The first aggregation pheromone components from bark beetles were identified in 1966 as a mixture of ipsdienol, ipsenol and verbenol. Since then, a number of additional components have been identified as both aggregation and anti-aggregation pheromones, with many of them being monoterpenoids or derived from monoterpenoids. The structural similarity between the major pheromone components of bark beetles and the monoterpenes found in the host trees, along with the association of monoterpenoid production with plant tissue, led to the paradigm that most if not all bark beetle pheromone components were derived from host tree precursors, often with a simple hydroxylation producing the pheromone. In the 1990s there was a paradigm shift as evidence for de novo biosynthesis of pheromone components began to accumulate, and it is now recognized that most bark beetle monoterpenoid aggregation pheromone components are biosynthesized de novo. The bark beetle aggregation pheromones are released from the frass, which is consistent with the isoprenoid aggregation pheromones, including ipsdienol, ipsenol and frontalin, being produced in midgut tissue. It appears that exo-brevocomin is produced de novo in fat body tissue, and that verbenol, verbenone and verbenene are produced from dietary α-pinene in fat body tissue. Combined biochemical, molecular and functional genomics studies in Ips pini yielded the discovery and characterization of the enzymes that convert mevalonate pathway intermediates to pheromone components, including a novel bifunctional geranyl diphosphate synthase/myrcene synthase, a cytochrome P450 that hydroxylates myrcene to ipsdienol, and an oxidoreductase that interconverts ipsdienol and ipsdienone to achieve the appropriate stereochemistry of ipsdienol for pheromonal activity. Furthermore, the regulation of these genes and their corresponding enzymes proved complex and diverse in different species. Mevalonate pathway genes in pheromone producing male I. pini have much higher basal levels than in females, and feeding induces their expression. In I. duplicatus and I. pini, juvenile hormone III (JH III) induces pheromone production in the absence of feeding, whereas in I. paraconfusus and I. confusus, topically applied JH III does not induce pheromone production. In all four species, feeding induces pheromone production. While many of the details of pheromone production, including the site of synthesis, pathways and knowledge of the enzymes involved are known for Ips, less is known about pheromone production in Dendroctonus. Functional genomics studies are under way in D. ponderosae, which should rapidly increase our understanding of pheromone production in this genus. This chapter presents a historical development of what is known about pheromone production in bark beetles, emphasizes the genomic and post-genomic work in I. pini and points out areas where research is needed to obtain a more complete understanding of pheromone production.  相似文献

We treated Norway spruce (Picea abies) stems with methyl jasmonate (MeJA) to determine possible quantitative and qualitative effects of induced tree defenses on pheromone emission by the spruce bark beetle Ips typographus. We measured the amounts of 2-methyl-3-buten-2-ol and (S)-cis-verbenol, the two main components of the beetle's aggregation pheromone, released from beetle entrance holes, along with phloem terpene content and beetle performance in MeJA-treated and untreated Norway spruce logs. As expected, phloem terpene levels were higher and beetle tunnel length was shorter (an indication of poor performance) in MeJA-treated logs relative to untreated logs. Parallel to the higher phloem terpene content and poorer beetle performance, beetles in MeJA-treated logs released significantly less 2-methyl-3-buten-2-ol and (S)-cis-verbenol, and the ratio between the two pheromone components was significantly altered. These results suggest that host resistance elicited by MeJA application reduces pheromone emission by I. typographus and alters the critical ratio between the two main pheromone components needed to elicit aggregation. The results also provide a mechanistic explanation for the reduced performance and attractivity observed in earlier studies when bark beetles colonize trees with elicited host defenses, and extend our understanding of the ecological functions of conifer resistance against bark beetles.  相似文献

