Blood is one of the widely used specimens for biological trace element research because of its biological significance and ease of sampling. We have conducted a study of the blood of the Kalpakkam township population for trace and minor elements. For this purpose, analytical methods have been developed and standardized in our laboratory for the elemental analysis of blood plasma and red cells. Inductively coupled plasma-mass spectrometry (ICP-MS), a relatively new technique, has been applied for the analysis of trace elements. Details regarding spectral interference and matrix interference encountered in the analysis of blood and the methods of correcting them have been discussed. Flame atomic absorption spectrometry (AAS)/atomic emission spectrometry (AES) has been applied for the determination of minor elements. Precision and accuracy of these methods have also been discussed.  相似文献

In this study, a number of selected trace elements and clinically relevant parameters were compared between thoracic empyemata and the corresponding sera for a better understanding of the trace element distribution between these two compartments. Serum-empyema pairs were obtained from 13 patients and quantified for selected and essential trace elements, namely copper (Cu), zinc (Zn), manganese (Mn), rubidium (Rb), and magnesium (Mg), by inductively coupled plasma-mass spectrometry (ICP-MS). In addition, the concentrations of the following clinical laboratory parameters were analyzed by standard methods: total protein, leukocyte count, lactate dehydrogenase, glucose, pH, and the C-reactive protein. Individual concentrations of the elements determined in the empyemata were frequently higher than in pleural effusions of any other benign or malignant condition except for Cu. Serum Cu exceeded the normal range (600–1400 μg/kg) in 6 out of 13 patients (median 1410 μg/kg). In the empyemata, Zn concentrations (median 2000 μg/kg) were characteristically higher than in the sera (median 450 μg/kg) and exceeded the upper limit for serum (1200 μg/kg) in 8 of the 13 patients. Manganese concentrations in the empyemata (median 2.7 μg/kg) were also higher compared to corresponding sera, although they stayed within the limits considered normal for serum of healthy adults (upper limit 2.9 μg/kg). Rubidium was also moderately higher in most empyemata (median 290 μg/kg) and exceeded the upper limit for serum (560 μg/kg) in two patients. The median concentration of the essential element magnesium was higher in the empyemata (23 mg/kg) than in the sera (21 mg/kg). However, all serum Mg concentrations except three remained within the normal range (17–22 mg/kg). Removal of large amounts of empyematous fluid may deprive the body of trace elements and can cause suboptimal or deficient trace element status and homeostasis. Recuperation will be accelerated by compensatory supplementation of trace elements. Therefore, selective medication with adequate trace element compounds in patients with thoracic empyema can be generally recommended for zinc. The other elements need not necessarily be monitored or substituted, because of their stable concentrations in the serum. Information concerning the chemical state of trace elements in biological systems generally has not been available. Such information for toxic elements and metals in metalloproteins could prove extremely valuable in the elucidation of their metabolism and other biological processes. The shielding of core electrons by binding electrons affect the energy required for creating inner-shell holes. Furthermore, the molecular binding and symmetry of the local environment of an atom affect the absorption spectrum in the neighborhood of the absorption edge. X-ray absorption near-edge structure (XANES) using synchrotron radiation excitation can be used to provide chemical speciation information for trace elements at concentrations as low as 10 ppm. The structure and position of the absorption curve in the region of an edge can yield vital data about the local structure and oxidation state of the trace element in question. Data are most easily interpreted by comparing the observed edge structure and position with those of model compounds of the element covering the entire range of possible oxidation states. The pathogenesis of some heart diseases has been associated with changes in the balance of certain trace elements. However, whether blood trace element changes exist that are related to changes in the cardiovascular system are, in most cases, unknown. In this study, blood trace element levels were analysed in 46 patients with non-rheumatic aortic valve sclerosis that were previously shown to have a disturbed trace element balance in their valve tissue, including 11/15 elements. Results showed significant changes of blood levels of 8/15 trace elements in these patients when compared with blood levels in 46 healthy controls. Of these elements, Cd and Mg were the only elements that increased in both blood and valves. Cu and Se were increased in blood but decreased in valves, whereas Co and Zn were decreased in blood but increased in valves. Several elements (As, Ca, Fe, Pb, and V) were unchanged in blood although changed in valves. Although Mn and Hg showed changes in blood, this was not evident in the valves. Al and Ag were the only elements that did not change in both blood and valves. Significant covariation in blood and valve levels was only observed for Al and Pb. The recorded pattern of trace element changes indicates a complex competition/exchange between body compartments in this disease, where the increased blood Cu/Zn ratio suggests an ongoing infectious/inflammatory process.  相似文献

