Summary Root formation on excised mustard cotyledons was found to be determined by cotyledon age at excision and by light. Light treatment after excision enhanced root production in cotyledons excised 6 hours to 6 days after the start of imbibition and was essential for root production in cotyledons older than 6 days at excision. Three or more 12 hour light periods after excision were sufficient to induce root production in almost 100% of 7 day old cotyledons. The interposition of up to 5 days of darkness between excision and subsequent culture in the light did not reduce the final rooting percentage but delayed the onset of rooting by a time equivalent to the length of the dark period. The dry weight values of the cotyledons were directly related to the total light energy received after excision.The age and light effects on rooting may be mediated by their effects on the rate of breakdown and on the total levels of available reserves present in the cotyledons.  相似文献

Root formation was found to extend the life-span of excised cotyledons of Helianthus annuus L. markedly. Excised cotyledons of 12-day-old plants attained longer life-span, higher root number and total root length than cotyledons excised before or after 12 days. Protein content of attached cotyledons reached a maximum level 12 days after the commencement of germination followed by a decrease. Cotyledons excised 8 days after sowing showed maximum level of protein content 44 days after excision followed by a decrease. The increase was correlated with the full development of roots. The incorporation of 14C-glycine into protein followed a pattern similar to the protein content, both in attached and detached cotyledons.  相似文献

Root initiation in Cucurbita cotyledons excised at various ages was studied in nutrient solution and water. The time required for root initiation depends on the age of the cotyledon and on the rooting medium. Nutrient solution favoured root formation. Maximum number of roots, maximal total root length, and maximum life-span were found with cotyledons excised at an age of 14 days. Excision earlier or later resulted in less roots, less total root length, and shorter life-span.  相似文献

Excision of both cotyledons of Lupinus albus L. (L. termis Forssk.) seedlings at different morphological stages during the first 8 days after sowing, retarded significantly the growth of 16-day-old seedlings and decreased the content of IAA in the shoots. The effect was more obvious at the early excision (4 days after sowing). A pattern similar to that of control plants with intact cotyledons was attained when cotyledons were excised later than 8 days after sowing. It is believed that the cotyledons control the IAA content of the shoot during the early stages of development. At day 36 flower primordia were observed in controls but not in cotyledon-less plants. At this time the IAA content of the shoot apices and the growth of cotyledon-less plants were the same as in controls except for plants that lost cotyledons 4 days after sowing, which showed similar values at week 11, but did not produce flower primordia. The other cotyledon-less plants produced flower primordia 4–11 days after controls. It is postulated that the cotyledons convey certain factor(s) to the developing axis at an early stage of the development which promote flowering.  相似文献

Pea plants grown under controlled conditions for 28 days were sampled nine times during this growth period. On each sampling day 80 plants were harvested. Cotyledons were excised from 40 plants and the root-shoot axis allowed to continue growth. The other 40 plants were divided into roots, shoots, and cotyledons; the dry weights of alcohol-water extracts and residues from these organs were determined. Shoot lengths, numbers of nodes, buds, flowers, and fruits of 28-day-old plants, whose cotyledons were excised at different times, were determined. These data indicate that cotyledon excision after the 13th day has no apparent effect on seedling growth, whereas excision before the 13th day reduces growth. Glucose-U-C14 supplied to attached cotyledons of 6- and 16-day-old seedlings resulted in no major difference in the distribution pattern of recovered label throughout the plant even though 16-day-old cotyledons were depleted of their normal endogenous food reserve.  相似文献

The effect of (±)-cis-chrysanthemic acid on indole-3-acetic acid (IAA)-induced rooting was additive. Rooting in excised cucumber cotyledons was significantly promoted by several isomers of chrysanthemic acid and sodium (±)-cis-chrysanthemate at 0.18 mM. Rooting in mung bean (Phaseolus radiatus L.) hypocotyls was also stimulated by the sodium salt at 0.06–0.6 mM. Rooting of kidney bean (Phaseolus vulgaris L.) hypocotyls was also clearly enhanced by sodium (±)-cis-chrysanthemate at 0.18–6 mM.  相似文献

