木糖还原酶是重组酿酒酵母工程菌利用木糖生成乙醇代谢途径中的关键酶, 该关键酶在利用木糖时依赖NADPH而不是NADH是导致酿酒酵母代谢木糖生成乙醇的最终产率低的主要原因之一。为了改变树干毕赤氏酵母木糖还原酶的辅酶依赖性, 对它的第21位赖基酸Lys进行了突变。利用质粒载体pET28b分别将突变后的基因K21A-XYL1、K21R-XYL1及野生基因WT-XYL1在大肠杆菌E. coli BL21(DE3)中进行表达, 表达后的蛋白经His-Tag纯化柱纯化后测定酶学性质。结果表明: K21R突变子的辅酶依赖性没有改变, 但K21A突变子的辅酶依赖性由NADPH完全逆转为NADH。  相似文献

毕赤酵母木糖还原酶定点突变改善其对双辅酶的亲和力   总被引:1,自引:0,他引:1  
通过毕赤酵母(Pichia stipitis)木糖还原酶(xylose reductase, XR)基因定点突变,获得NADH高亲和力的毕赤酵母木糖还原酶(PsXR),改善了辅酶不同而导致的酿酒酵母胞内氧化还原失衡.同时克隆了PsXR编码基因,通过BLAST工具进行同源性搜索,并用生物软件进行序列比对和结构分析,确定突变位点.用融合PCR方法进行定点突变,并在大肠杆菌表达系统中进行融合表达,且对表达产物进行HIS-TAG亲和纯化,分光光度法检测酶活性,计算比活力.本研究成功获得突变XR编码基因,并收集了纯化的突变蛋白.酶活性检测和比活力计算显示,3种突变酶对2种辅酶的亲和力在一定程度上都发生了变化.与未突变的PsXR相比,3种突变酶对辅酶NADPH的亲和力均显著下降,突变酶M3对辅酶NADH的亲和力未发生变化,而突变酶M1和M4对辅酶NADH的亲和力显著升高,其中突变酶M1对NADH的亲和力明显提高,对NADPH的亲和力明显下降,其活性主要依赖辅酶NADH,提示K270R位点在XR与辅酶结合中起关键作用.  相似文献

在导入表达毕赤酵母(Pichia stipitis)木糖还原酶(xylose reductase,XR)和木糖醇脱氢酶(xylitol dehydrogenase,XDH)基因的重组酿酒酵母中,木糖还原酶活性主要依赖辅酶NADPH,木糖醇脱氢酶活性依赖辅酶 NAD+,两者的辅助因子不同导致细胞内电子氧化还原的不平衡,是造成木糖醇积累,影响木糖代谢和乙醇产量的主要原因之一.将经过基因工程改造获得的NADH高亲和力的木糖还原酶突变基因m1,与毕赤酵母木糖醇脱氢酶(PsXDH)基因xyl2共转染酿酒酵母AH109,以转染毕赤酵母木糖还原酶(PsXR)基因xyl1和xyl2重组质粒的酵母细胞为对照菌株,在SC/-Leu/-Trp营养缺陷型培养基中进行筛选,获得的阳性转化子分别命名为AH-M-XDH和AH-XR-XDH.重组酵母在限制氧通气条件下对木糖和葡萄糖进行共发酵摇瓶培养,HPLC检测发酵底物的消耗和代谢产物的产出情况.结果显示,与对照菌株AH-XR-XDH相比,AH-M-XDH的木糖利用率明显提高,乙醇得率增加了16%,木糖醇产生下降了41.4%.结果证实,通过基因工程改造的木糖代谢关键酶,可用于酿酒酵母发酵木糖生产乙醇,其能通过改善酿酒酵母细胞内氧化还原失衡的问题,提高木糖利用率和乙醇产率.  相似文献

