Effects of the herbicide linuron on photosynthesis of the freshwater macrophytes Elodea nuttallii (Planchon) St. John, Myriophyllum spicatum L., Potamogeton crispus L., Ranunculus circinatus Sibth., Ceratophyllum demersum L. and Chara globularis (Thuill.), and of the alga Scenedesmus acutus Meyen, were assessed by measuring the efficiency of photosystem II electron flow using chlorophyll fluorescence. In a series of single-species laboratory tests several plant species were exposed to linuron at concentrations ranging from 0 to 1000 μg l−1. It was found that the primary effect of linuron, inhibition of photosystem II electron flow, occurred with a half-lifetime of about 0.1 to 1.9 h after addition of linuron to the growth medium. The direct effect of the herbicide on photosynthesis appeared to be reversible. Complete recovery from the inhibition occurred with a half-lifetime of 0.5 to 1.8 h after transfer of linuron treated plants to linuron free medium. The EC50,24h of the inhibition of photosystem II electron transport by linuron was about 9–13 μg l−1 for most of the macrophytes tested. For S. acutus the EC50,72h for inhibition of photosystem II electron flow was about 17 μg l−1 for the free suspension, and 22 μg l−1 for cells encapsulated in alginate beads. In a long-term indoor microcosm experiment, the photosystem II electron flow of the macrophytes E. nuttallii, C. demersum and the alga Spirogyra sp. was determined during 4 weeks of chronic exposure to linuron. The EC50,4weeks for the long-term exposure was 8.3, 8.7 and 25.1 μg l−1 for E. nuttallii, C. demersum and Spirogyra, respectively. These results are very similar to those calculated for the acute effects. The relative biomass increase of E. nuttallii in the microcosms was determined during 3 weeks of chronic exposure and was related to the efficiency of photosystem II electron transport as assessed in the different treatments. It is concluded that effects of the photosynthesis inhibiting herbicide on aquatic macrophytes, algae and algae encapsulated in alginate beads can be conveniently evaluated by measuring photosystem II electron transport by means of chlorophyll fluorescence. The primary and secondary effects of methabenzthiazuron on plankton were studied in outdoor microcosms (6 m3) for five months. Macrophytes, phytoplankton, zooplankton and the emergence of insects were examined. The herbicide was added to nine of the twelve mesocosms in concentrations ranging from 10 to 3371 μg a.i. l−1 in a single application. The other three ponds were used as untreated controls. Phytoplankton population dynamics fluctuated depending on light intensity, temperature, nutrients, competition and herbicide concentrations. Primary production was temporarily inhibited at test concentrations from 89 to 3371 μg l−1. Cryptophyta (Phytoflagellata) showed also a recovery at the higher concentrations during the study period. Other primary producers did not recover at these concentrations, as expected from the disappearance curve of the active ingredient at these concentrations. This was also indicated by structural indices such as the diversity, eveness and similarity index. The lower concentrations evoked no or only short-term, weak responses. Chemical water parameters were strongly influenced by the absence of macrophytes (Elodea canadensis) at the high application rates, which also caused significant changes in zooplankton communities. No effects were observed on macro-invertebrates and benthic organisms. In conclusion, noticeable effects were detected only at concentrations clearly above the effect threshold known from standard laboratory tests with aquatic primary producers. A micropropagation protocol was developed for the conservation of the critically endangered Western Australian shrub,Symonanthus bancroftii. It was necessary to screen antioxidant treatments to prevent the occurrence of lethal browning of explants upon excision. Potassium citrate and citric acid (0.1% w/v in a 4:1 ratio) prevented oxidative browning and was superior to the untreated control or other antioxidant treatments tested. Half strength Murashige and Skoog (MS) medium containing 0.5 μM kinetin and 0.25 μM benzyladenine produced three-fold multiplication compared to 1.75×, 1.5×, 1.8× and 1× multiplication for 2.5 μM kinetin + 0.25 μM benzyladenine, 0.5 