Quantitative study of the cytochrome c acting in the photosyntheticsystem of the blue-green alga Anabaena variabilis (M-2) wasdone with membrane fragments and intact cells. Membrane fragments highly active in the NADP+-Hill reaction(above 200 µmoles/mg chl.a;-hr) retained photoresponsivecytochrome c equal only one-tenth that of P700, while the plastocyanincontent was almost equal to that of P700. The cytochrome contentin intact cells was a little larger than that in membrane fragmentson the chlorophyll a basis. However, the values relative toP700 (1/9) and plastocyanin (1/10) were identical with thosein membrane fragments. The content was also far smaller thanthat of reaction center II's (1/6). If the cytochrome mediatesall electrons from reaction center II, the cytochrome oxidation-reductionshould have a rate constant of 2.4?102 sec–1 which isone order above of the rate constant of the cytochrome reduction(2.3 to 3.5?101sec–1). These quantitative relationshipsindicate that in Anabaena variabilis (M-2), c-type cytochrome,either cytochrome f or algal cytochrome c, cannot function inthe main electron flow between two reaction centers. Dark-adapted intact spinach chloroplasts exhibited two peaks,P and M1, at the early phase of fluorescence induction and atransient reduction of cytochrome f shortly after its initialphotooxidation and in parallel to the appearance of P. Analysisof the peak P and the transient reduction of cytochrome f indicatedthat electron transport in intact spinach chloroplasts was regulatedby light: electron transport was inactivated at the reducingside of photosystem I in the dark-adapted chloroplasts but rapidlyreactivated by illumination. The fluorescence peak M1 was correlatedto the proton gradient formed across the thylakoid membrane. Effects on P and transient reduction of cytochromef of NO2,3-phosphoglycerate (PGA) and oxalacetate (OAA), which can penetrateinto intact chloroplasts and accept electrons at different sitesafter photosystem I, were studied to determine the site of thelight regulation. NC2, which receives electrons fromreduced ferredoxin, markedly diminished both P and the transientreduction of cytochrome.f, whereas PGA and OAA, the reductionsof which are NADP-dependent, failed to affect the two transients.The ineffectiveness of PGA and OAA could not be attributed tothe dark inactivation of glyceraldehyde-3-phosphate and malicdehydrogenases, because dark-adapted chloroplasts still retainedsufficiently high levels of the enzyme activities. The resultsindicate that electron transport in intact spinach chloroplastsis regulated by light after ferredoxin but before NADP, i.e.,at the reducing terminal of the electron transport chain. Temperature, food and the availability of mates may all limitrates of egg production is freshwater diaptomids. We have usedthe reproductive phases of Diaptomus paliidus to develop anindex of food limitation (f) and an index of mate limitation(M), each of which responds independently of temperature. Theresponse of the food limitation index to high and low temperaturesand high and low food concentrations was examined in a two-wayfactorial experiment in the laboratory. The index was highlyresponsive to a change in food concentration, stable duringa change in temperature alone at the high food level and responsiveto the synergistic interaction of food and temperature effectswhen both factors were limiting. Laboratory data indicate thatthe f index should not be biased by mate limitation unless maledensities are 6.7 x 10–l0 males 1–1 or less. Themate limitation index was highly responsive to mate availability,and ranged from a low of 9–19 in the presence of abundantmales to 100 in tbe absence of males. Changes in carbon fixation rate and the levels of photosyntheticproteins were measured in fourth leaves of Lolium temulentumgrown until full expansion at 360 µmol quanta m–2s–1 and subsequently at the same irradiance or shadedto 90 µmol m–2 s–1. Ribulose-1,5-bisphosphatecarboxylase/oxygenase (Rubisco), light-harvesting chlorophylla/b protein of photosystem II (LHCII), 65 kDa protein of photosystemI (PSI), cytochrome f (Cytf) and coupling factor 1 (CF1) declinedsteadily in amount throughout senescence in unshaded leaves.In shaded leaves, however, the decrease in LHCII