Soybean [Glycine max (L.) Merrill] plants that had been subjectedto 15 d of nitrogen deprivation were resupplied for 10 d with1.0 mol m–3 nitrogen provided as NO3, NH4+, orNH4++NO3 in flowing hydroponic culture. Plants in a fourthhydroponic system received 1.0 mol m–3 NO3 duringboth stress and resupply periods. Concentrations of solublecarbohydrates and organic acids in roots increased 210 and 370%,respectively, during stress. For the first day of resupply,however, specific uptake rates of nitrogen, determined by ionchromatography as depletion from solution, were lower for stressedthan for non-stressed plants by 43% for NO3- resupply, by 32%for NH4+ + NO3 resupply, and 86% for NH4+ resupply. Whenspecific uptake of nitrogen for stressed plants recovered torates for non-stressed plants at 6 to 8 d after nitrogen resupply,carbohydrates and organic acids in their roots had declinedto concentrations lower than those of non-stressed plants. Recoveryof nitrogen uptake capacity of roots thus does not appear tobe regulated simply by the content of soluble carbon compoundswithin roots. Solution concentrations of NH4+ and NO3 were monitoredat 62.5 min intervals during the first 3 d of resupply. Intermittent‘hourly’ intervals of net influx and net effluxoccurred. Rates of uptake during influx intervals were greaterfor the NH4+ -resupplied than for the NO3 -resuppliedplants. For NH4+ -resupplied plants, however, the hourly intervalsof efflux were more numerous than for NO3 -resuppliedplants. It thus is possible that, instead of repressing NH4+influx, increased accumulation of amino acids and NH4+ in NH4+-resupplled plants inhibited net uptake by stimulation of effluxof NH4+ absorbed in excess of availability of carbon skeletonsfor assimilation. Entry of NH4+ into root cytoplasm appearedto be less restricted than translocation of amino acids fromthe cytoplasm into the xylem. Shelp, B. J. 1987. The composition of phloem exudate and xylemsap from broccoli (Brassica oleracea var. italica) suppliedwith NH+4, NO3 or NH4NO3.—J. exp. Bot. 38: 1619–1636. The detailed composition of xylem sap and exudate from stemincisions of attached inflorescences of broccoli (Brassica oleraceavar. italica) was compared in plants supplied with NH+4, NO3or NH4NO3. A phloem origin for the exudate was suggested fromthe high levels of sugars (71–133 mg cm-3), amino acids(8·1-26·7 mg cm3) and K. (2·3–3·8mg cm3), the low levels of NO3 and Ca, the high C: N (w/w) ratios(8·3–33), and the alkaline pH (7·2–7·3).In contrast, the xylem sap was mildly acidic (pH 5·6–6·0),and possessed lower levels of all organic and inorganic solutesbut NO3 and Ca, and lower ratios of K: Ca, Mg: Ca and C: N (0·6–4·4). Glutamine was the predominant o-phthalaldehyde-reactive aminocompound in both transport fluids with the next most abundantamino acids dependent on sap type and N-form. Together witharginine, -aminobutyric acid, which was found only in the xylemstream, was enhanced by NH+4compared to NO3 -nutrition suggestingthat glutamate metabolism was stimulated in the roots. Underlimiting N the amino acid concentrations in the transport fluidswere greater with NH+4 than with NO3. NO3 reduction occurredin both the root and shoot with the latter site predominatingover the entire N range (0-300 mol m3). Even though the compositionof nitrogenous solutes in the xylem was dependent on cultivarand N source, the composition of the phloem streams supplyingthe developing inflorescence was relatively unaffected. The data on the element composition of organs and phloem sapare interpreted to suggest that, in spite of the restrictedmobility of some elements such as B and Mn, a significant proportionof their total supply to developing sinks is carried in thephloem stream. Seedlings of Ricinus communis L. cultivated in quartz sand weresupplied with a nutrient solution containing either 