利用不同强度的启动子调控木糖代谢关键酶活性,构建稳定代谢葡萄糖和木糖产乙醇的重组酿酒酵母。以本实验室专利菌株Saccharomyces cerevisiae Y5为宿主菌,将树干毕赤酵母Pichia stipitis CBS6054的木糖还原酶基因XYL1和木糖醇脱氢酶基因XYL2置于磷酸甘油酸激酶基因启动子(PGKp)控制下,酿酒酵母Y5内源的木酮糖激酶基因XKS1分别由己糖激酶基因启动子(HXK2p)及其内源启动子(XKS1p)控制。这3个基因连同各自表达元件导入宿主细胞中,打通其木糖上游代谢途径。酶活测定结果显示,HXK2p对木酮糖激酶表现出更强的启动效率。重组菌Y5-X3-1中木糖还原酶/木糖醇脱氢酶/木酮糖激酶(XR/XDH/XK)的酶活比值为1∶5∶4,其木糖消耗量是宿主菌的5倍,最高乙醇产量为24.35 g/L,达到理论值的73%。结果表明,通过调节XYL1、XYL2及XKS1启动子的强度,调控其表达水平,进而改变3种酶的活性水平,对于提高重组酿酒酵母利用木糖发酵产乙醇有明显效果。  相似文献

为了使酿酒酵母较好地利用木糖产生乙醇,将来自Thermus thermophilus的木糖异构酶基因XYLA和酿酒酵母自身的木酮糖激酶基因XKS1,构建到酵母表达载体pESC-LEU中,导入酿酒酵母YPH499中,同时成功表达了两种酶基因。该菌以木糖为唯一碳源进行限氧发酵,木糖的利用率为9.64%,为宿主菌的4.17倍,产生2.22 mmol.L-1的乙醇。同时初步探讨了两种酶基因的表达量对酿酒酵母发酵木糖生成乙醇的影响。木糖异构酶对木糖的利用起关键性的作用,木酮糖激酶的过量表达不利于乙醇生成。  相似文献

【目的】通过系统研究一个、两个及多个非氧化磷酸戊糖(PP)途径基因组合过表达对酿酒酵母木糖代谢的影响,以优化重组菌株的构建过程,构建高效的木糖代谢酿酒酵母菌株。【方法】在酿酒酵母中双拷贝过表达上游代谢途径的关键酶(木糖还原酶XR,木糖醇脱氢酶XDH,木酮糖激酶XKS),在此基础上构建了一系列PP途径基因过表达菌株,并对其木糖发酵性能进行比较研究。【结果】木糖发酵结果显示,不同组合过表达PP途径基因能不同程度改善重组菌株的木糖发酵性能。其中,过表达PP途径全部基因(RKI1,RPE1,TAL1和TKL1)使菌株的发酵性能最优,其乙醇产率和产量较对照菌株分别提高了39.25%和12.57%,同时较其他基因组合过表达菌株也有不同程度的改善。【结论】通过构建PP途径基因不同组合过表达酿酒酵母菌株,首次对PP途径基因对酿酒酵母木糖代谢的影响进行了系统研究,结果表明,不同组合强化PP途径基因对重组菌株木糖代谢的影响存在差异,相对于其他基因过表达组合,同步过表达PP途径全部基因最有利于碳通量流向乙醇。  相似文献

木糖利用困难是秸秆燃料乙醇产业化的制约因素,改造酿酒酵母使其能够代谢木糖生成乙醇是当前研究的热点之一。为了研究CIT2基因对遗传改良酿酒酵母C5D-P-M菌株中葡萄糖与木糖共发酵过程中木糖利用的影响,设计引物并应用重叠PCR技术获得敲除CIT2基因的敲除组件,通过同源重组的方法将C5D-P-M中的CIT2基因敲除得到突变菌株C5D-P-M-CIT2Δ。通过对出发菌株和突变菌株的生长速率、葡萄糖利用率、木糖利用率及乙醇产量进行研究,结果表明突变菌株C5D-P-M-CIT2Δ的上述指标比出发菌株C5D-P-M均有一定程度的提高。因此推测,酿酒酵母中CIT2基因是影响木糖利用的因素之一,CIT2基因的敲除可提高酿酒酵母的木糖利用率。  相似文献

