The aim of this study was to evaluate the response of a wide battery of biomarkers in two native species, the freshwater shrimp Palaemonetes argentinus and the macrophyte Potamogeton pusillus, experimentally exposed to zinc in order to establish the potential use of selected species as bioindicators of aquatic pollution. For this purpose, we propose the use of integrated biomarker index (IBR) with a previous selection of biomarkers using boosted regression trees (BRTs) as a new tool in ecotoxicology. Organisms were collected from a reference site, acclimated and exposed at relevant environmental zinc levels (control, 5, 50 and 500 μg Zn L−1) for 96 h. Biomarkers were measured in cephalothorax and abdomen of shrimp as well as in leaf, stem and root of plants.Significant zinc accumulation was observed in cephalothorax of P. argentinus from 50 μg Zn L−1 and from 5 μg Zn L−1 in stem and root of P. pusillus, when compared with control condition. Those effect biomarkers with significant differences among treatments were pre-selected to run out the BRTs model for each species. In P. argentinus, microsomal acetylcholinesterase activity, metallothioneins and superoxide dismutase activity measured in cephalothorax, as well as glutathione reductase activity in abdomen, showed the higher capacity to explain or predict the zinc exposure concentration. On the contrary, in P. pusillus, only chlorophyll a measured in leaf and H2O2 measured in root were the more representative of exposure concentrations, at least, within the biomarkers tested in the present study. Thereafter, IBR was calculated with the selected biomarkers in P. argentinus and showed in a sole value the organism stress, which also correlates with zinc exposure and accumulation.Natives species tested displayed a sensitive response to metal exposure, which represents an important characteristic for biomonitoring programs. Our findings suggest that the BRTs and IBR are useful and robust run tools to select the better biomarkers in toxicological studies and indicate the organism stress.  相似文献

The uptake and accumulation of microcystin-LR (MC-LR) in the shrimp Palaemonetes argentinus was investigated using both laboratory and field assays. Shrimps were exposed in aquarium during 1, 2, 3 and 7 days to 1, 10 and 50 μg L−1 MCLR. Accumulation (0.7 ± 0.2 μg MC-LR g−1) was observed after three days exposures to 50 μg L−1 toxin. Then, shrimps were relocated in fresh water (free of MCLR) to verify the detoxification dynamic, showing a drop to 0.18 ± 0.01 μg MCLR g−1 after three days. The activity of glutathione-S-transferase, measured in the microsomal fraction (mGST), was significantly increased during the exposure period, with further increment during the detoxification period. Furthermore, cytosolic GST (sGST) and glutathione reductase (GR) increased their activities during detoxification, while inhibition was observed for catalase (CAT) with no significant changes for glutathione peroxidase (GPx). Current results suggest that GSH conjugation could be an important MC detoxification mechanism in P. argentinus and that MCLR induce oxidative stress in this shrimp.Field exposures were carried out in San Roque Reservoir (Córdoba, Argentina) after a cyanobacteria bloom. Nodularin (Nod) presence was measured for the first time in this waterbody (0.24 ± 0.04 μg L−1), being the first report of Nod in South America freshwaters. Nod was also detected in Palaemonetes argentinus (0.16 ± 0.03 μg g−1) after three weeks of exposure in this reservoir, with the concomitant activation of mGST, sGST and CAT.Although internal doses of Nod were low throughout the exposure, they were enough to cause biochemical disturbances in Palaemonetes argentinus.Further laboratory studies on Nod accumulation and antioxidant responses in Palaemonetes argentinus are necessary to fully understand these field results. P. argentinus should be considered a potential vector for transferring cyanotoxins to higher trophic levels in aquatic environments.  相似文献

