为了建立冬虫夏草中虫草酸含量的检测方法,比较冬虫夏草繁育品和野生冬虫夏草中的虫草酸含量,文中采用超声水提法研究了料液比和提取时间对虫草酸含量的影响,筛选出料液比为1∶50、提取时间为5 min时可高效提取冬虫夏草样品中的虫草酸。检测方法经方法学考察,专属性、重复性、准确度、稳定性均符合要求;21批冬虫夏草繁育品样本中的虫草酸含量为9.04%~14.14%;21批野生冬虫夏草虫草酸含量为6.16%~13.94%。通过t检验统计分析显示,冬虫夏草繁育品和野生冬虫夏草的虫草酸含量差异不显著,具有良好一致性。  相似文献

人工冬虫夏草的氨基酸含量及其营养价值评价   总被引:3,自引:0,他引:3  
目的为明确人工和野生冬虫夏草在氨基酸方面的差异,并为进一步开发利用人工冬虫夏草提供理论依据,对室内全人工培殖冬虫夏草和产自四川康定的野生冬虫夏草的氨基酸营养价值进行分析评价。方法用日立L-8800型全自动分析仪检测氨基酸含量并根据FAO/WHO的氨基酸评分标准模式和鸡蛋蛋白模式进行氨基酸营养价值评价。结果人工冬虫夏草氨基酸种类齐全,其全草氨基酸总量高于产自康定的野生冬虫夏草;其全草的鲜味氨基酸高于产自康定的野生冬虫夏草;其全草必需氨基酸总量低于产自康定的野生冬虫夏草。人工冬虫夏草必需氨基酸与氨基酸总量的比值高于WHO/FAO评分模式而低于鸡蛋蛋白模式。结论人工冬虫夏草氨基酸不仅种类组成合理,而且具有较高的营养价值。这一结果为人工冬虫夏草的进一步开发利用提供了理论基础。  相似文献

冬虫夏草具有珍贵的药用价值,是我国传统的稀有名贵中药材。由于生长环境受到限制,其数量较为稀少且因人为过度开采,冬虫夏草野生资源受到威胁,冬虫夏草资源的可持续性受到越来越多的关注。本文对近年冬虫夏草的人工培植技术、人工固体发酵培植培养技术、人工液态发酵培植培养技术及冬虫夏草发酵调控进行了综述。培养基成分、培养条件的优化及针对性地添加特定的外源物质均可提高冬虫夏草的产量及质量,这为冬虫夏草的人工培养提供了理论基础。  相似文献

对人工培养蛹虫草与青海产野生冬虫夏草的氨基酸含量进行测定,结果表明:野生冬虫夏草中所含有的各种氨基酸,在人工培养虫草的子实体及菌丝体中都有存在;且后者的精氨酸及脯氨酸含量明显高于野生冬虫夏草。由此为鉴定人工培养虫草的质量,进一步为工业规模开发与利用人工培养虫草,提供了科学理论依据。  相似文献

冬虫夏草菌是珍稀濒危名贵中药材冬虫夏草的无性型菌种,是侵染蝠蛾幼虫的唯一菌种,其侵染后形成名贵中药材冬虫夏草。冬虫夏草菌在不同营养及条件下生长形态不同,主要有丝状体和菌球体两种生长形态。冬虫夏草菌丝状菌体能够侵染蝠蛾幼虫,有可能作为一种新的接种体被广泛应用;但丝状菌体对营养要求苛刻、生长缓慢、菌丝体得率低的特点,阻碍了冬虫夏草菌丝状菌体作为侵染蝠蛾幼虫的接种体的开发及应用,从而阻碍了冬虫夏草人工培殖产业化的进程。为了提高冬虫夏草菌丝状菌体的生物表达量,本文对营养及培养条件进行了优化研究,得出了最佳条件为:葡萄糖质量浓度40 g/L、酵母粉质量浓度65 g/L、培养温度17℃、MgSO_4质量浓度3 g/L、KH_2PO_4质量浓度1.5 g/L、培养时间12 d,按最优培养条件,冬虫夏草菌丝状菌体干重得率15.67 g/L,为下一步新的接种体的制备提供菌源保障。  相似文献