Pine-feeding bark beetles (Coleoptera: Scolytidae) interact chemically with their host pines (Coniferales: Pinaceae) via the behavioral, physiological, and biochemical effects of one class of isoprenoids, the monoterpenes and their derivatives. Pine monoterpenes occur in the oleoresin and function as behaviorally active kairomones for pine bark beetles and their predators, presenting a classic example of tri-trophic chemical communication. The monoterpenes are also essential co-attractants for pine bark beetle aggregation pheromones. Ironically, pine monoterpenes are also toxic physiologically to bark beetles at high vapor concentrations and are considered an important component of the defense of pines. Research over the last 30 years has demonstrated that some bark beetle aggregation pheromones arise through oxygenation of monoterpenes, linking pheromone biosynthesis to the host pines. Over the last 10 years, however, several frequently occurring oxygenated monoterpene pheromone components (e.g., ipsenol, ipsdienol and frontalin) have also been shown to arise through highly regulated de novo pathways in the beetles (reviewed in Seybold and Tittiger, 2003). The most interesting nexus between these insects and their plant hosts involves the late-stage reactions in the monoterpenoid biosynthetic pathway, during which isomeric dimethylallyl diphosphate and isopentenyl diphosphate are ultimately elaborated to stereospecific monoterpenes in the trees and to hydroxylated monoterpenes or bicyclic acetals in the insects. There is signal stereospecificity in both production of and response to the monoterpenoid aggregation pheromones of bark beetles and in response to␣the monoterpenes of the pines. In the California fivespined ips, Ips paraconfusus, we have discovered a number of cytochome P450 genes that have expression patterns indicating that they may be involved in detoxifying monoterpene secondary metabolites and/or biosynthesizing pheromone components. Both processes result in the production of oxygenated monoterpenes, likely with varying degrees of stereospecificity. A behavioral analysis of the stereospecific response of I. paraconfusus to its pheromone is providing new insights into the development of an efficacious bait for the detection of this polyphagous insect in areas outside the western United States. In contrast, a Eurasian species that has arrived in California, the Mediterranean pine engraver, Orthotomicus (Ips) erosus, utilizes both a monoterpenoid (ipsdienol) and a hemiterpenoid (2-methyl-3-buten-2-ol) in its pheromone blend. The stereospecificity of the response of O. erosus to the monoterpenoid appears to be the key factor to the improved potency of the attractant bait for this invasive species.Dedicated to Professor David L. Timing of arrival at a resource often determines an individual’s reproductive success. Tree-killing bark beetles can reproduce in healthy trees by attacking in adequate numbers to overcome host defences that could otherwise be lethal. This process is mediated by aggregation and antiaggregation pheromones. Beetles that arrive early in such a “mass attack” must contend with undiminished tree defences, and produce enough pheromones to attract more beetles, but have a head start on gallery construction and egg-laying. Beetles that arrive late may be impeded by competition and diminishing availability of phloem, but should experience fewer costs associated with pheromone production and battling tree defences. We investigated relationships between timing of arrival, body size, pheromone production and fitness in the southern pine beetle, Dendroctonus frontalis. In field experiments, we captured beetles that arrived early (pioneers) and late on slash pine trees, Pinus elliottii, and measured pheromone amounts in their hindguts. We marked gallery entrances of beetles as they landed on a tree and measured their reproductive success after the attack terminated. We found no difference in body size or pheromone amounts between early and late arrivers. Most beetles arrived at the middle of the attack sequence, and excavated longer galleries per day than early arrivers. The number of offspring produced per day by beetles that established galleries midway through mass attack was higher than those that arrived early or very late in the sequence. Our results suggest that beetles do not exhibit adaptive phenotypic plasticity in pre-landing pheromone production, depending on the extent of previous colonisation of a host. Rather, it appears that stabilising selection favours beetles that attack in the middle of the sequence, and contributes to attack synchrony. Synchronous attack on trees is essential before population booms characteristic of tree-killing bark beetles can occur in nature.  相似文献

A summary of the present knowledge of variation in individual quality within a bark beetle population is given, with emphasis on the spruce bark beetle Ips typogrophus , A major causal factor is density, mediated by competition during larval development. Density negatively influences individual quality measured as weight, fat content and pheromone production. Together with decreasing mean values at higher densities, the skewness of the frequency distributions goes from negative to positive, while variance changes little. High densities, which often occur in the field, thus result in a large fraction of "low quality beetles". They have lower reproductive capacity and presumably lesser dispersal ability, lower survival, and earlier response to pheromone. This might concentrate the population in the next generation with increased competition as a result. It is suggested that a decrease in beetle "quality" due to increasingly intense intraspecific competition can contribute to the decline of an epidemic population.  相似文献

