1. Experiments were carried out to decide whether a homogeneous phase (solubility) theory or a micellar-structural theory more adequately describes the behavior of dried collodion membranes with solutions of strong electrolytes. 2. A number of dried collodion membranes were prepared from an electrochemically inactive collodion preparation (state I); the characteristic concentration potentials across them were low, about 30 mv. The membranes were activated by oxidation (state II) to give maximum or nearly maximum concentration potentials (about 50 mv.). The oxidized membranes are dried, dissolved in alcohol-ether, and a new set of dry collodion membranes prepared from this solution (state III). The concentration potentials across these membranes are low. 3. Since the properties of a homogeneous phase should not be influenced by a rearrangement of its constituent particles, the experimental results do not support a homogeneous phase (solubility) theory, but they agree with the predictions of the micellar-structural theory. The characteristic behavior of dried collodion membranes in solutions of strong inorganic electrolytes is therefore due to the micellar character of its interstices.  相似文献

1. Experiments were carried out to decide whether or not the electromotive properties of dried collodion membranes depend upon their thickness. 2. A number of dried collodion membranes of varying thickness, 3–160 µ, were prepared from collodion preparations of different electrochemical activity. The characteristic concentration potentials across them were measured and the means of these values determined for each thickness. 3. The characteristic concentration potentials across dried collodion membranes are a function of their thickness. The thinnest membranes yield in all cases the lowest concentration potentials; increasingly thicker membranes give increasingly higher potential values, until a constant value is reached which is characteristic of the particular collodion preparation used. With electrochemically active collodion, characteristic concentration potentials approaching the thermodynamically possible maximum are obtained with membranes of only 10 µ thickness, thinner membranes giving appreciably lower values. With two rather inactive commercial collodion preparations the characteristic concentration potential increases from about 30 mv. for membranes 3 µ thick to about 42 mv. for 20 µ membranes; still thicker membranes do not show a significant increase in the potential values. With a highly purified collodion preparation the constant maximum value was found to be about 32 mv., 4 µ thick membranes giving only about 22 mv. 4. These results do not support the homogeneous phase theory as applied to the dried collodion membrane. They are readily compatible with the micellar-structural theory. Several special possible cases of the latter as applied to the dried collodion membrane are discussed.  相似文献

Removing cultures from roller tubes before staining eliminates the destaining which often occurs when the cells are first stained and then removed by embedding in collodion. The cells are fixed in situ, dehydrated, and covered with collodion (Merk's flexible) for 10 min. The collodion is poured off, the fluid residue lining the tube allowed to dry for 10 min, and the tube is filled with tap water. The collodion cast containing the cells is loosened and removed, cut into strips, placed on slides and blotted into firm contact. The collodion is then dehydrated and dissolved with absolute alcohol followed by a 1:1 mixture of alcohol and ether. The slides can then be rehydrated and stained by conventional methods.  相似文献

Summary Select medium and substratum conditions were investigated for their effects on semiconservative DNA synthesis in essentially pure primary cultures of bile ductular epithelial cells that were initially isolated from cholestatic rat livers at 6 to 10 wk after bile duct ligation. DNA synthesis in these cultured cells was serum-dependent, being at its highest level when the concentration of fetal bovine serum present in the medium was maintained at 10%. This serum-dependent DNA synthesis was completely inhibited when 10 mM hydroxyurea was also included in the medium, and bile ductular cells cultured in the continued presence of 1.0% fetal bovine serum showed only marginal DNA synthesis during 8 to 10 d of primary culture when compared with no-serum controls. Maximum rates of serum-dependent DNA synthesis were obtained when the bile ductular cells were cultured for 7 to 14 d on tissue culture plastic coated with obtained when the bile ductular cells were cultured for 7 to 14 d on tissue culture plastic coated with either fibronectin from bovine plasma or type I rat-tail collagen. Cells cultured on plastic coated with basement membrane Matrigel exhibited the lowest levels of DNA synthesis, whereas those on plastic alone had intermediate amounts. Furthermore, the addition of epidermal growth factor (50 ng·ml−1·d−1) to medium supplemented with 1.0% fetal bovine serum greatly enhanced the rate of DNA synthesis in bile ductular cells after 6 d in primary culture on type I collagen-coated plastic over that measured in solvent control cultures. These findings indicate that our bile ductular epithelial cell culture model is potentially useful in the study of biliary cell growth regulation and carcinogenesis. The objective of this study was to establish a novel surgical model for the grafting of cultured epidermal sheets in mice in order to optimize studies on the behavior of these grafts. Graft-related skin immunology and wound-healing studies also would benefit from this adapted surgical approach. Adapted tie-over surgical procedures were established for mice and promptly applied. Early-stage observation of grafts was made possible by replacing the cotton dressing with a saddle-like plastic tube dressing with a screw cap. We grafted normal Balb/c mice and athymic nude (nu/nu) mice with cultured human epidermis. Evaluation of graft rejection was carried out with the first group, whereas the second provided information on epidermal stratification and terminal differentiation. This innovation permitted direct evaluation of the grafted tissue at any time. Advances in applied transplantation research will certainly provide additional tools for human applications.  相似文献

