Adenylate kinase (AK) mediates the reversible transfer of phosphate groups between the adenylate nucleotides and contributes to the maintenance of their constant cellular level, necessary for energy metabolism and nucleic acid synthesis. The AK were purified from crude extracts of two sulfate-reducing bacteria (SRB), Desulfovibrio (D.) gigas NCIB 9332 and Desulfovibrio desulfuricans ATCC 27774, and biochemically and spectroscopically characterised in the native and fully cobalt- or zinc-substituted forms. These are the first reported adenylate kinases that bind either zinc or cobalt and are related to the subgroup of metal-containing AK found, in most cases, in Gram-positive bacteria. The electronic absorption spectrum is consistent with tetrahedral coordinated cobalt, predominantly via sulfur ligands, and is supported by EPR. The involvement of three cysteines in cobalt or zinc coordination was confirmed by chemical methods. Extended X-ray absorption fine structure (EXAFS) indicate that cobalt or zinc are bound by three cysteine residues and one histidine in the metal-binding site of the “LID” domain. The sequence 129Cys-X5-His-X15-Cys-X2-Cys of the AK from D. gigas is involved in metal coordination and represents a new type of binding motif that differs from other known zinc-binding sites of AK. Cobalt and zinc play a structural role in stabilizing the LID domain.  相似文献

Adenylate kinases (AK) from Gram-negative bacteria are generally devoid of metal ions in their LID domain. However, three metal ions, zinc, cobalt, and iron, have been found in AK from Gram-negative bacteria. Crystal structures of substrate-free AK from Desulfovibrio gigas with three different metal ions (Zn2+, Zn-AK; Co2+, Co-AK; and Fe2+, Fe-AK) bound in its LID domain have been determined by X-ray crystallography to resolutions 1.8, 2.0, and 3.0 Å, respectively. The zinc and iron forms of the enzyme were crystallized in space group I222, whereas the cobalt-form crystals were C2. The presence of the metals was confirmed by calculation of anomalous difference maps and by X-ray fluorescence scans. The work presented here is the first report of a structure of a metal-containing AK from a Gram-negative bacterium. The native enzyme was crystallized, and only zinc was detected in the LID domain. Co-AK and Fe-AK were obtained by overexpressing the protein in Escherichia coli. Zn-AK and Fe-AK crystallized as monomers in the asymmetric unit, whereas Co-AK crystallized as a dimer. Nevertheless, all three crystal structures are very similar to each other, with the same LID domain topology, the only change being the presence of the different metal atoms. In the absence of any substrate, the LID domain of all holoforms of AK was present in a fully open conformational state. Normal mode analysis was performed to predict fluctuations of the LID domain along the catalytic pathway.  相似文献

We present a total of approximately 15 kb of DNA sequences, encompassing four chorion genes Ccs18, Ccs15, Ccs19, Cc16 and their flanking DNA in the medfly C. capitata. Comparison of coding regions, introns and intergenic sequences in five Dipteran species, D. melanogaster, D. subobscura, D. virilis, D. grimshawi and C. capitata documented an extensive divergence in introns and coding regions, but few well conserved elements in the proximal 5′ flanking regions in all species. These elements are related to conserved regulatory features of three of the genes, including tissue- and temporal regulation. In the fourth, gene s15, significant alterations in the 5′ flanking region may be responsible for its changed temporal regulation in C. capitata. One long intergenic sequence, located in the distal 5′ flanking region of gene s18, is homologous to ACE3, a major amplification control element and contains an 80-bp A/T-rich sequence, known to stimulate strong binding of the origin recognition complex (ORC) in D. melanogaster. Analysis of the nucleotide composition of all chorion genes in C. capitata and D. melanogaster showed that C. capitata exhibit less biased representation of synonymous codons than does D. melanogaster.  相似文献

