An electrochemiluminescence (ECL) approach for methamphetamine determination was developed based on a glassy carbon electrode modified with a Ru(bpy)32+‐doped silica nanoparticles/Nafion composite film. The monodispersed nanoparticles, which were about 50 nm in size, were synthesized using the water‐in‐oil microemulsion method. The ECL results revealed that Ru(bpy)32+ doped in silica nanoparticles retained its original photo‐ and electrochemical properties. The ECL intensity was found to be proportional to methamphetamine concentration over the range from 1.0 × 10?7 to 1.0 × 10?5 mol L?1, and the detection limit was found to be 2.6 × 10?8 mol L?1.  Glucose oxidase-catalyzed reduction of cis[MIII (LL)2Cl2]+ (M=Os and Ru) complexes to cis[MII (LL)2Cl2] (LL=2,2′-bipyridine and 1,10-phenanthroline type ligands) by d-glucose is a first-order process in the complex and the enzyme in aqueous buffered solution. The reaction follows MichaelisMenten kinetics in d-glucose and the rate is independent of d-glucose concentration above 0.03 M. The reactivity decreases in the series [Ru(bpy)2Cl2]+ > [Os(phen)2Cl2]+ > [Os(4,4′-Me2bpy)2Cl2]+ > [Os(4,7Me2phen)2Cl2]+. The measured second-order rate constant for the oxidation of reduced glucose oxidase by [Os(phen)2Cl2]+ in air equals 1.2×105 M–1 s–1 at pH 6.7, [d-glucose] 0.05 M, and 25  °C, which is ca. 20% less than that when the reaction solutions are purged with argon. In the case of [Ru(bpy)2Cl2]+ the rate constant equals 1.8×105 M–1 s–1 under similar conditions in air, showing higher reactivity of Ru complexes compared with Os ones. The reduction is pH-dependent with a maximum around 7. Added for solubilization of poorly soluble metal complexes, surfactants decrease the rates of the enzymatic reaction. The retardation effect increases in the series: cetyltrimethylammonium bromide < Triton X-100 < sodium dodecyl sulfate, i.e. on going from positively charged to neutral and then to negatively charged surfactants. The behavior of the OsIII and RuIII complexes toward reduced glucose oxidase contrasts to that of recently studied ferricenium cations. As opposed to the latter, the former do not show kinetically meaningful binding with the enzyme, and the Michaelis kinetics typical of the ferricenium case is not realized for the OsIII, and RuIII species. The systems OsIII- or RuIII-glucose oxidase are convenient for routine "one pot" spectrophotometric monitoring of the d-glucose content in samples, since the metal reduction to MII is accompanied by a strong increase in absorbance in the visible spectral region. Quenching effects of bergenin, based on the electrochemiluminescence (ECL) of the tris(2,2′‐bipyridyl)‐ruthenium(II) (Ru(bpy)32+)/tri‐n‐propylamine (TPrA) system in aqueous solution, is been described. The quenching behavior can be observed with a 100‐fold excess of bergenin over Ru(bpy)32+. In the presence of 0.1 m TPrA, the Stern–Volmer constant (KSV) of the ECL quenching is as high as 1.16 × 104 M?1 for bergenin. The logarithmic plot of the inhibited ECL versus logarithmic plot of the concentration of bergenin was linear over the range 3.0 × 10?6–1.0 × 10?4 mol/L. The corresponding limit of detection was 6.0 × 10?7 mol/L for bergenin (S/N = 3). In the mechanism of quenching it is believed that the competition of the active free radicals between Ru(bpy)32+/TPrA and bergenin was the key factor for the ECL inhibition of the system. Photoluminescence, cyclic voltammetry, coupled with bulk electrolysis, supports the supposition mechanism of the Ru(bpy)32+/TPrA–bergenin system. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