1. It is shown that collodion membranes which have received one treatment with a 1 per cent gelatin solution show for a long time (if not permanently) afterwards a different osmotic behavior from collodion membranes not treated with gelatin. This difference shows itself only towards solutions of those electrolytes which have a tendency to induce a negative electrification of the water particles diffusing through the membrane, namely solutions of acids, acid salts, and of salts with trivalent and tetravalent cations; while the osmotic behavior of the two types of membranes towards solutions of salts and alkalies, which induce a positive electrification of the water particles diffusing through the membrane, is the same. 2. When we separate solutions of salts with trivalent cation, e.g. LaCl3 or AlCl3, from pure water by a collodion membrane treated with gelatin, water diffuses rapidly into the solution; while no water diffuses into the solution when the collodion membrane has received no gelatin treatment. 3. When we separate solutions of acid from pure water by a membrane previously treated with gelatin, negative osmosis occurs; i.e., practically no water can diffuse into the solution, while the molecules of solution and some water diffuse out. When we separate solutions of acid from pure water by collodion membranes not treated with gelatin, positive osmosis will occur; i.e., water will diffuse rapidly into the solution and the more rapidly the higher the valency of the anion. 4. These differences occur only in that range of concentrations of electrolytes inside of which the forces determining the rate of diffusion of water through the membrane are predominantly electrical; i.e., in concentrations from 0 to about M/16. For higher concentrations of the same electrolytes, where the forces determining the rate of diffusion are molecular, the osmotic behavior of the two types of membranes is essentially the same. 5. The differences in the osmotic behavior of the two types of membranes are not due to differences in the permeability of the membranes for solutes since it is shown that acids diffuse with the same rate through both kinds of membranes. 6. It is shown that the differences in the osmotic behavior of the two types of collodion membranes towards solutions of acids and of salts with trivalent cation are due to the fact that in the presence of these electrolytes water diffuses in the form of negatively charged particles through the membranes previously treated with gelatin, and in the form of positively charged particles through collodion membranes not treated with gelatin. 7. A treatment of the collodion membranes with casein, egg albumin, blood albumin, or edestin affects the behavior of the membrane towards salts with trivalent or tetravalent cations and towards acids in the same way as does a treatment with gelatin; while a treatment of the membranes with peptone prepared from egg albumin, with alanine, or with starch has no such effect.  相似文献