The SMXA recombinant inbred mouse strain set was produced by systematic inbreeding from the F2 generation of a cross between two progenitor inbred strains, A/J and SM/J, which differed markedly with respect to the patterns of infection with Angiostrongylus costaricensis. We have applied this set to genetic analysis of mouse susceptibility to this nematode infection. The mortality was variable among substrains of the SMXA RI strains, indicating the involvement of multiple genes. Linkage analysis showed several chromosomal regions closely linked to mortality; chromosome 6 (D6Rik86, 87; P0.001), 10 (D10Rik66D10Mit12; P=0.0058), 13 (D13Rik79, 80; P=0.0096) and 17 (D17Mit28D17Rik76; P=0.0088).  相似文献

金钟藤(Decalobanthus boisianus)是林业有害植物, 其暴发生长和扩散对森林生态系统造成了严重破坏。本文以海南岛48个金钟藤典型分布群落为研究对象, 用方差比率法和贡献定律法探究群落的稳定性; 用χ 2统计量、联结系数(AC)、共同出现百分率(PC)、Ochiai指数和Dice指数分析金钟藤与伴生物种的种间联结关系; 用生态位宽度、生态位相似性系数和生态位重叠指数研究群落中各物种的生态位特征, 以期为金钟藤生物防治的植物物种筛选提供借鉴。结果表明: (1)金钟藤所在48个群落共有156种伴生植物, 其中大戟科、茜草科、桑科、无患子科和樟科植物占优势; (2)群落中优势物种呈正联结关系, 植物种类累积倒数百分比与累积相对频度交点坐标为(44.53, 55.47), 远离稳定交点坐标(20, 80), 说明群落处于不稳定状态; (3)金钟藤与芳槁润楠(Machilus suaveolens)、黄椿木姜子(Litsea variabilis)、岭南山竹子(Garcinia oblongifolia)、显脉杜英(Elaeocarpus dubius)、鸭脚木(Schefflera octophylla)和银柴(Aporusa dioica)都紧密关联, 说明金钟藤与这些物种的资源利用方式较相似; (4)金钟藤的生态位宽度最大, 与伴生物种间的生态位重叠度较高, 但伴生物种间的生态位重叠度较低。金钟藤的入侵导致群落处于不稳定状态, 并与伴生物种间存在激烈的竞争关系。因此, 建议在金钟藤已入侵的群落中大量栽种芳槁润楠、黄椿木姜子、显脉杜英、鸭脚木和银柴, 以遏制其蔓延; 大量栽种翻白叶树(Pterospermum heterophyllum)、海南菜豆树(Radermachera hainanensis)、九节(Psychotria rubra)和肉实树(Sarcosperma laurinum)用于金钟藤入侵群落的植被恢复。  相似文献

Aims Dissolved organic matter (DOM) is the most active component of organic matters in soils, and plays an important role in carbon cycles. It is a mixed organic compound with varying molecular sizes and weights. We aimed to explore the impacts of Dicranopteris dichotoma coverage on quantity and structure of DOM after vegetation restoration in severely eroded red soil region. Methods A typical sequence of vegetation restoration (Y0, without ecological restoration; Y13, ecological restoration for 13 years; Y31, ecological restoration for 31 years) was selected as the research object in Hetian Town, Changting County, Fujian Province, China. At each experimental site, soils were subject to three treatments—NRd, not removed D. dichotoma; Rd, removed D. dichotoma; and CK, control, and the effects of D. dichotoma on the spectral characteristics of DOM were evaluated.Important findings The results indicated that the quantity of soil DOC under NRd treatment of the Y0, Y13 and Y31 was 7.61, 4.83, and 5.47 times higher than their CK treatment, respectively. The Rd treatment had significantly lower DOC than that under NRd treatments, and it was 1.84, 4.12, and 4.73 times higher than their CK treatments, respectively. Thus the D. dichotoma had exerted significant effects on the quantity of soil DOM. The Aromaticity index (AI), emission fluorescence spectrum humification index (HIXem) and synchronous fluorescence spectrum humification index (HIXsyn) of DOM under the NRd treatment were significantly higher than those of the CK treatments in Y13 and Y31, respectively. However, the ratio of ultraviolet-visible light absorption photometric quantity at 250 nm wavelength to ultraviolet-visible light absorption photometric quantity at 365 nm wavelength (E2:E3) had an opposite trend. It showed that the DOM structure in soils covered by D. dichotoma contained more aromatic nucleus and had higher aromaticity and humification, and DOM molecular was larger. In addition, the AI and humification index (HIX) of DOM under the Rd treatment was significantly decreased compared with the NRd treatment. Similar results were observed by analysis of emission and synchronous fluorescence spectrum, and by the Fourier infrared transmission spectrum analysis. These results suggest that D. dichotoma had positive impacts on the complexity of DOM structure, but it was a long and slow process. The DOM spectral analysis showed that the soil DOM covered by D. dichotoma had a stable and complex structure and was easily adsorbed by soil colloid. As a result, Dicranopteris dichotoma had a positive effect on the accumulation of soil organic carbon.  相似文献