The present work was carried out in greenhouse conditions at the Centro de Investigación en Alimentación y Desarrollo AC in Delicias, Chihuahua, México. Four different concentrations (0, 25, 50 and 100 μM L−1) of Zn chelate and sulfate were used to study the antioxidant system of Phaseolus vulgaris L. Three genes related with antioxidant activity [superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT)] were selected for expression study. Results showed that when Zn chelate at 50 and 100 μM L−1 were applied SOD was repressed and GSH-Px expression was low at 0, 25 and 100 μM L−1 while with sulfate form SOD expression was low and GSH-Px expression was strong in all treatment. CAT was highly expressed in all form and treatments. For a biochemical study the same enzymes were spectrophotometrically measured. SOD activity shows differences in both forms of Zn, chelate form was different at 25, 50 and 100 μM L−1 with less activity at 100 μM L−1 and sulfate treatment shows differences in all concentrations used. GSH-Px activity shows significant differences with sulfate form at 25, 50 μM L−1 where at 50 μM the activity was higher and low at 100 μM L−1, CAT does not exhibit significant differences but with chelate treatment at 50–100 μM L−1 the activity was higher compared to sulfate. Finally, to raise the Zn concentration in bean under biofortification program is a promising strategy in cropping systems in order to increase the ingestion of zinc and antioxidant capacity in the general population and provided the benefits that this element offered in human health.  相似文献

The coastal shrub Limoniastrum monopetalum is capable of growth in soil containing extremely high concentrations of heavy metals. A greenhouse experiment was conducted in order to investigate the effects of a range of Zn concentrations (0–130 mmol l−1) on growth and photosynthetic performance, by measuring relative growth rate, total leaf area, plant height, gas exchange, chlorophyll fluorescence parameters and photosynthetic pigment concentrations. We also determined the total zinc, nitrogen, phosphorus, sulphur, calcium, magnesium, sodium, potassium, iron and copper concentrations in the plant tissues. The study species demonstrated hypertolerance to Zn stress, since survival was recorded with leaf concentrations of up to 1700 mg Zn kg−1 dry mass when treated with 130 mmol Zn l−1. L. monopetalum exhibited little overall effects on photosynthetic function at Zn levels of up to 90 mmol l−1. At greater external Zn concentration, plant growth was negatively affected, due in all probability to the recorded decline in net photosynthetic rate, which may be linked to the adverse effect of the metal on photosynthetic electron transport. Growth parameters were virtually unaffected by leaf tissue concentrations as high as 1400 mg Zn kg−1 dry mass thus indicating that this species could play an important role in the phytoremediation of Zn-polluted areas.  相似文献

This study was carried out to determine the median lethal concentrations (LC50) of Zinc nanoparticles (ZnNPs) on Oreochromis niloticus and Tilapia zillii. The biochemical and molecular potential effects of ZnNPs (500 and 2000 μg L−1) on the antioxidant system in the brain tissue of O. niloticus and T. zillii were investigated. Four hundred fish were used for acute and sub-acute studies. ZnNP LC50 concentrations were investigated in O. niloticus and T. zillii. The effect of 500 and 2000 μg L−1 ZnNPs on brain antioxidants of O. niloticus and T. zillii was investigated. The result indicated that 69 h LC50 was 5.5 ± 0.6 and 5.6 ± 0.4 for O. nilotica and T. zillii, respectively. Fish exposed to 500 μg L−1 ZnNPs showed a significant increase in reduced glutathione (GSH), total glutathione (tGSH) levels, superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR), glutathione peroxidase (GPx) and glutathione-S-transferase (GST) activity and gene expression. On the contrary, malondialdehyde (MDA) levels significantly decreased. Meanwhile, fish exposed to 2000 μg L−1 ZnNPs showed a significant decrease of GSH, tGSH levels, SOD, CAT, GR, GPx and GST activity and gene expression. On the contrary, MDA levels significantly increased. It was concluded that, the 96 h LC50 of ZnNPs was 5.5 ± 0.6 and 5.6 ± 0.4 for O. nilotica and T. zillii, respectively. ZnNPs in exposure concentrations of 2000 μg/L induced a deleterious effect on the brain antioxidant system of O. nilotica and T. zillii. In contrast, ZnNPs in exposure concentrations of 500 μg L−1 produced an inductive effect on the brain antioxidant system of O. nilotica and T. zillii.  相似文献