1 mol m–3NO3 or 1 mol m–3 NH+4 as the nitrogen source. Duringthe period between 41 and 51 d after sowing, the flows of N,C and inorganic ions between root and shoot were modelled andexpressed on a fresh weight basis. Plant growth was clearlyinhibited in the presence of NH+4. In the xylem sap the majornitrogenous solutes were nitrate (74%) or glutamine (78%) innitrate or ammonium-fed plants, respectively. The pattern ofamino acids was not markedly influenced by nitrogen nutrition;glutamine was the dominant compound in both cases. NH+4 wasnot transported in significant amounts in both treatments. Inthe phloem, nitrogen was transported almost exclusively in organicform, glutamine being the dominant nitrogenous solute, but theN-source affected the amino acids transported. Uptake of nitrogenand carbon per unit fresh weight was only slightly decreasedby ammonium. The partitioning of nitrogen was independent ofthe form of N-nutrition, although the flow of nitrogen and carbonin the phloem was enhanced in ammonium-fed plants. Cation uptakerates were halved in the presence of ammonium and lower quantitiesof K+, Na+ and Ca2+ but not of Mg2+ were transported to theshoot. As NH+4 was balanced by a 30-fold increase in chloride in thesolution, chloride uptake was increased 6-fold under ammoniumnutrition. We concluded that ammonium was predominantly assimilated inthe root. Nitrate reduction and assimilation occurred in bothshoot and root. The assimilation of ammonium in roots of ammonium-fedplants was associated with a higher respiration rate. In non-nodulated soybean [Glycine max (L.) Merrill cv. Ransom]plants that were subjected to 15 d of nitrogen deprivation inflowing hydroponic culture, concentrations of nitrogen declinedto 1.0 and 1.4mmol Ng–1 dry weight in shoots and roots,respectively, and the concentration of soluble amino acids (determinedas primary amines) declined to 40µmol g–1 dry weightin both shoots and roots. In one experiment, nitrogen was resuppliedfor 10 d to one set of nitrogen-depleted plants as 1.0 mol m–3NH4+ to the whole root system, to a second set as 0.5 mol m–3NH4+ plus 0.5 mol m–3 NO3 to the whole root system,and to a third set as 1.0 mol m–3 NH4+ to one-half ofa split-root system and 1.0 mol m–3 NO3 to theother half. In a second experiment, 1.0 mol m–3 of nitrogenwas resupplied for 4 d to whole root systems in NH4+ : NO3ratios of 1:0, 9:1, and 1:1. Nutrient solutions were maintainedat pH 6.0. When NH4+ was resupplied in combination with NO3 to thewhole root system in Experiment I, cumulative uptake of NH4+for the 10 d of resupply was about twice as great as when NH4+was resupplied alone. Also, about twice as much NH4+ as NO3was taken up when both ions were resupplied to the whole rootsystem. When NH4+ and NO3 were resupplied to separatehalves of a split-root system, however, cumulative uptake ofNH4+ was about half that of NO3. The uptake of NH4+,which is inhibited in nitrogen-depleted plants, thus is facilitatedby the presence of exogenous NO3, and the stimulatingeffect of NO3 on uptake of NH4+ appears to be confinedto processes within root tissues. In Experiment II, resupplyof nitrogen as both NH4+ and NO3 in a ratio of either1:1 or 9:1 enhanced the uptake of NH4+. The enhancement of NH4+uptake was 1.8-fold greater when the NH4+: NO3-resupplyratio was 1:1 than when it was 9:1; however, only 1.3 timesas much NO3 was taken up by plants resupplied with the1 :1 exogenous ratio. The effect of NO3 on enhancementof uptake of NH4+ apparently involves more than net uptake ofNO3 itself and perhaps entails an effect of NO3uptake on maintenance of K+ availability within the plant. Theconcentration of K+ in plants declined slightly during nitrogendeprivation and continued to decline following resupply of nitrogen.The greatest decline in K+ concentration occurred when nitrogenwas resupplied as NH4+ alone. It