目的:研究低聚异麦芽糖对模拟失重大鼠肠道益生菌的影响以及与骨钙代谢变化的关系。方法:30只雄性SD大鼠随机分为3组(每组10只)FC组;普通对照组(饲喂普通饲料);SC组,模拟失重对照组(饲喂普通饲料);SS组,模拟失重低聚异麦芽糖组(饲喂普通饲料 低聚异麦芽糖),实验期21d。结果:SC组大鼠肠道益生菌(主要是双歧杆菌和乳杆菌)数量、饲料钙的表观吸收率、股骨骨密度(BMD),骨钙含量以及骨钙素(BGP)的水平显著低于FC组,而血钙水平明显高于FC组;SS组大鼠肠道益生菌数量,饲料钙的表观吸收率,骨密度,骨钙含量以及骨钙素水平较SC组高,血钙水平显著低于SC组。结论:低聚异麦芽糖可以促进模拟失重大鼠肠道益生菌的增殖,减少股骨骨质的丢失,提高股骨BMD,增加骨形成,对骨代谢产生一定的有益影响。  相似文献

我们以前的工作提示,在模拟失重所引起的血管区域特异性适应变化中,局部肾素.血管紧张素系统(local reninangiotensin system,L-RAS)可能发挥关键调控作用。本文以losartan慢性阻断血管紧张素Ⅱ1型受体(angiotensin Ⅱtypelreceptor,AT1R),观察模拟失重是否仍能引起血管的这种适应性改变,并检测大血管管壁L-RAS主要成分的表达是否也发生相应变化。以尾部悬吊大鼠模型模拟失重的生理影响。制作基底动脉、胫前动脉、颈总动脉和腹主动脉的HE染色切片,在光学显微镜下进行形态观测:用免疫组织化学技米测量颈总动脉和腹主动脉壁的血管紧张素原(angiotensinogen,AGT)及AT-R的表达变化。结果表明:4周模拟失重引起大鼠基底动脉中膜和颈总动脉管壁各平滑肌肌层肥厚,而胫前动脉和腹主动脉则发生萎缩性改变;给予losartan4周引起上述4种血管皆发生萎缩性变化;阻断AT1R,模拟失重仍然能引起基底动脉、颈总动脉发生相对肥厚性改变和腹主动脉萎缩加重。4周模拟失重还引起颈总动脉壁中AGT和AT1R表达上调,而腹主动脉壁及血管周围组织中AGT和AT1R表达下调;给予losartan4周仅引起腹主动脉壁中AGT和AT1R表达减少;阻断AT1R,模拟失重使腹主动脉壁AT1R表达进一步减少。结果提示,4周模拟失重引起大鼠脑、颈部与后身大、中动脉血管的形态结构改变和L-RAS主要成分表达发生上调或下调,血管L-RAS在其中可能发挥关键性调控作用;但在慢性阻断AT1R的条件下,其它调控机制仍可能在脑血管适应性调节中发挥一定作用。  相似文献

