At six sites in central Germany consequences of SO2, NOX and O3 deposition and of acid precipitation on canopy throughfall of sulphate, nitrate, ammonium, organic acids and of metal cations from Norway spruce crowns were investigated in the field. Measured canopy throughfall rates (mmol ion kg-1 needle dw a-1 are separated in (i) background ion throughfall rates in clean air and (ii) trace gas-(or acid interception)-dependent throughfall rates at ambient trace gas concentrations. Based on synchronously measured pollution, precipitation and canopy throughfall data, statistical response functions are given, which allow the separate estimation of annual rates of sulphur and nitrogen deposition into spruce canopies if only annual means of SO2 or NO2 concentrations in air are known. The specific SO2 deposition rate of (0.841±0.214) mmol S kg-1 needle dw a-1 (nPa SO2 Pa-1)-1 is 2.3 times higher than the specific stomatal SO2 uptake. The NO2-dependent nitrogen deposition of (2.464±0.707) mmol N kg-1 needle dw a-1 (nPa NO2 Pa-1)-1 is 2.2 times higher than the specific stomatal NOX (NO2+NO) uptake. These ratios (2.32.2) are explained by the percentage of annual hours with open needle stomata. The shape of observed epicuticular SO2 and NOX deposition curves and of stomatal SO2 and NOX uptake curves are congruent. As for stomatal NOX uptake, there is an apparent compensation point at (5 to 8) nPa NO2 Pa-1. There is significant SO2-dependent canopy throughfall of Ca>K>Al>Mg>Fe in this order of relative importance. NOX deposition in spruce canopies reduces K+ throughfall and it weakly promotes throughfall of Mn2+ and Zn2+. There was no significant codeposition of sulphate and ammonium and no ion exchange of intercepted H3O+ with nutrient cations at the measured ambient pH values of the precipitation water. In the presence of O3, throughfall of Mn2+ is reduced and throughfall of K+, Ca2+ and Al3+ is enhanced. In the cooperative presence of SO2, NO2 and O3 pollution in the field there is a 1.3-fold increase of the annual K+ demand and a 1.5-fold Mg2+ demand of spruce canopies relative to the situation in clean air. This trace gas-dependent additional cation demand of spruce canopies corresponds to a needle loss percentage of (23 to 33)% if the additional K+ and Mg2+ throughfall could not be recycled in spruce ecosystems. The emission of reduced volatile sulfur compounds from twigs of Norway spruce (Picea abies (L.) Karst.) was measured in the field by cryosampling and gaschromatographic analysis. Trees were growing in the Erzgebirge (E-Germany) at Oberbärenburg and at the Kahleberg and at a third stand in NW-Bavaria (S-Germany). Emission rates were also measured for Scotch pine (Pinus sylvestris L.) and Blue spruce (Picea pungens Engelm.) at the Kahleberg. Twigs still attached to the trees were enclosed in a flow-through gas exchange cuvette. H2S was detected as the predominant reduced sulfur compound emitted from the twigs. The mean H2S emission rate from twigs of Norway spruce varied between 0.04 pmol kg-1 dw s-1 at Würzburg and 6.21 pmol kg-1 dw s-1 at the Kahleberg. Comparing different species at the Kahleberg, the mean H2S emission rate was almost the same from twigs of Norway spruce (6.2 pmol kg-1 dw s-1) and Blue Spruce trees (5.9 pmol kg-1 dw s-1) but it was approximately 18 times higher for Scotch pine (110 pmol kg-1 dw s-1). The percentage of SO2-exclusion via H2S-emission of the tree species investigated at the Kahleberg is calculated on the basis of data on SO2 fluxes. It is very small for Norway spruce and Blue spruce. However, for Scotch pine, H2S emission contributes about 10% to the detoxification of SO2.  相似文献

