目的:探讨白介素-18(IL-18)基因启动子区-137G/C(rs187238)位点和-607A/C(rs1946518)位点的等位基因、基因型、单体型与黑龙江省汉族人群心房颤动发病风险的相关性。方法:选取56例心房颤动患者和26例对照者,心房颤动患者按持续时间分为阵发房颤组和持续房颤组。采用聚合酶链式反应(PCR)和直接测序法(DS)对所选2个SNPs位点的基因型进行检测。结果:1黑龙江省地区汉族人群中IL-18基因启动子区-607A/C位点存在AA、AC、GG三种基因型,-137C/G位点存在CC、GC、GG三种基因型。2各心房颤动患者组与对照组间IL-18基因启动子区-137G/C(rs187238)位点和-607A/C(rs1946518)位点的基因型和等位基因频率比较均无显著性差异(P0.05)。3IL-18基因启动子区-137G/C(rs187238)位点和-607 A/C(rs1946518)位点有CA、CC、GA、GC四种单倍体型,各组单倍体型分布频率比较均无统计学差异(P0.05。4IL-18基因启动子区-137G/C(rs187238)位点和-607 A/C(rs1946518)位点的基因型和等位基因频率与AF患者的发病年龄均无统计学相关性(P0.05)。结论:IL-18基因启动子区-607A/C(rs1946518)位点和-137G/C(rs187238)位点不是黑龙江省汉族人群心房颤动的易感基因,可能与其心房颤动的发病风险无关。  相似文献

目的:分析广西壮族人群EBI3基因rs6613A/T、rs4905A/G多态性分布特点。方法:采用单碱基延伸的PCR技术对168例广西壮族人群EBI3 rs6613 A/T和EBI3 rs4905A/G进行多态性检测,对比国际人类基因组计划(Hap Map)公布的中国北京人、日本人、非洲人和意大利人的SNP分型数据,分析5个人群rs6613 A/T、rs4905A/G位点的基因型和等位基因频率差异。结果:在广西壮族人群中,EBI3基因rs6613 A/T位点AT基因型最常见,约为49.4%;T等位基因频率最高,约为52.1%;rs4905A/G多态性位点AC基因型最常见,约为48.2%;C等位基因频率最高,约为50.9%。EBI3基因型及等位基因频率分布于性别无显著相关性(P0.05)。广西壮族人群EBI3基因rs6613A/T位点基因型和等位基因频率与北京人差异无统计学意义(P0.05),但与非洲人、日本人、意大利人差异具有统计学意义(P0.05);EB-13基因rs4905A/G位点基因型和等位基因频率与北京人和日本人差异无统计学意义(P0.05),但与非洲人和意大利人比较差异具有统计学意义(P0.01)。结论:EBI3基因rs6613 A/T和EB-13 rs4905A/G多态性位点基因型和等位基因在广西壮族人群中的分布频率与其他种族和地区人群相比存在差异,这种差异可能是导致某些疾病在不同人群发病率和临床表现存在差异的原因之一。  相似文献

目的:探讨BRCA1基因启动子区rs11655505、rs73625095位点单核苷酸多态性与散发性乳腺癌易感性的关系。方法:采用ASA-PCR方法对200例乳腺癌患者(均经病理确诊)及200例正常女性BRCA1基因启动子区rs11655505(A/G)、rs73625095(A/G)位点单核苷酸多态性(SNP)进行分析,并将其PCR产物进行测序。结果:乳腺癌患者BRCA1基因启动子区rs11655505位点的A/G基因型频率为75%,显著高于正常人的40%;A/A基因型频率为7%,G/G基因型频率为18%,分别低于正常人的30%、30%。此位点的A或G等位基因在乳腺癌病例组及对照组中均无差别(x2=2.427,P=0.119);rs73625095位点的A/G基因型频率为68%,显著高于正常人的15%;G/G基因型频率为32%,低于正常人的84%;乳腺癌病例组中BRCA1基因启动子区rs11655505、rs73625095位点的A/G基因型与淋巴结转移与否相比,差别均有统计学意义(x2=7.321,P=0.026、x2=4.782,P=0.029)。结论:BRCA1基因rs11655505位点、rs736...  相似文献