μM kinetin + 5 μM gibberellic acid, 1 μM kinetin + 3 μM gibberellic acid and half strength MS with no plant growth regulators, over 4 weeks. Root production was achieved with indole-3-butyric acid (IBA) and α-naphthaleneacetic acid (NAA) at 0.5/0.5 μM (31% rooting) and 1.0/1.0 μM (36% rooting), after four weeks. Paclobutrazol (PBZ) at 0, 3.4 (1 mg 1−1), 10.2 (3 mg 1−1), or 17 μM (5 mg 1−1) improved tolerance to desiccation after transfer ofin vitro rooted shoots to soil. PBZ at 10.2 μM increased survival to 90% compared to 50% for those plantlets not treated with PBZ. The acclimatisation period from the glasshouse to the shadehouse was 1 week for plantlets treated with PBZ compared to 4 weeks for plantlets without any PBZ. The abundance and composition of phytoplankton were investigated at six stations along a transect from the Barguzin River inflow to the central basin of Lake Baikal in August 2002 to clarify the effect of the river inflow on the phytoplankton community in the lake. The water temperature in the epilimnion was high near the shore at Station 1 (17.3°C), probably due to the higher temperature of the river water, and gradually decreased offshore at Station 6 (14.5°C). Thermal stratification developed at Stations 2–6, and a thermocline was observed at a 17–22-m depth at Stations 2–4 and an 8–12-m depth at Stations 5 and 6. The concentrations of nitrogen and phosphorus nutrients in the epilimnion at all stations were <1.0 μmol N l−1 and <0.16 μmol P l−1, respectively. Relatively high concentrations of nutrients (0.56–7.38 μmol N l−1 and 0.03–0.28 μmol P l−1) were detected in the deeper parts of the euphotic zone. Silicate was not exhausted at all stations (>20 μmol Si l−1). The chlorophyll a (chl. a) concentration was high (>10 μg l−1) near the shore at Station 1 and low (<3 μg l−1) at five other stations. The <2 μm fraction of chl. a in Stations 2–6 ranged between 0.80 and 1.85 μg l−1, and its contribution to total chl. a was high (>60%). In this fraction, picocyanobacteria were abundant at all stations and ranged between 5 × 104 and 5 × 105 cells ml−1. In contrast, chl. a in the >2 μm fraction varied significantly (0.14–11.17 μg l−1), and the highest value was observed at Station 1. In this fraction, the dominant phytoplankton was Aulacoseira and centric diatoms at Station 1 and Cryptomonas, Ankistrodesmus, Asterionella, and Nitzschia at Stations 2–6. The present study demonstrated the dominance of picophytoplankton in the pelagic zone, while higher abundance of phytoplankton dominated by diatoms was observed in the shallower littoral zone. These larger phytoplankters in the littoral zone probably depend on nutrients from the Barguzin River.  相似文献

The influence of brackish phytoplankton cell classes upon the response of urea decomposition was investigated in Lake Nakaumi. The urea decomposition rate was 5 to 350 μmol urea m−3 h−1 in the light and 3 to 137 μmol urea m−3 h−1 in the dark. The urea decomposition rates in the light were obviously higher than in the dark. An extremely high rate (350 μmol urea m−3 h−1) was observed in Yonago Bay. The rate in the smaller fraction (<5 μm) exceeded that in the middle (5–25 μm) and larger fractions (>25 μm). The chlorophyll- and photosynthesis-specific rates for urea decomposition in the light were 0.5 to 3.9 μmol urea mg chl.a −1 h−1 and 0.3 to 1.3 μmol urea mg photo.C−1. The specific urea decomposing activities were higher in the smaller fraction than in the other two fractions. The present results suggest that in brackish waters urea decomposition occurred with coupling to the standing crop and photosynthetic activity of phytoplankton. Two series of experiments were conducted to determine suitable growth factors for the mass propagation of the local algal isolate Amphora sp. A higher growth rate of 0.2 doubling (μ) day−1 was attained at a lower photosynthetic photon flux density (PPFD; 11.4 μmol photon m−2s−1) compared to cultures exposed to higher levels of PPFD (16.1 μmol photon m−2s−1, −0.1 μ day −1; 31.3 μmol photon m−2s−1, 0.0 μ day−1). Cultures located inside the laboratory had a significantly higher cell density (133 × 104 cells cm−2) and growth rate (0.3 μ day−1) compared to those located outdoors (100 × 104 cells cm−2, 0.2 μ day−1). A comparison of nutrient medium across two locations showed that lipid content