Concentrations of ions were measured in soil solutions from beech (Nothofagus) forests in remote areas of New Zealand and in solutions from beech (Fagus sylvatica) and Norway spruce (Picea abies) forests in North-East Bavaria, West Germany, to compare the chemistry of soil solutions which are unaffected by acid deposition (New Zealand) with those that are affected (West Germany). In New Zealand, soil solution SO4 2– concentrations ranged between <2 and 58 mol L–1, and NO3 concentrations ranged between <1 and 3 mol L–1. In West Germany, SO4 2– concentrations ranged between 80 and 700 mol L–1, and NO3 concentrations at three of six sites ranged between 39 and 3750 mol L–1, but was not detected at the remaining three sites. At all sites in New Zealand, and at sites where the soil base status was moderately high in West Germany, pH levels increased, and total Al (Alt) and inorganic monomeric Al (Ali) levels decreased rapidly with increasing soil depth. In contrast, at sites on soils of low base status in West Germany, pH levels increased only slightly, and Al levels did not decline with increasing soil depth.Under a high-elevation Norway spruce stand showing severe Mg deficiency and dieback symptoms in West Germany, soil solution Mg2+ levels ranged between 20 and 60 mol L, and were only half those under a healthy stand. Alt and Ali levels were substantially higher the healthy stand than under the unhealthy stand, indicating that Al toxicity was not the main cause of spruce decline.  相似文献

2015年12月—2016年10月,每月小潮日原位定期向闽江口塔礁洲淡水感潮野慈姑(Sagittaria trifolia L.)湿地施加剂量为60、120 kg S hm~(-2)a~(-1)的K_2SO_4溶液(分别记做S-60和S-120),探讨模拟硫酸根(SO_4~(2-))沉降对河口淡水感潮湿地甲烷(CH4)排放通量及间隙水SO_4~(2-)浓度的影响。对照、S-60和S-120处理组CH_4排放通量年均值分别为(7.88±1.00)mg h~(-1)m~(-2)、(6.55±0.97)mg h~(-1)m~(-2)和(6.66±1.49)mg h~(-1)m~(-2)。在年尺度上,两个高强度模拟SO_4~(2-)沉降处理组均未显著降低闽江口淡水感潮野慈姑湿地CH_4排放通量(P0.05),即高强度SO_4~(2-)沉降不会对河口淡水感潮湿地CH_4排放通量产生类似于其对泥炭湿地和水稻田的显著抑制效应。在年尺度以及秋、冬季,两个施加K_2SO_4溶液处理显著增加了野慈姑湿地10 cm深度土壤间隙水SO_4~(2-)浓度。对于各个处理组,温度较高的夏、秋季CH_4排放通量均显著高于温度相对较低的冬、春季(P0.05)。不同处理组CH_4排放通量均与土壤温度呈显著正相关关系,温度仍然是影响亚热带河口淡水感潮湿地CH_4排放通量的重要环境因子。  相似文献

In order to quantify N2-emissions from a spruce and a beech site at the Höglwald Forest, a new measuring system was developed, that allowed simultaneous, direct determination of N2- and N2O-emission with high accuracy (detection limit approx. 10 g N m–2 h–1 for N2 and <1 g for N2O) using a gas-flow core method. This method requires exchange of the soil atmosphere with an artificial atmosphere, that differs only in that N2 is substituted by He. The measuring system, the methodology of measurements and validation experiments are described in detail. Due to the huge heterogeneity of denitrification activity in different soil cores taken from our forest sites, no general trends of N2 and N2O production in relation to soil moisture and temperature could be demonstrated. Based on reasonable number of measurements, this work gives for the first time an estimate of the magnitude of N2-losses from temperate forest soils. Both the magnitude of N2-emissions (spruce: 7.2±0.7 kg N2-N ha–1 yr–1; beech: 12.4±3.1 kg N2-N ha–1 yr–1), as well as the N2O–N2 ratio (spruce: 0.136±0.04; beech: 0.52±0.19) were significantly higher for soils from the beech sites as compared to soils from the spruce site. The results suggests that N2-emissions from N-saturated forest soils, still receiving high loads of atmospheric N-deposition, are approx. 30% of atmospheric N-input at the spruce site, and approx. 50% at the beech site. Our results demonstrate that losses of nitrogen in the form of N2 cannot be neglected in the context of calculating N-balances for given forest sites.  相似文献

The effects of light and several photosynthetic inhibitors on the rate of sulfite metabolism in cells obtained from Cucumis sativus L. cotyledons was studied. The cells were treated with 200 M Na2SO3 and the disappearance of sulfite was monitored using either dithiobisnitrobenzoic acid or fuchsin. The rate of sulfite disappearance in light was double the dark rate. Disalicylidene propanediamine at 1 mM increased this light-enhanced metabolism approx. 