Interspecific hybridizations were carried out between the two tetraploidsElymus caucasicus andE. longearistatus, and 23 tetraploids and hexaploids ofElymus containing SH, SY, SYH, and SYW genomes and representing various geographical regions. Meiotic pairing was studied in the two target species and their hybrids. It is concluded from this study that (i) interspecific hybridization is fairly easy to perform although strong reproductive barriers exist between the species; (ii)Elymus caucasicus andE. longearistatus are allotetraploids, and share the diverged SY genomes; (iii) the divergence of SY genomes is correlated with the geographic distance between theElymus spp. studied.  相似文献

Post-embedding immunocytochemical techniques using peroxidase–antiperoxidase as markers for light microscopy or immunoglobulin G-gold for electron microscopy respectively were used for the localization of cytokinins [9--D-ribofuranosil-N 6-(2-isopentenil) adenina ([9R]iP), 9--D-ribofuranosyl-zeatin ([9R]Z) and 9--D-ribofuranosyl-dihydrozeatin ([9R](diH)Z)] in kiwifruit (Actinidia deliciosa) meristematic cells of the second nodal segment. Immunolocation at the cellular level was carried out in cells from explants grown during 16 and 72h in liquid medium. Subcellular immuno-localization was performed in cells from explants grown for 35d on agar solidified-medium and for 30min, 4 and 16h in liquid medium with cellulose plugs as explant support. Taken as a whole, the results obtained for Actinidia deliciosa show that the studied cytokinins change their location during the culture period, although they can always be found to a greater or lesser extent in the nucleus and the cytoplasm. For instance, [9R]Z appears in the cytoplasm and in the nucleus during the first hours of culture and later is the only one that appears located mainly in nucleus. On the other hand, [9R](diH)Z changes from being predominantly located in the nucleus to practically appearing only in the cytoplasm at the end of the culture period. [9R]iP is principally found up to 4h of culture in the cytoplasm, and at 16h is evenly distributed in all the subcellular compartments except in the chloroplast. The existence of a large amount of cytokinins in the nucleus during the first hours of culture compared with the immunolabelling density at 35d is probably due to the activation of cell cycle mechanisms leading to organogenic development at the beginning of culture.  相似文献

Summary Activation kinetics of single high-threshold inactivating (HTI orN-type) calcium channels of cultured dorsal root ganglion cells from mouse embryos was studied using a patchclamp method. Calcium channels displayed bursting activity. The open-time histogram was single exponential with an almost potential-independent mean open time op. The closed-time histogram was multicomponent; at least three of the components were associated with the activation process. The fast exponential component with the potential-independent time constant cl f included all intraburst gaps, while two slower ones with potential-dependent time constants cl vs described shut times between bursts and between clusters of bursts. The burst length histogram was biexponential. The fast component with a relatively potential-independent time constant bur f described short, isolated channel openings while the slow component characterized real bursts with a potential-dependent mean life time. The waiting-time histogram could be fitted by a difference of two exponentials with time constants being the same as cl s and cl vs . The data obtained were described in the frame of a 4-state sequential model of calcium channel activation, in which the first two stages are formally attributed to potential-dependent transmembrane transfer of two charged gating particles accompanying the channel transitions between three closed states, and the third one to fast conformational changes in channel protein leading to the opening of the channel. The rate constants for all transitions were defined. The validity of the proposed model for both low-threshold inactivating (LTI orT-type) and high-threshold noninactivating (HTN orL-type) calcium channels is discussed.  相似文献

Zusammenfassung Im Nucleolus der Leberzellen von Ratten ist elektronenmikroskopisch — mit Formalinfixierung — kontrastreiches Chromatin und autoradiographisch — mit H3-Thymidin — eine DNS-Synthese nachweisbar. Das Chromatin ist nicht in die netzigen Anteile der Nucleolarsubstanz (Nucleolonema), sondern in die dazwischen liegenden Aufhellungen eingelassen, doch sind nur einige dieser Räume und auch diese oft nur unvollständig von Karyoplasma (= Chromosomensubstanz) ausgefüllt. Die einzelnen Chromatin-Einschlüsse erreichen normalerweise die lichtmikroskopischesichtbarkeitsgrenzenicht. Lediglich in hepatozellulärenTumornucleolen sind bereits in gewöhnlichen Präparaten feulgenpositive Strukturen festzustellen.Aus den Befunden wird gefolgert: der Nucleolus der Somazellen ist generell von — meist sublichtmikroskopischen — Anteilen aufgelockerter und aufgesplitterter Chromosomen durchzogen. Dabei handelt es sich um die nucleolusorganizer-Region der Nucleolarchromosomen, denen der organisierte Nucleolus auch in der Intermitose verhaftet bleibt.  相似文献

