In addition to several cytochromes three iron sulfur proteins were detected in mixotrophically grown cells of Ectothiorhodospira shaposhnikovii, a member of the Chromatiaceae. They were identified as a bacterial ferredoxin and two high potential iron sulfur proteins (HIPIPs). The two HIPIPs were purified and characterized. They were named according to their differing retention times on a DEAE-cellulose column using a continuous NaCl gradient: early and late HIPIP. The HIPIPs contain 4 mol of non-heme iron and 4 mol of acid labile sulfur per mol protein. Under the conditions of purification the early HIPIP (E m, 7+270 mV) was present in a semi-reduced state. Using ion-exchange chromatography the early HIPIP could be split into a reduced green-brown (pI=3.7) and an oxidized red-brown (pI=3.9) fraction. Summary Four types of striated muscle fibers with distinctive ultrastructure were defined in the Atlantic hagfish (Myxine glutinosa, L.): white, intermediate, and red fibers of m. parietalis, and red fibers of m. craniovelaris.White fibers are thick, contain very few mitochondria and fat vacuoles, and possess distinct and separate myofibrils with thin Z-disks and distinct M-lines. Intermediate fibers are thinner, possess largely similar myofibrils that often are even better separated due to a higher content of fat vacuoles and especially mitochondria and glycogen granules. Red fibers of m. parietalis contain large amounts of mitochondria, fat vacuoles, and glycogen granules. Their myofibrils possess M-lines, and although branching more, the myofibrils of red fibers conform with a Fibrillenstruktur pattern like those of white and intermediate fibers. Red fibers of m. craniovelaris are very thin, possess many smaller fat vacuoles, and large amounts of mitochondria and glycogen granules. The myofibrils are significantly thinner than in m. parietalis fibers, run as quite independent well separated units, possess thicker Z-disks, and lack M-lines. The mitochondrial membrane potential (deltapsi(m)) in apoptosis; an update   总被引:14,自引:0,他引:14  
Mitochondrial dysfunction has been shown to participate in the induction of apoptosis and has even been suggested to be central to the apoptotic pathway. Indeed, opening of the mitochondrial permeability transition pore has been demonstrated to induce depolarization of the transmembrane potential (m), release of apoptogenic factors and loss of oxidative phosphorylation. In some apoptotic systems, loss of m may be an early event in the apoptotic process. However, there are emerging data suggesting that, depending on the model of apoptosis, the loss of m may not be an early requirement for apoptosis, but on the contrary may be a consequence of the apoptotic-signaling pathway. Furthermore, to add to these conflicting data, loss of m has been demonstrated to not be required for cytochrome c release, whereas release of apoptosis inducing factor AIF is dependent upon disruption of m early in the apoptotic pathway. Together, the existing literature suggests that depending on the cell system under investigation and the apoptotic stimuli used, dissipation of m may or may not be an early event in the apoptotic pathway. Discrepancies in this area of apoptosis research may be attributed to the fluorochromes used to detect m. Differential degrees of sensitivity of these fluorochromes exist, and there are also important factors that contribute to their ability to accurately discriminate changes in m.  相似文献

On the basis of symposium contributions onChlorella, Hibbertia, Eucalyptus, Ambrosia and on numerical approaches some fundamental problems of (bio)systematics, evolution, and taxonomic categories are discussed: Methods available for analysing affinities; conflicting evidence from phenetic, biochemical, cytogenetic and other analyses; further classification problems in cases of intermediacy, etc. While sibs of various levels and their natural hierarchy often can be objectively defined, this appears impossible for particular taxonomic levels itself (e. g. species). A single objective taxonomic system of organisms is unrealistic. Glycyrrhizic acid and its 30-methyl ester were conjugated with 2-amino-1,3,4,6-tetra-O-acetyl-2-deoxy--D-glucopyranose, 2,3,4,6-tetra-O-acetyl--D-glucopyranosyl amine, 2,3,4-tri-O-acetyl--L-arabinopyranosyl amine, 2-acetamido-2-deoxy--D-glucopyranosyl amine, and -D-galactopyranosyl amine using N,N-dicyclohexylcarbodiimide and its mixtures with N-hydroxybenzotriazole. Structures of the conjugates were confirmed by IR, UV, 1H, and 13C NMR spectroscopy. The glycoconjugate with the residues of 2-acetamido-2-deoxy--D-glucopyranosyl amine in the carbohydrate part of its molecule exhibited antiviral activity (ID50 4 g/ml) toward the herpes simplex type 1 virus (HSV-1) in the VERO cell culture. Two compounds demonstrated anti-HIV-1 activity (50–70% inhibition of p24) in a culture of MT-4 cells at concentrations of 0.5–20 g/ml.  相似文献

