Growth and pigment concentrations of the, estuarine dinoflagellate, Prorocentrum mariae-lebouriae (Parke and Ballantine) comb. nov., were measured in cultures grown in white, blue, green and red radiation at three different irradiances. White irradiances (400–800 nm) were 13.4, 4.0 and 1.8 W · m?2 with photon flux densities of 58.7 ± 3.5, 17.4 ± 0.6 and 7.8 ± 0.3 μM quanta · m?2· s?1, respectively. All other spectral qualities had the same photon flux densities. Concentrations of chlorophyll a and chlorophyll c were inversely related to irradiance. A decrease of 7- to 8-fold in photon flux density resulted in a 2-fold increase in chlorophyll a and c and a 1.6- to 2.4-fold increase in both peridinin and total carotenoid concentrations. Cells grown in green light contained 22 to 32% more peridinin per cell and exhibited 10 to 16% higher peridinin to chlorophyll a ratios than cells grown in white light. Growth decreased as a function of irradiance in white, green and red light grown cells but was the same at all blue light irradiances. Maximum growth rates occurred at 8 μM quanta · m?2· s?1 in blue light, while in red and white light maximum growth rates occurred at considerably higher photon flux densities (24 to 32 μM quanta · m?2· s?1). The fastest growth rates occurred in blue and red radiation. White radiation producing maximum growth was only as effective as red and blue light when the photon flux density in either the red or blue portion of the white light spectrum was equivalent to that of a red or of blue light treatment which produced maximum growth rates. These differences in growth and pigmentation indicate that P. mariae-lebouriae responds to the spectral quality under which it is grown.  相似文献

Uptake and assimilation of nitrogen and phosphorus were studied in Olisthodiscus luteus Carter. A diel periodicity in nitrate reductase activity was observed in log and stationary phase cultures; there was a 10-fold difference in magnitude between maximum and minimum rates, but other cellular features such as chlorophyll a, carbon, nitrogen, C:N ratio (atoms) · cell?1 were less variable. Ks values (~2 μM) for uptake of nitrate-N and ammonium-N were observed. Phosphorus assimilated · cell?1· day?1 varied with declining external phosphorus concentrations; growth rates <0.5 divisions · day?1 were common at <0.5 μM PO4-P. Phosphate uptake rates (Ks= 1.0–1.98 μM) varied with culture age and showed multiphasic kinetic features. Alkaline phosphatase activity was not detected. Comparisons of the nutrient dynamics of O. luteus to other phytoplankton species and the ecological implications as related to the phytoplankton community of Narragansett Bay (Rhode Island) are discussed.  相似文献

Clones of Skeletonema costatum (Grev.) Cl. isolated from Narragansett Bay, R.I., during different seasons were grouped according to their electrophoretic banding patterns. The growth rates, pg chlorophyll · cell?1, carbon uptake · cell?1· h?1, and carbon uptake · pg chl?1· h?1 were measured at 20°C, in a 14:10 h L:D cycle at 180 μE · m?2· s?1. Statistically significant sources of variation were found among groups of clones in growth rate, pg chl · cell?1, and carbon uptake · pg chl?1· h?1. It was concluded that there is a significant relationship between the physiological characteristics of clones isolated from populations in different seasons and patterns of genetic variation inferred from the electrophoretic studies. However, genetic diversity detected by banding patterns tends to underestimate the total genetic diversity in natural populations. The groups of clones most common in summer bloom populations had significantly higher growth rates, lower values of pg chl · cell?1, and higher rates of carbon uptake · pg chl?1· h?1 at 20°C than did the group of clones most common in winter bloom populations. However, differences among groups in these parameters at 20°C alone cannot account for the seasonal cycling of genetically variable populations of Skeletonema in Narragansett Bay. The range of growth rates among clones of this species is 0.1–5.0 divisions · d?1 under a single set of temperature and light conditions. Chlorophyll concentrations range from 0.2–1.7 pg chl · cell?1 and carbon uptake · pg chl?1· h?1 varies by a factor of 7 among clones. The range of physiological variation in this species means that it is difficult to use laboratory studies of single clones to analyze the responses of natural populations of Skeletonema.  相似文献

