Investigations have been made to study the production of phenolic compounds (total phenolics, flavonoids and phenylpropanoids) and total antioxidant capacity in 27 Macedonian traditional medicinal plants to improve its potential as a source of natural antioxidants. Antioxidant potential of plant extracts was analyzed by five different assays: cupric reducing antioxidant capacity (CUPRAC), phosphomolybdenum method (PM), reducing power (RP), 2,2-diphenyl-1-picrylhydrazyl (DPPH·) and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS·+) radical scavenging activity. Origanum vulgare extract consistently exhibited the highest content of phenolic compounds and the strongest antioxidant capacity based on the tests performed, and can be proposed as a promising source of natural antioxidants. Melissa officinalis and Salvia ringens were also identified as valuable sources of antioxidant compounds. A positive linear correlation between antioxidant activity and total phenolics, flavonoids and phenylpropanoids indicates that these compounds are likely to be the main antioxidants contributing to the observed activities of evaluated plants. These findings suggest that the medicinal plants studied in this paper are good sources of bioactive compounds for the food and pharmaceutical industries.  相似文献

Gnidia glauca and Dioscorea bulbifera are traditional medicinal plants that can be considered as sources of natural antioxidants. Herein we report the phytochemical analysis and free radical scavenging activity of their sequential extracts. Phenolic and flavonoid content were determined. Scavenging activity was checked against pulse radiolysis generated ABTS•+ and OH radical, in addition to DPPH, superoxide and hydroxyl radicals by biochemical methods followed by principal component analysis. G. glauca leaf extracts were rich in phenolic and flavonoid content. Ethyl acetate extract of D. bulbifera bulbs and methanol extract of G. glauca stem exhibited excellent scavenging of pulse radiolysis generated ABTS•+ radical with a second order rate constant of 2.33×106 and 1.72×106, respectively. Similarly, methanol extract of G. glauca flower and ethyl acetate extract of D. bulbifera bulb with second order rate constants of 4.48×106 and 4.46×106 were found to be potent scavengers of pulse radiolysis generated OH radical. G. glauca leaf and stem showed excellent reducing activity and free radical scavenging activity. HPTLC fingerprinting, carried out in mobile phase, chloroform: toluene: ethanol (4: 4: 1, v/v) showed presence of florescent compound at 366 nm as well as UV active compound at 254 nm. GC-TOF-MS analysis revealed the predominance of diphenyl sulfone as major compound in G. glauca. Significant levels of n-hexadecanoic acid and octadecanoic acid were also present. Diosgenin (C27H42O3) and diosgenin (3á,25R) acetate were present as major phytoconstituents in the extracts of D. bulbifera. G. glauca and D. bulbifera contain significant amounts of phytochemicals with antioxidative properties that can be exploited as a potential source for herbal remedy for oxidative stress induced diseases. These results rationalize further investigation in the potential discovery of new natural bioactive principles from these two important medicinal plants.  相似文献

Subcritical water extraction was used to extract bioactive phenolic compounds from Vaccinium dunalianum Wight leaves. The optimal extraction conditions were determined as an extraction temperature of 150 °C, an extraction time of 40 min, and a liquid-solid ratio of 35 : 1 mL/g. The total phenolic content reached 21.35 mg gallic acid /g, which was 16 % higher than that by hot water extraction. The subcritical water extraction extract exhibited strong scavenging activity of DPPH free radical and ABTS+ free radical, as well as significant tyrosinase inhibitory activity. The study suggests that subcritical water extraction can alter the composition of the extracts, leading to the production of various phenolic compounds, effective antioxidants, and tyrosinase inhibitors from Vaccinium dulciana Wight leaves. These findings confirm the potential of Vaccinium dunalianum Wight as a natural antioxidant molecule source for the medicine and food industries, and for the therapy of skin pigmentation disorders.  相似文献

