Zusammenfassung Fünf Sommergersten, die gegen Rasse 24 von Puccinia striiformis resistent sind, wurden in fast allen möglichen Kombinationen miteinander und mit drei anfälligen Formen gekreuzt. Die F 2-Generationen, in zwei Fällen auch die F 3, wurden im Gewächshaus mit Rasse 24 infiziert. Zusätzlich konnten F 2-Generationen einiger Kreuzungen auch auf dem Feld mit Rasse 24 geprüft werden. Eine sichere Klassifikation und damit die eindeutige Analyse der Ergebnisse war nur im Gewächshaus möglich. Die Feldversuche stützen zwar diese Ergebnisse, hätten aber allein keine sicheren Schlüsse erlaubt.Die Resistenz aller fünf untersuchten Gersten ist monogen rezessiv bedingt. Drei von ihnen sind bezüglich des Resistenzgens allel: Bigo — yr, Ab. 14 — yr, BBA 2890 — yr. In Abed Binder 12 liegt yr2 vor, in I 5 yr3. Die Ergebnisse deuten auf Unabhängigkeit der drei Gene, enge Kopplung kann ausgeschlossen werden.
Genetic studies on the resistance of barley to yellow rust (Puccinia striiformis West.)
Summary Five spring barley varieties resistant to race 24 of Puccinia striiformis were crossed with each other and with three susceptible varieties (Table 1) in almost all possible combinations. The F 2 generations and in two cases also the F 3, were inoculated in the greenhouse with race 24. F 2 generations of some of the crosses were, in addition, tested with race 24 in the field. Dependable classification of resistance and, consequently, unambiguous interpretation of results was possible only for the experiments carried out in the greenhouse. Although the field tests confirmed these results, they would not have been conclusive by themselves.The resistance of all of the five varieties studied is due to single recessive genes (Table 2). Three of the varieties have alleles at the same locus, the other two varieties have other resistance genes (Tables 3 and 4): Bigo — yr, Ab. 14 — yr, BBA 2890 — yr; Abed Binder 12 — yr2; I 5 — yr3. The results suggest that the three genes are inherited independently, close linkage can be excluded.

Summary Polymerization of various nucleoside-5-phosphorimidazolides has been conducted in neutral aqueous solution using divalent metal ions as catalysts. Oligonucleotide formation took place from each of the ribonucleoside-5-phosphorimidazolides, ImpC, ImpU, ImpA, ImpG, and ImpI. The yields and distributions of the resulting oligonucleotides varied depending on the difference of the nucleic acid base and the metal ions used. The catalytic effect of divalent metal ions on the formation of oligocytidylates occurred in the following order: Pb2+>Zn2+>Co2+, Mn2+>Cd2+>Cu2+>Ni2+>Ca2+, Mg2+, none >Hg2+. The order changes slightly for other types of oligoribonucleotide formation. Oligoribonucleotides up to hexamers were obtained in 35–55% overall yield, when Pb2+ ion was used as a catalyst. Zn2+ ions yielded oligoribonucleotides up to tetramers in 10–20% overall yield. The resulting oligonucleotides contained mainly 2–5 internucleotide linkages.Little or no oligonucleotide was obtained from nucleoside-5-phosphorimidazolides modified in the sugars, Imp(3-dA), Imp(2-dA), Imp(Ara), Imp(Aris), and Imp(Nep). The results indicate that a ribosyl system is required for the metal ion-catalyzed synthesis of oligonucleotides. Abbreviations. Short peptides connecting-helices and-strands have been analyzed in 240 proteins refined at resolutions of 0.25 nm or better. Connecting peptides of lengths between one and five residues have been classified as part of supersecondary motifs of four types:, , , and. Careful consideration has been given to the definition of secondary structures on the basis of hydrogen bonds and main-chain conformational angles. Using five classes of residue conformation—a, b, e, l, t—in the nonregular structure regions of, space, 34 classes of supersecondary motifs occurring at least five times have been identified. Among these 34 classes, 11 classes that occur more than 25 times are commonly occurring supersecondary structure motifs. The patterns and conformations of the 11 commonly occurring supersecondary structure motifs have been characterized, demonstrating that patterns and conformations adopted by supersecondary structure motifs are limited. The results have relevance to structure prediction, comparative modeling, and protein folding.  相似文献

