This study was conducted to elucidate the effect of P fertilisation on the function of field communities of arbuscular mycorrhizal fungi (AMF) measured as P transport to flax. Two methods were applied to soil from a long-term field experiment with NaHCO3-extractable soil P levels of 24 and 50 mg kg-1in an experiment under controlled conditions: i) Measurement of plant growth and P uptake in the presence or absence of the fungicide benomyl and ii) measurement of hyphal P transport from a root-free compartment labelled with 32P. Benomyl successfully prevented mycorrhizal function. The absolute contribution of AMF to plant P uptake was of the same magnitude with or without P fertilisation at 27 days after sowing. Therefore, even though plants grown at the higher soil P level had greater P uptake, the relative contribution of AMF to P uptake was greater at the lower P level than at the higher P level (77 and 49% of total P uptake, respectively). The AMF in P-fertilized soil transported less P32 from the root-free compartment to the plant after 23 days than the AMF in unfertilized soil, but this difference disappeared in plants harvested after 27 and 32 days. The production of hyphae was largely similar in both fertilization treatments, indicating that the capacity for P uptake and transport by hyphae of the two AMF communities was similar. Aims

Arbuscular mycorrhizal fungi (AMF) can control root-knot nematode infection, but the mode of action is still unknown. We investigated the effects of AMF and mycorrhizal root exudates on the initial steps of Meloidogyne incognita infection, namely movement towards and penetration of tomato roots.

Methods

M. incognita soil migration and root penetration were evaluated in a twin-chamber set-up consisting of a control and mycorrhizal (Glomus mosseae) plant compartment (Solanum lycopersicum cv. Marmande) connected by a bridge. Penetration into control and mycorrhizal roots was also assessed when non-mycorrhizal or mycorrhizal root exudates were applied and nematode motility in the presence of the root exudates was tested in vitro.

Results

M. incognita penetration was significantly reduced in mycorrhizal roots compared to control roots. In the twin-chamber set-up, equal numbers of nematodes moved to both compartments, but the majority accumulated in the soil of the mycorrhizal plant compartment, while for the control plants the majority penetrated the roots. Application of mycorrhizal root exudates further reduced nematode penetration in mycorrhizal plants and temporarily paralyzed nematodes, compared with application of water or non-mycorrhizal root exudates.

Conclusions

Nematode penetration was reduced in mycorrhizal tomato roots and mycorrhizal root exudates probably contributed at least partially by affecting nematode motility.  相似文献

An investigation was carried out to test whether the mechanism of increased zinc (Zn) uptake by mycorrhizal plants is similar to that of increased phosphorus (P) acquisition. Maize (Zea mays L.) was grown in pots containing sterilised calcareous soil either inoculated with a mycorrhizal fungus Glomus mosseae (Nicol. and Gerd.) Gerdemann and Trappe or with a mixture of mycorrhizal fungi, or remaining non-inoculated as non-mycorrhizal control. The pots had three compartments, a central one for root growth and two outer ones for hyphal growth. The compartmentalization was done using a 30-m nylon net. The root compartment received low or high levels of P (50 or 100 mg kg–1 soil) in combination with low or high levels of P and micronutrients (2 or 10 mg kg–1 Fe, Zn and Cu) in the hyphal compartments.Mycorrhizal fungus inoculation did not influence shoot dry weight, but reduced root dry weight when low P levels were supplied to the root compartment. Irrespective of the P levels in the root compartment, shoots and roots of mycorrhizal plants had on average 95 and 115% higher P concentrations, and 164 and 22% higher Zn concentrations, respectively, compared to non-mycorrhizal plants. These higher concentrations could be attributed to a substantial translocation of P and Zn from hyphal compartments to the plant via the mycorrhizal hyphae. Mycorrhizal inoculation also enhanced copper concentration in roots (135%) but not in shoots. In contrast, manganese (Mn) concentrations in shoots and roots of mycorrhizal plants were distinctly lower, especially in plants inoculated with the mixture of mycorrhizal fungi.The results demonstrate that VA mycorrhizal hyphae uptake and translocation to the host is an important component of increased acquisition of P and Zn by mycorrhizal plants. The minimal hyphae contribution (delivery by the hyphae from the outer compartments) to the total plant acquisition ranged from 13 to 20% for P and from 16 to 25% for Zn.  相似文献