Marine toxic dinoflagellates of the genus Gambierdiscus are the causative agents of ciguatera fish poisoning (CFP), a form of seafood poisoning that is widespread in tropical, subtropical and temperate regions worldwide. The distributions of Gambierdiscus australes, Gambierdiscus scabrosus and two phylotypes of Gambierdiscus spp. type 2 and type 3 have been reported for the waters surrounding the main island of Japan. To explore the bloom dynamics and the vertical distribution of these Japanese species and phylotypes of Gambierdiscus, the effects of light intensity on their growth were tested, using a photoirradiation-culture system. The relationship between the observed growth rates and light intensity conditions for the four species/phylotypes were formulated at R > 0.92 (p < 0.01) using regression analysis and photosynthesis-light intensity (P-L) model. Based on this equation, the optimum light intensity (Lmax) and the semi-optimum light intensity range (Ls-opt) that resulted in the maximum growth rate (μmax) and ≥80% μ max values of the four species/phylotypes, respectively, were as follows: (1) the Lmax and Ls-opt of G. australes were 208 μmol photons m−2 s−1 and 91–422 μmol photons m−2 s−1, respectively; (2) those of G. scabrosus were 252 and 120–421 μmol photons m−2 s−1, respectively; (3) those of Gambierdiscus sp. type 2 were 192 and 75–430 μmol photons m−2 s−1, respectively; and (4) those of Gambierdiscus sp. type 3 were ≥427 and 73–427 μmol photons m−2 s−1, respectively. All four Gambierdiscus species/phylotypes required approximately 10 μmol photons m−2 s−1 to maintain growth. The light intensities in coastal waters at a site in Tosa Bay were measured vertically at 1 m intervals once per season. The relationships between the observed light intensity and depth were formulated using Beer’s Law. Based on these equations, the range of the attenuation coefficients at Tosa Bay site was determined to be 0.058–0.119 m−1. The values 1700 μmol photons m−2 s−1, 500 μmol photons m−2 s−1, and 200 μmol photons m−2 s−1 were substituted into the equations to estimate the vertical profiles of light intensity at sunny midday, cloudy midday and rainy midday, respectively. Based on the regression equations coupled with the empirically determined attenuation coefficients for each of the four seasons, the ranges of the projected depths of Lmax and Ls-opt for the four Gambierdiscus species/phylotypes under sunny midday conditions, cloudy midday conditions, and rainy midday conditions were 12–38 m and 12–54 m, 1–16 m and 1–33 m, and 0 m and 0–16 m, respectively. These results suggest that light intensity plays an important role in the bloom dynamics and vertical distribution of Gambierdiscus species/phylotypes in Japanese coastal waters.  相似文献

Cloud point extraction (CPE) was used to simultaneously preconcentrate trace-level cadmium, nickel and zinc for determination by flame atomic absorption spectrometry (FAAS). 1-(2-Pyridilazo)-2-naphthol (PAN) was used as a complexing agent, and the metal complexes were extracted from the aqueous phase by the surfactant Triton X-114 ((1,1,3,3-tetramethylbutyl)phenyl-polyethylene glycol). Under optimized complexation and extraction conditions, the limits of detection were 0.37 μg L−1 (Cd), 2.6 μg L−1 (Ni) and 2.3 μg L−1 (Zn). This extraction was quantitative with a preconcentration factor of 30 and enrichment factor estimated to be 42, 40 and 43, respectively. The method was applied to different complex samples, and the accuracy was evaluated by analyzing a water standard reference material (NIST SRM 1643e), yielding results in agreement with the certified values.  相似文献