was significantly higher in cultures enriched with F/2MTM (macronutrients + trace metals) and F/2MV (macronutrients + vitamins). Saturated fatty acids were also present in high concentrations in cultures enriched with F/2M (macronutrients only). Significantly higher amounts of saturated fatty acids were observed in cultures located outdoors (33.1%) compared to those located indoors (26.6%). The protein, carbohydrates and n-6 fatty acid content of Amphora sp. were influenced by the location and enrichment of the cultures. This study has identified growth conditions for mass culture of Amphora sp. and determined biochemical composition under those culture conditions. Seasonal variability of inorganic and organic nitrogen in the North Sea   总被引:1,自引:1,他引:0  
This study considers the cycling of nitrogen in the waters of the North Sea, particularly focussing on organic nitrogen. Dissolved inorganic nitrogen (DIN), dissolved organic nitrogen (DON) and particulate organic nitrogen (PON) were measured in the North Sea over a one-year period (autumn 2004–summer 2005). The surface water concentrations of nitrate, ammonium, DON and PON during the present study ranged from <0.1–7.2 μM, <0.1–2.0 μM, 1.9–11.2 μM and 0.3–5.6 μM, respectively, with DON the dominant fraction of total nitrogen at all times. These nutrients concentrations were significantly lower compared to previous studies in the southern North Sea. The seasonal variations showed high mean surface concentrations of nitrate (4.7 ± 0.6 μM) and DON (8.9 ± 0.9 μM), low ammonium (<0.1 μM) and PON (0.8 ± 0.1 μM) in winter, shifting to low nitrate (0.3 ± 0.3 μM) and DON (4.2 ± 1.2 μM) in summer, with high ammonium (0.8 ± 0.4 μM) in autumn and PON (2.5 ± 1.2 μM) in spring. Highest mean surface DON concentration was measured in winter and may be due to resuspension of the organic matter from the bottom sediments. For autumn and spring, phytoplankton DON release was likely to be the most significant source of DON as shown by high concentrations of low molecular weight (LMW) DON and its positive correlation to chlorophyll a. Low total and LMW DON concentrations during summer were likely to be due to the uptake of the LMW DON fraction by phytoplankton and bacteria and the stratification of the water column. DON is therefore shown to be a potentially important source of nitrogen in shelf seas especially after the spring bloom has depleted nitrate to limiting concentrations. Handling editor: L. Abundance and biomass of pico- (<2 μm) and nanoplankton (2–20 μm) were investigated in relation to hydrography in Kongsfjorden, Svalbard (79°N, 12°E) during late summer 2006. Autotrophic and heterotrophic picoplankton abundance ranged from 0.1 × 106 to 35.2 × 106 cells L−1 and from 0.4 × 106 to 20.3 × 106 cells L−1, respectively. The highest number of bacteria in the entire water column was recorded at station 2 at 10 m (22.3 × 108 cells L−1); the lowest concentration was observed at station 1 (6.0 × 108 cells L−1). The abundance of autotrophic and heterotrophic nanoplankton varied from 0.4 × 105 cells L−1 to 46 × 105 cells L−1 and from 0.3 × 106 to 9.1 × 106 cells L−1, respectively. Our results demonstrated that heterotrophic nanoflagellates and bacteria in Kongsfjorden microbial community were relatively important. The structure of plankton communities integrated with environmental variables could act as indicators of the variability of the inflow of Atlantic Water into Kongsfjorden.  相似文献

Variations of phytoplankton assemblages were studied in November–December 2001, in surface waters of the Southern Ocean along a transect between the Sub-Antarctic Zone (SAZ) and the Seasonal Ice Zone (SIZ; 46.9°–64.9°S; 142°–143°E; CLIVAR-SR3 cruise). Two regions had characteristic but different phytoplankton assemblages. Nanoflagellates(<20 μm) and pico-plankton (∼2 μm) occurred in similar concentrations along the transect, but were dominant in the SAZ, Sub-Antarctic Front (SAF), Polar Front Zone (PFZ) and the Inter-Polar Front Zone (IPFZ), (46.9°–56.9°S). Along the entire transect their average cell numbers in the upper 70 m of water column, varied from 3 × 105 to 1.1 × 106 cells l−1. Larger cells (>20 μm), diatoms and dinoflagellates, were more abundant in the Antarctic Zone-South (AZ-S) and the SIZ, (60.9°–64.9°S). In AZ-S and SIZ diatoms ranged between 2.7 × 105 and 1.2 × 106 cells l−1, dinoflagellates from 3.1 × 104 to 1.02 × 105 cells l−1. A diatom bloom was in progress in the AZ-S showing a peak of 1.8 × 106 cells l−1. Diatoms were dominated by Pseudo-nitzschia spp., Fragilariopsis spp., and Chaetoceros spp. Pseudo-nitzschia spp. outnumbered other diatoms in the AZ-S. Fragilaropsis spp. were most numerous in the SIZ. Dinoflagellates contained autotrophs (e.g. Prorocentrum) and heterotrophs (Gyrodinium/Gymnodinium, Protoperidinium). Diatoms and dinoflagellates contributed most to the cellular carbon: 11–25 and 17–124 μg C l−1, respectively. Small cells dominated in the northern region characterized by the lowest N-uptake and new production of the transect. Larger diatom cells were prevalent in the southern area with higher values of N-uptake and new production. Diatom and nanoflagellate cellular carbon contents were highly correlated with one another, with primary production, and productivity related parameters. They contributed up to 75% to the total autotrophic C biomass. Diatom carbon content was significantly correlated to nitrate uptake and particle export, but not to ammonium uptake, while flagellate carbon was well correlated to ammonium uptake, but not to export. Diatoms have contributed highly to particle export along the latitudinal transect, while flagellates played a minor role in the export.  相似文献

Castor bean (Ricinus communis L.) plants were grown for 5–7 weeks in a controlled environment at 350 μl l−1 or 700 μl l−1 CO2. Carbon assimilation, assimilate deposition, dark respiration and assimilate mobilization were measured in leaves 2, 3 and 4 (counted from the base of the plant), and a balance sheet of carbon input and export was elaborated for both CO2 concentrations. Carbon dioxide assimilation was nearly constant over the illumination period, with only a slight depression occurring at the end of the day in mature source leaves, not in young source leaves. Assimilation was ca. 40% higher at 700 μl l−1 than at 350 μl l−1 CO2. The source leaves increased steadily in weight per unit area during the first 3 weeks, more at 700 μl l−1 than at 350 μl l−1 CO2. On top of an irreversible weight increase, there was a large gain in dry weight during the day, which was reversed during the night. This reversible weight gain was constant over the life time of the leaf and ca. 80% higher at 700 μl l−1 than at 350 μl l−1. Most of it was due to carbohydrates. The carbon content (as a percentage) was not altered by the CO2 treatment. Respiration was 25% higher in high-CO2 plants when based on leaf area, but the same when based on dry weight. The rate of carbon export via the phloem was the same during the daytime in plants grown at 350 μl l−1 and 700 μl l−1 CO2. During the night the low-CO2 plants had only 50% of the daytime export rate, in contrast to the high-CO2 plants which maintained the high export rate. It was concluded that the phloem loading system is saturated during the daytime in both CO2 regimes, whereas during the night the assimilate supply is reduced in plants in the normal CO2 concentration. Two-thirds of the carbon exported from the leaves was permanently incorporated as plant dry matter in the residual plant parts. This “assimilation efficiency” was the same for both CO2 regimes. It is speculated that under 350 μl l−1 CO2 the growing Ricinus plant operates at sink limitation during the day and at source limitation during the night. Microzooplankton grazing was investigated in surface waters of the Indo-Pacific and Atlantic sectors of the Southern Ocean by the dilution method. Phytoplankton growth varied mainly between 0.1 and 0.4 day−1, and microzooplankton grazing between 0.0 and 0.3 day−1. Great fluctuations in phytoplankton growth rate were observed at one station within 3 weeks and between closely spaced stations. Microzooplankton grazing rates were similar to phytoplankton growth rate despite the variation of phytoplankton growth rates, although in some cases, phytoplankton growth overwhelmed microzooplankton grazing. These observations suggest that microzooplankton are the main consumers of primary producers, and that steady state between phytoplankton growth and microzooplankton grazing is usually established in the Southern Ocean in austral summer. Shallow lakes often alternate between two possible