探讨白介素17A基因多态性与胃癌预后的关系。129例研究对象纳入生存分析,分成死亡和存活两组,用基因测序的方法检测血液样本IL-17A基因SNP位点rs3748067、rs17880588基因型分布。rs3748067位点有3种基因型T/T、C/T、C/C,rs17880588位点有2种基因型A/G、G/G。比较存活组和死亡组之间的基因型分布频率和单点等位基因分布频率,发现rs3748067的基因型C/T在死亡组的分布频率较存活组高,基因型T/T和C/C在死亡组的分布频率低于存活组,两组之间分布频率差异有统计学意义(P0.05)。杂合型C/T基因型在存活组分布低于死亡组(OR=2.051,CI=0.016~1.420),该位点基因型杂合可能为胃癌预后的一种危险因素。rs17880588的两种基因型A/G、G/G在存活组和死亡组的分布频率差异无统计学意义(P0.05)。结论:IL-17A基因rs3748067位点SNP与胃癌预后可能有相关性。  相似文献

目的:研究新疆地区HPV16 E6、E7、LCR基因突变情况,分析HPV16变异体在宫颈癌及癌前病变发生发展中的作用。方法:选择HPV16阳性的宫颈癌及癌前病变患者,提取基因组DNA,利用PCR扩增HPV16 DNA E6、E7基因及LCR区核苷酸片段,正反向测序。与HPV16基因序列分析比对,分析核苷酸突变位点。结果:E6基因突变率为80.00%(92/115)主要突变位点T350G(59.78%)、T178G(18.47%);E7突变率为54.78%(63/115),主要突变位点A647G(33.33%)、T846C(26.98%);LCR突变率为23.48%(27/115),主要突变位点为C24T(74.07%)、C13T(25.92%)。维吾尔族T350G突变率较汉族妇女显著升高,而汉族A647G、T846C、C24T突变率显著高于维吾尔族,差异具有统计学意义(P0.05)。维吾尔族宫颈癌组T350G突变率显著高于炎症组(P0.05),且随病变严重程度增加突变率上升,汉族T350G、A647G、T846C、C24T突变率炎症组、宫颈病变组显著高于宫颈癌组(P0.05),维吾尔族C24T突变率炎症组显著高于宫颈癌组(P0.05),差异均具有统计学意义(P0.05)。结论:HPV16E6、E7突变可能与宫颈病变进展有关,T350G突变可能是维吾尔族宫颈癌高发的原因之一。  相似文献

目的系统分析江浙沪三地的乙肝病毒B、C基因型S基因突变和选择压力情况,以期为乙型肝炎的防治提供理论依据。方法采用NCBI数据库提供的乙肝病毒序列,分为B、C基因型两组,分析突变情况,同时采用Datamonkey进行选择压力分析和Bioedit进行氨基酸置换熵值分析。结果S基因226个氨基酸位点突变分析中,产生突变有位点122个,位点突变率C基因型(45.58%)高于B基因型(30.09%)(χ~2=11.523,P=0.001);总突变率C基因型(1.36%)高于B基因型(0.80%)(χ~2=46.642,P=0.000);α决定簇突变率C基因型(2.40%)高于B基因型(0.96%)(χ~2=20.524,P=0.000)。B基因型α决定簇突变率高于总突变率(χ~2=0.735,P=0.391);C基因型α决定簇突变率高于总突变率(χ~2=44.467,P=0.000)。B基因型氨基酸突变最多的位点为200、213、161和21位点,C基因型氨基酸突变最多的位点为126、68、3、53和194位点。两个基因型dN/dS均值均小于1。B基因型没有发现正向选择位点,发现8个负向选择位点。C基因型发现7个正向选择位点,17个负向选择位点。B基因型仅发现一个易突变位点(200位),C基因型也仅发现一个易突变位点(126位),大多数氨基酸位点熵值0.4。结论江浙沪地区S基因突变率水平较低,其中C基因型位点突变率、α决定簇突变率和总突变率高于B基因型,α决定簇突变率高于总突变率。C基因型经历外界环境免疫选择压力和自身进化压力双重影响,自身进化压力强于外界环境免疫选择压力;而B基因型主要遭受自身进化压力。尤其要格外关注C基因型的进化进程。  相似文献

