Summary The thyroid gland of adult salamanders, Hynobius nebulosus, in the breeding season was studied by electron microscopy. The follicular cells are different in cell height and fine structures; the taller cells with many cell organelles and granules and the lower cells with a few cell organelles and granules are both present in the same follicle. In the cytoplasm, three types of membrane-bounded granules, namely, cytosomes, colloid droplets, and vacuolar bodies and circular membrane complexes occur. The vacuolar bodies are subdivided into two types; the ordinary type having loosely distributed particles and the specific type containing tubules and/or closely packed filaments, crystalloid structures, except for the particles. The chromophobe colloids within the Bensley-cells correspond to extremely large, ordinary type vacuolar bodies, while the Langendorff-colloid cells possess increased numbers of granular cisternae of endoplasmic reticulum and a ribosome-rich, dense cytoplasmic matrix but not extremely large colloid. The intracytoplasmic circular membrane complexes appear in the Golgi area of cytosome-rich cells. It is suggested that they originate from the Golgi apparatus which was activated to produce many cytosomes. Intranuclear inclusions consisting of microtubules and filaments and tight junctions between two adjacent lateral plasma membranes are occasionally encountered.  相似文献

The subcommissural organ (SCO) of the golden hamster (Mesocricetus auratus) was studied by conventional electron microscopy, freeze-fracture technique, zinc-iodide-osmium (ZIO) and acid phosphatase cytochemical reactions. The ultrastructure of hamster SCO cells shows a few flattened cisternae of rough endoplasmic reticulum (ER) without dilated ones in the cytoplasm. The Golgi apparatus is very well developed. Freeze-fracture studies also indicate only short profiles of flattened ER in the cytoplasm endorsing the absence of dilated ER cisternae. After the treatment with ZIO mixture, reaction products were observed over flattened cisternae of the ER and the nuclear envelope. The Golgi apparatus was also reactive toward the ZIO mixture. Acid phosphatase activities are localized in the inner one or two saccules of the Golgi apparatus and dense bodies. From these results we suggest that (1) hamster SCO cells do not accumulate secretory material in the cytoplasm in the form of discrete secretory granules or dilated cisternae of ER, and (2) hamster SCO cells may possess extremely high secretory activity or may not be actively involved in secretory function at all as in rats or other rodents.  相似文献

The fine structure of the nerve cell types in the white planarian Procotyla fluviatilis were described. Ganglion cells comprise the major portion of the brain. These cells are irregular in shape with several cytoplasmic processes and contain ribosomes, a sparse endoplasmic reticulum, microtubules, lysosomes, and a Golgi apparatus with numerous small vesicles. Granule-containing cells are situated in the peripheral regions of the brain and along the nerve cords. These cells contain ribosomes, rough-surfaced endoplasmic reticulum and a Golgi apparatus with associated dense granules. The granules occupy most of the cytoplasm and are ~ 750A in diameter with moderately dense contents, ~ 750A with opaque contents, and ~ 1000A with contents of medium density. These granules are similar to those in the nervous systems of higher animals that contain epinephrine, norepinephrine, and neurosecretory substance, respectively. Each cell contains predominantly one type of granule although there is some intermixing of granules and intermediate types between the three most abundant granules. Small clear vesicles, resembling cholinergic synaptic vesicles, and all types of dense granules occur in the neuropil and within nerve endings.  相似文献