50%; neither 1 M 3,4-dichlorophenyl-N,N-dimethylurea nor 0.1 mM cyanazine, which completely inhibited CO2-dependent oxygen evolution, affected the rate of sulfite metabolism. Addition of 200 M Na2SO3 to the cells partially inhibited 14CO2 fixation. The rate of sulfite consumption by the cells did not affect this inhibition. We conclude that light-dependent sulfite metabolism is cucumber cells may utilize reduced ferredoxin generated as a result of photosynthetic electron transport. Desulfovibrio vulgaris (Marburg) was grown on H2 plus sulfate and H2 plus thiosulfate as the sole energy sources and acetate plus CO2 as the sole carbon sources. Conditions are described under which the bacteria grew exponentially. Specific growth rates () and molar growth yields (Y) at different pH were determined. and Y were found to be strongly dependent on the pH. Highest growth rates and molar growth yields were observed for growth on H2 plus sulfate at pH 6.5 (=0.15h-1; Y SO 4 2- =8.3g·mol-1) and for growth on H2 plus thiosulfate at pH 6.8 (=0.21h-1; Y S 2O 3 2 =16.9g·mol-1).The growth yields were found to increase with increasing growth rates: plots of 1/Y versus 1/ were linear. Via extrapolation to infinite growth rates a Y SO4 2- /max of 12.2g·mol-1 and a YS2O 3 2- /max of 33.5g·mol-1 was obtained.The growth yield data are interpred to indicate that dissimilatory sulfate reduction to sulfide is associated with a net synthesis of 1 mol of ATP and that near to 3 mol of ATP are formed during dissimilatory sulfite reduction to sulfide.  相似文献

Mitochondria from etiolated corn possess a much greater Ca2+ uptake capacity per mg protein than microsomes from the same source. Differences in energy requirements, sensitivity to specific inhibitors, and sedimentation properties enabled us to study both Ca2+ uptake mechanisms without mutual contamination. The microsomal Ca2+ uptake does not vary much among different plants as compared to the mitochondrial Ca2+ uptake; this is also true for different organs of the same plant. Mitochondrial Ca2+ uptake is more dependent on the age of the seedlings than microsomal uptake, because of changes in active Ca2+ uptake activity rather than of changes in efflux. Intactness and the oxidative and phosphorylative properties of the mitochondria remained unchanged during this time period. Chloroplasts with high rates of photosynthetic O2 evolution (up to 120 mol O2· (mg Chl)-1·h-1 compared with 130 mol O2· (mg Chl)-1·h-1 of whole cells) were isolated from Chlamydomonas reinhardtii cells grown in high and low CO2 concentrations using autolysine-digitonin treatment. At 25° C and pH=7.8, no O2 uptake could be observed in the dark by high- and low-CO2 adapted chloroplasts. Light saturation of photosynthetic net oxygen evolution was reached at 800 mol photons·m-2·s-1 for high- and low-CO2 adapted chloroplasts, a value which was almost identical to that observed for whole cells. Dissolved inorganic carbon (DIC) saturation of photosynthesis was reached between 200–300 M for low-CO2 adapted chloroplasts, whereas high-CO2 adapted chloroplasts were not saturated even at 700 M DIC. The concentrations of DIC required to reach half-saturated rates of net O2 evolution (Km(DIC)) was 31.1 and 156 M DIC for low- and high-CO2 adapted chloroplasts, respectively. The uptake and conversion of NO 2 - and the effect of NO 2 - on the uptake and reduction of NO 3 - were examined in N-depleted Phaseolus vulgaris L. Nitrite uptake at 0.1 mmol dm-3 was against an electrochemical gradient and became constant after one or two initial phases. Steadystate uptake declined with increasing ambient NO 2 - concentration (0–0.7 mmol dm-3). In this concentration range root oxygen consumption was unaffected by NO 2 - , indicating that the decrease of NO 2 - uptake was not related to respiration. After 6 h NO 2 - supply, about one-third of the absorbed NO 2 - had accumulated, mainly in the root system. Oxidation of NO 2 - to NO 3 - was not observed. The apparent induction period for NO 3 - uptake was about 6 h in control plants and 3.5 h in plants that were pretreated for 18 h with NO 2 - . In contrast, the time course of NO 2 - uptake was unaffected by pretreatment with NO 3 - . Steadystate NO 3 - uptake was less affected by NO 2 - than was steady-state NO 2 - uptake by NO 3 - . Nitrate reductase activity (NRA) in leaves and roots was induced by both NO 3 - and NO 2 - . In roots, induction with NO 2 - was faster than with NO 3 - , but there was no difference in NRA after 5 h. Nitrite inhibited NRA in the roots of NO 3 - -induced plants and thus seems to stimulate the induction, but not the activity of induced nitrate reductase. In view of the observed differences in time course and mutual