When leaves of three Grevillea species were macerated, hydrogencyanide was only released from the soft, immature ones. Thebark but not wood of young stems was weakly cyanogenic. Thegreatest concentration of cyanoglucosides occurred in peduncles,flower buds, open flowers, immature seeds and germinants. Nocyanide was released from nectar, pedicels, dry fruit, elaiosomesand dry walls mature seeds. Addition of ß-glucosidase,and sometimes water only, increased cyanogenesis in young leaves,bark, some seed and fruit walls, and embryos of mature seeds.The endogenous levels of ß-glucosidase were sufficientto release all the cyanide present in at least 80% of cases. Young leaves and germinants of G. bipinnatifida (resprouter)released more cyanide than those of G. banksii, while flowerbuds, stigmatic region, styles, perianths and ovaries of G.banksii (non-sprouter) released far more cyanide than thoseof G. bipinnatifida. The interspecific and intraplant distributionof cyanoglucosides is consistent with the expected impact ofbeneficial and herbivorous animals on long term fitness of thesespecies. The F1 hybrid (G. The reproductive biology of Grevillea robusta growing underexotic conditions in Kenya and Australia is reported. The speciesshowed both protandry and a self-incompatibility mechanism.The stigma was wet and papillate with a distinct groove in themiddle. The anthers dehisced prior to anthesis, when the perianthopened. Stigmatic receptivity began 1 d after anthesis, withthe greatest pollen germination rates and longest pollen tubesobtained 2 d after anthesis. Nectar secretion commenced withpollen dehiscence and was abundant at anthesis. Most stigmaticgrooves opened widely 1–2 d after anthesis and stigmasshowed taller papillae and abundant secretion. Controlled pollinationsgave a greater fruit set from cross-pollination (5.9% in Apriland 17.5% in July) than open-pollination (0.1% in April and3.3% in July). No fruit set from self-pollination was obtainedin April, and very few fruit set for geitonogamous (two outof 1622; 0.1%) or for autogamous (one out of 2707 flowers; 0.04%)pollination treatments in July. Following self-pollination,growth of pollen tubes was poorer than in other treatments,and was generally arrested in the upper style. Cross-pollinatedflowers produced normal and straight pollen tubes, while self-pollentubes had growth abnormalities. Most of the open-pollinatedflowers were found without pollen or with only self-pollen ontheir stigmas indicating that the amount of cross-pollen reachingthe stigma under open-pollination may be a factor limiting seedproduction. Flowers shed soon after the fertilization phasewere those with ungerminated pollen or no pollen. The various pathways of pollen development were investigatedin cultured anthers of Lolium temulentum, Festuca pratensisand the L. multiflorum x F. pratensis hybrid ‘Elmet’.In all three, development from the vegetative cell was the predominantpathway of pollen callus development. However, there were characteristicdifferences in the behaviour of the generative cell. In L. temulentumit remained attached to the pollen wall and degenerated, whereasin F. pratensis it divided. In ‘Elmet’ it detachedfrom the pollen wall and remained undivided. Both polarizedand unpolarized partitioned calluses were observed. Developmentof the fusion product of the vegetative and generative nucleiwere also observed in anthers of L. temulentum. Anomalous grainswere not found to be major source of pollen calluses. Sections of anthers of L. temulentum were used to investigatethe origin of S pollen grains, the small pale-staining grainswhich denote pollen dimorphism. Such grains form out of contactwith the tapetum and are therefore determined before or duringmeiosis (i.e. before harvest of anthers for culture). Sectionswere also used to demonstrate the influence of the durationof pretreatment on the development of the middle layer of theanther wall. In a compatible pollination in Crocus, pollen tube tips enterthe stigma papillae after the enzymic erosion of the cuticle,and the tubes continue downward growth towards the ovary betweenthe cuticle and the underlying pectocellulosic wall. The cuticleof the receptive zone of the stigma papilla is chambered, thechambers containing a secretion accumulated during the maturationof the stigma. Pollen exudates contain various acid hydrolases,but are incapable alone of eroding stigma cutin. Furthermore,there is no penetration when the proteins of the wall-held stigmasecretions are degraded enzymically. These facts are taken toindicate that the pollen contributes a ‘cutinase’precursor which is activated by a factor or factors held inthe stigma secretion. Pollens of certain Cruciferae producetubes capable of penetrating the Crocus stigma cuticle, suggestingthat notwithstanding the taxonomic remoteness of Cruciferaeand Iridaceae the enzyme activation systems are quite similar.  相似文献

