Single-stage nitritation–anammox combines the growth of aerobic ammonium-oxidizing bacteria (AOB) and anaerobic ammonium oxidizing bacteria (AnAOB) in one reactor. The necessary compromise of their milieu conditions often leads to the growth of nitrite-oxidizing bacteria (NOB). For this study, a sequencing batch reactor (SBR) for nitritation–anammox was operated for 180 days with sewage sludge reject water (removal capacity, 0.4 kg?N?m?3?day?1). The growth of NOB was favored by enhanced oxygen supply rather than extended aerobic phases. Suspended-type biomass from this SBR was taken regularly and sieved into three size fractions (all of them <1,000 μm). Batch experiments as well as fluorescence in situ hybridization were performed to study the distribution and activity of AnAOB, AOB, and NOB within those size fractions. Both the measured conversion rates and detected abundances decreased with increasing size fraction. The highest anammox conversion rates (15 g NH4 +–N per kilogram VSS per hour) and the highest abundances of Brocadia fulgida were found in the medium size fraction (100–315 μm). The batch experiments proved to be accurate tools for the monitoring of multiple processes in the reactor. The results were representative for reactor performance during the 6 months of reactor operation.  相似文献

Partial nitrification to nitrite (nitritation) can be achieved in a continuous process without sludge retention by wash out of nitrite oxidising bacteria (NOB) while retaining ammonia oxidising bacteria (AOB), at elevated temperatures (the SHARON process) and, as demonstrated in this paper, also at low dissolved oxygen (DO) concentrations. Enriched AOB was attained at a low DO concentration (0.4 mg l−1) and a dilution rate of 0.42 day−1 in a continuous process. A higher oxygen affinity of AOB compared to NOB seemed critical to achieving this. This was verified by determining the oxygen half saturation constant, K o, with similar oxygen mass transfer resistances for enriched AOB and NOB as 0.033 ± 0.003 mg l−1 and 0.43 ± 0.08 mg l−1, respectively. However, the extent of nitritation attained was found to be highly sensitive to process upsets.  相似文献

To achieve an appropriate mixture of ammonium and nitrite for anaerobic ammonium oxidation (ANAMMOX), 50% partial nitritation was optimized in a fixed bed biofilm reactor treating synthetic wastewater. Results suggested that 50% partial nitritation could be achieved by stepwise increases of influent NH4+-N at pH of 7.8 ± 0.2, temperature of 30 ± 1 °C and dissolved oxygen (DO) of 0.5-0.8 mg l−1. Hydraulic retention time (HRT) and influent alkalinity did significantly affect partial nitritation. At HRT 12 h, 50% partial nitritation could be kept stable, regardless of influent NH4+-N variation, by controlling the influent HCO3/NH4+ molar ratio at 1:1. The fluorescent in situ hybridization (FISH) results indicated the abundance of evolution of ammonia-oxidizing bacteria (AOB) and the nitrite-oxidizing bacteria (NOB) coincided well with the performance of partial nitritation. Furthermore, the AOB were highly affiliated with Nitrosomonas spp. and Nitrosospira spp. dominated (64.1%) in the biofilm with a compact structure during the stable 50% partial nitritation period.  相似文献

Membrane‐aerated biofilm reactors performing autotrophic nitrogen removal can be successfully applied to treat concentrated nitrogen streams. However, their process performance is seriously hampered by the growth of nitrite oxidizing bacteria (NOB). In this work we document how sequential aeration can bring the rapid and long‐term suppression of NOB and the onset of the activity of anaerobic ammonium oxidizing bacteria (AnAOB). Real‐time quantitative polymerase chain reaction analyses confirmed that such shift in performance was mirrored by a change in population densities, with a very drastic reduction of the NOB Nitrospira and Nitrobacter and a 10‐fold increase in AnAOB numbers. The study of biofilm sections with relevant 16S rRNA fluorescent probes revealed strongly stratified biofilm structures fostering aerobic ammonium oxidizing bacteria (AOB) in biofilm areas close to the membrane surface (rich in oxygen) and AnAOB in regions neighbouring the liquid phase. Both communities were separated by a transition region potentially populated by denitrifying heterotrophic bacteria. AOB and AnAOB bacterial groups were more abundant and diverse than NOB, and dominated by the r‐strategists Nitrosomonas europaea and Ca. Brocadia anammoxidans, respectively. Taken together, the present work presents tools to better engineer, monitor and control the microbial communities that support robust, sustainable and efficient nitrogen removal.  相似文献