is proposed that decreasedavailability of K+ within the NH4+-resup-plied plants inhibitedNH4+ uptake through restricted transfer of amino acids fromthe root symplasm into the xylem. The processes of NO3 uptake and transport and the effectsof NH4+ or L-glutamate on these processes were investigatedwith excised non-mycorrhizal beech (Fagus sylvatica L.) roots.NO3 net uptake followed uniphasic Michaelis-Menten kineticsin a concentration range of 10µM to 1 mM with an apparentKm of 9.2 µM and a Vmax of 366 nmol g–1 FW h–1.NH4+, when present in excess to NO3, or 10 mM L-glutamateinhibited the net uptake of NO3 Apparently, part of NO3taken up was loaded into the xylem. Relative xylem loading ofNO3 ranged from 3.21.6 to 6.45.1% of NO3 netuptake. It was not affected by treatment with NH4+ or L-glutamate.16N/13N double labelling experiments showed that NO3efflux from roots increased with increasing influx of NO3and, therefore, declined if influx was reduced by NH4+ or L-glutamateexposure. From these results it is concluded that NO3net uptake by non-mycorrhizal beech roots is reduced by NH4+or L-glutamate at the level of influx and not at the level ofefflux. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. The effects of NO-3 and NH+4 nutrition on hydroponically grownwheat (Triticum aestivum L.) and maize (Zea mays L.) were assessedfrom measurements of growth, gas exchange and xylem sap nitrogencontents. Biomass accumulation and shoot moisture contents ofwheat and maize were lower with NH+4 than with NO-3 nutrition.The shoot:root ratios of wheat plants were increased with NH+4compared to NO-3 nutrition, while those of maize were unaffectedby the nitrogen source. Differences between NO-3 and NH+4-fedplant biomasses were apparent soon after introduction of thenitrogen into the root medium of both wheat and maize, and thesedifferences were compounded during growth. Photosynthetic rates of 4 mM N-fed wheat were unaffected bythe form of nitrogen supplied whereas those of 12 mM NH+4-fedwheat plants were reduced to 85% of those 12 mM NO-3-fed wheatplants. In maize supplied with 4 and 12 mM NH+4 the photosyntheticrates were 87 and 82% respectively of those of NO-3-fed plants.Reduced photosynthetic rates of NH+4 compared to NO-3-fed wheatand maize plants may thus partially explain reduced biomassaccumulation in plants supplied with NH+4 compared to NO-3 nutrition.Differences in the partitioning of biomass between the shootsand roots of NO-3-and NH+4-fed plants may also, however, arisefrom xylem translocation of carbon from the root to the shootin the form of amino compounds. The organic nitrogen contentof xylem sap was found to be considerably higher in NH+4- thanin NO-3-fed plants. This may result in depletion of root carbohydrateresources through translocation of amino compounds to the shootin NH+4-fed wheat plants. The concentration of carbon associatedwith organic nitrogen in the xylem sap of maize was considerablyhigher than that in wheat. Seedlings of Italian ryegrass (Lolium multiflorum Lam. cv. RVP)and clonal stolon cuttings of white clover (Trifolium repensL. cv. Blanca) were grown for 19 d in flowing solution culture,with N supplied as either 250 mmol m–3 NO3 or NH3+.Rates of net uptake, influx and translocation of NO3and NH4+ were then determined using 15N and 13N labelling techniques:between 3–5 h into the photoperiod following 8 h darknessfor white clover (CL), and for ryegrass plants that were eitherentire (IL) or with shoots excised 90 min prior to 13N influx(IC); and 75 min into the photoperiod following 37–39h darkness for ryegrass (ID). Rates of net uptake, influx andefflux of NH4+ exceeded those of NO3 in IL and IC ryegrassplants: the opposite occurred in white clover (CL). The decreasein net uptake following defoliation of ryegrass was greaterfor NH4+ (62%) than NO3 (40%). For NH4+ this was