To develop tools for modeling diazotrophic growth in the open ocean, we determined the maximum growth rate and carbon content for three diazotrophic cyanobacteria commonly observed at Station ALOHA (A Long‐term Oligotrophic Habitat Assessment) in the subtropical North Pacific: filamentous nonheterocyst‐forming Trichodesmium and unicellular Groups A and B. Growth‐irradiance responses of Trichodesmium erythraeum Ehrenb. strain IMS101 and Crocosphaera watsonii J. Waterbury strain WH8501 were measured in the laboratory. No significant differences were detected between their fitted parameters (±CI) for maximum growth rate (0.51 ± 0.09 vs. 0.49 ± 0.17 d?1), half‐light saturation (73 ± 29 vs. 66 ± 37 μmol quanta · m?2 · s?1), and photoinhibition (0 and 0.00043 ± 0.00087 [μmol quanta · m?2 · s?1]?1). Maximum growth rates and carbon contents of Trichodesmium and Crocosphaera cultures conformed to published allometric relationships, demonstrating that these relationships apply to oceanic diazotrophic microorganisms. This agreement promoted the use of allometric models to approximate unknown parameters of maximum growth rate (0.77 d?1) and carbon content (480 fg C · μm?3) for the uncultivated, unicellular Group A cyanobacteria. The size of Group A was characterized from samples from the North Pacific Ocean using fluorescence‐activated cell sorting and real‐time quantitative PCR techniques. Knowledge of growth and carbon content properties of these organisms facilitates the incorporation of different types of cyanobacteria in modeling efforts aimed at assessing the relative importance of filamentous and unicellular diazotrophs to carbon and nitrogen cycling in the open ocean.  相似文献

The effects of acute stressors on physiological responses of juvenile great sturgeon or beluga Huso huso L. were investigated in two experiments. In the first experiment, fish were handled by placing them in containers at either low density (LD, one fish l?1) or high density (HD, four fish l?1) for 60 s. Concentrations of plasma cortisol, glucose and lactate were determined from blood collected at 0, 1, 3, 6 and 12 h after application of the stressor. Plasma cortisol concentrations increased after the disturbance in H. huso from both handling treatments, but changes were not significant. Plasma glucose rose significantly by 22·9 and 31·6% in LD and HD handling treatments, respectively, after 3 h. Significant increases in plasma lactate occurred within 1 h in both treatment groups, but that of the HD group was much higher. In the second experiment, fish were held at two different densities, LD (2 kg m?2 tank bottom surface area) and HD (7 kg m?2), for 8 weeks and then subjected to an aerial emersion handling stressor in a net for 60 s; blood samples were taken before handling (resting, 0 h) and at 1, 3, 6 and 9 h after handling. Plasma cortisol increased significantly in fish from the HD treatment from 8·8 ± 0·3 to 19·2 ± 2·4 ng ml?1 (mean ±s.e. ) by 1 h after stress, but post‐handling changes in the LD group were not significant. Significant increases in both plasma glucose and lactate were observed by 1 h in both treatment groups, with peak levels of plasma glucose evident at 3 h [69·4 ± 2·9 and 60·9 ± 1·7 mg dl?1 (mean ±s.e .) in LD and HD groups, respectively]. Plasma glucose levels were significantly higher in the LD group than in the HD group at 3 and 6 h. Post‐handling haemoglobin content increased by 1 h and white blood cell numbers were reduced by 3 and 6 h in the HD treatment group compared with resting values, but changes in these blood features in the LD group were not significant. Acute handling did not affect haematocrit in either treatment. The results suggest that H. huso is relatively resistant to handling and confinement, and could tolerate normal hatchery practices associated with aquaculture. Because changes in cortisol concentrations were relatively low compared with those in most teleosts, glucose and lactate concentrations may be more useful as stress indicators in juvenile H. huso. This study also demonstrated that prior exposure to a chronic stressor, specifically high stocking density, could alter the physiological response to subsequent acute handling in H. huso.  相似文献