The electrochemical behavior of membranes prepared from commercial collodion preparations varies widely, some preparations showing very high, other ones very low electrochemical efficiency ("activity"). 2. The electrochemical activity of a collodion membrane depends entirely upon impurities of an acidic nature contained in the collodion used for casting the membrane. 3. The active acidic impurities are substantially due to partial oxidation which occurs in the manufacturing process. Sulfuric acid compounds; e.g., acid sulfuric acid esters play only a minor rôle, if any. 4. The electrochemical behavior of collodion membranes in solutions of strong electrolytes is decisively dependent upon the acidic groups built permanently into the collodion surfaces. 1. The electrochemical behavior ("activity") of collodion membranes depends upon acidic, dissociable groups located in the interstices of the membranes. The active groups can be determined by base exchange measurements. High base exchange capacity is always found with preparations of great "electrochemical activity;" medium and low base exchange capacities occur with electrochemically active as well as with inactive preparations. The observed base exchange capacity is determined by two factors: the inherent acidity of the collodion (its mean equivalent weight) and the submicroscopic micellar structure of the collodion. A comparison of the base exchange capacity of various collodion preparations and their inherent acidities therefore allows certain conclusions to be drawn concerning the relative availability of the micellar surfaces in the different preparations. 2. The inherent acidity of various collodion preparations, their "acid number," was determined by electrometric titration. Collodion in the acidic state, i.e. after exchange of all other cations for H+ ions, was titrated in an organic solvent mixture with alcoholic KOH using a quinhydrone electrode. Details of the experimental procedure are given in the paper. The acid numbers, expressed in milliliters of 0.01 N KOH per gram dry collodion, vary from 1.0 for a highly purified collodion preparation of very low electrochemical activity to 3.3 for a highly oxidized sample of very high activity. Acid numbers of about 1.5 (corresponding to an equivalent weight of about 67,000) are found both with inactive commercial and with fairly active oxidized preparations. The base exchange capacity of the same preparations in the fibrous state as measured after 48 hours of exchange time varies from 0.0013 ml. 0.01 N NaOH per gm. dry collodion for the most inactive preparation up to 0.26 ml. 0.01 N NaOH per gm. for the most active preparation. Thus the acid numbers over the whole range investigated differ only in the ratio of 1:3.3, whereas the base exchange values differ in the range of 1:200. 3. In the inactive preparation only one in 770 acid groups is available for base exchange, in the most active collodion one group in 13; values between these extremes are found with commercial and alcohol purified oxidized preparations. 4. The high base exchange capacity of the electrochemically active preparations is not so much due to their higher acid number as to their more open structure. This difference in structure is ascribed to the presence of a small fraction of low molecular weight material which inhibits normal formation and arrangement of the micelles. 5. Short time base exchange experiments with fibrous collodion indicate that the number of acid groups available for the typical electrochemical membrane functions may be estimated to be about 50 to 1000 times less numerous than those found in the 48 hour base exchange experiments. It is estimated that in membranes prepared even from the most active collodion not more than one in 500 acid groups may be available for the typical membrane functions; with the less active preparations this ratio is estimated to be as high as one in 1,000,000 or more.  相似文献

Lamellar ichthyosis (LI, MIM# 242300) is a severe autosomal recessive genodermatosis present at birth in the form of collodion membrane covering the neonate. Mutations in the TGM1 gene encoding transglutaminase-1 are a major cause of LI. In this study molecular analysis of two LI Tunisian patients revealed a common nonsense c.788G>A mutation in TGM1 gene. The identification of a cluster of LI pedigrees carrying the c.788G>A mutation in a specific area raises the question of the origin of this mutation from a common ancestor. We carried out a haplotype-based analysis by way of genotyping 4 microsatellite markers and 8 SNPs flanking and within the TGM1 gene spanning a region of 6 Mb. Haplotype reconstruction from genotypes of all members of the affected pedigrees indicated that all carriers for the mutation c.788G>A harbored the same haplotype, indicating common ancestor. The finding of a founder effect in a rare disease is essential for the genetic diagnosis and the genetic counselling of affected LI pedigrees in Tunisia.  相似文献

Bone cells in vivo exist in direct contact with extracellular matrix, which regulates their basic biological processes including metabolism, development, growth and differentiation. Thus, the in vitro activity of cells cultured on tissue culture treated plastic could be different from the activity of cells cultured on their natural substrate. We selected MC3T3-E1 pre-osteoblastic cells to study the effect of extracellular matrix on cell proliferation because these cells undergo a progressive developmental sequence of proliferation and differentiation. MC3T3-E1 cells were cultured on plastic or plastic coated with ECM, fibronectin, collagen type I, BSA or poly l-lysine and their ability to proliferate was assessed by incorporation of [3H]dT or by enumeration of cells. Our results show that (1) ECM inhibits incorporation of [3H]dT by MC3T3-E1 cells; (2) collagen type I, but not BSA, poly l-lysine or fibronectin also inhibits incorporation of [3H]dT; (3) the level of ECM inhibition of [3H]dT incorporation is directly related to the number of cells cultured, but unrelated to the cell cycle distribution or endogenous thymidine content; (4) the kinetic profile of [3H]dT uptake suggest that ECM inhibits transport of [3H]dT from the extracellular medium, and (5) cell counts are similar in cultures whether cells are grown on plastic or ECM. These results suggest that decreased incorporation of [3H]dT by cells cultured on ECM is not reflective of bone cell proliferation.  相似文献