 Root colonization by arbuscular mycorrhizal fungi (AMF) may affect protein and lipid composition of plants by altering P nutrition or by eliciting other metabolic responses in the host plant. This study was conducted to determine the effects of an AMF and soil P on seed protein and lipid contents and yield of two genotypes of durum wheat (Triticum durum L.). Plants were grown in a greenhouse using soil: sand mixes with different levels of P, and with or without the AMF Glomus mosseae [(Nicol. and Gerd.) Gerd. and Trappe]. Percentage AMF root colonization decreased as P added to soil increased. The wheat genotype CR057 had higher AMF root colonization but lower seed P and protein concentrations than CR006. Without added soil P, protein concentration was significantly lower and lipid concentration and seed dry weight higher in arbuscular mycorrhizal (AM) than in nonAM plants. Seed lipid and protein contents were highly correlated with P content of plants. In nonAM plants, seed lipid and protein contents were low with no added soil P and did not differ with added soil P. Seed protein/lipid (Pro/L) concentration ratios of AM plants were higher than those of nonAM plants only when no P was added to the soil. The data indicate different patterns of seed P accumulation and different relationships between seed P and protein and lipid in AM and nonAM plants.  High salt levels in soil and water can limit agricultural production and land development in arid and semiarid regions. Arbuscular mycorrhizal fungi (AMF) have been shown to decrease plant yield losses in saline soils. The objective of this study was to examine the growth and mineral acquisition responses of greenhouse-grown tomato to colonization by the AMF Glomus mosseae [(Nicol. And Gerd.) Gerd. and Trappe] under varied levels of salt. NaCl was added to soil in the irrigation water to give an ECe of 1.4 (control), 4.7 (medium) and 7.4 dS m–1 (high salt stress). Plants were grown in a sterilized, low P (silty clay) soil-sand mix. Mycorrhizal colonization was higher in the control than in saline soil conditions. Shoot and root dry matter yields and leaf area were higher in mycorrhizal than in nonmycorrhizal plants. Total accumulation of P, Zn, Cu, and Fe was higher in mycorrhizal than in nonmycorrhizal plants under both control and medium salt stress conditions. Shoot Na concentrations were lower in mycorrhizal than in nonmycorrhizal plants grown under saline soil conditions. The improved growth and nutrient acquisition in tomato demonstrate the potential of AMF colonization for protecting plants against salt stress in arid and semiarid areas. Symbiotic mycorrhizal fungi play an important role in the absorption of soil nutrients and water by most plants. It has been suggested that hydraulically lifted water might maintain the integrity of the external mycorrhizal mycelium during drought. We tested this hypothesis in the obligately mycorrhizal species, coast live oak (Quercus agrifolia), using a microcosm system that separated the effects of hydraulic lift in roots from those in the external mycelium. Mycorrhizal oak seedlings were established in microcosms comprising three discrete compartments for (1) upper roots, (2) tap roots, and (3) external fungal mycelium. Eight months after planting, a drought treatment was initiated: irrigation to the upper root and fungal chambers was terminated and only irrigation to the taproot compartment was maintained. After 3, 12, 30, 50, 70 and 80 days of drought, tracers were injected into the taproot compartment at dusk. At dawn the following morning, mycorrhizal hyphae (EM and AM) and spores (AM) in upper root and fungal compartments were extensively labeled with the tracers. In contrast, no labeling was observed when tracers were injected into the taproot compartment during daytime. Nocturnal water translocation from plant to mycorrhizal fungi occurred in association with hydraulic lift. Saprotrophic/parasitic fungi in the microcosms were not labeled, suggesting a direct water transfer from plants to their mycorrhizal mutualists and not to other fungi in the soil. Even after prolonged drought (70-80 days), mycorrhizal hyphae persisted in soils with water potential values as low as -20 MPa. Maintaining mycorrhizal activity through direct water translocation could potentially improve the nutrient status of deep-rooted plants during periods when the fertile upper soil is dry.  相似文献