Protoplasts were prepared from Ulva fasciata Delile, and their photosynthetic performance was measured and compared with that of thalli discs. These protoplasts maintained maximal rates of photosynthesis as high as those of thalli (up to 300 μmol O2·mg chlorophyll?1·h?1) for several hours after preparation and were therefore considered suitable for kinetic studies of inorganic carbon utilization. The photosynthetic K1/2(inorganic carbon) at pH 6.1 was 3.8 μM and increased to 67, 158, and 1410 μM at the pH values 7.0, 7.9, and 8.9, respectively. Compared with these protoplasts, thalli had a much lower affinity for CO2 but approximately the same affinity for HCO3?. Comparisons between rates of photosynthesis and the spontaneous dehydration of HCO3? (at 50 μM inorganic carbon) revealed that photosynthesis of both protoplasts (which lacked apparent activity of extracellular/surface-bound carbonic anhydrase) and thalli (which were only 25% inhibited by the external carbonic anhydrase inhibitor acetazolamide) could not be supported by CO2 formation in the medium at the higher pH values, indicating HCO3? uptake. Since both protoplasts and thalli were sensitive to 4,4′-diisothiocyanostilbene-2,2′-disulfonate, we suggest that HCO3? transport was facilitated by the membrane-located anion exchange protein recently reported to function in certain Ulva thalli. These findings suggest that the presence of a cell wall may constitute a diffusion barrier for CO2, but not for HCO3?, utilization under natural seawater conditions.  相似文献

Two axenic, in vitro liquid suspension cultures were established for Agardhiella subulata (C. Agardh) Kraft et Wynne, and their growth characteristics were compared. This study illustrated how reliable routes for the development of suspension cultures of macrophytic red algae of terete thallus morphology can be achieved for biotechnology applications. Undifferentiated filament clumps of 2–8 mm diameter were established by induction of callus-like tissue from thallus explants, and lightly branched microplantlets of 2–10 mm length were established by regeneration of filament clumps. The filament clumps were susceptible to regeneration. Adventitious shoot formation was reliably induced from 40% to 70% of the filament clumps by gentle mixing at 100 rev min?1 on an orbital shaker. The specific growth rate of the microplantlets was higher than the filament clumps in nonagitated well plate culture (4%–6% per day for microplantlets vs. 2%–3% per day for filament clumps) at 24° C and 8–36 μmol photons·m?2·s?1 irradiance (10:14 h LD cycle) when grown on ASP12 artificial seawater medium at pH 8.6–8.9 with 20%–25% per day medium replacement. Oxygen evolution rate vs. irradiance measurements showed that relative to the filament clumps, microplantlets had a higher maximum specific oxygen evolution rate (Po,max= 0.181 ± 0.035 vs. 0.130 ± 0.023 mmol O2·g?1 dry cell mass·h?1), but comparable respiration rate (Qo= 0.040 ± 0.013 vs. 0.033 ± 0.017 mmol O2·g?1 dry cell mass·h?1), compensation point (Ic= 3.8 ± 2.4 vs. 5.7 ± 1.2 μmol photons·m?2·s?1), and light intensity at 63.2% of saturation (Ik= 17.5 ± 3.9 vs. 14.9 ± 2.6 μmol photons·m?2·s?1). The microplantlet culture was more suitable for suspension culture development than the filament clump culture because it was morphologically stable and exhibited higher growth rates.  相似文献

Using time-course, natural-light incubations, we assessed the rate of carbon uptake at a range of light intensities, the effect of supplemental additions of nitrogen (as NH4+ or urea) on light and dark carbon uptake, and the rates of uptake of NH4+ and urea by phytoplankton from Vineyard Sound, Massachusetts from February through August 1982. During the winter, photoinhibition was severe, becoming manifested shortly after the start of an incubation, whereas during the summer, there was little to no evidence of photoinhibition during the first several hours after the start of an incubation. At light levels which were neither photoinhibiting nor light limiting, rates of carbon uptake normalized per liter were high and approximately equal during winter and summer (22–23 μg C·l?1 · h?1), and low during spring (<10 μgC·l?1· h?1). In contrast, on a chlorophyll a basis, rates of carbon fixation were as high during spring (15–20μg C·μg Chl a?1·h?1), when concentrations of chlorophyll a were at the yearly minimum (<0.5 μg · l?1) as during the summer, when chlorophyll a concentrations were substantially higher (0.8–1.3 μg · l?1). Highest rates of NH4+ and urea uptake were observed during summer, and at no time of the year was there evidence for severe nitrogen deficiency, although moderate nitrogen nutritional stress was apparent during the summer months.  相似文献

