Forty varieties of vegetable, cereal, fodder and grass seeds at 10, 13 and 16% moisture content were fumigated with methyl bromide at 10 and 18oC, to determine the maximum safe moisture content required to avoid unacceptable phytotoxicity. Dosages (concentration x time products) of 200 and 400 mg h/1 of technical and commercial methyl bromide were used and treated seeds were compared with controls in germination tests. Results were not consistent, but in c. 30 % of the comparisons lower moisture contents, lower temperature and lower dosage each resulted in better germination than the corresponding higher values. The reductions in germination of treated samples were usually less than 5 % and the overall average reduction was only 2.3%. This work showed that under controlled conditions the treatments are safe for most of the tested varieties of seed, but for some seeds, particularly white clovers, Agrostis and runner beans, the highest moisture content (16%) should be avoided.  相似文献

Magnolia dealbata is a deciduous tree species in danger of extinction. Seed ecology data and successful germination treatments are important for conservation and species reintroduction programs. This study reports about seed ecology and germination treatments of M. dealbata Zucc. The results showed that the seeds, influence of vertebrates excluded, persisted viable after 1 year, suggesting the presence of a semi-persistent seed pool. There was no significant difference (p>0.05) in survival of seeds with or without sarcotesta after a year. Significant differences (p<0.05) were found in the number of seeds surviving after a year at different levels of burial. There was no significant interaction upon survival (p>0.05) between sarcotesta presence or absence in the seed and burial depth. If vertebrates were not excluded the seeds had a removal rate of 100% 8 months after being exposed. Seed survival curve was of Type II, that is, with constant mortality throughout the time. The most successful germination treatment (p<0.05) with 100% germination was soaking of seeds without sarcotesta for 24 h at room temperature (18–20 °C).  相似文献

Summary Immunolocalization of alkaloids in lupin seeds (Lupinus spp.) has been performed by cryofixation and conventional methods. Alkaloids were localized in the protein bodies of the cotyledon cells. Some immunogold particles in the walls of these cells were also observed. There were no differences in the sites of localization between the two mentioned methods. X-ray microanalysis of elements showed the presence of P, Mg, S, and K in the protein bodies of cotyledon cells in lupin seeds. The role of K+ in alkaloids transport is discussed.  相似文献

Larvae of Utetheisa ornatrix (L.)(Lepidoptera: Arctiidae) are found mainly inside unripe pods of several alkaloid‐bearing Crotalaria (Fabaceae) species. Although eggs are laid on the leaves, the larvae are usually found feeding on unripe seeds in the pods. In this work, we investigated the selective pressures that could explain why U. ornatrix larvae feed primarily on unripe pods with seeds and not on leaves. Our results showed that larval survivorship in the laboratory was unaffected by feeding on leaves or unripe seeds, and that larval development up to the pupal stage was better in larvae that fed on unripe seeds, although perforating unripe pods to reach seeds was costly in terms of survivorship. Females were also heavier when fed on unripe seeds, but there was no significant difference in the fecundity of females fed either of the two diets. Feeding on unripe seeds in pods had other benefits for U. ornatrix, including a lower predation rate for larvae that fed inside compared to larvae that fed outside the pods. Similarly, adults derived from larvae that fed on unripe seeds were preyed upon less frequently by the orb‐weaving spider Nephila clavipes than were adults that fed on leaves. The latter benefit may be closely related to the high concentration of pyrrolizidine alkaloids in unripe seeds, which is about five times more than in leaves. These alkaloids are sequestered by the larvae and transferred to adults, which then become chemically protected. However, this chemical defence does not protect the larvae against ants such as Ectatomma quadridens and Camponotus crassus. Pods with unripe seeds that confer physical protection to larvae and pyrrolizidine alkaloids that confer chemical protection to adults limit the use of leaves by U. ornatrix larvae.  相似文献