The time constant of the process producing the delay in Na inactivation development as determined by the two pulse method (delay) was extracted and compared to that of the slowest Na activation process 3 for the I Na during the conditioning pulse of that same determination. delay and two pulse inactivation c values were computer generated using a nonlinear least squares algorithm. h and single pulse inactivation h values were independently generated for each determination also with the aid of the computer using the same non-linear least squares algorithm. In one determination at 2 mV, c was 4.68 and delay 0.494 ms while h was 4.70 and 3 0.491 ms for a c/h of 0.996 and a delay/3 of 1.006. Mean delay/3 from five determinations in four axons, both Cs and K perfused, and spanning a potential range of-27 to 2mV was 1.068. The precursor process to inactivation is channel opening. Some fraction of channels presumably inactivate via another route where prior channel opening is not required.  相似文献

Summary A fungus identified as Cunninghamella blakesleeana (Lendner) can carry out 15-hydroxylation of lithocholic acid to a new bile acid (3,15-dihydroxy-5-cholanic acid). By optimizing the fermentation conditions, the amount of the product increased from 0.17 g/l to 1.2 g/l. Three classes of inner mitochondrial membrane (IMM) channel activities have been defined by direct measurement of conductance levels in membranes with patch clamp techniques in 150 mM K Cl. The 107 pS activity is slightly anion selective and voltage dependent (open with matrix positive potentials). Multiple conductance channel (MCC) activity includes several levels from about 40 to over 1000 pS and can be activated by voltage or Ca2+. MCC may be responsible for the Ca2+-induced permeability transition observed with mitochondrial suspensions. A low conductance channel (LCC) is activated by alkaline pH and inhibited by Mg2+. LCC has a unit conductance of about 15 pS and may correspond to the inner membrane anion channel, IMAC, which was proposed from results obtained from suspension studies. All of the IMM channels defined thus far appear to be highly regulated and have a low open probability under physiological conditions. A summary of what is known about IMM channel regulation and pharmacology is presented and possible physiological roles of these channels are discussed.  相似文献

Plantlet formation through organogenesis in callus cultures of Himalayan yellow poppy,Meconopsis paniculata D.Don (Prain), a threatened taxon of ornamental value, is described. Hypocotyl segments from 3-month-old laboratory-raised seedlings produced callus on agar-solidified Murashige and Skoog medium (MS) containing 10 M -naphthaleneacetic acid and 1 M kinetin. Shoots differentiated best from callus on MS containing 10 M indolebutyric acid (IBA) and 1 M 6-benzyladenine. The regenerated shoots rooted best on MS medium containing 10 M IBA. Over the last seven years, solid-state NMR has been widely employed to study structural and functional aspects of the nicotinic acetylcholine receptor. These studies have provided detailed structural information relating to both the ligand binding site and the transmembrane domain of the receptor. Studies of the ligand binding domain have elucidated the nature and the orientation of the pharmacophores responsible for the binding of the agonist acetylcholine within the agonist binding site. Analyses of small transmembrane fragments derived from the nicotinic acetylcholine receptor have also revealed the secondary structure and the orientation of these transmembrane domains. These experiments have expanded our understanding of the channels structural properties and are providing an insight into how they might be modulated by the surrounding lipid environment. On the basis of symposium contributions onChlorella, Hibbertia, Eucalyptus, Ambrosia and on numerical approaches some fundamental problems of (bio)systematics, evolution, and taxonomic categories are discussed: Methods available for analysing affinities; conflicting evidence from phenetic, biochemical, cytogenetic and other analyses; further classification problems in cases of intermediacy, etc. While sibs of various levels and their natural hierarchy often can be objectively defined, this appears impossible for particular taxonomic levels itself (e. g. species). A single objective taxonomic system of organisms is unrealistic. The behavior of individual allocyclic chromosomes has been analyzed in lymphocytes of a sister and a brother with Bloom's syndrome. Of 4,633 diploid cells, 115 showed allocyclic chromosomes, and 74 of these had 44, 45 or 46 normal metaphase chromosomes accompanied by one or two allocyclic chromosomes. Of 56 tetraploid cells, 9 contained such chromosomes. The allocyclic chromosomes appeared pulverized or extended corresponding to S or G2 PCC. We have proposed the hypothesis that individual allocyclic chromosomes do not, as a rule, come from micronuclei, as has often been assumed, but have been left behind in their cycle. This would be caused by a mutation or deletion of a hypothetical coiling center situated near the centromere of each chromosome arm. The following observations agree with our explanation but less well or not at all with the idea of micronuclei: (1) In only 9.6% of the cells does the allocyclic chromosome lie at the edge of the metaphase plate. (2) In 24 cells a part of a chromosome is pulverized while the rest is in metaphase. (3) Both a pulverized and an extended chromosome were present in the same cell. (4) A pulverized acrocentric is often nose-to-nose with a normal D or G chromosome. (5) No allocyclic chromosomes corresponding to G1 PCC have been found in our material. (6) When a ring is replaced by an allocyclic chromosome, it is usually a member of a 46-chromosome complement. Furthermore, the occurrence of allocyclic chromosomes is correlated with that of other chromosome anomalies which do not follow a Poisson distribution. Allocyclic chromosomes are also more frequent (16%) in tetraploid than in diploid cells (2%).  相似文献