Contamination in a trace element laboratory can come from a variety of sources, including laboratory gloves. Therefore, vinyl and latex gloves were obtained from as many manufacturers as would supply gloves. These gloves were either prepared for acid-washing and subsequent soaking in an acid solution, or immersed in an acid solution for a duration of either 1 min or 1 h. Incubation washes were analyzed for a variety of trace elements by flame atomic abosrption spectroscopy (AAS) or inductively coupled mass spectrometry (ICP-MS). Results indicated that only three brands of vinyl gloves were acceptable for use in a trace element laboratory, whereas others had contamination of different elements. Latex gloves contained such high levels of biologically important elements that they were not considered suitable for routine trace element work. Vinyl gloves of choice should be routinely acid-washed before use in a trace element laboratory.  相似文献

不同利用强度下绿洲农田土壤微量元素有效含量特征   总被引:13,自引:0,他引:13  
以地处极端干旱背景下的塔里木盆地南缘策勒绿洲为研究区,以绿洲化为视角,根据当地农民耕作习惯于2005年选择4块试验农田,分别代表当地典型的农田利用强度。基于单项指数(Ei)和综合指数(Ec)相结合的评价方法,分析了绿洲农田在不同利用强度下的土壤Fe、Cu、Mn、Zn等微量元素有效性,探讨了土壤微量元素与人为耕作管理措施之间的关系。结果表明:人为耕作管理强度会对土壤微量元素有效含量产生重要影响,绿洲不同位置农田因施肥强度和管理方式的差别,其土壤微量元素有效含量特征存在显著差异。绿洲内部农田土壤有效Fe、Cu、Mn、Zn含量显著高于绿洲边缘各样地,新垦农田土壤有效Fe、Cu、Zn含量均低于对照样地;绿洲内部农田土壤微量元素有效性最高,新垦农田最低;农田土壤微量元素有效性与土壤有机质存在显著的正相关关系。  相似文献

It has become evident over the last two decades that there is an intimate relationship between the trace elements and cancer. Some trace elements have been shown to be carcinogens, others appear to provide protection against cancer. Profound changes in trace element concentrations and distribution occur in patients with cancer, but most changes remain undefined. A review of a number of studies of trace element changes in patients with cancer demonstrates that simple correlations of trace element levels in disease are of only limited use. Such reports underscore the need for large-scale studies that consider the many variables of malignancies and of trace element chemistry. The variables that must be considered for cancer include tissue of origin; histologic, pathologic and clinical staging; nutritional status as reflected by serum levels of calcium, iron, magnesium, phosphorus, the electrolytes, pH, albumen, and globulin; endocrine balance, effects of previous and concurrent therapies such as surgery, chemotherapy, hormonal manipulation, immunotherapy, and radiotherapy; history of exposure to toxic agents; and the presence of other disease. Similarly, trace element studies entail variables that must be considered and controlled prospectively, including timing and techniques of sampling, storage, and analysis, and simultaneous measurement of at least the majority of possibly interrelated elements rather than studying one element at a time. The various national cooperative oncology groups such as ECOG, SWOG, and SEOG now offer unusually well-studied groups of cancer patients who are managed according to carefully and prospectively defined protocols in participating institutions. With present knowledge, it is now time to approach these groups with a proposal to incorporate trace element studies in their protocols. A potential protocol will be discussed.  相似文献

