Introduction

The aphid Rhopalosiphum padi L. is a vector of Barley yellow dwarf virus (BYDV) in wheat and other economically important cereal crops. Increased atmospheric CO2 has been shown to alter plant growth and metabolism, enhancing BYDV disease in wheat. However, the biochemical influences on aphid metabolism are not known.

Objectives

This work aims to determine whether altered host-plant quality, influenced by virus infection and elevated CO2, impacts aphid weight and metabolism.

Methods

Untargeted 1H NMR metabolomics coupled with multivariate statistics were employed to profile the metabolism of R. padi reared on virus-infected and non-infected (sham-inoculated) wheat grown under ambient CO2 (aCO2, 400 µmol mol?1) and future, predicted elevated CO2 (eCO2, 650 µmol mol?1) concentrations. Un-colonised wheat was also profiled to observe changes to host-plant quality (i.e., amino acids and sugars).

Results

The direct impacts of virus or eCO2 were compared. Virus presence increased aphid weight under aCO2 but decreased weight under eCO2; whilst eCO2 increased non-viruliferous (sham) aphid weight but decreased viruliferous aphid weight. Discriminatory metabolites due to eCO2 were succinate and sucrose (in sham wheat), glucose, choline and betaine (in infected wheat), and threonine, lactate, alanine, GABA, glutamine, glutamate and asparagine (in aphids), irrespective of virus presence. Discriminatory metabolites due to virus presence were alanine, GABA, succinate and betaine (in wheat) and threonine and lactate (in aphids), irrespective of CO2 treatment.

Conclusion

This study confirms that virus and eCO2 alter host-plant quality, and these differences are reflected by aphid weight and metabolism.

Key message

The CO 2 effect on the root production of a broad-leaved community was insignificant when grown in brown forest soil, however, it was positively large when grown in volcanic ash soil.

Abstract

We evaluated the root response to elevated CO2 fumigation of 3 birches (Betula sp.) and 1 deciduous oak (Quercus sp.) grown in immature volcanic ash soil (VA) or brown forest soil (BF). VA is a nutrient-poor, phosphorus-impoverished soil, broadly distributed in northern Japan. Each species had been exposed to either ambient (375–395 μmol mol?1) (aCO2) or elevated (500 μmol mol?1) (eCO2) CO2 during the daytime (more than 70 μmol m?2 s?1) over 4 growing seasons. The results suggest that eCO2 did not cause an increase in total root production when the community had grown in fertile BF soil, however, it did cause a large increase when the community was grown in infertile VA soil. Yet, carbon allocation to plant roots was not affected by eCO2 in either the BF or VA soils. Rhizo-morphogenesis appeared to occur to a greater extent under eCO2. It seems that the saplings developed a massive amount of fine roots under the VA and eCO2 conditions. Unexpectedly, eCO2 resulted in a larger total root mass when the community was grown in VA soil than when grown in BF soil (eCO2 × VA vs. eCO2 × BF). These results may hint to a site-specific potential of communities to sequester future atmospheric carbon. The growing substance of plants is an important factor which root response to eCO2 depends on, however, further studies are needed for a better understanding.

Aims

Despite extensive studies on effects of elevated CO2 concentration ([CO2]e) on plant growth, few studies have investigated the responses of native grassland plant species to [CO2]e in terms of nutrient acquisition.

Methods

The effects of [CO2]e (769 ± 23 ppm) on Artemisia frigida and Stipa krylovii, two dominant species in Inner Mongolia steppe were investigated by growing them for 7 weeks in Open-Top Chambers (OTC).

Results

Exposure to [CO2]e enhanced shoot and root growth of A. frigida and S. krylovii. Elevated [CO2] increased photosynthetic rates (Pn) by 34 % in A. frigida but decreased Pn by 52 % in S. krylovii. Moreover, root-secreted acid phosphatase activity in A. frigida was stimulated by [CO2]e, while exudation of malate from roots of S. krylovii was suppressed by [CO2]e. Exposure to [CO2]e led to a decrease in P concentration in shoots and roots of A. frigida and S. krylovii, but total amount of P accumulated in shoots and roots of both species was increased by [CO2]e.