Summary Phytophthora infestans has three kinds of somatic nuclei: an oval shaped nucleus (approx. 3.1×2.7 ) which stains diffusely except for a crescent shaped Feulgen positive cap which stains intensely; a granular nucleus whose contents are organized into a fairly constant number of stained bodies, and, a deeply staining condensed nucleus. The capped nucleus is thought to be metabolic or resting and the granular nucleus is thought to be dividing as it is most commonly found in hyphal tips. Attenuated forms of all three kinds of nuclei are found.Nuclear division is mitotic and intranuclear. Eight—ten chromosomes are seen at metaphase.Sporangia have a mean of 6.3 nuclei which is constant for age and strain of culture. Sporangia become multinucleate as a result of nuclear migration and not by division in the developing sporangium. Zoospores are usually uninucleate.The nuclear cap is persistent throughout nuclear division when it also divides. It is associated with flagella production and nuclear migration and has some of the properties of a blepharoplast.  相似文献

The concentration of airborne spores of Cladosporium and Alternaria has been investigated at five monitoring stations situated in cities from the foot of the Tatra Mountains to central Poland along a south-north transect (Zakopane, Kraków, Ostrowiec witokrzyski, Warszawa, Pozna) i.e. from a height of 900 m to 80 m above sea level. The aerobiological monitoring of fungal spores was performed by means of five Burkard volumetric spore traps.Cladosporium spores were dominant at all the stations. The highest Cladosporium and Alternaria spore concentrations were observed at all the sites in July and August, except at Warszawa in both years and at Pozna in 1995 where the maximum of Cladosporium spores occurred in June and July, and at Ostrowiec witokrzyski in 1995 where the maximum was found in July, August and September.Fungal spore concentrations in Zakopane and Kraków were significantly lower than those in Ostrowiec witokrzyski, Warszawa and Pozna and periods of abundant Cladosporium spore occurrence were different in these two groups of monitoring stations.  相似文献

Summary 1. Anaerobic amino-acid breakdown in a garden soil was accompanied by the formation of large quantities of volatile fatty acid and ammonia (plus indole in the case of tryptophan).2. Hydroxyproline, leucine, methionine, phenylalanine, proline, threonine, tryptophan, tyrosine, and valine were found to be resistant and decomposed more slowly than did alanine, arginine, aspartic acid, cystine, glutamic acid, glycine, histidine lysine, and serine. Casein hydrolysate was rapidly and completely deaminated.3. The slow rates of breakdown of the resistant acids when tested singly could not be attributed either to the effects of adsorption on the soil surface or to elimination of an essential Stickland reaction.4. The effect of glucose and of nitrate on anaerobic amino-acid metabolism have been tested. With the exception ofdl-methionine nitrate acted as an hydrogen acceptor in the anaerobic deamination of the resistant acids, glucose had no effect on their anaerobic metabolism.  相似文献

Mild cerebral anoxic/ischemic/stress insults promote tolerance and thereby protect the brain from subsequent lethal anoxic/ischemic insults. We examined whether specific activation of PKC , , , or isoforms is associated with ischemic preconditioning (IPC) in rat brain. IPC was produced by a 2-minute global cerebral ischemia. Membrane and cytosolic fractions of the hippocampi were immunoblotted using specific antibodies for PKC, , , and . PKC showed a significant translocation to the membrane fraction from 30 min to 4 h and PKC at 4 h following IPC. In contrast, the membrane/cytosol ratio of PKC showed a tendency to decrease at 30 min and 8 h, and the membrane/cytosol ratio of PKC was significantly decreased from 30 min to 24 h following IPC. These findings indicate PKC isoform-specific membrane translocations in the hippocampus after brief global brain ischemia and suggest that activation of PKC and PKC may be associated with IPC-induced tolerance in the rat hippocampus.  相似文献

