生物标志物(Biomarkers)在海洋环境监测中的研究与进展   总被引:3,自引:0,他引:3  
污染物的存在给海洋环境和生态健康带来了巨大的压力,如何监测这些污染物并对其毒性进行科学的评价,成为当今环境科学关心的热点问题。生物标志物(Biomarkers)是毒理学研究的重要工具,它能对污染事件进行早期预警,并能在一定程度上评估生态风险。对近十多年来生物标志物的研究与应用进行了回顾,总结了不同水平(分子、细胞、个体、系统)生物标志物的种类、特征、检测方法和应用特点,并对现存挑战和未来发展趋向进行了展望,旨在系统的认识生物标志物,并为其在海洋环境监测中的合理应用提供理论借鉴。  相似文献

2011-2012年,在广西西部沿海设置5个站点,以野生文蛤作为指示生物进行3次采样监测.从个体、细胞和分子水平选择了钻沙所需时间、吞噬能力、溶酶体膜稳定性、血浆三价铁还原力、乙酰胆碱酯酶活性及彗星率6项指标,利用综合生物标志物响应指数模型(IBR),整合上述生物标志物指标,并转化为形象直观的星状图,对调查站点的环境状况进行风险评估.结果表明: 各站点的综合生物标志物响应指数值(IBR/n)介于2.30~8.68,茅尾海的环境压力最大,北仑河口最小.虽然不同的生物标志物对污染压力的响应存在差异,但利用IBR可以有效区分文蛤所在区域的环境压力状况.利用生物标志物的监测结果与化学监测结果基本吻合.  相似文献

应用陆栖无脊椎动物的生物标志物对土壤生态系统中污染物的暴露和效应进行评价日益受到重视,并取得了显著的研究进展,文中介绍了溶酶体、胁迫蛋白和金属硫蛋白(MTs)3种主要生物标志物,体腔细胞内溶酶体膜稳定性用中性红保持时间(NRR)进行检测;胁迫蛋白类多采其中的Hsp70和Hsp60;金属硫蛋白不同同分异构体的定量分析可用于反映不同的金属污染胁迫,对3种生物标志物机理、特性、检测实例以及在污染土壤生态毒理诊断中的应用前景进行了评述。  相似文献

根据2007年9月和10月福建南日岛生态调查资料,从海水水质、海水营养结构与营养水平、生物多样性等3个方面诊断与评价了南日群岛海域环境现状,利用综合质量指数法对海洋生态环境质量进行综合评价,并探讨了不同评价指数的关系及合理性.结果表明:海域的pH、溶解氧(DO)、化学需氧量(COD)、Pb、Cd、Hg、As含量均符合第二类海水水质标准.71%站位的磷酸盐、14%站位的无机氮和7%站位的石油类污染物超第二类海水水质标准.海水水质总体属于较好等级,营养结构表现为氮限制,大部分海域处于富营养化状态,依据浮游生物的多样性指数评价结果为轻污染-清洁水平.生态环境综合质量指数评价结果表明南日群岛海域生态环境总体处于良好水平.利用不同的评价指数对海域环境健康状况进行诊断的结果存在一定差异.在实际评价中应综合运用化学指标和生物指标,才能得到相对客观的结论.  相似文献

典型平原河网地区底栖动物生物指数筛选及评价基准研究   总被引:1,自引:0,他引:1  
2011年夏、秋季在上海市全境主要河流的83个断面进行大型底栖动物的采样, 共获取底栖动物20个分类单位(种), 其中软体动物、环节动物和节肢动物分别占50%、30% 和20%。选取常见的28个生物指数分别进行计算, 并采用非参数统计检验方法筛选出8个敏感生物指数。通过记分法对8种敏感生物指数统一量纲后, 获得数值范围为8-40的综合生物指数, 运用四分法划分了判别河道水环境质量的生物基准: 32-40, 较好(Good); 24-31, 一般(Fair); 16-23, 污染(Poor); 8-15, 严重污染(Very poor)。9个断面未发现活体生物, 定为超严重污染(Super poor)。依照生物基准比较上海市41个代表性断面近5年主要水质理化指标, 基本反映出水质理化指标的空间差异性, 该综合生物指数及判别基准也可适用于评价平原河网地区的河道水质状况。    相似文献