为了探讨一种具有玉型血小板反应蛋白的去整合素和金属蛋白酶12 (ADAMTS-12)基因rs25754G/A单核苷酸多态性(SNPs)与颈动脉斑块稳定性的关系,本研究选择78例颈动脉易损斑块患者和101例颈动脉稳定斑块患者作为本次实验的受试对象,应用荧光标记-限制性内切酶酶切法对ADAMTS-12基因rs25754G/A位点进行SNPs基因型检测,运用SPSS16.0统计学软件进行等位基因和基因型频数分布的分析。研究结果显示,颈动脉易损斑块组ADAMTS-12基因rs25754G/A多态位点G+AA基因型和A等位基因频率(98.72%和87.82%)明显高于颈动脉稳定斑块组(90.10%和62.38%)(p0.05)。本研究初步结论表明,ADAMTS-12基因rs25754G/A多态位点与颈动脉易损斑块的发生有相关性。  相似文献

目的:研究中国北方人群TCF7L2基因rs11196218和rs290487多态性的分布特点及其与血脂谱的相关性。方法:对1255例中国北方人群TCF7L2基因进行单核苷酸多态性检测,同时检测其血脂水平,分析血脂水平与上述基因的相关性。结果:在中国北方人群中TCF7L2基因rs11196218位点AA,AG和GG基因型频率分别为6.61%,39.68%和53.71%,等位基因A、G频率分别为26.45%和73.55%;而rs290487位点TT,CT和CC基因型频率分别为37.45%,45.98%和16.57%,等位基因T,C频率分别为60.44%和39.56%。rs11196218A/G与血清低密度脂蛋白、总胆固醇水平具有相关性(P≤0.05),而rs290487C/T与血脂水平无相关性(P0.05)。结论:在中国北方人群中存在TCF7L2基因rs11196218A/G和rs290487C/T单核苷酸多态性,且其变异频率大,rs11196218A/G与血脂异常相关。  相似文献

目的:探讨新疆地区维吾尔族和汉族草酸钙结石与钙敏感受体(calcium sensitive receptor,Ca SR)基因多态性之间的关系。方法:选择398例临床确诊泌尿系草酸钙结石患者(200例维吾尔族,198例汉族)和399例正常对照者(200例维吾尔族,199例汉族),应用Sna Pshot方法对Ca SR基因两位点(rs1042636,rs1801726)的基因型及等位基因频率进行检测,并分析其与草酸钙结石发病的相关性以及对血钙、24 h尿钙水平的影响。结果:各组2个位点的基因型分布均符合Hardy-Weinberg平衡。汉族结石组与汉族对照组及维吾尔族结石族与维吾尔族对照组rs1042636、rs1801726位点基因型分布及基因频率差异均无统计学意义(P0.05)。维吾尔和汉族rs1042636基因型及等位基因频率比较差异有统计学意义(P0.05),且维吾尔族人群携带rs1042636等位基因A的风险高于汉族人群(病例组中OR值=2.145,%95CI=[1.602~2.866],P0.01;对照组中OR值=1.773,%95CI=[1.332~2.359],P0.01),其中维/汉病例组中等位基因频率分别为A=278(69.5%)/204(51.5%),G=122(30.5%)/192(48.5%);维/汉对照组中等位基因频率分别为A=264(66.0%)/208(52.3%),G=136(34.0%)/190(47.7%)。而病例组和对照组rs1801726基因型频率差异无统计学意义(P0.05);汉族病例组、对照组发现GG+AG基因型较AA基因型有较高的尿钙水平(病例组:P=0.007和对照组:P=0.006),维吾尔族人群该位点与两项指标无相关性。结论:Ca SR基因2个基因位点rs1042636、rs1801726可能不是新疆地区维吾尔族和汉族草酸钙结石发病的危险因子,两族rs1042636基因多态性分布存在差异,rs1042636位点基因多态性能影响汉族人群尿钙排泄,可能汉族调节钙排泄的遗传因素之一。  相似文献