Navicula incerta is a marine microalga distributed in Baja California, México, commonly used in aquaculture nutrition, and has been extended to human food, biomedical, and pharmaceutical industries due to its high biological activity. Therefore, the study aimed to optimize culture conditions to produce antioxidant pigments. A central composite experimental design and response surface methodology (RSM) was employed to analyze the best culture conditions. The medium (nitrogen-deficient concentrations), salinity (PSU = Practical Salinity Unity [g/kg]), age of culture (days), and solvent extraction (ethanol, methanol, and acetone) were the factors used for the experiment. Chlorophyll a (Chl a) and total carotenoids (T-Car), determined spectroscopically, were used as the response variables. The antioxidant capacity was evaluated by DPPH? and ABTS?+ radical inhibition, FRAP, and anti-hemolytic activity. According to the overlay plots, the optimum growth conditions for Chl a and T-Car production were the following conditions: medium = 0.44 mol·L-1 of NaNO3, salinity = 40 PSU, age of culture: 3.5 days, and solvent = methanol. The pigment extracts obtained in these optimized conditions had high antioxidant activity in ABTS?+ (86.2–92.1% of inhibition) and anti-hemolytic activity (81.8–96.7% of hemolysis inhibition). Low inhibition (33–35%) was observed in DPPH?. The highest value of FRAP (766.03 ± 16.62 μmol TE/g) was observed in the acetonic extract. The results demonstrated that RSM could obtain an extract with high antioxidant capacity with potential applications in the biomedical and pharmaceutical industry, which encourages the use of natural resources for chemoprevention of chronic-degenerative pathologies.  相似文献

Thamnidium elegans CCF 1456, a filamentous fungus, was used to enhance the total phenolic content and radical scavenging activity of maize via solid-state fermentation. Thamnidium fermented maize (TFM) and unfermented maize (UFM) grains were extracted with 65% ethanol and dimethylsulfoxide (DMSO). Total phenolic content (TPC), and radical scavenging capacity — determined with 1,1-diphenyl-2-picrylhydrazyl (DPPH) and radical cations of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS·+) found for TFM — were significantly (P < 0.05) higher on the 5th day of incubation than that of UFM extracts. A linear correlation was observed among TPC, DPPH and ABTS scavenging activities, and also among TPC and various carbohydrate-cleaving enzymes (α-amylase, β-glucosidase and xylanase), suggesting that this? strategy may help to enrich? TFM with improved phytochemical properties and antioxidant activities.  相似文献

研究了蓝莓果渣提取物总酚含量及其抗氧化活性。分别采用水、40%乙醇及纤维素酶辅助乙醇超声提取蓝莓果渣,并用Folin-Ciocalteu试剂对3种提取物的总酚含量进行评估;并采用DPPH清除实验及O2—.清除实验对3种提取物的抗氧化活性进行研究。实验结果表明,纤维素酶辅助超声提取蓝莓果渣的总酚含量最高,可达425.36±15.21 mg GAE.100 g-1DW,远远高于水提物(169.46±9.75 mg GAE.100 g-1DW)及醇提物(218.39±12.54mg GAE.100 g-1DW)中的总酚含量。且纤维素酶辅助乙醇超声提取物对DPPH的清除能力为2.67±0.13 gVc.100 g-1DW,对O—.2的清除能力2.48±0.14 g Vc.100 g-1DW,明显好于醇提物及水提物抗氧化活性。  相似文献

The current study was designed to evaluate the antioxidant, anti‐inflammatory and antimicrobial activities of Alchemilla mollis (Buser ) Rothm . (Rosaceae) aerial parts extracts. Chemical composition was analyzed by spectrophotometric and chromatographic (HPLC) techniques. The antioxidant properties assessed included DPPH· and ABTS·+ radical scavenging, β‐carotene‐linoleic acid co‐oxidation assay. Antimicrobial activity was evaluated with disc diffusion and micro dilution method. In order to evaluate toxicity of the extracts, with the sulforhodamine B colorimetric assay L929 cell line (mouse fibroblast) was used. The anti‐inflammatory activities of the potent antioxidant extracts (methanol, 70% methanol, and water extracts) were determined by measuring the inhibitory effects on NO production and pro‐inflammatory cytokine TNF‐α levels in lipopolysaccharide stimulated RAW 264.7 cells. 70% methanol and water extracts which were found to be rich in phenolic compounds (184.79 and 172.60 mg GAE/g extract) showed higher antioxidant activity. Luteolin‐7‐O‐glucoside was the main compound in the extracts. Ethyl acetate and 70% methanol extracts showed higher antibacterial activity against Staphylococcus aureus and Salmonella enteritidis with MIC value of 125 μg/ml. 70% methanol extract potentially inhibited the NO and TNF‐α production (18.43 μm and 1556.22 pg/ml, respectively, 6 h).  相似文献