 Plant phosphorus uptake via external hyphae of arbuscular mycorrhizal fungi has been measured using compartmented systems where a hyphal compartment is separated from a rooting compartment by a fine mesh. By labelling the soil within the hyphal compartment with a radioactive phosphorus (P) isotope, hyphal uptake of P into the plant can be traced. The objective of this growth chamber study was to test two hyphal compartments of different design with respect to their suitabilities for measurement of hyphal P uptake. One hyphal compartment was simply a nylon mesh bag filled with 32P-labelled soil. The labelled soil in the other hyphal compartment was completely surrounded by an 8–10 mm layer of unlabelled soil that served as a buffer zone. Mycorrhizal and non-mycorrhizal subterranean clover plants were grown in pots with a centrally positioned hyphal compartment. Uptake of radioactive P by non-mycorrhizal control plants was 25% of that by mycorrhizal plants with the mesh bag but only 3% when including the buffer zone. Based on this good control of non-mycorrhizal P uptake from within the hyphal compartment and its greater ease of handling once produced, we judged the hyphal compartment including a buffer zone to be superior to the mesh bag. Sweet potato plants were grown with or without Glomus intraradices in split-root pots with adjacent root compartments containing a soil with a low availability of phosphate. One fungal tube, from which root growth was excluded, was inserted into each root compartment. During 4 weeks before harvest, the soil moisture level in either both or only one of the two root-compartments of each pot was decreased. Controls remained well watered. Low soil moisture generally had a negative effect on the amount of extraradical mycelium of G. intraradices extracted from the fungal tubes. Sporulation in the fungal tubes was much higher compared with the soil in the root compartment, but remained unaffected by the soil moisture regime. Concentrations of P in extraradical mycelium were much lower than usually found in plants and fungi, while P concentrations in associated mycorrhizal host plant tissues were in an optimum range. This suggests efficient transfer of P from the extraradical mycelium to the host plant. Despite the negative effect of a low soil moisture regime on extraradical G. intraradices development, the symbiosis indeed contributed significantly to P uptake of plants exposed to partial rootzone drying. The possibility that extraradical arbuscular mycorrhizal fungal development was limited by P availability under dry soil conditions is discussed.  相似文献

Although arbuscular mycorrhizal fungi (AMF) are known for their positive effect on flax growth, the impact of genetic manipulation in this crop on arbuscular mycorrhiza and plant performance was assessed for the first time. Five types of transgenic flax that were generated to improve fiber quality and resistance to pathogens, through increased levels of either phenylpropanoids (W92.40), glycosyltransferase (GT4, GT5), or PR2 beta-1,3-glucanase (B14) or produce polyhydroxybutyrate (M50), were used. Introduced genetic modifications did not change the degree of mycorrhizal colonization as compared to parent cultivars Linola and Nike. Arbuscules were well developed in each tested transgenic type (except M50). In two lines (W92.40 and B14), a higher abundance of arbuscules was observed when compared to control, untransformed flax plants. However, in some cases (W92.40, GT4, GT5, and B14 Md), the mycorrhizal dependency for biomass production of transgenic plants was slightly lower when compared to the original cultivars. No significant influence of mycorrhiza on the photosynthetic activity of transformed lines was found, but in most cases P concentration in mycorrhizal plants remained higher than in nonmycorrhizal ones. The transformed flax lines meet the demands for better quality of fiber and higher resistance to pathogens, without significantly influencing the interaction with AMF.  相似文献

We investigated uptake of Cd by arbuscular mycorrhizal (AM) maize inoculated with Glomus mosseae from a low-P sandy calcareous soil in two glasshouse experiments. Plants grew in pots containing two compartments, one for root and hyphal growth and one for hyphal development only. Three levels of Cd (0, 25 and 100 mg kg–1) and two of P (20 and 60 mg kg–1) were applied separately to the two compartments to assess hyphal uptake of Cd. Neither Cd nor P addition inhibited root colonization by the AM fungus, but Cd depressed plant biomass. Mycorrhizal colonization, P addition and increasing added Cd level led to lower Cd partitioning to the shoots. Plant P uptake was enhanced by mycorrhizal colonization at all Cd levels studied. When Cd was added to the plant compartment and P to the hyphal compartment, plant biomass increased with AM colonization and the mycorrhizal effect was more pronounced with increasing Cd addition. When P was added to the plant compartment and Cd to the hyphal compartment, plant biomass was little affected by AM colonization, but shoot Cd uptake was increased by colonization at the low Cd addition rate (25 mg kg–1) and lowered at the higher Cd rate (100 mg kg–1) but with no difference in root Cd uptake. These effects may have been due to immobilization of Cd by the fungal mycelium or effects of the AM fungus on rhizosphere physicochemical conditions and are discussed in relation to possible phytostabilization of contaminated sites by AM plants.  相似文献