associatedwith a large decrease in influx from 110 to 6.0µmol h–1g–1 root fr. wt; but for NO3, influx only decreasedfrom 42 to 37 µmol h–1 g–1. Prolonged exposureto darkness (ID plants) also lowered net uptake of NO3and NH4+ by, respectively, 86% and 95% of IL levels. For NH4+this was characterized by a large decrease in influx and a smalldecrease in efflux; whilst for NO3 the effect of a largedecrease in influx was reinforced by a smaller increase in efflux. The data were used to estimate the translocatory fluxes of NO3(03–20µmol h–1 g–1) and NH4+ (003–0.4µmolh–1 g–1), assimilation in the roots of NO3(02–26µmol h–1 g–1) and NH+4 (05–89 µmolh–1 g–1), and the concentrations of NO3 (9–15mol m–3) in the cytoplasmic compartment of the roots.The relevance of variable influx and efflux to models for theregulation of N uptake is discussed. Concentrations of inorganic cations are often lower in plantssupplied with NH4+ as compared with NO3. To examine whetherthis is attributable to impaired root uptake of cations or lowerinternal demand, the rates of uptake and translocation of K,Mg, and Ca were compared in maize plants (Zea mays L.) withdifferent growth-related nutrient demands. Plants were grownin nutrient solution with either 1·0 mol m–3 NO3or NH4+ and the shoot growth rate per unit weight of roots wasmodified by varying the temperature of the shoot base (SBT)including the apical shoot meristem. The shoot growth rate per unit weight of roots, which was takenas the parameter for the nutrient demand imposed on the rootsystem, was markedly lower at 12°C than at 24°C SBT.As a consequence of the lower nutrient demand at 12°C SBT,uptake rates of NO3 and NH4+ declined by more than 50%Compared with NO3 supply, NH4+ nutrition depressed theconcentrations of K and particularly of Ca in the shoot, bothin plants with high and with low nutrient demand. This indicatesa control of cation concentration by internal demand ratherthan by uptake capacity of the roots. Translocation rates of K, Mg and Ca in the xylem exudate werelower in NH4+- than in NO3-fed plants. Net accumulationrates of Ca in the shoot were also decreased, whereas net accumulationrates of K in the shoot were even higher in NH4+-fed plants.It is concluded that reduced cation concentrations in the xylemsap of plants supplied with NH4+ are due to the lower demandof cations for charge balance. The lower K translocation tothe shoot is compensated by reduced retranslocation to the roots.For Ca, in contrast, decreased translocation rates in NH4+-fedplants result in lower shoot concentration. Hordeum vulgare cv. California Mariout was established in sandculture at two different NaCl concentrations (0.5 mol m–3‘control’ and 100 mol m–3) in the presenceof 6.5 mol m–3 K +. Between 16 and 31 d after germination,before stem elongation started, xylem sap was collected by useof a pressure chamber. Collections were made at three differentsites on leaves 1 and 3: at the base of the sheath, at the baseof the blade, i.e. above the ligule, and at the tip of the blade.Phloem sap was collected from leaf 3 at similar sites throughaphid stylets. The concentrations of K +, Na+, Mg2+ and Ca2+were measured. Ion concentrations in xylem sap collected at the base of leaves1 and 3 were identical, indicating there was no preferentialdelivery of specific ions to older leaves. All ion concentrationsin the xylem decreased from the base of the leaf towards thetip; these gradients were remarkably steep for young leaves,indicating high rates of ion uptake from the xylem. The gradientsdecreased with leaf age, but did not disappear completely. In phloem sap, concentrations of K+ and total osmolality declinedslightly from the tip to the base of leaves of both controland salt-treated plants. By contrast, Na+ concentrations inphloem sap collected from salt-treated plants decreased drasticallyfrom 21 mol m–3 