To achieve nitritation from complete-nitrification seed sludge at room temperature of 19 ± 1 °C, a lab-scale sequencing batch reactor (SBR) treating domestic wastewater with low C/N ratios was operated to investigate the control and optimization of nitrifying communities. Ammonia oxidizing bacteria (AOB) dominance was enhanced through the combination of low DO concentrations (<1.0 mg/L) and preset short-cycle control of aeration time. Nitritation was successfully established with NO2?-N/NOx?-N over 95%. To avoid the adverse impact of low DO concentrations on AOB activities, DO concentrations were increased to 1–2 mg/L. At the normal DO levels and temperatures, on-line control strategy of aerobic durations maintained the stability of nitritation with nitrite accumulation rate over 95% and ammonia removal above 97%. Fluorescence in-situ hybridization (FISH) analysis presented that the maximal percentage of AOB in biomass reached 10.9% and nitrite oxidizing bacteria (NOB) were washed out.  相似文献

Aim

To provide deeper insights into nitrification process within aerobic bioreactors containing supplemental physical support media (hybrid bioreactors).

Methods and Results

Three bench‐scale hybrid bioreactors with different media size and one control bioreactor were operated to assess how biofilm integrity influences microbial community conditions and bioreactor performance. The systems were operated initially at a 5‐day hydraulic retention time (HRT), and all reactors displayed efficient nitrification and chemical oxygen demand (COD) removal (>95%). However, when HRT was reduced to 2·5 days, COD removal rates remained high, but nitrification efficiencies declined in all reactors after 19 days. To explain reduced performance, nitrifying bacterial communities (ammonia‐oxidizing bacteria, AOB; nitrite‐oxidizing bacteria, NOB) were examined in the liquid phase and also on the beads using qPCR, FISH and DGGE. Overall, the presence of the beads in a reactor promoted bacterial abundances and diversity, but as bead size was increased, biofilms with active coupled AOB–NOB activity were less apparent, resulting in incomplete nitrification.

Conclusions

Hybrid bioreactors have potential to sustain effective nitrification at low HRTs, but support media size and configuration type must be optimized to ensure coupled AOB and NOB activity in nitrification.

Significance and Impact of the Study

Emiliania huxleyi (strain L) expressed an exceptional P assimilation capability. Under P limitation, the minimum cell P content was 2.6 fmol P·cell?1, and cell N remained constant at all growth rates at 100 fmol N·cell?1. Both, calcification of cells and the induction of the phosphate uptake system were inversely correlated with growth rate. The highest (cellular P based) maximum phosphate uptake rate (VmaxP) was 1400 times (i.e. 8.9 h?1) higher than the actual uptake rate. The affinity of the P‐uptake system (dV/dS) was 19.8 L·μmol?1·h?1 at μ = 0.14 d?1. This is the highest value ever reported for a phytoplankton species. Vmax and dV/dS for phosphate uptake were 48% and 15% lower in the dark than in the light at the lowest growth rates. The half‐saturation constant for growth was 1.1 nM. The coefficient for luxury phosphate uptake (Qmaxt/Qmin) was 31. Under P limitation, E. huxleyi expressed two different types of alkaline phosphatase (APase) enzyme kinetics. One type was synthesized constitutively and possessed a Vmax and half‐saturation constant of 43 fmol MFP·cell?1·h?1 and 1.9 μM, respectively. The other, inducible type of APase expressed its highest activity at the lowest growth rates, with a Vmax and half‐saturation constant of 190 fmol MFP·cell?1·h?1 and 12.2 μM, respectively. Both APase systems were located in a lipid membrane close to the cell wall. Under N‐limiting growth conditions, the minimum N quotum was 43 fmol N·cell?1. The highest value for the cell N‐specific maximum nitrate uptake rate (VmaxN) was 0.075 h?1; for the affinity of nitrate uptake, 0.37 L·μmol?1·h?1. The uptake rate of nitrate in the dark was 70% lower than in the light. N‐limited cells were smaller than P‐limited cells and contained 50% less organic and inorganic carbon. In comparison with other algae, E. huxleyi is a poor competitor for nitrate under N limitation. As a consequence of its high affinity for inorganic phosphate, and the presence of two different types of APase in terms of kinetics, E. huxleyi is expected to perform well in P‐controlled ecosystems.  相似文献