Abstract

Box–Behnken Design (BBD) was used to optimize the extraction conditions of polyphenols from Malva (Malva parviflora L.) leaves. The effect of ethanol concentration (20–80%), solvent/leaf powder ratio (10:1 to 30:1, v/w) and extraction time (5–45?min) on the polyphenols yield and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the obtained extracts were investigated. Quadratic models fit well. The optimal conditions (53.40% ethanol, solvent/leaf powder ratio 20:1 (v/w), and 15?min) resulted in an extract with a maximum yield of polyphenols (1098.4?mg GAE/100?g leaf powder) and high inhibition percentage of DPPH radical (33.31%) with desirability 0.742. High Performance Liquid Chromatography (HPLC) analysis indicated that the major identified polyphenol compounds extracted at the optimal conditions were naringenin, ρ-coumaric acid, apigenin-7-glucoside, luteolin, and cinnamic acid. These findings indicate that M. parviflora leaf extracts possess DPPH radical scavenging activity and could be used as a natural source for bioactive products.  相似文献

Passiflora alata Curtis, commonly known as sweet passion fruit, is one of the commercially cultivated species of the genus Passiflora, whose fruits can be consumed in natura due to their sweet taste. It is also used worldwide as an ornamental and in folk medicine. The goal of this work was the evaluation of the antioxidant potential of extracts from in vivo plants, and in vitro-derived materials of P. alata. Leaves from in vivo plants were used for the optimization of parameters that affect the efficiency of extraction of antioxidant compounds (proportions of ethanol:water, maceration period, solvent:plant tissue ratio, and number of extraction stages), by employing the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The antioxidant activity and the extract yields were significantly influenced by the proportion of ethanol:water and maceration period. The optimized protocol was applied to obtain the extracts of in vitro-derived materials. Total phenolic content was determined using the Folin–Ciocalteu method. Higher antioxidant activities and phenolic contents were observed in extracts from leaves of in vivo-seed derived and from acclimatized plants when compared to in vitro plants, calluses and suspension cultures. Differences in the reaction kinetics of DPPH scavenging activity were also observed.  相似文献

The aim of this study is to examine phytochemical composition and evaluation of antimicrobial, antioxidant and cytotoxic activity of Anchusa officinalis plant extracts in different solutions: ethanol, chloroform, petroleum, acetone and ethyl acetate. A comparative analysis has shown that ethanol extract had the highest concentration of phenols (104.03?±?0.63 mgGA/g), and the highest concentration of flavonoids (30.26?±?0.40 mgRU/g). The highest concentration of the condensed tannins recorded in chloroform extract (74.65?±?0.57 mg GA/g). The ethanol extract showed the strongest antioxidant and the best antimicrobial activity as compared to all other tested extracts, while the chloroform and the acetone extracts showed the best cytotoxic activity on cell line of mouse fibroblast carcinoma (L2OB). This is the first report of citotoxic activity of extract A. officinalis plant from Balkan region on tumor cell lines. HPLC analysis of A. officinalis plant extracts confirmed that the predominant polyphenol components were: rosmarinic acid, chlorogenic acid, naringenin, lutein-glycoside and rutin.  相似文献