为了使牛体外受精卵能通过体外早期发育阻滞期,我们建立了卵丘细胞单层(A)和输卵管上皮单层(B)两种共同培养系统。A1共同培养实验是在本实验室进行的,A2和B共同培养实验均在日本岗山大学农学部动物繁殖学研究室进行,牛输卵管组织的分离使用0.76%EDTA—PBS溶液,共同培养系统均使用含10%小牛血清的TCM—199(Earle's salts)作为培养液。培养的卵泡卵母细胞体外成熟率为100%,体外受精率为99—100%。在A1共同培养实验中,越过阻滞期发育到16—细胞以上的胚胎占卵裂胚的35.7%,与A2共同培养实验中越过阻滞期的发育率(40.1%)无显著差异(P<0.05)。A1和A2共同培养实验,在卵裂基础上得到的桑椹胚和囊胚发育率分别为23.7%和27.9%。每百枚培养的卵母细胞,在A1共同培养实验中可获得桑椹胚和囊胚15.1枚,在A2共同培养实验中可获桑椹胚和囊胚的20.5枚。B共同培养实验中桑椹胚和囊胚发育率为54.1%,显著高于A1或A2共同培养实验的相应发育率(P<0.001),使用B共同培养系统每百枚培养的卵母细胞可以获得37枚桑椹胚和囊胚。  相似文献

Lipid-impregnated collodion (nitrocellulose) films have been frequently used as a fusion substrate in the measurement and analysis of electrogenic activity in biological membranes and proteoliposomes. While the method of fusion of biological membranes or proteoliposomes with such films has found a wide application, little is known about the structures formed after the fusion. Yet, knowledge of this structure is important for the interpretation of the measured electric potential. To characterize structures formed after fusion of membrane vesicles (chromatophores) from the purple bacterium Rhodobacter sphaeroides with lipid-impregnated collodion films, we used near-field scanning optical microscopy. It is shown here that structures formed from chromatophores on the collodion film can be distinguished from the lipid-impregnated background by measuring the fluorescence originating either from endogenous fluorophores of the chromatophores or from fluorescent dyes trapped inside the chromatophores. The structures formed after fusion of chromatophores to the collodion film look like isolated (or sometimes aggregated, depending on the conditions) blisters, with diameters ranging from 0.3 to 10 microm (average approximately 1 microm) and heights from 0.01 to 1 microm (average approximately 0.03 microm). These large sizes indicate that the blisters are formed by the fusion of many chromatophores. Results with dyes trapped inside chromatophores reveal that chromatophores fused with lipid-impregnated films retain a distinct internal water phase.  相似文献

The nuchal ligament of unborn calves contains a neutral endopeptidase that is biochemically and immunologically similar to the neutral endopeptidase (NEP), or enkephalinase, from human kidney. Enzymatic activity was inhibited more than 90% by phosphoramidon (1 microM). The specific activity in membrane fractions, as determined by hydrolysis of the dansylated substrate, DAPGN, was similar in tissue from fetuses of gestational ages ranging from 100 to 280 days. NEP activity in adult ligament tissue, however, was less than 10% of that in fetal tissue. Fibroblasts dissociated from ligament tissue by collagenase displayed less NEP activity than did preparations of intact ligament, and activity was even lower in cultured cells. By contrast, fibroblasts cultured from fetal calf lungs had NEP activity comparable to that in the ligament tissue. When ligament fibroblasts were cultured on subcellular matrices derived from fetal lung fibroblasts the NEP activity increased relative to those cultured on plastic alone. These studies confirm the presence of neutral endopeptidase (NEP) in the nuchal ligament of the fetal calf. The consistent activity through a range of gestational ages and the influence of the subcellular matrix suggest that this enzyme might be involved in growth of the ligament during fetal life.  相似文献