Summary Serratia strain CV produces spontaneously about 0,5% auxotrophic mutants of different types. By means of a special auxanographic technique, among about 60 monoauxotrophs, 5 were found which frequently and spontaneously mutate to a certain other auxotrophic state loosing the first one (arg 1 nia ;arg 2 his ,ad, hyx ;thr met ;prol his ;leu ad, hyx ). It could be shown that this auxotrophic leap happens in ohne step without a prototrophic intermediate step. By backmutation experiments with the mutationsleu ad it was shown that the second auxotrophy acts as a suppressor mutation to the first auxotrophy. Since the biochemical reaction sequence blocked by the first mutation seems not to be related to the chain blocked by the second mutation, and since the second mutants do not feed the first mutants syntrophically, it is not probable that the suppression of the first auxotrophy is caused by a substance accumulated as an effect of the second mutation, removing the first auxotrophy by intracellular feeding. Rather, the second auxotrophy-gene (in its mutated, suppressing allelic state, e. g.ad ) seems to contribute information to the production of the enzyme determined usually by the first auxotrophy-gene, which information was lost by the first mutation (e. g. leu ). Thus, the information on the specificity of an enzyme would be stored not only in one single gene but in several genes.  相似文献

Alzheimer's disease is the most prevalent form of dementia worldwide. Therapies are desperately needed to prevent and cure the disease. Mouse models of amyloid- deposition [APP and PSAPP transgenic (tg) mice] have been useful in determining the role of amyloid- (A) in both the pathogenesis and cognitive changes in AD. In addition, they have allowed scientists to investigate potential AD therapies in living animals. Active and passive A immunizations have been employed successfully in APP and PSAPP tg mice to lower cerebral A levels and improve cognition. Optimization of immunization protocols and characterization of immune responses in wildtype mice have been reported. Based on the promising results of A immunization studies in mice, a clinical trial was initiated for A vaccination in humans with AD. Although no adverse effects were reported in the Phase I safety trials, about 5% of AD patients in the phase II clinical trial developed meningoencephalitis, ending the trial prematurely in March 2002. Studies in AD mouse models and wildtype mice may help elucidate the mechanism for these unwanted side effects and will be useful for testing newer, safer vaccines for future use in human clinical trials.  相似文献