Using the technique of allele-specific priming of the polymerase chain reaction (PCR), the C-T substitution in codon 39 was identified as the cause of -thalassaemia in an Irish family. Analysis of the restriction fragment length polymorphisms (RFLPs) in the -globin gene cluster established linkage of the -thalassaemia mutation to a particular -haplotype but indicated that a recombinational event had occurred in the paternal chromosome in the younger of two affected children. Non-paternity was excluded by DNA fingerprinting analysis with hypervariable minisatellite probes. This is the fourth case of recombination in the -globin gene cluster to be reported. The event has occurred 5 of the polymorphic RsaI site at position-550 bp upstream of the -globin gene mRNA Cap site, within the 9.1-kb region that has been shown to be a hot spot for recombination in the -globin gene cluster.  相似文献

The predacious watermold Zoophagus insidians traps loricate rotifers on short, lateral branches of the main, hyphal axis. These branches or traps are packed at their distal ends with a number of vesicles filled with an electron-dense matrix. Electron micrographs of the mycelium disclose a two-layered wall; the outer layer is electron dense and the inner, electron transparent. The outer dense layer on the tip of the trap is organized into a number of fine ridges and occasional discontinuities. Thin sections through recently trapped rotifers indicate that the cilia of the animals are stuck to the trap by a glue. This adhesive is derived from secretion of the matrix of the vesicles aggregated in the tip of the trap. The secretion mechanism is triggered by the animal and is accompanied by: 1. The separation of the two layers of the wall, 2. fusion of the vesicles with the cell membrane and 3. extrusion of the glue through pits in the tip of the inner wall of the trap. After snaring a rotifer, the previously arrested branch grows as a haustorium into the body cavity of the animal. The host tissues disintegrate within a few hours and appear to be the main nutrient source of the fungus.  相似文献

Summary A study was made of the effect of genotype and temperature (25 and 17°C) on sex ratio in the hybrids D. virilis Sturt. X D. littoralis Sokolov. A genetic system has been found controlling sex-differential viability. In the F1 of the reciprocal hybrids D. virilis X D. littoralis the sex ratio is normal, though at 17°C females are slightly excessive. The abnormal sex ratio is observed only in the progeny of test crosses.The major gene causing the death of female progeny of the cross [ (, D. virilis x , D. littoralis) x D. virilis] x D. littoralis is located on chromosome 2 of D. virilis. It is expressed as a lethal if chromosome 5 is heterogeneous virilis-littoralis. Chromosome 3 of D. virilis bears a modifier-enhancer and chromosome 5, a suppressor, of this lethal found in chromosome 2. This genetic system has a maternal effect and functions at 25°C, interacting with the X-chromosome of D. littoralis. If the maintainance temperature is lowered to 17°C, the progeny of the cross hybrid FB1 x D. littoralis is predominantly female. Partial death of males is accounted for by a disturbance in the interaction between the genes of X-chromosome in certain combinations with the D. virilis autosomes and the Y-chromosome of the paternal species D. littoralis.Sex-differential mortality in the hybrids D. virilis x D. littoralis is one of the isolating factors between these species which does not appear to act until the second and subsequent F1 generations due to the formation of the recombination load.  相似文献