High bulk extracellular phosphatase activity (PA) suggested severe phosphorus (P) deficiency in plankton of three acidified mountain lakes in the Bohemian Forest. Bioavailability of P substantially differed among the lakes due to differences in their P loading, as well as in concentrations of aluminum (Al) and its species, and was accompanied by species‐specific responses of phytoplankton. We combined the fluorescently labeled enzyme activity (FLEA) assay with image cytometry to measure cell‐specific PA in natural populations of three dinophyte species, occurring in all the lakes throughout May–September 2007. The mean cell‐specific PA varied among the lakes within one order of magnitude: 188–1,831 fmol · cell?1 · h?1 for Gymnodinium uberrimum (G. F. Allman) Kof. et Swezy, 21–150 fmol · cell?1 · h?1 for Gymnodinium sp., and 22–365 fmol · cell?1 · h?1 for Peridinium umbonatum F. Stein. To better compare cell‐specific PA among the species of different size, the values were normalized per unit of cell biovolume (amol · μm?3 · h?1) for further statistical analysis. A step‐forward selection identified concentrations of total and ionic Al together with pH as significant factors (P < 0.05, Monte Carlo permutation test), explaining cumulatively 57% of the total variability in cell‐specific PA. However, this cell‐specific PA showed an unexpected reverse trend compared to an overall gradient in P deficiency of the lake plankton. The autecological insight into dinophyte cell‐specific PA therefore suggested other factors, such as light availability, mixotrophy, and/or zooplankton grazing, causing further PA variations among the acidified lakes.  相似文献

Activated sludge obtained from two municipal wastewater treatment facilities (WWTF) was used as seed sludge for enriched nitrifiers, which were later entrapped in polyvinyl alcohol. Seed sludge from one WWTF was acclimated to high ammonia level (1813 mg NH3-N l?1) through the return of sludge digester supernatant back to primary clarifier while seed sludge from the other WWTF was un-acclimated. To elucidate on how to control partial nitrification by entrapped cells, which could be different from suspended cells, kinetics of entrapped enriched nitrifiers were studied using a respirometric assay. The community of nitrifiers within the entrapment matrix, which was observed by fluorescence in situ hybridization (FISH) technique, was related to the nitritation and nitratation kinetics based on oxygen uptake rate. Maximum oxygen uptake rate, and substrate and oxygen affinities of both ammonia oxidizing bacteria (AOB) for nitritation and nitrite oxidizing bacteria (NOB) for nitratation in entrapped cells were lower than those of corresponding suspended cells. Under dissolved oxygen (DO) limiting conditions, nitratation was more suppressed than nitritation for suspended cells, while for the entrapped cells, the results were the contrary. A free ammonia (FA) inhibition affected only the un-acclimated sludge. Either FA inhibition or DO limitation might not be a sole effective control parameter to achieve partial nitrification by entrapped cells. FISH results revealed that Nitrosomonas europaea was the dominant AOB while Nitrobacter species was the dominant NOB in all cases. Heterotrophs were also present in the entrapment at 22.8 ± 18.6% and 41.5 ± 4.3% of total bacteria for acclimated and un-acclimated originated sludge. The availability of substrate and oxygen governed the distributions of AOB, NOB and heterotrophs within the entrapment and nitritation kinetics of entrapped nitrifiers.  相似文献