,-Dehydroamino acids are useful peptide modifiers. However, their stereoelectronic properties still remain insufficiently recognized. Based on FTIR experiments in the range of s(N-H), AI, AII and s(C=C) and ab initio calculations with B3LYP/6-31G*, we studied the solution conformational preferences and the amide electron density perturbation of Ac-Xaa-NHMe, where Xaa = Ala, (E)-Abu, (Z)-Abu, (Z)-Leu, (Z)-Phe and Val. Each of these dehydroamides adopts a C5 structure, which in Ac-Ala-NHMe is fully extended and accompanied by the strong C5 hydrogen bond. Interaction with bond C=C lessens the amidic resonance within the flanking amide groups. The N-terminal C=O bond is noticeably shorter, both amide bonds are longer than the corresponding bonds in the saturated entities and the N-terminal amide system is distorted. Ac-Ala-NHMe constitutes an exception. Its C-terminal amide bond is shorter than the standard one and both amide systems are ideally planar. Ac-(E)-Abu-NHMe shares stereoelectronic features with both Ac-Ala-NHMe and (Z)-dehydroamides.  相似文献

Summary Sarcoplasmic reticulum (SR) vesicles from frog leg muscle were fused with a planar phospholipid bilayer by a method described previously for rabbit SR. As a result of the fusion, K+-selective conduction channels are inserted into the bilayer. Unlike the two-state rabbit channel, the frog channel displays three states: a nonconducting (closed) state and two conducting states and . In 0.1m K+ the single-channel conductances are 50 and 150 pS for and , respectively. The probabilities of appearearance of the three states are voltage-dependent, and transitions between the closed and states proceed through the state. Both open states follow a quantitatively identical selectivity sequence in channel conductance: K+>NH 4 + >Rb+>Na+>Li+>Cs+. Both open states are blocked by Cs+ asymmetrically in a voltage-dependent manner. The zero-voltage dissociation constant for blocking is the same for both open states, but the voltage-dependences of the Cs+ block for the two states differ in a way suggesting that the Cs+ blocking site is located more deeply inside the membrane in the than in the state.  相似文献

Physiological tests, redetermination of G+C values with HPLC and DNA-DNA hybridization were used to determine the taxonomic affiliation of Spirillum 5175. This facultatively sulfur-reducing bacterium was compared to the type strains of the phenotypically most similar species Wolinella succinogenes and Campylobacter sputorum biovar bubulus. In addition to morphology, the following physiological properties were in common: use of elemental sulfur, nitrate, nitrite, aspartate, fumarate or malate as electron acceptor for growth with hydrogen or formate under anoxic conditions; microaerobic growth with 2% (v/v) oxygen. The G+C content of Wolinella succinogenes (51.8 mol%) and Campylobacter sputorum biovar bubulus (30.4 mol%) differs about 10 mol% from the G+C content of Spirillum 5175 (40.6 mol%). No significant DNA homology could be detected between the three strains. These differences excluded affiliation of Spirillum 5175 with the genera Wolinella or Campylobacter despite phenotypic similarities. On the basis of our results and DNA-rRNA hybridization studies by other authors, we established the new genus Sulfurospirillum for the freeliving Campylobacter-like bacteria Spirillum 5175 and Campylobacter spec. DSM 806. Strain Spirillum 5175 is described as the type strain of the new genus and species Sulfurospirillum deleyianum.Dedicated to R. S. Wolfe on the occasion of his 70th birthday  相似文献

This is the first report on -tubulin and microtubule arrays during microsporogenesis in a gymnosperm. Meiosis in Ginkgo biloba is polyplastidic, as is typical of the spermatophyte clade, and microtubule arrays are organized at various sites during meiosis and cytokinesis. In early prophase, a cluster of -tubulin globules occurs in the central cytoplasm adjacent to the off-center nucleus. These globules diminish in size and spread over the surface of the nucleus. A system of microtubules focused on the -tubulin forms a reticulate pattern in the cytoplasm. As the nucleus migrates to the center of the microsporocyte, -tubulin becomes concentrated at several sites adjacent to the nuclear envelope. Microtubules organized at these foci of -tubulin give rise to a multipolar prophase spindle. By metaphase I, the spindle has matured into a distinctly bipolar structure with pointed poles. In both first and second meiosis, -tubulin becomes distributed throughout the metaphase spindles, but becomes distinctly polar again in anaphase. In telophase I, -tubulin moves from polar regions to the proximal surface of chromosome groups/nuclei where interzonal microtubules are organized. No cell wall is deposited and the interzonal microtubules embrace a plate of organelles between the two nuclear cytoplasmic domains (NCDs) of the dyad. Following second meiosis, phragmoplasts that form between sister and non-sister nuclei fuse to form a complex six-sided structure that directs simultaneous cytokinesis. -Tubulin becomes associated with nuclei after both meiotic divisions and is especially conspicuous in the distal hemisphere of each young microspore where an unusual encircling system of cortical microtubules develops.  相似文献