The membraneless bioelectrochemical reactor (Ml-BER) is useful for dark hydrogen fermentation. The effect of the electrochemical reaction on microorganisms in the Ml-BER was investigated using glucose as the substrate and compared with organisms in a membraneless non-bioelectrochemical reactor (Ml-NBER) and bioelectrochemical reactor (BER) with a proton exchange membrane. The potentials on the working electrode of the Ml-BER and BER with membrane were regulated to ?0.9 V (versus Ag/AgCl) to avoid water electrolysis with a carbon electrode. The Ml-BER showed suppressed methane production (19.8?±?9.1 mg-C·L?1·day?1) and increased hydrogen production (12.6?±?3.1 mg-H·L?1·day?1) at pHout 6.2?±?0.1, and the major intermediate was butyrate (24.9?±?2.4 mM), suggesting efficient hydrogen fermentation. In contrast, the Ml-NBER showed high methane production (239.3?±?17.9 mg-C·L?1·day?1) and low hydrogen production (0.2?±?0.0 mg-H·L?1·day?1) at pHout 6.3?±?0.1. In the cathodic chamber of the BER with membrane, methane production was high (276.3?±?20.4 mg-C·L?1·day?1) (pHout, 7.2?±?0.1). In the anodic chamber of the BER with membrane (anode-BER), gas production was low because of high lactate production (43.6?±?1.7 mM) at pHout 5.0?±?0.1. Methanogenic archaea were not detected in the Ml-BER and anode-BER. However, Methanosarcina sp. and Methanobacterium sp. were found in Ml-NBER. Prokaryotic copy numbers in the Ml-BER and Ml-NBER were similar, as were the bacterial community structures. Thus, the electrochemical reaction in the Ml-BER affected hydrogenotrophic and acetoclastic methanogens, but not the bacterial community.  相似文献

Light intensity and temperature interactions have a complex effect on the physiological process rates of the filamentous bluegreen alga Anabaena variabilis Kütz. The optimum temperature for photosynthesis increased with increasing light intensity from 10°C at 42 μE·m?2·s?1 to 35°C at 562 μE·m?2·s?1. The light saturation parameter, IK, increased with increasing temperatures. The maximum photosynthetic rate (2.0 g C·g dry wt.?1·d?1) occurred at 35°C and 564 μE·m?2·s?1. At 15°C, the maximum rate was 1.25 g C·g dry wt.?1·d?1 at 332 μE·m?2·s?1. The dark respiration rate increased exponentially with temperature. Under favorable conditions of light intensity and temperature the percent of extracellular release of dissolved organic carbon was less than 5% of the total C fixed. This release increased to nearly 40% under combinations of low light intensity and high temperature. A mathematical model was developed to simulate the interaction of light intensity and temperature on photosynthetic rate. The interactive effects were represented by making the light-saturation parameters a function of temperature.  相似文献

The stress response of Oncorhynchus mykiss in high‐altitude farms in central Mexico was investigated over two seasons: the cool (9·1–13·7° C) dry winter season, and the warmer (14·7–15·9° C), wetter summer season. Fish were subjected to an acute stress test followed by sampling of six physiological variables: blood cortisol, glucose, lactate, total antioxidant capacity, haemoglobin concentration and per cent packed cell volume (VPC%). Multivariate analyses revealed that lactate and total antioxidant capacity were significantly higher in the summer, when water temperatures were warmer and moderate hypoxia (4·9–5·3 mg l?1) prevailed. In contrast, plasma cortisol was significantly higher in the winter (mean ± s.e .: 76·7 ± 4·0 ng ml?1) when temperatures were cooler and dissolved oxygen levels higher (6·05–7·9 mg l?1), than in the summer (22·7 ± 3·8 ng ml?1). Haemoglobin concentrations (mg dl?1) were not significantly different between seasons, but VPC% was significantly higher in the summer (50%) than in the winter (35%). These results suggest that in summer, effects of high altitude on farmed fish are exacerbated by stresses of high temperatures and hypoxia, resulting in higher blood lactate, increased total antioxidant capacity and elevated VPC% levels.  相似文献

