Phosphate uptake kinetics of Synechococcus sp. WH7803 and Thalassiosiraweissflogii were studied in axenic batch culture. Phosphate-repleteSynechococcus sp. WH7803 cells have a lower affinity for inorganicphosphate (Pi) (Ks = 67 µmol l–1) than Pi-starvedcells (Ks = 3.1 µmol l–1). The Ks of Pi-starvedcells increased  相似文献

Surveys were conducted along the northeast coast of the USA.between Portsmouth, NH, and the Chesapeake Bay in 1988 and 1990,to determine the population distribution of Aureococcus anophagefferens, the chrysophyte responsible for massive and destructive ‘browntides’ in Long Island and Narragansett Bay beginning in1985. A species-specific immunofluorescent technique was usedto screen water samples, with positive identification possibleat cell concentrations as low as 10–20 cells ml–1.Both years.A.anophagefferens was detected at numerous stationsin and around Long Island and Barnegat Bay, NJ, typically athigh cell concentrations. To the north and south of thus ‘center’,nearly half of the remaining stations were positive for A.anophagefferens, but the cells were always at very low cell concentrations. Manyof the positive identifications in areas distant from Long Islandwere in waters with no known history of harmful brown tides. The species was present in both open coastal and estuanne locations,in salinities between 18 and 32 practical salinity units (PSU).The observed population distributions apparently still reflectthe massive 1985 outbreak when this species first bloomed, giventhe number of positive locations and high abundance of A.anophagefferensin the immediate vicinity of Long Island. However, the frequentoccurrence of this species in waters far from this population‘center’ is disturbing. Aureococcus anophagefferensis more widely distributed than was previously thought. Numerousareas thus have the potential for destructive brown tides suchas those associated with the sudden appearance of the speciesin 1985.  相似文献

Phycoerythrin-containing Synechococcus species are considered to be major primary producers in nutrient-limited gyres of subtropical and tropical oceanic provinces, and the cyanophages that infect them are thought to influence marine biogeochemical cycles. This study begins an examination of the effects of nutrient limitation on the dynamics of cyanophage/Synechococcus interactions in oligotrophic environments by analyzing the infection kinetics of cyanophage strain S-PM2 (Cyanomyoviridae isolated from coastal water off Plymouth, UK) propagated on Synechococcus sp. WH7803 grown in either phosphate-deplete or phosphate-replete conditions. When the growth of Synechococcus sp. WH7803 in phosphate-deplete medium was followed after infection with cyanophage, an 18-h delay in cell lysis was observed when compared to a phosphate-replete control. Synechococcus sp. WH7803 cultures grown at two different rates (in the same nutritional conditions) both lysed 24 h postinfection, ruling out growth rate itself as a factor in the delay of cell lysis. One-step growth kinetics of S-PM2 propagated on host Synechococcus sp. WH7803, grown in phosphate-deplete and-replete media, revealed an apparent 80% decrease in burst size in phosphate-deplete growth conditions, but phage adsorption kinetics ofS-PM2 under these conditions showed no differences. These results suggested that the cyanophages established lysogeny in response to phosphate-deplete growth of host cells. This suggestion was supported by comparison of the proportion of infected cells that lysed under phosphate-replete and-deplete conditions, which revealed that only 9.3% of phosphate-deplete infected cells lysed in contrast to 100% of infected phosphate-replete cells. Further studies with two independent cyanophage strains also revealed that only approximately 10% of infected phosphate-deplete host cells released progeny cyanophages. These data strongly support the concept that the phosphate status of the Synechococcus cell will have a profound effect on the eventual outcome of phage-host interactions and will therefore exert a similarly extensive effect on the dynamics of carbon flow in the marine environment.  相似文献