We explored different treatments to enhance the probability of sowed seeds of two early successional (ES, Cecropia obtusifolia and Ochroma pyramidale) and two late successional (LS, Brosimum costaricanum and Dialium guianense) species to escape predation and germinate in abandoned cattle‐raising pasture fields in Southeastern Mexico. ES species were sown in groups of 50 seeds under three treatments: invertebrate exclusion, burial, and exposition to seedeaters. LS species were sown in groups of 10 seeds under three treatments: vertebrate exclusion, burial, and exposition to seedeaters. We registered seed predation and germination 2, 4, 8, 16, 32, and 64 days after the initial sowing. Overall, ES showed higher predation rates (mean ± SE = 0.45 ± 0.07 seed seed?1 day?1; n = 3) than LS species (0.09 ± 0.02 seed seed?1 day?1). Cecropia obtusifolia was completely predated in all treatments after 8 days. Burial and exclusion treatments reduced final predation in circa 6% for O. pyramidale, relative to that of exposed seeds (85% after 8 days); most germination occurred in buried seeds (3.7%). In B. costaricanum, burial enabled germination by 10%; exposed and excluded seeds were removed 100%. Dialium guianense showed 12% germination in buried seeds and circa 20% of the seeds were not removed after 64 days. Direct sowing would be a recommended rainforest restoration practice for species with relatively large seeds if deposited in groups and buried. Studies which address variation across numerous sites are necessary in order to generate more consistent seed predation patterns and rainforest restoration principles in tropical pastures.  相似文献

Seeds of Datura ferox were collected in soybean fields, grouped into four categories according to the degree of fruit maturity and placed just level with the soil surface or buried to a depth of 7 or 15 cm. Seed survival after 8 months was c. 30% when the seeds were left on the soil surface, but between 40 and 90% when the seeds were buried. The riper seeds were the more persistent. A similar pattern was observed after 20 months. Seedling emergence was negatively related with the degree of seed maturity and depth of burial, but it was never great enough to explain seed losses. In another experiment seeds from ripe capsules were superficially sown and the soil was: (0) left undisturbed, (1) cultivated in late winter or (2) cultivated in late winter and late spring. In all cases there was a small flush of seedlings at the beginning of the first spring; other flushes occurred only after soil cultivation and were larger during the second spring than during the first. In the plots cultivated twice a year (2) the seedlings that emerged in the first spring represented c. 4% of the initial seed bank. Survival after 20 months was about 25% in all treatments. In an arable field under soybean cropping the seedling flushes during the spring months were related to the pattern of soil cultivation. The seedlings that emerged after crop drilling represented c. 18% of the seed bank (estimated before drilling). Implications for weed management are discussed.  相似文献

With many degraded environments undergoing restoration efforts, there is a growing need for the optimization of direct seeding practices. Seeds planted on wildlands are often consumed by rodents, leading to reduced plant establishment. Coating seeds in rodent aversive products may prevent seed‐predation. We tested 10 seed‐coating formulations containing products expected to deter rodents, namely: ghost and cayenne pepper powders; essential oils from bergamot, neem, and pine; methyl‐nonyl‐ketone, anthraquinone, activated carbon, beta‐cyclodextrin, and a blank coating containing no rodent deterrents to serve as a control treatment. Each treatment was applied to Pseudoroegneria spicata (bluebunch wheatgrass) seeds. These seeds germinated similarly to uncoated control seeds unless the coating contained methyl‐nonyl‐ketone which reduced germination. When seeds were offered to Ord's kangaroo rats (Dipodomys ordii), they strongly avoided the treatments in favor of uncoated control seeds. Notably, the blank coating, lacking active ingredients, still elicited 99% avoidance. However, these results indicated behavior when alternative food sources are readily available, a scenario rare in nature. To address this, a second feeding experiment was conducted to observe D. ordii's behavior under calorie‐restricted conditions. D. ordii were subjected to a fast period, then offered only one treatment. Under these conditions, many subjects chose to consume coated seeds, but to a lesser degree than subjects offered control seeds. Seeds coated in ghost pepper, neem oil, and activated carbon reduced consumption by 47–50%. Given these lab results, we would expect these treatments to increase native plant establishment following the direct seeding of wildlands by protecting seeds from rodent predation.  相似文献