In this study special interest was given to trace elements recognized as to be carcinogenic to humans. The kidney tissue sections were analyzed in order to determine the concentrations of elements present in the sample. The Synchrotron Radiation Induced X-ray Emission (SRIXE) technique was applied using a white photon microbeam. The results from cancerous parts of the kidney tissues were compared to non-cancerous parts and to the control group. In addition the iron concentration level was determined in the serum of those patients. Two-dimensional scans are presented to illustrate the differences between perfused and not-perfused tissues. According to this study there is no significant difference in the Mn concentration between cancerous and non-cancerous parts of the kidney, but the concentrations of Cd, Cr, Ti, V, Cu, Se, and Zn are at a lower concentration level in the cancerous parts than in the non-cancerous parts. A converse observation has been made for Fe. This may be associated with different metabolism and dynamics of the cancer process and both higher vascularization and need of higher blood supply in the cancerous tissue. The two-dimensional scanning of thin kidney sections showed differences in the trace element distributions depending on the analyzed samples: perfused and non-perfused. Perfusion removed blood mostly from the peritubular capillaries while in the glomerulus some capillaries had a relatively high Fe content. A low Fe concentration was observed in nephron tubules while a converse observation has been made for Cd. This may indicate that Cd is localized in the cells but not in the blood.  相似文献

微量元素如铁、锌、铜等对维持生物体代谢和健康至关重要,其含量失衡会造成代谢异常甚至死亡,因此生物体存在复杂机制维持这些微量元素的稳态代谢平衡(homeostasis)。近年来国际上一些实验室尝试用模式脊椎生物斑马鱼来开展该领域的研究,展示出斑马鱼的特有优势。特别是大规模正向遗传学筛选的成功开展,一系列微量元素代谢异常的突变体(如:weissherbst、chardonnay、chianti、shiraz、gavi、calamity和catastrophe)相继发现,为研究离子代谢调控机制和相关疾病的发病机理,提供了整体动态的活体模型。铁代谢相关基因加,2J和grx5都己在斑马鱼中成功定位克隆,斑马鱼铜载体基因atp7a突变体calamity的深入研究,进一步阐明了Menkes病的发病机理。利用斑马鱼的优势,结合小鼠模型和人群来研究微量元素的体内稳态代谢平衡将是微量元素代谢机制研究的新方向。  相似文献

微量元素包括铁、锌、铜、锰、硒、碘等,在机体生命活动中发挥着极其重要的生理学功能,参与蛋白质结构,并通过与蛋白质和其他有机基团结合参与酶、激素和维生素等生物大分子合成。微量元素代谢失衡会发生系列病理改变并最终导致疾病。生物微量元素研究是多学科交叉的科学研究领域,业已成为生命科学的国际研究热点。该领域发展对人类生存与健康具有重要意义。经过几代科学家的不懈努力,我国的生物微量元素研究已经从无到有,逐步取得令国际同行瞩目的科研成果,尤其近年来,我国该领域基础研究快速发展,更是相继产生了诸多高水平原创性科研成果。这种迅速发展与壮大,得益于大批海外归国优秀人才,更离不开国家和社会在科研经费和政策等方面的大力支持。简介了近年来我国生物微量元素研究领域的科学家及其主要科研成果,期望广大读者能全面了解国内生物微量元素研究现状以及可喜的发展趋势。衷心期待着中国的生物微量元素研究能有更大发展,能有更大成就。  相似文献

Specific plant species that can take up and accumulate abnormally high concentrations of elements in their aboveground tissues are referred to as “hyperaccumulators”. The use of this term is justified in the case of enormous element-binding capacity of plants growing in their natural habitats and showing no toxicity symptoms. An increasing interest in the study of hyperaccumulators results from their potential applications in environmental biotechnology (phytoremediation, phytomining) and their emerging role in nanotechnology. The highest number of plant species with confirmed hyperaccumulative properties has been reported for hyperaccumulators of nickel, cadmium, zinc, manganese, arsenic and selenium. More limited data exist for plants accumulating other elements, including common pollutants (chromium, lead and boron) or elements of commercial value, such as copper, gold and rare earth elements. Different approaches have been used for the study of hyperaccumulators – geobotanical, chemical, biochemical and genetic. The chemical approach is the most important in screening for new hyperaccumulators. This article presents and critically reviews current trends in new hyperaccumulator research, emphasizing analytical methodology that is applied in identification of new hyperaccumulators of trace elements and its future perspectives.  相似文献