Despite the increasing knowledge of Synechococcus spp. and their co-occurring cyanophages in oceanic and coastal water, little is known about their abundance, distribution, and interactions in the Chesapeake Bay estuarine ecosystem. A 5-year interannual survey shows that Synechococcus spp. and their phages are persistent and abundant members of Chesapeake Bay microbial communities. Synechococcus blooms (106 cells ml−1) were often observed in summer throughout the Bay, contributing 20 to 40% of total phytoplankton chlorophyll a. The distribution of phycoerythrin-containing (PE-rich) Synechococcus cells appeared to mostly correlate with the salinity gradient, with higher abundances at higher salinities. Cyanophages infectious to Synechococcus were also abundant (up to 6 × 105 viruses ml−1 by the most probable number assay) during summer months in the Bay. The covariation in abundance of Synechococcus spp. and cyanophages was evident, although the latitude of observed positive correlation varied in different years, mirroring the changing environmental conditions and therefore the host-virus interactions. The impacts of cyanophages on host Synechococcus populations also varied spatially and temporally. Higher phage-related Synechococcus mortality was observed in drought years. Virus-mediated host mortality and subsequent liberation of dissolved organic matter (DOM) may substantially influence oceanic biogeochemical processing through the microbial loop as well as the microbial carbon pump. These observations emphasize the influence of environmental gradients on natural Synechococcus spp. and their phage population dynamics in the estuarine ecosystem.  相似文献

Synechococcus species are important primary producers in coastal and open‐ocean ecosystems. When nitrate was provided as the sole nitrogen source, nickel starvation inhibited the growth of strains WH8102 and WH7803, while it had little effect on two euryhaline strains, WH5701 and PCC 7002. Nickel was required for the acclimation of Synechococcus WH7803 to low iron and high light. In WH8102 and WH7803, nickel starvation decreased the linear electron transport activity, slowed down QA reoxidation, but increased the connectivity factor between individual photosynthetic units. Under such conditions, the reduction of their intersystem electron transport chains was expected to increase, and their cyclic electron transport around PSI would be favored. Nickel starvation decreased the total superoxide dismutase (SOD) activity of WH8102 and WH7803 by 30% and 15% of the control, respectively. The protein‐bound 63Ni of the oceanic strain WH8102 comigrated with SOD activity on nondenaturing gels and thus provided additional evidence for the existence of active NiSOD in Synechococcus WH8102. In WH7803, it seems likely that nickel starvation affected other metabolic pathways and thus indirectly affected the total SOD activity.  相似文献

The temporal and spatial variability in the quality and quantityof settling phytoplankton material in relation to concurrentprimary production was studied using sediment traps at threecoastal stations from a semi-enclosed bay (Pojo Bay) throughthe outer archipelago to the open Gulf of Finland. The fluxof settling phytoplankton was high (9.3 g C m–2period–1)in Pojo Bay, especially in spring, and lower in the archipelago(8.1 g C m–2 period–1) and open-sea area (5.2 gC m"2 period"1), although the primary production followed theopposite pattern. A large influx of allochthonous material intoPojo Bay in spring brought allochthonous phytoplankton cellsinto the traps, but limited primary production. Diatoms werethe most abundant settled phytoplankton at all stations, butthe species composition varied between Pojo Bay (Aulacoseiraspp., Rhizosolenia minima) and the outer stations (Skeletonemacostatum, Chaetoceros spp.)At the outer stations, migratingdinoflagellates (Peridiniella catenate) comprised part of thesettling material in spring. The high settling flux of the cyanophyteAphanizomenon flos-aquae is discussed. The species compositionof the phytoplankton assemblage influenced the proportion ofthe total organic carbon sedimentation that consisted of phytoplanktoncarbon.  相似文献

Seasonal variation in the phylogenetic composition of Synechococcus assemblages in estuarine and coastal waters of Hong Kong was examined through pyrosequencing of the rpoC1 gene. Sixteen samples were collected in 2009 from two stations representing estuarine and ocean-influenced coastal waters, respectively. Synechococcus abundance in coastal waters gradually increased from 3.6 × 103 cells ml−1 in March, reaching a peak value of 5.7 × 105 cells ml−1 in July, and then gradually decreased to 9.3 × 103 cells ml−1 in December. The changes in Synechococcus abundance in estuarine waters followed a pattern similar to that in coastal waters, whereas its composition shifted from being dominated by phycoerythrin-rich (PE-type) strains in winter to phycocyanin-only (PC-type) strains in summer owing to the increase in freshwater discharge from the Pearl River and higher water temperature. The high abundance of PC-type Synechococcus was composed of subcluster 5.2 marine Synechococcus, freshwater Synechococcus (F-PC), and Cyanobium. The Synechococcus assemblage in the coastal waters, on the other hand, was dominated by marine PE-type Synechococcus, with subcluster 5.1 clades II and VI as the major lineages from April to September, when the summer monsoon prevailed. Besides these two clades, clade III cooccurred with clade V at relatively high abundance in summer. During winter, the Synechococcus assemblage compositions at the two sites were similar and were dominated by subcluster 5.1 clades II and IX and an undescribed clade (represented by Synechococcus sp. strain miyav). Clade IX Synechococcus was a relatively ubiquitous PE-type Synechococcus found at both sites, and our study demonstrates that some strains of the clade have the ability to deal with large variation of salinity in subtropical estuarine environments. Our study suggests that changes in seawater temperature and salinity caused by the seasonal variation of monsoonal forcing are two major determinants of the community composition and abundance of Synechococcus assemblages in Hong Kong waters.  相似文献

