The reaction rate of reduced monomeric cytochrome f with oxidizedplastocyanin, both purified from Japanese radish, was determinedby a stopped-flow method. The oxidation rate constant was 6.0x 107 M1sec1 at pH 7.0 and 25°C, which is slightly higherthan the value reported by Wood [(1974) Biochim. Biophys. Acta357 : 370] for oligomeric parsley cytochrome f Thermodynamicparameters also were determined to be 56 KJ M–1 for activationenthalpy and 90 J M–1 K–1 for activation entropy.Neither a pH from 6 to 9 nor the addition of NaCl, polylysine,histone or polyaspartate affected the rate constant. 1Present address: The National Institute for Environmental Studies,Yatabe, Ibaraki 305, Japan. Post-embryonic development time, egg production rate and adultbody size of calanoid copepod Eurytemora affinis from Lake Ohnuma,Japan, were determined under six temperature-food conditions(103,5 x 103, 104 and 5 x 104 cells ml–1 at 15°C,5 x 104 cells ml–1 at 10and20°C) in the laboratory.The measured parameters varied with both temperature and foodconcentration. Development time from hatching to adult femalewas 9.2, 11.4 and 22.8 days at 20, 15 and 10°C respectively,at the highest food concentration. The males developed to adultat one to two days earlier than the females. An effect of foodshortage on development time occurred at the lowest food concentration.This development time was 24.8 days even at 15°C, beingtwice as long as that at the highest food concentration. Prosomelength of these food-limited females was approximately 75% ofwell-fed ones, which reduced by only 10% with increasing temperaturefrom 10 to 20°C. Clutch size (C, eggs clutch–1) ofwell-fed individuals depended on prosome length of the adultfemale (L, mm), and was expressed as an equation: C = 65.2 L383. Clutch size of individuals reared at less than 104 cellsml–1, however, mostly laid below the estimated curve,especially at the lowest food concentration being only 10% ofthat at the highest food concentration. These results suggestthat food availability is a more important factor affectingpopulation growth of E.affinis in Lake Ohnuma than variationof temperature.  相似文献

