Survival of airborne virus influences the extent of disease transmission via air. How environmental factors affect viral survival is not fully understood. We investigated the survival of a vaccine strain of Gumboro virus which was aerosolized at three temperatures (10°C, 20°C, and 30°C) and two relative humidities (RHs) (40% and 70%). The response of viral survival to four metrics (temperature, RH, absolute humidity [AH], and evaporation potential [EP]) was examined. The results show a biphasic viral survival at 10°C and 20°C, i.e., a rapid initial inactivation in a short period (2.3 min) during and after aerosolization, followed by a slow secondary inactivation during a 20-min period after aerosolization. The initial decays of aerosolized virus at 10°C (1.68 to 3.03 ln % min(-1)) and 20°C (3.05 to 3.62 ln % min(-1)) were significantly lower than those at 30°C (5.67 to 5.96 ln % min(-1)). The secondary decays at 10°C (0.03 to 0.09 ln % min(-1)) tended to be higher than those at 20°C (-0.01 to 0.01 ln % min(-1)). The initial viral survival responded to temperature and RH and potentially to EP; the secondary viral survival responded to temperature and potentially to RH. In both phases, survival of the virus was not significantly affected by AH. These findings suggest that long-distance transmission of airborne virus is more likely to occur at 20°C than at 10°C or 30°C and that current Gumboro vaccination by wet aerosolization in poultry industry is not very effective due to the fast initial decay.  相似文献

The hygropreference of adult Cryptopygus antarcticus and Alaskozetes antarcticus was investigated over 2 h at 5, 10 and 20 degrees C, along humidity gradients (9-98% RH) established by means of different salt solutions. Two chamber arrangements were employed, linear and grid, to determine any influence of thigmotactic behaviour on distribution within the RH gradient. The humidity preference of both species varied with temperature. At 5 and 10 degrees C, C. antarcticus distributed homogeneously showing no clear RH preference. At 20 degrees C, this species preferred the highest humidity (98% RH). A. antarcticus demonstrated a preference for the lowest humidity (9% RH) at 5 degrees C, but at 10 degrees C its distribution differed between the two arena types. At 20 degrees C, A. antarcticus showed no clear humidity preference. Assays to control for experimental asymmetries along the gradient; thigmotactic behaviour; and aggregative behaviour exclude these factors as explanations for the observed results. The mean initial water content of samples did not differ significantly between temperature regimes (C. antarcticus: 68.6, 71.1 and 74.3%; A. antarcticus: 68.1, 70.1 and 68.6% at 5, 10 and 20 degrees C respectively), but the level of water loss increased significantly with temperature. The influence of desiccation tolerance and the ecological significance of the observed humidity preferences are discussed.  相似文献

The spotted stemborer, Chilo partellus (Swinhoe) (Lepidoptera: Crambidae), is one of the most important insect pests attacking maize and sorghum in Ethiopia. Recent studies have indicated that the pest is spreading to new locations where it was not reported before. In the current study, laboratory investigations were carried out to determine the combined effect of different levels of relative humidity and temperature regimes on the development and fecundity of C. partellus, as these physical factors are known to play an important role in the life cycle of insects and adaptability to local climate. Developmental time, longevity, potential fecundity and realized fecundity of C. partellus were measured under controlled conditions. Three temperature regimes (22°C, 26°C and 30°C) and three relative humidity levels (40%, 60% and 80%) were tested. It was found that temperature, relative humidity (RH) and their interaction significantly affected the developmental time, adult longevity, potential fecundity and realized fecundity of the pest. Developmental time was inversely related to temperature. Mean duration of C. partellus life cycle was 70.2 days at 22°C and 80% RH, whereas it took only 26.5 days to complete its life cycle at 30°C and 40% RH. Male and female longevity were similar in most cases. The adult life span ranged between 6.9-11.1 days at 22°C and 3.1-7.2 days at 30°C for different levels of relative humidity. The most suitable conditions for C. partellus development and fecundity were 26-30°C temperatures regimes and 60-80% RH levels.  相似文献