Influence of trace elements in body metabolism and their physiological importance in various diseases have motivated their accurate and quantitative determination in biological tissues and fluids. Instrumental Neutron Activation Analysis (INAA) using short and long term irradiation has been employed to determine five minor elements (Cl, K, Na, Mg, P) and 15 trace elements (As, Br, Co, Cr, Cs, Cu, Fe, Hg, Mn, Rb, Sb, Se, Sc, Sr, and Zn) in cancerous and normal breast tissue from 30 patients of four clinical stages. Several elements show enhancement in cancerous breast tissue. Selenium shows maximum enhancement of 94.7% followed by K (81.6%), Sc (66.7%), Cu (58.2%), Na (48.5%), P (44.4%), and Zn (39.2%). Some element, such as Fe, Cr, and Mn, are depressed by 30.8, 30.1, and 12.8%, respectively. These elements compete for binding sites in the cell, change its enzymatic activity and exert direct or indirect action on the carcinogenic process accelerating the growth of tumors. This is further evidenced by histopathological examination of cancerous cells showing poor cytological differentiation. An attempt has been made to correlate trace element concentrations of Se, Cu, Zn, Rb, Br, Hg, As, Co, Fe, Cr, and Mn and the ratios of Se/Zn, K/P, Cu/Zn, Na/K, and Se/Fe with the clinical stages of cancer. Inhibition of enzymatic activity caused by variation in trace element concentrations results in immunological breakdown of the body system.  相似文献

Trace elements are known to have a key role in myocardial metabolism. The accumulation (cobalt, arsenic, copper) or deficiency (selenium, zinc) of trace elements may be responsible for idiopathic dilated cardiomyopathy. We investigated the trace element concentrations (Cu, Zn, Mg) in sera from patients with dilated cardiomyopathy by atomic absorption spectrophotometry. We observed that patients with dilated cardiomyopathies have higher copper and lower zinc concentrations in serum than healthy controls. The magnesium concentrations of patients did not differ significantly from that of control subjects.  相似文献

The distributions of trace elements in Shield, Ace and Burton Lakes of the Vestfold Hills were investigated. Three aspects are discussed as follows: (1) the vertical distribution of 18 trace elements in the three lakes, (2) the behaviour of trace elements in the lakes, especially that of manganese in Shield Lake, and (3) the origin of trace elements in antarctic saline lakes.High concentrations of trace elements were found in these coastal saline lakes, when compared to open ocean water.We suggest that the peak of total extractable manganese, found at 20 m in Shield Lake, was related to the oxic/anoxic water interface brought about by microbiological activity. Solid phase manganese at the upper oxic layer may have precipitated and then reached the anoxic boundary to be there reduced to manganese ion. This dissolved manganese may then have diffused upwards to be reoxidized to a solid form. This cycle, repeated many times, may have produced the Mn profile.The alkali, alkaline earth elements and Cl were probably derived from relict seawater. Other elements were present in similar concentration ratios to those of South Polar aerosols. Residence time calculations indicate that fallout of aerosol particles, themselves derived from various sources, is capable of accounting for the measured concentrations of some trace elements in Shield Lake. This source of trace elements may be significant for other antarctic saline lakes.  相似文献