Conclusions

The two dominant species in temperate steppes differed in their responses to [CO2]e, such that A. frigida was more adapted to [CO2]e than S. krylovii under low availability of soil P.

Impact of different levels of elevated CO 2 on the activity of Frankia (Nitrogen-fixing actinomycete) in Casuarina equisetifolia rooted stem cuttings has been studied to understand the relationship between C. equisetifolia, Frankia and CO2. The stem cuttings of C. equietifolia were collected and treated with 2000 ppm of Indole Butyric Acid (IBA) for rooting. Thus vegetative propagated rooted stem cuttings of C. equisetifolia were inoculated with Frankia and placed in the Open top chambers (OTC) with elevated CO2 facilities. These planting stocks were maintained in the OTC for 12 months under different levels of elevated CO2 (ambient control, 600 ppm, 900 ppm). After 12 months, the nodule numbers, bio mass, growth, and photosynthesis of C. equisetifolia rooted stem cuttings inoculated with Frankia were improved under 600 ppm of CO2. The rooted stem cuttings of C. equisetifolia inoculated with Frankia showed a higher number of nodules under 900 ppm of CO2 and cuttings without Frankia inoculation exhibited poor growth. Tissue Nitrogen (N) content was also higher under 900 ppm of CO2 than ambient control and 600 ppm levels. The photosynthetic rate was higher (17.8 μ mol CO2 m?2 s?1) in 900 ppm of CO2 than in 600 ppm (13.2 μ mol CO2 m?2 s?1) and ambient control (8.3 μ mol CO2 m?2 s?1). This study showed that Frankia can improve growth, N fixation and photosynthesis of C. equietifolia rooted stem cuttings under extreme elevated CO2 level conditions (900 ppm).  相似文献

Background and aims

Peatland moss communities play an important role in ecosystem function. Drivers such as fire and atmospheric pollution have the capacity to influence mosses via multiple pathways. Here, we investigate physical and chemical processes which may influence establishment and growth of three key moss species in peatlands.

Methods

A controlled factorial experiment investigated the effects of different peat bulk density, ash deposition and rainwater chemistry treatments on the growth of Sphagnum capillifolium, S. fallax and Campylopus introflexus.

Results

Higher peat bulk density limited growth of both Sphagnum species. S. capillifolium and C. introflexus responded positively to ash deposition. Less polluted rain limited growth of C. introflexus. Biomass was well correlated with percentage cover in all three species.

Conclusions

Peat bulk density increases caused by fire or drainage can limit Sphagnum establishment and growth, potentially threatening peatland function. Ash inputs may have direct benefits for some Sphagnum species, but are also likely to increase competition from other bryophytes and vascular plants which may offset positive effects. Rainwater pollution may similarly increase competition to Sphagnum, and could enhance positive effects of ash addition on C. introflexus growth. Finally, cover can provide a useful approximation of biomass where destructive sampling is undesirable.

Experiments employing free-air CO2 enrichment (FACE) facilities have indicated that elevated atmospheric carbon dioxide (eCO2) stimulates growth in diverse terrestrial ecosystems. Studies of the effects of eCO2 on wetland plants have indicated a similar response, but these studies were mostly performed in growth chambers. We conducted a 2-year FACE experiment [CO2 ≈ 582 µmol mol?1] in a marsh in Spain to test whether the common reed (Phragmites australis) responds to carbon enrichment, as previously reported in other macrophytes. More specifically, we tested the effect of eCO2 on P. australis growth, photosynthesis, transpiration, and biomass, its effect on modifying plant and soil ratios of carbon, nitrogen, and phosphorus, and whether the strong environmental variability of this wetland modulates these responses. Our findings show that effects of eCO2 in this wetland environment are more complex than previously believed, probably due to hydrological effects. The effects of eCO2 on reed plants were cumulative and manifested at the end of the growing season as increased 38–44% instantaneous transpiration efficiency (ratio of net photosynthesis to transpiration), which was dependent on plant age. However, this increase did not result in a significant increase in biomass, because of excessive root exudation of carbon. These observations contrast with previous observations of wetland plants to increased atmospheric CO2 in growth chambers and shed new light on the role of wetland plants as a carbon sink in the face of global climate change. The combined effects of water stress, eCO2, and soil carbon processes must be considered when assessing the function of wetlands as a carbon sink under global change scenarios.  相似文献