and the 65kDa protein was delayed until later in senescence whereas theamount of Cyt f protein decreased rapidly following transferto shade and was lower than that of unshaded leaves at the earlyand middle stages of senescence. Decreases in the Rubisco andCF1 of shaded leaves occurred at slightly reduced rates comparedwith unshaded leaves. These results indicate that chloroplastproteins in fully-expanded leaves are controlled individually,in a direction appropriate to acclimate photosynthesis to agiven irradiance during senescence. From the membrane fragments of the green alga Bryopsis maxima,a cytochrome which resembles cytochrome f of higher plants wassolubilized with methyl ethyl ketone. The cytochrome was partlypurified by ammonium sulfate fractionation, followed by gelfiltration. Its properties were similar to those of the algalcytochrome f reported by Wood (26). The approximate molar ratioof cytochromes f, c-553 and chlorophyll in B. maxima was 1 :1 : 600–700. 1 In this communication, according to the recommendation byWood (26), cytochrome f is the membrane-bound c-component andcytochrome c-55 the soluble one. Partially purified homoserine dehydrogenase was prepared frompea seedlings. The optimum pH for this enzyme is approximately 5.4. The Kmvaluesfor ASA and TPNH are 4.6xl0–4Af and 7.7xl0–5M, respectively.This enzyme can also utilize DPNH but less effectively thanTPNH. In contrast with yeast homoserine dehydrogenase whichis insensitive to — SH reagents, the pea enzyme is inhibitedalmost completely by 10–4MPCMB and 10–5MHgCl2, theinhibition being removed by 10–2M thioglycolate. Homoserinedehydrogenase was found not only in decotylized seedlings, butalso in cotyledons. The significance of this enzyme in homoserine biosynthesis ingerminating pea seeds has been discussed. The relationship between thef-ratio [NO3 uptake/(NO3+ NH4+) uptake] and ambient nitrate concentration was evaluatedfor eight data sets from coastal waters. The f-ratio increasedasymptotically with increase in nitrate concentration in mostdata sets. However, the rate at which f-ratio increased at lownitrate concentration (slope = m) and the maximum attained f-ratio(fmax) varied among regions; the initial slope varied most withvalues ranging in excess of an order of magnitude. The datawere analyzed in relation to environmental factors and methodologicalconsiderations known to influence the f-ratio. Ambient ammoniumconcentration was important in accounting for regional differencesin the f versus NO3 relationship. A further analysisof the data, relating f-ratio to the ratio of NO3/(NO3+ NH4+) concentrations yielded a much more regionally consistentand approximately linear relationship; slopes varied by lessthan a factor of two in the extreme cases. Inclusion of knownalternative (aside from NH4+) sources of reduced-N (e.g. urea)and correction for methodological/computational errors (isotopedilution) systematically reduce f-ratio estimates. Other factors,e.g. reduced-N uptake by microheterotrophs, may systematicallyincrease the f-ratio.  相似文献

A polygalacturonase-inhibiting protein (PGIP) was purified fromimmature raspberry fruits using ion exchange chromatography.The protein was composed of a single polypeptide chain withMr of 38·5 kDa and a pI residing above pH 10. Kineticstudies suggested that the inhibition was of a non-competitivenature. The PGIP inhibited two endopolygalacturonases (endo-PG)purified from Botrytis cinerea and an endo-PG produced by Aspergillusniger to varying degrees but did not inhibit two exo-PGs purifiedfrom B. cinerea, bacterial endopectate lyases and bacterialendo-PGs. The concentration of PGIP at various stages of flowerand fruit development was determined. The inhibitor was notdetected in the flower, but reached a maximum of 69 units g–1in the immature green fruit decreasing to 9 units g–1as fruits matured. Ion absorption by Chara corallina from solutions containingpredominantly KC1 or RbCl at up to 100 mol m–3 resultedin accumulation of salts and turgor regulation. Turgor regulationdid not occur in solutions containing Na+ or Li+salts. Duringion absorption from various salts of K+ and Rb+ vacuolar cationconcentration exceeded Cl concentration. This differencewas shown to be balanced by the synthesis and accumulation