Five pregnant female growing rats have been orbited for five days aboard the Cosmos 1514 soviet biological satellite. They were compared to five female rats kept in vivarium and five female conditioned rats in synchronised way. Histomorphometric studies were performed in order to investigate: 1. The early effects of weightlessness on the bone mass in loading (tibiae and femur) and unloading bones (thoracic and lumbar vertebrae). 2. The changes of osteoblastic and osteoclastic activities. A short exposure does not induce changes in the bone mass and the inner structure of loading and unloading bones. These results fit in well with human data available in the literature: they show that weightlessness doesn't change bone mass in the early phase of a spaceflight. However extrapolation of animal results to men is discussed. In unloading bones (vertebrae) osteoblastic activity was not measurable. Osteoclasts detected by histoenzymologic method don't change as far as their number per mm3 of trabecular bone is concerned. However the number per mm2 of trabecular area increases. It seems likely that an increase of the osteoclastic population occurs in trabecular bone. In loading bones, formation activity (appreciated by the measurement of osteoid seam thickness) and total osteoclastic resorption surfaces were not modified. Exposure to microgravity (weightlessness) is known to cause rapid bone and muscle losses. We have used the hind limb-suspended (HLS) rat model to simulate microgravity-induced musculoskeletal losses in order to assess resulting hormonal changes and to develop a novel pharmacological countermeasure. Previously, we demonstrated significant decreases in circulatory hormonal levels [serum thyroxin, 1,25(OH)2 vitamin D (p<0.05), and serum testosterone (p<0.001)] in HLS rats. Both thyroxin and 1,25(OH)2 vitamin D levels returned to normal soon after removal from HLS, while testosterone levels matched normal levels only after a further 3-4 weeks. However, even by day 42, bone mineral density (BMD) remained significantly lower, although serum hormones were back to normal. Because serum testosterone levels become undetectable in HLS rats, we hypothesized that the replacement of testosterone during HLS could prevent musculoskeletal losses. Based on these data, an intervention study was carried out to assess the efficacy of testosterone and synthetic anabolic steroid, nandrolone decanoate (ND), in prevention of weightlessness-induced musculoskeletal losses. HLS rats (control) had a significant reduction of muscle volume (42.9 -/+ 3.0, versus 56 -/+ 1.8 in ground control rats; p<0.01). Both testosterone and ND treatments prevented this muscle loss (51.5 -/+ 2 cm(3) and 51.6 -/+ 1.2, respectively; a 63% improvement, p<0.05). Similarly, BMD of the placebo-treated HLS rats was significantly lower than that of ground control rats (0.416 -/+ 0.011 versus 0.354 -/+0.014, p<0.05), and testosterone and ND prevented this bone loss (0.404 -/+ 0.013 versus. 0.409 -/+ 0.011, respectively). These data suggest that both testosterone and ND therapy can minimize the musculoskeletal losses associated with exposure to simulated weightlessness. Experiments using the combination of bisphosphonate and testosterone demonstrated complete protection of both muscle and bone in these HLS rats. Therefore, considering that: 1) testosterone is anabolic to osteoblasts and muscle cells and also decreases the rate of bone turnover, 2) serum testosterone levels are markedly suppressed in simulated weightlessness, and 3) testosterone replacement therapy prevented musculoskeletal losses in HLS rats, we propose that the musculoskeletal losses observed in this animal model (i.e., simulated microgravity) are related to their testosterone deficiency. Since serum sex hormones levels are markedly reduced in this model of simulated microgravity, androgen replacement with a bisphosphonate seems to be a rational counter.  相似文献

The purpose of this study was to evaluate the effects of isolated alterations in mineral content on mouse bone torsional properties. The femora and tibiae from 25 eight-week-old male A/J strain mice were divided into five groups and selectively decalcified from 5% to 20%. The right femora were then tested to failure in torsion while the tibiae were ashed to determine final mineral content of the decalcified bones. Contralateral femora were serially cross-sectioned to determine geometric properties, and effective material properties were then calculated from the geometric and structural properties of each femoral pair. We found that the relationship between ash content and effective shear modulus or maximum effective shear stress could best be characterized through a power law, with an exponential factor of 6.79 (R2 = 0.85) and 4.04 (R2 = 0.67), respectively. This indicates that in a murine model, as with other species, small changes in ash content significantly influence effective material properties. Furthermore, it appears that (in adolescent A/J strain mice) effective shear modulus is more heavily affected by changes in mineralization than is maximum effective shear stress when these properties are derived from whole bone torsional tests to failure.  相似文献