以豆科植物紫花苜蓿为试验材料,应用三室(供体室-间隔室-受体室)培养系统,研究在供体和受体紫花苜蓿根系之间菌丝网络形成的时间效应以及间隔室中不同植物对菌丝网络建成的介导作用.第一个试验在供体和受体植物生长8、10、12、14周之后进行收获以检验菌丝网络形成的时间效应;第二个试验则在间隔室分别种植紫花苜蓿、羊草和独行菜,以考察菌根依赖性不同的植物对菌丝网络形成的介导作用.试验结果显示:(1)接种丛枝菌根真菌的供体紫花苜蓿根系能够形成良好的菌根共生,其外延菌丝可穿过尼龙网和间隔室侵染受体植物根系;植物生长8周后,在受体植物根系检测到菌根侵染,证实供体和受体植物间形成了根间菌丝网络;10周后,尽管供体室和受体室植物的侵染率已无差异,但二者的生物量和地上部磷浓度差异却加大,表现出菌丝网络对植物种内竞争影响的不对称性.(2)试验条件下,不同介导植物对受体植物的菌根侵染及生物量均无明显影响,但显著降低了供体植物生物量和地上部磷浓度;间隔室无介导植物或种植独行菜时,受体植物地上部和根系生物量显著低于供体植物,而当介导植物为紫花苜蓿和羊草时,受体和供体植物生物量无显著差异.研究表明,植物根间菌丝网络的形成受时间和介导植物的影响,同时也具有调节植物间资源分配和植物相互作用的功能.  相似文献

【背景】丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)能促进植物的养分吸收及生长发育。入侵中国华南地区的外来入侵植物南美蟛蜞菊(Wedelia trilobata)常分布于养分匮乏的荒地,却能迅速生长并排挤本地植物而快速扩张领地。【目的】探究丛枝菌根真菌是否促进南美蟛蜞菊的生长与竞争能力。【方法】采用南美蟛蜞菊及其同属本地植物蟛蜞菊(Wedelia chinensis)的盆栽对比控制试验,并设置接种及不接种AMF变形球囊霉(Glomus versiforme)、不同磷营养水平以及单种或混种的种植方式3种处理对两种植物的生长及竞争能力进行比较。【结果】AMF均能侵染上述两种植物,并且AMF对南美蟛蜞菊根系的侵染率显著高于其对蟛蜞菊根系的侵染,尤其是在低磷水平下南美蟛蜞菊的菌根侵染率更高,而且AMF的侵染显著促进了低磷水平下南美蟛蜞菊的生长及其对蟛蜞菊的竞争能力。【结论】丛枝菌根真菌能够促进南美蟛蜞菊的生长,增强其对本地植物的竞争优势,该效应很可能对外来植物南美蟛蜞菊的成功入侵产生一定的作用。  相似文献

A split root device was designed to assess the possible role of AMF in translocation and detoxification of As by maize plants. Half of each maize root system grew in As-amended or unamended soil and the remainder was inoculated with either Glomus mosseae or Acaulospora morrowiae. External mycelium was collected from a third compartment. Neither shoot nor root As concentrations were affected by inoculation with either fungus. Soil As amendment produced higher As concentrations in roots in the second compartment and in the external mycelium. The As concentrations in the matrix solution of the second root compartment were lower in mycorrhizal treatments with no differences in soluble As in the hyphal compartments. Mycorrhiza exerted little effect on As translocation within plants but may have influenced root As efflux. Deposition of As in external mycelium indicates a possible role of mycorrhizal fungi in the detoxification of As in the host plants.  相似文献