Abstract.  Water deprivation tolerance is investigated in the last larval stadium of Libellula depressa under various conditions of relative humidity (60–100% relative humidity; RH). Most of the larvae maintained at 100% RH emerge and, at lower RH levels show some resistance to dehydration because they die after a mean period ranging from 1.4 days at 60% RH up to 6.7 days at 90% RH. In dual-choice chambers with humidity gradients from 63–74% RH and from 68–84% RH, larvae spend most of the time in the moist side of the chamber. In a Y-tube olfactometer, the larvae reveal a positive hygrotaxis to two airstreams carrying different amounts of water vapour (98% vs. 50%) and spend most of their time in the 'humid' arm. The ecological significance of desiccation tolerance and hygropositive response in the last larval stadium of L. depressa is discussed in relation to the presence of hygroreceptors in dragonfly larvae.  相似文献

The root-feeding flea beetle Longitarsus bethae Savini & Escalona, was introduced into South Africa as a candidate biological control agent for the noxious and invasive weed, Lantana camara L. As part of the study to predict the beetles' survival in its new range, the influence of climatic conditions on its egg development and reproductive performance were investigated in the laboratory. The threshold temperature (T degrees) and degree-days (DD) required for egg hatch were determined after exposing the eggs to various constant temperatures (12, 17, 22, 27 and 32 degrees C) in separate growth chambers. The DD required for egg hatch was 178.6, and the temperature threshold required for egg hatch was 11.3 degrees C. Survival of eggs varied from 27 to 56% at 32 and 17 degrees C, respectively, and was optimum between 17 and 25 degrees C. Oviposition was examined under high and low relative humidity (RH) regimes while egg hatch was determined at six RH levels, each maintained in a separate controlled growth chamber set at a constant temperature (25 degrees C). Whilst RH had no influence on oviposition, eggs were highly susceptible to aridity, and continuous exposure to relative humidity below 63% for more than three days was wholly lethal at 25 degrees C. Optimum egg hatch occurred at RH between 85 and 95% for up to 12 days. The effect of day length on oviposition and subsequent egg hatch was investigated under two photoperiod regimes. Neither oviposition nor subsequent egg hatch was influenced by photoperiod. The knowledge obtained will be useful for mass rearing as well as field release programmes for L. bethae.  相似文献

Abstract The effects of relative humidity (RH) on cocoon formation and survival in the braconid parasitoid wasp Cotesia glomerata (L.) (Hymenoptera: Braconidae) are investigated under various humidity conditions (50, 75, 90, 95 and 100% RH) at 20 °C and under an LD 16 : 8 h photoperiod. The mortality rate at the time of egression from hosts under 100% RH is significantly higher than for other RHs. Cocoon clusters formed at 100% RH spread significantly more than those formed at 50, 75, or 90% RH. Developmental periods differ significantly among RHs under which wasps developed. The mean period from the egression from hosts to adult emergence is 8.7 days when developed at 50–95% RHs, and 8.0 days at 100% RH. The emergence rates of C. glomerata that are maintained under the same humidity conditions after egression from hosts are not significantly different among RHs. However, emergence rates from cocoons that are transferred from 100% RH to 50 and 75% RH are < 70%, although the rates are > 90% in most cases. Some wasps do not emerge from cocoons: more than 60% die after adult eclosion at all RHs; the relative frequency of adult deaths is approximately 90% at 50% RH. Relative humidity influences the cluster and cocoon status strongly: both good clusters and cocoons are formed at low RHs. Emergence rates from cocoons of different ranks are significantly different: the rates of low‐rank cocoons are low at low RHs. The survival of C. glomerata is affected strongly by RH through cocoon formation.  相似文献

Eleven samples of Ottawa variety hard red winter wheat were inoculated with a standardized suspension of Salmonella montevideo. The contaminated wheat samples were placed in constant relative humidity (RH) chambers held at 25 C. Relative humidities were 7, 11, 22, 33, 43, 53, 62, 75, 84, 92, and 98%. Constant RH at 25 C was maintained with different saturated salt solutions in the sealed chambers. Periodic counts of viable S. montevideo cells per gram of wheat were made over a 28-week sampling period. Viable counts of S. montevideo on wheat held at 7, 11, and 22% RH decreased from an initial 106 cells/g of wheat to a final count of 104 cells/g in each sample. Samples stored at 33, 43, 53, and 62% RH decreased from 106 viable cells/g to 3.6 × 103, 103, 102, and 20 viable cells/g, respectively. No viable S. montevideo cells were detected in the samples held at 75, 84, 92, and 98% RH after 22, 16, 26, and 16 weeks, respectively.  相似文献