The interaction between the gravitational field, the position of the body, and the functional characteristics of the blood vessels determines the distribution of intravascular volume. In turn, this distribution determines cardiac pump function. One of the most profound circulatory changes that occurs in man during exposure to weightlessness is a cephalad redistribution of fluid caused by the lack of hydrostatic pressure in this microgravitative environment. The cephalad redistribution of fluid results in a loss of blood volume and then induces a decrease in preload. Recently, a decrease in sensitivity of arteriole to catecholamine has reported in rats of simulated weightlessness. This change in arteriole may reduce afterload. As a result, cardiovascular system may be shifted to a hypokinetic state during weightlessness condition for long-term. Echocardiographic data from astronauts during space flight showed an increase in heart rate, a 12 % decrease in stroke volume, and a 16 % decrease in left end diastolic volume. Electron-microscopic studies have shown changes in cardiac morphology in rats after exposure to microgravity for 7-12.5 days. After the COSMOS 2044 flight for 14 days, the light-microscopic studies have shown an atrophy of papillary muscles in rats left cardiac ventricle. It is not clear whether the function of atrophic myocardium is impaired. The data in three aspects as mentioned above suggest that weightlessness or simulated weightlessness may decrease the myocardial function. However, definite changes in cardiac performance have been hard to prove due to many limits. This studies were to answer two questions: Is the myocardial contractility depressed in rats subjected to simulated weightlessness for four weeks? To assess the performance of femoral orthopedic implants, they are often attached to cadaveric femurs, and biomechanical testing is performed. To identify areas of high stress, stress shielding, and to facilitate implant redesign, these tests are often accompanied by finite element (FE) models of the bone/implant system. However, cadaveric bone suffers from wide specimen to specimen variability both in terms of bone geometry and mechanical properties, making it virtually impossible for experimental results to be reproduced. An alternative approach is to utilize synthetic femurs of standardized geometry, having material behavior approximating that of human bone, but with very small specimen to specimen variability. This approach allows for repeatable experimental results and a standard geometry for use in accompanying FE models. While the synthetic bones appear to be of appropriate geometry to simulate bone mechanical behavior, it has not, however, been established what bone quality they most resemble, i.e., osteoporotic or osteopenic versus healthy bone. Furthermore, it is also of interest to determine whether FE models of synthetic bones, with appropriate adjustments in input material properties or geometric size, could be used to simulate the mechanical behavior of a wider range of bone quality and size. To shed light on these questions, the axial and torsional stiffness of cadaveric femurs were compared to those measured on synthetic femurs. A FE model, previously validated by the authors to represent the geometry of a synthetic femur, was then used with a range of input material properties and change in geometric size, to establish whether cadaveric results could be simulated. Axial and torsional stiffnesses and rigidities were measured for 25 human cadaveric femurs (simulating poor bone stock) and three synthetic "third generation composite" femurs (3GCF) (simulating normal healthy bone stock) in the midstance orientation. The measured results were compared, under identical loading conditions, to those predicted by a previously validated three-dimensional finite element model of the 3GCF at a variety of Young's modulus values. A smaller FE model of the 3GCF was also created to examine the effects of a simple change in bone size. The 3GCF was found to be significantly stiffer (2.3 times in torsional loading, 1.7 times in axial loading) than the presently utilized cadaveric samples. Nevertheless, the FE model was able to successfully simulate both the behavior of the 3GCF, and a wide range of cadaveric bone data scatter by an appropriate adjustment of Young's modulus or geometric size. The synthetic femur had a significantly higher stiffness than the cadaveric bone samples. The finite element model provided a good estimate of upper and lower bounds for the axial and torsional stiffness of human femurs because it was effective at reproducing the geometric properties of a femur. Cadaveric bone experiments can be used to calibrate FE models' input material properties so that bones of varying quality can be simulated.  相似文献

In this study changes in the properties of natural organic matter (NOM) were studied during composting of sewage sludge in a laboratory experiment using the pile method. Typical physicochemical parameters were measured during 53 days of composting including humic fractions. The effects of humification on the molecular properties of humic acids (HA) were investigated by 13C CP/MAS NMR spectroscopy. On the basis of chemical analyses, 53 days of composting sewage sludge with structural material can be divided into three phases: (i) domination of rapid decomposition of non-humic, easily biodegradable organic matter (two to three weeks), (ii) domination of organic matter humification and formation of polycondensed, humic-like substances (the next two weeks), (iii) stabilization of transformed organic material and weak microbial activity. Spectroscopic characterization (13C NMR) of compost humic acids reveals changes in their structures during maturation. The changes are highly correlated with the processes taking place in bulk compost.  相似文献

The purpose of the study was to compare indentation modulus (IM) and hardness of condylar bone in young and adult dogs. In addition we desired to examine histologic sections for bone formation activity in the two groups. Mandibular condyles were obtained from adult (1- to 2-year-old) and young (approximately 5-m old) dogs. Two sections/condyle were obtained and one was processed for histomorphometry and the other for mechanical analyses. Indents were made on moist condylar trabecular bone to a depth of 500 nm at a loading rate of 10 nm/s using a custom-made hydration system to obtain IM and hardness. Histomorphometric analyses measured the bone volume/total volume (BV/TV%) and ratio of labeled to unlabeled bone within the condyle. Data were analyzed using a repeated-measures factorial analysis of variance and Tukey-Kramer method. Overall, the IM of the adult condyles (10.0+/-3.4 GPa, Mean+/-SD) were significantly (P<0.0001) higher than in young dogs (5.6+/-2.6 GPa). There was a greater bone mass in the young (60.2%) versus the adult condyles (42%). Also, significantly more labeled bone in the young (66.1%) condylar bone suggested higher bone forming activity than in adult condyles (27.5%). With age there is a change in mass and material properties in the trabecular bone of the mandibular condyle in dogs.  相似文献