MHC class II genes are major genetic components in rats developing autoimmunity. The majority of rat MHC class II sequencing has focused on exon 2, which forms the first external domain. Sequence of the complete open reading frame for rat MHC class II haplotypes and structure-based alignment is lacking. Herein, the complete open reading frame for RT1-B, RT1-B, RT1-D, and RT1-D was sequenced from ten different rat strains, covering eight serological haplotypes, namely a, b, c, d, k, l, n, and u. Each serological haplotype was unique at the nucleotide level of the sequenced RT1-B/D region. Within individual genes, the number of alleles identified was seven, seven, six, and three and the degree of amino-acid polymorphism between allotypes for each gene was 22%, 16%, 19%, and 0.4% for RT1-B, RT1-B, RT1-D, and RT1-D, respectively. The extent and distribution of amino-acid polymorphism was comparable with mouse and human MHC class II. Structure-based alignment identified the 65–66 deletion, the 84a insertion, the 9a insertion, and the 1a–1c insertion in RT1-B previously described for H2-A. Rat allele-specific deletions were found at RT1-B76 and RT1-D90–92. The mature RT1-D polypeptide was one amino acid longer than HLA-DRB1 due to the position of the predicted signal peptide cleavage site. The neutral sugars (glucose, fructose, and sucrose) and the sugar phosphates (glucose 6-phosphate, glucose 1-phosphate and fructose 6-phosphate) soluble in hot aqueous 80% methanol from the fibres of cotton — Gossypium arboreum L., G. barbadense L., and G. hirsutum L. — were determined at various stages of fibre development. In addition, the (13)--D-glucan content was measured and in the case of G. arboreum the rate of (13)--D-glucan and cellulose synthesis was determined with [14C]sucrose as the precursor. For each of the species a similar chronology was obtained for the changes in content of the various non-structural carbohydrates. At the early stages of secondary wall formation, glucose and fructose exhibited a maximum which was closely followed by a maximum in the (13)--D-glucan content and in the sugar phosphates. On the other hand, the sucrose content increased regularly until fibre maturity. Zusammenfassung Die Bostrychia-Caloglossa-Assoziation wächst vorwiegend auf Mangroven oder krautiggen Halophyten. Innerhalb des Bostrychietum wachasen die kleineren Assozienten auf den grossen. Zwischen Mangroven und Bostrychietum können Balaniden als zoologisches Substrat zwischengeschaltet sein. Auf der Bostrychia-Caloglossa-Assoziation siedeln nicht nur obligat Diatomeen und fakultativ Infusorien — suktorien — sondern Muschellarven setzen sich z.B. auf Bostrychia arbuscula zur ontogenetischen Ausbildung fest. Caloglossa — bevorzugt ältere Thalli — wird von Schnecken abgeweidet (von Ratnasabapathy in Südneuseeland beobachtet) und regeneriert anschliessend. Analog bilden hochkant stehende verrottete Blätter flutender Caloglossa ogasawaraensis — Räschen in strömendem Wasser marginale Adventivspro\serien, die zu neuen Horsten werden. Rheo-Wirkung kann sich bei Caloglossa — ausser generell langgestreckten Formen — auswirkwen in Torsion, Blatt-randwellung, Polykladie und zangenartigem Zusammenneigen ihrer Gabelspitzen (C. leprieurii). Bei Bostrychia — ausser Auflockerung der Versweeigungssysteme — in Ausbildung nurmehr latenter Hapteren (Flagellifulcratae) oder von langstieligen Puschelhapteren (Ramifulcratae), deren breite Terminalzone beim Fassen eines Halms als