The efficacy of an essential oil obtained from Pimpinella anisum fruits and its major compound, trans-Anethole, was tested on the eggs, larvae and adults of Culex quinquefasciatus. While causing no significant mortality on eggs, other tested stages were very sensitive to the essential oil and trans-Anethole. LC50 for the 2nd to 4th instar larvae was estimated as 26–27 μL·L 1 and 15–19 μL·L 1 for the essential oil and trans-Anethole, respectively. As for the essential oil applied on adults, LC(LD)50 was estimated as 9.3 μL mL 1 (spray test), 1.9 μL L 1 (fumigation test) and 0.6 μg cm 2 (tarsal test), and for trans-Anethole as 8.1 μL mL 1 (spray test), 2.1 μL L 1 (fumigation test) and 0.4 μg cm 2 (tarsal test). The time needed to achieve 50% mortality after application of LC(LD)99 of the essential oil was significantly different; for example, in larvicidal assays it ranged from 15 to 235 min depending on the larval instar, and from 9 to 180 min when applied to adults, depending on the mode of application. It was also found that temperature had an important effect on the larvicidal efficacy of the essential oil, and oviposition deterrent activity was studied.The essential oil and trans-Anethole were toxic for Daphnia magna (62–92% mortality) and significantly reduced its fertility at high concentrations (35–50 μL mL 1) and long exposure (48 h). However, no negative effect on Daphnia mortality or fertility was found at shorter exposure times (6 h) and/or lower concentrations (20 μL mL 1).Based on the results of this study, we can recommend the essential oil from P. anisum as a suitable active substance for potential botanical insecticides.  相似文献

The aim of the present study was to assess the temporal variation of the heavy metal content (Co, Cu, Fe, Mn, Ni, Pb, and Zn) in surface water and sediments in relation to agricultural practices in the Xanaes River (Córdoba, Argentina). A second objective was to analyze possible relationships between the input of heavy metals on surface water and sediment, heavy metal accumulation and physiological changes in the aquatic plant Myriophyllum aquaticum. Samples were taken from the river at two contrasting sites (between April 2010 and August 2010): (1) a pristine area (mountain site), and (2) an area with intensive agricultural activity located at 60 km down river (agricultural site). The total concentration of heavy metals in surface water was higher in samples collected at the agricultural site but in sediments only the Mn concentration was higher than at the mountain site. The Fe and Mn concentrations in surface water at the agricultural site exceeded the recommended values for Argentinean Legislation of 300 μg L−1 for Fe and 100 μg L−1 for Mn. The accumulations of Zn and Mn in M. aquaticum were higher at the agricultural site and more elevated than the Zn and Mn concentrations in sediments at the same sites and sampling times. At the agricultural site, temporal variations of Cu, Fe and Zn were relatively similar for plants and water column, but the levels of the metals in plants were displaced over time. These results suggest that the levels of pollutants in the river came in pulses from the riverbank. These results show the potential use of M. aquaticum as a suitable accumulation biomonitor at the early stages of heavy metal pollution in rivers.  相似文献

Muscodor cinnamomi was selected and investigated for its in vitro ability to produce indole-3-acetic acid (IAA) to solubilize different toxic metal (Ca, Co, Cd, Cu, Pb, Zn)-containing insoluble minerals and tolerance to metals, herbicides and an insecticide. The results indicated that this fungus is able to produce IAA (45.36 ± 2.40 μg ml−1) in liquid media. This phytohormone stimulated coleoptile elongation, and increased seed germination and root elongation of tested plants. The metal tolerance and solubilizing ability depended on the type of insoluble minerals. M. cinnamomi showed the highest growth tolerance on Ca-containing media at 150 mM, followed by Zn-containing media at 100 mM and Cd-containing media at 10 mM. This fungus tolerated the three herbicides (2,4-d-dimethylammonium, glyphosate and paraquat dichloride) and an insecticide (methomyl) at the recommended dosages for field application. Moreover, M. cinnamomi completely controlled Rhizoctonia solani AG-2 root rot in tomato plants, and increased root length, shoot dry weight and root dry weight. This is the first report of in vitro IAA production, solubilization of insoluble metal minerals, and tolerance to herbicides, an insecticide and metals as well as the plant growth promoting ability of M. cinnamomi.  相似文献