Chemical communication in scarab beetles involves female-released long-distance sex pheromones. Electrophysiological recordings using tungsten microelectrodes demonstrated two types of olfactory receptor neurons in the scarab beetle Anomala cuprea, each specific for one of the two pheromone components (R)-buibuilactone and (R)-japonilure, respectively. No neurons were found that responded specifically to enantiomers of the pheromone compounds, i.e. (S)-buibuilactone and (S)-japonilure. Pheromone receptor neurons are present in high numbers on both the male and the female antenna, with a lower sensitivity in the females. As in bark beetles and moths, the pheromone receptor neurons in A. cuprea are very sensitive and selective. The difference in response thresholds between (R)- and (S)-enantiomers is almost three orders of magnitude. Pheromone receptor neurons are found in sensilla placodea located in a defined area on each lamella in the antennal club. (R)-buibuilactone and (R)-japonilure neurons are always found in different sensilla. Both types of sensilla contain two neurons, with the pheromone-sensitive neuron displaying a high spike amplitude and the second neuron, not responding to any of the tested compounds, always with a lower spike amplitude. Summary Two species of predatory beetles that locate their prey, Ips pini, by responding to its aggregation pheromone have different chiral preferences to ispdienol than does the herbivore. This suggests that chiral disparity may provide some escape for bark beetles from predation, and that geographic variation in herbivore communication systems may be partially due to predator — imposed selection pressures. These results also suggest ways in which the semiochemical and biological control of North America's most damaging group of forest insects can be improved.  相似文献

An analysis of semiochemical communication between host trees, bark beetles and commensal or entomophagous insects discloses five principal means by which semiochemicals can influence the population dynamics of bark beetles. These are: mediation of aggregation and mass attack on new hosts, cessation of aggregation and shifting of attack to uninhabited hosts, induction of aggregation by competing species, inhibition of aggregation by competing species, and mediation of host finding by commensal and entomophagous insects. Further analysis suggests major points of natural vulnerability which lead to six fundamental strategies for potential pest management: prevention of production of aggregation pheromones, sabotage of olfactory perception, exploitation of semiochemical-based secondary attraction, exploitation of antiaggregation pheromones, exploitation of repellent allomones, and exploitation of the kairomonal response by entomophagous insects. Investigations of the many possible tactics arising from these strategies have led to three types of operational pest management programs: prevention of pheromone production by excluding bark beetles from their hosts; suppression of bark beetle populations through the utilization of semiochemical-baited traps, trees or logs; and the use of antiaggregation pheromones to protect vulnerable hosts from attack.  相似文献

Abstract 1 Host plant terpenes can influence attraction of conifer bark beetles to their aggregation pheromones: both synergistic and inhibitory compounds have been reported. However, we know little about how varying concentrations of individual monoterpenes affect responses. 2 We tested a gradient of ratios of α‐pinene, the predominant monoterpene in host pines in the Great Lakes region of North America, to Ips pini's pheromone, racemic ipsdienol plus lanierone. 3 Ips pini demonstrated a parabolic response, in which low concentrations of α‐pinene had no effect on attraction to its pheromone, intermediate concentrations were synergistic and high concentrations were inhibitory. These results suggest optimal release rates for population monitoring and suppression programmes. 4 Inhibition of bark beetle attraction to pheromones may be an important component of conifer defences. At terpene to pheromone ratios emulating emissions from trees actively responding to a first attack, arrival of flying beetles was low. This may constitute an additional defensive role of terpenes, which are also toxic to bark beetles at high concentrations. 5 Reduced attraction to a low ratio of α‐pinene to pheromone, as occurs when colonization densities become high and the tree's resin is largely depleted, might reflect a mechanism for preventing excessive crowding. 6 Thanasimus dubius, the predominant predator of I. pini, was also attracted to ipsdienol plus lanierone, but its response differed from that of its prey. Attraction increased across all concentrations of α‐pinene. This indicates that separate lures are needed to sample both predators and bark beetles effectively. It also provides an opportunity for maximizing pest removal while reducing adverse effects on beneficial species. This disparity further illustrates the complexity confronting natural enemies that track chemical signals to locate herbivores.  相似文献