The population abundances and rates of biomass production ofheterotrophic nanoplankton (HNAN) in Georgia coastal waterswere evaluated by epifluorescence microscopy. HNAN populations(mostly non-pigmented microflagellates <10 µm in diameter)ranged from 0.3 x 103 cells ml–1 in shelf waters 15 kmoffshore to 6.3 x 103 cells ml–1 in waters 0.25 km fromthe coast. There was a strong correlation (r = 0.83) betweenHNAN and free bacterioplankton population abundances, but noapparent relation (r = 0.38) between HNAN and phototrophic nanopLankton(PNAN) abundances. HNAN biomass production in estuarine andnearshore shelf waters, as estimated from increases in HNANpopulations during laboratory incubations of natural water samples, ranged from 0.10 to 0.79 mg C m–3 h–3, with populationgeneration times of 9.7 to 26.5 h. There was a significant linearrelation (r = 0.95) between HNAN biomass and HNAN productivity. We calculated that HNAN may graze at least 30% to 50% of dailybacterioplankton production in Georgia coastal waters.  相似文献

Grazing by microzooplankton on autotrophic and heterotrophicpicoplankton as well as >0.7 µm phytoplankton (as measuredby chlorophyll a) was quantified during July, August, October,January and April in the surface layer of Logy Bay, Newfoundland(47°38'14'N, 52°39'36'W). Rates of growth and grazingmortality of bacteria, Synechococcus and >0.7 µm phytoplanktonwere measured using the sea water dilution technique. Microzooplanktoningested 83–184, 96–366 and 64–118% of bacterial,Synechococcus and >0.7 µm phytoplankton daily potentialproduction, respectively and 34–111, 25–30 and 16–131%of bacterial, Synechococcus and >0.7 µm phytoplanktonstanding stocks, respectively. The trends in prey net growthrates followed the seasonal cycles of prey biomass, suggestingthat microzooplankton are important grazers in Newfoundlandcoastal waters. Ingestion was lowest during January and October(~2 µg C l–1 day–1) and highest in August(~20 µg C l–1 day–1). Aside from April when>0.7 µm phytoplankton represented the majority (~80%)of carbon ingested, bacterioplankton and <1 µm phytoplanktonrepresented most of the carbon ingested (~40–100%). Althoughmicrozooplankton have here-to-fore been unrecognized as an importantgrazer population in Newfoundland coastal waters, these resultssuggest that they play an important role in carbon flow withinthe pelagic food web, even at low temperatures in Logy Bay.  相似文献

Relatively warm (12.50–12.75°C) and high-salinity[<35.640 practical salinity units (PSU)] water flowing eastwardwas detected at the shelf-break during a cruise carried Outin the southern Bay of Biscay in Spring 1987. The slope currentinduced the formation of a convergent front separating well-mixedoceanic waters from haline-stratified coastal waters. Very highconcentrations of dissolved oxygen (295 µmol kg–1)and chlorophyll a(>4.5 mg m–3) were found at the outeredge of the frontal boundary. Small autotrophic flagellatesdominated the phytoplankton community. Primary production peakedat the boundary region. Estimated phytoplankton growth ratesindicated that active growth was taking place, with lower turnovertimes integrated over the water column at the frontal station(2.5–5 days) than at coastal (1.5–2.8 days) or oceanic(1.5–3.5 days) stations. The lowest doubling times (1–2days) were calculated for surface frontal populations. Accumulationof zooplankton was also observed associated with the convergentphysical structure, although this relationship was less markedthan for phytoplankton. Copepods, mainly Paracalanus parvus,Acartia clausi and Oithona helgolandica, formed the bulk ofthe mesozooplankton biomass. Compatibility between the sizeof phytoplankton cells and copepod size spectra indicate highfood availability for these animals, particularly in the vicinityof the front. The distribution of fish eggs and fish larvaewas also coupled with the slope current-induced front. Sardinelarvae were more abundant at the coastal side of the front, whereas larval stages of blue whiting reached the highest densitiesat off-shelf stations. Larvae of lamellibranch molluscs andbryozoa were restricted to nearshore waters, as the frontalboundary prevented larval dispersion to the open ocean. Theresults presented in this paper suggest that the Iberian slopecurrent and its associated shelf-break frontal structure werecrucial in controlling phytoplankton primary production, activityof grazers, distribution of larvae of fishes and benthic invertebrates, and ultimately in determining the structure of the pelagic foodweb in the southern Bay of Biscay during the seasonal periodof vertical mixing.  相似文献

