Abstract

Phyto-recurrent selection is an established method for selecting tree genotypes for phytoremediation. To identify promising Populus (poplar) and Salix (willow) genotypes for phytotechnologies, our objectives were to (1) evaluate the genotypic variability in survival, height, and diameter of poplar and willow clones established on soils heavily contaminated with nitrates; and (2) assess the genotypic stability in survival and diameter of selected poplar clones after one and eleven growing seasons. We tested 27 poplar and 10 willow clones planted as unrooted cuttings, along with 15 poplar genotypes planted as rooted cuttings. The trees were tested at an agricultural production facility in the Midwestern, United States. After 11 growing seasons, using phyto-recurrent selection, we surveyed survival and measured the diameter of 27 poplar clones (14 unrooted, 13 rooted) that were selected based on superior survival and growth throughout plantation development. Overall, willow exhibited the greatest survival, while poplar had the greatest height and diameter. At 11 years after planting, superior clones were identified that exhibited above-average diameter growth at the establishment- and rotation-age, most of which had stable genotypic performance over time. Selection of specific clones was favorable to genomic groups, based on the geographic location and soil conditions of the site.  相似文献

Cadmium accumulation and tolerance in Populus nigra and Salix alba   总被引:1,自引:0,他引:1  
Rooted cuttings of Populus nigra L. clone Poli and Salix alba L. clone SS5 were treated for three weeks with 50 μM CdSO4 in nutrient solution. The willow showed a far higher Cd tolerance, expressed as tolerance index (Ti), than the poplar in both roots and leaves. The root Cd content was higher in poplar than in willow, whereas in leaves the opposite was found. As a consequence, the translocation factor (Tf) revealed a greater ability of Cd transport in willow than in poplar. Cd treatment enhanced cysteine, γ-glutamylcysteine and reduced glutathione contents in roots of both species, whereas in leaves they were only enhanced in poplar. Furthermore, only poplar leaves showed an enhanced content of phytochelatins, whereas malic and citric acids rose in response to Cd only in the willow leaves. Cd treatment increased putrescine, spermidine and spermine contents in both roots and leaves of the willow, whereas in poplar only the putrescine content was enhanced in roots.  相似文献

The objective of this study was to evaluate seven woody plant species on four growth media for their potential contribution to moose habitat and establishment of viable plant communities on a proposed mine site in southcentral Alaska. Populus balsamifera (balsam poplar), Salix alaxensis (feltleaf willow), S. barclayi (Barclay willow), S. bebbiana (Bebb willow), Alnus tenuifolia (thinleaf alder), Betula papyrifera (paper birch), and Picea glauca (white spruce) were selected for their functions in moose habitat, ease of propagation, and presence in the existing native vegetation. Three native soils were selected for biological characteristics such as different potential to form mycorrhizae and to regenerate local plant species, both of which are governed partly by existing vegetation. The fourth growth medium, glacial till or overburden, was expected to have little or no biological activity. A mining disturbance was simulated on three sites by removing existing vegetation from the plots, stripping the native soils, and then spreading these soil materials over the respective study plots. Rooted cuttings of the Salicaceae and nursery seedlings of the other species were planted in each of the four growth media. Height and survival of all plant species were greater on the three soil media than on the glacial till during the second and third years. Percentage of ectomycorrhizal infection on transplants was similar among growth media, although lower ectomycorrhizal infection occurred on volunteers in grassland soils than in the other growth media. Browsable plants were produced within three years on the disturbed native soils but not on the glacial till.  相似文献

Species and hybrids between species belonging to the genera Populus (poplar) and Salix (willow) have been used successfully for phytoremediation of contaminated soils. Our objectives were to: 1) evaluate the potential for establishing genotypes of poplar and willow on soils heavily contaminated with petroleum hydrocarbons and 2) identify promising genotypes for potential use in future systems. We evaluated height, diameter, and volume after first year budset by testing 20 poplar clones and two willow clones. Unrooted cuttings, 20 cm long, were planted in randomized complete blocks at 0.91- x 0.91-m spacing at Gary, IN, USA (41.5 degrees N, 87.3 degrees W). Four commercial poplar clones (NM6, DN5, DN34, and DN182) were planted as 20- and 60-cm cuttings. Sixty-cm cuttings exhibited greater height and diameter than 20-cm cuttings; however, we recommend continued use and testing of different combinations of genotype and cutting length. We identified promising genotypes for potential use in future systems and we recommend allocating the majority of resources into commercial poplar clones, given their generalist growth performance. However, further utilization and selection of experimental clones is needed. Specific clones rather than genomic groups should be selected based on the geographic location and soil conditions of the site.  相似文献