competition, a common uptake mechanism for NO 2 - and NO 3 - seems unlikely. Expression of the NO 2 - effect on the induction of NO 3 - uptake required more time than the induction itself. Factors influencing the rate of superoxide (O 2 - ) production by thylakoids were investigated to determine if increased production of the radical was related to injury induced by chilling at a moderate photon flux density (PFD). Plants used were Spinacia oleracea L., Cucumis sativus L. and Nerium oleander L. grown at either 200° C or 45° C. Superoxide production was determined by electron-spin-resonance spectroscopy of the (O 2 - )-dependent rate of oxidation of 2-ethyl-1-hydroxy-2,5,5-trimethyl-3-oxazolidine (OXANOH) to the corresponding oxazolidinoxyl radical, OXANO ·. For all plants, the steady-state rate of O 2 - production by thylakoids, incubated at 25° C and 350 mol photon · m–2 · s–1 (moderate PFD) with added ferredoxin and NADP, was between 7.5 and 12.5 mol · (mg chlorophyll)–1 · h–1. Incubation at 5° C and a moderate PFD, decreased the rate of O 2 - production 40% and 15% by thylakoids from S. oleracea and 20° C-grown N. oleander, chillinginsensitive plants, but increased the rate by 56% and 5% by thylakoids from C. sativus and 45° C-grown N. oleander, chilling-sensitive plants. For all plants, the addition of either ferredoxin or methyl viologen increased the rate of O 2 - -production at 25° C by 75–100%. With these electron acceptors, lowering the temperature to 5° C caused only a slight decrease in O 2 - production. In the absence of added electron acceptors, thylakoids produced O 2 - at a rate which was about 45% greater than that when ferredoxin and NADP were present. The addition of 3-(3,4-dichlorophenyl)-1,1-dimethylurea reduced O 2 - production under all conditions tested. The results show that the rate of O 2 - production increases in thylakoids when the rate of electron transfer to NADP is reduced. Soil flooding damages shoot systems by inhibiting root functioning. An example is the inhibition of water uptake brought about by decreased root hydraulic conductance. The extent of any resulting foliar dehydration this causes is limited by partial stomatal closure that begins within 4 h and is maintained for several days. Root to shoot signals that promote closure in flooded tomato plants have remained elusive but may include changes in solute delivery to the shoot by transpiration. Accordingly, we examined total osmolites and selected mineral ions in samples of xylem sap flowing at rates approximating whole plant transpiration. After 2.5 h flooding,delivery of total osmolites and of PO4 3-SO4 2-Ca2+K+NO3 and H+strongly decreased while Na+ remained excluded. Several hours later, deliveries of osmolites, PO4 3-, SO4 2-, Ca2+, and Na+ rose above control values, suggesting that, after approximately 10 h, root integrity became degraded and solute uptake de-regulated. Deliveries of NO3 remained below control values. Reducing or eliminating the supply of K+ to detached leaves to test the potential of decreased K+ delivery to close stomata proved negative. Decrease in H+ delivery was associated with sap alkalisation. However, raising the pH of buffer from 6.0 or 6.5 to 7.0 did not close stomata when tested in the presence of abscisic acid (ABA) at a concentration (10 mol m–3) typical of the transpiration stream of flooded plants. It is concluded that despite their rapidity and scale, negative messages in the form of increased pH and decreased solute delivery from roots to shoots are, themselves, unlikely initiators of stomatal closure in flooded tomato plants.  相似文献