The epididymis of the fan-throated lizard Sitana ponticeriana was examined with light and transmission electron microscopy to understand the cellular mechanisms of fabrication of secretion granules in epithelial principal cells, granule release into the lumen, and the fate of the dense structured granules after reaching the lumen. Principal cells of the ductus epididymis, except at the cauda, secrete electron-dense biphasic granules copiously, which decrease in abundance from the initial segment to corpus. The principal cell possesses a prominent Golgi apparatus and all versions of endoplasmic reticulum (ER), rough, smooth, and sparsely granulated. The material of the dense portion of the secretion granules, after processing at the Golgi apparatus, appears to accumulate in large ER cisternae in the supranuclear cytoplasm. It undergoes condensation when the cisternae become condensing vacuoles. Mitochondria appear to play a role in dense granule formation. The condensing vacuoles are displaced toward the apical cytoplasm when the material of the less dense portion is added to the condensing vacuoles at the Golgi area. Thus, the less dense and dense portions of the secretion granules are secreted and added to the condensing vacuoles separately. The composite granules are released into the lumen by exocytosis when the less dense portion merges with the luminal content, whereas the dense portion maintains its structured identity. The latter, initially measuring 1-2 microm in diameter, increases in size several times. It is inferred that these granules release their content gradually, resulting in the appearance of vacuoles, and suggesting that the granules have an insoluble matrix in which there is a sparingly soluble material. The substance leaching out of the granules appears to contribute to keeping the sperm quiescent and alive during storage in the male reproductive tract.  相似文献

The normal parathyroids of six humans and a Virginia deer were studied by light and electron microscopy. The parenchyma of the deer parathyroid is composed of uniform chief cells, which contained 100 to 400 mµ electron-opaque, membrane-limited granules, presumed to be secretory granules, in addition to the usual cytoplasmic organelles. Desmosomes are present between adjacent cells, and rare cilia are observed protruding from the chief cells into the intercellular space. The human parathyroids contain chief cells in two phases—active and inactive—as well as oxyphil cells. Active chief cells have a large Golgi apparatus, sparse glycogen, numerous secretory granules, and rare cilia. Inactive chief cells contain a small Golgi apparatus, abundant glycogen, and few secretory granules. Both forms have the usual cytoplasmic organelles and, between adjacent cells, desmosomes. Oxyphil cell cytoplasm is composed of tightly packed mitochondria and glycogen granules, with rare secretory granules. Cells with cytoplasmic characteristics intermediate between chief and oxyphil cells, possibly representing transitional cells, have been observed. Secretory granules of both man and deer are composed of 100 to 200 A particles and short rods, and the granules develop from prosecretory granules in the Golgi region of the cell. The human secretory granules are smaller and more variable in shape than those of the deer. The granules are iron and chrome alum hematoxylin-positive, argyrophilic, and aldehyde fuchsin-positive, permitting light microscopic identification. They are also found in the capillary endothelial cells of the parathyroid and in its surrounding connective tissue. The secretory granules of the parathyroid cells can thus be followed from their formation in the Golgi apparatus almost to their extrusion into the blood stream.  相似文献