at the tip to 7.5 mol m–3 at thebase. Data of K/Na ratios in xylem and phloem sap were used to constructan empirical model of Na+ and K+ flows within xylem and phloemduring the life cycle of a leaf, indicating recirculation ofNa+ within the leaf. Growth of two actinorhizal species was studied in relation tothe form of N supply in water culture. Non-nodulated bog myrtle(Myrica gale) and grey alder (Alnus incana) were grown withNH4+, NH4NO3 or NO3 (4 mol m–3 N). A nodulatedseries of bog myrtle was also cultivated in N-free medium. Relative growth rate (RGR), utilization rate of N, and shoot/rootratio were highest for the two species with the N completelysupplied as NH4+. In both species, nitrate was largely reducedin the roots and the presence of NO3 in combined-N supplyalways affected the RGR and N utilization rate negatively. BothN2 fixation and complete NO3 nutrition represented conditionsof relative N-deficiency resulting in relatively low tissue-Nconcentrations and a greater allocation of dry mass to the roots.The physiological N status of nodulated M. gale plants was comparativelygood, as indicated by a normal nodule weight ratio and a relativelyhigh N2-fixing rate per unit nodule mass. However, whole-plantN2-fixing capacity remained relatively low in comparison withacquisition rates of N in combined-N plants. The anion charge from the nitrate reduction was largely directlyexcreted as an OH efflux. H + /N ratios generally agreedwith the theory. In comparison with NH4+ nutrition, carboxylateconcentrations were higher in N2-fixing M. gale plants and theH + /N ratio in nodulated plants was less than unity below thevalue for ammonium plants as previously found for other actinorhizalspecies. Therefore, NH4+ should be an energetically more attractiveN source for actinorhizal plants than N2. The results agree with commonly accepted views on energeticsof N uptake and assimilation in higher plants and support theconcept of a basically similar physiological behaviour betweennon-legumes and legumes. Plant acid-base homeostasis is achieved when the mmoles of hydroxylions produced in the plant equal the mmoles of protons. Reductionof nitrate to ammonia is the major source of hydroxyl ions whereasammonium uptake-assimilation and the metabolism of neutral sugarsto organic acids are the primary sources of protons. Soybean[Glycine max (L.) Merr plants were grown hydroponically on mediumsupplemented with 3·0 mol m–3 nitrogen providedas various combinations of KNO3 and NH4NO3 Plant growth consumedessentially all available nitrogen in each case; however, onlyin flasks supplemented with approximately 1·8 minolesof KNO3 plus 0·6 mmole of NH4NO3 was the pH of the mediumunchanged. Thus, for every mmole of nitrogen assimilated, approximately0·6 mmole of dissociable protons must have been producedby the conversion of neutral sugars to carboxylic acids. Also,it was shown that a plant obtaining all of its nitrogen fromnitrate must neutralize or excrete approximately 0·5mmole of hydroxyl ion d–1. Conversely, the plant derivingall of its nitrogen from dinitrogen must excrete or neutralizeat least 0·8 mmole of hydrogen ion d–1 whereasthe plant deriving all of its nitrogen from aminonium must excreteor neutralize approximately 2·1 mmoles of hydrogen iond–1. Nevertheless, plants grown on medium supplementedwith 2·4 mol m–1 nitrate plus 0·6 mol m–3ammonium did not achieve a higher growth rate than plants grownon 3·0 mol m–3 nitrate. Merr.] is inhibited by continuous growth in the presenceof 4· mol m–3 KNO3 The presence of 4·0 molm–3 ‘starter nitrate’ for 3-6 d during noduledevelopment, however, subsequently stimulates nodule dry weightaccumulation and nitrogenase activity. These stimulations occureven though 4· mol m–3 nitrate temporarily delaysnodule development, i.e. the late steps of nodule developmentare reversibly inhibited by