Adult male T. castaneum (Herbst) and T. confusum (du Val) secretes an aggregation pheromone that is attractive to both sexes. Orientation behaviour of the two Tribolium species responding to different concentrations of synthetic aggregation pheromone in still and moving air was studied in an 2.5 m×0.4 m olfactometer. Analysis of Tribolium tracks indicated that the aggregation pheromone stimulated the beetles to walk faster at higher concentrations to increase the frequency and magnitude of turning and to decrease track reversal distances and distances between turns. The mean walking speed of both species was lowest at the highest air speed. The behavioural responses of the beetles to the pheromone in still and moving air were similar, indicating chemotaxis as the major orientation mechanism used by both species to locate an odour source. The beetles showed greater orientation efficiency within a discrete pheromone plume than a diffuse plume.  相似文献

Abstract:  Semiochemical interactions between the spruce bark beetle Ips typographus , its predators Medetera setiventris , Thanasimus formicarius and Thanasimus femoralis , and the host Norway spruce, Picea abies , were studied in the field. The chemicals S - cis -verbenol, 2-methyl-3-buten-2-ol, ipsdienol, (+)- α -pinene, (−)- α -pinene, (±)- α -pinene, limonene, camphor and their naturally occuring mixtures were used as trap baits in a multiple-choice design that allowed for comparison of their attractivity for the focal species. Medetera was attracted to both the prey aggregation pheromone and its multifunctional component, ipsdienol. On the contrary, both Thanasimus species responded predominantly to ipsdienol and less to the prey aggregation pheromone. In the case of I. typographus , the attractivity of aggregation pheromone seems to be increased by the addition of a mixture of monoterpenic tree volatiles, and by addition of ipsdienol. Bark beetles and predators showed species-specific responses to volatile mixtures representing different stages of tree decay and different stages of bark beetle colony establishment. These responses correlates with the optimal foraging habitat of each species. None of the predator species responded to 2-methyl-3-buten-2-ol, a substantial component of I. typographus pheromonal bouquet, thus it is hypothesized that only substances of monoterpenic origin attract predators.  相似文献

Abstract.  Two-day-old male cowpea weevils, Callosobruchus maculatus, fly upwind to a point source of female sex pheromone at three wind speeds. All beetles initiating flight along the pheromone plume make contact with the pheromone source. Analysis of digitized flight tracks indicates that C. maculatus males respond similarly to moths tested at several wind speeds. Beetles' mean net upwind speeds and speeds along their track are similar ( P  > 0.05) across wind speeds, whereas airspeeds increase ( P <  0.01) with increasing wind speed. Beetles adjust their course angles to fly more directly upwind in higher wind speeds, whereas track angles are almost identical at each wind speed. The zigzag flight paths are generally narrow compared with most moth flight tracks and interturn distances are similar ( P  > 0.05) at the wind speeds employed. The frequency of these counterturns across the wind line is almost constant regardless of wind speed, and there is little variation between individuals. The upwind flight tracks are more directly upwind than those typically seen for male moths flying upwind toward sex pheromone sources. Male moths typically produce a bimodal distribution of track angles to the left and right of the windline, whereas C. maculatus males' track angles are centred about 0°. Preliminary examination of two other beetle species indicates that they fly upwind in a similar fashion.  相似文献

Sex and aggregation pheromones consist of species-specific blends of chemicals. The way in which different species' blends have evolved has been the subject of some debate. Theoretical predictions suggest that differences between species have arisen not through the accruing of small changes, but through major shifts in chemical composition. Using data on the aggregation pheromones of 34 species of bark beetle from two genera, Dendroctonus and Ips, we investigated how the distributions of the chemical components of their pheromone blends mirror their phylogenetic relationships. We tested whether there were consistent patterns that could be used to help elucidate the mode of pheromone evolution. Although there were obvious differences in pheromone blends between the two genera, the differences between species within each genus followed a less clear phylogenetic pattern. In both genera, closely related species are just as different as more distantly related species. Within Dendroctonus, particularly, most chemical components were distributed randomly across the phylogeny. Indeed, for some chemicals, closely related species may actually be more different than would be expected from a random distribution of chemical components. This argues strongly against the idea of minor shifts in pheromone evolution. Instead, we suggest that, within certain phylogenetic constraints, pheromone evolution in bark beetles is characterized by large saltational shifts, resulting in sibling species being substantially phenotypically (i.e. pheromonally) different from one another, thus agreeing with theoretical predictions.  相似文献