Salix alba L. and Populus×euroamericana cv. Robusta cuttings were grown in 10 μM Cd(NO3)2 (direct treatment) or in Knop solution and afterwards in Cd(NO3)2 (indirect treatment). Cd impact on rooting of directly treated plants and its impact on normally formed roots and shoots of indirectly treated plants were studied. The cumulative length, number and biomass of willow roots, pigment and starch contents, leaf net photosynthetic rate and dry mass/leaf area ratio of willow leaves were positively influenced by indirect treatment. However, indirectly treated poplars were more sensitive to Cd than directly treated ones. Indirect treatment lowered root Cd uptake in willow, Cd accumulation in cuttings of both species and Cd accumulation in poplar shoots. Cd-caused structural changes were similar in both species and in both treatments. Root apices, rhizodermis and cortex were the most seriously damaged root parts. In directly treated willow, the structure of central cylinder (0.5 – 1 cm from apex) remained unchanged in contrast to indirectly treated plants. Formation of cambium close to the apex indicated shortening of root elongation zone of indirectly treated plants. Directly Cd-treated poplar roots exhibited unusual defence activity of root apical meristem and accumulation of darkly stained material around central cylinder. Efficient in vitro regeneration systems for Vaccinium species   总被引:1,自引:0,他引:1  
Efficient protocols for shoot regeneration from leaf explants suitable for micropropagation as well as for the development of transgenic plants were developed for blueberry (Vaccinium corymbosum) and lingonberry (Vaccinium vitis-idaea) cultivars. Nodal segments were used to initiate in vitro shoot cultures of lingonberry cultivar ‘Red Pearl’ and southern highbush blueberry cultivar ‘Ozarkblue’. In order to develop an optimized regeneration procedure, different types and concentrations of plant growth regulators were tested to induce adventitious shoot regeneration on excised leaves from micropropagated shoots of both cultivars. The effect on percentage regeneration and number of shoots per explant was investigated. Results indicated that zeatin was superior to TDZ and meta-topolin in promoting adventitious shoot formation. A concentration of 20 μM zeatin was most effective in promoting shoot regeneration in both cultivars, in case of ‘Red Pearl’ along with 1 μM NAA. Shoots were either allowed to root in vitro on medium containing IBA or NAA or ex vitro in a fog tunnel. IBA was superior to NAA for induction of root development in vitro in both Vaccinium cultivars. Ex vitro rooting under high humidity was tested with cuttings from mature field-grown plants, from acclimatized tissue culture derived plants and with unrooted in vitro proliferated shoots planted directly. It was found that in vitro shoots rooted better under fog than cuttings from the other plant sources and rooting was equivalent to that achieved in vitro.  相似文献

A common approach to re‐establishing cottonwood–willow habitat along regulated rivers is through installing dormant, rootless cuttings, yet there is little published information exploring floodplain characteristics that optimize growth of southwestern riparian willows planted in this manner. The goal of this project was to evaluate relationships between growth attributes of Salix exigua and soil texture and soil water availability. Monitoring plots were established in five willow swales planted with dormant S. exigua cuttings along the banks of the Middle Rio Grande in central New Mexico. Data analysis revealed significantly higher aerial cover, height, and stem density for S. exigua plants installed in plots with intermediate levels (15–25%) of fine textured soils distributed through the soil profile. Similar relationships were found in relation to soil water availability. Regression analysis of percent fines and available water at different depth increments provided limited explanation of variability in willow growth attributes at different plots. Findings indicate that S. exigua plants established from cuttings can achieve heights and aerial cover values similar to naturally established willow bars if the floodplain soil profile contains intermediate levels of fine textured soils and the maximum depth to groundwater is within 1.5 m of the ground surface. Where sites are dominated by coarse sand, S. exigua growth may be improved if maximum depth to groundwater is within 1 m of the ground surface.  相似文献