对太湖8个区域共60样点进行底栖大型无脊椎动物采样,共获得底栖动物24种,其中软体动物14种,节肢动物5种,甲壳动物5种.根据Hilsenhoff 生物指数确定了15个相对清洁的样点和45个污染样点,然后进行21种生物指数的综合评估,结果表明,总分类单元数、(甲壳动物 软体动物)分类单元数、%(甲壳动物 软体动物)、%腹足纲、Goodnight-Whitley指数、Hilsenhoff 生物指数和%直接收集者等7个底栖动物生物指数可以用作判别太湖水质的敏感生物指数.通过5, 3, 1记分法对6种生物指数统一量纲后,获得变化范围为7~35的综合生物指数,运用四分法划分了太湖水质判别的生物基准:7~14很差,15~21差,22~28一般,29~35好,并对60个样点进行重新记分,获得了太湖水质的基本生态分区现状,太湖的东南区属水质较好的区域,而西北区属水质较差的区域.该水质生物基准基本适合评价太湖不同区域的水质状况.  相似文献

藻类对水体环境变化敏感,其种类和数量与环境因素有密切联系并因环境的变化而发生变化,因此藻类常作为河流健康评价的指示生物。水体中的藻类根据生活习性不同分为着生藻类和浮游藻类,在河流健康评价中,以往的工作中有的采用浮游藻类用于河流健康评价,有的采用着生藻类用于评价,但浮游藻类和着生藻类究竟何者用于河流健康评价更适宜,抑或是二者在用于评价的适宜性上没有明显差别,迄今为止未开展过深入研究。选择三峡库区内的两条河流嘉陵江和乌江作为研究对象,于2015年9月,在两条河流上共布设11个研究断面,对嘉陵江、乌江的水环境理化因子、着生藻类和浮游藻类群落进行调查研究,应用着生藻类生物完整性指数(Periphytic algal index of biological integrity,Pe-IBI)和浮游藻类生物完整性指数(Phytoplankton index of biological integrity,Ph-IBI),并结合水体综合污染指数(Comprehensive pollution index,CPI),对嘉陵江、乌江的健康状况进行评价。研究结果表明,采用着生藻类生物完整性评价(Pe-IBI)能筛选出7个核心生物参数(藻类总分类单元数、蓝藻总分类单元数、绿藻总分类单元数、菱形藻比例、优势分类单元比例、香农多样性指数、均匀度)用于河流健康评价,着生藻类生物完整性指数(Pe-IBI)与水体综合污染指数(CPI)具有极显著的负相关关系,并且与水体中总氮、铅含量也有极显著的负相关关系,表明水环境质量越低则着生藻类的生物完整性越差;当采用浮游藻类生物完整性(Ph-IBI)用于河流健康评价时却只能筛选出1个核心生物参数(藻类密度),而且浮游藻类生物完整性指数(Ph-IBI)与水体综合污染指数(CPI)及12个水体环境指标并无明显的相关性。本研究表明,在河流水体中,与浮游藻类相比,着生藻类更能反映水体环境的状况,对河流水体环境的反映更为准确。因此,在以流动水体为特征的河流的健康评价中,采用着生藻类比采用浮游藻类更为适宜。  相似文献

本文应用PFU法对常德水系四季微型生物群落的结构和功能进行了分析。根据MacArthur-Wilson平衡模型提出了3个功能参数,测定了微型生物的2个结构参数。同步进行了16个站四季的水质分析,提出化学综合污染指数(P)。经统计学处理,P和微型生物群落的HI、S_(eq)、G、T_(90%)等指数均呈显著性相关。物种多样性(d)也在一定程度上反映出城市水体的不同污染程度。再次证明PFU微型生物监测方法的科学性,为生物监测提供了快速、经济、正确的新方法。  相似文献