Little information is available concerning the efficacy of chelates applied to biosolids (sewage-sludge)-treated soil for heavy-metal removal. The purpose of the experiment was to determine the availability to sunflower (Helianthus annuus L.) and hybrid poplar (Populus deltoides Marsh. × P. nigra L.) seedlings, of non-essential (Cd, Ni, Pb) and essential heavy metals (Cu, Fe, Mn, Zn) in field soil injected with biosolids since 1976 and treated with ethylenediamine-tetraacetic acid (EDTA) in 2001. Sunflower was grown at two densities, 20000 and 60000 plants/ha, and poplar at 10000 plants/ha. The tetrasodium salt of EDTA was applied at rates of 0, 0.5, 1, and 2 g EDTA salt per kg surface (25-cm depth) soil. The EDTA did not affect uptake by poplar of the three non-essential (Cd, Ni, Pb) and four essential (Cu, Fe, Mn, Zn) heavy metals. For sunflower, the 1.0 g/kg rate of chelate addition resulted in maximal removal of the three non-essential heavy metals (Cd, Ni, Pb). Uptake of the essential heavy metals by sunflower was little affected by the EDTA. At the 20000 plants/ha density, leaves of sunflower grown with 1.0 g EDTA Na42H2O per kg soil accumulated more Cd, Ni, and Pb than leaves of sunflower grown without the EDTA salt. At this density, concentrations of Cd in leaves of sunflower without EDTA and with 1.0 g/kg EDTA salt were 2.2 and 6.5 g/g, respectively; for Ni, they were 6.7 and 19.2 g/g, respectively; and for Pb, they were 15.6 and 46.9 g/g, respectively. At the 60000 plants/ha density, stems of sunflower grown with 1.0 g EDTA Na42H2O per kg soil accumulated more Cd, Ni, and Pb than stems of sunflower grown without the EDTA salt. At this density, concentrations of Cd in stems of sunflower without EDTA and with 1.0 g/kg EDTA salt were 0.6 and 4.6 g/g, respectively; for Ni, they were 1.7 and 17.6 g/g, respectively; and for Pb, they were 5.2 and 42.8 g/g, respectively. Removal of the non-essential heavy metals by sunflower was greater at the higher plant density (60000 plants/ha) compared to the lower one (20000 plants/ha).  相似文献

Kinks or bends introduced in peptides and proteins by helical distorter residues such as proline, other imino acids and glycine, especially when these are in close proximity in the sequence, are increasingly recognized as playing an essential role in the gating of channel-forming peptides as well as of physiological ion channels. Peptaibols are useful simple models for the much more complex biological ion channels, especially voltage-gated ones. In this short review, we compare the monomeric structures of three selected peptaibols (alamethicin, trichotoxin and antiamoebin) that widely differ with regards their near-central kink angles and dipolar moment orientations. Arachidonic acid has been shown to activate K+-selective, mechanosensitive ion channels in cardiac, neuronal and smooth muscle cells. Since the cardiac G protein (G K )-gated, muscarinic K+ (KACh) channel can also be activated by arachidonic acid, we investigated whether the KACh channel was also sensitive to membrane stretch. In the absence of acetylcholine (ACh), KACh channels were not active, and negative pressure failed to activate these channels. With ACh (10 m) in the pipette, applying negative pressure (0 to –80 mm Hg) to the membrane caused a reversible, pressure-dependent increase in channel activity in cell-attached and inside-out patches (100 m GTP in bath). Membrane stretch did not alter the sensitivity of the KACh channel to GTP. When G K was maximally activated with 100 m GTPS in inside-out patches, the KACh channel activity could be further increased by negative pressure. Trypsin (0.5 mg/ ml) applied to the membrane caused activation of the KACh channel in the absence of ACh and GTP; KACh channel activity was further increased by stretch. These results indicate that the atrial muscarinic K+ channels are modulated by stretch independently of receptor/G protein, probably via a direct effect on the channel protein/ lipid bilayer.  相似文献