Saugscheibe fungiert. Die an rheo-Standorten zurücktretende Catenella ist nurmehr in toto langgestreckt fluitans entwickelt.F. libera von Murrayella periclados von recht tiefen Horizonten wie submerse Mangrovewurzeln wirkt gegenüber f. genuina etiolieer und ist als gigas-Form ausgebildet. Als Modus sekundären Wachstums kann — die ekortikate — Murrayella Hyphenrinde ausbilden, wie die unberindete subantarktische Bostrychia tenuis, während bei Bostrychia moritziana durch Längenwachstum die Zweigabgänge sekundär in die oberen Ecken der Rhachis-Perizentralen verschoben werden. Auch in sehr alten Bostrychia tenella-Rhachides von Ghana weisen gesprengte Zentralzellen auf sekundäres Längenwachstum und breite Querstreifen-Strukter überalterter Bostrychia scorpioides von Sierra Leone auf Dehnung des zentralen Sipho und hiermit in Zusammenhang stehendes sekundäres Dickenwachstum.Das für das schattige obere Litoral charakteristische Bostrychietum vergrünt bei Lichtexponierung. Die — berindeten — Bostrychien werden nun schwammartig und/oder krümmen ihre Spro\systeme dorsiventral beiderseits zur Rhachis und bodenwärts, hierdurch die Fruktifikationsorgane schützend.Am euryoxybionen Standort — Mangrove — bildt Caloglossa zur Schlammkompensierung Stämmchen aus (C. leprieurii var. hookeri). Ramifulkrate Bostrychien stelzen mittels ihre positiv geotroischen Hapterenzweige auf den stark verschalammten Borken der Mangroven, über die die grösseren Catenellen — durch den gleichen Hapterenmondus — gleichsam zu schweben scheinen.Für den west-venezolanischen Lago Maracaibo (s.1.) wurde erstmalig-für die Felsfazies — Bostrychia binderi , Bostrychia tenella , Caloglossa leprieurii und Catenella opuntia festgestellt, ferner Bostrychia radicans von Brückenbalken (lignikol).Antheridien für Mangrove-Catenellen bei C. impudica für die Monate Januar, April, Juli, bei C. nipae für Süd-Mai, Süd-Juni und August. Growth rate estimates () of phytoplankton populations that were sampled from nitrogen-limited continuous cultures and then incubated for short durations in batch culture with added14C-HCO3 were significantly different than steady-state growth rates () for 3 of 5 marine phytoplankton species. Two diatoms,Thalassiosira weissflogii andChaetoceros simplex, displayed virtually identical growth rates (=) over a wide range of, whereas for a third diatom,Phaeodactylum tricornutum, was overestimated by an average of 40% compared to. In contrast, was underestimated by the14C technique for the two remaining species: up to 40% at a steady-state of 1.0 day–1 for the chlorophyteDunaliella tertiolecta and up to 100% at of 1.4 day–1 for the haptophytePavlova lutheri. For the latter two species the divergence between and appeared to increase with increasing steady-state. A simple model of labeled and total carbon flow between the aqueous phase and cellular biomass was constructed to demonstrate that respiration was negligible when=, but was significant when>. In the cases in which<, a rapid physiological alteration presumably took place once the steady state was disturbed and cells were placed in the incubation chambers, which perhaps was related to the nutritional state of the cultures at the time of sampling. Questions thus are raised regarding our ability to measure accurately primary productivity from shipboard experiments with confined samples of phytoplankton from nutrient-impoverished waters that probably are less hardy than the laboratory cultures used in these studies.  相似文献