Summary For the purpose used in understanding thyroid phylogenesis, the fine structure and the iodine metabolism of the endostyle of Ascidians,Ciona intestinalis, was studied by electron microscopy and electron microscopic autoradiography. There are 8 kinds of zones in the endostyle.Zone 1, 3, and 5 cells, especially zone 1 cells, are characterized by numerous long cilia. These cells which show no indications of protein-secretion but numerous small vesicles and cytoplasmic filaments might play a role in catching and transporting food, absorption of liquid and supporting the endostylar construction.Zone 2, 4, and 6 cells are large and characterized by well developed rough endoplasmic reticulum and numerous electron-dense secretory granules which are considered to be synthesized in the rough endoplasmic reticulum and transported to the Golgi apparatus to mature. They, which are somewhat similar to the pancreatic exocrine cells in fine structure, are believed to secrete the proteinous or mucoproteinous substances which might be related to the digestion of food.Zone 7 and 8 cells which might be homologous to the thyroid cell of the higher vertebrate contains poorly developed rough endoplasmic reticulum, small Golgi apparatus, a few multivesicular bodies, a few lysosomes, and numerous small vesicles. In addition zone 8 cells bear cilia on their apical surface. The cytoplasmic characteristics of these cell types, especially of zone 8 cells, are fairly similar to those of type 2C and type 3 cells of the endostyle of a larval lamprey, though the rough endoplasmic reticulum is not so well developed. By electron microscopic autoradiography numerous silver grains were observed on the apical cell membrane region of zone 7 and 8 cells, especially of zone 8 cells, 1, 4, 6, 16 and 24 hours after immersion in sea water containing125I. This fact suggests that the iodination takes place in the apical cell membrane region of these cells. The materials in the endostylar lumen is washed away during the fixation and dehydrating processes of the tissue. Therefore, the possibility of iodination of thyroglobulin-like substances taking place within the endostylar lumen cannot be ruled out. Grains were also found in the multivesicular bodies and lysosomes after 4, 6, 16 and 24 hours, especially 16 and 24 hours. It seems that the organic iodine might be reabsorbed into the cytoplasm of these cells.This investigation was supported by research grant from Dr. Henry C. Summary The fine structure of the main excretory duct epithelium of the male mouse submandibular glands was investigated by scanning and transmission electron microscopy. Three principal cell-types were observed: type I and II, and basal cells. This epithelium was characterized by the presence of intercellular canaliculi. Type-I cells were the most numerous. They had an abundance of mitochondria, well-developed Golgi apparatus, a few electron-lucent lipid-containing granules and poorly developed basal infoldings. These cells were also characterized by many glycogen granules throughout the cytoplasm and abundant smooth endoplasmic reticulum in the apical cytoplasm. Type-II cells were the second most numerous. Their most characteristic feature was the presence of abundant heterogeneous lipid-containing granules having acid phosphatase activity at the periphery. They were concentrated in the infra- and supranuclear cytoplasm. The granules may be derived from mitochondrial transformation and seem to be a special kind of secondary autolysosome. Type-II cells also contained abundant mitochondria throughout the cytoplasm, much smooth endoplasmic reticulum in the apical cytoplasm, a well developed Golgi apparatus adjacent to the heterogeneous lipid-containing granules and no basal infoldings. Basal cells were situated adjacent to the basal lamina. They had a large nucleus and the cytoplasm was filled with glycogen granules.  相似文献

Ultrastructural changes of hepatocytes were examined in the sand lamprey,Lampetra reissneri, during various phases of the life cycle. In hepatocytes of ammocoetes, the rough endoplasmic reticulum was composed of short cisternae and the Golgi apparatus were scarcely developed, showing no sexual differences at this stage of life cycle. In hepatocytes of female lampreys at the metamorphic stages 4 to 5, the rough endoplasmic reticulum was developed to form long parallel cisternae and the Golgi apparatus were well-developed. The rough endoplasmic reticulum developed further to form stacks of long parallel cisternae extending over the cytoplasm in hepatocytes of females at the young adult stage, and became composed of both long parallel and vesicular cisternae in the cells of females at the adult stage. The Golgi apparatus were invariably welldeveloped in hepatocytes of young adult and adult females. No consipcuous development was observed in profiles of the rough endoplasmic reticulum and the Golgi apparatus in hepatocytes of males during and after metamorphosis. The ultrastructural changes of the rough endoplasmic reticulum and the Golgi apparatus observed in hepatocytes of female sand lampreys are considered to have an intimate relation to the activity of vitellogenin synthesis in the liver, and it is suggested that the hepatocytes begin to rapidly synthesize vitellogenin in the sand lamprey at the metamorphic stages 4 to 5.  相似文献