In this study, the variation of biomass, kinetic parameters, and stoichiometric parameters for ammonia-oxidizing bacteria (AOB) and nitrite-oxidizing bacteria (NOB) in TNCU3 process were explored at different aerobic hydraulic retention time (AHRT). The results indicated that the growth rate constants of AOB were 0.92, 0.88, and 0.95 days?1, respectively, meanwhile, those of NOB were 2.58 1.41, and 1.40 days?1, respectively, when AHRT was 5, 6, and 7 h. The lysis rate constants for AOB and NOB were 0.13 and 0.17 days?1, respectively. When AHRT was 5, 6, and 7 h, the yield coefficients of AOB were 0.20, 0.23, and 0.28 g COD g?1 N, respectively, meanwhile those of NOB were 0.23, 0.19, and 0.22 g COD g?1 N, respectively. The average percentage of AOB was 0.44, 0.61, and 0.64%, respectively, while that of NOB was 0.46, 0.61, and 0.74%, respectively. The relation between the biomass percentage of AOB and AHRT was in a good agreement with first type hyperbolic curve. The relation between the biomass percentage of NOB and AHRT was in a good agreement with seven types of curve including simple exponential curve, power exponential curve, and first type hyperbolic curve etc. When the AHRT increased from 5 to 7 h, the removal efficiency of NH4 +–N increased from 80.2 to 94.8%, or by 14.6%. Meanwhile, the removal efficiency of total nitrogen increased from 63.6 to 70.9%, or by 7.3%.  相似文献

Lab‐scale experiments were conducted to investigate the effects of ferrous iron on nutrient removal performance and variations in the microbial community inside aerobic granular sludge for 408 days. Two reactors were simultaneously operated, one without added ferrous iron (SBR1), and one with 10 mg Fe2+ L?1 of added ferrous iron (SBR2). A total of 1 mg Fe2+ L?1 of added ferrous iron was applied to SBR1 starting from the 191st day to observe the resulting variations in the nutrient removal performance and the microbial community. The results show that ammonia‐oxidizing bacteria (AOB) could not oxidize ammonia due to a lack of iron compounds, but they could survive in the aerobic granular sludge. Limited ferrous iron addition encouraged nitrification. Enhanced biological phosphorus removal (EBPR) from both reactors could not be maintained regardless of the amount of ferrous iron that was applied. EBPR was established in both reactors when the concentration of mixed liquor suspended solid (MLSS) and the percentage of Accumulibacteria increased. A total of 10 mg Fe2+ L?1 of added ferrous iron had a relatively adverse effect on the growth of AOB species compared to 1 mg Fe2+ L?1 of added ferrous iron, but it encouraged the growth of Nitrospira sp. and Accumulibacteria, which requires further study. It could be said that the compact and stable structure of aerobic granular sludge preserved AOB and NOB from Fe‐deficient conditions, and wash‐out during the disintegration period. © 2017 American Institute of Chemical Engineers Biotechnol. To reveal the succession procedure of ammonia-oxidizing bacteria (AOB) and nitrite-oxidizing bacteria (NOB) community structure in sequencing biofilm batch reactor (SBBR), the molecular biological techniques of denaturing gradient gel electrophoresis (DGGE), cloning, and real-time PCR were applied. DGGE showed that the structural diversity of the bacterial community increased during the biofilm formation period, and some kinds of populations had been highly preponderant consistently. The results of cloning and sequencing revealed that Nitrosomonas was the dominant species. The real-time PCR analysis indicated that the amount of the AOB increased significantly after the cultivation period, and the NOB gradually decreased. The AOB content on the 25th day was 17 times that of the 6th day. It also showed the biofilm formed successfully with accumulating nitrite and prepared to achieve the achievement of simultaneous nitrification and denitrification in SBBR. Furthermore, the ammonia-oxidizing rate was in correspondence with the NH4 +-N removal efficiency.  相似文献

A kinetic study was carried out in a saline medium to assess the effect of O2 on the two-step nitrification process: for the first nitritation step, 2–26 mg dissolved O2 (DO) l–1 was used and for the second nitratation step, 0.5–24 mg DO l–1 was used. Nitritation rate was measured in the presence of sodium azide so as to inhibit nitratation. Ammonia-oxidizing (AOB) and nitrite-oxidizing (NOB) bacterial in the mixed culture were determined by 16 S rRNA hybridization. The affinity constants for oxygen O2 of the AOB and the NOB were 1.66 mg O2 l–1 and 3 mg O2 l–1 respectively. The larger than the previously reported values of these constants might be due to the high salt content in the medium. High O2 concentrations did inhibit the nitrification rate.  相似文献