The Cyt f and P700 contents in leaves of three Sorghum, varietieswere measured, in relation to their carbon assimilation, underdifferent light intensities during growth. At the maximum irradiationused (1,800 µE m–2 s–1) the ratio of P700to Cyt f was close to unity, whereas under low irradiation (450µE m–2 s–1) the ratio of P700 to Cyt f rangedfrom two to three. A strikingly positive correlation existedbetween the P700 contents of the leaves and their rates of carbondioxide fixation, dry matter production and Cyt f contents,only when the plants were grown under high light intensities.The P700 content of the leaves in plants grown under low irradiationwas unrelated to the contents of Cyt f. Thus, at a high lightintensity there is a close relationship between the Cyt f andP700 levels, but at low intensities the amounts of electroncarriers and the reaction centre are independent. The fecundity and somatic growth rates of Calanus agulhensisand Calanoides carinatus, the dominant large calanoid copepodsin the southern Benguela upwelling system, as well as the fecundityof several other common copepods, were measured between Septemberand March of 1993/94 and 1994/95. Mean egg production of mostcopepods was low at >30 eggs female-1 day-1 {Calanoides carinatus23.7, Calanus agulhensis 19.0, Neocalanus tonsus 16.1 and Rhincalanusnasutus 26.1), whereas the mean fecundity of Centropages brachiatuswas significantly greater (83.6 eggs female–1 day-1).This study also presents the first comprehensive field estimatesof the fecundity of Nanno-calanus minor (mean: 26.1 eggs female–1day–1, range: 0.0–96.2 eggs female–1 day–1)and of somatic growth of N6 and all copepodite stages of Calanoidescarinatus (decreasing from 0.58 day–1 for N6 to 0.04 day–1for C5). Somatic growth rates of Calanus agulhensis also declinedwith age: from 0.57 day1 for N6 to 0.09 day1 for C5. Data ongrowth rates were used to assess the relative importance offood [as measured by total chlorophyll (Chi) a concentration],phytoplankton cell size (proportion of cells >10 µm)and temperature to the growth of copepods. Multiple regressionresults suggested that fecundity and somatic growth rates werepositively related to both Chi a concentration and phytoplanktoncell size, but not to temperature. Although it was not possibleto separate the effects of Chi a concentration and phytoplanktoncell size, data from previous laboratory experiments suggestthat copepod growth is not limited by small cells per se, butby the low Chi a concentrations that are associated with theseparticles in the field. Despite growth not being directly relatedto temperature, a dome-shaped relationship was evident in somespecies, with slower growth rates at cool (<13°C) andwarm (>18°C) temperatures. The shape of this relationshipmirrors that of Chi a versus temperature, where poor Chi a concentrationsare associated with cool and warm temperatures. It is concludedthat the effect of food limitation on growth of copepods outweighsthat of temperature in the southern Benguela region. Plants of six contrasting genotypes of barley were raised fromvernalized (imbibed at 1 °C for 30 d) or non-vernalizedseeds and grown in 12 different controlled environments comprisingfactorial combinations of three photoperiods (10, 13 and 16h d–1), two day temperatures (18 and 28 °C) and twonight temperatures (5 and 13 °C). Except at longer daysfor Athenais or Arabi Abiad, the 28 °C day temperature wasgenerally supra-optimal and delayed awn emergence. At lowertemperatures and in photoperiods shorter than the critical value,PC, which delay awn emergence, the time from sowing to awn emergencefor five of the genotypes conformed to the equation 1/f=a +bT{macron}+cPwhere f is the time to awn emergence (d), T{macron} is meandiurnal temperature (°C), P is photoperiod (h d–1)and a, b and c are genotype-specific constants. In Arabi Abiad,however, significant responses to temperature were not detected.The low temperature pre-treatment of the seeds reduced the subsequenttime to awn emergence in Athenais and the autumn-sown genotypesAger, Arabi Abiad and Gerbel B, especially in longer days, buteither had no effect or tended to delay awn emergence in thespring-sown types Emir and Mona. In the spring-sown types PCwas outside the range investigated (i.e. > 16 h d–1),but in Ager it was approx. 13 h d–1 and in Gerbel B justover 13 h d–1. Ingestion and clearance rates, feeding behaviors and life historyvariables of the marine cladoceran Penilia avirostris were evaluatedover a range of food concentrations encountered in nature (0.01–3.0mm3 1–1 of Isochrysis galbana). Ingestion rates increasedand clearance rates decreased with increasing food concentration.No maximum feeding thresholds were observed over the range ofalgal concentrations offered. Weight-specific ingestion ratesdecreased with increasing body weight. Feeding behaviors suchas mandibular activity, abdominal claw rejections of cloggedfeeding structures and feeding appendage activity decreasedat a food level of 0.3 mm3 l–1 of l. galbana. Peniliaavirostris had very poor survivorship at extremely low (0.01mm3 l–1 and high (3.00 mm3 l–1) food levels. The amount of fucoxanthin, a taxonomically diagnostic carotenoid,recovered after passage through the guts of the copepods Acartiacalifomiensis and Calanus pacificus, was determined after thecopepods had fed on low (50 µg Cl–1) and high (350µg C1–1 for Acartia; 500 ug C H for Calanus) concentrationsof the diatom Thalassiosira weissftogii, during spring (May)and winter (December). Changes in pigment concentrations andcell abundances were assessed in experimental (with copepods)and control (without copepods) samples by standard incubationexperiments. Pigment recovery was assessed by (i) comparingthe amount of ingested pigment recovered in the experimentalgroups with that predicted to have been ingested from cell countdata and (ii) comparing fuco-xanthin/cell ratios in controland experimental samples. Both techniques suggested that pigmentloss is substantial (usually 60–100%), regardless of species,food availability or season. Patterns of pigment conservationdiffered between species, although pigment recovery was alwayshigher at high, than at low, food concentrations. Pigment recoveryin Acartia was higher (9.4–28.0%) in the spring than duringthe winter (0 recovery), regardless of food concentration. InCalanus, however, pigment recovery was always higher at high(34.9–67.8%) than at low (0 recovery) food concentrations,regardless of season.  相似文献