Human activities have recently caused severe destruction of Sphagnum wetlands in subtropical high-mountain regions, calling for urgent efforts to restore Sphagnum wetlands. Through a greenhouse experiment in western Hubei, China, we studied the effects of different substrate types (peat and mountain soil) and different levels of nitrogen (N) (0, 2, 4, 6, 10 g m?2 year?1) and phosphorus (P) (0, 0.2, 0.5, 1, 2 g m?2 year?1) on the growth of Sphagnum palustre, which was evaluated by four growth indicators: length growth, number of capitula, coverage change and biomass. We aimed to determine the optimal nutrient conditions for S. palustre growth, which would contribute to the rapid colonization and restoration of Sphagnum wetlands. The results showed that the different substrates significantly influenced S. palustre growth. Compared with those of peat, the acidic properties of the local yellow brown soil in the subtropical high-mountain regions were more favorable for S. palustre growth. As N addition increased, the four growth indicators responded inconsistently to the different substrates. While the number of capitula markedly increased, the other three indicators significantly decreased in the mountain soil or exhibited no definitive changes in the peat. The addition of P markedly promoted S. palustre growth in both substrates. However, a threshold for P fertilization existed; the highest productivity occurred at P additions of 0.2 and 0.5 g m?2 year?1 in the peat and mountain soil, respectively. The N and P contents in the capitula increased in parallel as the N and P fertilization rates increased, suggesting that these nutrients were absorbed proportionately and were used during the growth of S. palustre.  相似文献

In this work, the structural, compositional, optical, and dielectric properties of Ga2S3 thin films are investigated by means of X-ray diffraction, scanning electron microscopy, energy dispersion X-ray analysis, and ultraviolet—visible light spectrophotometry. The Ga2S3 thin films which exhibited amorphous nature in its as grown form are observed to be generally composed of 40.7 % Ga and 59.3 % S atomic content. The direct allowed transitions optical energy bandgap is found to be 2.96 eV. On the other hand, the modeling of the dielectric spectra in the frequency range of 270–1,000 THz, using the modified Drude-Lorentz model for electron-plasmon interactions revealed the electrons scattering time as 1.8 (fs), the electron bounded plasma frequency as ~0.76–0.94 (GHz) and the reduced resonant frequency as 2.20–4.60 ×1015 (Hz) in the range of 270–753 THz. The corresponding drift mobility of electrons to the terahertz oscillating incident electric field is found to be 7.91 (cm 2/Vs). The values are promising as they nominate the Ga2S3 thin films as effective candidates in thin-film transistor and gas sensing technologies.  相似文献

The classical definition of mesophyll conductance (g m) represents an apparent parameter (g m,app) as it places (photo)respired CO2 at the same compartment where the carboxylation by Rubisco takes place. Recently, Tholen and co-workers developed a framework, in which g m better describes a physical diffusional parameter (g m,dif). They partitioned mesophyll resistance (r m,dif = 1/g m,dif) into two components, cell wall and plasmalemma resistance (r wp) and chloroplast resistance (r ch), and showed that g m,app is sensitive to the ratio of photorespiratory (F) and respiratory (R d) CO2 release to net CO2 uptake (A): g m,app = g m,dif/[1?+?ω(F?+?R d)/A], where ω is the fraction of r ch in r m,dif. We herein extend the framework further by considering various scenarios for the intracellular arrangement of chloroplasts and mitochondria. We show that the formula of Tholen et al. implies either that mitochondria, where (photo)respired CO2 is released, locate between the plasmalemma and the chloroplast continuum or that CO2 in the cytosol is completely mixed. However, the model of Tholen et al. is still valid if ω is replaced by ω(1?σ), where σ is the fraction of (photo)respired CO2 that experiences r ch (in addition to r wp and stomatal resistance) if this CO2 is to escape from being refixed. Therefore, responses of g m,app to (F?+?R d)/A lie somewhere between no sensitivity in the classical method (σ =1) and high sensitivity in the model of Tholen et al. (σ =0).  相似文献