Structural characteristics of the femur are compared in preagricultural (2200 B.C.–A.D. 1150) and agricultural (A.D. 1150–1550) subsistence strategy groups from the Georgia coast. Using an automated technique, cross-sectional geometric properties used in structural analyses (areas, second moments of area) were determined at midshaft and distal to the lesser trochanter in 20 adults from each group. A significant decline in magnitude of almost every geometric property occurs in the agricultural group. The differences between groups are reduced but still significant for many properties after standardizing for bone length differences. In addition, a remodeling of the femoral cortex to one of relatively smaller medullary and subperiosteal diameter, as well as a more circular cross-sectional shape, is characteristic of agricultural femora. Thus, while the relative cross-sectional area of bone remains the same, the spatial distribution of bone area is different in the two groups. The results strongly suggest a relative reduction in mechanical loadings of the femur in the agricultural group, implying different levels and possibly types of activity involving the lower limb in the two groups. The data are also compared with similar data available for the Pecos Pueblo (agricultural) sample. The comparison indicates that types of activity may have been more similar in the two agricultural samples, but that general levels of activity were more similar in the Pecos Pueblo and Georgia coast preagricultural samples.  相似文献

Laying hens develop a type of osteoporosis that arises from a loss of structural bone, resulting in high incidence of fractures. In this study, a comparison of bone material properties was made for lines of hens created by divergent selection to have high and low bone strength and housed in either individual cages, with restricted mobility, or in an aviary system, with opportunity for increased mobility. Improvement of bone biomechanics in the high line hens and in aviary housing was mainly due to increased bone mass, thicker cortical bone and more medullary bone. However, bone material properties such as cortical and medullary bone mineral composition and crystallinity as well as collagen maturity did not differ between lines. However, bone material properties of birds from the different type of housing were markedly different. The cortical bone in aviary birds had a lower degree of mineralization and bone mineral was less mature and less organized than in caged birds. These differences can be explained by increased bone turnover rates due to the higher physical activity of aviary birds that stimulates bone formation and bone remodeling. Multivariate statistical analyses shows that both cortical and medullary bone contribute to breaking strengthThe cortical thickness was the single most important contributor while its degree of mineralization and porosity had a smaller contribution. Bone properties had poorer correlations with mechanical properties in cage birds than in aviary birds presumably due to the greater number of structural defects of cortical bone in cage birds.  相似文献

Fracture risk in type 2 diabetes is increased despite normal or high bone mineral density, implicating poor bone quality as a risk factor. Raloxifene improves bone material and mechanical properties independent of bone mineral density. This study aimed to determine if raloxifene prevents the negative effects of diabetes on skeletal fragility in diabetes-prone rats. Adult Zucker Diabetic Sprague-Dawley (ZDSD) female rats (20-week-old, n = 24) were fed a diabetogenic high-fat diet and were randomized to receive daily subcutaneous injections of raloxifene or vehicle for 12 weeks. Blood glucose was measured weekly and glycated hemoglobin was measured at baseline and 12 weeks. At sacrifice, femora and lumbar vertebrae were harvested for imaging and mechanical testing. Raloxifene-treated rats had a lower incidence of type 2 diabetes compared with vehicle-treated rats. In addition, raloxifene-treated rats had blood glucose levels significantly lower than both diabetic vehicle-treated rats as well as vehicle-treated rats that did not become diabetic. Femoral toughness was greater in raloxifene-treated rats compared with both diabetic and non-diabetic vehicle-treated ZDSD rats, due to greater energy absorption in the post-yield region of the stress-strain curve. Similar differences between groups were observed for the structural (extrinsic) mechanical properties of energy-to-failure, post-yield energy-to-failure, and post-yield displacement. These results show that raloxifene is beneficial in preventing the onset of diabetes and improving bone material properties in the diabetes-prone ZDSD rat. This presents unique therapeutic potential for raloxifene in preserving bone quality in diabetes as well as in diabetes prevention, if these results can be supported by future experimental and clinical studies.  相似文献

