Genetic control of the level of blood catalase activity was first demonstrated in 1927. At present, such control has been demonstrated or suggested for nine different species, including man, the most studied. The development of an acatalasemic strain of mice has permitted a wide variety of experimental approaches, including most of those used in humans. Among those approaches which cannot readily be applied to man but have been used in acatalasemic mice are investigations of sensitivity to radiation lethality, mechanism of awareness to radiation, possible use as a model for replacement therapy for inborn errors of metabolism, and catalase in tissues other than erythrocytes. These are described, together with genetic, immunological, and other studies comparable to similar work on acatalasemic humans.This paper was presented at a symposium entitled Genetic Control of Mammalian Metabolism held at The Jackson Laboratory, Bar Harbor, Maine, June 30–July 2, 1969. Mechanisms are discussed by which several radiation-induced lethal mutations in the mouse control enzyme activity and morphogenesis. These mutations behave as alleles at the albino locus in chromosome I; furthermore, all of them suppress glucose 6-phosphatase activity, resulting in perinatal death of albino homozygotes. Additional pleiotropic effects, and the absence of a dosage effect in heterozygotes, are not easily explained by a mutational change in the structural gene for glucose 6-phosphatase. The mutations might affect membrane proteins and thus render membrane structure abnormal; such membranes may lack the ability to bind tyrosinase as well as glucose 6-phosphatase and may also be responsible for morphogenetic abnormalities.This paper was presented at a symposium entitled Genetic Control of Mammalian Metabolism held at The Jackson Laboratory, Bar Harbor, Maine, June 30–July 2, 1969. Studies on the genetic mechanisms in control of mouse liver tyrosine aminotransferase expression were of three general types: (1) studies on strain variance in endogenous enzyme activity and of various factors affecting the basal enzyme level, (2) purification of the enzyme and studies of its properties, and (3) studies of strain variance in enzyme regulation dealing primarily with glucocorticoid induction and with the starvation-induced enzyme adaptation. Tyrosine aminotransferase (l-tyrosine: 2-oxoglutarate aminotransferase, E.C. 2.6.1.5) was purified 400 to 600-fold from livers of C57BL/6J and DBA/2J inbred mice. Several of the properties of the mouse liver enzyme were similar to those known for the rat liver enzyme although the apparent K m (l-tyrosine) was lower, calculated at 6.25×104 M. Disc gel electrophoresis of the enzyme from 105,000 g supernatant fluid after induction by hydrocortisone indicated three bands of enzyme activity with strain variance in electrophoretic mobility between the C57BL/6J and DBA/2J mice. The administration of glucose to fasting C57BL/6J mice repressed the starvation-induced increase in enzyme activity, but did not prevent the hydrocortisone induction of enzyme activity. DEAE-cellulose chromatography of purified enzyme from fasting DBA/2J and C57BL/6J mice which had been labeled in vivo with C 14 -l-leucine revealed strain differences in the elution patterns for both enzyme activity and radioactivity. Two peaks of enzyme activity were detected in the enzyme preparations from fasting mice. The marked strain variance in the enzyme activity and the quantity of radioactivity associated with the first enzyme peak may indicate differential rates of protein turnover for different isozymic forms of tyrosine aminotransferase. Flumethasone, a potent difluoro synthetic glucocorticoid, was used in studies on the hormonal regulation of tyrosine aminotransferase in obese mutant mice of the C57BL/6J-ob strain. Two electrophoretically distinct components of glucuronidase have been resolved in extracts of murine liver. A mutation which results in a ten-fold reduction in liver glucuronidase activity leads to a reduction in intensity of both components. Since the substrate dependencies of both mutant (gg) and wild-type (GG) glucuronidase have previously been shown to be identical, it is suggested that the mutation results in a decreased concentration of liver glucuronidase molecules. Furthermore, the anodal electrophoretic component is shown to be preferentially localized in lysosomes, while the cathodal component is primarily microsomal. Summary Simple pseudo-3D modifications to