Stomatal conductance (g s) of mature trees exposed to elevated CO2 concentrations was examined in a diverse deciduous forest stand in NW Switzerland. Measurements of g s were carried out on upper canopy foliage before noon, over four growing seasons, including an exceptionally dry summer (2003). Across all species reductions in stomatal conductance were smaller than 25% most likely around 10%, with much variation among species and trees. Given the large heterogeneity in light conditions within a tree crown, this signal was not statistically significant, but the responses within species were surprisingly consistent throughout the study period. Except during a severe drought, stomatal conductance was always lower in trees of Carpinus betulus exposed to elevated CO2 compared to Carpinus trees in ambient air, but the difference was only statistically significant on 2 out of 15 days. In contrast, stomatal responses in Fagus sylvatica and Quercus petraea varied around zero with no consistent trend in relation to CO2 treatment. During the 2003 drought in the third treatment year, the CO2 effect became reversed in Carpinus, resulting in higher g s in trees exposed to elevated CO2 compared to control trees, most likely due to better water supply because of the previous soil water savings. This was supported by less negative predawn leaf water potential in CO2 enriched Carpinus trees, indicating an improved water status. These findings illustrate (1) smaller than expected CO2-effects on stomata of mature deciduous forest trees, and (2) the possibility of soil moisture feedback on canopy water relations under elevated CO2.  相似文献

Do short-term fluctuations in CO2 concentrations at elevated CO2 levels affect net CO2 uptake rates of plants? When exposed to 600 μl CO2 l?1, net CO2 uptake rates in shoots or leaves of seedlings of two tropical C3 tree species, teak (Tectona grandis L. f.) and barrigon [Pseudobombax septenatum (Jacq.) Dug.], increased by 28 and 52% respectively. In the presence of oscillations with half-cycles of 20 s, amplitude of ca. 170 μl CO2 l?1 and mean of 600 μl CO2 l?1, the stimulation in net CO2 uptake by the two species was reduced to 19 and 36%, respectively, i.e. the CO2 stimulation in photosynthesis associated with a change in exposure from 370 to 600 μl CO2 l?1 was reduced by a third in both species. Similar reductions in CO2-stimulated net CO2 uptake were observed in T. grandis exposed to 40-s oscillations. Rates of CO2 efflux in the dark by whole shoots of T. grandis decreased by 4.8% upon exposure of plants grown at 370 μl CO2 l?1 to 600 μl CO2 l?1. The potential implications of the observations on CO2 oscillations and dark respiration are discussed in the context of free-air CO2 enrichment (FACE) systems in which short-term fluctuations of CO2 concentration are a common feature.  相似文献

Background

The greenhouse gas (GHG) mitigation is one of the most important environmental benefits of using bioenergy replacing fossil fuels. Nitrous oxide (N2O) and methane (CH4) are important GHGs and have drawn extra attention for their roles in global warming. Although there have been many works of soil emissions of N2O and CH4 from bioenergy crops in the field scale, GHG emissions in large area of marginal lands are rather sparse and how soil temperature and moisture affect the emission potential remains unknown. Therefore, we sought to estimate the regional GHG emission based on N2O and CH4 releases from the energy crop fields.

Results

Here we sampled the top soils from two Miscanthus fields and incubated them using a short-term laboratory microcosm approach under different conditions of typical soil temperatures and moistures. Based on the emission measurements of N2O and CH4, we developed a model to estimate annual regional GHG emission of Miscanthus production in the infertile Loess Plateau of China. The results showed that the N2O emission potential was 0.27 kg N ha?1 year?1 and clearly lower than that of croplands and grasslands. The CH4 uptake potential was 1.06 kg C ha?1 year?1 and was slightly higher than that of croplands. Integrated with our previous study on the emission of CO2, the net greenhouse effect of three major GHGs (N2O, CH4 and CO2) from Miscanthus fields was 4.08 t CO2eq ha?1 year?1 in the Loess Plateau, which was lower than that of croplands, grasslands and shrub lands.

Conclusions

Our study revealed that Miscanthus production may hold a great potential for GHG mitigation in the vast infertile land in the Loess Plateau of China and could contribute to the sustainable energy utilization and have positive environmental impact on the region.