2013年对淮北采煤塌陷湖(乾隆湖和临涣湖)大型底栖动物群落结构进行了季节性调查。共采集到大型底栖动物26种, 隶属于3门5纲17科. 乾隆湖和临涣湖年平均密度分别为230.85和215.80 ind./m2, 年平均生物量分别为56.11和36.38 g/m2。两湖大型底栖动物以摇蚊幼虫、霍甫水丝蚓和苏氏尾鳃蚓为优势类群, 底栖动物最高密度均出现在夏季(乾隆湖为 278.0 ind./m2, 临涣湖为288.2 ind./m2); 生物量则主要以软体动物和水生昆虫为主, 夏季和秋季大型底栖动物生物量明显高于春季和冬季。运用Shannon多样性指数(H')、Margalef多样性指数(D)和Biotic Index生物学指数(BI)对乾隆湖和临涣湖进行水质生物评价研究, 结果显示Shannon多样性指数不适宜于该两湖的水质生物评价, BI的评价结果与水质状况基本相符。综合 Margalef多样性指数和BI生物指数评价的结果表明: 乾隆湖和临涣湖春季和冬季处于轻度污染状态, 夏季和秋季处于中度污染状态。  相似文献

蚯蚓在土壤中行使了很多重要的生态功能,蚯蚓生物标记物常用作土壤污染风险评价研究。这篇综述的目的是探讨当前蚯蚓生物标记物研究是否可以应用到实际的土壤污染风险评价。1)讨论了蚯蚓生物标记物在土壤污染风险评价体系中的重要性,认为它是化学分析方法的有益补充,可以提供更为全面和客观的土壤污染信息;2)综述了相关研究中所使用的蚯蚓类型,土壤类型和生物标记物类型,及其它试验设计要素和最后结果的变异,认为目前蚯蚓生物标记物研究以实验室基础研究为主,筛选出了大量的生物标记物,一定程度上揭示了生物标记物的对各类典型污染物及其组合的应答机制;同时也认为,未来的蚯蚓生物标记物研究应该重点探讨将其应用到实际的土壤污染风险评价中的可行性及如何应用;3)目前不同研究之间从试验设计到结果都具有很大的变异,难以通过综合比较获得完全可靠的具有实践意义的结论和成果,因此,有必要通过建立标准化的蚯蚓生物标记物研究方法,推动生物标记物的研究工作;4)提出了蚯蚓生物标记物研究方法标准化的具体建议,推荐了蚯蚓生物标记物走向实际应用所需要解决的问题。  相似文献

A suite of biomarkers was measured in caged mussels at areas impacted by different anthropogenic activities along the Greek coastline to assess biological effects of environmental pollution. Mussels were caged at coastal sites in the vicinity of major cities, in areas influenced by major industries, agricultural practices and in islands away from known sources of pollution. Biomarkers indicative of neurotoxicity (acetylcholinesterase, AchE), oxidative stress (catalase, CAT), phase II biotransformation of xenobiotics (glutathione S-transferase, GST), metal exposure (metallothioneins, MTs) and protein synthesis (RNA:DNA ratio) were measured to assess effects of various types of pollutants. AchE activity proved to be the most responsive biomarker with decreased values at sites influenced by agricultural, urban and industrial activities. Decreased CAT and GST activities and increased MTs levels were recorded at a number of anthropogenic-impacted sites. RNA:DNA ratio showed a biphasic response as both high and low values were found at impacted sites. Principal component analysis clearly distinguished sites receiving pollution inputs from non-polluted sites. The combination of the selected biomarkers used in caged mussels resulted useful in the assessment of the effects of environmental pollution.  相似文献

根据压力-状态-响应(PSR)框架模型,从广义上定义河口区生态系统,将河口及毗邻的陆域、海域生态系统作为一个整体,从压力指标、状态指标、响应指标3个方面构建了黄河河口区生态系统健康评价的指标体系,以研究区1991年数据和相关国家标准为基准,2013年代表现况,利用综合指数法(CEI)评价了黄河河口区的生态系统健康状况。结果显示:黄河河口区生态系统健康评价的响应指数最高(0.9055),压力指数居中(0.8288),状态指数最低(0.6458),综合指数为0.7427。总体来看,与1991年相比,目前黄河河口区生态系统仍处于"健康"状态,但健康状况明显下降,其中状态指数下降最为严重。从区域轻度开发到人类活动强烈干扰阶段,黄河河口区存在过度捕捞、湿地不合理开发、浅海养殖过度及污染物排放等一系列影响生态系统健康的问题,应进行区域的生态恢复和科学管理。  相似文献