Inter-organ control of greening in etiolated cucumber (Cucumis sativus L. cv. Aonagajibae) cotyledons was investigated. Four- or six-day-old excised or intact etiolated cucumber cotyledons were illuminated under aerobic conditions. Excised cotyledons without hypocotyl hooks produced chlorophyll without a prolonged lag phase and the rate of chlorophyll formation was not depressed if they were illuminated immediately after excision. If the excised cotyledons were incubated in the dark before illumination, chlorophyll accumulation at the end of 6 h of continuous illumination was remarkably lowered as the dark period lengthened, especially in 6-day-old cotyledons. The rapid loss of chlorophyll-forming capacity of excised cotyledons during dark preincubation suggests a stimulatory effect of hypocotyls on the greening in cotyledons. The treatment of excised cotyledons with bleeding sap in the dark for 18 h resulted in the promotion of chlorophyll formation during subsequent continuous illumination. Partial fractionation of bleeding sap with organic solvents and paper chromatography indicates that the active substances showed the same behavior as cytokinins. These facts add weight to the hypothesis that cytokinins from roots flow into cotyledons and stimulate greening.  相似文献

Six to seven week old red clover plants (Trifolium pratense L. cv Merviot) were used to investigate the time-course of root senescence following complete and permanent excision from the shoot. Plants were grown in sand culture watered with nutrient solution. After excision of the shoots, roots were left in situ and sampled over a period of up to 42 days. Respiration rate began to decrease immediately after excision, reaching 50% of its initial value after 24 h. The decline involved a reduction in the capacity of the respiratory pathways as measured in the presence of an uncoupler (FCCP) and exogenous glucose. The reduction in respiration could be prevented by supplying 100 mM sucrose to excised roots incubated in nutrient solution at the time of excision, but not 4–5 days after excision. There was a steady reduction in the protein and soluble sugar concentrations from the time of excision and a smaller reduction in starch. Free amino acid concentrations increased immediately after excision, but the temporal dynamics differed between individual amino acids. The total concentration of free amino acids rose to a maximum value 6–13 days after excision, before declining. Under these conditions roots survived for a remarkably long period of time. Depending on the experiment, cell viability, measured as the percentage of cells with positive turgor, was unchanged for at least 20 days, and complete loss of viability was not observed until 34–42 days after excision. There was no appreciable loss of N from the roots until cell viability declined significantly. The potential implications of these results for modelling and management of N cycling in cropping systems is discussed briefly.  相似文献

Sucrose was supplied to stock plants of Pisum sativum L. cv. Alaska grown at different levels of irradiance. There was no significant effect on the rooting of the cuttings by sucrose supply to intact plants regardless of the irradiance. However, an increase in the number of roots per cutting was obtained at increasing concentrations of sucrose when the stock plants had been grown at 4 W m?2 and their cotyledons had been removed two days before the cuttings were excised. Cotyledons were removed from stock plants at different times before the excision of cuttings with the intent to regulate the endogenous supply of carbohydrate. The number of roots per cutting was reduced by removal of the cotyledons and this reduction was correlated to the number of days the stock plants had grown without cotyledons as well as to the irradiance pre-treatment. A greater reduction occurred in cuttings from plants grown under 4 W m?2 than from those grown under 38 W m?2. The growth of the stock plants and the subsequent stem growth of the cuttings was determined by the irradiance to the stock plants and by the time of removal of the cotyledons. Exogenous supply of sucrose had no effect on the stem growth of the cuttings.  相似文献

Initiation of roots on hypocotyl cuttings of Pinus contorta in vitro   总被引:1,自引:0,他引:1  
The origin of roots and wound tissue after treatments for induction of roots on hypocotyl cuttings of three-week-old Pinus contorta Dougl. ex Loud, is discussed. The cuttings were cultured in vitro and treated with 1.2 μ M to 1.5 M IBA (indole-3-butyric acid) for 6 h to 10 days. The control, which was not treated with IBA developed a wound tissue from which roots formed. Cuttings treated with IBA developed roots directly from the hypocotyl. Direct rooting was faster than indirect rooting via a wound tissue. Rooting was considered to be optimal if more than 80% of the cuttings rooted within 19 days and half of the cuttings which possessed roots after one month had acquired them within 14 days. This type of rooting was obtained after treatment with either 80 μ M IBA for 4 to 6 days or 1.25 to 5.0 m M IBA for 6 h. Suboptimal treatments gave lower rooting percentages and superoptimal treatments resulted in delayed rooting. In IBA-treated cuttings, large increases in mitotic activity (number of mitoses per mm hypocotyl) were found in the pericycle and parenchyma inside endodermis. However, the control also had similar mitotic activities as the IBA-treated cuttings but closer to the cut surface. This led us to the conclusion that similar tissues may produce either wound tissue or roots. Almost all roots obtained through direct rooting originated outside resin ducts.  相似文献