Shoot proliferation and rooting of three cranberry (Vaccinium macrocarpon Ait.) cultivars Bergman, Pilgrim, and Stevens were obtained in vitro on a modified nutrient medium containing zeatin following a one-step procedure. Bergman and Stevens differed in terms of shoot height, leaf number per shoot, rooting frequency, root number per explant, and root length; this was manifested with various concentrations of zeatin. Shoots proliferated and roots developed best when nodal segments were cultured in the medium supplemented with very low concentration of zeatin (2–4 μM). Such zeatin-induced tissue culture (TC) shoots of Bergman, Pilgrim, and Stevens were rooted ex vitro and compared with those propagated by conventional softwood cuttings (SC) for growth and morphology over four growth seasons. Significant interactions for leaf number per upright were observed among the treatments. The cultivars differed in terms of runner number per plant, upright length, number of leaves per upright, and shoot vigor. The propagation method had an effect on morphology of cranberry plants. The TC plants produced more runners and uprights with more leaves per upright than the conventional cuttings. This increase in vegetative growth of in vitro-derived plants over stem cuttings varied among genotypes. In vitro culture on zeatin-containing nutrient medium apparently induces the juvenile branching characteristics that favored enhanced vegetative growth with more shoots and leaf production.  相似文献

Small diameter fine, fibrous roots of willow are exceedingly difficult to remove from most soils. Where field retrieval of entire plants including clean roots may be important, porous membrane rooting envelopes are a method of choice.Purple osier willow (Salix purpurea L.) stem cuttings (clone SP3) were field rooted in buried porous membrane envelopes permeable to water and nutrients. Ramets were grown for two and four months, then separated into component parts for dry weight analysis.At two months, cuttings grown in rooting envelopes were no different in shoot dry weight than plants grown in clay pots or in soil. Tests indicated that rooting envelopes may be reused at least once without affecting shoot or total plant dry weight. Likewise, membrane pore sizes of 0.45, 1.2 and 3 m did not affect SP3 dry weight in four month tests. The importance of matching plant material and experiment duration to envelope size was illustrated by the limitation of growth by envelope edges at four months compared to two months.  相似文献

Black willow (Salix nigra) uses periodic flood events for dispersal of vegetative propagules, subjecting them to periods of soaking before their deposition along the streambank. It was hypothesized that this life history trait results in optimal conditions for willow growth and survival. To test this hypothesis, a greenhouse experiment was conducted using 1.2‐m‐long black willow cuttings (posts) with a basal diameter of 5 cm. Cuttings were subjected to three soaking treatments (0, 3, and 10 days) and then grown under four soil moisture regimes (control, drought, permanently flooded, and intermittently flooded). Growth, biomass, and survival were recorded. Results showed that soaking posts for 10 days was most beneficial in the control soil moisture regime, enhancing root, shoot, leaf, and total biomass production. Shoot abundance and growth were also enhanced by 10 days of soaking in the control and permanently flooded moisture regimes. Finally, survival increased significantly in control and drought moisture regimes in response to the 10‐day soaking treatment. Results clearly demonstrated that soaking had significant effects on willow post success when evaluated across all soil moisture regimes. Posts subjected to the 10‐day soaking treatment consistently had greatest growth and biomass and displayed a doubling of the survival rate. Therefore, soaking willow cuttings before planting is a simple, inexpensive technique that may be used to bolster streambank restoration success.  相似文献