The reaction between glucose and methylene blue, catalyzed by glucose oxidase (GOD)was analysed calorimetrically. The amount of heat produced under saturating methylene blue concentrations ( > 10?2 mol/1)was measured with glucose concentration and time as parameters (kinetic procedure) Kinetic constants (pseudo one substrate kinetics) were derived from the experimental data: KM(glucose)= 1.18 × 10?3 mol/l and Vmax = 0.085 J/mg GOD min (3.89 · 10?6 mol/mg GOD min) Comparison of caloric with optical measurements gave an enthalpy of reaction of 22.52 kJ/mol. Considering the observed substrate inhibition, glucose determinations are possible up to glucose concentrations of 0.1 mol/l.  相似文献

The dimethylaminopyridine (DMAP) promoted reaction between [Os(bpy)2(CO)(OTf)]OTf (where ) and methylene chloride is reported. C-Cl bond breaking of a solvent molecule leads to the formation of the [Os(bpy)2(CO)(Cl)]OTf complex. The reactivity and redox properties of [Os(bpy)2(CO)(OTf)]OTf were investigated by means of room- and low-temperature electrochemical experiments. In CH2Cl2, at low temperature, the complex undergoes two 1e electrochemical and chemical reversible reductions (ErEr mechanism), but at room temperature a more complex electrochemical mechanism is observed, leading to the electro-synthesis of [Os(bpy)2(CO)(Cl)]OTf via electrochemical reversible and chemical irreversible reduction processes (ErCi mechanism). The DMAP nucleophilicity was used to produce the new [Os(bpy)2(CO)(Br)]OTf and [Os(bpy)2(CO)(I)]OTf complexes which have been fully characterized.  相似文献

Glucose potentiometric biosensor was prepared by immobilizing glucose oxidase on iodide-selective electrode. The hydrogen peroxide formed after the oxidation of glucose catalysed by glucose oxidase (GOD) was oxidized by sodium molybdate (SMo) at iodide electrode in the presence of dichlorometane. The glucose concentration was calculated from the decrease of iodide concentration determined by iodide-selective sensor. The sensitivity of glucose biosensor towards iodide ions and glucose was in the concentration ranges of 1.0 × 10?1–1.0 × 10?6 M and 1.0 × 10?2?1.0 × 10?4 M, respectively. The characterization of proposed glucose biosensor and glucose assay in human serum were also investigated.  相似文献

Four complexes [Pd(L)(bipy)Cl]·4H2O (1), [Pd(L)(phen)Cl]·4H2O (2), [Pt(L)(bipy)Cl]·4H2O (3), and [Pt(L)(phen)Cl]·4H2O (4), where L = quinolinic acid, bipy = 2,2’-bipyridyl, and phen = 1,10-phenanthroline, have been synthesized and characterized using IR, 1H NMR, elemental analysis, and single-crystal X-ray diffractometry. The binding of the complexes to FS-DNA was investigated by electronic absorption titration and fluorescence spectroscopy. The results indicate that the complexes bind to FS-DNA in an intercalative mode and the intrinsic binding constants K of the title complexes with FS-DNA are about 3.5?×?104 M?1, 3.9?×?104 M?1, 6.1?×?104 M?1, and 1.4?×?105 M?1, respectively. Also, the four complexes bind to DNA with different binding affinities, in descending order: complex 4, complex 3, complex 2, complex 1. Gel electrophoresis assay demonstrated the ability of the Pt(II) complexes to cleave pBR322 plasmid DNA.  相似文献