The possible involvement of chemotropic factors influencingpollen tube growth in the style and ovary of Salpiglossis wasstudied. Pollen deposited at the base of excised styles germinatedreadily, with pollen tubes penetrating the transmitting tissueand growing towards the stigma, implying that no chemotropicrequirement exists for pollen tube growth in the style. However,the position and high efficiency of seed set following limited-pollinationof intact pistils would support the involvement of localizedchemotropism operating within the ovary. This paper describes the characteristics of the pollen and thereceptive surfaces of the stigmas in the three marine angiospermsincluded in the Hydrocharitaceae. The pollen in Enhalus acoroidesand Thalassia hemprichii is spherical and has an ornamentedexine. An exine layer is not found in Halophila stipulacea wherereniform pollen grains are contained within transparent moniliformtubes. Cytochemical tests show that the pollen wall in the threespecies contains acidic and neutral polysaccharides and acidhydrolase acitivity is detected in the intine of H. stipulaceaand T. hemprichii. In Thalassia, one of the intine enzymes,acid phosphatase, is unambiguously associated with cytoplasmicinclusions. Flowering in Thalassia is coincident with the spring tides andthe pollen is released as a mass suspended in a thecal slimewhich contains approximately 5 per cent by weight carbohydrate,the principal mono-saccharide being mannose. Electrophoreticanalysis of the pollen-free slime shows a single glycoproteincomponent. The stigmas of the three seagrasses are papillate and of the‘dry’ type possessing a continuous protein-aceouspellicle subtended by a cuticle. The stigma pellicle exhibitscytochemically detectable esterase activity and binds the lectinconcanavalin A. Acid phosphatase activity is localized beneaththe cuticle at the tips of the stigma papillae. The discoveries show that the characteristics of the pollenand stigmas in the seagrasses are comparable with those foundin terrestrial flowering plants. The similarity in enzymaticproperites of the pollen wall and stigma pellicle suggests that,intriguingly, a similar mechanism of cuticle erosion might wellfollow compatible pollination both on land and in the sea. When ripe, viable pollen of Pinus contorta, Pinus nigra, Piceasitchensis, Abies alba and Cedrus deodara is germinated on asuitable artificial substrate, the process of pollen tube growthbegins 12–36 h later, according to the species. The tubeemerges at the leptoma, a structurally specialized site in thepollen wall, and the early tube wall is continuous with thetwo microfibrillar intine strata within the grain. Later inpollen tube development, when cytoplasmic zonation has beenestablished, only the inner of these two wall layers is differentiated. Cytochemical methods show that, during hydration and throughoutthe period of tube growth in culture, proteins are releasedfrom the pollen grains; before germination most conspicuouslyfrom the leptoma, subsequently from the tube itself. The emissioncontains a number of hydrolytic enzymes and a non-catalyticmoiety. Gel-electrofocusing reveals that the hydrolases releasedfrom germinating Pinus contorta pollen include several acidphosphatase and esterase isozymes, and that there are differencesbetween the composition of the emission and the spectrum ofsoluble proteins extracted from the pollen grains before germination.Analysis by immunodiffusion demonstrates that two componentswith antigenic characteristics present in the quiescent pollengrains are represented in the emission. The possible utilityof the released components in the biology of conifer reproductionis discussed. High resolution light and fluorescence microscopy were usedto investigate the structural and cytochemical features of thekiwifruit(Actinidia deliciosa)pistil and to follow the pollentube pathway after pollination. The multicarpellary ovary issyncarpous only at the ovary level thus leaving 30–40free styles on top. The fusion of the longitudinally-foldedcarpels to form the syncarpous ovary forms a central parenchymatousaxis, the columella, from which the ovules radiate outwardsinto the ovary cavity. A prominent cup shaped depression onthe columella at the top end of the ovary, termed the pollentube distributor cup (PTDC) was detected. Pollen tubes fromthe stigma travel through the transmitting tract and enter thePTDC from where they are distributed towards the ovary. Evenwhen pollination is restricted to two stigmas, the PTDC seemsto ensure that the pollen tubes are evenly distributed aroundthe ovary resulting in an even distribution of seeds. Saururus cernuus, a species belonging to the primitive herbaceousangiosperm family Saururaceae, exhibits high rates of self-sterility.We investigated the structural and functional aspects of pollen-carpelinteractions following cross and self pollination to assessthe tissue specific site and timing of self-sterility and factorsimportant for successful cross pollen tube growth. Self-sterilitywas due to inhibition of self pollen germination at a dry stigma.Self pollination was associated with anomalous foot formation,reduced cell wall expansion and secretory activity of stigmaticpapillae, and callose production in stigmatic papillae. Followinggermination, cross compatible pollen tubes entered a solid coreof transmitting tissue and grew to the base of a short style.Entry of cross pollen tubes into the ovary was coincident withovule enlargement which placed the micropyle in the proximityof cross pollen tube tips. Ovule enlargement also occurred followingself pollination. Cross pollen tubes either entered an exudate-filledmicropyle directly from the style, or growth in the ovary waslocalized to the epidermis of the locule and outer integumentprior to entry into the micropyle. Prior to