Summary Growth of selected clones of birch and willow obtained from nitrogen deficient soils was compared with that of unselected controls in pot experiments using three levels of nitrogen. Unselected controls of both genera continued shoot growth, albeit very slowly, with a very low level of nitrogen (5 ppm), while selected clones of birch grew significantly more than the controls. Selected clones of willow, on the other hand, ceased shoot growth after 10 weeks with this low nitrogen treatment. Nevertheless, they remained healthy, their leaves containing similar concentrations of nitrogen to those of the plants which continued stem growth throughout the experiment. Furthermore, they had very high root: shoot ratios compared with those of control willows and both selected and unselected birch. The two genera may have developed different mechanisms for tolerating low nitrogen, birch producing a small, relatively efficient root system; willow a larger but less efficient one. Both appear equally effective in ensuring survival on low-nitrogen sites in the field since all the selected clones were obtained from such sites and have survived well in field trials on similar sites. Both birch and willow responded toincreased nitrogen availability with increasing shoot growth and a relative decline in root growth. However, whereas in willow the unselected plants responded significantly more than selected clones, a similar but less markeddifference was found in birch. It appears that in both genera, as in herbaceous plants originating from nitrogen deficient sites, selected clones are less able to respond to increasing nitrogen supply than control plants from more fertile habitats. Attempts to correlate the response of the selected clones to nitrogen in this experiment with that to added nitrogen fertilizer in field trials has been unsuccessful. Further work is required to determine the importance of the many interacting factors which influence the response of young trees to nitrogen under the unusual field conditions associated with restored mineral workings.  相似文献

The effects of four indole-3-butyric acid (IBA) concentrations and two propagation methods were studied in a lowbush blueberry (Vaccinium angustifolium Ait.) clone collected from natural stands in Newfoundland and Labrador, Canada. Lowbush blueberry cultures were established in vitro from nodal explants on a modified cranberry (V. macrocarpon Ait.) tissue culture medium containing zeatin (2 μM). Blueberry plants propagated by in vitro shoot proliferation (TC) and by conventional softwood cuttings (SC) were evaluated for growth and morphology. Significant interactions for morphological characteristics were observed among the treatments. The IBA concentration had an effect on morphology of propagated plants, increasing the concentration of IBA increased stem length and leaves per stem across propagation methods. Stems per plant increased with IBA concentration up to 20 μM in SC plants, but not in TC plants. Plant vigor was affected by neither IBA concentration nor propagation method. The TC plants produced longer and more stems with more leaves per stem than the conventional cuttings. In vitro culture on zeatin-containing nutrient medium apparently induces the juvenile branching characteristics that favored enhanced vegetative growth with more stems and leaf production. It is suggested that IBA may serve as a physiologically active form of auxin in contributing to increased stem and leaf production in lowbush blueberry SC plants but not in TC plants.  相似文献

Two divergent populations of T. repens cv. Haifa developed from two generations of recurrent selection for shoot chloride concentration, were grown in the greenhouse at 0 and 40 mol m–3 NaCl. Over two harvest cycles at 40 mol m–3 NaCl, the population selected for a low concentration of chloride in the shoot maintained a significantly lower chloride and sodium concentration compared with those plants selected for a high shoot chloride concentration. The distribution of chloride in the shoots was further examined in a subsample of plants from both populations. In all plants, concentrations of chloride were lower in the expanding and fully expanded leaves than in the older leaf tissue or petioles.While there were no significant differences in the photosynthetic rates between lines, shoot yields and relative leaf expansion rates were higher in the low chloride population. Plant death was greater in plants selected for high shoot chloride. These results suggest that selections based on measurements of low shoot chloride concentrations may be successful in developing a cultivar of T. repens with improved salt tolerance.  相似文献

In recent decades, invasive willows and poplars (Salicaceae) have built dense floodplain forests along most of the rivers in Patagonia, Argentina. These invasion processes may affect Salix humboldtiana as the only native floodplain tree species in this region. It is assumed, that the property to reproduce vegetatively can play an important role in the establishment of invasive species in their new range. Thus, in order to contribute to a better understanding of willow and poplar invasions in riparian systems and to assess the potential impacts on S. humboldtiana the vegetative reproduction capacities of native and invasive Salicaceae were analysed. In a greenhouse experiment, we studied cutting survival and growth performance of the three most dominant invasive Salicaceae of the Patagonian Río Negro region (two Salix hybrids and Populus spec.), as well as S. humboldtiana, taking into account three different moisture and two different soil conditions. In a subsequent experiment, the shoot and root biomass of cuttings from the former experiment were removed and the bare cuttings were replanted to test their ability to re-sprout. The two invasive willow hybrids performed much better than S. humboldtiana and Populus spec. under all treatment combinations and tended to re-sprout more successfully after repeated biomass loss. Taking into account the ecology of vegetative and generative recruits of floodplain willows, the results indicate that the more vigorous vegetative reproduction capacity can be a crucial property for the success of invasive willow hybrids in Patagonia being a potential threat for S. humboldtiana.  相似文献