A biosensor for trace metal ions based on horseradish peroxidase (HRP) immobilized on maize tassel-multiwalled carbon nanotube (MT-MWCNT) through electrostatic interactions is described herein. The biosensor was characterized using Fourier transform infrared (FTIR), UV–vis spectrometry, voltammetric and amperometric methods. The FTIR and UV–vis results inferred that HRP was not denatured during its immobilization on MT-MWCNT composite. The biosensing principle was based on the determination of the cathodic responses of the immobilized HRP to H2O2, before and after incubation in trace metal standard solutions. Under optimum conditions, the inhibition rates of trace metals were proportional to their concentrations in the range of 0.092–0.55 mg L−1, 0.068–2 mg L−1 for Pb2+ and Cu2+ respectively. The limits of detection were 2.5 μg L−1 for Pb2+ and 4.2 μg L−1 for Cu2+. Representative Dixon and Cornish-Bowden plots were used to deduce the mode of inhibition induced by the trace metal ions. The inhibition was reversible and mixed for both metal ions. Furthermore, the biosensor showed good stability, selectivity, repeatability and reproducibility.  相似文献

The effects of two anion/Cl? channel inhibitors, Zn2+ and niflumic acid (NA), on seedling photosynthetic and fluorescent parameters of two Glycine soja populations (salt-tolerant BB52; salt-sensitive N23227) and Glycine max cultivar (salt-tolerant Lee68) were studied and compared under salt stress. Treatments with Zn2+ and NA only (10, 20 μmol L?1) were also imposed for comparisons. Results showed that, there were non-toxic and non-nutritional effects of Zn2+ and NA treatments alone on seed germination and seedling growth of soybeans. Under 150 mmol L?1 NaCl for 6 d, leaf chlorophyll and carotenoid contents, net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO2 concentration (Ci), transpiration rate (Tr), and the maximum photochemical efficiency of photosystem II (PS II) (Fv/Fm) except the stomatal limitation (Ls) significantly decreased in three kinds of soybean seedlings when compared with their control plants. The NaCl stress plus additional 20 μmol L?1 Zn showed an obvious enhancement of leaf chlorophyll and carotenoid contents, Pn, Gs, Ci and Tr, especially for the G. max cultivar Lee68, but the supplementation of 20 μmol L?1 NA showed the reverse effects.  相似文献

In this study, zinc (Zn) and cadmium (Cd) tolerance, accumulation and distribution was conducted in Potentilla griffithii H., which has been identified as a new Zn hyperaccumulator found in China. Plants were grown hydroponically with different levels of Zn2+ (20, 40, 80 and 160 mg L?1) and Cd2+ (5, 10, 20 and 40 mg L?1) for 60 days. All plants grew healthy and attained more biomass than the control, except 40 mg L?1 Cd treatment. Zn or Cd concentration in plants increased steadily with the increasing addition of Zn or Cd in solution. The maximum metal concentrations in roots, petioles and leaves were 14,060, 19,600 and 11,400 mg kg?1 Zn dry weight (DW) at 160 mg L?1 Zn treatment, and 9098, 3077 and 852 mg kg?1 Cd DW at 40 mg L?1 Cd treatment, respectively. These results suggest that P. griffithii has a high ability to tolerate and accumulate Cd and Zn, and it can be considered not only as Zn but also as a potential cadmium hyperaccumulator. Light microscope (LM) with histochemical method, scanning electron microscope combined with energy dispersive spectrometry (SEM-EDS) and transmission electron microscope (TEM) were used to determine the distribution of Zn and Cd in P. griffithii at tissue and cellular levels. In roots, SEM-EDS confirmed that the highest Zn concentration was found in xylem parenchyma cells and epidermal cells, while for Cd, a gradient was observed with the highest Cd concentration in rhizodermal and cortex cells, followed by central cylinder. LM results showed that Zn and Cd distributed mainly along the walls of epidermis, cortex, endodermis and some xylem parenchyma. In leaves, Zn and Cd shared the similar distribution pattern, and both were mostly accumulated in epidermis and bundle sheath. However, in leaves of 40 mg L?1 Cd treatment, which caused the phytotoxicity, Cd was also found in the mesophyll cells. The major storage site for Zn and Cd in leaves of P. griffithii was vacuoles, to a lesser extent cell wall or cytosol. The present study demonstrates that the predominant sequestration of Zn and Cd in cell walls of roots and in vacuoles of epidermis and bundle sheath of leaves may play a major role in strong tolerance and hyperaccumulation of Zn and Cd in P. griffithii.  相似文献