Tidal influences on appendicularian densities were observedat North Inlet, South Carolina, by sampling along a transectwhich ran from a tidal creek to a station 5 km offshore. Oikopleuradioica was the dominant species in North Inlet, while Oikopleuralongicauda and Appendicularia sicula contributed marginallyto appendicularian numbers during midsummer and fall. A strongtide-dependent density pattern was clear for inshore waters.Low-tide densities of all three species showed a dramatic increasein an offshore direction. At high tide, densities were similarbetween all stations for O. dioica, while O. longicauda andA. sicula showed a less pronounced density gradient than atlow tide. Population densities within the inlet were greateron spring tides than neap tides and tidal influences were generallyconsistent between seasons. Appendicularians enter the estuaryin densities as high as 20 072 animals m–3, indicatingthat tidal currents may be an important mechanism for exchangeof appendicularian biomass between coastal and estuarine waters. 1Present address: Allan Hancock Foundation, University of SouthernCalifornia, Los Angeles, CA 90007, USA.  相似文献

We measured egg production rates of the estuarine calanoid copepodAcartia tonsa in Mobile Bay, an estuary in the northern Gulfof Mexico. Two stations were sampled approximately monthly, one at the mouth of the bay and the other just beyond the mouthin the salinity front between bay and coastal waters. Over thewhole year, temperature was the most important environmentalvariable controlling egg production. Rates increased with temperatureup to 30°C and 140 eggs female–1 day–1. We foundno evidence of food limitation. There was no correlation betweenegg production and phytoplankton abundance, nor increased eggproduction in response to supplements of phytoplankton addedto natural food, suggesting that non-phytoplankton food wasimportant in the diet. At the highest egg production rates, the amount of the storage lipid triacylglycerol (TAG) in adultfemales was greatly diminished, to <50 ng female–1.This suggests that lipids in the diet can be very tightly coupledto egg production. Both egg production and TAG content of femalesshowed significant variability on spatial scales of 3–15km, especially in relation to the salinity front separat ingwater outwelling from the bay from open coastal water. For organismsthat are using copepods and their eggs as food, this variabilitywould result in a heterogeneous food environment, both in termsof the abundance of food and its nutritional content.  相似文献

The composition and positional distribution of fatty acids inmonogalactosyldiacylglycerol, digalactosyldiacylglycerol, phosphatidylglyceroland sulphoquinovosyldiacylglycerol from two cyanobacteria, Anacystisnidulans and Synechococcus sp. grown at 25°C have been determinedand compared with measurements of the phase separation temperaturesof the lipids. Only monogalactosyldiacylglycerol in Anacystisand sulphoquinovosyldiacylglycerol in Synechococcus showed phaseseparation temperatures above 0°C. The phase transitiontemperature of a sample of sulphoquinovosyldiacylglycerol containingover 90% of the dihexadecanoyl molecular species has been determinedto be 43°C for the Na+ salt and 38°C for the Mg++ salt. *Deceased. We present a numerical model of nutrient uptake and photosynthesisduring migrations of the marine diatom Rhizosolenia that wasdeveloped to estimate fluxes of carbon and nitrogen due to thesemigrations in the open ocean. The predicted specific rate ofincrease of Rhizosolenia was 0.11–0.15 day1, whereas thetotal time for one migration cycle ranged between 3 and 5 days.Using published estimates of Rhizosolenia abundance, we estimatethat new primary production due to Rhizosolenia migrations rangesbetween 0.018 and 0.033 mmol N m–2 day–1. Thesevalues represent up to 17% of new production due to turbulentdiffusive fluxes of nitrate into the euphotic zone and are ofthe same order of magnitude as new production due to nitrogenfixation in tropical oceans. Large-scale contributions of Rhizosoleniato oceanic new production are limited by their relatively lowstanding crop. Variations in the formulation of losses withdepth greatly affected gross and net fluxes of carbon and nitrogen.Better characterization of losses of Rhizosolenia and improvedestimates of its abundance will help determine more accuratelythe contributions of Rhizosolenia to global biogeochemical cycles. 1Present address: Department of Agricultural and EnvironmentalScience, The Queen's University of Belfast New forge Lane, BelfastBT95PX, UK  相似文献