Electromagnetic induction heating (EMIH) assisted extraction of phenolic compounds from Rosmarinus officinalis L, and the antioxidant and antimicrobial activities of the plant extract were examined in this study. The extraction yield acquired with this process was found to be 25.1?±?2%, with maximum amounts of phenolic compounds: 127.87?±?2.1 mg Gallic acid equivalents per g dry weight and total flavonoids contents 14.48?±?1.5 mg quercetin equivalents per g dry weight, under optimum extraction conditions (extraction time 2 h, ratio of raw material to liquid 1:2 and 0% of NaCl). The antioxidant activity was assessed by the 1,1-diphenyl-2-picrylhydrazyl (DPPH); 2, 2′-azinobis (3-ethylbenzothiazoline-6- sulfonic acid) radical cation (ABTS+) and ferric reducing power (FRAP) methods. The results indicate the extract derived through EMIH showed a strong antioxidant ability (89.25%; EC50 of 0.0148 µg/mL). Besides, the antimicrobial bioassay demonstrated that the extract possessed a good antimicrobial activity against all tested fungi and bacteria.  相似文献

Streptomyces mediolani sp. AC37 was isolated from the root system of higher plant Taxus baccata and produced metabolite identified as (?)-8-O-methyltetrangomycin according to LC/MS/MS analysis. In our screening program for improvements of bioactive secondary metabolites from plant associate streptomycetes, mutation was used as a tool for the induction of genetic variations for selection of higher (?)-8-O-methyltetrangomycin producers of isolates. S. mediolani sp. AC37 was treated with UV irradiation and chemical mutagenic treatment (N-nitroso-N-methyl-urea). The radical scavenging and antioxidant capacity of (?)-8-O-methyltetrangomycin and extracts isolated from mutants were tested using EPR spin trapping technique and ABTS·+ assay. Comparison of electron microscopic images of Streptomyces sp. AC37 and mutant strains of Streptomyces sp. AC37 revealed substantial differences in morphology and ultrastructure.  相似文献

This work aimed to discuss the effects of exogenous abscisic acid (ABA) on the root growth regulation of maize seedlings under chilling stress. The roots of the maize cultivar Zhengdan 958 were irrigated with ABA (10?7, 10?6, 10?5 and 10?4 M) at the third true leaf stage under chilling duration (0, 2, 4, 6, and 8 days). The biomass, the phenylalanine ammonia lyase (PAL), and polyphenol oxidase (PPO) enzyme activities, total phenolic and flavonoid contents, the ferric reducing ability of plasma (FRAP) antioxidant capacity, and 2,2-azinobis (3-ethlbenzothiazo-line-6-sulfonic acid) diammonium salt radical (ABTS·+) scavenging capacity of the roots of maize seedlings were measured after the treatment. The results showed that appropriate concentrations of exogenous ABA effectively enhanced root biomass, increased PAL and PPO enzyme activities, and significantly increased total phenolic contents and flavonoid contents. Moreover, the ABA markedly improved the FRAP antioxidant capacity and ABTS·+ scavenging capacity under low-temperature stress. These results indicate that ABA-treated maize seedlings are resistant to chilling stress and that the optimum concentration of ABA is 10?5 M. Exogenous applications of ABA have a concentration effect in alleviating chilling stress, in which low concentrations have a promoting effect and high concentrations have an inhibiting effect.  相似文献