通过盆栽试验研究了接种丛枝菌根真菌(AM真菌)、种植共生植物(三叶草)、添加重金属螯合物EDTA和磷肥磷酸钙对铅污染下瞿麦生长和品质的影响,为科学种植中药材提供理论依据.结果表明: 接种AM真菌可以显著抑制铅的吸收(P<0.05),促进根部发育,使根冠比增大,且活性成分累积最多,大黄素含量为6.5 mg·g-1;与三叶草共生后,抑制铅吸收的效果不佳且药材品质下降,大黄素含量低于对照组,降至3.2 mg·g-1.但在AM真菌共同参与后瞿麦的生长量和活性成分有所增加,铅含量达到最低,低至1.3 mg·g-1;添加重金属螯合剂会使瞿麦生长量下降并促进根部对铅的吸收,铅含量最高可达340.0 mg·g-1;磷酸钙可固定土壤中其他重金属元素,故更适宜在复合型污染时使用.综合考虑,在保护中药材安全、品质方面AM真菌具有良好的应用价值.  相似文献

The general features of the tegument of Dactylogyrus amphibothrium and D. hemiamphibothrium, gill parasites of the freshwater fish Acerina cernua, resemble those of other monogeneans. However, in D. amphibothrium the tegument bears spike-like structures associated with tegumental sacs and these have not previously been reported in monogeneans. The origin of the sacs and the possible functions of the spikes and sacs are discussed. It has been found that the tegument covering the two pads located on the ventral surface of D. amphibothrium does not differ in structure from the general body tegument; the pad regions are elevated above the general body surface because of the presence of large glandular sub-tegumentary organs which communicate with the receptaculum seminis. This indicates that the pads play a part in the reproductive biology of the parasite. In spite of the fact that the micro-environments of D. amphibothrium and D. hemiamphibothrium are the same or broadly similar, D. hemiamphibothrium has no spikes, tegumentary sacs or glandular pads and has only two kinds of tegumentary secretory inclusion compared with three kinds in D. amphibothrium. One of the three kinds of cyton in D. amphibothrium is restricted to the ventro-lateral regions of the posterior half of the body. The tegument of both D. amphibothrium and D. hemiamphibothrium differs from that of other adult monogeneans so far investigated in the presence of granular endoplasmic reticulum in the outer syncytial layer.  相似文献

