioclimatically equivalent meteorological stations (86) to produce a rain-temperature climagram. Tortoises attain 1900 m in the High Atlas range. Tortoises occur where mean annual rainfall (P) is up to 1062 mm, and even 1112 mm where mean minimum monthly temperature for the coolest month (m) is below 0oC and they are usually absent unless associated (90.0%) with Quercus ilex L. and Quercus suber L. woodland. Tortoises are absent from the Arctic-Alpine and Arid or Saharan bioclimates and where, without riverine habitat, Emberger's Quotient (Q) is below 19.5 (P = 139 mm) in the Steppe bioclimate. Below Q_= 39.1, they occur (88.9%) where there is Argania spinosa (L.) Maire woodland in the south-west and other broad-leaved and mixed wood- and scrubland species elsewhere. The Moroccan climagram includes and is extended by further stations (36) in the North African and southern European ranges generally ( T. g. graeca is reported in NW Egypt). The Moroccan range potentially occupies 167 000 m2. Of a total of 45 stations below 550 m in NW Europe, the Western Mediterranean climagram only overlaps 12 in more oceanic Britain and Ireland; 11 are coastal or insular and 11 where Quercus woodland extends the natural range with m below 0oC in montane areas. Tortoises do not occur where mean annual sunshine much exceeds 3250 h in northern Africa or below c. 2400 h in NW Europe. Tortoises introduced from the Mediterranean region by the pet trade are unable to reproduce if they survive in NW Europe for insolation is insufficient for egg incubation and hatchling survival.  相似文献