为研究模拟失重是否可以引起大鼠心肌与血管组织HSP70的诱导表达发生改变,用尾部悬吊大鼠模型模拟失重,以研究失重对生理的影响,用Northern杂交与Western印迹分析检测4周模拟失重大鼠热应激后并在室温下恢复1h(SUS-H1)或2h(SUS-H2_心肌,血管组织HSP70表达的变化,结果表明,热应激后,各组大鼠心肌组织的HSP72 mRNA表达的均显著增加,但SUS-H2大鼠心肌组织的表达显著低于CON-H2组;各组大鼠心肌组织HSP72表达也均显著增加,但SUS-H1与SUS-H2大鼠的表达与相应对照组相比,则仅呈降低趋势,其底动脉血管组织的HSP72 mRNA与HSP72诱导表达均显著增高,而在股动脉则两者仅呈降低趋势,上述结果提示,模拟失重可导致大鼠心肌发生类似衰老的心肌改变;身体前,后部血管组织HSP70的诱导表达变化可能与血管的分化性适应方向一致。  相似文献

许多数据表明长期失重以后立位耐力降低可能与压力感受性反射功能的改变有关。本文比较了两组被试者15天低动力卧床前后的立位耐力。以血压调节模型为基础分析了两种不同方式卧床前后单纯立位和下身负压加立位时压力感受性反射功能的改变,并用颈部加压及下身负压对中枢调节功能改变进行了观察。结果表明严格的头低位卧床后,立位耐力下降及压力感受性反射功能改变明显大于半日平卧半日倚坐者。而压力感受性反射功能的改变,特别是中枢神经系统调节功能的紊乱,是卧床后立位耐力降低的主要原因。从这种考虑为基础,作者提出了改变失重或模拟失重状态下的血液分布,调整对压力感受器的刺激,可能是预防心血管失调的有效方法。  相似文献

本工作旨在探讨短、中期模拟失重下人鼠肠系膜小动脉血管平滑肌细胞(vascular smooth muscle cells,VSMCs)电压依赖性钙离子通道(voltage-dependent calcium channels,VDC)功能的改变。以尾部悬吊大鼠模型模拟失重对不同部位血管的影响。采用全细胞膜片钳实验技术,以Ba^2 作为载流子,测定1周及4周模拟失重人鼠肠系膜小动脉VSMCs的VDC电流密度、稳态激活与失活曲线及有关参数,并与对照组结果进行比较。研究表明,本实验所记录到的内向电流主要为钡离子通过长时程VDC(L-VDC)所形成的电流。与对照组相比,1周模拟失重大鼠肠系膜小动脉VSMCs的L-VDc电流密度仪呈降低趋势;但4周模拟失重人鼠肠系膜小动脉VSMCs的L-VDC电流密度则已显著降低。此外,与对照组相比,1、4周模拟失重大鼠肠系膜小动脉VSMCs的膜电容、翻转电位与L-VDC的一些动力学特征值,如通道的开放与关闭速率,通道电流稳态激活与火活曲线及其特征拟合参数V0.5与K的值,均末见有显著改变。结果提示:模拟失重下后身小动脉VSMCs的VDC功能降低可能是模拟失重引起人鼠后身动脉收缩反应性降低及适应性萎缩变化的电生理机制之一。  相似文献

The aim of this study was to investigate the effects of the clinostat-simulated weightlessness on biological characteristics of MLO-Y4 osteocyte-like cells. MLO-Y4 cells were incubated for 24 h, then randomly divided into 3 groups and rotated in a clinostat as a model of simulated weightlessness for 12 h, 24 h and 48 h. The morphology, cytoskeleton, and secretion of soluble molecules of MLO-Y4 cells were observed and detected. The results show that clinostat culture affects the number of dendrites/cell, cytoskeleton distribution, and secretion of nitric oxide and prostaglandin E2 in MLO-Y4 cells. These results may provide some clue to explore the cellular mechanism of bone loss caused by weightlessness.  相似文献

Differential response of rat limb bones to strenuous exercise   总被引:4,自引:0,他引:4  
We examined the influence of a strenuous exercise regimen on tibial and metatarsal bones to show not only how the geometric, histological, and mechanical properties of immature bone respond to strenuous exercise but also how long bones within the same limb may respond differentially to exercise. Female Sprague-Dawley rats (8 wk old) were divided randomly into two groups: a sedentary control (n = 15) and an exercised group (n = 15). The exercise intensity was 80-90% of maximum oxygen capacity 5 days/wk for 10 wk. Mechanical properties of tibia and second metatarsus (MT) were determined with three-point bending, and contralateral bones were used for geometric and histological analyses. Length and middiaphyseal cross-sectional geometry of the exercised tibiae were significantly less than controls, but material properties were not different. The exercised tibiae had significantly lower structural properties (e.g., loads at the proportional limit and maximum and energy at failure load). The middiaphyseal dorsal cortex of exercised MT was significantly thicker than controls, but tensile stress at the proportional limit and elastic modulus of exercised MT were significantly less than controls. The average number of osteons and osteocytes per unit area of the tibial middiaphysis was significantly greater in the exercised group--especially in lateral and posterior cortices. The number of osteons and osteocytes per unit area in the MT, however, was significantly less in the exercised group. The differential effects of strenuous exercise on tibia and MT suggest that local loading and bone-specific responses have important roles in modulating the response of immature bone to strenuous exercise.  相似文献