The present study was aimed at determining total phenolic and flavonoid contents and studying the antioxidant activity of ginger (Zingiber officinale Rosc.) rhizome and callus, 6-gingerol and 6-shogaol and callus treated with elicitors. Petroleum ether (PE) and chloroform: methanol (1:1, v/v) (CM) extracts were prepared by maceration. Highest total phenolic content was obtained from the CM extract (60.34?±?0.43?mg gallic acid/g) of rhizome while callus showed lower content detected in the CM extract (33.6?±?0.07?mg gallic acid/g). Flavonoids were only detected in rhizome (CM extract 40.25?±?0.21?mg quercetin/g). Both rhizome extracts exhibited good antioxidant activity with higher activity recorded in PE extract (IC50 value 8.29?±?1.73?μg/mL). Callus extracts revealed lower antioxidant activity (IC50 value 1265.49?±?59.9?μg/mL obtained from CM extract). 6-gingerol and 6-shogaol displayed high antioxidant activity in both assays with IC50 4.85?+?0.58DPPH and 5.35?±?0.33ABTS μg/mL for the former and IC50 7.61?±?0.81DPPH and IC50 7.05?±?0.23ABTS μg/mL for the latter. Treatment of callus with elicitors showed significant (p?<?0.05) effects in enhancing phenolic content and related antioxidant activity. The highest significant increase in phenolic content (37% and 34%) and antioxidant activity in DPPH assay (34% and 30%) was observed in callus treated with 100?mg/L yeast extract and 50?mg/L salicylic acid respectively. Therefore, studying the effect of the elicitation of ginger cultured tissues in phenolic accumulation would be of immense importance for pharmacological, cosmetic and agronomic industries.  相似文献

The study explored on the commonly available weed plant Commelina nudiflora which has potential in-vitro antioxidant and antimicrobial activity. The different polar solvents such as ethanol, chloroform, dichloromethane, hexane and aqueous were used for the soxhlet extraction. The extracts were identified pharmacologically as important bioactive compounds and their potential free radical scavenging activities, and antimicrobial properties were studied. C. nudiflora extracts were monitored on their in-vitro antioxidant ability by DPPH and ABTS radical scavenging assay. Aqueous extract shows significant free radical scavenging activity of 63.4 mg/GAE and 49.10 mg/g in DPPH and ABTS respectively. Furthermore, the aqueous crude extract was used in antibacterial studies, which shows the highest inhibitory activity against Pseudomonas aeruginosa, Escherichia coli and Salmonella typhi. Among all the extracts, aqueous extract of C. nudiflora has significant control over free radical scavenging activity and inhibition of the growth of food pathogenic bacteria. Also, the aqueous extract contains abundance of phenolics and flavonoids higher than other extracts. A comprehensive phytochemical study of Juniperus turbinata (Cupressaceae) collected from La Maddalena Archipelago (Sardinia, Italy) is reported. Both the essential oil and the ethanolic extract obtained from the aerial parts were analyzed. The essential oil appears to belong to a new chemotype compared to other Mediterranean juniper accessions, as it was favored by geographic isolation of the isles. It showed a low content of monoterpene hydrocarbons and α‐terpineol, ent‐manoyl oxide, 1,10‐di‐epi‐cubenol as the major constituents. The ethanolic fraction contained mainly diterpenoids. Among these, 15‐formyloxyimbricatolic acid ( 7 ) is a new natural product since it has hitherto been obtained only by synthetic route. The phenolic fraction contained biflavonoids: cupressuflavone ( 9 ), followed by minor amounts of amentoflavone ( 10 ) and hinokiflavone ( 11 ). The essential oil and six purified compounds ( 1 – 4 , 8 and 9 ) were assessed for biological activities, namely antioxidant (assessed by DPPH·, ABTS·+ and FRAP methods) and cytotoxic effects towards selected human tumor cell lines (MDA‐MB 231, A375 and HCT116 cells). Compound 3 exhibited higher radical scavenging activity against ABTS·+ radical than the reference Trolox. Noteworthy, compound 8 showed powerful effects towards tumor cell lines, with IC50 values in the range of 0.060 – 0.201 μm , which make it a promising anticancer drug candidate.  相似文献