Summary Certain aspects of selective toxicity of 1,3,7-trimethylxanthine (1,3,7-T), a natural herbicide, were studied inPhaseolus mungo andAmaranthus spinosus. Treatment of seeds ofA. spinosus with 1,3,7-T (sub-lethal concentration) caused a marked decrease (29.5%) in amylase activity. Kinetic studies with respect to substrate saturation and Km values as well asin vitro treatments of the cell-free enzyme indicated no effect on its catalytic property. The inhibitory effect of 1,3,7-T on amylase activity could not be counteracted with treatments of GA3. A general biochemical analysis revealed that the starch/sugar ratio increased in the seedlings grown from treated seeds. Analysis also showed a higher ratio for insoluble/soluble nitrogen. It is suggested that inhibition of germination is caused by reduced synthesis of amylase which is not mediated through gibberelic acid. Treatment of seeds ofPhaseolus mungo with, 1,3,7-T. caused no change in amylase activity,in vivo nitrate reductase activity, ethanol soluble and insoluble nitrogen and protein content.  相似文献

The quantitative and qualitative aspects of seed dispersal by the western lowland gorilla (Gorilla gorilla gorilla) were investigated in Gabon. Fresh faeces were collected and washed to identify and count the seeds. Seed germinability after gut passage was estimated with trials in a nursery at the study site. To assess the impact of gut passage on germination success and delay, comparative trials were run with four treatments: (i) gut passed seeds cleaned of faeces, (ii) gut passed seeds within a faecal matrix, (iii) seeds from fresh fruits surrounded by pulp, and (iv) seeds from fresh fruits cleaned of pulp. The analysis of 180 faecal units resulted in the identification of 58 species of seed. Germination trials were realized for 55 species and the mean germination success reached 46%. The impact of gut passage was investigated for Santiria trimera and Chrysophyllum lacourtianum; both species displayed higher germination success after ingestion. This study shows that gorillas effectively disperse seeds of numerous plant species, many of which provide timber or nontimber forest products or are typical of Gabonese forests. Considering the high‐quality of gorilla deposition sites, gorillas is thought to play a unique role in the dynamics of Central African forest.  相似文献

以野鸦椿(Euscaphis japonica)种壳和胚为材料,甲醇浸提得到种壳浸提液和胚浸提液,配制浸提液浓度梯度为原浸提液浓度的10%、20%、30%、40%,研究不同浓度的种壳和胚浸提液对白菜、小麦、绿豆种子底物酶活性、发芽率、幼苗根长和苗高的影响,萃取和分离种壳和胚甲醇浸提液中的内源抑制物质,探讨野鸦椿种子内源抑制物质的活性与成分。结果表明:随着种壳和胚浸提液浓度的增加,白菜种子酸性磷酸酶活性和发芽率均显著降低(P0.05),表现为抑制作用递增,种壳的抑制作用小于胚,而幼苗的根长和苗高则表现为低促高抑,在10%浸提液处理下根长和苗高达最大值,种壳的促进效果弱于胚;小麦种子淀粉酶活性及幼苗的根长和苗高递减(P0.05),表现为抑制作用递增,而发芽率则在浓度≤20%时差异不明显(P0.05),浓度为30%时显著降低(P0.05),40%时发芽率为0,种壳的抑制作用大于胚;绿豆种子蛋白酶活性、发芽率、幼苗根长和苗高均在浸提液浓度≥20%时显著下降(P0.05),且种壳的作用效果小于胚。种子内源抑制物萃取及分离表明,外壳中含酚酸类较胚多,含碱类较胚少。综上认为,野鸦椿种壳和胚中均含有较高活性的内源抑制物,但性质、成分及含量存在差异,外壳内源抑制物主要作用对象为淀粉类物质,胚乳内源抑制物主要作用于油脂类和蛋白类物质。  相似文献