a short-term exposure to 4·0mol m–3 nitrate. On the other hand, treatment with 4·0mol m–3 nitrate in excess of 14 d significantly reducesnodule dry weight Thus, extended growth in the presence of 4·0mol m–3 KNO3 seems to block both early and late stepsof nodule development. Nodulation of hydroponically grown soybeansis also inhibited by continuous growth in the presence of 2·0mol m–3 (NH4)2SO4 This inhibition is not caused by acidityof the growth medium. Plants of Lupinus albus L., cv. Ultra, were grown hydroponicallywith NO3-nutrition for 51 d under control (0.05 mol m–3Na+ and 10 mol m–3 Cl) and saline (40 mol m–3NaCI) conditions. Plants were harvested 41 and 51 d after germinationand analysed for content and net increment of C, N and the mineralcations K+, Na+, Mg2+, and Ca2+ and the anions Cl, NOJ,malate, phosphate, and SO42–. Roots, stem interaodes,petioles and leaflets were analysed separately. During the studyperiod net photosynthesis, respiratory losses of CO2 from shootand root and the composition of the spontaneously bleeding phloemsap and the root pressure xylem exudate were also determined.Using molar ratios of C over N in the transport fluids, incrementsof C and N, and photosynthetic gains as well as respiratorylosses of C, the net flows of C and N in the xylem and phloemwere then calculated as in earlier studies (Pate, Layzell andMcNeill, 1979a). Knowing the carbon flows, the ratios of ionto carbon in the phloem sap, and ion increments in individualorgans, net flows of K+, Na+, and Cl over the study periodwere also calculated. Salt stress led to a general decrease of all partial componentsof C and N partitioning indicating that inhibitions were notdue to specific effects of NaCI salinity on photosynthesis oron NO3 uptake. However, there were differences between variouslyaged organs, and net phloem export of nitrogenous compoundsfrom ageing leaves was substantially enhanced under saline conditions.In addition, NO3reduction in the roots was specificallyinhibited. Uptake and xylem transport of K+ was more severelyinhibited than photosynthetic carbon gain or NO3 uptakeby the root. K+ transport in the phloem was even more severelyrestricted under saline conditions. Na+ and Cl flowsand uptake, on the other hand, were substantially increasedin the presence of salt and, in particular, there were thenmassive flows of Na in the phloem. The results are discussedin relation to the causes of salt sensitivity of Lupinus albus.The data suggest that both a restriction of K+ supply and astrongly increased phloem translocation of Na+ contribute tothe adverse effects of salt in this species. Restriction ofK+ supply occurs by diminished K+ uptake and even more by reducedK+ cycling within the plant. Two approaches to quantifying relationships between nutrientsupply and plant growth were compared with respect to growth,partitioning, uptake and assimilation of NO3 by non-nodulatedpea (Pisum sativum L. cv. Marma). Plants grown in flowing solutionculture were supplied with NO3 at relative addition rates(RAR) of 0·03, 0·06, 0·12, and 0·18d–1, or constant external concentrations ([NO3)of 3, 10, 20, and 100 mmol m–3 over 19 d. Following acclimation,relative growth rates (RGR)approached the corresponding RARbetween 0·03–0.12 d-1, although growth was notlimited by N supply at RAR =0.18 d-1. Growth rates showed littlechange with [NO3–] between 10–100 mmol m–3(RGR=0·15 –0·16 d-1). The absence of growthlimitation over this range was suggested by high unit absorptionrates of NO3, accumulation of NO3 in tissues andprogressive increases in shoot: root ratio. Rates of net uptakeof NO3 from 1 mol m–3 solutions were assessed relativeto the growth-related requirement for NO3, showing thatthe relative uptake capacity increased with RGR between 0·03–0·06d–1 , but decreased thereafter to a theoretical minimumvalue at RGR  相似文献