目的:探讨白血病融合基因亲嗜性病毒整合位点1(ecotropic viral integration site-1,EVI1)的多态性与白血病发生风险的相关性。方法:选取本院2017年2月~2019年2月收治的骨刺患儿90例作为研究组,同期选择健康人群83例作为对照组。清晨空腹抽取两组入选者的外周静脉血2 mL,采用PCR方法检测两组入选者EVI1的多态性情况,调查一般资料并进行相关性分析。结果:EVI1 rs17561基因共有CC、CA、AA三种基因型,两组入选者的EVI1 rs17561基因分布均符合Hardy-Weinberg平衡定律,研究对象具有群体代表性。两组入选者EVI1 rs17561基因型分布差异具有统计学意义(P0.05),研究组的EVI1 rs17561基因CC基因型显著高于对照组(90.0%vs. 75.9%, P0.05),研究组的等位基因C频率(显著高于对照组(96.7%vs. 80.7%, P0.05)。在90例骨刺患儿中,6例患儿确诊为白血病,检出率为6.7%,均为CC基因型。研究组患儿EVI1 rs17561基因的CC基因型与血小板计数、危险度分层、诊断分型显著相关(P0.05)。多元Logistic回归分析显示血小板计数、危险度分层、诊断分型为影响EVI1rs17561CC基因型的主要因素(P0.05)。结论:白血病患儿融合基因EVI1多态性比较常见,多表现为rs17561CC等位基因,此等位基因可能与白血病患者的血小板计数、危险度分层、诊断分型显著相关,其中血小板计数、危险度分层、诊断分型为影响EVI1rs17561CC基因型的主要因素。  相似文献

Objective

Sepsis is an inflammatory syndrome caused by infection, and both its incidence and mortality are high. Because interferon-gamma (IFN-γ) plays an important role in inflammation, this work assessed IFN-γ single nucleotide polymorphism (SNPs) that may be associated with sepsis.

Methods

A total of 196 patients with pneumonia-induced sepsis and 213 age- and sex-matched healthy volunteers participated in our study from July 2012 to July 2013 in Guangzhou, China. Patient clinical information was collected. Clinical pathology was assessed in subgroups defined based on clinical criteria, APACHE II (acute physiology and chronic health evaluation) and SOFA (sepsis-related organ failure assessment) scores and discharge rate. Four functional SNPs, −1616T/C (rs2069705), −764G/C (rs2069707), +874A/T (rs2430561) and +3234C/T (rs2069718), were genotyped by Snapshot in both sepsis patients and healthy controls. Pearson’s chi-square test or Fisher’s exact test were used to analyze the distribution of the SNPs, and the probability values (P values), odds ratios (OR) and 95% confidence intervals (CIs) were calculated.

Results

No mutations in the IFN-γ −764G/C SNP were detected among the participants in our study. The +874A/T and +3234C/T SNPs were in strong linkage disequilibrium (LD) (r2 = 0.894). The −1616 TC+TT, +874 AT+AA genotype and the TAC haplotype were significantly associated with sepsis susceptibility, while the CTT haplotype was associated with protection against sepsis incidence. Genotype of −1616 TT wasn’t only protective against severity of sepsis, but also against higher APACHE II and SOFA scores as +874 AA and +3234 CC. The TAC haplotype was was protective against progression to severe sepsis either.