A new species of sulfate-reducer, Desulfosporosinus meridiei, was recently isolated from gasoline-contaminated anaerobic groundwater in which degradation of toluene and other hydrocarbons occurred. Ground-water from inside (three sites) and outside (one site) the contaminant plume was probed with specific polyclonal antibodies raised against two strains of D. meridiei (strains T2 and S6). Molecular 16S rRNA probes designed to hybridize with cells of D. meridiei were also used. Cell counts using antistrain T2 antibodies (specific for all strains of D. meridiei and two strains of D. orientis) were similar (103 cells/mL) both inside and outside the plume as were total DAPI counts (106 cells/mL). The numbers of cells stained with antibodies specific for Group B strains of D. meridiei varied between locations. The molecular probes DSP477A and DSP477B were designed for, and were effective on, pure cultures of D. meridiei and were able to distinguish this species from Desulfovibrio desulfuricans in hybridization experiments. No cells were seen to hybridize with probe DSP477B in groundwater samples. Cell counts in groundwater using the universal eubacterial probe, EUB338, were only 8% to 29% of DAPI counts. Fluorescence intensity was poor and auto fluorescence of particles made counting difficult. This study showed that molecular probing using techniques commonly employed in many laboratories was of little use for evaluating microbial populations in this groundwater. Polyclonal antibodies were considerably more useful for identifying populations of specific cells. The lack of difference in cell numbers between contaminated and nearby uncontami-nated groundwater suggests that cell counts will not always be useful as indicators of intrinsic remediation.  相似文献

Disa cardinalis and three populations within the D. tripetaloides species complex contain variation in their chloroplast DNA (cpDNA) variability. All four taxa possessed unique cpDNAs and sequence divergence values ranged from 0.34 to 1.03%. A phylogeny of these genomes was reconstructed, along with the genomes of three other species, D. racemosa, D. uniflora and D. venosa, all of which are also section Disa and series RAcemosae, to determine the relationship of these closely related species to the D. tripetaloides complex. A phylogeny of the taxa using morphological data was also reconstructed. Outgroup comparison was made with D. sagittalis, a member of section Coryphaea. Although the molecular and morphological data were not completely congruent, both data types revealed D. cardinalis, rather than D. tripetaloides ssp. aurata, to be more closely allied with D. tripetaloides ssp. tripetaloides, suggesting that D. tripetaloides ssp. aurata should be elevated to species rank. Additionally, the high sequence divergence observed between the Natal and Cape populations, coupled with their geographical isolation and alternate flowering seasons, suggests that these two D. tripetaloides ssp. tripetaloides populations may, in fact, be more appropriately ranked as subspecies.  相似文献

茎柔鱼(Dosidicus gigas)广泛分布于东太平洋海域,在海洋生态系统中扮演着重要角色。本研究根据2019年6—12月我国鱿钓生产船在东太平洋公海采集的62尾茎柔鱼样本,对其胃含物残留的4131个耳石和75个角质颚样本进行鉴定分析。结果表明: 茎柔鱼主要摄食鱼类10种、头足类4种。从摄食饵料的出现频率和数量百分比来看,主要摄食的鱼类饵料为荧串光鱼、朗明灯鱼和墨西哥尾灯鱼,荧串光鱼在不同胴长组的茎柔鱼胃内均有出现,是茎柔鱼最重要的饵料;摄食的头足类主要有茎柔鱼、鸢乌贼和爪乌贼。随着胴长的增大,茎柔鱼摄食的饵料种类不断增加,头足类和灯笼鱼饵料比例增加,且摄食的饵料尺寸增大,摄食营养结构发生改变,摄食营养水平随之提高。研究结果可为评估不同饵料生物在茎柔鱼食性转变过程中的食物贡献提供基础资料。  相似文献