On the basis of an europium (III)‐doped Prussian blue analog film modifying platinum electrode as the working electrode, a Ru(bpy)32+‐based electrochemiluminescence (ECL) assay coupled with capillary electrophoresis has been first established for the determination of ketotifen fumarate (KTF). Analytes were injected onto a separation capillary of 50 cm length (50 μm i.d., 360 μm o.d.) by electrokinetic injection for 10 s at 10 kV. Parameters related to the separation and detection were discussed and optimized. It was proved that 15 mm phosphate buffer at pH 8.0 could achieve the most favorable resolution, and the highest sensitivity of detection was obtained using the detection potential at 1.25 V and 5 mm Ru(bpy)32+ in 100 mm phosphate buffer at pH 8.0 in the detection reservoir. Under the optimized conditions, the ECL intensity was in proportion to KTF concentration over the range from 3.0 × 10?8 to 5.0 × 10?6 g mL?1 with a detection limit of 2.1 × 10?8 g mL?1 (3σ). The relative standard deviations of the ECL intensity and the migration time were 0.95 and 0.26%, respectively. Three mononuclear CuII complexes, [CuCl(naph‐pa)] ( 1 ), [Cu(bipy)(naph‐pa)]Cl ( 2 ), and [Cu(naph‐pa)(phen)]Cl ( 3 ) ((naph‐pa)=Schiff base derived from the condensation of 2‐hydroxynaphthalene‐1‐carbaldehyde and 2‐picolylamine (=2‐(aminomethyl)pyridine), bipy=2,2′‐bypiridine, and phen=1,10‐phenanthroline) were synthesized and characterized. Complex 1 exhibits square‐planar geometry, and 2 and 3 exhibit square pyramidal geometry, where Schiff base and bipy/phen act as NNO and as NN donor ligands, respectively. CT (Calf thymus)‐DNA‐binding studies revealed that the complexes bind through intercalative mode and show good binding propensity (intrinsic binding constant Kb: 0.98×105, 2.22×105, and 2.67×105 M ?1 for 1 – 3 , resp.). The oxidative and hydrolytic DNA‐cleavage activity of these complexes has been studied by gel electrophoresis: all the complexes displayed chemical nuclease activity in the presence and absence of H2O2. From the kinetic experiments, hydrolytic DNA cleavage rate constants were determined as 2.48, 3.32, and 4.10 h?1 for 1 – 3 , respectively. It amounts to (0.68–1.14)×108‐fold rate enhancement compared to non‐catalyzed DNA cleavage, which is impressive. The interaction of native fish salmon DNA (FS-DNA) with [Eu(bpy)3Cl2(H2O)]Cl, where bpy is 2,2′-bipyridine, is studied at physiological pH in Tris-HCl buffer by spectroscopic methods, viscometric techniques as well as circular dichroism (CD). These experiments reveal that Eu(III) complex has interaction with FS-DNA. Moreover, binding constant and binding site size have been determined. The value of Kb has been defined 2.46 ± .02 × 105 M?1. The thermodynamic parameters are calculated by Van’t Hoff equation, the results show that the interaction of the complex with FS-DNA is an entropically driven phenomenon. CD spectroscopy followed by viscosity as well as fluorescence and UV––Vis measurements indicate that the complex interacts with FS-DNA via groove binding mode. Also, the synthesized Eu(III) complex has been screened for antimicrobial activities.  相似文献

New Os(II) complexes including [Os(dpop′)2](PF6)2 (dpop′= dipyrido(2,3-a;3′,2′-j)phenazine) and a series of mixed ligand [Os(dpop′)(N-N)Cl]PF6 (N-N = 2,2′-bipyridine(bpy); 2,2′-bipyrimidine(bpm) and 2,3-bis(2-pyridyl)pyrazine(dpp)) were synthesized. The Os dπ → dpop′ π MLCT transitions for [Os(dpop′)2]2+ are observed at lower energy than for Os dπ → tpy π (tpy = 2,2′:6′,2″-terpyridine) and Os dπ → tppz π (tppz = 2,3,5,6-tetrakis(2-pyridyl)pyrazine) (The ligand abbreviations tpd, tpp and tpypz have also appeared in the literature for 2,3,5,6- tetrakis(2-pyridyl)pyrazine in addition to tppz.) MLCT transitions in the comparative [Os(tpy)2]2+ and [Os(tppz)2]2+ complexes. The Os dπ → dpop′ π MLCT transitions are observed at lower energy in mixed bidentate ligand N-N systems compared with [Os(dpop′)2]2+. Cyclic voltammetry shows more positive osmium oxidation, and less negative ligand reduction potentials for [Os(dpop′)2]2+ as compared to [Os(tpy)2]2+ and [Os(tppz)2]2+ complexes. The osmium oxidation potentials in mixed ligand [Os(dpop′)(N-N)Cl]+ complexes are at less positive potential than for the [Os(dpop′)2]2+ ion. NMR results show different chemical shifts for ring protons either trans or cis to dpop′ in mixed ligand systems, and also show two geometrical isomers for the [Os(dpop′)(dpp)Cl]+ complex. The [Os(dpop′)(dpp)Cl]+ geometric isomer with the pyrazine ring of dpp trans to dpop′ is found more predominate by 1.0/0.7 over the isomer with the pyrazine ring of dpp cis to dpop′ and that inter-conversion of geometric isomers does not occur in room temperature solution on the NMR timescale.  相似文献