Cellular nutrient concentrations and nutrient uptake rates of Cladophora glomerata (L.) Kuetzing were determined during summer and fall in 1989–1990 at a site on the upper Clark Fork of the Columbia River, Montana. Both physiological tests indicated that Cladophora growth is likely to be limited by nitrogen during late summer-early fall. Maximum uptake rates of ammonia-N and nitrate-N were 5935–6991 and 507–984 μg · g DW?1· h?1, respectively, during July–October when dissolved inorganic nitrogen (DIN) concentrations in the river were less than 10 μg · L?1. During November-December, when DIN was 72–376 μg · L?1, maximum ammonia-N uptake was 1137–1633 μg · g DW?1· h?1 and maximum nitrate-N uptake was 0–196 μg · g DW?1· h?1. Cellular nitrogen during summer–early fall was 0.78–1.80% of Cladophora dry weight, frequently at or below 1.1%, a level suggested as a critical minimum N concentration for maximum growth. In contrast, cellular P was 0.18–0.36% of dry weight, 3–6 times the suggested critical P concentration of 0.06%. Molar ratios of cellular N:P (< 16:1) and DIN: SRP (< 4:1) during late summer-early fall also indicated potential N limitation. Cellular N and P from Cladophora collected from a second site influenced by a municipal wastewater discharge in 1990 displayed similar seasonal trends. At both sites, seasonal fluctuations in DIN were closely tracked by changes in cellular N, Cellular P, however, increased through the growing season despite declining levels of SRP in the river.  相似文献

Baseline, post‐angling and maximum attainable blood lactate concentrations were measured for the fishery species redthroat emperor Lethrinus miniatus to gain insight into the condition of fish released following c. 30 s angling and <45 s air exposure. Mean ± s.d . baseline blood lactate was 1·5 ± 0·6 mmol l?1, which increased and plateaued around 6 mmol l?1 at 15–30 min post‐angling. These values were significantly lower than those obtained from fish maximally exhausted with a prolonged chase and air exposure protocol following capture (10·9 ± 1·8 mmol l?1), suggesting that L. miniatus is not maximally exhausted during standard angling practices.  相似文献

The relative importance of respiration and organic carbon release to the efficiency of carbon specific growth of Skeletonema costatum (Grev.) Clave was evaluated over a light range from 1500–15 μE · m?2· s?1. Net growth efficiency ranged from 0.45–0.69 with a maximum at 130 μE · m?2· s?1. Respiration was 93% or more of the variations in growth efficiency. Organic carbon release ranged from 0–7% of gross production and increased with light intensity. Carbon specific particulate production was a hyperbolic function of incident light intensity and was related exponentially to particulate carbon production per unit chlorophyll a. Full sunlight conditions, 1500 μE · m?2· s?1, did not induce photoinhibition of gross production. Variations in the efficiency of growth of S. costatum were minimized over a wide range of light intensities mainly because of variations in cellular pigments which permitted the efficient utilization of available light energy, and a reduction in the losses of carbon which increases the growth rate, possibly as a consequence of the recycling of respired carbon within the cell.  相似文献

Cultures of Trichodesmium NIBB 1067 were grown in the synthetic medium AQUIL with a range of iron added from none to 5 × 10?7 M Fe for 15 days. Chlorophyll-a, cell counts, and total cell volume were two or three times higher in medium with 10?7 M Fe than with no added Fe. Oxygen production rate per chlorophyll-a was over 60% higher with higher iron. Increased iron stimulated photosynthesis at all irradiances from about 12–250 μE · m?2· s?1. Nitrogen fixation rate, estimated from acetylene reduction, for 10?7 and 10?8 M Fe cultures was approximately twice that of the cultures with no added Fe. The range of rates of O2 production and N2 fixation in cultures at the iron concentrations we used were similar to the rates from natural samples of Trichodesmium from both the Atlantic, and the Pacific oceans. This similarity may allow this clone to be used, with some caution, for future physiological ecology studies. This study demonstrates the importance of iron to photosynthesis and nitrogen fixation and suggests that Trichodesmium plays a central role in the biogeochemical cycles of iron, carbon and nitrogen.  相似文献