中华蚊母树(Distylium chinense)是华中地区特有种, 在三峡库区局部地段占优势形成群落。对三峡库区不同生境下中华蚊母树群落特征及其与土壤环境因子的关系进行研究可为其保护及消落带植被恢复提供科学依据。运用数据分类和排序等方法, 对中华蚊母树群落物种组成、植物区系、物种多样性及其与土壤环境因子的关系进行研究。结果表明, 共发现维管植物56种, 隶属于36科54属, 植物区系以世界分布、泛热带分布和北温带分布为主, 生活型以草本和灌木为主。自然消落带灌木层重要值最大的是中华蚊母树, 草本层重要值最大的是藤本植物地果(Ficus tikoua), 主要伴生种为具有一定水淹耐受性的灌木和多年生草本, 如小梾木(Swida paucinervis)、白茅(Imperata cylindrica)等; 反季节消落带灌木层仅中华蚊母树一种, 草本层重要值最大的物种是狗牙根(Cynodon dactylon), 主要伴生种为苍耳(Xanthium sibiricum)、苘麻(Abutilon theophrastic)等一年生草本。双向聚类分析将调查的6个样地分为四大类群: 中华蚊母树+小梾木群落、中华蚊母树+地果群落、中华蚊母树+细叶水团花(Adina rubella)群落、中华蚊母树+狗牙根群落。典范对应分析表明, 海拔高度和土壤pH是影响中华蚊母树群落物种分布的主要环境因子, 土壤有机质、全氮、全磷和速效钾含量是次要因素, 但它们是影响群落物种多样性指数的主要因子, 其中土壤氮是群落的限制因子。因此, 在反季节消落带植被重建中, 对中华蚊母树群落进行构建时, 除考虑细叶水团花、小梾木等作为高海拔消落带伴生物种外, 还应增加土壤氮和钾的供给, 使中华蚊母树群落保持较高的物种多样性, 维持消落带生态系统稳定。  相似文献

A novel adenylate kinase (AK) has recently been purified from Desulfovibrio gigas and characterized as a Co(2+)/Zn(2+)-containing enzyme: this is an unusual characteristic for AKs from Gram-negative bacteria, in which these enzymes are normally devoid of metals. Here, we studied the conformational stability of holo- and apo-AK as a function of temperature by differential scanning calorimetry (DSC), circular dichroism (CD), and intrinsic fluorescence spectroscopy. The thermal unfolding of AK is a cooperative two-state process, and is sufficiently reversible in the 9-11 pH range, that can be correctly interpreted in terms of a simple two-state thermodynamic model. The spectral parameters as monitored by ellipticity changes in the CD spectra of the enzyme as well as the decrease in tryptophan intensity emission upon heating were seen to be good complements to the highly sensitive but integral DSC-method.  相似文献

The sensitivity of the catalytic activities of the D. salina chloroplast coupling factor 1 (CF1) to chemical modification by N-ethylmaleimide has been investigated. (i) When D. salina thylakoid membranes are treated with N-ethylmaleimide, both photophosphorylation and the inducible CF1 ATPase activity are partially (approx. 60%) inhibited. The inhibition of both activities does not require the presence of a proton-motive force, and the inhibition of photophosphorylation is directly related to the N-ethylmaleimide-covalent modification of CF1 as shown by (a) the time-course for the inhibition and (b) the maximal extent of inhibition. (ii) Treatment of the purified, latent, D. salina CF1 with low concentrations of N-ethylmaleimide also results in the partial (approx. 60%) inhibition of the inducible ATPase activity (I50 ≈ 50 μM). The inhibition does not require the presence of the chemical modifier during the activation of the enzyme. (iii) N-ethylmaleimide-induced inhibition of the ATPase activity of either membrane-bound or solubilized CF1 is partially reversed by either (a) prolonged incubation at low concentrations of N-ethylmaleimide or (b) short incubation times at high concentrations of N-ethylmaleimide. The results are interpreted as indicating multiple binding sites on the D. salina CF1 that have different rates of reactivity with N-ethylmaleimide. Those sites (or site) that react rapidly with N-ethylmaleimide cause(s) an inhibition of both ATP synthase and ATPase activities, whereas those sites (or site) that react more slowly partially restore(s) the original-ATPase activity. The effects of N-ethylmaleimide on the catalytic activity of D. salina CF1 are probably mediated by N-ethylmaleimide-induced conformational changes of the enzyme.  相似文献