Reaction of ctc-OsBr2(RaaiR)2 [RaaiR=1-alkyl-2-(arylazo)imidazole, p-R-C6H4-NN-C3H2-NN-1-R, where R=H (a), Me (b), Cl (c) and R=Me (2), Et (3) and CH2Ph (4)] with 2,2-bipyridine (bpy) in presence of AgNO3 in EtOH followed by the addition of NH4PF6 afforded a mixed ligand complex [Os(bpy)(RaaiR)2](PF6)2. The structure of the complex, in one case [Os(bpy)(MeaaiMe)2](PF6)2 · 4H2O, has been confirmed by X-ray crystallography. The complexes are diamagnetic (low spin d6, s=0) and they show intense MLCT transition in the visible region (480-525 nm) and a weak transition at longer wavelength (>850 nm) in CH3CN solution. Cyclic voltammetry of the complexes show two metal oxidation, Os(II)/Os(III) at 0.72-0.76 V and Os(III)/Os(IV) at 1.34-1.42 V and three successive ligand reductions.  相似文献

Perturbation of the tris(2,2′‐bipyridine)ruthenium(II) [Ru(bpy)32+]‐catalyzed Belousov–Zhabotinsky (BZ) oscillating chemiluminescence (CL) reaction induced by l ‐cysteine was observed in the closed system. It was found that the CL intensity was decreased in the presence of l ‐cysteine. Meanwhile, oscillation period and oscillating induction period were prolonged. The sufficient reproducible induction period was used as parameter for the analytical application of oscillating CL reaction. Under the optimum conditions, the changes in the oscillating CL induction period were linearly proportional to the concentration of l ‐cysteine in the range from 8.0 × 10?7 to 5.0 × 10?5 mol L?1 (r = 0.997) with a detection limit of 4.3 × 10?7 mol L?1. This study describes the quenching effects of p‐aminobenzenesulfonic acid (p‐ABSA) based on electrochemiluminescence (ECL) of the tris (2,2‐bipyridyl)‐ruthenium(II)(Ru(bpy)32+)/tri‐n‐propylamine (TPrA) system in aqueous solution. Quenching behaviours were observed with a 200‐fold excess of p‐ABSA over Ru(bpy)32+. In the presence of 0.1 M TPrA, the Stern‐Volmer constant (KSV) of ECL quenching was as high as 1.39 × 104 M‐1 for p‐ABSA. The logarithmic plot of inhibited ECL versus concentration of p‐ABSA was linear over the range of 6.0 × 10‐6 ‐3.0 × 10‐4 mol/L. The corresponding limit of detection was 1.2 × 10‐6 mol/L for p‐ABSA (S/N = 3). The mechanism of quenching is believed to involve an energy transfer from the excited‐state luminophore to a dimer of p‐ABSA and the adsorption of free radicals of p‐ABSA at the electrode surface that impeded the oxidation of the Ru(bpy)32+/TPrA system. Given the increase in the incidence of insulin resistance, obesity, and type 2 diabetes in children and adolescents, it would be of paramount importance to assess quantitative indices of insulin secretion and action during a physiological perturbation, such as a meal or an oral glucose‐tolerance test (OGTT). A minimal model method is proposed to measure quantitative indices of insulin secretion and action in adolescents from an oral test. A 7 h, 21‐sample OGTT was performed in 11 adolescents. The C‐peptide minimal model was identified on C‐peptide and glucose data to quantify indices of β‐cell function: static φs and dynamic φd responsivity to glucose from which total responsivity φ was also measured. The glucose minimal model was identified on glucose and insulin data to estimate insulin sensitivity, SI, which was compared to a reference measure, SIref, provided by a tracer method. Disposition indices, which adjust insulin secretion for insulin action, were then calculated. Indices of β‐cell function were φs = 51.35 ± 8.89 × 10?9min?1, φd = 1,392 ± 258 × 10?9, and φ = 82.09 ± 17.70 × 10?9min?1. Insulin sensitivity was SI = 14.19 ± 2.73 × 10?4, not significantly different from SIref = 14.96 ± 3.04 × 10?4 dl/kg·min per µU/ml, and well correlated: r = 0.98, P < 0.0001, thus indicating that SI can be accurately measured from an oral test. Disposition indices were DIs = 1,040 ± 201 × 10?14 dl/kg/min2 per pmol/l, DId = 33,178 ± 10,720 × 10?14 dl/kg/min per pmol/l, DI = 1,844 ± 522 × 10?14 dl/kg/min2 per pmol/l. Virtually the same minimal model assessment was obtained with a reduced 3 h, 9‐sample protocol. OGTT interpreted with C‐peptide and glucose minimal model has the potential to provide novel insight regarding the regulation of glucose metabolism in adolescents, and to evaluate the effect of obesity and interventions such as diet and exercise.  相似文献