Conclusion

Our results suggest that functional IFN-γ SNPs and their haplotypes are associated with pneumonia-induced sepsis.  相似文献

According to previous investigations, CD14 is suggested to play a pivotal role in initiating and perpetuating the pro-inflammatory response during sepsis. A functional polymorphism within the CD14 gene, rs2569190, has been shown to impact the pro-inflammatory response upon stimulation with lipopolysaccharide, a central mediator of inflammation in sepsis. In this study, we hypothesized that the strong pro-inflammatory response induced by the TT genotype of CD14 rs2569190 may have a beneficial effect on survival (30-day) in patients with sepsis. A total of 417 adult patients with sepsis (and of western European descent) were enrolled into this observational study. Blood samples were collected for rs2569190 genotyping. Patients were followed over the course of their stay in the ICU, and the 30-day mortality risk was recorded as the primary outcome parameter. Sepsis-related organ failure assessment (SOFA) scores were quantified at sepsis onset and throughout the observational period to monitor organ failure as a secondary variable. Moreover, organ support-free days were evaluated as a secondary outcome parameter. TT-homozygous patients were compared to C-allele carriers. Kaplan-Meier survival analysis revealed a higher 30-day mortality risk among C-allele carriers compared with T homozygotes (p = 0.0261). To exclude the effect of potential confounders (age, gender, BMI and type of infection) and covariates that varied at baseline with a p-value < 0.2 (e.g., comorbidities), we performed multivariate Cox regression analysis to examine the survival time. The CD14 rs2569190 C allele remained a significant covariate for the 30-day mortality risk in the multivariate analysis (hazard ratio, 2.11; 95% CI, 1.08-4.12; p = 0.0282). The 30-day mortality rate among C allele carriers was 23%, whereas the T homozygotes had a mortality rate of 13%. Additionally, an analysis of organ-specific SOFA scores revealed a significantly higher SOFA-Central nervous system score among patients carrying the C allele compared with T-homozygous patients (1.9±1.1 and 1.6±1.0, respectively; p = 0.0311). In conclusion, CD14 rs2569190 may act as a prognostic variable for the short-term outcome (30-day survival) in patients with sepsis.  相似文献

目的:探讨北部湾人群C型凝集素-1(Dectin-1)基因多态性与马尔尼菲青霉菌病易感性的相关性。方法:选取北部湾地区的马尔尼菲青霉菌(PM)病患者71例为病例组,另选北部湾地区的71例体检正常者为对照组,直接测序检测rs16910526、rs16910527位点的基因型及等位基因频率,并分析其与马尔尼菲青霉菌病易感性的相关性。结果:(1)对照组和病例组之间rs16910526有三种基因型GG、GT、TT,两组之间基因型和等位基因频率比较差异不显著(P0.05)。(2)对照组和病例组之间rs16910527有三种基因型AA、AC、CC,且病例组AC的基因型频率显著高于对照组(P0.05)。(3)局限性、播散性PM患者rs16910526、rs16910527基因型和等位基因频率比较差异不显著(P0.05)。(4)rs16910526、rs16910527的4种单倍型:GT、AC、AT、TT,位于同一连锁不平衡区域内,且对照组和病例组A/C的分布频率比较差异具有统计学意义(P0.05)。结论:北部湾人群Dectin-1的rs16910527位点与马尔尼菲青霉菌病易感性相关,且A/C能提高马尔尼菲青霉菌病的易感性。  相似文献

摘要 目的:探讨与分析血清白细胞介素8(IL-8)基因多态性与食管鳞癌(ESCC)根治术后的相关性。方法:2017年8月到2020年6月选择在本院诊治的食管鳞癌患者98例作为研究对象,检测血清IL-8表达水平。所有患者都给予根治手术治疗,随访患者的预后并进行相关性分析。结果:所有患者术后随访到2021年7月,平均随访时间为25.69±2.58个月,死亡28例,死亡率为28.6 %(死亡组)。两组血清IL-8表达水平表达具有差异(P<0.05)。所有患者的基因型频率均符合Hardy-Weinberg这一平衡法则,表明本文所选取的样本均具有群体代表性。IL-8基因启动子rs4073A/T的AA基因型较死亡组高,TT基因型较死亡组低,两组A、T等位基因频率分布对比有差异(P<0.05)。直线相关性分析显示:IL-8基因启动子rs4073A/T的AA基因型、A等位基因、血清IL-8表达水平与预后死亡率存在相关性(P<0.05)。多因素logistic回归分析显示:IL-8基因启动子rs4073A/T的AA基因型、A等位基因、血清IL-8表达水平为导致患者随访死亡的主要因素(OR=2.051,3.094,P<0.05)。结论:食管鳞癌根治术后患者依然存在一定的死亡率,患者死亡与血清IL-8基因多态性存在相关性,同时多伴随有IL-8的高表达。IL-8基因启动子rs4073A/T的AA基因型、A等位基因、血清IL-8表达水平为导致患者死亡的主要因素。  相似文献