To evaluate the role of the gill chloride cells in regulating metabolic alkalosis in rainbow trout (Oncorhynchus mykiss), the surface area of branchial chloride cells was altered experimentally using combined cortisol/ovine growth hormone injections. Long-term (10-day) treatment of fish with cortisol/ovine growth hormone caused an increase in the two-dimensional chloride cell fractional surface area when compared to uninjected fish (from 8.4 to 29.7%). This was the combined result of an increase in the size of individual cells (from 34.6 to 59.2 m2) and increased numbers of cells (from 2368 to 5006 cells · mm-2). Metabolic alkalosis was induced by intra-arterial infusion of 140 mmol · l-1 NaHCO3; control fish were infused with 140 mmol · l-1 NaCl. Blood pH and plasma [HCO3 -] increased in both the untreated and the cortisol/ovine growth hormone-treated fish. However, the increases in pH (from 8.05 to 8.53) and [HCO3 -] (from 5.9 to 22.2 mmol · l-1) in the untreated fish were significantly greater than in the cortisol/ovine growth hormone-treated fish (pH increased from 7.78 to 8.11; [HCO3 -] increased from 5.5 to 13.9 mmol · l-1). In all fish, NaHCO3 infusion elicited an increase in the rate of branchial basic equivalent excretion (acidic equivalent uptake) which, in turn, was caused by decreases and increases in branchial Na+ uptake and Cl- uptake, respectively. In the untreated fish, there was a pronounced increase (75%) in chloride cell surface area during NaHCO3 infusion. The attenuation of the metabolic alkalosis during HCO3 - infusion in the cortical/ovine growth hormone-treated fish was caused, at least in part, by an enhancement of branchial basic equivalent excretion. In these fish that already displayed a proliferation of chloride cells, there was no further increase in chloride cell surface area. The changes in Na+ influx and Cl- influx were quantitatively similar during NaHCO3 infusion in both groups. Summary Elodea canadensis grows over a wide range of inorganic carbon, nutrient, and light conditions in lakes and streams. Affinity for HCO 3 - use during photosynthesis ranged from strong to weak in Elodea collected from seven localities with different HCO 3 - and CO2 concentrations. The response to HCO 3 - was also very plastic in plants grown in the laboratory at high HCO 3 - concentrations and CO2 concentrations varying from 14.8 to 2,200 M. Bicarbonate affinity was markedly reduced with increasing CO2 concentrations in the growth medium so that ultimately HCO 3 - use was not detectable. High CO2 concentrations also decreased CO2 affinity and induced high CO2 compensation points (360M CO2) and tenfold higher half-saturation values (800 M CO2).The variable HCO 3 - affinity is probably environmentally based. Elodea is a recently introduced species in Denmark, where it reproduces only vegetatively, leaving little opportunity for genetic variation. More important, local populations in the same water system had different HCO 3 - affinities, and a similar variation was created by exposing one plant collection to different laboratory conditions.Bicarbonate use enabled Elodea to photosynthesize rapidly in waters of high alkalinity and enhanced the carbon-extracting capacity by maintaining photosynthesis above pH 10. On the other hand, use of HCO 3 - represents an investment in transport apparatus and energy which is probably not profitable when CO2 is high and HCO 3 - is low. The carbon-dioxide response of photosynthesis of leaves of Quercus suber, a sclerophyllous species of the European Mediterranean region, was studied as a function of time of day at the end of the summer dry season in the natural habitat. To examine the response experimentally, a standard time course for temperature and humidity, which resembled natural conditions, was imposed on the leaves, and the CO2 pressure external to the leaves on subsequent days was varied. The particular temperature and humidity conditions chosen were those which elicited a strong stomatal closure at midday and the simultaneous depression of net CO2 uptake. Midday depression of CO2 uptake is the result of i) a decrease in CO2-saturated photosynthetic capacity after light saturation is reached in the early morning, ii) a decrease in the initial slope of the CO2 response curve (carboxylation efficiency), and iii) a substantial increase in the CO2 compensation point caused by an increase in leaf temperature and a decrease in humidity. As a consequence of the changes in photosynthesis, the internal leaf CO2 pressure remained essentially constant despite stomatal closure. The effects on capacity, slope, and compensation point were reversed by lowering the temperature and increasing the humidity in the afternoon. Constant