Munns, R. 1985. Na+, K+ and Cl in xylem sap flowing toshoots of NaCl-treated barley.—J. exp. Bot. 36: 1032–1042. Na+, Cl and K+ concentrations were measured in xylemsap obtained by applying pressure to the roots of decapitatedbarley plants grown at external [NaCl] of 0, 25, 50, 100, 150and 200 mol m–3. For any given NaCl treatment, ion concentrationsin the xylem sap were hyperbolically related to the flux ofwater. Ion concentrations in sap collected at very low volumefluxes (without applied pressure) were 5–10 times higherthan in sap collected at moderate fluxes (under pressure). Fora given moderate volume flux, Na+ concentration in the xylemsap, [Na+]x, was only 4.0 mol m–3 at external [NaCl] of25–150 mol m–3, and increased to 7.0 mol m–3at 200 mol m–3. [Cl-]x showed a similar pattern. Thisshows there would be little difference in the rate of uptaketo the shoot of plants at 25–150 mol m–3 externalNaCl and indicates little change even at 200 mol m-3 NaCl becausetranspiration rates would be much lower. Thus the reduced growthof the shoot of plants at high NaCl concentrations is not dueto higher uptake rates of Na+ or Cl. The fluxes of Na+, Cl and K increased non-linearlywith increasing volume flux indicating little movement of saltin the apoplast. The flux of K+ increased even when [K+]x wasgreater than external [K+], indicating that membrane transportprocesses modify the K+ concentration in the transpiration streamas it flows through the root system. Potassium-Ammonium Uptake Interactions in Tobacco Seedlings   总被引:6,自引:0,他引:6  
Short-term (< 12 h) uptake experiments were conducted with6–7-week-old tobacco (Nicotiana tabacum L. cv. Ky 14)seedlings to determine absorption interactions between K+ andNH4+. At equal solution concentrations (0.5 mol m–3) netK+ uptake was inhibited 30–35% by NH4+ and NH4+ uptakewas decreased 9–24%. Removal of NH4+ resulted in completerecovery in K+ uptake rate, but NH4+ uptake rate did not recoverwhen K+ was removed. In both cases, inhibition of the uptakerate of one cation saturated as the concentration of the othercation was increased up to 0.5 mol m–3. The relative effectof K+-NH4+ interactions was not altered when Cl- was replacedwith SO42–, but the magnitudes of the uptake rates wereless in the absence of Cl-. The Vmax for NH4+ uptake was reducedfrom 128 to 105 µmol g–1 dry wt. h–1 in thepresence of 0.5 mol m–3 K+ and the Km for NH4+ doubledfrom 12 to 27 mmol m–3 in the presence of K+. The resultsof these K+-NH4+ experiments are interpreted as mixed-noncompetitiveinteractions. However, an enhanced efflux of K+ coupled to NH4+influx via an antiporter cannot be ruled out as contributingto the decrease in net K+ uptake. This work aimed to study the impacts of acquisition and assimilationof various nitrogen sources, i.e. NO3, NH4+ or NH4NO3,in combination with gaseous NH3 on plant growth and acid-basebalance in higher plants. Plants of C3 Triticum aestivum L.and C4 Zea mays L. grown with shoots in ambient air in hydroponicculture solutions with 2 mol m–3 of nitrogen source asNO3, NH4+ or NH4NO3 for 21 d and 18 d, respectively,had their shoots exposed either to 320 µg m–3 NH3or to ambient air for 7 d. Variations in plant growth (leaves,stubble and roots), and OH and H+ extrusions as wellas the relative increases in nitrogen, carbon and carboxylatewere determined. These data were computed as H+/N, H+/C, (C-A)/N,and (C-A)/C to analyse influences of different nitrogen sourceson acid-base balance in C3 Triticum aestivum and C4 Zea maysplants. Root growth in dry weight gain was significantly reduced bytreatment with 320 µg m–3 NH3 in Triticum aestivumand Zea mays growing with different N-forms, whereas leaf growthwas not significantly affected by NH3. In comparison with C3Triticum aestivum, non-fumigated C4 Zea mays had low ratiosof OH/N in NO3–3-grown plants and of H+/N in NH4+- and NH 