In this paper we use a dynamical systems approach to prove the existence of a unique critical value c * of the speed c for which the degenerate density-dependent diffusion equation u ct = [D(u)u x ] x + g(u) has: 1. no travelling wave solutions for 0 < c < c *, 2. a travelling wave solution u(x, t) = (x - c * t) of sharp type satisfying (– ) = 1, () = 0 *; '(*–) = – c */D'(0), '(*+) = 0 and 3. a continuum of travelling wave solutions of monotone decreasing front type for each c > c *. Summary Oligomerization of 5-TMP in water pools entrapped by dodecyl-ammonium chloride surfactant aggregates in benzene: hexane in the presence of dicyanodiimide at temperatures ranging from 21°–72° resulted in the formation of linear and cyclic oligonucleotides containing up to pentamers. Effects of temperature, time and surfactants have been examined. Rate constants for the formation of oligomers have been determined at five different temperatures. These data afforded values of H = 11.8 ± 1.9 Kcal mole–1, S=–53 6 e.u. and G = 27.4 4.0 Kcal mole–1. Prebiotic significance of these results are discussed.  相似文献

Summary The ultrastructure of spermatogenesis is described for the first time in an aspidogastrean. The zone of differentiation which is usually formed during digenean spermiogenesis was not observed inMulticotyle purvisi. Instead, spermatid components are assembled within the common cytoplasmic mass before the outgrowth of spermatids. Microtubules, mitochondrion, nucleus and axonemes including their basal body regions, migrate from the cytoplasm into the spermatid which is pinched off at the level of the arching membrane. An unusual, complex structure of the basal body region is described. Intercentriolar bodies and striated rootlets are left behind and quickly disappear from the residual cytoplasm. Recombinant Penicillium citrinum -1,2-mannosidase, expressed in Aspergillus oryzae, was employed to carry out regioselective synthesis of -d-mannopyranosyl-(12)-d-mannose. Yields (w/w) of 16.68% disaccharide, 3.07% trisaccharide and 0.48% tetrasaccharide were obtained, with 12 linkages present at 98.5% of the total linkages formed. Non-specific -mannosidase from almond was highly efficient in reverse hydrolysis and oligosaccharide yields of 45–50% were achieved. The products of the almond mannosidase were a mixture of disaccharides (30.75%, w/w), trisaccharides (12.26%, w/w) and tetrasaccharides (1.89%, w/w) with 12, 13 and 16 isomers. -1,2-linkage specific mannosidase from P. citrinum and -1,6-linkage-specific mannosidase from Aspergillus phoenicis were used in combination to hydrolyse the respective linkages from the mixture of isomers, resulting in -d-mannopyranosyl-(13)-d-mannose in 86.4% purity. The synthesised oligosaccharides can potentially inhibit the adhesion of pathogens by acting as "decoys" of receptors of type-1 fimbriae carried by enterobacteria.  相似文献

Summary Chris wheat possessed genes Sr5, Sr7a, Sr8a, Sr9g and Sr12. W3746, derived from the cross Chris/Baart, possessed Sr7a and Sr12. The response conferred by Sr7a was influenced by the genetic background. Although Sr7a or Sr12 alone conferred no observable resistance upon adult plants, the adult resistances of Chris and W3746 to predominant pathotypes appeared to be associated with the interaction of Sr7a and Sr12, or genes at closely linked loci.  相似文献

A highly purified natural -interferon (nIFN) was tested in vitro for direct and indirect antiproliferative activity against renal cell carcinoma (RCC), using a modified human tumor clonogenic assay and clinically achievable concentrations. In preclinical experiments, the indirect (cytokine-mediated) antiproliferative activity of nIFN was investigated using ACHN cells (established human RCC cell line). Continuous exposure to nIFN at concentrations of more than 5 IU/ml in the presence of feeder cells (a mixture of 5×104 monocytes/dish and 5×105 lymphocytes/dish, obtained from healthy donors) significantly inhibited colony formation of ACHN cells in comparison with growth inhibition in the absence of feeder cells (P<0.05). Various cytokines were measured in the supernatants lying over the medium on the feeder-layer agarose containing the same conditioned feeder cells. With IFN at 500 IU/ml, tumor necrosis factor (TNF) and IFN were detected at markedly high levels for 2–24 h. Neutralizing anti-TNF monoclonal antibody significantly reduced the indirect antiproliferative activity. Using our modified human tumor clonogenic assay technique, sufficient numbers of colonies for drug testing were observed in 19 of 31 surgical specimens (61.3%). In these clinical materials, nIFN at a clinically achievable concentration (50 IU/ml) significantly inhibited colony growth in the presence of feeder cells consisting of 5×104 monocytes/dish and 5×105 lymphocytes/dish, obtained from the patient whose tumor was examined (P<0.05). In colony-forming cases, a significant correlation between the percentage colony survival and TNF concentration in the supernatant was observed (r=–0.95,P<0.01). These results suggest that this assay system may be an appropriate technique for evaluating the antiproliferative activities of nIFN involving cytokine-mediated action, and that TNF may play an important role in this cytokine-mediated activity.This work was supported in part by a grant-in-aid for promoting research (no. 02771010) from the Ministry of Education  相似文献