Summary Experiments were carried out to study the iodine metabolism of the endostyle of the larval lamprey which is considered to be homologous to the thyroid gland. Larval lampreys, ammocoetes of Lampetra japonica were intraperitoneally injected with 200 c of Na 125I; their endostyles were removed 30 minutes, 1, 2, 4, 6, 8 and 24 hours after the treatment. Type 1 and type 4 cells (Marine) were almost inactive in binding iodine. Silver grains appeared within 30 minutes after the injection over the apical cell membrane including the surfaces of microvilli and cilia of type 2 c and type 3 cells. These grains increased in number until 2 hours. A few of apical small vesicles of the same cells were labeled 1 to 2 hours after the injection. Small dense granules large dense bodies, and multivesicular bodies in the type 2 c and type 3 cells were labeled especially at 6 to 24 hours. The ratio in number of the labeled dense granules, or bodies to the unlabeled ones tended to increase markedly with time. Large or small vacuoles, dense or light in the cytoplasm of some type 5 cells which lack indications of protein-synthesis sign in the cytoplasm were labeled 6 to 24 hours after the injection of 125I, and the number of the labeled vacuoles increased with time. From these facts, we conclude that: (1) iodination of the thyroglobulin of type 2c and type 3 cells takes place almost entirely at the apical cell membrane region, (2) the thyroglobulin-like protein contained in the apical small vesicles of type 2c and type 3 cells is slightly iodinated, (3) although it is difficult to determine whether the dense granules and bodies, which might be lysosomes, are secretory substances or reabsorbed materials, the possibility of the occurrence of reabsorption and hydrolysis of the thyroglobulin in the type 2c and type 3 cells should be considered, and (4) reabsorption of the thyroglobulin from the endostylar lumen by some type 5 cells should be also considered.  相似文献

The effect of insulin (I), cortisol (F) and prolactin (P) on the ultrastructural morphology of epithelial cells of cultured mammary explants from virgin ovariectomized (OV-X) goats were studied. The epithelial cells showed little structural organization and were devoid of fat droplets and secretory protein granules at zero time of culture. The cytoplasm contained few profiles of smooth and rough endoplasmic reticulum and the Golgi apparatus was rudimentary. After being cultured in Waymouth's medium without added hormones the epithelial cells were indistinguishable from epithelial cells of uncultured explants. The addition of I induced changes mainly in the appearance of nucleoli. The nucleoli were enlarged and fibrillogranular areas with light spaces were observed. The most obvious cytological changes of epithelial cells of explants cultured in the presence of I and F are translocation of the nucleus into the basal cytoplasm, increase of rough endoplasmic reticulum, an increase in the size of the Golgi apparatus, presence of one or two lipid droplets and in some cells vacuoles with protein granules were present. Mitochondria were more abundant. The epithelial cells of explants cultured in the presence of I, F and P were characterized by the polarization of organelles within the cytoplasm and by the formation and release of protein granules and small and large fat droplets. The cell nucleus was in the basal cytoplasm, the Golgi apparatus was supranuclear. The rough endoplasmic reticulum was extensively developed and formed large sacs. Intracellular dense bodies, cytoplasmic matrices, and luminal colloids in the thyroid follicle of cyclostomes, hagfish and lamprey were examined to identify the distribution of iodine using an energy dispersive X-ray microanalyzer attached to a scanning transmission electron microscope. High level of iodine was detected only in the large dense body of the hagfishes, while it was too slight in quantity to detect by this method in the cytoplasmic matrix as well as in the luminal colloid. In the adult lamprey thyroid, an appreciable amount of iodine was detected in a few large dense bodies. In mice and rats, it is very hard to detect iodine in the luminal colloid, intracellular colloid droplet, and in the lysosomal dense granule by this method, though these structures have been well known to contain a fairly large amount of iodine which is combined with thyroglobulin. These facts mean that intracellular dense bodies in the thyroid follicular epithelium of the cytoclostome, especially of the hagfish have an extremely larger amount of iodine. These bodies are considered to be secondary lysosomes or residual bodies containing reabsorbed colloid materials which are highly condensed.  相似文献

Summary An electron microscopical study of the epithelium of the uterine tube was carried out in the newborn. Among the epithelial cells at least two morphologically well defined types can be distinguished: ciliated and non-ciliated cells.The ultrastructure of the cilia and related structures corresponds to what has been described by other authors in ciliated cells of various organs and of different species. Near the basal bodies of the cilia there is a concentration of vesicular mitochondria, which is thought to be evidence of a high metabolic activity in this region of the cell. Large opaque granules in the supranuclear zone of the ciliated cells are, it is suggested, paraplasmatic inclusions, perhaps supporting material for the ciliokinetic processes. There was no evidence of a secretory function of the ciliated cells.Among the non-ciliated cells, which in general show a straight lined luminal border with few microvilli, there are some cells containing dense granules, which are distributed throughout the cytoplasm and concentrated in the luminar side of the cell. The apical parts of these cells are protruding and sometimes digitated or branched; they contain accumulated granular materials and are separated from the rest of the cell after the formation of an intracellular plasmalemma. A similar detachment was found in an other cell type, but here the protruded apical parts of the cells are edematous and do not contain any visible secretory materials. It is uncertain if the detached cytoplasmic substances form a part of a specific secretory product; there are no secretory granules within the cytoplasm. On the contrary, the detachment of cytoplasmic parts may only accompany the excessive proliferation of cells which takes place during this period of growth.  相似文献