Buoyancy properties of the giant diatom Ethmodiscus   总被引:2,自引:0,他引:2
Net-collected Ethmodiscus spp. were examined and found to becapable of positive buoyancy with ascent rates averaging 1.4m h–1 on individually measured cells, and 0.3 m h–1using a homogeneous settling technique. These data, togetherwith published reports of positive buoyancy in Rhizosolenia, indicate that upward movement is a widespread phenomenon inlarge oceanic diatoms. These observations also suggest thatpublished size-rate relationships for marine diatoms requirerevision to include the effects of positive buoyancy.  相似文献

Laboea sirobila Lohmann, a marine oligotrich ciliate, was presentin Long Island Sound, USA, from March through June 1984. Peakabundance occurred in mid-June, when it was estimated that 10.2x 106 individuals m–2 were present in the 37-m deep watercolumn. When preserved specimens were examined with epifluorescencemicroscopy, the photosynthetic pigments chlorophyll and phycoerythrinwere observed within chloroplasts inside the ciliate. Thesepigments were also present in Myrionecta (formerly Mesodinium)rubrum Jankowski, a photosynthetic ciliate which co-occurredwith L. strobila during the present study. Vertical profilesshowed both ciliates to be most abundant in the upper few metersof the water column, especially during periods of strong thermalstratification.  相似文献

In two contrasting regions off the coast of Brazil, picoplankton(<1 µm) responses to removal of larger grazers andto the additions of glucose and amino acids were determined.Effects of glucose and amino acid additions (1 µM) onparticulate nitrogen and chlorophyll a concentrations, and onrates of NH4+ uptake and regeneration, were observed after 5h pre-incubation. In the oceanic waters, removal of the >1µm fraction had no significant effect on the chlorophylla of the picoplankton after 5 h. However, the addition of glucosestimulated both uptake and regeneration by a mean of 27%, andthe addition of arginine led to significant decreases in therates of NH4+ uptake and regeneration. In contrast, in the coastalwaters, significant increases in chlorophyll and particulatenitrogen concentrations were found after 5 h incubation in boththe amended samples and in the controls, and mean rates of NH4+uptake and regeneration were affected to a lesser degree bythe additions of either glucose or amino acids than in the oceanicwaters. The oceanic responses were suggestive of carbon limitationof heterotrophic bacteria. In the coastal region, on the otherhand, the supply of organic carbon and nitrogen was likely tohave been sufficient to meet the nutritional requirements ofthe heterotrophic bacteria and cyanobacteria. Grazing by largerorganisms on the picoplankton appeared to play a more significantrole in the nitrogen cycle in the coastal waters than in theoceanic waters.  相似文献

Seasonal variations in diversity and biomass of tintinnids (Ciliophora:Tintinnida) were investigated at two fixed stations in the innerpart of the Bahía Blanca Estuary (38°42' S, 61°50'W) during an annual cycle. The variations were analysed in relationto surface temperature, salinity, transparency, solar radiationand chlorophyll a (Chl a)concentration. Biomass was calculatedin terms of biovolume and carbon units. Diversity was estimatedas the number of species and the Shannon Index (H', ln based).Density of tintinnids ranged from 100 to 7800 individuals L–1H' ranged from 0 to 1.81. The biomass varied from 0.3 to 127.78x 106 µm3 L–1 (0.02–39.4 µg C L–1).Density was significantly related to temperature, solar radiationand Secchi distance (P < 0.01); diversity was significantlyrelated to temperature (P < 0.01) and solar radiation (P< 0.05). Biomass was significantly related only to temperature(P < 0.01) in one of the stations. According to principalcomponents analysis (PCA) tintinnids exhibited a segregationof three groups: winter, spring–summer and autumn forthe most internal station and winter, spring and summer–autumnfor the most external station. H' values were lower than thoseobserved in other coastal systems found at about the same latitudein the northern hemisphere.  相似文献