To investigate the influence of experimental diets on morphological and mechanical characteristics of immature bone, this study thoroughly examined the nutrition–bone connection. A non-destructive evaluation method involving high-resolution in-vivo micro-computed tomography and finite element (FE) analysis was used to investigate the relationship between obesity and osteopenia—two disorders of body composition. Correlation of nutritional deficiency with bone characteristics was also investigated. Some recent studies have shown that both obesity and osteopenia share several common genetic and environmental factors. However, there have been few studies correlating these pathologies in-vivo from a structural and biomechanical point of view. In the present study, detailed changes in morphological and mechanical characteristics of trabecular bone architecture were detected and tracked by longitudinal studies of morphometric parameters and simulated compression testing. Rats were randomized into three groups: overeaten diet (OD) for formation of obesity, normal diet (ND), and restricted diet (RD) in which rats received 65% of the normal diet. In the OD and ND groups, all structural parameters changed significantly (p<0.05). The degree of alteration in the structural parameters of the ND group was similar to that of the RD group (p<0.05). In simulated compression tests using FE models, the effective modulus of the OD group significantly decreased, depending on measuring time (p<0.05), whereas that of the ND and RD groups significantly increased (p>0.05). The key finding of the present study is that fat mass is morphologically and mechanically inversely correlated with bone mass when the mechanical loading effects of greater body weight on bone mass are applied.  相似文献

建立在失重环境下NK细胞的培养体系,探讨失重培养条件对NK细胞活性的影响。取C57BL/6小鼠脾,分离NK细胞,在正常重力和失重状态下旋转细胞两种培养体系中培养NK细胞48 h。以Yac-1细胞为靶细胞,采用MTT法检测其杀伤活性;半定量PCR分析穿孔素、颗粒酶的转录水平。与正常重力培养对照相比,失重培养的NK细胞杀伤活性显著降低,其穿孔素和颗粒酶B的转录水平均显著低于正常重力培养组,穿孔素/GAPDH结果为失重培养组0.625±0.042;正常重力培养组1.054±0.036(P<0.01);得到颗粒酶B/GAPDH结果为失重培养组0.700±0.042;正常重力培养组1.068±0.058(P<0.01)。成功建立了NK细胞体外培养体系,在失重环境下NK细胞杀伤活性降低。  相似文献

Background

Warfarin, a widely used anticoagulant, is a vitamin K antagonist impairing the activity of vitamin K-dependent Bone Gla Protein (BGP or Osteocalcin) and Matrix Gla Protein (MGP). Because dabigatran, a new anticoagulant, has no effect on vitamin K metabolism, the aim of this study was to compare the impact of warfarin and dabigatran administration on bone structure and vascular calcification.

Methods

Rats with normal renal function received for 6 weeks warfarin, dabigatran or placebo. Bone was evaluated immuno-histochemically and hystomorphometrically after double labelling with declomycin and calcein. Aorta and iliac arteries were examined histologically.

Results

Histomorphometric analysis of femur and vertebrae showed significantly decreased bone volume and increased trabecular separation in rats treated with warfarin. Vertebra analysis showed that the trabecular number was higher in dabigatran treated rats. Osteoblast activity and resorption parameters were similar among groups, except for maximum erosion depth, which was higher in warfarin treated rats, suggesting a higher osteoclastic activity. Therefore, warfarin treatment was also associated with higher bone formation rate/bone surface and activation frequency. Warfarin treatment may cause an increased bone turnover characterized by increased remodelling cycles, with stronger osteoclast activity compared to the other groups. There were no differences among experimental groups in calcium deposition either in aortic or iliac arteries.

Conclusions

These findings suggest for the first time that dabigatran has a better bone safety profile than warfarin, as warfarin treatment affects bone by reducing trabecular size and structure, increasing turnover and reducing mineralization. These differences could potentially result in a lower incidence of fractures in dabigatran treated patients.  相似文献