The aim of this study was to investigate under a controlled environment, the effect of temperature on the survival and infectivity of Pseudotheraptus devastans Distant, a cassava anthracnose disease vector. The insect P. devastans was collected from young cassava (Manihot esculenta Crantz) field plots, at the International Institute of Tropical Agriculture, (IITA), Ibadan, Nigeria. A mixture of the different developmental stages of eggs, first to fifth instar nymphs, and adults, were incubated in controlled environment chambers, under various constant temperatures of: 15, 17, 22, 25, 27, 30, and 35°C. Relative humidity at different temperature conditions were recorded and maintained at 90%, 85%, 80%, 75%, 70%, 65%, and 60%, respectively. A significant increase in insect survival was observed between 22 and 27°C temperature conditions while a significant decrease in survival was observed at 15°C and above 30°C. Lesion number, lesion diameter and infectivity among the insect stages varied as a function of temperature and relative humidity. Infectivity was highest at 22–25°C maintained at 75–80% RH and lowest at 15°C and above 30°C maintained respectively, at 65% RH and 90% RH. There was considerable low vector infectivity due to low survival of the insects at extreme temperatures.  相似文献

Engineered materials to improve the shelf-life of desiccated microbial strains are needed for cost-effective bioaugmentation strategies. High temperatures and humidity of legume-growing regions challenge long-term cell stabilization at the desiccated state. A thermostable xeroprotectant core and hydrophobic water vapour barrier shell encapsulation technique was developed to protect desiccated cells from the environment. A trehalose core matrix increased the stability of desiccated Bradyrhizobium by three orders of magnitude over 20 days at 32°C and 50% relative humidity (RH) compared to buffer alone; however, the improvement was not deemed sufficient for a shelf-stable bioproduct. We tested common additives (skim milk, albumin, gelatin and dextran) to increase the glass transition temperature of the desiccated product to provide further stabilization. Albumin increased the glass transition temperature of the trehalose-based core by 40°C and stabilized desiccated Bradyrhizobium for 4 months during storage at high temperature (32°C) and moderate humidity (50% RH) with only 1 log loss of viability. Although the albumin-trehalose core provided exceptional protection against high temperature, it was ineffective at higher humidity conditions (75%). We therefore incorporated a paraffin shell, which protected desiccated cells against 75% RH providing proof of concept that core and shell encapsulation is an effective strategy to stabilize desiccated cells.  相似文献

The use of mulch in urban landscapes has increased in the United States for the past decade. Tree-based organic mulches can supply Coptotermes formosanus Shiraki with food, moisture, and shelter. The current research contributes to mulch management technology in termite control. A choice test arena was designed to determine the repellence and mortality caused by commercial mulches treated with different concentrations of the fungus Metarhizium anisopliae (Metschnikoff) against C. formosanus. Each of six tree-based mulches (pine bark, pine straw, bald cypress, eucalyptus, water oak, and melaleuca) was coated with six conidial concentrations ranging from 1 x 10(3) to 1 x 10(8) conidia/ml. The foragers of C. formosanus were repelled significantly by the fungal-treated mulch substrates; the proportion of termites on fungal-treated mulch was usually <20% during the 28-d test. By day 28, >99% of the termites were killed in test arenas containing a chamber with mulch treated with 10(7) or 10(8) conidia/ml. M. anisopliae significantly reduced mulch consumption by 34-71%. Mulch consumption by the termites was negatively correlated with fungal concentration, and the type of mulch also affected consumption. The differences in termite foraging activities, mortality, and food consumption among mulches were usually confounded by differences in fungal concentrations of M. anisopliae. The results indicate that repellence and virulence of M. anisopliae conidia should significantly reduce the suitability of these six mulches as a habitat for C. formosanus.  相似文献

Ophraella communa, an unintentionally introduced leaf beetle in China, has good control efficiency on ragweed, Ambrosia artemisiifolia. Aspects of the climatic requirements for development, survival, longevity and fecundity of O. communa were studied under the conditions of constant temperature (25 ± 1°C), photoperiod of 14 L:10 D and three relative humidities (60%, 75% and 90% RHs). The results showed that the developmental periods of O. communa at different stages shortened along with the increasing relative humidity, except that of the pupal stage. Although no differences were observed in the pupal survival rate, ovipositional period, fecundity, longevity and adult female age-specific survivorship of O. communa under the three humidity conditions, the survival rates during the egg, larva and entire immature stage were significantly higher at 75% RH and 90% RH than at 60% RH. The innate rate of increase (r m), net reproductive rate (R 0), finite rate of increase (λ) reached the maximum at 75% RH, with values of 0.181, 1116.4 and 1.198, respectively. These results indicated that the optimum relative humidity for the development of O. communa ranged from 75% RH to 90% RH. Thus O. communa prefers moist microclimate habitats. Its population may expand rapidly during mid-May to late August in south, east and central China, when the humidity is relatively high.  相似文献

