Thrips tabaci (Lindeman) is an important polyphagous pest, and vector of Tomato spotted wilt virus responsible for sporadic, but devastating epidemics in potato. T. tabaci shows significant preferential differences between potato cultivars that may be important for field resistance. To investigate the role of visual cues in host choice we tested colour preference using two-choice assays. Thrips tabaci showed a significant preference for mid-green over red, blue, and white coloured cards, a preference for both light-green and mid-green over dark-green, and light-green over yellow, but no preferential difference between mid-green and yellow, and between light-green and mid-green. Analysis of the spectral reflectance of potato cultivars differing in thrips preference, revealed significant differences within the 400–700 nm wavelengths. In most cases cultivars preferred by T. tabaci had lighter green foliage and higher reflectance at 552 nm, and thus colour preference may be important for host selection. Oviposition choice of T. tabaci for potato cultivars was determined from counts of larvae and unhatched eggs from leaf disks under choice and no-choice conditions. In contrast to the colour choice experiments, onion thrips showed oviposition preference for cultivars with darker green foliage and lower reflectance at 552 nm.  相似文献

Two aspects of the within-plant distribution of Thrips tabaci Lindeman (Thysanoptera: Thripidae) on onion, Allium cepa L., plants were investigated: 1) diurnal variations in the distribution of adults and larvae between basal and upper sections of onion leaves, and 2) between-leaf and within-leaf distribution of the eggs. The diurnal investigations showed that higher proportions of larvae than of adults congregated at the basal sections of plants, particularly when plants were young and thrips density was low. As plants matured and thrips density increased, the larvae became more dispersed. Regardless of plant size, there were always more adults in the upper than basal plant sections. There were no clear time-windows during the 24-h diurnal cycle when more thrips were in the upper plant parts. T. tabaci eggs were laid everywhere in the plant. Leaves of intermediate ages had more eggs than older or younger leaves. Within leaves, the white leaf sheath received the least eggs and leaf tips received slightly more eggs than leaf sheaths. The highest egg density was found between the green leaf base and the leaf tips. Regardless of plant size, more than half of all eggs were laid above the basal sections. The percentage increased to >95% in mature plants. Except when plants were small the outer leaves were preferred over inner leaves and upper leaf sections preferred over lower leaf sections as egg-laying sites by adults. Implications of the results in the management of T. tabaci are discussed.  相似文献

Development of insecticide resistance in onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), populations in onion (Allium spp.) fields and the incidence of the T. tabaci transmitted Iris yellow spot virus have stimulated interest in evaluating alternative management tactics. Effects of straw mulch applied in commercial onion fields in muck areas of western New York were assessed in 2006 and 2007 as a possible onion thrips management strategy. In trials in which no insecticides were applied for thrips control, straw mulch-treated plots supported significantly lower T. tabaci populations compared with control plots. In both years, the action thresholds of one or three larvae per leaf were reached in straw mulch treatments between 7 and 14 d later than in the control. Ground predatory fauna, as evaluated by pitfall trapping, was not increased by straw mulch in 2006; however, populations of the common predatory thrips Aeolothrips fasciatus (L.) (Thysanoptera: Aeolothripidae) were significantly lower in straw mulch plots in both years. Interference of straw mulch in the pupation and emergence of T. tabaci was investigated in the lab and their emergence was reduced by 54% compared with bare soil. In the field the overall yield of onions was not affected by the straw mulch treatment; however, the presence of jumbo grade onions (>77 mm) was increased in 2006, but not in 2007. These results indicate that populations of T. tabaci adults and larvae can be significantly reduced by the use of straw mulch without compromising overall onion yield. The use of this cultural practice in an onion integrated pest management program seems promising.  相似文献

During the past two decades, onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), has become a global pest of increasing concern in commercial onion (Allium cepa L.), because of its development of resistance to insecticides, ability to transmit plant pathogens, and frequency of producing more generations at high temperatures. T. tabaci feeds directly on leaves, causing blotches and premature senescence as well as distorted and undersized bulbs. T. tabaci can cause yield loss > 50% but can be even more problematic when it transmits Iris yellow spot virus (family Bunyaviridae, genus Tospovirus, IYSV). IYSV was identified in 1981 in Brazil and has spread to many important onion-producing regions of the world, including several U.S. states. IYSV symptoms include straw-colored, dry, tan, spindle- or diamond-shaped lesions on the leaves and scapes of onion plants and can cause yield loss up to 100%. Here, we review the biology and ecology of T. tabaci and discuss current management strategies based on chemical, biological, and cultural control as well as host resistance. Future directions for research in integrated pest management are examined and discussed.  相似文献