In the present study of 9,000 individuals representative of the general population, we have considered whether the addition of common single nucleotide polymorphisms (SNPs) in the promoter region of Apolipoprotein E (APOE) improve the statistical explanation of variation in lipid traits and test the hypothesis that the estimated genotype effects are independent of factors indexed by gender and age. To address these questions, we have asked, for each gender and for each 20-year age strata (young: 20–39 years; middle-aged: 40–59 years; old: 60–79 years; very old: 80–100 years), how much trait variation is associated with the traditional 2, 3, and 4 allelic variations defined by the g.2059TC and g.2197CT SNPs in the fourth exon of the APOE gene, and how much additional trait variation is associated with genotypes defined by combining the g.2059TC and g.2197CT SNPs with one, two, or three promoter SNPs. Our study demonstrates that the pleiotropic effects of genotype variation defined by the traditional 2, 3, and 4 alleles on five plasma measures of lipid metabolism manifest differently in women and men and change significantly during the life cycle for high-density lipoprotein cholesterol in women. Multi-site genotypes defined by adding SNPs located in the 5 promoter region to the traditional g.2059TC and g.2197CT SNPs doubled the estimate of genetic variance of high-density lipoprotein and apolipoprotein Al in middle-aged females.  相似文献

Primers were developed for 118 microsatellites isolated from grape (Vitis vinifera) genomic libraries enriched for (AC)n repeats. Only one microsatellite sequence matched other grape SSR-sequences in the GeneBank database. Genotyping was carried out in the parental lines and four offspring of two pseudo-test-cross populations, Cabernet Sauvignon x Seyval and Chardonnay x Bianca, and a further six other grape genotypes (V. vinifera Sultanina, Merlot, Syrah, Müller-Thurgau, Vitis Regent and V. riparia Gloire de Montpellier). A total of 108 microsatellites showed easily scorable alleles and 100 of them segregated according to a configuration suitable for mapping in either cross. A further 8 SSRs, although unsuitable for mapping in those crosses, showed polymorphism in the other genotypes tested. This set of markers was used, along with 75 microsatellites of other repeat-types, to fingerprint 46 offspring of the cross Chardonnay x Bianca. For each full-sib, individual heterozygosity and distance in repeat units between pairs of alleles at each locus (mean d2) were calculated as a tool for predicting highly outbred recombinant individuals. Six microsatellites with segregation ratios significantly distorted towards the lack of homozygous sibs were identified and mapped to linkage groups LG 3 and LG 5. Estimation of heterozygosity at genome-wide level and genotyping at loci for which homozygous sibs are discriminated against are discussed for marker-assisted background selection in outcrossing grapevines.  相似文献

Summary Four types of chromosomes with a deletion between the human embryonic and globin genes were identified among 2.8% of 321 Black Americans from Georgia. Two deletions of approximately 11 kb which differed by about 300 bp occurred on chromosomes with or without a polymorphic Xba I site 5 to the globin gene [(X+) or (X-)]. The deletions are identifiable in Xba I digests of genomic DNA using an or a globin gene probe which yield fragments of 23 kb from (X+)–* chromosomes or 27 kb from (X)–* chromosomes. Digestion with other enzymes and probing with both and probes gave fragments typical of the two globin gene deletions previously identified in Polynesians. Among Black Americans, these globin gene deletions have been found in combination with globin gene deletions in trans but not in cis. Homozygotes have not been found. Hematologic data on carriers of the globin gene deletions in association with Hb AS, SS, and SC suggest that these deletions have no effect on the function of the adult globin genes.  相似文献