为了研究亚高温下不同空气湿度对番茄植株光合作用及物质积累的影响,本试验利用人工气候室,在11:00—15:00平均温度为33℃的亚高温条件下,设置3个空气相对湿度处理,分别为70%~80%(高湿)、50%~60%(中湿)和不加湿的30%~40%(低湿)。结果表明:在处理25d时,高湿处理番茄叶片叶绿素含量、净光合速率显著高于低湿处理,而低湿处理果实空洞率比高湿处理高18.4%(P<0.05);在33℃亚高温条件下,70%~80%的相对湿度有利于光合作用的增强和果实品质的提高。  相似文献

The stability of a paper-immobilized antibody was investigated over a range of temperatures (40-140 °C) and relative humidities (RH, 30-90%) using both unmodified filter paper and the same paper impregnated with polyamide-epichlorohydrin (PAE) as supports. Antibody stability decreased with increasing temperature, as expected, but also decreased with increasing RH. At 40 °C, the half-life was more than 10 days, with little dependence on RH. However, at 80 °C, the half-life varied from ~3 days at low RH to less than half an hour at 90% RH, demonstrating that hydration of the antibody promotes unfolding. Antibody stability was not influenced by the PAE paper surface treatment. This work shows that antibodies are good candidates for development of bioactive paper as they have sufficient stability at high temperature to withstand printing and other roll-to-roll processing steps, and sufficient low temperature stability to allow long-term storage of bioactive paper materials.  相似文献

The influence of ambient conditions on the development of Metarhizium anisopliae chlamydospores in tick eggs is reported for the first time. The infection of tick eggs by M. anisopliae involves common events, such as adhesion, conidial germination, appressoria formation, invasion, and development within the eggs. However, the final stage of fungal development differs according to the environmental conditions. At high humidity (close to 100%) and moderate temperature (25°C) the fungus emerged from the eggs and formed conidiophores and conidia externally on the dead eggs. Elevating the temperature to 30°C or reducing humidity to 55-75% induced the production of chlamydospores inside the eggs, without conidiogenesis. When eggs with mature chlamydospores were returned to the appropriate conditions (25°C and 100% RH), conidiogenesis was recovered. Formation of chlamydospores, observed by means of histology and TEM, began with the thickening and septation of hyphae. As the chlamydospore wall thickened a new external undulated wall layer appeared. The mature chlamydospore in eggs has an oval shape (5.3 ± 0.9 microm long, 2.5 ± 0.2 microm wide); its wall comprises three distinct layers. The ability of M. anisopliae to produce chlamydospores under harsh conditions is advantageous and should be considered in application.  相似文献

The rate of spinning of silkworm larva (Bombyx mori.L) was slow at 22 degrees C and fast at 38 degrees C. The time taken for completion of cocoon was longest at 98 +/- 2% and least at 40 +/- 2% RH. However, good quality cocoons were spun at 22 degrees C and 65 +/- 5% RH, hence it would be advantageous to maintain this temperature and relative humidity at the time of cocoon spinning in the case of CB race of silkworm.  相似文献

在相对湿度(RH)为55%、65%、75%、85%和95%条件下,观察湿度对三角新小卷蛾Olethreutes leucaspis Meyrick各虫态生长、发育及存活的影响,组建三角新小卷蛾实验种群生命表。结果表明:在RH为55%-95%条件下,三角新小卷蛾卵、幼虫、蛹和世代的发育历期在RH为85%时最短,发育历期分别为1.67、12.46、7.67和25.42 d。高湿条件下卵、幼虫和蛹的存活率较高。三角新小卷蛾各虫态发育历期、存活率与相对湿度呈抛物线关系。在RH为55%-95%条件下该虫内禀增长力(rm)均大于0,种群趋势指数(I)均大于1,该虫种群呈增长趋势。三角新小卷蛾发育适宜的RH为75%。  相似文献