ofmalate. Vacuolar malate concentration reached 48 mol m3,with accumulation occurring at rates of up to 0.45 mol m–3h–1. Malate accumulation was inhibited by low externalpH and was dependent upon external HCO3 concentration.The synthesis of malic acid and its subsequent dissociationimposed a severe acid load on the cell. Biophysical regulationof cellular pH was achieved by a H+efflux at a rate of about40 nmol m–2 s–1from the cell. The results presentedargue against cytoplasmic Cl, HCO3 or pH regulatingmalate accumulation in Chara and it is suggested that malatetransport across the tonoplast may regulate malate accumulation. Acclimation of single plants of Lolium temulentum to changing[CO2] was studied on plants grown in controlled environmentsat 20°C with an 8 h photoperiod. In the first experimentplants were grown at 135 µ;mol m–2 s–1 photosyntheticphoton flux density (PPFD) at 415µl l–1 or 550µll–1 [CO2] with some plants transferred from the lowerto the higher [CO2] at emergence of leaf 4. In the second experimentplants were grown at 135 and 500 µmol m–2 s–1PPFD at 345 and 575 µl l–1 [CO2]. High [CO2] during growth had little effect on stomatal density,total soluble proteins, chlorophyll a content, amount of Rubiscoor cytochrome f. However, increasing [CO2] during measurementincreased photosynthetic rates, particularly in high light.Plants grown in the higher [CO2] had greater leaf extension,leaf and plant growth rates in low but not in high light. Theresults are discussed in relation to the limitation of growthby sink capacity and the modifications in the plant which allowthe storage of extra assimilates at high [CO2]. The Cyt f and P700 contents in leaves of three Sorghum, varietieswere measured, in relation to their carbon assimilation, underdifferent light intensities during growth. At the maximum irradiationused (1,800 µE m–2 s–1) the ratio of P700to Cyt f was close to unity, whereas under low irradiation (450µE m–2 s–1) the ratio of P700 to Cyt f rangedfrom two to three. A strikingly positive correlation existedbetween the P700 contents of the leaves and their rates of carbondioxide fixation, dry matter production and Cyt f contents,only when the plants were grown under high light intensities.The P700 content of the leaves in plants grown under low irradiationwas unrelated to the contents of Cyt f. Thus, at a high lightintensity there is a close relationship between the Cyt f andP700 levels, but at low intensities the amounts of electroncarriers and the reaction centre are independent. Cell counts from samples taken at weekly intervals, from 14May to 22 October 1969, in a T. canadensis stand in Massachusetts,U.S.A., showed that the width of the annual ring was correlatedwith the rate of cell production and that only the least vigoroustrees (c. 20 tracheids year–1) had a shorter growing season.The time required for completion of a cell-division cycle inthe cambial zone decreased during the course of the season,from 35 to 20 days for the less vigorous trees (25–45tracheids year–1) and from 28 to 10 days for the morevigorous trees (45–100 tracheids year–1). The timerequired for the completion of radial growth of the tracheidsdecreased from 18 to 9 days, with no evidence of any changeswith tree vigour. The actual radial growth-rate of the tracheidswas constant within the range 1.5–3 um day–1. Thetime required for deposition of the secondary cell wall increasedfrom 10 to 50 days, with little evidence of any changes withtree vigour. The actual rate of deposition of cell wall materialwas about 0.15 µ2 µ–1 day–1 and seemedto show little change during the course of the season. The timeperiod required for lysis of the cytoplasm was about 4 days,with no evidence of any changes with tree vigour and littleevidence of any changes during the course of the season.  相似文献