4NO3-grown plants. Utilization of NH3 from the atmospherereduced both the OHN ratios in NO3 -grown plantsand the H+/N ratio in NH4+ - and NH4NO3 -grown plants of bothspecies. Furthermore, Zea mays had higher ratios of (C-A)/Nin NH4+ - and NH4NO3-grown plants than Triticum aestivum. Thismeans that C4 Zea mays had synthesized more organic anion perunit increase in organic N than C3 Triticum aestivum plants.Within both species, different nitrogen sources altered theratios of (C-A)/N in the order: NH4NO3>NH4+>NO3.Fumigation with NH3 increased organic acid synthesis in NO3- and NH4+ - grown plants of Triticum aestivum, whereas it decreasedorganic acid synthesis in Zea mays plants under the same conditions.Furthermore, these differences in acid-base regulation betweenC3 Triticum aestivum and C4 Zea mays plants growing with differentnitrogen sources are discussed. Changes in net photosynthesis, respiration, transpiration andcontents of total C, NO3-N and reduced N were followed throughoutthe life of leaf 6 of nitrate-dependent plants of castor beanexposed to moderate salinity stress (71 mol m–3 NaCl).Salt treatment was applied for measuring mineral flows in aparallel study (Jeschke and Pate, 1991b). Concurrent measurementswere made of solute composition and C: N molar ratios and concentrationsof reduced N and collected NO3-N in phloem sap bleeding fromshallow incisions in the top and at the base of petioles andin xylem exudates from flaps of proximal leaf midribs followingpressurization of the root system. The resulting data were usedto construct empirical models of the respective economies ofC, total N, NO3 and reduced N for a sequence of defined phasesof leaf life. Water use efficiency increased 3-fold from emergenceto a maximum of 1·5 mmol CO2 mol–1 H2O before decliningto 0·5 mmol CO2 mol–1 H2O at senescence. Xylemmolar ratios of C:N varied from 1·2–2·8,with nitrate always a smaller component than reduced N. Phloemsap C:N increased from 10–40 with leaf expansion and wasthen maintained in the range of 40–50 until falling steeplyto 20 at leaf senescence. Nitrate comprised less than 1% oftotal N in all phloem sap samples. The models of C uptake, flow,and utilization showed a major role of phloem import and thenincreasingly of laminar photosynthesis in providing C for leafgrowth. The carbon budget was thereafter characterized by ratesof phloem export closely matched to net rates of CO2 fixationby the lamina. Corresponding data for total N depicted an earlymajor role of both xylem and phloem import, but the eventualdominance of xylem import as the N source for leaf growth. Cyclingof N by xylem to phloem exchange commenced before the leaf hadachieved maximum N content, and was the major contributor tophloem export until leaf senescence when mobilized N providedmost exported N. The nitrate economy of the leaf was characterizedby early establishment of tissue pools of the ion in the petioleand to a lesser extent in the lamina, continued high rates ofnitrate reduction in the lamina but negligible assimilationin the petiole, and a release through xylem of previously accumulatedNO3 from petiole to lamina. Related data for reduced N illustratedthe much greater importance of this form of N than nitrate intransport, storage and cycling of N at all stages of leaf andpetiole life. Xylem to phloem interchanges of reduced N in petiolewere minimal in comparison with cycling through the lamina.The ratio of CO2 reduction to NO3 reduction in the lamina wasat first low (57 mol mol–1) increasing to a peak valueof 294 during mature leaf functioning before declining to 190during the presenescence phase of leaf development. This patternreflected age-related effects on water use efficiency, changesin NO3 levels in the xylem stream entering the lamina, and therelatively low photosynthetic performances of very young andsenescent laminae.