根据NCBI中的木糖还原酶基因序列设计引物,利用高保真聚合酶克隆树干毕赤酵母木糖还原酶基因,加A后克隆到质粒pGM-T中,测序验证.然后将目的基因克隆到舍有强启动子的穿梭表达载体p424GPD中,构建含有XYL1基因的重组质粒p424GPD-XYL1.将p424GPD-XYL1转化到大肠杆菌中,提取总蛋白,聚丙烯酰胺凝胶电泳分析.酶活测定确定木糖还原酶基因XYL1在大肠杆菌中得到活性表达,表明表达载体构建成功.表达载体的成功构建为后续构建重组酿酒酵母利用木糖发酵奠定基础.  相似文献

Background  

The NAD(P)H-dependent Pichia stipitis xylose reductase (PsXR) is one of the key enzymes for xylose fermentation, and has been cloned into the commonly used ethanol-producing yeast Saccharomyces cerevisiae. In order to eliminate the redox imbalance resulting from the preference of this enzyme toward NADPH, efforts have been made to alter the coenzyme specificity of PsXR by site-directed mutagenesis, with limited success. Given the industrial importance of PsXR, it is of interest to investigate further ways to create mutants of PsXR that prefers NADH rather than NADPH, by the alternative directed evolution approach.  相似文献

A major problem when xylose is used for ethanol production is the intercellular redox imbalance arising from different coenzyme specificities of xylose reductase (XR) and xylitol dehydrogenase. The residue Lys21 in XR from Pichia stipitis was subjected to site-directed mutagenesis to alter its coenzyme specificity. The N272D mutant exhibited improved catalytic efficiency when NADH was the coenzyme. Both K21A and K21A/N272D preferred NADH to NADPH, their catalytic efficiencies for NADPH were almost zero. The catalytic efficiency of K21A/N272D for NADH was almost 9-fold and 2-fold that of K21A and the wild-type enzyme, respectively. Complete reversal of coenzyme specificity toward NADH and improved catalytic efficiency were achieved. Xylose reductase catalyzes the NAD(P)H-dependent reduction of xylose to xylitol and is essential for growth on xylose by yeasts. To understand the nature of coenzyme binding to the Pichia stipitis xylose reductase, we investigated the role of the strictly conserved Lys270 in the putative IPKS coenzyme binding motif by site-directed mutagenesis. The Lys270Met variant exhibited lower enzyme activity than the wild-type enzyme. The apparent affinity of the variant for NADPH was decreased 5–16-fold, depending on the substrate used, while the apparent affinity for NADH, measured using glyceraldehyde as the substrate, remained unchanged. This resulted in 4.3-fold higher affinity for NADH over NADPH using glyceraldehyde as the substrate. The variant also showed a 14-fold decrease in Km for xylose, but only small changes were observed in Km values for glyceraldehyde. The wild-type enzyme, but not the Lys270Met variant, was susceptible to modification by the Lys-specific pyridoxal 5′-phosphate. Results of our chemical modification and site-directed mutagenesis study indicated that Lys270 is involved in both NADPH and d-xylose binding in the P. stipitis xylose reductase.  相似文献

Xylose reductase from the xylose-fermenting yeast Pichia stipitis was purified to electrophoretic and spectral homogeneity via ion-exchange, affinity and high-performance gel chromatography. The enzyme was active with various aldose substrates, such as DL-glyceraldehyde, L-arabinose, D-xylose, D-ribose, D-galactose and D-glucose. Hence the xylose reductase of Pichia stipitis is an aldose reductase (EC 1.1.1.21). Unlike all aldose reductases characterized so far, the enzyme from this yeast was active with both NADPH and NADH as coenzyme. The activity with NADH was approx. 70% of that with NADPH for the various aldose substrates. NADP+ was a potent inhibitor of both the NADPH- and NADH-linked xylose reduction, whereas NAD+ showed strong inhibition only with the NADH-linked reaction. These results are discussed in the context of the possible use of Pichia stipitis and similar yeasts for the anaerobic conversion of xylose into ethanol.  相似文献