Background

The protein C pathway plays an important role in the maintenance of endothelial barrier function and in the inflammatory and coagulant processes that are characteristic of patients on dialysis. We investigated whether common single nucleotide variants (SNV) in genes encoding protein C pathway components were associated with all-cause 5 years mortality risk in dialysis patients.

Methods

Single nucleotides variants in the factor V gene (F5 rs6025; factor V Leiden), the thrombomodulin gene (THBD rs1042580), the protein C gene (PROC rs1799808 and 1799809) and the endothelial protein C receptor gene (PROCR rs867186, rs2069951, and rs2069952) were genotyped in 1070 dialysis patients from the NEtherlands COoperative Study on the Adequacy of Dialysis (NECOSAD) cohort) and in 1243 dialysis patients from the German 4D cohort.

Results

Factor V Leiden was associated with a 1.5-fold (95% CI 1.1–1.9) increased 5-year all-cause mortality risk and carriers of the AG/GG genotypes of the PROC rs1799809 had a 1.2-fold (95% CI 1.0–1.4) increased 5-year all-cause mortality risk. The other SNVs in THBD, PROC, and PROCR were not associated with 5-years mortality.

Conclusion

Our study suggests that factor V Leiden and PROC rs1799809 contributes to an increased mortality risk in dialysis patients.  相似文献

This study was undertaken to investigate whether sequential measurement of blood interleukin (IL)-6 levels using chemiluminescent enzyme immunoassay (CLEIA) would be useful for the management of patients with systemic inflammatory response syndrome (SIRS)/sepsis. Forty consecutive patients with SIRS/sepsis admitted to ICU were involved in the study. Blood IL-6 level was measured everyday throughout their ICU stay at the clinical laboratory by CLEIA method. The platelet count and the sequential organ failure assessment (SOFA) score were measured consecutively. The blood IL-6 levels were elevated in SIRS/sepsis patients and were extremely high in patients with septic shock. There was no significant difference in the blood IL-6 level on admission between survivors (n=27) and non-survivors (n=13). However, the mean blood IL-6 level during ICU stay was significantly higher in the non-survivors (p<0.05). There were significant correlation between the peak IL-6 blood level and the lowest platelet count, and between the peak IL-6 blood level and the maximum SOFA score, respectively. The platelet count became lowest 2.0+/-2.0 days later on average, and the SOFA score became maximal 2.5+/-1.4 days later on average following the day when IL-6 reached its peak value. Sequential measurement of blood IL-6 levels by CLEIA is useful in evaluating the severity and in predicting the outcome of the patients with SIRS/sepsis.  相似文献

Genetic and environmental factors are important for the development of nonalcoholic fatty liver disease (NAFLD). The aim of the present study was to examine the single nucleotide polymorphism (SNP) -129C/T (rs17883901) in glutamate-cysteine ligase catalytic subunit (GCLC) and SNPs I128T (rs3816873) and Q95H (rs61733139) in microsomal triglyceride transfer protein (MTTP) in NAFLD. Eighty-three patients with a diagnosis of NAFLD and 93 healthy subjects were included in the study. Tetra amplification refractory mutation system-polymerase chain reaction was designed to detect the SNPs. There were no significant differences in the polymorphism of -129C/T (rs17883901) of the GCLC gene among NAFLD and control groups (p?>?0.05).