We have determined by an immunological method the endogenous levels of three cytokinins: dihydrozeatin riboside (DHZR), transzeatin riboside (tZR) and isope-ntenyladenosine (IPA) in watermelon (Citrullus vulgaris Schrad., cv. Fairfax) cotyledons that were either attached to the seedling or excised from the seed after imbibition and then grown on water. Both seedlings and cotyledons were grown either for 5 days in continuous light or for 3 days in the dark and 2 days in light. Our aim was to verify whether endogenous cytokinin levels are lower in excised than in attached cotyledons as could be expected since excised cotyledons are much more sensitive to exogenous cytokinin application. The levels of the three cytokinins were very low immediately after imbibition, but gradually increased during the following days. They were higher in excised cotyledons after 5 days of culture in the dark than in cotyledons of the same age that had developed on the seedling. Dihydrozeatin riboside was by far the most abundant of the three cytokinins in cotyledons as well as in the hypocotyl and the root.
Irradiation reduced the level of DHZR, negating the concept that light promotes cotyledon development by increasing endogenous cytokinins. Transzeatin riboside when supplied exogenously, stimulated cotyledon development at a lower concentration than the other two cytokinins. Exogenous supply of ben-zyladenine (BA) induced a strong increase in endogenous tZR already after 24 h.  相似文献

Cotyledons excised without the hypocotyl hook from 6-day-old etiolated cucumber ( Cucumis sativus L. var. Elem) seedlings accumulated a significantly higher amount of chlorophyll than cotyledons excised with hooks or intact cotyledons. It was found that maximum ehancement of greening was achieved after 2 h of dark incubation following excision. Pretreatments with red light effected an additive rise in chlorophyll level in subsequent white light after a dark incubation, suggesting that the effects of excision and phytochrome on greening act independently. Etiolated seedlings were variously dissected before greening and it was found that enhancement occurred only when cotyledons were excised at the level of the hypocotyl hook or above it. Similar results were obtained when the dissected plants were pre-treated with red light.  相似文献

Summary An experimental rooting system was developed to study in vitro adventitious root formation in hazelnut cotyledons. Experiments involveda) assay of several culture media,b) use of different developmental status of the seeds (germinated and ungerminated),c) cotyledons subjected to various light regimes, andd) different size of cotyledons slices. It was observed that higher rooting was induced in cotyledonary portions of 5- or 7-mm thickness (250 and 350 mm3, respectively) cultured on half-strength basal medium supplemented with 50 μM indole-3-butyric acid and 5 μM kinetin. Rooting was affected by light and the developmental state of seeds. Preinitiation, initiation, and root manifestation stages were defined according to specific culture periods and in relation with morphologic and histologic changes. The first histologic changes (cell division and root primordia induction) were observed after 12 days in culture. At 30 days of culture in rhizogenic medium root primodia were fully differentiated with well-developed vascular tissues.  相似文献