pollination, thetransmitting tract was void of secretions except for exudatein the micropyle. Growth of pollen tubes on the locule and integumentwas associated with exudate apparently arising from transmittingcells adjacent to growing pollen tubes. The present study providesthe first evidence in a primitive herbaceous species of stigmaticself-incompatibility (SI) in association with a dry stigma,pollination-induced signalling events affecting developmentof carpellary tissues, and micropylar exudates. Brassica campestris Male Fertility 2 (BcMF2) is a putative polygalacturonase(PG) gene previously isolated from the flower bud of Chinesecabbage (Brassica campestris L. ssp. chinensis Makino, syn.B. rapa ssp. chinensis). This gene was found to be expressedspecifically in tapetum and pollen after the tetrad stage ofanther development. Antisense RNA technology was used to studythe function of BcMF2 in Chinese cabbage. Scanning and transmissionelectron microscopy revealed that there were deformities inthe transgenic mature pollen grains such as abnormal locationof germinal furrows. In addition, the homogeneous pectic exintinelayer facing the exterior seemed to be overdeveloped and predominantlyoccupied the intine, thus reversing the normal proportionaldistribution of the internal endintine layer and the externalexintine layer. Since it is a continuation of the intine layer,the pollen tube wall could not grow normally. This resultedin the formation of a balloon-like swelling structure in thepollen tube tip in nearly 80% of the transgenic pollen grains.Premature degradation of tapetum was also found in these transgenicplants, which displayed decreased expression of the BcMF2 gene.BcMF2 might therefore encode a new PG with an important rolein pollen wall development, possibly via regulation of pectin'sdynamic metabolism. The pollen tube pathway has been studied in unpollinated andpollinated pistils of Lycopersicon peruvianum using histochemicalstains for detection of proteins, lipids and arabinogalactansby bright-field microscopy, and decolourized aniline blue fordetection of pollen tubes by epifluoresence microscopy. Thepollen tube pathway is a continuous tract of mucilage from thestigma surface to the ovule micropyles, and is associated witha continuous tract of specialized, protein-rich transmittingcells comprising the stigmatic papillae, vertical files of stylartransmitting cells and the placental epithelium within the ovary.The superficial exudate of the stigma is hydrophobic and richin lipids. The mucilage of the style and ovary is hydrophilicand rich in arabinogalactans but low in proteins. Pollen tubesgrow between cells through the mucilage of the stigma and stylartransmitting tract, and across the surface of the placenta inthe ovarian mucilage. The structure of the junction of the stylartransmitting tract with the top of the ovary placenta assistseffective distribution of pollen tubes within the ovary. Pollen tube growth, fertilization and early embryo and endospermdevelopment were studied using light microscopy in the avocadovarieties Fuerte and Hass. The ovule was penetrated by a pollen tube by 24 h after pollination.On reaching the ovary, the pollen tube grew along the surfaceof the inner ovary wall. It then grew around the funicle, throughthe micropyle in the inner integument and between the papillatecells at the apex of the nucellus. It entered the embryo sacvia a synergid. Sperm nuclei were present in the embryo sacat 48 h after pollination and fusion of the polar and spermnuclei took place before fusion of the egg and sperm. The endospermnucleus was the first to divide and cell wall formation occurredfollowing division. The first division of the zygote occurredat 5 or 6 days after pollination. In the variety Fuerte less than 20 per cent of the 1- and 2-day-oldembryo sacs had been penetrated by a pollen tube although tubeswere often observed in the integument or nucellus. In the varietyHass over 60 per cent of the embryo sacs were penetrated. Inwas concluded that low yields of the variety Fuerte may be partlyattributable to the failure of the pollen tube to penetratethe embryo sac. Aniline blue fluorescence was used to study the growth of maizepollen tubes in the stigmas of 13 diverse sorghum accessions.In 12, only short maize pollen tubes were formed, but in thesingle exception (Sorghum nervosum Nr481) maize pollen tubesgrew at least as far as the base of the style. The S. bicolorgenotypes S9B and CMS (a cytoplasmic male sterile line) werehybridized with Nr481, and analysis of maize pollen tube growthin F1 plants, and BC1 plants using Nr481 as the recurrent parent,suggested that differences in inhibition of pollen tube growthwere due to variation at a single locus, which we propose todesignate lap (Inhibition of alien pollen tubes). AccessionNr481 appears to be homozygous for a recessive allele permittingmaize pollen tube growth. Attempts were made to produce sorghumx maize hybrids using Nr481 and CMS derivatives which were knownto allow maize pollen tube growth to the base of the style.A putative hybrid endosperm was obtained in one Nr481 x Seneca60 maize cross, but this was not repeatable and no hybrid plantswere produced. Gladiolus has a dry type of stigma. Compatible pollen grainsalight and germinate on the receptive surface of the papillae,penetrate the cuticle and grow towards the style through a