the constant-time HSQC and HCACO experiments are described that allow accurate (±0.5 Hz) measurement of one bond JCH coupling constants in proteins that are uniformly enriched with 13C. An empirical ,-surface is calculated which describes the deviation of 1JCH from its random coil value, using 203 1JCH values measured for residues in the proteins calmodulin, staphylococcal nuclease, and basic pancreatic trypsin inhibitor, for which and are know with good precision from previous X-ray crystallographic studies. Residues in -helical conformation exhibit positive deviations of 4–5 Hz, whereas deviations in -sheet are small and, on average, slightly negative. Data indicate that 1JCH depends primarily on , and that 1JCH may be useful as a qualitative probe for secondary structure. Comparison of 1JCH coupling constants measured in free calmodulin and in its complex with a 26-aminoacid peptide fragment of myosin light-chain kinase confirm that the calmodulin secondary structure is retained upon complexation but that disruption of the middle part of the central helix is even more extensive than in free calmodulin. A biocultural study of the side effects of chronic lithium treatment among 70 Hong Kong Chinese psychiatric patients, using a self-report 33-item checklist and semi-structured interviews, revealed an imperfect correspondence between biomedically prescribed and culturally endorsed psychotropic side effects. Although polidipsia and polyuria (47%) were the biomedically most real side effects, they were not usually regarded as bothersome or translated into metaphors to express undesirable side effects.Complaints such as tiredness (38%), drowsiness (36%) and poor memory (31%) were also common but their frequency was significantly lower than that of normal control subjects. The item loss of creativity had no conceptual equivalent in Chinese and was usually misinterpreted. As no patient was aware that lithium was a metal, the side effect metallic taste was variously labelled. Contrary to Western findings, complaints of missing of highs, loss of assertiveness and fear of weight gain were rarely encountered. Active elicitation was required for indigenous complaints, with 38% of patients considering lithium to cause mild hotness. This was readily neutralized by drinking more water which had a cooling effect. Expectedly, concurrent neuroleptics and antidepressants amplified most lithium side effects.This study affirms Western data on the biomedically universalizable effects of chronic lithium treatment, but also supports the thesis that culturally constituted cognitive styles affect patients' recognition, labelling, experiencing and reporting the total drug effect. Further, it demonstrates that the lived experience and clinical negotiation of lithium associated side effects reproduce, authenticate, and at times critique, core cultural and moral premises of Western and Chinese societies.  相似文献

The genetic locus for alleles (+k and k) that determine the presence and absence of the muscle enzyme, phosphorylase kinase, has been located on the X chromosome of the mouse. The inheritance of glycogen content in resting skeletal muscle follows the Mendelian pattern, and the genes which determine it must also be sex-linked. Evidence is presented which strongly suggests that one of the major determinants of glycogen concentration is phosphorylase kinase; inverse correlations of the enzyme and glycogen were found during neonatal development and among hybrid females, where content of phosphorylase kinase in muscle is highly variable. This variability in kinase content also determines the degree of the epinephrine effect (formation of phosphorylase a) in these hybrid females. The hybrid mice (F1, F2, and first backcross) were obtained from crosses of I/FnLn and C57BL/FnLn mice. Radiolabeled human 2-microglobulin (2m) can bind to mouse histocompatibility (H-2) antigens on the cell surface or to partially purified H-2 antigens in solution. The complexes containing human 2m and H-2 antigens from C3H (H-2k) mice could be immunoprecipitated specifically with alloantisera, rabbit anti-H-2 xenoantisera, and with monoclonal H-2-specific antibodies. Specific association with H-2 antigens was also observed with other haplotypes. The only exception was B10.D2 (H-2 d ) from which complexes containing human 2M could only be precipitated with anti-H-2 xenosera. Thus radiolabeled human 2M can be used as a specific label for mouse H-2 antigens in precipitation and