Abstract

Environmental exposure is a growing public health burden associated with several negative health effects. An estimated 4.2 million deaths occur each year from ambient air pollution alone. Biomarkers that reflect specific exposures have the potential to measure the real integrated internal dose from all routes of complex environmental exposure. MicroRNAs (miRNAs), small non-coding RNAs that regulate gene expression, have been studied as biomarkers in various diseases and have also shown potential as environmental exposure biomarkers. Here, we review the available human epidemiological and experimental evidence of miRNA expression changes in response to specific environmental exposures including airborne particulate matter. In doing so, we establish that miRNA exposure biomarker development remains in its infancy and future studies will need to carefully consider biological and analytical ‘design rules’ in order to facilitate clinical translation.  相似文献

Over the past decade, molecular, biochemical and cellular markers have been extensively used in pollution monitoring of aquatic environments. Biochemical markers have been selected among early molecular events occurring in the toxicological mechanisms of main contaminants. This paper assesses the marine environment quality along the Tunisian coasts using a statistical approach. Clams (Ruditapes decussatus) were collected during the four seasons of 2003 on seven different sites from the Tunisian coasts. Oxidative stress was evaluated in gills using catalase activity (Cat), neutral lipids and malonedialdehyde accumulation. Glutathione S-transferase activity is related to the conjugation of organic compounds and was evaluated in both, gills and digestive glands. Acetylcholinesterase activity was evaluated as the biomarker of exposure to organophosphorous, carbamate pesticides and heavy metals. For each biomarker, a discriminatory factor was calculated and a response index allocated. For each site, a global response index was calculated as the sum of the response index of each biomarker. Discriminant analysis shows significant differences between sites and seasons compared with control sample. Faroua (site 1) and Menzel Jemile (site 2) seem to be the less polluted with respect to the other sites for all seasons. Gargour (site 6) shows the highest Multimarker Pollution Index during the four seasons, indicating higher contamination level.  相似文献

Glutathione S-transferase (GST) is a phase II enzyme that functions as a detoxicant by catalyzing the conjugation of reduced glutathione with a variety of xenobiotics via cysteine thiol. Molecular genetic approaches using gene biomarkers show substantial relevance as sensitive biomarkers for the indication of pollution levels. In order to use GSTs as molecular biomarkers for marine pollution monitoring, we cloned and sequenced the full-length cDNA of seven GST genes from the marine polychaete Perinereis nuntia. The deduced amino acid sequence of Pn-GSTs showed a high similarity to those of other species that clustered into the same clades in a phylogenetic analysis. In addition, to evaluate Pn-GSTs as useful biomarkers on effects after cadmium (Cd) exposure, we exposed sublethal concentrations of Cd (5, 50, and 500 μg/L) to P. nuntia, and they showed relatively different but significantly increases, depending on exposure time and Cd concentrations. Particularly, Pn-GST-omega and Pn-GST-sigma genes were highly sensitive with a clear dose-dependent manner on mRNA expression. The total GST activities also have significantly increased levels at higher concentrations of Cd exposure. These results indicate that Pn-GSTs play important roles in Cd-induced oxidative stress in terms of the physiological changes relating to metabolism and cell protection, and those genes would have great potential as molecular biomarkers to monitor marine environmental health.  相似文献

Numerous molecular, cellular, and physiological biomarkers have been used to assess the responses of marine animals to petroleum compounds. To be used in ecological risk assessment after an oil spill, a biomarker response needs to be linked to petroleum exposure and not strongly influenced by internal and external confounding factors. Biomarker responses to petroleum PAH, dominated by alklated two-and three-ringed aromatics, can be quite different than responses to pyrogenic PAH, dominated by four-and five-ringed aromatics. In many field sites there is a mixture of petrogenic and pyrogenic PAH, along with other contaminants, making it difficult to relate biomarker responses to a particular contaminant class. Biomarkers used to assess marine animal responses in the field include the cytochrome P450 system, heat stress protein, histopathology, and bile fluorescent compounds (FAC). Other biomarkers, including DNA/chromosomal damage and phase 2 enzymes, have been shown to respond after laboratory exposure, but more work needs to be done to demonstrate their usefulness in the field. One of the most useful biomarkers of petroleum exposure are the FAC responses in fish, which can be used to distinguish between petrogenic and pyrogenic PAH exposure. Few of the presently used biomarkers are linked to higher order biological effects, e.g. toxicity, reproductive failure.  相似文献