A magnetophoretic harvesting agent, a polypyrrole/Fe3O4 magnetic nanocomposite, is proposed as a cost and energy efficient alternative to recover biomass of the microalgae Botryococcus braunii, Chlorella protothecoides, and Chlorella vulgaris from their culture media. The maximal recovery efficiency reached almost 99 % for B. braunii, 92.4 % for C. protothecoides, and 90.8 % for C. vulgaris. The maximum adsorption capacity (Q 0) of the magnetic nanocomposite for B. braunii (63.49 mg dry biomass mg?1 PPy/Fe3O4) was higher than that for C. protothecoides (43.91 mg dry biomass mg?1 PPy/Fe3O4) and C. vulgaris (39.98 mg dry biomass mg?1 PPy/Fe3O4). The highest harvesting efficiency for all the studied microalgae were at pH 10.0, and measurement of zeta-potential confirmed that the flocculation was induced by charge neutralization. This study showed that polypyrrole/Fe3O4 can be a promising flocculant due to its high efficacy, low dose requirements, short settling time, its integrity with cells, and with great potential for saving energy because of its recyclability.  相似文献

To investigate the effects of atmospheric CO2 enrichment on physiology and autumnal leaf phenology, we exposed 3-year-old sugar maple (Acer saccharum Marsh.) seedlings to 800 (A8), 600 (A6), and 400 μL(CO2) L–1 (AA) in nine continuous stirred tank reactor (CSTR) chambers during the growing season of 2014. Leaf abscission timing, abscised leaf area percentages, leaf number, light-saturated net photosynthetic rate (PNmax), leaf area, accumulative growth rates, and biomass were determined and assessed. The results suggested the following: (1) no significant differences were found in the timing of leaf abscission in the three CO2-concentration treatments; (2) PNmax was continuously stimulated to the greatest extent in A8 at 319% and 160% in A6 until the end of the growing season, respectively; and (3) leaf number, leaf area, and accumulative height growth all significantly increased by elevated CO2, which led to a 323% increase in A8 biomass and 235% in A6 biomass after 156-d fumigation. In summary, the results suggest, the timing of leaf abscission of sugar maple in fall was not modified by CO2 enrichment, the increased carbon gain by elevated CO2 was mainly due to increased leaf area, more leaves, and the continuously enhanced high photosynthesis throughout the growing season instead of the leaf life span.  相似文献

Soil salinity is a major abiotic stress that affects global agricultural productivity. Exploring the mechanisms that halophytes employ to thrive and flourish under saline environments is essential to increase the salt tolerance in sensitive crop species. Of the three halophytes used in this study Salicornia brachiata and Suaeda maritima belong to the same family Chenopodiaceae, while Sesuvium portulacastrum, a mangrove-associated halophyte, belongs to the family Aizoaceae. Assuming that halophytes of same family share similar salt tolerance mechanisms, we generated a suppression subtractive hybridization (SSH1) cDNA library from salt-treated leaf tissues of S. brachiata as tester and that of S. maritima as driver to identify salt-responsive genes unique to S. brachiata. To elucidate the difference in salt-tolerance mechanisms, and to identify salt-tolerance mechanisms amongst different families of halophytes, SSH2 library was generated from salt-treated leaf tissue of S. brachiata as tester and that of S. portulacastrum as driver. Totally, 87 and 49 EST clones representing unique genes were obtained from SSH1 and SSH2 libraries, respectively. Examination of the expression patterns of 17 (SSH1) and 15 (SSH2) differentially expressed genes using semi-quantitative RT-PCR confirmed up-regulation of these genes in shoots in response to salt treatment and elevated CO2 condition, but to a different extent. This study has provided insights into the molecular responses of S. brachiata to salt stress and elevated CO2 conditions.  相似文献

The behavior of Myrothecium verrucaria, artificially inoculated on spinach, was studied under seven different temperature conditions (from 5 to 35 °C) and under eight different combinations of temperature and CO2 concentration (14–30 °C and 775–870 or 1550–1650 mg/m3). The isolate used for this study was growing well on spinach, and the mycotoxins verrucarin A and roridin E were produced under all tested temperature and CO2 conditions. The maximum levels of verrucarin A (18.59 ng/g) and roridin E (49.62 ng/g) were found at a temperature of 26–30 °C and a CO2 level of 1550–1650 mg/m3. Rises in temperature as well as in temperature and CO2 concentrations had a significant effect by increasing Myrothecium leaf spots on spinach. The biosynthesis of verrucarin A was significantly increased at the highest temperature (35 °C), while roridin E was influenced by the CO2 concentration. These results show that a positive correlation between climate condition and macrocyclic trichothecene production is possible. However, because of the ability of M. verrucaria to produce mycotoxins, an increase in temperature could induce the spread of M. verrucaria in temperate regions; this pathogen may gain importance in the future.  相似文献