states: one clear with submerged macrophytes, and another one turbid, dominated by phytoplankton. A third type of shallow lakes, the inorganic turbid, result from high contents of suspended inorganic material, and is characterized by low phytoplankton biomass and macrophytes absence. In our survey, the structure and photosynthetic properties (based on 14C method) of phytoplankton were related to environmental conditions in these three types of lakes in the Pampa Plain. The underwater light climate was characterized. Clear-vegetated lakes were more transparent (K d 4.5–7.7 m−1), had high DOC concentrations (>45 mg l−1), low phytoplankton Chl a (1.6–2.7 μg l−1) dominated by nanoflagellates. Phytoplankton productivity and photosynthetic efficiency (α ~ 0.03 mgC mgChla −1 h−1 W−1 m2) were relatively low. Inorganic-turbid lakes showed highest K d values (59.8–61.4 m−1), lowest phytoplankton densities (dominated by Bacillariophyta), and Chl a ranged from 14.6 to 18.3 μg l−1. Phytoplankton-turbid lakes showed, in general, high K d (4.9–58.5 m−1) due to their high phytoplankton abundances. These lakes exhibited the highest Chl a values (14.2–125.7 μg l−1), and the highest productivities and efficiencies (maximum 0.56 mgC mgChla −1 h−1 W−1 m2). Autotrophic picoplankton abundance, dominated by ficocianine-rich picocyanobacteria, differed among the shallow lakes independently of their type (0.086 × 105–41.7 × 105 cells ml−1). This article provides a complete characterization of phytoplankton structure (all size fractions), and primary production of the three types of lakes from the Pampa Plain, one of the richest areas in shallow lakes from South America. During January 1989, phytoplankton biomass and species composition were studied in a north / south transect at the Weddell / Scotia Confluence (47°W), between 57° and 61°30′S. Results showed a diatom bloom in the Scotia Sea (chlorophyll a 1.9 μg l−1, particulate organic carbon 239 μg l−1), dominated by Fragilariopsis cylindrus, Dactyliosolen antarcticus and Chaetoceros dichaeta. Low chlorophyll a / phaeopigments ratios (about 1.4) and silicate concentrations (15 μmol l−1) suggested that this was an advanced bloom phase, probably linked to high grazing pressure. Minimum chlorophyll a values of 0.1–0.2 μg l−1 and particulate organic carbon 46 μg l−1 were found at the Weddell / Scotia Front and in a subsurface layer of the Weddell Sea Water. In the southern part of the transect (61°30′S), in the Weddell Sea, a second surface maximum was found (chlorophyll a 0.9 μg l−1, particulate organic carbon 120 μg l−1), but with a different species composition, with Cryptomonas sp. dominant. Our results show a succession within the diatom community in the Weddell / Scotia Confluence Waters when comparing the three EPOS legs. In the Weddell Sea from spring to summer, nanoflagellates, with only a minor contribution from diatoms, persist over a long period with little change in the community structure. We suggest that the frontal system, together with the receding ice edge and the grazing pressure of either krill or protozooplankton, are mainly responsible for the phytoplankton distribution patterns found. The composition and ecological role of ciliates and dinoflagellates were investigated at one station in Kongsfjorden, Svalbard, during six consecutive field campaigns between March and December 2006. Total ciliate and dinoflagellate abundance mirrored the seasonal progression of phytoplankton, peaking with 5.8 × 104 cells l−1 in April at an average chlorophyll a concentration of 10 μg l−1. Dinoflagellates were more abundant than ciliates, dominated by small athecates. Among ciliates, aloricate oligotrichs dominated the assemblage. A large fraction (>60%) of ciliates and dinoflagellates contained chloroplasts in spring and summer. The biomass of the purely heterotrophic fraction of the ciliate and dinoflagellate community (protozooplankton) was with 14 μg C l−1 highest in conjunction with the phytoplankton spring bloom in April. Growth experiments revealed similar specific growth rates for heterotrophic ciliates and dinoflagellates (<0–0.8 d−1). Food availability may have controlled the protozooplankton assemblage in winter, while copepods may have exerted a strong control during the post-bloom period. Calculations of the potential grazing rates of the protozooplankton indicated its ability to control or heavily impact the phytoplankton stocks at most times. The results show that ciliates and dinoflagellates were an important component of the pelagic food web in Kongsfjorden and need to be taken into account when discussing the fate of phytoplankton and biogeochemical cycling in Arctic marine ecosystems.  相似文献