The activities of ribulose bisphosphate carboxylase (RuBPC) and phosphoenolpyruvate carboxylase (PEPC), as indicators of autotrophic and nonautotrophic CO2 fixation, were measured in excised cotyledons of Pinus radiata D. Don cultured for 21 days under shoot-forming (SF) and nonshoot-forming (NSF) conditions. The activity of RuBPC was found to increase in both SF and NSF cultures during the initial 5 days of culture. However, it leveled off from day 5 to day 10 and subsequently began to decrease until the end of the culture period under the SF conditions. In contrast, in the NSF cultures, RuBPC activity increased until day 15, when it was twofold higher than the maximum activity found in the SF cultures. An increase in PEPC activity of about 2.5 times the level of activity in freshly excised cotyledons was observed during the initial 5 days of culture under the SF conditions. PEPC activity began to decline after day 5 until it reached the level of activity seen in NSF cotyledons by day 15. In contrast, the activity of PEPC did not show any significant increase during the initial 10 days of culture under the NSF conditions. The Km (phosphoenolpyruvate) of PEPC from SF cotyledons was about 35% higher than that of NSF cotyledons. Cotyledons from two culture periods (days 5 and 15) were incubated for 15 seconds with NaH14CO3. The label in the malate and asparatate fractions as a percentage of total 14C incorporation was 3 times higher in the SF cotyledons than in the NSF cotyledons. A higher incorporation of 14C into products of photosynthesis under the NSF conditions was also observed.  相似文献

Summary A method is described for multiple shoot and plantlet formation from zygotic embryos of Taxus brevifolia. Adventitious bud primordia were best induced by culturing zygotic embryos on 1/2B5 medium supplemented with 10 M BA for 14 days. Further vegetative buds were produced following subculture to half-strength McCown's basal salt medium containing 1.0% activated charcoal. Individual adventitious shoots were excised and approximately 5% of these formed roots. Rooting frequency was increased to 58% by a single treatment with ABT rooting powder. Vigorous growing Taxus brevifolia plants were established after transfer to plant growth medium.  相似文献

 对采自广西海岸的木榄(Bruguiera gymnorrhiza)和红海榄(Rhizophora stylosa)胚轴进行品质检测,相关数据可作为该地区胚轴评价的参考指标。不同盐度海水水培和淡水沙培试验的结果表明:较之海水盐度为20‰和30‰的高盐度海水,10‰以下的低盐度海水对木榄胚轴萌根和发芽有明显促进作用,使始萌根日缩短一半时日(6d),平均萌根日数快7.1~4.8d,萌根率提高20%~30%,并且较早开始发芽。红海榄胚轴萌根的最佳盐度条件为20‰左右的高盐度,此时的萌根率和平均萌根率分别达到90%和68%,淡水条件下红海榄的萌根率只有40%。试验还发现木榄胚轴连续萌根率具有双峰值现象,红海榄胚轴顶端的发芽可能会受下胚轴曝光的抑制。  相似文献

Removal of the main root system of sunflower ( Helianthus annuus ) initiates adventitious root development on the lower portion of the hypocotyl. The first cytological changes (enlarged nuclei in the interfascicular parenchymatous cells adjacent to the phloem and some cell divisions) are observed 24 h after root excision. On the basis of experiments in which (a) roots, apical buds and various amounts of cotyledonary tissue were removed, (b) cuttings were subjected to various light regimes, (c) benzyladenine oas applied to cotyledons to create an artificial sink, it was concluded that the roots normally produce factors inhibiting to adventitious rooting and might be a sink for stimulatory substances produced in the shoots. The cotyledons seem to be the major source of these stimulators. Application of aqueous and ethanolic extracts of cotyledons and hypocotyls to cuttings promoted adventitious rooting.  相似文献

The initiation and development of root primordia in detachedcotyledons of Sinapis alba (white mustard) and Raphanus sativus(radish) are studied, together with the inhibitory effects ofsucrose and mineral nutrients on these processes. Root primordium initiation on petioles of excised mustard cotyledonscultured in petridishes in water commenced after 3 days andwas completed after 5 days in culture, by which time a numberof the primordia had extended and emerged from the petiole.Both sucrose and mineral nutrient solution had an inhibitoryeffect which was most marked when the cotyledonswere culturedin the solution from the time of excision. The total numberof primordia initiated, their rate of development, and the finaltotal number of emerged roots were all reduced. The later thetime of transfer from water either to sucrose or to nutrient,the less marked the inhibition. Indeed, nutrient solution enhancedroot growth in mustard when cotyledons were transferred after5 days in water when root emergence had just commenced. The effects of sucrose and nutrients in relation to applicationbefore and after initial meristem formation has taken placeare discussed, together with the ways in which these two solutionsmay exert their effect on root initiation and development.  相似文献