Determination of fourteen alkaloids, toxic Aconitum alkaloids, aconitine, mesaconitine, jesaconitine, hypaconitine and deoxyaconitine, and their hydrolysis products, benzoylaconines and aconines, have been established using capillary liquid chromatography (LC) fast atom bombardment mass spectrometry (FAB-MS) with a frit interface. Protonated molecular ions were observed as base peaks in the FAB-MS for these fourteen alkaloids. All the alkaloids were simultaneously quantified with linear gradient LC elution by solvent mixture of acetonitrile and 0.3% trifluoroacetic acid using selected ion monitoring of the protonated molecular ions. The calibration curves of these alkaloids were linear in injection amounts ranging from 5 to 500 pg, and their detection limits were 1 pg per injection (S/N=3). Solid-phase extraction using Sep-Pak Plus PS-1 was also investigated to clean-up and concentrate alkaloids in blood and urine samples, and showed satisfactory recoveries. This capillary LC–frit-FAB-MS method enables determination of low levels of Aconitum alkaloids in blood and urine samples, coupled with solid-phase extraction.  相似文献

Aims: The efficacy of a commercial seed washer and 1 and 3% peroxyacetic acid or 20 000 ppm calcium hypochlorite for reducing Salmonella on alfalfa seeds was investigated. Methods and Results: Alfalfa seeds were inoculated with Salmonella Stanley to achieve c. 5 log CFU g?1. Seeds were then treated with 1 or 3% peroxyacetic acid or 20 000 ppm calcium hypochlorite for 15 min in a commercial seed washer that uses air to enhance contact of the sanitizer with the seed. Experiments were also conducted using industry and laboratory methods. An c. 1‐log reduction in number of Salm. Stanley was demonstrated regardless of the chemical treatment or method of treatment. Although this 1‐log reduction was significant (P < 0·05), differences among the treatments were not significant. Treating the seed with 1 and 3% peroxyacetic acid resulted in similar Salm. Stanley reductions of 1·77 and 1·34 log, respectively, not being statistically significant (P > 0·05). Conclusions: These results suggest that under conditions tested, 1 or 3% peroxyacetic acid solutions are equally effective as 20 000 ppm of Ca(OCl)2 in the reduction of Salm. Stanley on alfalfa seed when used in conjunction with a commercial seed washer. Significance and Impact of the Study: A 1% peroxyacetic acid solution could potentially be used in place of 20 000 ppm of Ca(OCl)2 for treatment of seeds used for sprouting. The commercial seed washer did not enhance removal of Salm. Stanley from alfalfa seeds, but did facilitate removal of excess soil from seeds.  相似文献

  • Crop wild relatives can be a useful source of genotypes that maximise crop survival and yield in specific habitats. Lupinus angustifolius is an annual forb with crop varieties derived from a narrow genetic basis but that are cultivated worldwide. Its seeds have high nutritional value, but they naturally contain alkaloids with anti‐nutritive factors. The study of its wild populations can be useful to find genotypes that contribute to higher climate resilience and greater yield under stressing environmental conditions.
  • Using a common garden, we evaluated ecotypic differentiation in four natural populations from two contrasting latitudes in terms of plant biomass, seed mass and number, alkaloid content in seeds for the three main alkaloids present in the plant and seed colour, including its possible influence on post‐dispersal predation. Correlations among traits were also assessed.
  • We found differences among populations for all traits except final biomass. Northern populations had lighter seeds and a tendency to yield more seeds when they produced white seeds, compared to southern populations and variegated seeds. Regardless of latitude, populations showed differences in alkaloid concentration, with all three alkaloids found generally in high or low concentrations in each population. Proportion of white seed morphs varied in each population. Seed colour did not influence predator preference. In addition, white seed colour was related to a low alkaloid content.
  • Our results evidence the existence of natural ecotypic differentiation in L. angustifolius not only due to latitudinal range, but also to local environmental factors. White seed coat colour could be used as a visual clue for identification of low‐alkaloid genotypes, a priority trait in L. angustifolius breeding programmes.