Benthic dinoflagellates of the genus Ostreopsis are found all over the world in temperate, subtropical, and tropical coastal regions. Our recent studies revealed that a putative “cryptic” species of Ostreopsis ovata is present widely along Japanese coasts. This organism, Ostreopsis sp. 1, possesses palytoxin analogs and thus its toxic blooms may be responsible for potential toxification of marine organisms. To evaluate the bloom dynamics of Ostreopsis sp. 1, the present study examined the growth responses of Ostreopsis sp. 1 strain s0716 to various light intensities (photon flux densities: μmol photons m−2 s−1) using a newly devised photoirradiation-culture system. This novel system has white light-emitting diodes (LEDs) capable of more closely simulating the wavelength spectrum of light entering the oceanic water column than do fluorescent tubes and halogen lamps. In this system, the light intensity of the white LEDs was reduced through two polarizing filters by varying the rotation angles of the filters. Thereby, the new system was capable of culturing microalgae under well-controlled light intensity conditions. Ostreopsis sp. 1 grew proportionally when light intensity was increased from 49.5 to 199 μmol photons m−2 s−1, but its growth appeared to be inhibited slightly at ≥263 μmol photons m−2 s−1. The relationship between observed growth rates and light intensity was calculated at R > 0.99 (P < 0.01) using a regression analysis with a modified equation of the photosynthesis-light intensity (P-L) model. The equation determined the critical light intensities for growth of Ostreopsis sp. 1 and the organism's growth potential as follows: (1) the threshold light intensity for growth: 29.8 μmol photons m−2 s−1; (2) the optimum light intensity (Lm) giving the maximum growth rate (μmax = 0.659 divisions day−1): 196 μmol photons m−2 s−1; (3) the optimum light intensity range (Lopt) giving ≥95% μmax: 130–330 μmol photons m−2 s−1; (4) the semi-optimum range (Lsopt) giving ≥80% μmax: 90 to over 460 μmol photons m−2 s−1. The Lsopt represents 4.5–23% ambient light intensity present in surface waters off of a temperate region of the Japanese coast, Tosa Bay; putatively, this semi-optimum range of light intensity appears at depth of 12.9–27.8 m. Considering these issues, our data indicate that Ostreopsis sp. 1 in coastal environments may form blooms at ca. ∼28 m depth in regions along Japanese coasts.  相似文献

Conidiation and lytic enzyme production by Trichoderma viride at different solids concentration of pre-treated municipal wastewater sludge was examined in a 15-L fermenter. The maximum conidia concentration (5.94 × 107 CFU mL−1 at 96 h) was obtained at 30 g L−1 suspended solids. The maximum lytic enzyme activities were achieved around 12–30 h of fermentation. Bioassay against a fungal phytopathogen, Fusarium sp. showed maximum activity in the sample drawn around 96 h of fermentation at 30 g L−1 suspended solids concentration. Entomotoxicity against spruce budworm larvae showed maximum value ≈17290 SBU μL−1 at 30 g L−1 suspended solids concentration at the end of fermentation (96 h). Plant bioassay showed dual action of T. viride, i.e., disease prevention and growth promotion. The rheological analyses of fermentation sludges showed the pseudoplastic behaviour. In order to maintain required dissolved oxygen concentration ≥30%, the agitation and aeration requirements significantly increased at 35 g L−1 compared to 30 and 25 g L−1. The oxygen uptake rate and volumetric oxygen mass transfer coefficient, kLa at 35 g L−1 did not increase in comparison to 30 g L−1 due to rheological complexity of the broth during fermentation. Thus, the successful fermentation operation of the biocontrol fungus T. viride is a rational indication of its potential for mass-scale production for agriculture and forest sector as a biocontrol agent.  相似文献