Loliun perenne L. (cv.S. 23) was grown on vermiculite in winterin a heated greenhouse for 8 weeks under factorial combinationsof two potassium regimes (nominally 6 parts/106 and 156 parts/106in Hewitt's solution) and three densities of artificially supplementedvisible radiation flux (36.1, 7.3, and 2.2 W m–2). Growthand potassium uptake were studied through the calculation ofvarious growth functions from fitted curves. There was little effect of potassium treatment but the experimentalmaterial responded markedly to light. Leaf-area ratio in thethree treatments showed extreme plasticity in increasing from2–3 x 10–2 through 6 x 10–2 to 8–9 x10–2 m2 g–1 as light intensity decreased. Correspondingdecreases in unit leaf rate, however, caused over-all reductionsin relative growth rate. Specific absorption rates for potassium (AK, dry-weight basis)were strongly reduced at the lower light intensities but alsodisplayed complex ontogenetic drifts. Values of the allometricconstant, k (the ratio of root and shoot relative growth rates),decreased from c. 0.7 at 36.1 W m–2 through c. 0.3 at7.3 W m–2 to a value not significantly different fromzero (P < 0.05) at 2.2 W m–2. In material grown under the two higher light intensities a constantinverse relationship was found between the mass ratio of rootand shoot and the corresponding activity ratio. The resultsconform to this model: Mass ratio = –0.001+45.0 (1/activityratio) where activity ratio is expressed as specific absorptionrate for potassium (in µg g root–1 h–1)/unitshoot rate (rate of increase of whole-plant dry weight per unitshoot dry weight, in mg g shoot–1 h–1). Cytoplasmic pH (pHc) in Chara corallina was measured (from [14C]stribution)as a function of external pH (pH0)and temperature. With pH0near 7, pHc at 25?C is 7.80; pHcincreases by 0.005 pH units?C–1 temperature decrease, i.e. pHc at 5 ?C is 7.90. WithpH? near 5.5, the increase in pHc with decreasing temperatureis 0.015 units ?C–1 between 25 and 15?C, but 0.005 units?C–1 between 15 and 5?C. This implies a more precise regulationof pHc with variations in pHo at 5 or 15 ?C compared with 25?C. The observed dp Hc/dT is generally smaller than the –0.017units ?C–1 needed to maintain a constant H+/OH–1,or a constant fractional ionization of histidine in protein,with variation in temperature. It is closer to that needed tomaintain the fractional ionization of phosphorylated compoundsor of CO2–HCO3 The value of dpHc/dT has importantimplications for several regulatory aspects of cell metabolism.These include (all as a function of temperature) the rates ofenzyme reactions, the H+ at the plasmalemma(and hence the energy available for cotransport processes),and the mechanism for pHc regulation by the control of bidirectionalH+ fluxes at the plasmalemma.  相似文献

Bot. 38: 42–52 Oilseed rape (Brassica napus L. cv. Bien venu) was grown for49 d in flowing nutrient solution at pH 6?0 with root temperaturedecrementally reduced from 20?C to 5?C; and then exposed todifferent root temperatures (3, 5, 7, 9, 11, 13,17 or 25?C)held constant for 14 d. The air temperature was 20/15?C day/nightand nitrogen was supplied automatically to maintain 10 mmolm–3 NH4NO3 in solution. Total dry matter production wasexponential with time and similar at all root temperatures givinga specific growth rate of 0?0784 g g–1 d–1. Partitioningof dry matter was influenced by root temperature; shoot: rootratios increased during treatment at 17?C and 25?C but decreasedafter 5 d at 3?C and 5?C. The ratio of shoot specific growthrate: root specific growth rate increased with the ratio ofwater soluble carbohydrates (shoot: root). Concentrations ofwater soluble carbohydrates in shoot and root were inverselyrelated to root temperature; at 3, 5 and 7?C they increasedin stem + petioles throughout treatment, coinciding with a decreasein the weight of tissue water per unit dry matter. These resultssuggest that the accumulation of soluble carbohydrates at lowtemperature is the result of metabolic imbalance and of osmoticadjustment to water stress. Cauliflower (Brassica oleracea L. botrytis) and broccoli (Brassicaoleracea L. italica) plants were grown in large pots in growthchambers for a range of temperatures (mean air temperaturesfrom 7.0-25.3 C) and irradi-ances (from 9.3-50.8 mol m–2d–1 or 4.7-25.4 MJ m–2 d–1). The extinctioncoefficient for PAR decreased with plant size reaching a valueof 0.55 in cauliflower and 0.45 in broccoli at plant leaf areasof 0.235 m2 and 0.227 m2, respectively. The leaf area expansionrate was unaffected by irradiance when compared at identicalleaf surface temperatures. The response of expansion rate tosurface temperature was fitted to a broken stick model witha base temperature of –0.7C and an optimum