radioimmunoassays. Vacuolar processing enzymes (VPEs) are responsible for the maturation of seed proteins. These processing enzymes belong to a novel group of cysteine proteinases with molecular masses of 37 to 39 kDa. We isolated two genes of VPEs from a genomic library of Arabidopsis. The gene products were designated -VPE and -VPE, and they were 56% identical in terms of amino acid sequence. The amino acid sequences of -VPE and -VPE were also 55% and 67% identical to that of castor bean VPE, respectively. The gene for -VPE had 7 introns, while that of -VPE had 8 introns. Northern blot analysis revealed that -VPE is expressed in rosette leaves, cauline leaves and stems of Arabidopsis, while -VPE is predominantly expressed in the flowers and buds. Neither -VPE nor -VPE is expressed in the siliques. This result strongly suggests that the isolated genes encode isozymes of VPE that are specific to vegetative organs.  相似文献

Barrier-raised transforming growth factor 1 (TGF1)-deficient mice consistently die before 35 days of age of a severe multiorgan inflammatory disease that can affect the skeletal muscle, heart, liver, pancreas, salivary gland, lung, oesophagus and stomach. The underlying cause of this disease is not known. To determine whether abnormal responsiveness of the immune system to the presence of enteric flora plays a causative role, a colony of TGF1-deficient and wild-type mice were raised in a sterile environment. Seven germ-free TGF1-deficient and 5 germ-free TGF1 wild-type mice were examined. Lesion development was analysed and compared with historical data on 50 barrier-raised TGF1 mutant mice and 32 barrier-raised wild-type mice. All germ-free TGF1-deficient mice died shortly after weaning, as do their barrier-raised counterparts. There was a significant delay in death in germ-free TGF1-deficient mice compared with barrier-raised mutant mice. However, there was no difference in the type, severity or incidence of lesions between TGF1 mutant mice raised under germ-free or barrier conditions. Germ- free wild-type mice had no lesions. Summary A procedure is described which simplifies the classification of skeletal muscle fibres in that it allows a simultaneous evaluation of both the oxidative capacity and the intensity of reversed ATPase of the fibres, and thus enables to distinguish three fibre types — SO, FOG and FG — in one tissue section. After preincubation at pH 4.1–4.2 the cryostat section is incubated for succinate dehydrogenase (SDH) and subsequently for reversed-ATPase. This is followed by the fixation with neutral buffered formaldehyde. The results of typing of chicken, minipig and rabbit fibres in a single muscle section stained with this technique are identical to those obtained with the usual method based on a comparison of serial sections of which one is stained for SDH activity the other for reversed-ATPase activity.  相似文献

Monoclonal antibodies (mAb) were prepared against conjugated transforming growth factor 1 (TGF1) peptides: amino acid positions 48–60 and positions 86–101. Two antibodies, mAb 16-3G1 [anti-(48–60)] and mAb 5-2G6 [anti-(86–101)] cross-reacted with native TGF1,-2 and-3 (16-3G1) or only with native TGF1 (5-2G6). Both mAb were used to characterize TGF-mediated effects on the metastatic potential in nude mice of human carcinoma cell line SLU-1 and its metastatic subline SLU-M1. Autocrine TGF1-mediated up-regulation of cell proliferation and its suppression by anti-TGF antibodies in vitro was recorded for SLU-M1 cells whereas SLU-1 cell proliferation in vitro appeared to be refractory to anti-TGF antibodies and exogenous TGF-1. However, the potential of s.c. tumours to develop distant metastases in nude mice was about the same for both cell lines. Development of primary tumours and distant metastases could be suppressed by treatment of mice with anti-TGF antibodies. Thus we assume that the metastatic potential of tumour cells is independent of TGF-mediated growth-regulation effects in vitro. The anti-TGF-induced suppression of tumour progression and metastasis in nude mice might rather result from stimulation of the immune surveillance. TGF-mediated autocrine down-regulation of MHC-unrestricted cytotoxicity of activated human monocytes and CD56+ LAK cells and its reversion by anti-TGF antibodies could be readily demonstrated. In all our experimental series, the neutralizing potential of both anti-TGF antibodies, though directed against opposite sites of the TGF1 molecule, was very similar.  相似文献