Endogeneous levels of zinc and copper were found to be 1.2±0.1×10−2 and 0.3±0.1×10−2 μg/A260 unit, respectively, in polysomal fractions from control animals; cadmium, however, was undetectable. In experimental animals (injected with cadmium) zinc, copper, and cadmium were found in polysomal fractions isolated by two different methods. One hour after a cadmium injection there was a rise in both the zinc and copper content of the polysomal fractions, which then declined steadily to below control levels by 16 h. Neither zinc nor cadmium were dialyzable from these fractions by a TRIS buffer; however, addition of 0.01M EDTA to the buffer resulted in removal of 75% of the zinc and all of the detectable cadmium. The addition of cadmium (CdCl2) to control supernatants (adjusted to the cadmium concentration present in supernatants 6 h after in vivo exposure) resulted in metal binding to polysomal fractions in levels comparable to those observed after in vivo exposures to the metal. When cadmium was added in the form of cadmium thionein, a smaller fraction of the metal was isolated with the polysomal fraction. Cadmium bound to polysomal fractions in vivo (24 h after exposure) was sensitive to release by protease digestion, but insensitive to release by ribonuclease digestion.  相似文献

Summary Three laboratory-scale water pipe systems were set up to study the effects of adding two levels of acetic acid (10 and 50 μg acetate eq-C l−1) on the bacterial regrowth in water pipes. The results of the water pipe test showed that nearly all carbon in the acetic acid could be readily utilized by bacteria and resulted in an increase in biomass concentration. The maximum heterotrophic plate counts in biofilm were equal to 3.5 × 104, 8.9 × 105 and 2.9 × 107 c.f.u. cm−2 while the maximum heterotrophic plate counts of free bacteria were equal to 1.2 × 103, 5.0 × 103 and 6.8 × 104 c.f.u. ml−1 for the blank and with addition of 10 and 50 μg acetate eq-C l−1. These results showed that addition of acetic acid to drinking water has a positive effect on the assimilable organic carbon content of drinking water and bacterial regrowth in the distribution system. This effect is enhanced with addition of high-level acetic acid. Batch tests were also conducted using water samples collected from a Taiwanese drinking water distribution system. The bacterial regrowth potentials of the blank were equal to 4.3 × 103, 1.5 × 104, 4.9 × 104 and 7.5 × 104 c.f.u. ml−1 for water samples collected from treatment plant effluent, commercial area, mixed area, and residential area, respectively. These results showed that the biological stability of drinking water is the highest in treatment plant effluent, followed by distributed water of the commercial area, distributed water of the mixed area, and then the distributed water of residential area.  相似文献

Production rates, abundance, chlorophyll a (Chl a) concentrations and pigment composition were measured for three size classes (<2 μm, 2–11 μm and >11 μm) of phytoplankton from May to December 2000 in deep, mesotrophic, alpine lake Mondsee in Austria. The study focuses on differences among phytoplankton size fractions characterised by their surface area to volume ratio ([mml−1: mm3l−1]), pigment distribution patterns and photosynthetic rates. Particular attention was paid to autotrophic picophytoplankton (APP, fraction <2 μm) since this size fraction differed significantly from the two larger size fractions. Among the three fractions, APP showed the highest surface area to volume ratios and a high persistence in the pattern of lipophilic pigments between temporarily and spatially successive samples (about 80% similarity of pigment composition between samples over seasons and depths). The epilimnetic abundance of APP varied seasonally with an annual maximum of 180 × 10cells ml−1 in June (at 4–9 m). The minimum (October at 12 m) was more than an order of magnitude lower (4.9 × 103 ml−1). APP peaked during autumn and contributed between 24% and 42% to the total area-integrated Chl a (10–23 mg m−2) and between 16% and 58% to total area-integrated production (5–64 mg m−2  h−1) throughout seasons.  相似文献