The internal lipid, carotenoid, and toxin concentrations of Karenia brevis (C. C. Davis) Gert Hansen and Moestrup are influenced by its ability to use ambient light and nutrients for growth and reproduction. This study investigated changes in K. brevis toxicity, lipid class, and carotenoid concentrations in low‐light, nitrate‐replete (250 μmol quanta · m?2 · s?1, 80 μM NO3); high‐light, nitrate‐replete (960 μmol quanta · m?2 · s?1, 80 μM NO3); and high‐light, nitrate‐reduced (960 μmol quanta · m?2 · s?1, <5 μM NO3) mesocosms. Reverse‐phase HPLC quantified the epoxidation state (EPS) of the xanthophyll‐cycle pigments diadinoxanthin and diatoxanthin, and a Chromarod Iatroscan thin layer chromatography/flame ionization detection (TLC/FID) system quantified changes in lipid class concentrations. EPS did not exceed 0.20 in the low‐light mesocosm, but increased to 0.65 in the high‐light mesocosms. Triacylglycerol and monogalactosyldiacylglycerol (MGDG) were the largest lipid classes consisting of 9.3% to 48.7% and 37.3% to 69.7% of total lipid, respectively. Both lipid classes also experienced the greatest concentration changes in high‐light experiments. K. brevis increased EPS and toxin concentrations while decreasing its lipid concentrations under high light. K. brevis may mobilize its toxins into the surrounding environment by reducing lipid concentrations, such as sterols, limiting competition, or toxins are released because lipids are decreased in high light, reducing any protective mechanism against their own toxins.  相似文献

Marine phytoplankton and macroalgae acquire important resources, such as inorganic nitrogen, from the surrounding seawater by uptake across their entire surface area. Rates of ammonium and nitrate uptake per unit surface area were remarkably similar for both marine phytoplankton and macroalgae at low external concentrations. At an external concentration of 1 μM, the mean rate of nitrogen uptake was 10±2 nmol·cm?2·h?1 (n=36). There was a strong negative relationship between log surface area:volume (SA:V) quotient and log nitrogen content per cm2 of surface (slope=?0.77), but a positive relationship between log SA:V and log maximum specific growth rate (μmax; slope=0.46). There was a strong negative relationship between log SA:V and log measured rate of ammonium assimilation per cm2 of surface, but the slope (?0.49) was steeper than that required to sustain μmax (?0.31). Calculated rates of ammonium assimilation required to sustain growth rates measured in natural populations were similar for both marine phytoplankton and macroalgae with an overall mean of 6.2±1.4 nmol·cm?2·h?1 (n=15). These values were similar to maximum rates of ammonium assimilation in phytoplankton with high SA:V, but the values for algae with low SA:V were substantially less than the maximum rate of ammonium assimilation. This suggests that the growth rates of both marine phytoplankton and macroalgae in nature are often constrained by rates of uptake and assimilation of nutrients per cm2 surface area.  相似文献

The effects of diurnal variations in light intensity on the biomass characteristics and the efficiency of daily growth of Skeletonema costatum (Grev.) Cleve were evaluated. The relative importance of changes in carbon specific rates of respiration and organic release to the efficiency of growth was determined. Light intensity was either constant at 130 μE · m?2 · s?1 during the light period or fluctuated throughout the light period from 500 to 10 μE · m?2 · s?1 at rates of either 1 or 12 cycles · day?1. Total daily light was equivalent for all light regimes at 5.6 E · m?2 · day?1.Daily rates of growth remained comparable at ≈ 1 · day?1 under constant and fluctuating light regimes. Cell size as daily mean carbon · cell?1, nitrogen · cell?1 and cellular volume was decreased under diurnally varying light whereas daily mean chlorophyll a · cell?1 was unaffected.Rates of respiration, organic release and gross production were elevated several fold under diurnally varying light in comparison to constant light. Net growth efficiency decreased from 0.69 under constant light to values of 0.50 and 0.38 under 1 and 12 cycles · day?1, respectively. Decreased efficiency of growth under diurnally fluctuating light resulted mostly from greater respiratory activity while organic release remained < 10% of gross production. Increased rates of gross production reflected enhancement in the efficiency of carbon fixation with fluctuating light.  相似文献