In culture, Gambierdiscus spp. have been shown to prefer irradiances that are relatively low (≤250 μmol photons m−2 s−1) versus those to which they are frequently exposed to in their natural environment (>500 μmol photons m−2 s−1). Although several behavioral strategies for coping with such irradiances have been suggested, it is unclear as to how these dinoflagellates do so on a physiological level. More specifically, how do long term exposures (30 days) affect cell size and cellular chlorophyll content, and what is the photosynthetic response to short term, high irradiance exposures (up to 1464 μmol photons m−2 s−1)? The results of this study reveal that cell size and chlorophyll content exhibited by G. carolinianus increased with acclimation to increasing photon flux density. Additionally, both G. carolinianus and G. silvae exhibited reduced photosynthetic efficiency when acclimated to increased photon flux density. Photosynthetic yield exhibited by G. silvae was greater than that for G. carolinianus across all acclimation irradiances. Bisexual populations of the charophyte Chara canescens (Desv. et Loisel. in Loisel., 1810) containing male and female individuals are rarely found. Two experiments were carried out to study whether male and female algae from the same site exhibit different physiological capacities, especially with respect to light acclimation.Algae from two different shore levels and from laboratory cultures acclimated to six irradiance conditions (35–500 μmol photons m−2 s−1) were compared. Field measurements showed that both female and male algae of C. canescens are able to acclimate to daily changes in solar irradiance. The quantum yield of Photosystem II (PSII) decreased with increasing irradiance in the morning and increased with decreasing irradiance in the afternoon. Growth experiments showed increasing growth rates from 35 μmol photons m−2 s−1 (∼7 mg FW) up to 500 μmol photons m−2 s−1 (∼27 mg FW) in female and male C. canescens. The irradiance saturation point for photosynthesis (Ek) was about 140 μmol m−2 s−1 for both sexes within the whole range of acclimation irradiances. The maximum photosynthesis rate at saturating irradiances (Pmax) of male algae was highest at Ek, whereas Pmax of female algae was highest at 500 μmol photons m−2 s−1. The photosynthetic efficiency in the light-limited range (α) increased in female C. canescens and decreased in male C. canescens. The ratio of the non-photochemical quenching parameter (NPQ) to the relative electron transport rates rETR(MT) increased in both sexes with irradiance, but showed a steeper increase in male than in female algae. Pigment analysis showed similar acclimation pattern for male and female C. canescens. Chl a/Chl b ratios of both sexes were constant over the whole range of Eg, whereas Chl a/carotenoid ratios in male and female C. canescens decreased from 70 μmol photons m−2 s−1 upwards. Pigment analysis pointed out that the carotenes α-, β- and γ-carotene were more prominent in male than in female algae.Our results indicate that female C. canescens are more efficient in light acclimation than male algae from the same site. Nevertheless, further investigations of bisexual C. canescens populations resolving CO2-uptake mechanisms and/or genetic differences are needed.  相似文献