Regulation by irradiance level of the mechanism for dissolved inorganic carbon (DIC) acquisition was examined in the red macroalga Gracilaria tenuistipitata Zhang et Xia. For this purpose, affinity for external DIC, carbonic anhydrase (CA; EC 4.2.1.1) activity and content of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco; EC 4.1.1.39) were determined in thalli grown at 45 and 500 μmol photons m−2 s−1. Oxygen evolution rates declined by 50% when the medium pH was changed from 8.1 to 8.7, and the pH compensation point attained was ca. 9.2. These characteristics were unaffected by the light treatments. In contrast, photosynthetic conductance for DIC at pH 8.7 was doubled in thalli grown at high irradiance compared with those grown at low irradiance (to 0.74 × 10−6 from 0.33 × 10−6 m s−1). Photosynthetic rates at saturating DIC concentration were also higher by 60% in thalli grown at high irradiance. These differences could not be attributed to changes in the use of external DIC, since external CA activity did not vary. Although the irradiance level did not modify the pool size of Rubisco, Rubisco content expressed on a chlorophyll a basis was almost doubled at high irradiance. These results likely indicate that the internal transport of DIC towards the active-site of Rubisco, rather than the external use of DIC, is enhanced in the thalli grown at high irradiance. Picoeukaryotes dominate the phytoplankton of Lake Balaton—the largest shallow lake in Central Europe—in the winter period. We examined the annual dynamics of picoplankton abundance and composition in the lake in order to establish if the picoeukaryotes merely survive the harsher winter conditions or they are able to grow in the ice-covered lake when the entire phytoplankton is limited by low light and temperature. Lake Balaton has an annual temperature range of 1–29°C, and it is usually frozen between December and February for 30–60 days. In the spring-autumn period phycocyanin and phycoerythrin rich Cyanobacteria are the dominant picoplankters, and picoeukaryotes are negligible. Our five-year study shows the presence of three types of picophytoplankton assemblages in Lake Balaton: (1) Phycoerythrin-rich Cyanobacteria—the dominant summer picoplankters in the mesotrophic lake area; (2) Phycocyanin-rich Cyanobacteria—the most abundant summer picoplankters in the eutrophic lake area and; (3) Picoeukaryotes—the dominant winter picoplankters in the whole lake. The observed winter abundance of picoeukaryotes was high (up to 3 × 105 cells ml−1), their highest biomass (520 μg l−1) exceeded the maximum summer biomass of picocyanobacteria (500 μg l−1). Our results indicate that the winter predominance of picoeukaryotes is a regular phenomenon in Lake Balaton, irrespective of the absence or presence of the ice cover. Picoeukaryotes are able to grow at as low as 1–2°C water temperature, while the total phytoplankton biomass show the lowest annual values in the winter period. In agreement with earlier findings, the contribution of picocyanobacteria to the total phytoplankton biomass in Lake Balaton is inversely related to the total phytoplankton biomass, whereas no such relationship was observable in the case of picoeukaryotes.  相似文献

Vital protoplasts from Spathiphyllum wallisii ‘Alain’ and Anthurium scherzerianum ‘238’ were isolated from both somatic embryos and leaves. The highest yields were obtained when 1.5% cellulase, 0.5% macerase and 0.5% driselase were used for Spathiphyllum wallisii leaves and 0.5% cellulase, 0.3% macerase and 0.5% driselase for Anthurium scherzerianum embryos. About 1 × 106 protoplasts g−1 and 1 × 105 protoplasts g−1 could be isolated from leaves and embryos, respectively. For protoplast fusion Spathiphyllum wallisii ‘Alain’ and Anthurium scherzerianum ‘238’ were mixed in a 1:1 ratio in a fusion solution containing 1 mM CaCl2·2H2O, 1 mM MES and 0.5 M mannitol. Fusion was performed by protoplast alignment under 500 V cm−1 alternating current for 60 s and subsequent generation of two pulses of 4500 V cm−1 direct current during 50 μs. Development until colony stage was achieved using agarose beads for protoplast culture.  相似文献