In this work we present the synthesis, structural and spectroscopic characterisation of a series of cobalt(II) and zinc(II) coordination compounds with benzimidazole (bz) and its 2-benzimidazole derivatives: 2-aminobenzimidazole (2ab), albendazole (abz) and tris(2-benzimidazolylmethyl)amine (ntb). The compounds were evaluated for their in vitro antimicrobial activity against Staphylococcus aureus, Micrococcus luteus, Salmonella typhi, Pseudomonas aeruginosa, Escherichia coli and Proteus vulgaris. Their cytotoxic activity was also evaluated using human cancer lines, HeLa, HCT-15 and SKLU-1. The halide tetrahedral compounds [Co(bz)2Br2] 3, [Zn(2ab)2Cl2] · 0.5H2O 11, [Co(abz)Cl2(H2O)] · 3H2O 14, [Co(abz)Br2(H2O)] 15, [Zn(abz)Cl2(H2O)] · 3H2O 17 and [Zn(abz)Br2(H2O)] · H2O 18 displayed similar minimal inhibition concentration (MIC) values against Micrococcus luteus and Escherichia coli, comparable to those of amoxicillin and chloramphenicol. Additionally, 11 showed a wide range of activity towards Gram(+) and Gram(−) microorganisms. The tetradentate ntb and its trigonal bipyramidal cobalt(II) and zinc(II) compounds were active, regardless of the anion present in the complex. Compound [Co(abz)Cl2(H2O)] · 3H2O 14 showed promising activity in HeLa cells, while [Co(ntb)Br]Br · H2O 21 inhibited Hela and HCT-15 cell lines.  相似文献

本文研究了兰科石斛属鼓槌石斛的地理分布与菌根真菌区系组成的相关性。采用克隆文库技术对云南省西双版纳和临沧市两个地区的8份野生鼓槌石斛根部样品进行了分析。结果表明,共从两个地区的鼓槌石斛根部获得了14个真菌的可操作分类单元(OTU),主要隶属于胶膜菌科Tulasnellaceae和小纺锤菌目Atractiellales。分析鼓槌石斛对菌根真菌的专一性和偏好性发现,鼓槌石斛对菌根真菌专一性较低,每个植株都能同时与多种菌根真菌共生;不同地理分布的鼓槌石斛菌根真菌类群存在明显差异,两个地区的鼓槌石斛的菌根真菌中只有1个OTU是相同的,其余均不同。说明菌根真菌可能和鼓槌石斛的生态适应存在一定的相关性。  相似文献

为探讨阿维菌素防治马尾松毛虫的抗性风险和抗性机制,本研究采用叶片浸药法测定了马尾松毛虫抗阿维菌素种群(AV)对多种杀虫剂的交互抗性及增效剂胡椒基丁醚(PBO)、磷酸三苯酯(TPP)和顺丁烯二酸二乙酯(DEM)对马尾松毛虫AV种群、秦岭接官亭种群(JGT)和敏感种群(S)的增效作用,并使用分光光度计测定了AV、JGT和S种群的羧酸酯酶(CarE)、谷胱甘肽转移酶(GST)和多功能氧化酶系(MFOs)活性。结果表明: 马尾松毛虫AV种群与甲氨基阿维菌素苯甲酸盐(抗性倍数RR50=25.0)、毒死蜱(RR50=19.0)和高效氯氟氰菊酯(RR50=15.4)存在中等水平交互抗性,与溴虫腈存在低水平交互抗性(RR50=8.1),与乙基多杀菌素、多杀菌素和氯虫苯甲酰胺不存在交互抗性。增效剂PBO与TPP对马尾松毛虫AV、JGT和S种群均有显著增效作用,DEM对马尾松毛虫AV、JGT和S种群均无显著增效作用。马尾松毛虫AV种群MFOs中细胞色素P450含量(3.5倍)、细胞色素b5含量(3.1倍)、O-脱甲基酶活性(4.1倍)和CarE活性(2.2倍)均显著高于S种群,AV种群GST活性与S种群不存在显著性差异。MFOs及CarE活性增强是马尾松毛虫对阿维菌素产生抗性的重要生化机制,推荐乙基多杀菌素、多杀菌素和氯虫苯甲酰胺与阿维菌素轮换使用防治马尾松毛虫。  相似文献