Nutritional requirements, functional response, development andreproduction of Cyclops vicinus were studied with exclusivelyalgal food. Phytoflagellates were found to be adequate foodresources for both juvenile development and egg production.Ingestion measurements were performed with Chlamydomonas reinhardii.The functional response data give evidence for low feeding efficiency,especially for the naupliar stages. A difference between naupliiand older instars was also found in their quantitative foodneeds. A higher threshold food concentration was observed fornaupliar development (0.4 mg C l–1) than for copepoditedevelopment (0.2 mg C l–1). The calculation of assimilationefficiencies suggests that the high food requirements are dueto low specific ingestion rates rather than poor assimilationefficiency. Development time decreased as algal density increasedand males developed more quickly than females at all food concentrations.Body size and carbon content increased with increasing foodconcentration. Continuous egg production was observed abovea food concentration of 0.5 mg C l–1. The results haveimplications for the life cycle of C. vicinus. Summer diapauseis interpreted as a strategy to avoid starvation of the juvenilestages. During ice-free periods, seston ranges from 10 to 600 and averages29.2 mg dry wt 1–1 in Lake Balaton, the largest shallowlake in Central Europe. Most (80–99%) of the seston consistsof 1–10 µm sized mineral particles. The abiosestoncauses permanent food limitation in the ingestion of the ediblephytoplankton (1–22 µm fraction) by Daphnia cucullataand D.galeata. The postembryonic development time of D.galeataincreases and its fecundity decreases with increasing abioseston.The fecundity and the mortality of D.galeata are balanced atan abioseston concentration of 20.0 mg dry wt 1–1.  相似文献

Acclimation of single plants of Lolium temulentum to changing[CO2] was studied on plants grown in controlled environmentsat 20°C with an 8 h photoperiod. In the first experimentplants were grown at 135 µ;mol m–2 s–1 photosyntheticphoton flux density (PPFD) at 415µl l–1 or 550µll–1 [CO2] with some plants transferred from the lowerto the higher [CO2] at emergence of leaf 4. In the second experimentplants were grown at 135 and 500 µmol m–2 s–1PPFD at 345 and 575 µl l–1 [CO2]. High [CO2] during growth had little effect on stomatal density,total soluble proteins, chlorophyll a content, amount of Rubiscoor cytochrome f. However, increasing [CO2] during measurementincreased photosynthetic rates, particularly in high light.Plants grown in the higher [CO2] had greater leaf extension,leaf and plant growth rates in low but not in high light. Theresults are discussed in relation to the limitation of growthby sink capacity and the modifications in the plant which allowthe storage of extra assimilates at high [CO2]. Our goal was to test our understanding of ingestion, assimilationefficiency and metabolism for Mnemiopsis mccradyi by formulatingand validating a simulation model of growth under differentconditions of food availability. The model was based on a carbonbudget approach using formulations derived from empirical results,including how each process was affected by food availabilityand ctenophore size. An experimentally measured carbon budgetfor pulsed food availability indicated that, relative to totalingestion, growth was high (17–48%), respiration plusorganic release was relatively low (24–48%) and little(<10%) of the ingested carbon was unaccounted for. New laboratoryinvestigations of feeding and assimilation efficiency were necessaryto refine the formulations so that model predictions comparedfavorably with a variety of laboratory measurements of growth,and growth efficiency, as well as the complete experimentallymeasured carbon budget. The refined model predicted a high ratioof growth to metabolism (>2) and a high gross growth efficiency(>30%) for smaller ctenophores at high food concentrations(>20 prey l–1). Both growth rates and growth efficiencieswere predicted to decrease for larger ctenophores. Model predictionswere generally consistent with experimental results, includinginvestigations using pulsed food availability to simulate environmentalpatchiness. Although the model underpredicted ctenophore growthin some experiments at low food densities, the model predictionof a minimum prey concentration of about 8 l–1 (24 µgC l–1) for sustaining a ctenophore population of reproductivesize agreed with field observations.  相似文献

The spawning rate of laboratory-reared Centropages typicus fedHymenomonas elongala increases with food concentration, up toa value of {small tilde}2800 µg C (16 500 cells) ml–1.An alternation of a low food (1000 cells ml–1) and highfood concentration (16 500 cells ml–1) is not favourableto egg release when its periodicity is 1 or 2 days, whereasit may be of advantage if it is longer (3–6 days). The effect of temperature on post-embryonic growth of Neomysisintermedia was investigated under unlimited food conditionsin the laboratory. The effect of temperature on the size ofnewly released animals was negligibly small, but body size wasinversely related to temperature in adults. This was mainlycaused by the difference in the number of molts before maturation.The specific growth rate of N. intermedia increased exponentiallywith a temperature coefficient, Q10 of 4.6 from 0.018 d–13C to 0.21 d–1 at 20C in juveniles, and with a temperaturecoefficient of 2.7 from 0.006 d–1 at 3C to 0.05 d–1at 25C in adults. The rate in juveniles levelled off above20C, and dropped at 29C. Brood size and brood interval decreasedwith temperature increase, while the daily specific reproductionrate increased. The specific growth rate of gravid females,including production of egg matter, increased exponentiallywith a temperature coefficient of 3.3 from 0.015 d–1 at10C to 0.093 d–1 at 25C. The present laboratory experiments confirmed the temperaturecontrol on the growth of N. intermedia suggested in a hyper-eutrophiclake.  相似文献