Royal jelly from Apis mellifera is a highly active natural biological substance and is probably one of the most interesting raw substances in natural product chemistry. Trace elements play a key role in the biomedical activities associated with royal jelly, as these elements have a multitude of known and unknown biological functions. For this reason concentrations of 28 trace (Al, Ba, Sr, Bi, Cd, Hg, Pb, Sn, Te, Tl, W, Sb, Cr, Ni, Ti, V, Co, Mo) and mineral (P, S, Ca, Mg, K, Na, Zn, Fe, Cu, Mn) elements were systematically investigated in botanically and geologically defined royal jelly samples. In addition, concentrations of 14 trace elements were measured in the associated honey samples--honey being the precursor of royal jelly. Concentrations of K, Na, Mg, Ca, P, S, Cu, Fe, Zn, Al, Ba and Sr in royal jelly were determined by inductively coupled plasma optical emission spectroscopy (ICP-OES), while concentrations of Bi, Cd, Hg, Pb, Sn, Te, Tl, W, Sb, Cr, Mn, Ni, Ti, V, Co and Mo in royal jelly were determined by double focusing magnetic sector field inductively coupled plasma mass spectrometry (SF-ICP-MS). In the honey samples, trace and mineral element concentrations strongly depended on botanical and geological origin, and substantial variation was found. In contrast, the concentrations of trace and mineral elements were highly constant in the associated royal jelly samples. The most important results were the homeostatic adjustments of trace and mineral element concentrations in royal jelly. This effect was evidently produced in the endocrine glands of nurse bees, which are adapted for needs of bee larvae. In conclusion, this research yielded a surprising and completely new finding--that royal jelly, as a form of lactation on the insect level, shows the same homeostatic adjustment as mammalian and human breast milk.  相似文献

Abstract

The toxicity, mobility, bioavailability and bioaccumulation of metals are dependent on the particular physico-chemical form in which the element occurs in the environment. Special attention has been paid to metals which are essential for the proper functioning of organisms if present in appropriate amounts but are toxic if in excess (i.e. Se, Cr), and also to non-essential elements (i.e. Hg, Pb, Cd, Sn and As). To assess the potential hazard to the health of marine organisms, qualitative and quantitative analyses of metal species accumulating along the food chain needs to be carried out. This paper reviews the available information on the speciation of trace elements in the food chain in marine ecosystems and the analytical tools used for acquiring reliable information in this field. Advantages and limitations of commonly used techniques indicate that all metal species in different samples need diverse extraction, separation and detection conditions. Although not recommending which procedure is the most suitable to determine a given compound, speciation analysis has the potential to be a powerful tool for the identification of trace element species in biological samples.  相似文献

矿物元素在畜禽生长发育、新陈代谢、神经活动、免疫功能、内分泌等几乎所有的生命活动过程中都发挥着重要的生理作用,分为钙、磷、钠、镁等常量元素和铁、锰、钴、铜等微量元素。某种矿物元素摄入不足或在体内过量蓄积、矿物元素间比例失调,都将引起严重后果。随着畜禽养殖业的集约化规模化发展,饲用矿物元素用量迅速增加,但存在不合理使用的情况,造成了资源浪费和环境污染。因此,在满足畜禽营养需要量的前提下,合理地使用矿物元素剂型及剂量,实现降低矿物元素经畜禽粪污向环境中的排放量,不仅利于集约化畜禽养殖业的发展,同时也能有效降低畜禽粪污中的重金属含量,为种植业提供优良的有机肥来源,维护生态系统的平衡,促进种植业和养殖业的可持续健康发展。基于此,以畜禽体内含量最高的常量元素钙磷和微量元素为切入点,介绍了饲用矿物元素的应用现状、畜禽矿物元素基础需要量及合适的添加范围,着重综述了新型饲用微量元素的开发与应用进展,并探讨了矿物元素动态饲喂模式的可行性及有效性,以期为合理利用矿物元素资源及进一步解决全球生态农业研究的畜禽矿物元素导致的环境污染提供参考。  相似文献

Trace elements are indispensable for the effective and proper functioning of biological systems. Recent years have demonstrated the conspicuous lack of knowledge about trace-element physiology. Establishment of reference values is a very difficult task, requiring the consideration of and compensation for a number of possible simultaneous phenomena. Peripheral blood has been used in medical diagnosis for a very long time, because, among other things, it is easily accessible. In the search for signs of lack or excess of minerals and trace elements in disease, the interest has been focused mainly on blood plasma or serum. The utilization of blood cells as a marker model is proposed here. The advent of the nuclear microprobe made possible the determination of elemental profiles of individual cells. The techniques of blood cell separation and preparation for microprobe analysis are presented and discussed. As an example of a possible diagnostic application, a set of reference data from a control group is compared to corresponding data from a group of patients suffering from acute myeloid leukemia.  相似文献