The ontogeny of an oviductal zona pellucida glycoprotein, designated ZP-0, in the golden hamster oviductal epithelium was investigated by means of light and electron microscopic immunolabeling studies using a monoclonal antibody. Light microscopic immunohistochemistry showed that ZP-0 appeared in the oviductal epithelium from 9.5 to 10.5 days after birth. At first, ZP-0 appeared in the Golgi region of the epithelial cells, and then increased in amount as development progressed to fill the supranuclear cytoplasm. Electron microscopic observations showed that rough endoplasmic reticulum and Golgi apparatus were well developed in non-ciliated cells of the ampullar and isthmic segments at 10.5 days after birth. Secretory granules appeared near the Golgi apparatus and markedly increased in number until 15.5 days after birth. Ultrastructural immunocytochemistry showed that ZP-0 appeared in the well-developed Golgi apparatus and then was stored within the secretory granules of developing secretory cells. ZP-0 was never present in the ciliated cells. These results indicate a coincidence between the beginning of ZP-0 biosynthesis and the morphological cytodifferentiation of secretory cells in the golden hamster oviduct during postnatal development. This leads us to the conclusion that ZP-0 is a possible differentiation marker of the golden hamster oviductal secretory cell.  相似文献

The intracellular location of basic fibroblast growth factor (bFGF) was determined in A431 human epidermoid carcinoma cells both on immunofluorescence and on immunoelectron microscopy using a monoclonal anti-bFGF antibody. The immunofluorescence was located in the cytoplasm in quiescent cells. Following the addition of FCS to the culture medium of quiescent sparse cells the growth factor was translocated to and accumulated in the nucleolus. Immunogold particles were dense near the ribosomes, but were not recognized in the cytoplasmic structures concerned with the usual secretory pathway such as the rough endoplasmic reticulum, the Golgi apparatus, and secretory granules. These results suggest that endogenous bFGF undergoes intracellular sorting and enters the nucleoli in A431 cells according to an extracellular growth signal.  相似文献

We investigated the oligosaccharide sequence of glycoconjugates, mainly sialoglycoconjugates, in the horse oviductal ampulla during oestrus by means of lectin and pre-lectin methods such as the KOH-neuraminidase procedure to remove sialic acid residues and incubation with N-glycosidase F to cleave N-linked glycans. Ciliated cells displayed N-linked oligosaccharides throughout the cytoplasm. The cilia glycocalyx expressed both N- and O-linked (mucin-type) oligosaccharides, both showing a high variety of terminal sequences. In the most non-ciliated cells, the whole cytoplasm contained N-linked oligosaccharides with terminal alphaGal as well as mucin-type glycans with terminal Forssman pentasaccharides. In a few scattered non-ciliated cells, the whole cytoplasm displayed sialylated N-linked oligosaccharides with terminal Neu5Ac-GalNAc and O-linked glycans terminating with neutral and/or alphaGalNAc, Neu5Ac alpha2,6Gal/GalNAc, Neu5AcGal beta1,3GalNAc. Supra-nuclear granules, probably Golgi zones, of non-ciliated cells showed mainly O-linked glycans rich in sialic acid residues. The luminal surface of non-ciliated cells showed N-linked oligosaccharides, containing terminal/internal alphaMan/alphaGlc, betaGlcNAc and terminal alphaGal, as well as mucin-type oligosaccharides terminating with a large variety of either neutral saccharides or sialylated sequences. Apical protrusions containing O-linked oligosaccharides with terminal Forssman pentasaccharide, Neu5Ac-Gal beta1,4GlcNAc, Neu5Ac-GalNAc were seen in non-ciliated cells scattered along the epithelium. These findings show the presence of sialoglycoconjugates in the oviductal ampulla epithelium of the mare and the existence of different lectin binding profiles between ciliated and non-ciliated (secretory) cells, as well as the presence of non-ciliated cell sub-types which might determine functional differences along the ampullary epithelium of mare oviduct.  相似文献