sub-cuticularpollen-tube guide of mucilage. This is secreted from epidermalcells of the stylodium and style canal. The cuticle, which coversthe pollen tube guide mucilage, is continuous through the stylecanal to the ovary. Pistil structure and pollen tube growth were investigated inLeptospermum myrsinoides and L. continentale (Myrtaceae). BothL. myrsinoides and L. continentale pistils consist of an ovarywith five locules, a style and a five-lobed dry, papillate stigma.A centrally located stigmatic cleft is present but extends onlyto the base of the stigma. Pollen germinates and grows intercellularlythrough the stigma into the central transmitting tissue of thestyle. Pollen tubes do not grow down the stigmatic cleft. Atthe base of the style the transmitting tissue separates intofive, each tract leading through the placenta to one of thefive locules. The pollen tubes continue to grow intercellularlythrough these five tracts entering the locules between the lobesof the placenta. Pollen tubes are smooth-walled and straightwhilst in the transmitting tissue of the style but produce shortlateral branches at regular intervals when in the locules. Branchingcontinues until pollen tubes enter ovules. It is suggested thatthe observed branching in the locules is a result of pollentubes following a chemotropic or thigmotropic pathway to theovules. The embryo-sac of Grevillea Banksii R. Br. develops accordingto the normal type. At the time of fertilization is has a veryirregular outline. The synergids show a well-marked filiformapparatus. The development of the embryo is similar to that in the twomembers of the family previously studied (Kausik, 1938, 1938a). The endosperm becomes cellular. The presence of the vermiformappendage of the endosperm described in Grevillea robusta (Kausik,1938), has been demonstrated here. Though similar in both, theendosperm of G. Banksii shows a narrow middle region with rowsof elongated cells which are absent in G. robusta. In conclusion, the writer has great pleasure in acknowledginghis indebtedness to Prof. M. A. The microfibrillar polysaccharide component of the pollen intinecan be isolated by progressive chemical digestion of the exineand the cellular contents and the extraction of the matrix materials.The resulting intine ‘ghosts’ reveal various characteristicstructural features. The microfibrils have apparent individualdiameters in the range of 5–15 nm, but they are commonlyassociated laterally to form ribbons, or aggregated in strandsor cables of dimensions great enough to be resolved with theoptical microscope. These often show preferred orientations,which can be associated with pollen grain shape and with thedisposition-of the germination apertures. The apertural intinemay be structurally complex, as in Abutilon hybridum, where,after the removal of the exine, the polysaccharide caps whichoverlie the protein storage sites of the pollen grain wall retainthe elaborate patterning of the original cytoplasmic evaginationsfrom the vegetative cell. A cDNA for a pathogenesis-related endo-ß-1,3-glucanaseisolated from soybean, was fused to an anther tapetum-specificpromoter (Osg6B promoter) isolated from rice and the resultingchimeric gene was introduced into tobacco. The Osg6B promoterbecame active in the anther tapetum during formation of tetradsand the tapetal glucanase activity in the transgenic plantscaused in a significant reduction in the number of fertile pollengrains. Most of the pollen grains were aberrant in shape, lackedgerminal apertures and aggregate of the pollen grains. Granulesof ß-1,3-glucan, which have not previously been reported,were often observed to adhere to the surface of the pollen grains.Further observations revealed that the callose wall was almostabsent in the pollen tetrads of transgenic plants. In wild-typeplants, by contrast, the tetrads were surrounded by callosethat was degraded soon after the tetrad stage to release freemicrospores. Thus, the introduced gene for endo-ß-1,3-endoglucanaseunder the control of the Osg6B promoter caused digestion ofthe callose wall at the beginning of the tetrad stage, a timethat was just a little earlier than the time at which endogenousglucanase activity normal appears. These results demonstratethat premature dissolution of the callose wall in pollen tetradscauses male sterility and suggest that the time at which tapetallyproduced glucanase is activate is critical for the normal developmentof microspores. Changes in the obturator of the peach (Prunus persica) havebeen investigated and related to pollen tube growth in thisregion. At anthesis, the cells of the obturator are active andrich in starch reserves. Twelve days after anthesis these cellsproduce a secretion that stains for carbohydrates and for proteins.As the secretion is produced, starch vanishes from these cellsand they degenerate and collapse as callose is accumulated.Secretion is independent of pollination as it takes place ina similar fashion both in pollinated and in unpollinated flowers. Pollen tube growth along the obturator surface depends on thissecretion for, although pollen tubes reach the base of the styleseven days after pollination, they cannot grow on the obturatoruntil five days later, when the secretion is produced. Thisdiscontinuous secretion taking place at the obturator may providea mechanism that controls the entrance of pollen tubes intothe ovary in the peach.