internal CO2 may aid in minimizing photoinhibition during stomatal closure at midday. Summary Leaves of two field growing co-occuring perennial shrubs (drought-deciduous Diplacus aurantiacus and the evergreen Heteromeles arbutifolia) from the Californian chaparral were exposed to small doses of SO2. During this exposure the leaf environment was manipulated to determine how the presence of SO2 alters the response of gas exchange to other environmental stresses. The data show that no direct changes in stomatal conductance (g) or net assimilation rate (A) could be attributed to short-term (7 h) SO2 (4.2 mol m-3, 0.1 l l-1) exposure. D. aurantiacus leaves possessed features which demonstrate that they were sensitive to changes in environment e.g. light flux and atmospheric relative humidity. The interspecific differences in stomatal sensitivity to water vapour were extremely important, as relative humidity is a major factor influencing carbon fixation and the rate of pollutant absorption. Conditions of high relative humidity and high xylem water potentials are suggested to pre-dispose leaves of D. aurantiacus to greater pollutant doses than the more stomatally-conservative evergreen, H. arbutifolia. In the presence of SO2 there was some indication of increased g for both D. aurantiacus and H. arbutifolia as W became smaller. This SO2-effect was only obvious as increasing atmospheric humidity induced further stomatal opening. Complete annual cycles of N2O and CH4 flux in forest soils at a beech and at a spruce site at the Höglwald Forest were followed in 1997 by use of fully automatic measuring systems. In order to test if on a microsite scale differences in the magnitude of trace gas exchange between e.g. areas in direct vicinity of stems and areas in the interstem region at both sites exist, tree chambers and gradient chambers were installed in addition to the already existing interstem chambers at our sites. N2O fluxes were in a range of –4.6–473.3 g N2O-N m–2 h–1 at the beech site and in a range of –3.7–167.2 g N2O-N m–2 h–1 at the spruce site, respectively. Highest N2O emissions were observed during and at the end of a prolonged frost period, thereby further supporting previous findings that frost periods are of crucial importance for controlling annual N2O losses from temperate forests. Fluxes of CH4 were in a range of +10.4––194.0 g CH4 m–2 h–1 at the beech site and in a range of –4.4––83.5 g CH4 m–2 h–1 at the spruce site. In general, both N2O-fluxes as well as CH4-fluxes were higher at the beech site. On a microsite scale, N2O and CH4 fluxes at the beech site were highest within the stem area (annual mean: 49.6±3.3 g N2O-N m–2 h–1; –77.2±3.1 g CH4 m–2 h–1), and significantly lower within interstem areas (18.5±1.4 g N2O-N m–2 h–1; –60.2±1.8 g CH4 m–2 h–1). Significantly higher values of total N, C and pH in the organic layer, as well as increased soil moisture, especially in spring, in the stem areas, are likely to contribute to the higher N2O fluxes within the stem area of the beech. Also for the spruce site, such differences in trace gas fluxes could be demonstrated to exist (mean annual N2O emission within (a) stem areas: 9.7±0.9 g N2O-N m–2 h–1 and (b) interstem areas: 6.2±0.6 g N2O-N m–2 h–1; mean annual CH4 uptake within (a) stem areas: –26.1±0.6 g CH4 m–2 h–1 and (b) interstem areas: –38.4±0.8 g CH4 m–2 h–1), though they were not as pronounced as at the beech site.  相似文献

Mass spectrometric measurements of 16O2 and 18O2 isotopes were used to compare the rates of gross O2 evolution (E0), O2 uptake (U0) and net O2 evolution (NET) in relation to different concentrations of dissolved inorganic carbon (DIC) by Chlamydomonas reinhardtii cells grown in air (air-grown), in air enriched with 5% CO2 (CO2-grown) and by cells grown in 5% CO2 and then adapted to air for 6h (air-adapted).At a photon fluence rate (PFR) saturating for photosynthesis (700 mol photons m-2 s-1), pH=7.0 and 28°C, U0 equalled E0 at the DIC compensation point which was 10M DIC for CO2-grown and zero for air-grown cells. Both E0 and U0 were strongly dependent on DIC and reached DIC saturation at 480 M and 70 M for CO2-grown and