Lipid content and lipid class composition were determined in stream periphyton and the filamentous green algae Cladophora sp. and Spirogyra sp, Sterols and phospholipids were compared to chlorophyll a (chl a) as predictors of biomass for stream periphyton and algae. Chlorophyll a, phospholipids, and sterols were each highly correlated with ash-free dry mass (AFDM) (r2 > 0.98). Stream periphyton exposed naturally to high light (HL) and low light (LL) had chl a concentrations (μg chl a-mg?1AFDM) of 7.9± 0.7 and 12.4 ± 2.9, respectively, while the sterol concentrations of these HL and LL stream periphyton (1.6 ± 0.4) were not significantly different (P > 0.05). Periphyton exposed to an irradiance of 300 μmol photons·m?2s?1 in the laboratory for 60 h had 5.6 ± 0.55 μg chl a·mg?1 AFDM, but the same periphyton exposed to 2% incident light for the same amount of time had 11.0 ± 0.56 μg chl mg?1 AFDM. Sterol concentrations in these periphyton communities remained unchanged (1.5 ± 0.3 μg·mg?1AFDM), Similar results (i.e. changes in chl a but stability of sterol concentrations in response to irradiance changes) were also found for Cladophora and Spirogyra in laboratory experiments. Sterols can be quantified rapidly from a few milligrams of algae and appear to be a useful predictor of eukaryote biomass, whereas cellular levels of chl a vary substantially with light conditions. Phospholipids (or phospholipid fatty acids) are considered to be a reliable measure of viable microbial biomass. Nevertheless, phospholipid content varied substantially and unpredictably among algae and periphyton under different light regimes. Irradiance also had a significant effect on storage lipids: HL Cladophora and HL periphyton had 2 × and 5 × greater concentrations of triacylglycerols, respectively, compared to their LL forms. HL and LL algae also differed in the concentration of several major fatty acids. These light-induced changes in algal lipids and fatty acids have important implications for grazers.  相似文献

The effects of irradiance on the biochemical composition of the prymnesiophyte microalga, Isochrysis sp. (Parke; clone T-ISO), a popular species for mariculture, were examined. Cultures were grown under a 12:12 h light: dark (L:D) regime at five irradiances ranging from 50 to 1000 μE·m 2·s?1 and harvested at late-logarithmic phase for analysis of biochemical composition. Gross composition varied aver the range of irradiances. The highest levels of protein were present in cells from cultures grown at 100 and 250 μE·m 3·s1, and minimum levels of carbohydrate and lipid occurred at 50 μE·m?2·s?1. Because the cell dry weight was reduced at lower irradiances, different trends were evident when results were expressed as percentage of dry weights. Protein percentages were highest at Wand 100 μE·m?2·s?1 and carbohydrate at 100 μE·m?2·s?1. The composition of amino acids did not differ over the range of irradiances. Glutamate and aspartate were always present in high proportions (9.0–13.5%); histidine. methionine, tryptophan, cystine, and hydroxy-proline were minor constituents (0.0–2.6%). Glucose was the predominant sugar in all cultures, ranging from 23.0% (50 μE·m?2·s?1) to 45.0% (100 μE·m?2·s?1) of total polysaccharide. No correlation was found between the proportion of any of the sugars and irradiance. The proportions of the lipid class components and fatty acids showed little change with irradiance. The main fatty acids were 14:0, 16:0, 16:1(n-7), 18:1(n-9), 18:3(n-3). 18:4(n-3), 18:5(n-3), and 22:6(n-3). Proportions of 22: 6(n-3) increased, whereas l8:3(n-3). 18:3(n-6). and 18:4(n-3) decreased, with increasing irradiance. Pigment concentrations were highest in cultures grown at 50 μE·m?2·s?1, except for fucoxanthin and diadinoxanthin (100 μE·m?2·s?1). The concentrations of accessory pigments correlated with chlorophyll a, which decreased in concentration with increasing irradiance. On the basts of biochemical composition, an irradiance of 100 μE·m?1·s?1 (12:12 h L:D cycle)for the culture of Isochrysis sp. (clone T-ISO) may provide optimal nutritional value for maricultured animals, although feeding trials are now necessary to substantiate this.  相似文献