在温室盆栽条件下,分别模拟单作、间作和尼龙网分隔种植,比较接种丛枝菌根(arbuscular mycorrhizal, AM)真菌Glomus intraradicesGlomus mosseae对菌根植物玉米和非菌根植物油菜生长和磷吸收状况的影响,并分析土壤中各无机磷组分的变化。结果发现,接种AM真菌可以促进土壤中难溶性磷(Ca10-P和O-P)向有效态磷转化,并显著降低总无机磷含量 (P<0.05),显著提高菌根植物玉米的生物量和磷吸收量(P<0.05),特别是在间作体系中使玉米的磷营养竞争比率显著提高了45.0%-104.1% (P<0.05),显著降低了油菜的生物量和磷吸收量(P<0.05),从而增强了了菌根植物的竞争优势,降低了非菌根植物与菌根植物的共存能力。揭示了石灰性土壤中AM真菌对植物物种多样性的影响,有助于更加全面地理解AM真菌在农业生态系统中的作用。  相似文献

Root colonization by arbuscular mycorrhizal fungi (AMF) was investigated in industrially polluted grassland characterized by exceptionally high phosphorus levels (up to 120 g kg(-1) soil). Along a pollution-induced nitrogen gradient, soil and tissue element concentrations of Artemisia vulgaris plants and their mycorrhizal status were determined. Additionally, we compared mycorrhization rates and above-ground biomass of A. vulgaris at N-fertilized and control plots in the N-poor area. Despite high soil and tissue P concentrations, plants from N-deficient plots, which were characterized by low tissue N concentrations and N : P ratios, were strongly colonized by AMF, whereas at a plot with comparable P levels, but higher soil and plant N concentrations and N : P ratios, mycorrhization rates were significantly lower. Correlation analyses revealed a negative relationship between percentage root colonization of A. vulgaris by AMF and both tissue N concentration and N : P ratio. Accordingly, in the fertilization experiment, control plants had higher mycorrhization rates than N-fertilized plants, whereas the species attained higher biomass at N-fertilized plots. The results suggest that N deficiency stimulates root colonization by AMF in this extraordinarily P-rich field site.  相似文献

Two experiments were carried out to investigate phosphorus efficiencies and mycorrhizal responsiveness in an improved cultivar (Clipper) and a landrace (Sahara) of barley (Hordeum vulgare L.). In experiment 1, two pot sizes were used to evaluate the effect of soil volume on P uptake and mycorrhizal responsiveness. In experiment 2, a compartmented ("cross-pot") system was used to monitor (32)P delivery by external hyphae of arbuscular mycorrhizal fungi (AMF) to the host plant. Results showed that, irrespective of growth conditions, Sahara had much larger root biomass than Clipper and consequently substantially more P was allocated to roots in Sahara than in Clipper. Specific root length in Clipper was much longer than in Sahara. Increase in soil volume enhanced percentage root length colonised by AMF, plant growth and P uptake, and Sahara was more sensitive to changes in soil volume than Clipper. Pot size (soil volume) used to assess responsiveness to AMF by different plant species or genotypes with different root/shoot ratios might be a confounding factor. Clipper was more responsive to AMF than Sahara in terms of tissue P concentrations, which is partly related to their differences in root/shoot ratios. However, increases in SPU [specific P uptake, mg P (g root biomass)(-1)] caused by AMF were bigger in Clipper, suggesting that AMF played a larger role in P uptake. In accordance with the larger increase in SPU, Clipper took up more (32)P via AMF hyphae than Sahara. The compartmented system using radioactive P might be an alternative approach to directly investigate mycorrhizal responsiveness of different plant species or varieties than conventional pot experiments, provided that the same AM fungus is used.  相似文献