Developmental and survival rates of the locally important diaspidid pest, Abgrallaspis cyanophylli (Signoret) reared on Solanum tuberosum L . tubers were examined under light and dark conditions; humidities of 33, 53, 62 and 75% relative humidity (RH); varying population densities; constant temperatures in the range of 20 to 30oC and at cycling temperatures of 12 h at 14°C and 12 h at 30°C. Developmental rate was slightly lower under constant light conditions but mortality was higher in the dark, particularly amongst the males. At 26°C, there were no differences in developmental rate in relation to the various humidity levels. However, survival was significantly lower at 33% RH, with females suffering higher mortality than males. Population density was found to have no effect on developmental rate or size of the females. Overall mortality increased in line with population density although the result was poorly correlated. Within the range 20-28°C developmental rate increased with rising temperature but decreased at 30°C. Thermal summation and polynomial regression data show a theoretical lower thermal threshold for development of 12.47°C. The thermal constant was 541.7 degree days. Survival was lowest at 20°C and 30°C and highest at temperatures in the median range and under cycling conditions. Male survival was significantly higher than that of the females at 30°C and under the cycling regime of 14/30°C. The results suggest that the optimum conditions for rearing A. cyanophylli on potatoes would be at temperatures in the range 24-26°C and humidities of 55-65% RH.  相似文献

Summary: The hygropreference of gardening workers of the leaf-cutting ant Atta sexdens rubropilosa was investigated in the laboratory using a gradient of relative humidity. Gardening workers were placed, together with pieces of fungus garden, in small, interconnected nest chambers offering four different relative humidities: 33 %, 75 %, 84 % and 98 % RH. Workers were allowed to move freely between them and to relocate the fungus following their humidity preference. While workers distributed themselves randomly in the nest chambers, they located the fungus gardens in the chamber with the highest humidity. These results indicate that gardening workers are able to sense differences in relative humidity, and that this ability is shown when they are engaged in fungus culturing. Humidity is discussed as one of the relevant variables that probably underlay the evolution of regulatory responses for the control of fungus growth in leaf-cutting ants.  相似文献

Experiments were conducted on laboratory populations of Eriworm, Philosamia ricini Boisduval to study the effect of temperature and humidity on development, growth, survival and oviposition. Experiments were performed at four different temperatures (22 +/- 2 degrees C, 26 +/- 2 degrees C, 28 +/- 2 degrees C and 34 +/- 1 degrees C) each with three different humidities (50%, 70% and 90% relative humidity). Shortest developmental period (42 days) was observed in 28 +/- 2 degrees C and 70% RH. Longest developmental period was (63.6 days) observed in 22 +/- 2 degrees C at 50% RH. Highest larval weight (9.1 g) was found in 28 +/- 2 degrees C at 70% RH. Best pupal weight was observed in 26 +/- 2 degrees C at 70% RH. Weight of silk material was found to be maximum in 26 +/- 2 degrees C at 70% RH. Survival was best in 28 +/- 2 degrees C and 26 +/- 2 degrees C at 70% RH. Oviposition was found to be highest in 28 +/- 2 degrees C at 70% RH.  相似文献

Four isolates of the entomopathogenic fungus Zoophthora radicans were compared in a laboratory study to evaluate the effect of relative humidity (RH) on duration of primary conidial viability. Primary conidia were showered onto agar-coated glass microscope slides within an enclosed chamber equilibrated to one of five test RH levels (60, 75, 80, 95, or 100%). Target RH levels were achieved by recirculating air through a glycerin/water solution, of controlled specific gravity, contained in a reservoir within the chamber. Conidial samples of each isolate incubated for 5, 10, 30, 60, 120, 180, or 240 min at each RH were removed and inspected using a technique of simultaneous vital fluorochrome staining to determine percentage conidial viability. At 60% RH, isolates did not differ significantly and average viability dropped to less than 10% within the first 60 min. At 75% RH, viability did not change significantly over 4 h. However, average viabilities at 75% RH differed significantly for the four isolates and ranged from 24 to 63%. At 80% RH, viability differed significantly among isolates and declined slowly over time, remaining above 80% for 2 h and above 50% for 4 h. At 95 and 100% RH, average viability was near 95% and did not vary significantly with time or isolate. These data can be used to assist selection of appropriate isolates for biological control.  相似文献