Silymarin, a known standardized extract obtained from seeds of Silybum marianum is widely used in treatment of several diseases of varying origin. In the present paper, we clarified the antioxidant activity of silymarin by employing various in vitro antioxidant assay such as 1,1-diphenyl-2-picryl-hydrazyl free radical (DPPH·) scavenging, 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, total antioxidant activity determination by ferric thiocyanate, total reducing ability determination by Fe3+ ? Fe2+ transformation method and Cuprac assay, superoxide anion radical scavenging by riboflavin/methionine/illuminate system, hydrogen peroxide scavenging and ferrous ions (Fe2+) chelating activities. Silymarin inhibited 82.7% lipid peroxidation of linoleic acid emulsion at 30 μg/mL concentration; butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), α-tocopherol and trolox indicated inhibition of 83.3, 82.1, 68.1 and 81.3% on peroxidation of linoleic acid emulsion at the same concentration, respectively. In addition, silymarin had an effective DPPH· scavenging, ABTS√+ scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, ferric ions (Fe3+) reducing power by Fe3+ ? Fe2+ transformation, cupric ions (Cu2+) reducing ability by Cuprac method, and ferrous ions (Fe2+) chelating activities. Also, BHA, BHT, α-tocopherol and trolox, were used as the reference antioxidant and radical scavenger compounds. Moreover, this study, which clarifies antioxidant mechanism of silymarin, brings new information on the antioxidant properties of silymarin. According to the present study, silymarin had effective in vitro antioxidant and radical scavenging activity. It could be used in the pharmacological and food industry because of its antioxidant properties.  相似文献

Abstract

This study was undertaken to investigate the effect of various solvents and techniques on the extractability of antioxidant compounds, particularly phenolics, from leaves and roots of Silene vulgaris subsp. macrocarpa grown wild in Morocco. Maceration and hot extraction with methanol or water and Soxhlet ethanol extraction were utilized. Aimed at establishing the potential safety of the extracts, Artemia salina lethality bioassay was performed. All the extracts were found to be non-toxic, except for the leaf Soxhlet ethanol. The antioxidant potential of the extracts was evaluated in vitro by DPPH, reducing power, and ferrous ions chelating activity assays. The leaf extracts displayed noticeable radical scavenging and chelating activities, and maceration with methanol (Mac-MeOH) resulted the most suitable extraction method for an effective recovery of antioxidants; further, the root Mac-MeOH extract demonstrated good chelating properties (IC50 = 335.49?±?0.70?µg/mL). Thus, leaf and root Mac-MeOH extracts were subjected to phytochemical investigations. The total phenolic, flavonoid and condensed tannin content was determined spectrophotometrically. Thirteen polyphenolic compounds were positively identified, by HPLC-PDA-ESI-MS, in the leaf extract for the first time, with p-coumaric acid derivatives being the most abundant ones (81%), whereas only catechin and procyanidin B1 were found in the root extract.  相似文献

Rice stems were subjected to a subcritical fluid treatment at 230 °C, using ethanol or acetone at a dilution of 0–100% in water. The obtained extracts were determined for their yield, carbohydrate content, phenolic content, DPPH radical scavenging ability, and color. The highest yield and carbohydrate content were achieved with the subcritical 20% (v/v) organic solvent, while the highest phenolic content was obtained with subcritical 80% (v/v) acetone. The highest radical scavenging ability was achieved with subcritical 60% (v/v) ethanol and 80% (v/v) acetone. The lightness of the extracts obtained with subcritical ethanol and acetone was negatively correlation with their radical scavenging ability (R=?0.85). The relationship between the lightness and phenolic content of the extracts was not significant, suggesting that other substances in the extract could also possess radical scavenging ability.  相似文献

The work was conducted with the purpose to evaluate antioxidant activity of Parmelia saxatilis (PS) by different analytical methods. Water and methanol were used as solvents and antioxidative effects were measured by a ferric thiocyanate method (FTC) and thiobarbituric acid test (TBA). The antioxidant activity increased with the increasing amount of extracts (from 50 to 250 μg) added to linoleic acid emulsion. The methanol extract of PS exhibited high antioxidative activity that was not significantly (P < 0.05) different from α-tocopherol, while aqueous extracts of PS showed low antioxidative activity. Similar trends of antioxidant activity were observed using either the FTC or TBA methods. Antioxidant activity, reducing power, free radical scavenging (DPPH·), superoxide anion radical scavenging, metal chelating and hydrogen peroxide scavenging activities of PS extracts showed dose dependence and increased with concentration of PS extract. The results obtained in the present study indicate that the PS might be a potential source of natural antioxidant.  相似文献