Summary The effects of added P and lime on Douglas fir and Scots pine seedlings, and poplar and willow cuttings growing in a podzolic soil (pH 3.8, 90 ppm total P) were studied in pot experiments. Conifer dry weights responded best to P applied in the absence of lime, whereas liming to pH 4.3 promoted the P response of the broadleaved species. Normal rates of P, and of lime (broad-leaved species), by promoting growth, also raised total contents of P and metals (Zn, Mn, Cu, Fe) in the various plant parts (stems, foliage, roots), but generally lowered the metal concentrations. The results strongly suggest that P interfered with the root to shoot translocation of Cu, Fe and Al (Al only estimated in Scots pine), but not with that of Zn and Mn. It is postulated that internal plant tolerance (promoted by P) plays a more important part in neutralizing toxic metal concentrations (Zn, and possibly also Fe) in the soil than do exclusion mechanisms. High applications of P without Cu may depress growth, as demonstrated for willow. Water-soluble soil P data may be misinterpreted if other limiting soil factors (pH, Cu status) have not been eliminated.  相似文献

Rooting in terminal shoot and lateral shoot cuttings from 10-year-old elite trees of Casuarina equisetifolia L. in different sex groups was achieved after 20 days when the basal ends of the cuttings were dipped for 3 h in 20 ppm indole-3-butyric acid (IBA). Shoots derived from male plants rooted better than their female and monoecious counterparts, and the lateral shoots were more responsive to rooting than the terminal shoots. During rooting, the metabolic activities varied in both lateral shoot and terminal shoot cuttings derived from plants under different sex groups. Peroxidase and polyphenoloxidase activities were high during root initiation and showed a sharp decline thereafter. The polyphenoloxidase activity was higher in the lateral shoot than the terminal shoot cuttings. The efficiency of the absorption from the medium and accumulation by plant of ions of the heavy metals depends on many factors including plant age and its genotype. The heavy metals accumulation by 1-, 2- and 3-year-old plants was studied in the aspect of reclamation and revitalization effectiveness of demoted areas. Results of this study answer the question concerning the accumulation of seven heavy metals (Cd, Co, Cr, Cu, Ni, Pb and Zn) by willow cuttings (Salix viminalis) in environments with different concentrations of these elements. Cuttings used were originally grown on a substrate not contaminated with the metals and rich in nutrients. In order to eliminate the effect of soil physicochemical factors, the experiment was carried out using a hydroponic system. Results indicated significant effects of the investigated metal concentrations on their accumulation by willow. The extent of metal accumulation as an indicator of the remediation capacity of willow depended on the age of the cuttings used at planting.  相似文献

Phenolic compounds are potential antimicrobial agents in willows ( Salix spp.). However, little is known about their role in willow‐pathogen interactions. Earlier studies have shown that phenolic levels may vary depending on genotype and environmental factors (such as nutrient availability). In the present study, the combined effects of nitrogen fertilization and infection by pathogenic rust ( Melampsora sp.) on phenolics and growth in willow ( salix myrsinifolia ) clones were investigated. Cutting-propagated plants of eight willow clones were subjected to low and optimum levels of nitrogen fertilization and inoculated with rust urediniospores or mock-inoculated with water. The plants were harvested 20 days after the inoculations and the dry weight of the leaves, stems and roots determined. Phenolics were analysed from leaf material using HPLC. The willow clones differed significantly in their phenolic content, rust frequency and phytomass production. Plants under optimum nitrogen availability showed increased shoot phytomass production and reduced root production. Rust infection increased the root phytomass of willow under both fertilization treatments, but had no consistent effect on the shoot growth of the clones. Low nitrogen availability increased the phenolic levels in willow leaves, but the rust frequency did not differ significantly between plants grown at low and optimum levels of nitrogen fertilization. In several clones, rust infection increased the concentrations of individual phenolic compounds, but there were no significant correlations between phenolic concentrations and the level of rust infection. The results indicate that resistance in compatible willow‐rust interactions may not be directly governed by phenolic compounds.  相似文献