本文测定了野生采集和人工培育冬虫夏草、人工培育冬虫夏草子实体、冬虫夏草发酵菌液、冬虫夏草寄主蝙蝠蛾昆虫食物(蕨麻Potentilla anserine和珠芽蓼Polygonum viviparum),以及野生冬虫夏草产地土壤中5种重金属元素(铜Cu、铅Pb、砷As、镉Cd、汞Hg)的含量。结果表明,人工培育冬虫夏草、人工培育冬虫夏草子实体、冬虫夏草发酵菌液、蕨麻块根、珠芽蓼种子的5种重金属元素含量符合《中医药-中药材重金属限量》ISO国际标准;而野生冬虫夏草,除砷含量(6.170 mg/kg)超标外,铜、铅、镉、汞元素含量均符合要求。按国家标准《土壤环境质量农用地土壤污染风险管控标准(试行)》(GB 1516-2018),四川产地土壤(KDX)与青海产地土壤(GD、HN)的5种重金属含量均低于农用土壤污染风险筛选值;而四川产地土壤(KDT),除镉含量(0.923 mg/kg)高于风险筛选值而低于风险管制值外,其余4种元素含量均低于风险筛选值。结果为人工培育冬虫夏草的质量控制提供了参考。  相似文献

冬虫夏草寄主昆虫选育及生殖退化研究   总被引:1,自引:0,他引:1  
选育发育速度快、繁殖能力强、中国被毛孢侵染率和冬虫夏草产出率高的寄主昆虫品系是冬虫夏草培植的基础,同时明确冬虫夏草的寄主昆虫蝙蝠蛾退化机制是保证冬虫夏草培植的条件。本文从冬虫夏草产区采集蝙蝠蛾种群种质资源,分别针对蝙蝠蛾发育速度、繁殖能力、中国被毛孢侵染率、冬虫夏草产出率4个方面通过杂交及四代选育,获得新种群(A1),该种群对冬虫夏草繁育环境的适应力强,发育整齐度高,繁育一代的种群扩繁倍数从8.2倍提高至26.3倍;对中国被毛孢的敏感性较好,侵染率高,冬虫夏草产出率从原始种群的11.2%提高至18.5%。同时,仿照高原生态环境长期饲养冬虫夏草的寄主昆虫蝙蝠蛾,其子代出现雌雄比失调、发育历期分化、生殖力降低等退化现象。经过研究分析得知卵孵化率下降的主要原因是雄性的生殖力下降。组织切片观察蝙蝠蛾雄性生殖系统,在幼虫后期、蛹、成虫期会出现生精囊减少及畸形等异常情况,推测此为雄性生殖力降低、卵孵化率下降的生理原因。研究结果为高效、稳定地饲养蝙蝠蛾保证冬虫夏草培植具有重要意义。  相似文献

本文对近年来冬虫夏草的培植,包括菌种分离培养、寄主饲养、侵染机理、接种方法及子实体发育条件等研究进展进行了综述。本研究组成功实现了冬虫夏草培植产业化,所培植的冬虫夏草与野生冬虫夏草在虫种、菌种、外观形态、显微结构和化学成分等方面一致。最后分析了冬虫夏草培植技术现阶段存在的问题,并对未来进行了展望。  相似文献

利用油混悬剂独有的剂型特性可提高人工培育冬虫夏草中的接种侵染率, 因而研制高活力冬虫夏草菌种油混悬剂可推动其产业化的发展。根据油混悬剂的特殊性, 对MTT法进行改进及优化, 结果为: 反应时间120 min、反应温度37°C、以甲苯为萃取剂, 活细胞量与琥珀酸脱氢酶活力线性相关。依据该方法筛选制剂中助剂的种类及浓度, 并通过正交试验进一步优化, 获得冬虫夏草菌种油混悬剂高活力配方为: 菌种量0.15 g/mL、硬脂酸铝60 mg/mL、司班-80 50 μL/mL。稳定性试验结果表明, 该制剂在4°C下保  相似文献