Based on the strong enhancement effect of procaterol hydrochloride on the electrochemiluminescence (ECL) of Ru(bpy)32+ (bpy = 2,2′‐bipyridine) in an alkaline H3PO4–NaOH buffer solution on a bare Pt electrode, a simple, rapid and sensitive method was developed for the determination of procaterol hydrochloride. The optimum conditions for the enhanced ECL have been developed in detail in this work. Under optimum conditions, the logarithmic ECL enhancement vs. the logarithmic concentration of procaterol hydrochloride is linear over a wide concentration range of 2.0 × 10?7 to 2.0 × 10?4 M (r =  0.9976), with a limit of detection of 1.1 × 10?8 M (S/N =  3), and a relative standard deviation of 2.1% (n =  7, c =  5.0 × 10?6 M). The proposed method was applied to the determination of this drug in tablets with recoveries of 89.7%–98.5%. In addition, a possible mechanism for the enhanced ECL of Ru(bpy)32+, which is caused by ProH, has also been proposed.  相似文献

Abstract

A new Ru(II) complex of [Ru(bpy)2(Hppip)]2+ {bpy = 2,2′-bipyridine; Hppip = 2-(4-(pyridin- 2-yl)phenyl)-1H-imidazo[4,5-f][1,10]phenanthroline} has been synthesized by grafting of 2-pyridyl to parent complex [Ru(bpy)2(Hpip)]2+ {Hppip = 2-(4-phenyl)-1H-imidazo[4,5-f] [1,10]phenanthroline}. The acid-base properties of [Ru(bpy)2(Hppip)]2+ studied by UV-visible and luminescence spectrophotometric pH titrations, revealed off-on-off luminescence switching of [Ru(bpy)2(Hppip)]2+ that was driven by the protonation/deprotonation of the imidazolyl and the pyridyl moieties. The complex was demonstrated to be a DNA intercalator with an intrinsic DNA binding constant of (5.56 ± 0.2) × 105 M?1 in buffered 50 mM NaCl, as evidenced by UV-visible and luminescence titrations, reverse salt effect, DNA competitive binding with ethidium bromide, steady-state emission quenching by [Fe(CN)6]4-, DNA melting experiments and viscosity measurements. The density functional theory method was also used to calculate geometric/electronic structures of the complex in an effort to understand the DNA binding properties. All the studies indicated that the introduction of 2-pyridyl onto Hpip ligand is more favorable for extension of conjugate plane of the main ligand than that of phenyl, and for greatly enhanced ct-DNA binding affinity accordingly.  相似文献