Sickle cell anaemia (SCA) exhibits significant variations in clinical presentation in different populations for which several genetic factors including SCA-associated -and -thalassaemias, G-6-PD deficiency and elevated Hb F level have been implicated as possible ameliorating factors. Saudi Arabia is unique in that mild and severe forms of the disease occur at a high frequency. We investigated the G/A ratio and Hb F level and correlated these values with the severity of SCA. The results showed that Hb F level varies significantly in both groups of patients with no evident correlation with the mild clinical manifestations. However, G/A ratio correlated significantly with the disease severity where a high ratio was observed in patients with the mild and a low ratio in patients with the severe disease. The results are evaluated and discussed in the light of correlation studies and regression analysis.  相似文献

Summary. We investigated the influence of hypothyroidism on homocysteine metabolism in rats, focusing on a hypothetical deficient synthesis of FAD by riboflavin kinases. Animals were allocated in control group (n=7), thyroidectomized rats (n=6), rats with diet deficient in vitamin B2, B9, B12, choline and methionine (n=7), thyroidectomized rats with deficient diet (n=9). Homocysteine was decreased in operated rats (2.6±1.01 vs. 4.05±1.0mol/L, P=0.02) and increased in deficient diet rats (29.56±4.52 vs. 4.05±1.0mol/L, P=0.001), when compared to control group. Erythrocyte-Glutathione-Reductase-Activation-Coefficient (index of FAD deficiency) was increased in thyroidectomized or deficient diet rats (P=0.004 for both). Methylenetetrahydrofolate-reductase and methionine-synthase activities were decreased in thyroidectomized rats but not in those subjected to deficient diet. Cystathionine--synthase was increased only in operated rats. Taken together, these results showed a defective re-methylation in surgical hypothyroidism, which was due in part to a defective synthesis of vitamin B2 coenzymes. This defective pathway was overcompensated by the increased Cystathionine--synthase activity.  相似文献

The seasonal fluctuations in numbers, sex ratios and the changes in the reproductive status of the Mexican fruit fly, Anastrepha ludens, caught in McPhail traps in citrus orchards and the effect of control measures on them, were examined in Belize, Central America. Evidence is presented that the seasonal increase in numbers was derived mainly from infestations in grapefruit and that harvesting played a significant part in controlling numbers by removing infested fruit. In orchards where sprays were not used to control the fruit fly, the sex ratio and the proportion of fecund females were very variable but there were always greater numbers of females — often two to four times as many. During spray programs the sex ratio approached unity and the proportion of fecund females decreased to zero. The sex ratio therefore gave an indication of the mean reproductive status of the female flies in the traps and thus the capacity of a local population to initiate infestations as well as the efficiency of control measures used.
Résumé Effectifs, taux sexuel, périodes de reproduction, ainsi que leurs variations au cours de l'année et après application de mesures de lutte ont été étudiés chez la mouche des fruits mexicaine A. ludens dans des vergers d'agrumes de Belize (Amérique centrale), à l'aide de pièges McPhail.Les effectifs d'A. ludens sont caractérisées par une progression régulière à partir d'un minimum entre juin et septembre, jusqu'à un maximum en février ou mars. Ces variations saisonnières dépendent de la présence de fruits mûrs, de pamplemousses notamment. Le taux sexuel des adultes capturés dans des vergers non traités est très variable, mais les sont toujours plus nombreuses (souvent 2 à 4 fois plus) que les mâles. Pendant la saison des pamplemousses, les peuvent pondre dans les fruits mûrs qui abondent; si la proportion de augmente, celle des fécondes diminue. Après un traitement efficace à l'aide d'un attractif insecticide à base d'hydrolisat de protéines, le taux sexuel tend vers 1 : 1, les fécondes tendant à disparaître et la majorité des caputées étant fraîchement émergées et immatures.Pendant la saison des pamplemousses, le taux sexuel donne des indications sur le potentiel reproducteur des , sur leur aptitude à provoquer une infestation, ainsi que sur l'efficacité et la persistance des attractifs insecticides utilisés pour la lutte contre la mouche des fruits.