Ophiocordyceps sinensis is one of the best known fungi in Traditional Chinese Medicine. Many efforts have been devoted to locating the production areas of this species resulting in various reports; however, its geographic distribution remains incompletely understood. Distribution of O. sinensis at the county level is clarified in this work based on both a literature search and fieldwork. More than 3600 publications related to O. sinensis were investigated, including scientific papers, books, and online information. Herbarium specimens of O. sinensis and field collections made by this research group during the years 2000-2010 were examined to verify the distribution sites. A total of 203 localities for O. sinensis have been found, of which 106 are considered as confirmed distribution sites, 65 as possible distribution sites, 29 as excluded distribution sites and three as suspicious distribution sites. The results show that O. sinensis is confined to the Tibetan Plateau and its surrounding regions, including Tibet, Gansu, Qinghai, Sichuan, and Yunnan provinces in China and in certain areas of the southern flank of the Himalayas, in the countries of Bhutan, India and Nepal, with 3,000 m as the lowest altitude for the distribution. The fungus is distributed from the southernmost site in Yulong Naxi Autonomous County in northwestern Yunnan Province to the northernmost site in the Qilian Mountains in Qilian County, Qinghai Province, and from the east edge of the Tibetan Plateau in Wudu County, Gansu Province to the westernmost site in Uttarakhand, India. The clarification of the geographic distribution of O. sinensis will lay the foundation for conservation and sustainable use of the species.  相似文献

中国被毛孢(Hirsutella sinensis)是天然冬虫夏草[Cordyceps sinensis(Berk)Sacc.]的真正无性型,其生长温度低(低于20℃),生长缓慢(发酵周期长达20~30d),对人工发酵的技术条件要求很高,因而大大增加了工业化发酵生产的成本和技术难度。为了解决上述问题,实验中从天然冬虫夏草子座中分离到中国被毛孢菌株BS-1,并对其进行了原生质体紫外诱变处理。经过粗筛和精筛,从中选出一株可常温下(25~28℃)发酵、生长迅速(发酵周期4~5 d)、有效成分(甘露醇和腺苷)含量显著高于原始菌株的诱变株BS-5962。通过连续10代PDA斜面继代培养和摇瓶试验,以及连续3批的1t罐的中试试验,检查结果表明所得诱变株性状优异而稳定。可为降低中国被毛孢的液体深层发酵成本和技术难度提供一种有效的解决方案。  相似文献

The entomopathogenic fungus Ophiocordyceps sinensis has been important in traditional Chinese medicine but has yet to be commercially cultivated. One bottleneck is the very low frequency of stromata formation from artificially infected moth larvae. The mating system of fungi is the determining factor for sexual reproduction, but mating-type genes of O. sinensis have not been previously investigated. In this study, the putative mating-type gene MAT1-2-1 within the MAT1-2 idiomorph was amplified by polymerase chain reaction (PCR) and was determined to consist of 859 nucleotides that encode 249 amino acids; genes within the MAT1-1 idiomorph, however, were not determined. The MAT1-2-1 gene contained the conserved high-mobility group (HMG) box, and MAT1-2-1 flanking sequences were subsequently obtained. Although no putative open reading frames of the MAT1-1 idiomorph were detected within the ca. 8-kb flanking sequences of MAT1-2-1, a putative DNA lyase gene (which is present next to both idiomorphs in some heterothallic fungi) was found ca. 3.0 kb downstream of MAT1-2-1. The intervening distance between MAT1-2-1 and the DNA lyase gene in O. sinensis is larger than that in Cordyceps militaris and Cordyceps takaomontana. In addition, O. sinensis showed low sequence similarities with C. militaris and C. takaomontana in both MAT1-2-1 and the DNA lyase gene. In the phylogenetic tree, different MAT1-2-1 haplotypes of O. sinensis clustered together with high bootstrap support. As a single-copy gene, MAT1-2-1 was detected in all examined O. sinensis isolates including tissue cultures and single-ascospore cultures. This report describes, for the first time, a mating-type gene of O. sinensis.  相似文献