Summary The granulosa cells of the ovarian follicle of the rat and the domestic fowl have been studied with the light and electron microscope. The nuclei of the granulosa cells were irregular with indentations and large in proportion to the cytoplasm of the cell. The mitochondria had a dense, dark matrix with only few cristac. The Golgi apparatus was moderately developed, located towards the oocyte in a juxtanuclear position. The endoplasmic reticulum was rather sparse. Lipid droplets were only occasionally encountered. Microtubules were regularly observed. The functions of the granulosa cells are discussed. Compared with the steroid-producing cells of the theca interna of the same follicles, the granulosa cells primarily are the nursing cells for the growing oocyte and mainly have the characteristics of protein forming cells.  相似文献

Summary Nitrogen mustard N-oxide was tried for the fixation of tissue for electron microscopy. A fixative consisting of 1% nitrogen mustard N-oxide, 1% glutaraldehyde and 1% paraformaldehyde buffered at pH 7.4 followed by 1% OsO4 buffered at pH 7.4 was found useful for the tissues examined: thyroid, anterior pituitary, adrenal gland and oviduct of mice.If the tissues are fixed and the sections are stained with uranyl acetate and lead acetate doubly, the follicle colloid, colloid droplets, and secretory granules containing thyroglobulin in the thyroid become higher in electron density. The cisterna of the maturing face of the Golgi apparatus, secretory granules, ribosomes, nucleolus and chromatin in the cells examined are extremely electron dense. Tubular elements of smooth endoplasmic reticulum in the adrenal cortical cell and microtubules in all the cells examined are also well preserved. The fixative containing nitrogen mustard N-oxide is useful also for cytochemistry. Using tissue fixed by this method and stained en bloc by uranyl acetate, the noradrenaline and adrenaline cells in the adrenal medulla are clearly distinguished by light microscopy.This study was supported by a grant from the Japan Educational Ministry  相似文献

The epididymis of the bull was divided into six regions, and morphological differences between regions were studied. The epithelium of all regions contained four cell types: principal and basal epithelial cells, and intraepithelial lymphocytes and macrophages. The epithelium of regions II-V also contained a few apical cells. Principal cells of all regions possessed an endocytotic apparatus including stereocilia underlain by canaliculi, coated vesicles, and subapical vacuoles (up to 1 micron in diameter); however, large vacuoles with a flocculent content and multivesicular bodies (up to 5 microns in diameter) were most numerous in regions II, III, and IV. The unique features of principal cells of region I were the presence of well-developed Golgi bodies, few lipid droplets, and whorls of smooth endoplasmic reticulum in the supranuclear cytoplasm. Numerous mitochondria, distended cisternae of rough endoplasmic reticulum, and dense granules characterized the infranuclear cytoplasm of the principal cells of regions II-VI; however, these features were more developed in region V. Apical cells were characterized by the apical location of the nucleus, many mitochondria in the apical cytoplasm, and few microvilli at the luminal border. Basal cells with few cytoplasmic lipid droplets were present throughout the length of the epididymis but appeared more numerous in region V. Intraepithelial lymphocytes were present at all levels of the epithelium but were never seen in the lumen. Intraepithelial macrophages containing heterogeneous granules, eccentric nuclei, and pseudopods were invariably seen near the basal area of the epithelium in all regions. These observations are discussed in an effort to define the role of each cell type in the epididymal epithelium.  相似文献