A study was conducted to evaluate the effect of N and P supply levels on mycorrhizal formation and nutrient uptake in corn hybrids with different architectures and to determine arbuscular mycorrhizal fungal (AMF) development in relation to shoot N/P ratio and shoot:root ratio. Corn pot cultures with a pasteurized medium of two parts sand and one part sandy loam soil were grown in the greenhouse. Marigold plants inoculated or not with Glomus intraradices Schenck & Smith were used to establish an AMF hyphal network in the designated soil pots. Corn hybrids were seeded after removal of the marigold plant. Mycorrhizal colonization of corn hybrids and the quantity of extraradical hyphae produced in soil were greatest at the lowest P level and at the intermediate N level. Root colonization was correlated with shoot N/P ratio only at the intermediate N level. The shoot concentrations of P, Mg, Zn and Cu were significantly higher in mycorrhizal plants than in non-mycorrhizal plants. The corn phenotype with the highest shoot:root ratio had the highest root colonization. The corn hybrid with a leafy normal stature architecture had a greater mycorrhizal colonization than that of other two corn hybrids. This experiment showed that N level in soil influenced shoot N/P ratio, root colonization and extraradical hyphal production, which in turn influenced uptake of other nutrients. The effects of liming and inoculation with the arbuscular mycorrhizal fungus, Glomus intraradices Schenck and Smith on the uptake of phosphate (P) by maize (Zea mays L.) and soybean (Glycine max [L.] Merr.) and on depletion of inorganic phosphate fractions in rhizosphere soil (Al-P, Fe-P, and Ca-P) were studied in flat plastic containers using two acid soils, an Oxisol and an Ultisol, from Indonesia. The bulk soil pH was adjusted in both soils to 4.7, 5.6, and 6.4 by liming with different amounts of CaCO3.In both soils, liming increased shoot dry weight, total root length, and mycorrhizal colonization of roots in the two plant species. Mycorrhizal inoculation significantly increased root dry weight in some cases, but much more markedly increased shoot dry weight and P concentration in shoot and roots, and also the calculated P uptake per unit root length. In the rhizosphere soil of mycorrhizal and non-mycorrhizal plants, the depletion of Al-P, Fe-P, and Ca-P depended in some cases on the soil pH. At all pH levels, the extent of P depletion in the rhizosphere soil was greater in mycorrhizal than in non-mycorrhizal plants. Despite these quantitative differences in exploitation of soil P, mycorrhizal roots used the same inorganic P sources as non-mycorrhizal roots. These results do not suggest that mycorrhizal roots have specific properties for P solubilization. Rather, the efficient P uptake from soil solution by the roots determines the effectiveness of the use of the different soil P sources. The results indicate also that both liming and mycorrhizal colonization are important for enhancing P uptake and plant growth in tropical acid soils.  相似文献

 Arbuscular mycorrhizal fungi (AMF) living symbiotically with host plants enhance plant growth by improving the acquisition of mineral nutrients and water relations. This study determined the effects of AMF inoculation on growth, benefit/cost and water-use efficiency (grams dry matter produced per kilogram water evapotranspired) in two durum wheat genotypes (drought sensitive and drought tolerant) under water-stressed and well-watered conditions. Plants were grown in a low-P silty clay (Typic Xerochrept) soil mix in a greenhouse. Shoot and root dry matter (DM) and root AMF colonization were higher for well-watered than for water-stressed plants. The mycorrhizal plants were more water-use efficient than nonmycorrhizal plants. Shoot DM differences between mycorrhizal and nonmycorrhizal plants represent the benefit derived by plants from AMF-root associations. Shoot DM differences between mycorrhizal and nonmycorrhizal plants under similar conditions of water treatment represent the cost to the plant of AMF-root associations. Values of benefit/cost for AMF-root associations were highest when plants were water-stressed and decreased under well-watered conditions. Mycorrhizas are ubiquitous symbioses that may have an important role in the movement of C from air to soil. Studies on the effects of climate change factors on mycorrhizas have been concentrated on the effects of atmospheric [CO2] whereas temperature effects have been neglected. Based on previous results showing no effect of varying atmospheric [CO2] on the development and P uptake of the arbuscular mycorrhizal fungi (AMF) colonizing plants growing in controlled conditions, we hypothesized that soil temperature would have a higher impact on AMF development and nutrient uptake than the effects of [CO2] on the host plant. Pea plants were grown in association with either a single isolate of Glomus caledonium or AMF from field soil in factorial combination with the corresponding current (10 °C) or elevated (15 °C) soil temperatures at current (350 p.p.m) or elevated (700 p.p.m) atmospheric [CO2]. 33P uptake by extraradical AMF hyphae was measured independently from root P uptake in a root exclusion compartment. Intraradical colonization developed well at both soil temperatures and almost duplicated from 10 to 15 °C. Extraradical mycelium developed only at 15 °C in the root exclusion compartment and hyphal P uptake could therefore be studied at 15 °C only. Hyphal P uptake differed markedly between inoculum types, but was not altered by growing the host plants at two atmospheric [CO2] levels. No significant [CO2] × soil temperature interactions were observed. The results suggested that, in the system tested, AMF development and function is likely more influenced by the temperature component of climate change than by its [CO2] component. We suggest that much more attention should be paid to temperature effects in future studies.  相似文献