AIMS: The present study comparatively investigates the optimal culture conditions for the production of exopolysaccharides (EPS) and cordycepin during submerged mycelial culture of two entomopathogenic fungi Cordyceps militaris and Cordyceps sinensis. METHODS AND RESULTS: Fermentations were performed in flasks and in 5-l stirred-tank fermenters. In the case of C. militaris, the highest mycelial biomass (22.9 g l(-1)) and EPS production (5 g l(-1)) were achieved in a medium of 40 g l(-1) sucrose, 5 g l(-1) corn steep powder at 30 degrees C, and an initial pH 8.0. The optimum culture conditions for C. sinensis was shown to be (in g l(-1)) 20 sucrose, 25 corn steep powder, 0.78 CaCl2, 1.73 MgSO4.7H2O at 20 degrees C, and an initial pH 4.0, where the maximum mycelial biomass and EPS were 20.9 and 4.1 g l(-1) respectively. Cordycepin, another bioactive metabolite, was excreted at low levels during the early fermentation period (maximum 38.8 mg l(-1) in C. militaris; 18.2 mg l(-1) in C. sinensis). CONCLUSIONS: The two fungi showed different nutritional and environmental requirements in their submerged cultures. Overall, the concentrations of mycelial biomass, EPS and cordycepin achieved in submerged culture of C. militaris were higher than those of C. sinensis. SIGNIFICANCE AND IMPACT OF THE STUDY: C. militaris and C. sinensis are representative insect-born fungi which have been longstanding and widely used as traditional medicines in eastern Asia. Comparative studies between two fungi are currently not available and this is the first report on the optimum medium composition for submerged culture of C. sinensis.  相似文献

中华卵索线虫Ovomermis sinensis感染棉铃虫Helicoverpa armigera 1天后,棉铃虫幼虫血淋巴中游离氨基酸总量大幅度下降,各种游离氨基酸的含量也是如此变化。感染2~4天后的棉铃虫血淋巴中游离氨基酸总量变化不大,各种游离氨基酸的含量有升有降。感染5~6天后的棉铃虫血淋巴中游离氨基酸总量和各种氨基酸的含量都急剧上升。研究表明,中华卵索线虫寄生棉铃虫时,棉铃虫血淋巴中游离氨基酸含量的变化朝着有利于线虫生长发育的方向进行。  相似文献

通过CTAB法从冬虫夏草菌株和天然冬虫夏草不同部位提取DNA,并用PCR扩增进行验证,证明了CTAB法适合从冬虫夏草子座、菌核和冬虫夏草菌培养物中提取DNA。首次报道1种将子囊孢子破壁直接进行PCR扩增的方法,并比较了该方法和CTAB法在提取冬虫夏草DNA方面的差异。2种方法获得的DNA用于PCR扩增均能得到较好的结果。  相似文献

冬虫夏草作为青藏高原特有的生物资源,对生长环境有着极其特殊的要求.通过调查青海省冬虫夏草资源分布状况,选取玉树、果洛、黄南、海南等地区的冬虫夏草进行氨基酸、微量元素含量检测,以反映冬虫夏草资源特征.研究结果表明,青海省冬虫夏草含有丰富的氨基酸和微量元素,且冬虫夏草不同部位的氨基酸、微量元素含量有明显差别.将青海省冬虫夏草与其他产区的冬虫夏草氨基酸含量进行对比分析,青海省冬虫夏草氨基酸含量低于四川、云南产区的冬虫夏草,和西藏产区的冬虫夏草氨基酸含量基本一致.  相似文献