Selectivity-size spectra, clearance and ingestion rates andassimilation efficiencies of Acartia clausi (Copepoda), Peniliaavirostris (Cladocera) and Doliolum denticulatum (Doliolida)from Blanes Bay (Catalan Sea, NW Mediterranean) were evaluatedin grazing experiments over a wide range of food concentrations(0.02–8.8 mm3 L–1 plankton assemblages from BlanesBay, grown in mesocosms at different nutrient levels). Acartiaclausi reached the highest grazing coefficients for large algae>70 µm (longest linear extension), P. avirostris forintermediate food sizes between 15 and 70 µm, and D. denticulatumfor small sizes from 2.5 to 15 µm. Penilia avirostrisand D. denticulatum acted as passive filter-feeders. Acartiaclausi gave some evidence for a supplementary raptorial feedingmode. Effective food concentration (EFC) decreased linearlywith increasing nutrient enrichment for D. denticulatum andfollowed domed curves for A. clausi and for P. avirostris withmaximum values at intermediate and high enrichment levels, respectively.Clearance rates of crustacean species showed curvilinear responseswith narrow modal ranges to increasing food concentration. Clearancerates of D. denticulatum increased abruptly and levelled intoa plateau at low food concentrations. Mean clearance rates were13.9, 25.5 and 64.1 mL ind.–1 day–1, respectively.No clearance could be detected for A. clausi at food concentrations<0.1 mm3 L–1 and for P. avirostris at food concentrations  相似文献

Food limitation for the cladoceran Daphnia galeta was measuredgrowing them with natural lake seston for one growing seasonunder standardized conditions in the laboratory. Growth rateswere related to several measures of food quantity. Particulateorganic carbon (POC) <30 µm was the best predictorof total food availability measured. It was better than chlorophyll-a(Chl-a) of the same size fraction, algal volume calculated fromcell counts or total particle volume from Coulter Counter measurements.Daphnids were also grown with Scenedesmus acutus under the sameconditions and their growth rates were compared. Sestonic carbonconcentrations were related to Scenedesmus carbon concentrations,that provided equal growth rates. The ratio of the carbon concentrationsof both food types (seston, Scenedesmus) represented the foodquality of seston. Thus, the nutritional quality of naturallake seston relative to a standard alga was determined overone growing season. Growth rates of seston fed animals revealedthat they were food limited during long periods of the year.This was also ascertained by standard clutch sizes of fieldanimals. However, the causes of food limitation varied withinthe duration of the studied period. Whereas during the clear-waterphase the food quantity was limiting (while the quality washigh: 100% Scenedesmus), shortly after on 10 June and 17 Junefood conditions were close to threshold concentrations due tolow food quality. Investigations of the reproductive biology of Cyclops vicinusrevealed that mated females oscillate between gravid and nongravidreproductive conditions. The gravid condition can be dividedin two recognizable phases: gravid/nonovigerous and gravid/ovigerous,the former phase being shortest. The maturation of new oocytestakes place when the old egg sacs are still being carried; thisensures a rapid clutch succession. Females which remain unmated,extrude few eggs, in no case complete egg sacs, and remain gravidthus conserving oocytes. Females which mate only once, showa similar reproductive pattern (clutch size and clutch succession)to those which remain combined with males, and thus have theopportunity to remate, but tend to produce fewer clutches. Malesare able to fertilize 3–4 females day–1. Matingcapacity of males is possibly limited by the time needed tofill a new spermatophore. Short-term starvation (5 days) lengthenedclutch-to-clutch periods and diminished clutch size. When thestarvation period started in the gravid/ovigerous phase, a normalclutch was extruded but no new oocytes matured during starvation,indicating that the energy for egg maturations is provided inthe first part of the reproductive cycle.  相似文献