We focused on the effects of a mutation of xylose reductase from Pichia stipitis (PsXR) on xylose-to-ethanol fermentation using recombinant Saccharomyces cerevisiae transformed with PsXR and PsXDH (xylitol dehydrogenase from P. stipitis) genes. Based on inherent NADH-preferring XR and several site-directed mutagenetic studies using other aldo-keto reductase enzymes, we designed several single PsXR mutants. K270R showing decreased NADPH-preferring activity without a change in NADH-preferring activity was found to be a potent mutant. Strain Y-K270R transformed with K270R PsXR and wild-type PsXDH showed a 31% decrease in unfavorable xylitol excretion with 5.1% increased ethanol production as compared to the control in the fermentation of 15 g l(-1) xylose and 5 g l(-1) glucose.  相似文献

Thermo-tolerant yeast Kluyveromyces marxianus is able to utilize a wide range of substrates, including xylose; however, the xylose fermentation ability is weak because of the redox imbalance under oxygen-limited conditions. Alleviating the intracellular redox imbalance through engineering the coenzyme specificity of NADPH-preferring xylose reductase (XR) and improving the expression of XR should promote xylose consumption and fermentation. In this study, the native xylose reductase gene (Kmxyl1) of the K. marxianus strain was substituted with XR or its mutant genes from Pichia stipitis (Scheffersomyces stipitis). The ability of the resultant recombinant strains to assimilate xylose to produce xylitol and ethanol at elevated temperature was greatly improved. The strain YZB014 expressing mutant PsXR N272D, which has a higher activity with both NADPH and NADH as the coenzyme, achieved the best results, and produced 3.55 g l?1 ethanol and 11.32 g l?1 xylitol—an increase of 12.24- and 2.70-fold in product at 42 °C, respectively. A 3.94-fold increase of xylose consumption was observed compared with the K. marxianus YHJ010 harboring KmXyl1. However, the strain YZB015 expressing a mutant PsXR K21A/N272D, with which co-enzyme preference was completely reversed from NADPH to NADH, failed to ferment due to the low expression. So in order to improve xylose consumption and fermentation in K. marxianus, both higher activity and co-enzyme specificity change are necessary.  相似文献

Xylose fermentation by Saccharomyces cerevisiae requires the introduction of a xylose pathway, either similar to that found in the natural xylose-utilizing yeasts Pichia stipitis and Candida shehatae or similar to the bacterial pathway. The use of NAD(P)H-dependent XR and NAD(+)-dependent XDH from P. stipitis creates a cofactor imbalance resulting in xylitol formation. The effect of replacing the native P. stipitis XR with a mutated XR with increased K(M) for NADPH was investigated for xylose fermentation to ethanol by recombinant S. cerevisiae strains. Enhanced ethanol yields accompanied by decreased xylitol yields were obtained in strains carrying the mutated XR. Flux analysis showed that strains harboring the mutated XR utilized a larger fraction of NADH for xylose reduction. The overproduction of the mutated XR resulted in an ethanol yield of 0.40 g per gram of sugar and a xylose consumption rate of 0.16 g per gram of biomass per hour in chemostat culture (0.06/h) with 10 g/L glucose and 10 g/L xylose as carbon source.  相似文献

The activities of xylitol dehydrogenase and xylose reductase in the yeasts Candida shehatae, C. didensiae, C. intermediae, C. tropicalis, Kluyveromyces marxianus, Pichia stipitis, P. guillermondii, Pachysolen tannophilus, and Torulopsis molishiama were studied at different oxygen transfer rates (OTRs) to the fermentation medium (0, 5, and 140 mmol O2/(1 h)). The activities of these enzymes were maximum in the yeasts P. stipitis and C. shehatae. The xylitol dehydrogenase of all the yeasts was NAD-dependent, irrespective of the intensity of aeration. The xylose reductase of the yeasts C. didensiae, C. intermediae, C. tropicalis, Kl. marxianus, P. guillermondii, and T. molishiama was NADPH-dependent, whereas the xylose reductase of P. stipitis, C. shehatae, and Pa. tannophilus was specific for both NADPH and NADH. The effect of OTR on the activities of the different forms of xylitol dehydrogenase and xylose reductase in the xylose-assimilating yeasts is discussed.  相似文献