As one of the promising species of microalgae for biofuel production, Chlorella vulgaris CS-42 was cultivated phototrophically in two cylindrical photobioreactors with aeration of 5 % (v/v) CO2 or air for 13 days to evaluate the effects of CO2 supplementation on biomass, CO2 fixation performance, and biochemical content. Significant increases of specific growth rate and total carbon content in biomass resulting in a higher CO2 fixation rate were found with 5 % CO2. The maximum biomass concentration, carbohydrate and fatty acid contents with 5 % CO2 were significantly higher than those with air, while carbohydrate biosynthesis was most affected as compared to other biochemical components. Cytomic analysis revealed a rapid accumulation of neutral lipid in the late growth phase with more lipid bodies visualized by confocal laser scanning microscopy (CLSM), when nitrate consumption was accelerated with CO2 supplementation. Gas chromatography mass spectrometry (GC-MS) analysis indicated that 5 % CO2 favored the formation of C18:2, which led to a decrease in the degree of lipid unsaturation (DLU). These results proved that CO2 supplementation was one of the most efficient methods to significantly prompt the growth of microalgae and increase the C/N ratio in the medium, which in turn regulated the carbon metabolic flux to enhance neutral lipid and fatty acid production in C. vulgaris.  相似文献

Sunflower, the fifth largest oilseed crop in the world, plays an important role in human diets. Recently, sunflower production in North America has suffered serious yield losses from newly evolved races of sunflower rust (Puccinia helianthi Schwein.). The rust resistance gene, designated R 14 , in a germplasm line PH 3 originated from a wild Helianthus annuus L. population resistant to 11 rust races. PH 3 has seedling with an extraordinary purple hypocotyl color. The objectives of this study were to map both the R 14 rust resistance gene and the purple hypocotyl gene-designated PHC in PH 3, and to identify molecular markers for marker-assisted breeding for sunflower rust resistance. A set of 517 mapped SSR/InDel and four SNP markers was used to detect polymorphisms between the parents. Fourteen markers covering a genetic distance of 17.0 cM on linkage group (LG) 11 were linked to R 14 . R 14 was mapped to the middle of the LG, with a dominant SNP marker NSA_000064 as the closest marker at a distance of 0.7 cM, and another codominant marker ORS542 linked at 3.5 cM proximally. One dominant marker ZVG53 was linked on the distal side at 6.9 cM. The PHC gene was also linked to R 14 with a distance of 6.2 cM. Chi-squared analysis of the segregation ratios of R 14 , PHC, and ten linked markers indicated a deviation from an expected 1:2:1 or 3:1 ratio. The closely linked molecular or morphological markers could facilitate sunflower rust-resistant breeding and accelerate the development of rust-resistant hybrids.  相似文献

We studied growth and photosynthesis of cucumber (Cucumis sativus) seedlings under two vapor-pressure deficit levels (VPD; 0.4 and 3.0 kPa), two salinity levels (0 mM and 34 mM NaCl), and two CO2 concentrations ([CO2]; 400 and 1,000 μmol mol–1). Relative growth rate (RGR) decreased with increasing VPD, but the causal factor differed between salinity levels and CO2 concentrations. Under ambient [CO2], RGR decreased with increasing VPD at low salinity mainly due to decreased leaf area ratio (LAR), and decreased net assimilation rate (NAR) at high salinity. The decrease in intercellular [CO2] (Ci) with decreasing stomatal conductance caused by high VPD did not significantly limit net photosynthetic rate (PN) at low salinity, but PN was potentially limited by Ci at high salinity. At high [CO2], high VPD reduced LAR, but did not affect NAR. This is because the decrease in Ci occurred where slope of PNCi curve was almost flat.  相似文献