air-grown algae respectively. U0 increased from DIC compensation to DIC saturation. The U0 values were about 40 (CO2-grown), 165 (air-adapted) and 60 mol O2 mg Chl-1 h-1 (air-grown). Above DIC compensation the U0/E0 ratios of air-adapted and air-grown algae were always higher than those of CO2-grown cells. These differences in O2 exchange between CO2- and air-grown algae seem to be inducable since air-adapted algae respond similarly to air-grown cells.For all algae, the rates of dark respiratory O2 uptake measured 5 min after darkening were considerably lower than the rates of O2 uptake just before darkening. Throughfall nitrogen of a 15-year-old Picea abies (L.) Karst. (Norway spruce) stand in the Fichtelgebirge, Germany, was labeled with either 15N-ammonium or 15N-nitrate and uptake of these two tracers was followed during two successive growing seasons (1991 and 1992). 15N-labeling (62 mg 15N m-2 under conditions of 1.5 g N m-2 atmospheric nitrogen deposition) did not increase N concentrations in plant tissues. The 15N recovery within the entire stand (including soils) was 94%±6% of the applied 15N-ammonium tracer and 100%±6% of the applied 15N-nitrate tracer during the 1st year of investigation. This decreased to 80%±24% and 83%±20%, respectively, during the 2nd year. After 11 days, the 15N tracer was detectable in 1-year-old spruce needles and leaves of understory species. After 1 month, tracer was detectable in needle litter fall. At the end of the first growing season, more than 50% of the 15N taken up by spruce was assimilated in needles, and more than 20% in twigs. The relative distribution of recovered tracer of both 15N-ammonium and 15N-nitrate was similar within the different foliage age classes (recent to 11-year-old) and other compartments of the trees. 15N enrichment generally decreased with increasing tissue age. Roots accounted for up to 20% of the recovered 15N in spruce; no enrichment could be detected in stem wood. Although 15N-ammonium and 15N-nitrate were applied in the same molar quantities (15NH 4 + : 15NO 3 - =1:1), the tracers were diluted differently in the inorganic soil N pools (15NH 4 + /NH 4 + : 15NO 3 - /NO 3 - =1:9). Therefore the measured 15N amounts retained by the vegetation do not represent the actual fluxes of ammonium and nitrate in the soil solution. Use of the molar ammonium-to-nitrate ratio of 9:1 in the soil water extract to estimate 15N uptake from inorganic N pools resulted in a 2–4 times higher ammonium than nitrate uptake by P. abies.  相似文献

Internodal and whorl (branch) cells of the green alga,Chara corallina Klein ex Willd., em. R.D.W., were studied with the extracellular vibrating probe for measuring transmembrane ion currents, and with an extracellular pH microprobe for measuring the surface pH profile. Bands of positive inward current (OH- efflux) 1–3 mm wide were separated by wider bands of outward current (HCO 3 - influx) along the length of the cell. The measured peaks of inward current ranged from 20 to 60 A cm-2 (98 m from the cell surface) which would correspond to a surface ionic flux of 270–800 pmol cm-2 s-1. The peaks of outward current (HCO 3 - influx) ranged from 10 to 30 A cm-2 which would correspond to a surface ionic flux of 140–400 pmol cm-2 s-1. The inward current bands matched the regions of surface alkalinity very well. The outward current (HCO 3 - influx) was reduced at least 10-fold in low-HCO 3 - medium, with a commensurate readjustment in the strength and pattern of inward current (OH- efflux). (Although these experiments involved a manipulation of the external pH, it is felt that the main adjustment in current patterns was in response to the reduction in exogenous HCO 3 - ). The presence of the vibrating probe perturbed the inward current region when vibrating with a 26-m amplitude, but this perturbation was eliminated when a 7-m amplitude was used. The perturbation was usually observed as a reduction in the number of inward current peaks with an increase (approximate doubling) in the amplitudes of the one or two remaining peaks. Both the inward and outward currents were light-dependent, falling off within seconds of light removal.  相似文献