Summary The tryptic peptides from the A and B chains of cat hemoglobins A and B have been isolated and the amino acid compositions determined. Differences between the two chains were found in two peptides,T-1 (GlySer) andT-14 (AsnSer and LysArg). The GlySer and LysArg substitutions are placed at-1 and-144 respectively from earlier work, and the third substitution, AsnSer at-139 is suggested from this work. In addition, the presence of a blocked amino terminus in B has been confirmed. Tentative sequences constructed by homology with known-chain structures suggest the occurrence of substitutions at 1 1 contacts in A and B that may be functionally significant. There are at least 18 differences in amino acid composition between cat A and dog-chains and 22 differences between cat A and normal adult human-chains.  相似文献

Summary The incidence of myosatellite cells associated with white and red muscle fibres of the parietal muscle and red fibres of the craniovelar muscle was estimated by quantitative electron microscopy in the Atlantic hagfish (Myxine glutinosa, L.). Myosatellite cell nuclei constitute 3, 11 and 23 % of the total number of nuclei inside the basal lamina of the three types of muscle fibres, respectively. However, the total number of nuclei is highest in white fibres, most of the nuclei belonging to striated muscle cells. Myosatellite cell profiles in transverse sections constitute 23, 41 and 61 % of the number of muscle fibre profiles of the three types, respectively. The intervals between adjacent myosatellite cells are 135 m in white fibres, 55 m in red parietal fibres, and only 25 m in craniovelar fibres. Since craniovelar fibres are also comparatively thin, myosatellite cells constitute a significant fraction of the volume inside the basal lamina in these fibres. The myosatellite cells are 30–50 m long and up to 5 m thick. Some myosatellite cells possess few organelles, whereas others appear to contain many free ribosomes, granular endoplasmic reticulum, prominent Golgi apparatus and lysosome-like bodies.This investigation was supported by the Norwegian Research Council for Science and the Humanities (NAVF grant No. C20.30–37). The primary structure of adult marmoset hemoglobin has been determined. The - and -chains of HbA were separated on a CM23 column in 8 M urea using a sodium phosphate gradient. Tryptic digests of the - and -chains were fractionated on a Dowex 50W-X2 column using a pH and pyridine acetate gradient. Large peptide fragments were obtained by the cyanogen bromide cleavage of the - and -chains, as well as by tryptic digestion of the maleylated - and -chains. The sequence was derived from the amino acid compositions and sequences of the individual tryptic peptide, automated sequence determination of intact - and -chains, as well as automated sequence determination of cyanogen bromide fragments and tryptic maleylated peptides derived from the - and -chains. The complete structure of marmoset adult hemoglobin is closely homologous to that of other primate hemoglobins. The sequence of the marmoset -chain differs from the -chain of human HbA at positions 8, 19, 23, 68, and 116. An examination of three inbred strains of mice differing with respect to liver and kidney catalase activity reveals two distinct genetic factors controlling the level of liver catalase activity. The first genetic factor controls the catalytic activity of the enzyme. Specific activity of purified enzyme from C57BL/6 and C57BL/Ha strains is 60% of that of the DBA/2 strain. The second factor controls the content of liver catalase. Liver catalase of C57BL/Ha is degraded in vivo at a rate one half that of liver catalase of DBA/2 and C57BL/6, resulting in the accumulation of twice as many catalase molecules in C57BL/Ha. The factor affecting turnover of catalase is apparently specific for catalase of liver since no differences exist in kidney catalase levels between C57BL/Ha and C57BL/6. Furthermore, this factor does not appear to alter the metabolism of total liver protein since no substantial difference in the turnover rate of liver protein is observed among the strains. It is particularly significant that the genetic factor affecting the amount of liver catalase does so by altering the rate of catalase degradation rather than the rate of synthesis, confirming the previously published report of Rechcigl and Heston (1967).