Poly(vinyl alcohol) (PVA) has been utilized as a support material for the immobilization of nitrifying bacteria without the comprehensive survey of partial nitritation. In the present study, the activities of nitrifiers and the maximum nitrogen conversion rate of partial nitritation with PVA sponge-cubes were specified according to different conditions. The selective enrichment of ammonia-oxidizing bacteria (AOB) on PVA sponge-cubes was achieved by the competition between AOB and nitrite-oxidizing bacteria for dissolved oxygen. The efficiency of ammonia oxidation was proportional to the concentration of HCO3 ? with the molar ratio of HCO3 ?-C/NH4 +-N = 1.91 and a half of the ratio was applied to the further experiments to ensure stable partial nitritation. The maximum nitrogen conversion rate of partial nitritation was dependent on the volume, not the size of sponge-cubes. The partial nitritation showed the superior rate performance of 3.09 kg N/m3 day with the packing ratio of 32 % of 5 × 5 × 5 mm3 PVA sponge-cubes.  相似文献

Aims: We undertook a series of experiments to investigate the susceptibility of Legionella pneumophila grown under extracellular and intracellular conditions and other water‐related bacteria to silver ions. Methods and Results: In this study, the antimicrobial effect of silver ions to intra‐ and extra‐cellular grown Legionella bacteria was investigated. The minimal inhibitory concentration (MIC) after 24 h exposure, leading to a 5 log reduction, was c. 64 μg l?1 AgNO3 for extracellular grown Legionella and other tested Gram‐positive and Gram‐negative bacteria. In contrast, the MIC for intracellularly grown Legionella was up to 4096 μg l?1 AgNO3 after 24 h. Furthermore, the heterotrophic bacteria grown within a biofilm model were killed at a concentration of 4–16 μg l?1 AgNO3. In contrast, biofilm‐associated Legionella were less sensitive (MIC 128–512 μg l?1 AgNO3). Conclusion: Intracellularly and biofilm‐grown legionellae are less sensitive against silver compared with agar‐grown bacteria. Significance and Impact of the Study: The reduced sensitivity of Legionella grown in amoebae might explain why the effect of silver decontamination requires an extended exposure in field trials.  相似文献

Up-flow oxygen-controlled biofilm reactors equipped with a non-woven fabric support were used as a single reactor system for autotrophic nitrogen removal based on a combined partial nitrification and anaerobic ammonium oxidation (anammox) reaction. The up-flow biofilm reactors were initiated as either a partial nitrifying reactor or an anammox reactor, respectively, and simultaneous partial nitrification and anammox was established by careful control of the aeration rate. The combined partial nitrification and anammox reaction was successfully developed in both biofilm reactors without additional biomass inoculation. The reactor initiated as the anammox reactor gave a slightly higher and more stable mean nitrogen removal rate of 0.35 (± 0.19) kg-N m−3 d−1 than the reactor initiated as the partial nitrifying reactor (0.23 (± 0.16) kg-N m−3 d−1). FISH analysis revealed that the biofilm in the reactor started as the anammox reactor were composed of anammox bacteria located in inner anoxic layers that were surrounded by surface aerobic AOB layers, whereas AOB and anammox bacteria were mixed without a distinguishable niche in the biofilm in the reactor started as the partial nitrifying reactor. However, it was difficult to efficiently maintain the stable partial nitrification owing to inefficient aeration in the reactor, which is a key to development of the combined partial nitrification and anammox reaction in a single biofilm reactor.  相似文献

Marine Crenarchaeota are among the most abundant groups of prokaryotes in the ocean and recent reports suggest that they oxidize ammonia as an energy source and inorganic carbon as carbon source, while other studies indicate that Crenarchaeota use organic carbon and hence, live heterotrophically. We used catalysed reporter deposition fluorescence in situ hybridization (CARD‐FISH) to determine the crenarchaeal and bacterial contribution to total prokaryotic abundance in the (sub)tropical Atlantic. Bacteria contributed ~50% to total prokaryotes throughout the water column. Marine Crenarchaeota Group I (MCGI) accounted for ~5% of the prokaryotes in subsurface waters (100 m depth) and between 10 and 20% in the oxygen minimum layer (250–500 m depth) and deep waters (North East Atlantic Deep Water). The fraction of both MCGI and Bacteria fixing inorganic carbon, determined by combining microautoradiography with CARD‐FISH (MICRO‐CARD‐FISH), decreased with depth, ranging from ~30% in the oxygen minimum zone to < 10% in the intermediate waters (Mediterranean Sea Outflow Water, Antarctic Intermediate Water). In the deeper water masses, however, MCGI were not taking up inorganic carbon. Using quantitative MICRO‐CARD‐FISH to determine autotrophy activity on a single cell level revealed that MCGI are incorporating inorganic carbon (0.002–0.1 fmol C cell?1 day?1) at a significantly lower rate than Bacteria (0.01–0.6 fmol C cell?1 day?1). Hence, it appears that MCGI contribute substantially less to autotrophy than Bacteria. Taking the stoichiometry of nitrification together with our findings suggests that MCGI might not dominate the ammonia oxidation step in the mesopelagic waters of the ocean to that extent as the reported dominance of archaeal over bacterial amoA would suggest.  相似文献