UDP-GlcNAc:Man1-3R 1-2-N-acetylglucosaminyltransferase I (GlcNAc-T I; EC 2.4.1.101) catalyses the conversion of [Man1-6(Man1-3)Man1-6][Man1-3]Man-O-R to [Man1-6(Man1-3)Man1-6] [GlcNAc1-2Man1-3]Man-O-R (R=1-4GlcNAc1-4GlcNAc-Asn-X) and thereby controls the conversion of oligomannose to complex and hybrid asparagine-linked glycans (N-glycans). GlcNAc-T I also catalyses the conversion of Man1-6(Man1-3)Man-O-octyl to Man1-6(GlcNAc1-2Man1-3)Man-O-octyl. We have therefore tested a series of synthetic analogues of Man1-6(Man1-3)Man-O-octyl as substrates and inhibitors for rat liver GlcNAc-T I. The 2-deoxy and the 3-, 4- and 6-O-methyl derivatives are all good substrates confirming previous observations that the hydroxyl groups of the Man1-6 residue do not play major roles in the binding of substrate to enzyme. In contrast, all four hydroxyl groups on the Man1-3 residue are essential since the corresponding deoxy derivatives either do not bind (2- and 3-deoxy) or bind very poorly (4- and 6-deoxy) to the enzyme. The 2- and 3-O-methyl derivatives also do not bind to the enzyme. However, the 4-O-methyl derivative is a substrate (K m =2.6mm) and the 6-O-methyl compound is a competitive inhibitor (K i=0.76mm). We have therefore synthesized various 4- and 6-O-alkyl derivatives, some with reactive groups attached to anO-pentyl spacer, and tested these compounds as reversible and irreversible inhibitors of GlcNAc-T I. The interaction of heme-free (o) and heme-containing (h) chains of human hemoglobin has been monitored in 0.1 M potassium phosphate buffer, pH 7 or 8, at [5°C. Soret zero and first-derivative spectra were consistent with a uniform association reaction. Stopped-flow investigations demonstrated association rates on the order of 107 M–1 s–1. This was 100-fold more rapid than the reported rate of combination of h and h proteins. This encounter-like rate of semi--hemoglobin (oh) formation was increased by raising the pH from 7 to 8. pH change is known to affect the spatial arrangement of AB—GH helical entities. Molecular graphic analysis of modeled o protein superimposed over native h protein revealed an apo Mb-like structure with well-defined AB—GH segments. Repositioning of these core helical segments, resulting in increased conformational freedom of the 11 interface, was apparently responsible for the enhanced association properties of the o protein.  相似文献

DNA polymorphism patterns linked to the A-globin gene were analyzed in healthy Japanese using four different restriction endonucleases. The chromosomes with the A-globin gene were mapped through an evaluation of the presence of seven different restriction sites (HincII 5 to ; HindIII in G and A; HincII in, and 3 to, 1; AvaII in ; Bam-HI 3 to ). Among 36 chromosomes analyzed, 20 chromosomes had a haplotype of [+–––––+]. Among 55 individuals examined, 7 possessed a homozygous haplotye of [+–––––+]. All Japanese with the AT-globin gene had a subhaplotype of [–++–+] 5 to the -globin gene. Their major haplotypes were [–++–+–+] and [–++–++–]. It was expected that the presence of the AT-globin gene in Japanese may be deduced from subhaplotypes 5 to the -globin gene.  相似文献

DNA polymerases II () and III() are the only nuclear DNA polymerases known to possess an intrinsic 3 5 exonuclease in Saccharomyces cerevisiae. We have investigated the spontaneous mutator phenotypes of DNA polymerase and 3 5 exonuclease-deficient mutants, pol3-01 and pol2-4, respectively. pol3-01 and pol2-4 increased spontaneous mutation rates by factors of the order of 102 and 101, respectively, measured as URA3 forward mutation and his7-2 reversion. Surprisingly, a double mutant pol2-4 pol3-01 haploid was inviable. This was probably due to accumulation of unedited errors, since a pol2-4/pol2-4 pol3-01/pol3-01 diploid was viable, with the spontaneous his7-2 reversion rate increased by about 2 × 103-fold. Analysis of mutation rates of double mutants indicated that the 3 5 exonucleases of DNA polymerases and can act competitively and that, like the 3 5 exonuclease of DNA polymerase the 3 5 exonuclease of DNA polymerase acts in series with the PMS1 mismatch correction system. Mutational spectra at a URA3 gene placed in both orientations near to a defined replication origin provided evidence that the 3 5 exonucleases of DNA polymerases and act on opposite DNA strands, but were in sufficient to distinguish conclusively between different models of DNA replication.  相似文献