In this paper, it was found that glucose oxidase (GOD) has been stably immobilized on glassy carbon electrode modified by ordered mesoporous silica-SBA-15 and Nafion. The sorption behavior of GOD immobilized on SBA-15 matrix was characterized by transmission electron microscopy (TEM), ultraviolet–visible (UV–vis), FTIR, respectively, which demonstrated that SBA-15 can facilitate the electron exchange between the electroactive center of GOD and electrode. The direct electrochemistry and electrocatalysis behavior of GOD on modified electrode were characterized by cyclic voltammogram (CV) which indicated that GOD immobilized on Nafion and SBA-15 matrices displays direct, nearly reversible and surface-controlled redox reaction with an enhanced electron transfer rate constant of 3.89 s−1 in 0.1 M phosphate buffer solution (PBS) (pH 7.12). Furthermore, it was also discovered that, in the absence of O2, GOD immobilized on Nafion and SBA-15 matrices can produce a wide linear response to glucose in the positive potential range. Thus, Nafion/GOD-SBA-15/GC electrode is hopeful to be used in the third non-mediator's glucose biosensor. In addition, GOD immobilized on SBA-15 and Nafion matrices possesses an excellent bioelectrocatalytic activity for the reduction of O2. The Nafion/GOD-SBA-15/GC electrode can be utilized as the cathode in biofuel cell.  相似文献

This study aimed to overexpress a glucose oxidase gene (GOD1) in Aureobasidium sp. P6 to achieve Ca2+-gluconic acid (GA) overproduction. The GOD1 gene was cloned, deleted, and overexpressed. A protein deduced from the GOD1 gene of Aureobasidium sp. P6 strain had 1824 bp that encoded a protein with 606 amino acids, with a conserved NADB-ROSSMAN domain and a GMC-oxred domain. Deleting the GOD1 gene made the disruptant GOK1 completely lose the ability to produce GA and GOD1 activity, whereas overexpressing the GOD1 gene rendered the transformant GOEX8 to produce considerably more Ca2+-GA (160.5?±?5.6 g/L) and higher GOD1 activity (1438.6?±?73.2 U/mg of protein) than its parent P6 strain (118.7?±?4.3 g/L of Ca2+-GA and 1100.0?±?23.6 U/mg of GOD1 protein). During a 10-L fermentation, the transformant GOEX8 grown in the medium containing 160.0 g/L of glucose produced 186.8?±?6.0 g/L of Ca2+-GA, the yield was 1.2 g/g of glucose, and the volumetric productivity was 1.7 g/L/h. Most of the produced GOD1 were located in the yeast cell wall. The purified product was identified to be a GA. The transformant GOEX8 overexpressing the GOD1 gene could produce considerably more Ca2+-GA (186.8?±?6.0 g/L) than its wild-type strain P6.  相似文献

A tris(2,2‐bipyridyl)ruthenium(II) (Ru(bpy)32+)‐based electrochemiluminescence (ECL) detection coupled with capillary electrophoresis (CE) method has been established for the sensitive determination of ephedrine for the first time. Under the optimized conditions [ECL detection at 1.15 V, 25 mmol/L phosphate buffer solution (PBS), pH 8.0, as running buffer, separation voltage 12.5 kV, 5 mmol/L Ru(bpy)32+ with 60 mmol/L PBS, pH 8.5, in the detection cell] linear correlation (r = 0.9987) between ECL intensity and ephedrine concentration was obtained in the range 6.0 × 10–8–6.0 × 10–6 g/mL. The detection limit was 4.5 × 10–9 g/mL (S:N = 3). The developed method was successfully applied to the analysis of ephedrine in human urine and the investigation of its interactions with three proteins, including bovine serum albumin (BSA), cytochrome C (Cyt‐C) and myoglobin (Mb). The number of binding sites and the binding constants between ephedrine and BSA, Cyt‐C and Mb were 8.52, 12.60, 10.66 and 1.55 × 104 mol/L, 6.58 × 103 mol/L and 1.59 × 104 mol/L, respectively.