Rat small intestinal epithelial cells and human colon adenocarcinoma cells cultured on Matrigel expressed the differentiation specific enzyme, sucrase-isomaltase, as determined by indirect immunofluorescence. Rat small intestinal epithelial cells, rat colonocytes, and human colon adenocarcinoma cells developed an altered morphology when cultured on Matrigel and became apoptotic within 24-48 h. Benzo[a]pyrene and 2,3,7,8-tetrachlorodibenzo-p-dioxin caused a 2- and 5-fold induction, respectively, of ethoxyresorufin-o-deethylase activity in rat small intestinal epithelial cells cultured on Matrigel. Benzo[a]pyrene- or 2,3,7,8-tetrachlorodibenzo-p-dioxin-induced ethoxyresorufin-o-deethylase activity in rat small intestinal epithelial cells cultured on plastic was not detected. 2,3,7,8-tetrachlorodibenzo-p-dioxin treatment caused a 14-fold induction of transfected, rat CYP1A1-promoter-luciferase activity in rat small intestinal epithelial cells cultured on Matrigel. Benzo[a]pyrene and 2,3,7,8-tetrachlorodibenzo-p-dioxin treatment induced ethoxyresorufin-o-deethylase activity by 6- and 1.6-fold, respectively in rat colonocytes cultured on Matrigel. Induction of ethoxyresorufin-o-deethylase activity was not observed in rat colonocytes cultured on plastic. CYP1A1-promoter-luciferase activity was induced 3-fold by 2,3,7,8-tetrachlorodibenzo-p-dioxin in rat colonocytes cultured on Matrigel. Induction of CYP1A1-promoter-luciferase activity in rat small intestinal epithelial cells or rat colonocytes cultured on plastic was not observed. Ethoxyresorufin-o-deethylase activity in human colon adenocarcinoma cells, cultured on either plastic or Matrigel, was induced 7-fold by benzo[a]pyrene. 2,3,7,8-Tetrachlorodibenzo-p-dioxin-induced ethoxyresorufin-o-deethylase activity was 2-fold greater in human colon adenocarcinoma cells cultured on Matrigel compared to cells cultured on plastic. Extracellular matrix-mediated differentiation and apoptosis of intestinal cells provide in vitro systems for study of the regulation of CYP1A1 expression, carcinogen activation in the gut and mechanism(s) of apoptosis of colon cancer cells.  相似文献

Stem explants, excised from greenhouse-grown Begonia rex plants, were cultured on basal medium (T. Murashige and F. Skoog, Physiol. Plant. 15: 473–497, 1962) contained in sterile Petri dishes. The medium was supplemented with benzyladenine (0.1 mg 1−1) naphthaleneacetic acid (0.01 mg 1−1) and, according to experimental requirements, with either sucrose (3%) or mannitol (3%). Histochemical and biochemical examination of the starch content of the explant was carried out over several days. There was no starch deposition or organogenesis in tissue cultured on mannitol and carbohydrate-free growth medium. The most dramatic finding was the heavy accumulation of starch in tissue cultured on sucrose medium. This copious accumulation preceded any organ formation and was mainly in regions which ultimately gave rise to shoot primordia. The heavy build-up of starch preceding organogenesis was also observed when explants previously cultured on mannitol medium were transferred to medium containing sucrose. During shoot primordia development there was a decrease in the starch content of the cultured tissue indicating the utilization of the polyglucan in the organogenic process.  相似文献

A 1% collodion solution in a 1:1 mixture of 95% alcohol and ether can be used to fix smears and to form a protective coating on them. Such coating permits transportation of dry slides over long distances to a laboratory for staining and diagnosis. The collodion solution can be used several times provided that it is filtered after each specimen is coated. With urine sediments, the application of the coating keeps a large number of cells on the slide, hence facilitates a thorough examination and more reliable diagnosis. Papanicolaou's method is recommended for staining the coated specimens, especially those for the diagnosis of cancer.  相似文献