Due to the exceptionally hot and dry summer in 2003 the ergot of that harvest was rather small and could only be separated from normal grain with increased efforts. Based on a clean-up procedure of Wolffet al. (1) and of Kluget al. (2), a HPLC-FLD-method for the determination of 12 ergot alkaloids (6 “In”-, 6 “Inin”-forms) was established and modified. Actually reference substances are commercially available only for 5 selected alkaloids. Because of the instability of the alkaloids a new standard preparation procedure was tested and implemented. The maximum allowed impurity with ergot (0.05%=1000 μg alkaloids/kg) was exceeded in samples of harvest 2003. Except for one sample, all exceedings were detected in conventionally grown products, unlike organically grown products. The aim of this study was to determine the nutrient digestibility of seeds of four varieties of narrow-leafed lupines (Lupinus angustifolius) and the possibility of soya bean meal (SBM) substitution by lupine seeds alone and in combination with rapeseed meal (RSM) in the diets of pigs. The seeds of the lupine varieties Kalif, Sonet, Zeus and Boruta were analysed. The apparent total tract digestibility (ATTD) was determined on 50 cross-bred pigs using the difference method with titanium dioxide as a marker. The substitution of SBM by lupine seeds alone (at 0 – 100%) was tested on 60 pigs (20–105 kg body weight (BW)) and by a combination of lupine seeds and RSM on 180 fattening pigs (35–80 kg BW). The chemical composition of lupine seeds differed considerably, especially in terms of crude protein and mineral content. All seeds contained less than 0.05% alkaloids and 9.3% oligosaccharides in dry matter. The ATTD of protein ranged from 70% to 74%, those of ether extract from 36% to 55% and those of gross energy from 77% to 84%. The entire replacement of SBM by lupine seeds (var. Sonet) did not have a negative effect on the performance of grower and fattener pigs. The substitution of SBM by a combination of lupines and RSM reduced the performance of growing and finishing pigs significantly.  相似文献

When seeds of two rice cvs. Ratna and Jaya were germinated under increasing levels of cadmium nitrate (0, 100 and 500 μM) in the medium, a marked decrease in germination percentage was observed with Cd treatments, as compared to controls. There was more absorbed Cd in embryo axes than in endosperms. More uptake resulted with increasing Cd levels in the growth medium in embryo axes. In both rice cultivars, during a germination period of 0 – 120 h, an increased level of protein as well as free amino acids was noted in Cd treatments. Protease activity in general decreased in both embryo axes as well as endosperms due to Cd treatment. In vitro studies showed an enhancement in protease activity in Cd treatments at low Cd levels (50–100 μM), whereas concentrations above this caused inhibition in enzyme activity. Under 500 μM Cd treatments in vivo there was about 30 to 50 percent decline in leucine aminopeptidase (LAP) activity in endosperms, however, carboxypeptidase activity showed a marked increase in endosperms beyond 24 h under Cd treatments. In embryo axes of germinating seeds there was always a decline in peptidase activities, under the influence of cadmium. The leucine amino peptidase and protease activity were always greater in embryo axes in cv. Ratna than cv. Jaya. However, the carboxypeptidase activity was higher in Jaya when compared to Ratna in endosperms under Cd treatments. The results suggest possible suppression of protease and peptidase activities due to Cd treatments in germinating rice seeds leading to altered levels of protein and amino acids.  相似文献

内生真菌是一类共生于植物体内,能够不同程度影响宿主植物生态适应性和竞争能力的微生物。分析内生真菌在豚草种子中的分布、种群结构,以及内生真菌发酵液对种子发芽和幼苗生长的作用。结果显示:发生于6个地区的豚草种子均能分离获得内生真菌,分离率在19%—92.63%之间,不同地区之间差异极显著(P0.01)。内生真菌主要存在于种子的总苞部位,分离率达到65.52%。发生于福建省长乐市松下镇的豚草种带内生真菌种群包含5个属,以链格孢属(Alternaria)真菌为优势菌群,占82.26%;其次为镰孢属(Fusarium)真菌,占9.68%;其它3个属的真菌发生较少,均低于5%。内生真菌主要以水平传播方式在豚草不同世代之间传播。供试的7个内生真菌菌株的发酵液均不同程度地抑制豚草种子发芽,降低幼苗地上部高度、根长度、根数量和总生物量,但不同菌株发酵液之间抑制程度差异明显,显示不同菌株对豚草种子发芽和幼苗生长产生不同的影响。内生真菌发酵液处理后的种子仍然保持较高程度的活力;不同内生真菌发酵液处理后,有活力的种子维持在50%—87.5%之间,均高于(或等于)清水处理的种子,说明内生真菌代谢产物只是抑制种子的发芽,但并不导致种子的腐烂和死亡。这些研究结果初步显示种子携带的内生真菌可能在豚草入侵生物学中发挥重要的作用。  相似文献