Pot experiments were performed to evaluate the phytoremediation capacity of plants of Atriplex halimus grown in contaminated mine soils and to investigate the effects of organic amendments on the metal bioavailability and uptake of these metals by plants. Soil samples collected from abandoned mine sites north of Madrid (Spain) were mixed with 0, 30 and 60 Mg ha−1 of two organic amendments, with different pH and nutrients content: pine-bark compost and horse- and sheep-manure compost. The increasing soil organic matter content and pH by the application of manure amendment reduced metal bioavailability in soil stabilising them. The proportion of Cu in the most bioavailable fractions (sum of the water-soluble, exchangeable, acid-soluble and Fe–Mn oxides fractions) decreased with the addition of 60 Mg ha−1 of manure from 62% to 52% in one of the soils studied and from 50% to 30% in the other. This amendment also reduced Zn proportion in water-soluble and exchangeable fractions from 17% to 13% in one of the soils. Manure decreased metal concentrations in shoots of A. halimus, from 97 to 35 mg kg−1 of Cu, from 211 to 98 mg kg−1 of Zn and from 1.4 to 0.6 mg kg−1 of Cd. In these treatments there was a higher plant growth due to the lower metal toxicity and the improvement of nutrients content in soil. This higher growth resulted in a higher total metal accumulation in plant biomass and therefore in a greater amount of metals removed from soil, so manure could be useful for phytoextraction purposes. This amendment increased metal accumulation in shoots from 37 to 138 mg pot−1 of Cu, from 299 to 445 mg pot−1 of Zn and from 1.8 to 3.7 mg pot−1 of Cd. Pine bark amendment did not significantly alter metal availability and its uptake by plants. Plants of A. halimus managed to reduce total Zn concentration in one of the soils from 146 to 130 mg kg−1, but its phytoextraction capacity was insufficient to remediate contaminated soils in the short-to-medium term. However, A. halimus could be, in combination with manure amendment, appropriate for the phytostabilization of metals in mine soils.  相似文献

α-Glucuronidase (EC 3.2.1.139) of family GH 115 from Scheffersomyces stipitis is a valuable enzyme for the modification of water-soluble xylan into insoluble biopolymers, due to its unique ability to act on polymeric xylans. The influence of growth rate on the production of α-glucuronidase by recombinant Saccharomyces cerevisiae MH1000pbk10D-glu in glucose-limited fed-batch culture was studied at 14 and 100 L scale. At and below the critical specific growth rate (μcrit) of 0.12 h−1 at 14 L scale, the biomass yield coefficient (Yx/s) remained constant at 0.4 g g−1 with no ethanol production, whereas ethanol yields relative to biomass (keth/x) of up to 0.54 g g−1 and a steady decrease in Yx/s were observed at μ > 0.12 h−1. Production of α-glucuronidase was growth associated at a product yield (kα-glu/x) of 0.45 mg g−1, with the highest biomass (37.35 g/L) and α-glucuronidase (14.03 mg/L) concentrations, were recorded during fed-batch culture at or near to μcrit. Scale-up with constant kLa from 14 to 100 L resulted in ethanol concentrations of up to 2.5 g/L at μ = 0.12 h−1. At this scale, α-glucuronidase yield could be maximised at growth rates below μcrit, to prevent localised high glucose concentration pockets at the feed entry zone that would induce oxido-reductive metabolism. This is the first report where recombinant production of α-glucuronidase (EC 3.2.1.139) by S. cerevisiae was optimised for application at pilot scale.  相似文献

Brazilian filamentous fungi Rhizopus sp. (SIS-31), Aspergillus sp. (SIS-18) and Penicillium sp. (SIS-21), sources of oxidases were isolated from Caatinga's soils and applied during the in situ cathodic oxygen reduction in fuel cells. All strains were cultivated in submerged cultures using an optimized saline medium enriched with 10 g L−1 of glucose, 3.0 g L−1 of peptone and 0.0005 g L−1 of CuSO4 as enzyme inducer. Parameters of oxidase activity, glucose consumption and microbial growth were evaluated. In-cell experiments evaluated by chronoamperometry were performed and two different electrode compositions were also compared. Maximum current densities of 125.7, 98.7 and 11.5 μA cm−2 were observed before 24 h and coulombic efficiencies of 56.5, 46.5 and 23.8% were obtained for SIS-31, SIS-21 and SIS-18, respectively. Conversely, maximum power outputs of 328.73, 288.80 and 197.77 mW m−3 were observed for SIS-18, SIS-21 and SIS-31, respectively. This work provides the primary experimental evidences that fungi isolated from the Caatinga region in Brazil can serve as efficient biocatalysts during the oxygen reduction in air-cathodes to improve electricity generation in MFCs.  相似文献