temperatureof 21.0C. The radiation conversion coefficient increased withair temperature below 13.8C and remained constant above this.The estimated radiation conversion coefficient above 13.8Cand for a PPFD of 20 mol m–2 d–1 was 0.77 g mol–1in cauliflower and 0.87 g mol–1 in broccoli. The radiationconversion coefficient declined with increasing irradiance levelfrom a maximum of 1.89 g mol–1 at near nil irradiancein cauliflower. In vitrorates of gross and net oxygen production were measuredas a function of light intensity in some plankton communitiescollected from Bedford Basin, Nova Scotia, and in a monoclonalculture of Synechococcus. The rate of gross oxygen productionwas measured by a technique in which the stable oxygen isotope,18O, serves as a photosynthetic tracer Net oxygen productionwas measured by automated Winkler technique. The rate of communityrespiration in the light was then determined by the differencebetween gross and net rates of oxygen production. In the naturalpopulations examined, neither gross nor net oxygen productionrates were significantly inhibited at the highest light intensitymeasured (500–800 µE m–2 s–1) In a samplein which the dark respiration rate was small relative to themaximal rate of production [Pmax;sensu Platt et al (1980) JMar. Res., 38, 687–701] the rates of ‘light’respiration were 3 times greater. In two other communities,with high rates of dark respiration relative to Pmaxthe ratesof ‘light’ respiration were closer to rates of darkrespiration. In the Synechococcus clone, both gross and netoxygen production rates were inhibited at high light intensities.Rates of ‘light’ respiration were found to varyas a function of light intensity. The greatest rates of respirationwere measured in samples incubated at light intensities thatwere just saturating (100 µE m–2 s–1). Ingestion, respiration, and molting loss rates were measuredover the 3 – 29°C range in Neomysis intermedia. Weightspecific rates of these physiological processes ranged from2 to 140% body C day–1 for ingestion, from 2 to 15% bodyC day–1 for respiration, and from 0.1 to 5% body C day–1for molting loss. All weight-specific rates showed a logarithmicdecrease with a logarithmic increase in body weight, and a logarithmicincrease with a linear increase in temperature below 20 or 25°C.The effect of temperature, however, was different between thephysiological rates, with a large temperature dependency foringestion (Q10 = 2.6 –3.9) and molting loss (Q10 = 2.9– 3.6) and a moderate temperature dependency for respiration(Q10 = 1.9 – 2.1). Calculated assimilation efficiencychanged with body size, but was constant over the temperaturerange examined. Allocation of assimilated materials varied witha change in temperature, reflecting the different temperaturedependence between physiological processes. It was deduced thatthe strong temperature dependency of the growth rate in N. intermediaobserved in the previous studies resulted from the large temperatureeffect on ingestion and assimilation rates, superimposed bythe different allocation of assimilated materials. 1Present address: Department of Botany, University of Tokyo,Hongo, Tokyo 113, Japan  相似文献

Effects of Cl and other anions on the rate of HILL reactionin Euglena chloroplasts were investigated. Cl acceleratedthe reaction rate with ferricyanide as HILL oxidant; Br,F and I were also effective; NO3, PO42–and SO42– were less effective. Divalent cations, Ca2+and Mg2+, were also highly effective. The promoting effectsof these ions were highly dependent on pH and the nature andconcentration of the HILL oxidant used. Accelerating effectsof the ion increased with decreasing concentrations of ferricyanide.Generally, the stimulating effect of Cl was much moremarked at pH 7–7.5, with little effect at pH 5. Thus,the pH-activity relationship in the HILL reaction is more orless markedly modified by addition of ions. Cl, and other anions, accelerated the reaction by affectingonly the dark rate-limiting portion of the HILL reaction; thelight reaction constant remained uninfluenced. We inferred thatsome reaction step, at which ferricyanide receives electronfrom photosystem 2, is accelerated by Cl and other ions.Cl effects were rather small, or undetectable, with DPIPor p-benzoquinone as oxidants.