Summary In comparison with two wild type barley cultivars, Sundance and Bomi, biochemical data show that the high-lysine mutant Hiproly contains abundant amounts of lysine-rich -amylase, whereas mutant Risø 1508, also a high-lysine mutant, contains negligible amounts of this enzyme. Immunocytochemical studies of germinating barley seeds, using both mono- and polyclonal antibodies to -amylase, support the biochemical findings of enzyme abundance in developing seeds. Three immunostaining methods for localization of -amylase were tested; of these, the avidin-biotin-peroxidase complex method, a relatively new procedure for study of plant tissues, is by far the most sensitive. -Amylase occurs predominantly in cytoplasm of the endosperm, with a minor, but previously unknown localization in the aleurone of the mid-region of the seeds. A spatial distribution of -amylase is seen. Endosperm in the upper and lower regions has the greatest amount of -amylase, with the amount decreasing toward the mid-region. In the mid-region, a limited aleurone localization of -amylase is found in all four barley strains. The function of this aleurone localization is unclear. In the endosperm, the abundance of -amylase appears to be inversely correlated with number of starch grains per unit area in Hiproly but not in Risø 1508, yet the rate of germination of the two mutants is essentially identical. The complete definition of the chemical structure of GD1b-ganglioside (GD1b) lactone isolated from human brain has been given by means of spectrometric and spectroscopic analyses. GD1h lactone contains a single ester linkage involving the external sialic acid carboxyl group and the C-9 hydroxyl group of the internal sialic acid unit. This report examines plasma amyloid proteins A40 and A42 and apolipoprotein E (apoE) levels and their relationships with age in non-demented older adults with (N = 32) or without the apoE-4 allele (N = 94). A levels did not differ between the groups whereas the 4 allele was associated with a significant reduction in plasma apoE. In subjects with the 4 allele, increasing age was associated with significant reduction in plasma A40. Subjects without the 4 allele showed a significant positive correlation between A40 and A42 levels. There was also a significant correlation between plasma A40 and apoE levels in all subjects.  相似文献

cDNAs coding for the HLA class II DR and DQ and chains of the diabetogenic haplotypes DR3 and DR4 were introduced into a mammalian expression vector and transfected into L-cell mouse fibroblasts to produce cells expressing individual human class II molecules. Stable L transfectants were generated expressing each of the DR or DQ isotypes of the cis-encoded and chains of the DR3 or DR4 haplotypes, as well as the trans-encoded and chains of the DQ molecules of the two haplotypes. However, isotype mismatched combinations (DR/DQ or DQ/DR) did not result in any stable transfectants. The stable DQ L-cell transfectants obtained, along with homozygous B-cell lines expressing the DQ2 and DQ8 specificities, were tested against a large panel of twentyone anti-HLA class II monoclonal antibodies (mAbs). Their unusual reactivity patterns are described including the failure of most pan-DQ mAbs to react with all DQ expressing L-cell transfectants. Interestingly, some mAbs react with certain heterodimers expressed on B-LCL but fail to recognize the same heterodimers expressed on the transfectants. This is suggestive of minor structural modifications that class II molecules undergo depending on the cells they are expressed on.The nucleotide sequence data reported in this paper have been submitted to the GenBank nucleotide sequence database and have been assigned the accession number U07848. The name DQB1 * 0202 was officially assigned by the WHO Nomenclature Committee in April 1994. This follows the agreed policy that, subject to the conditions stated in the most recent Nomenclature Report (Bodmer et al. 1992), names will be assigned to new sequences as they are identified. Summary A fungus identified as Cunninghamella blakesleeana (Lendner) can carry out 15-hydroxylation of lithocholic acid to a new bile acid (3,15-dihydroxy-5-cholanic acid). By optimizing the fermentation conditions, the amount of the product increased from 0.17 g/l to 1.2 g/l. Summary The histochemical activities of succinic dehydrogenase (SDH), myofibrillar Adenosine triphosphatase (ATPase) and alpha glycerophosphate dehydrogenase were studied in serial sections of rat vastus lateralis (red) (RVL), gastrocnemius and diaphragm muscles. Three main fibre-types were distinguished. The Type I fibres of RVL and gastrocnemius muscles fell into two distinct groups: one category-Type IA showed very low ATPase activity. The second category of Type IB fibres displayed moderate ATPase reaction. The Type IA fibres were divisible into two sub-groups when tested for SDH reaction. Type IA1 fibres possessed a homogenous distribution of diformazan·granules throughout the fibre: Type IA2 fibres displayed characteristic moth-eaten pattern of diformazan localization. The diaphragm muscle did not show either Type IB or Type IA2 varieties. The great majority of Type I fibres were sub-type IA1 in the three fast muscles studied. It is also demonstrated here that an inherent heterogeneity exists between Type I fibres of diaphragm and leg muscles in regard to -GPD localization. This histochemical data emphasizes the fact that subdivision of Type I striated muscle fibres of mammalian animals into two sub-types is only approximate and that a further subcategorization is possible.  相似文献