At concentrations of 0.01–1 mM, five synthetic multiring analogs of strigol were effective germination stimulants of intact and dehulled wild oat (Avena fatua L.) seeds. The effect was concentration-dependent and equaled or exceeded that produced by equimolar gibberellic acid (GA3). The most effective strigol analog treatments induced 55–80% germination within 7 days in intact wild oat seeds and resulted in 63–86% germination and normal seedling growth over 14 days. Intact wild oat controls germinated 14% after 14 days. The stimulation of wild oat germination by these synthetic strigol analogs demonstrates that these compounds, initially developed as germination stimulants for the seeds of the parasitic weed, witchweed (Striga asiatica L. Kuntz.), have bioregulatory activity in dormant seeds of monocots, as well as dicots. None of the compounds tested significantly affected the germination of nondormant cultivated oat seeds (Avena sativa L.). 为了解濒危物种南方红豆杉(Taxus chinensis var. mairei)种子内含物含量受温度和湿度层积的影响,设置4个季节、2种湿度(16%和24%)基质层积处理,对种子的可溶性糖、淀粉、可溶性蛋白和脂肪等内含物质的变化进行研究。结果表明,不同层积处理下种子贮藏物质的含量有显著变化,春季层积9个月后,可溶性蛋白含量达到最高值;可溶性糖含量呈现降低-升高-降低的变化趋势;淀粉和脂肪含量均随层积逐渐减少。秋季层积9个月后,淀粉含量降至最低。相比于24%湿度,16%湿度的春季、秋季、冬季层积9个月后,脂肪含量均减少较多,说明16%湿度下种子代谢活动更强。春季和秋季的暖温更能促进种子代谢,促进种子形态后熟。夏季温度过高,导致种子生活力下降,夏季层积处理3个月后,种子已经发霉和腐烂。层积过程中,种子内含物在相关酶的作用下,降解为可溶性蛋白、可溶性糖等,为种子萌发提供物质与能量。种子层积时间、温度和湿度及交互作用可作为种子内含物的调控因子。  相似文献

ABSTRACT Agricultural intensification is a key factor in the decline of many avian populations throughout the world, yet the exact mechanisms that contribute to these declines are relatively unresolved, especially for seed-eating species. We tested the hypothesis that forage quality, particularly protein content, limits productivity in birds consuming diets dominated by agricultural grains and lacking in native forages. We used white-winged doves (Zenaida asiatica) because columbids are highly granivorous and white-winged doves have declined in areas dominated by grain agriculture. We randomly assigned 52 pairs of captive doves to 1 of 2 treatments: we fed one treatment sorghum (Sorghum bicolor) and the second sorghum plus native seeds (croton [Croton sp.] and native sunflower [Helianthus sp.]). Birds with access to native seeds fledged 123% more young per pair, with young weighing 32% more at fledging than those fed only sorghum. Doves with access to native seeds selected a diet consisting of 43% sorghum, 32% croton, and 25% sunflower. We conclude that agricultural grains, which typically have high metabolizable energy and low to moderate protein concentrations, are not sufficient for normal productivity of white-winged doves. The lack of native forages with sufficient metabolizable energy and protein content may be a factor in the declining productivity of white-winged doves in the lower Rio Grande Valley of Texas, USA, as well as other species in regions dominated by agriculture.  相似文献