Temperatures in the forest litter of Norway spruce Picea abies were recorded throughout the day to obtain environmental parameters that could be used to design realistic flight-activity experiments in the laboratory. Flight activity and survival were monitored electronically in plastic chambers where the conditions were controlled by an environmental chamber. Flight attempts of the bark beetle Ips typographus were initially lower in chambers with forest duff but were prolonged compared with those of beetles in chambers with a metal screen substrate. Small bark slabs and spruce twigs in the duff were utilized as food and extended the period of flight and survival. A thermal gradient in duff from 25° at the surface down to 13.8°C at a depth of 4 cm also slightly increased the survival of beetles compared with a constant 25°. A daily ambient temperature cycle as well as the duff thermal gradient increased the survival from about 3 d to more than 8 d. The latter length in the laboratory agreed with survival rates of caged beetles in a clearcut forest area, while beetles caged in the forest survival for more than 14 d. Temperatures were monitored at the duff surface of the caged beetles and compared with the catches of beetles that were attracted to a pheromone trap and collected with an electronic fraction collector. Information on flight and survival during the dispersal period is necessary to the design of ecologically sound management programs for control of bark beetles.  相似文献

1 Host tree terpenes can influence attraction of conifer‐infesting bark beetles to their aggregation pheromones, and both synergistic and inhibitory effects have been reported. 2 We tested a gradient of ratios of (–)‐α‐pinene, the predominant monoterpene in Norway spruce, to the pheromone of Ips typographus, a major pest of Norway spruce. 3 Attraction of I. typographus increased as the release rate of (–)‐α‐pinene increased. The two highest (–)‐α‐pinene : pheromone ratios (526 : 1 and 2595 : 1) attracted twice as many I. typographus as pheromone alone, whereas low to intermediate ratios (56 : 1, 274 : 1) did not differ from pheromone alone. 4 Our results are in agreement with a proposed model, which suggests that bark beetles display unique response profiles to host terpenes depending on the physiological condition of the host trees that they typically colonize. Ips typographus, which is an aggressive species capable of colonizing and killing healthy trees, showed an increased attraction to monoterpene : pheromone ratios, and this may be high enough to inhibit attraction of less aggressive beetle species typically colonizing dead, dying or stressed trees. 5 Attraction of associates of I. typographus was also modified by (–)‐α‐pinene. Ips duplicatus, a competitor of I. typographus, showed increased attraction to the pheromone of I. typographus across all concentrations of (–)‐α‐pinene.  相似文献

The influence of population density and aggregation pheromone were investigated as possible flight initiating factors for Prostephanus truncatus (Horn) (Col.: Bostrichidae). Maize grains (175 g in glass jars) were infested with 20, 50, 150 or 300 beetles, all of which were removed after 2 weeks. Flight initiation of the progeny was observed over a period of 4 weeks, with and without the synthetic aggregation pheromone of P. truncatus. Addition of the synthetic pheromone had no influence on the number of beetles taking off. The number of beetles dispersing increased with growing initial and progeny densities. Dispersal rates (% of total progeny dispersing) rose with declining rate till they reached a maximum at ca. 33%. Of the beetles dispersing, 74% left the cultures between 18.00 and 20.00 hours. Surviving progeny per female decreased with increasing initial density. Sex ratios in the initial cultures and in the F1 were ca. 1:1, whereas a higher proportion of females (67%) was found among the dispersing beetles.  相似文献

ABSTRACT. Equal numbers of dispersing Ips grandicollis adults were caught on sticky traps held on susceptible and resistant Pinus radiata trees. When beetles were attracted to the same set of trees using baits of ipsenol (aggregation pheromone), some of the beetles that landed on the boles of the trees bored into the bark. The subsequent response of the tree determined whether the tree was accepted or rejected. In resistant trees, the exudation of resin prevented continued boring and led to the retreat of the beetles. In susceptible trees, continued boring led to the establishment of the beetles. Ipsenol was not detected in males that retreated after boring into resistant trees. Beetles that bored into P. radiata logs obtained from slash, continued to bore into the bark, due to the absence of any of the resistance mechanisms; ipsenol was detected in such males. Extracts of groups of male beetles caught from the dispersing population on pheromone-baited traps, contained little or no ipsenol.  相似文献