The photoprotective response in the dinoflagellate Glenodinium foliaceum F. Stein exposed to ultraviolet‐A (UVA) radiation (320–400 nm; 1.7 W · m2) and the effect of nitrate and phosphate availability on that response have been studied. Parameters measured over a 14 d growth period in control (PAR) and experimental (PAR + UVA) cultures included cellular mycosporine‐like amino acids (MAAs), chls, carotenoids, and culture growth rates. Although there were no significant effects of UVA on growth rate, there was significant induction of MAA compounds (28 ± 2 pg · cell?1) and a reduction in chl a (9.6 ± 0.1 pg · cell?1) and fucoxanthin (4.4 ± 0.1 pg · cell?1) compared to the control cultures (3 ± 1 pg · cell?1, 13.3 ± 3.2 pg · cell?1, and 7.4 ± 0.3 pg · cell?1, respectively). In a second investigation, MAA concentrations in UVA‐exposed cultures were lower when nitrate was limited (P < 0.05) but were higher when phosphate was limiting. Nitrate limitation led to significant decreases (P < 0.05) in cellular concentration of chls (chl c1, chl c2, and chl a), but other pigments were not affected. Phosphate availability had no effect on final pigment concentrations. Results suggest that nutrient availability significantly affects cellular accumulation of photoprotective compounds in G. foliaceum exposed to UVA.  相似文献

Fatty acids were measured in G. galatheanum grown either phototrophically, or mixotrophically with Storeatula major (Cryptophyceae) as prey. G. galatheanum, like many photosynthetic dinoflagellates, contains high amounts of n‐3 long‐chain‐polyunsaturated fatty acids (LC‐PUFA) such as docosahexaenoic acid (DHA, 22:6n‐3) and the hemolytic toxic fatty acid 18:5n‐3. We hypothesize that a benefit of phagotrophy in G. galatheanum is the acquisition of precursor linolenic acid (18:3n‐3) that fuels LC‐PUFA synthesis. Phototrophs grew at 0.37 d?1, while mixotrophs grew at 0.40 d?1 with a feeding rate of 0.62 d?1. Photosynthesis was lower in mixotrophs (3.7 pg C cell?1 h?1) than phototrophs (4.9 pg C cell?1 h?1). DHA levels were higher in mixotrophs [3.7 (+/? 0.11) pg cell?1] than phototrophs [3.0 (+/? 0.16) pg cell?1] and prey [0.4 (+/? 0.01) pg cell?1]. 18:5n–3 levels [1.7 (+/? 0.03) pg cell?1] were similar in phototrophs and mixotrophs. An intermediate in n‐3 LC‐PUFA synthesis, 20:4n‐3, accumulated in mixotrophs [0.6 (+/? 0.27) pg cell?1] relative to phototrophs (not detected) and prey [0.03 (+/? 0.002) pg cell?1]. Low ratios of linolenic acid to DHA in phototrophic G. galatheanum (0.14) relative to mixotrophic G. galatheanum (0.29) and prey (2.14) are consistent with substrate limitation of LC‐PUFA synthesis in phototrophs. Accumulation of 20:4n‐3 suggests incomplete conversion of linolenic acid to DHA, possibly due to conditions in batch culture. We conclude that precursors for n‐3 LC‐PUFA biosynthesis in G. galatheanum may be acquired through ingestion of S. major, and may partially control feeding/photosynthesis in mixotrophic populations.  相似文献