Dark-adapted intact spinach chloroplasts exhibited two peaks,P and M1, at the early phase of fluorescence induction and atransient reduction of cytochrome f shortly after its initialphotooxidation and in parallel to the appearance of P. Analysisof the peak P and the transient reduction of cytochrome f indicatedthat electron transport in intact spinach chloroplasts was regulatedby light: electron transport was inactivated at the reducingside of photosystem I in the dark-adapted chloroplasts but rapidlyreactivated by illumination. The fluorescence peak M1 was correlatedto the proton gradient formed across the thylakoid membrane. Effects on P and transient reduction of cytochromef of NO2,3-phosphoglycerate (PGA) and oxalacetate (OAA), which can penetrateinto intact chloroplasts and accept electrons at different sitesafter photosystem I, were studied to determine the site of thelight regulation. NC2, which receives electrons fromreduced ferredoxin, markedly diminished both P and the transientreduction of cytochrome.f, whereas PGA and OAA, the reductionsof which are NADP-dependent, failed to affect the two transients.The ineffectiveness of PGA and OAA could not be attributed tothe dark inactivation of glyceraldehyde-3-phosphate and malicdehydrogenases, because dark-adapted chloroplasts still retainedsufficiently high levels of the enzyme activities. The resultsindicate that electron transport in intact spinach chloroplastsis regulated by light after ferredoxin but before NADP, i.e.,at the reducing terminal of the electron transport chain. The relationship between thef-ratio [NO3 uptake/(NO3+ NH4+) uptake] and ambient nitrate concentration was evaluatedfor eight data sets from coastal waters. The f-ratio increasedasymptotically with increase in nitrate concentration in mostdata sets. However, the rate at which f-ratio increased at lownitrate concentration (slope = m) and the maximum attained f-ratio(fmax) varied among regions; the initial slope varied most withvalues ranging in excess of an order of magnitude. The datawere analyzed in relation to environmental factors and methodologicalconsiderations known to influence the f-ratio. Ambient ammoniumconcentration was important in accounting for regional differencesin the f versus NO3 relationship. A further analysisof the data, relating f-ratio to the ratio of NO3/(NO3+ NH4+) concentrations yielded a much more regionally consistentand approximately linear relationship; slopes varied by lessthan a factor of two in the extreme cases. Inclusion of knownalternative (aside from NH4+) sources of reduced-N (e.g. urea)and correction for methodological/computational errors (isotopedilution) systematically reduce f-ratio estimates. Other factors,e.g. reduced-N uptake by microheterotrophs, may systematicallyincrease the f-ratio.  相似文献

The population carbon budget and seasonality of Boeckella minutain a newly formed subtropical reservoir were examined 3 yearsafter the reservoir filled. Average daily biomass was 26.4 mgC m–3 and the annual population carbon budget was: consumption2470, egestion 1482, assimilation 988, production 493 and respiration495, mg C m–3 year–1, and the average P/B and P/Aratios were 0.08 and 0.5 respectively. Clutch size and reproductiveeffort (egg production/assimilation) were low, and the proportionof males decreased throughout the population cycle. The seasonalabundance pattern changed from perennial (pre-filling years)to a 7 month cycle. It is suggested that eutrophication andthe spring bloom of cyanobacteria may have accentuated a seasonaldecrease in reproductive effort and survival, leading to anabsence of planktonic stages during summer, and that restingeggs facilitated population survival during the summer periodof stratification.  相似文献

The number, size (mm3 pellet –1) and total volume of fecalpellets produced (mm3 h–1) by Boeckella titicacae increasedwith concentration of Chlorella and natural seston in threedifferent lakewaters from Bahia de Puno and Lago Grande of LakeTiticaca and nearby Laguna Arapa. The three lakewaters differedin the size and number of fecal pellets produced at seston concentrations>0.5 mg dry wt l–1. However, the total volume of fecalpellets egested varied little. Large-sized food (>10 µg)resulted in larger pellets but smaller total fecal production.Food concentration had parallel effects on the production offeces and on feeding rates. Both fecal production and feedingincreased linearly with food concentration with saturation between5 and 7.5 x 105 Chlorella ml–1 (3.7–5.6 mg dry wtl–1). Varied responses in fecal production when fed differentsize fractions of lake seston suggest Boeckella has a complexand flexible feeding behavior. Feeding preferences of B.titicacaeand B.occidentalis (Ivlev index) for two seston size fractionsare generally reflected by fecal production.  相似文献