Factors influencing the resistance of onion (Allium cepa) cultivars to onion fly (Delia antiqua) damage were investigated. These included different environments, methods of plant raising and egg inoculation, and plant size. A glasshouse compartment maintained at 14 to 37 °C was as satisfactory for assessing the resistance as a controlled environment. The basic test unit comprised 10 onion seedlings raised in a 400 × 50 × 50 mm deep plastic trough and inoculated with 50 onion fly eggs from a laboratory culture. Resistance was assessed by counting wilted plants every two or three days for 21 days after egg inoculation. The reciprocal of number of days for each plant to wilt (wilting rate) was an appropriate variate for statistical analysis, adjusted for the length of the longest leaf at the time of inoculation. The method enables cultivars or breeding lines of onion species to be reliably screened for resistance to larvae of this pest in a glasshouse so that only the most promising need be tested in the field.  相似文献

The Pseudomonas fluorescens isolate Pfl was found to inhibit the growth of pathogen Alternaria palandui, in vitro. In the present study, foliar application of a talc-based formulation of Pfl significantly reduced the incidence of leaf blight of onion, caused by A. palandui. Induction of defense-related proteins viz., chitinase, beta-1,3 glucanase, peroxidase (PO) and polyphenol oxidase (PPO) by application of Pfl, was studied against A. palandui infection in resistant (IHR 56) and susceptible (MDUI) onion cultivars. Chitinase in both cultivars, with or without challenge-inoculation of A. palandui revealed changes in the isoform pattern. The Native-PAGE of PO showed induction of PO2 isoform in both the cultivars, in response to inoculation of pathogen. Isoform analysis of PPO also exhibited induction in the Pfl-treated plants challenged with pathogen. Similarly, the activity of beta-1,3-glucanase was greatly induced in Pfl-treated plants, challenged with pathogen as compared to controls. Thus, the P. fluorescens-treated plants showed significant increase in the levels of the defense enzymes, in comparison to the plants challenged with the pathogen.  相似文献

Dicotyledonous plants were reliably inoculated with stem nematodes by placing drops of 1.3% carboxymethylcellulose containing the surface sterilised worms between the cotyledons or in the leaf axils of recently emerged seedlings raised in pots of cool, moist soil. Inoculating onion leaves, bean stems and potato tubers or potato leaves gave variable results and inoculating onion, tulip and narcissus bulbs and lucerne and red clover stems was usually unsuccessful. An attempt to characterise 67 stem nematode populations by the reactions of a number of different plants failed for lack of useful differential hosts. Lucerne was, however, resistant to all but lucerne populations. Multiplication of stem nematode populations varied greatly between cultivars of lucerne or red clover. Some cultivars were resistant to some populations of lucerne or red clover stem nematodes and susceptible to others. These differences could not be ascribed to differences in viability of the nematodes or to differences in success of the inoculations. They indicate the presence of different pathotypes or biotypes in different populations of a so-called ‘host race’ and indicate the need for new resistant cultivars to be tested against a range of populations before they are released for general use. Amongst lucerne cultivars tested, Vertus was resistant to some lucerne stem nematode populations and susceptible to others. The supposedly resistant lucerne cultivars Euver and Lifeuil were as susceptible as was Europe. Amongst red clover cultivars tested, Redhead, Kühn, Changins and Mt Calme were susceptible, Britta, Lucrum and Temara were the least resistant, Renova, Rittinova and Quin were intermediate and susceptible to one or more of the populations tested but Norseman and especially Sabtoron were very resistant.  相似文献

In modern agriculture there is a great demand for a rapid and objective screening method for stress resistance, because so far, the resistance of new cultivars is tested in time- and money consuming field experiments. Based on fluorescence ratios, and lifetime of fluorophores measured by fluorescence spectroscopy, we have postulated that an early discrimination of susceptible and resistant wheat cultivars to the leaf rust pathogen Puccinia triticina can be accomplished. As representative for leaf rust resistant and leaf rust susceptible wheat genotypes the cultivars Esket and Skalmeje, respectively, were chosen. Plants were grown under controlled environment conditions and inoculated with the leaf rust pathogen at the second-leaf-stage by single-droplet application. Fluorescence measurements were carried out from two to four days after inoculation (dai) by using a compact fibre-optic fluorescence spectrometer with nanosecond time-resolution. Experimental results indicated that UV laser-induced spectral characteristics as well as determination of fluorescence lifetime are suited to detect leaf rust two dai. For this purpose several ratios and wavelength can be considered. In general, the tested cultivars showed distinct responses to the pathogen development. In this context the ratio F451/F687 measured three dai and mean lifetimes at 500 nm and 530 nm are suited to differentiate the resistant Esket from the susceptible Skalmeje genotypes.  相似文献

Populations of onion thrips, Thrips tabaci Lindeman, from commercial onion fields in New York were evaluated for their susceptibility to the commonly used pyrethroid, lambda-cyhalothrin (Warrior T), using a novel system called the Thrips Insecticide Bioassay System (TIBS). To use TIBS, thrips are collected directly from the plant into an insecticide-treated 0.5-ml microcentrifuge tube that has a flexible plastic cap with a small well into which 0.08 ml of a 10% sugar-water solution with food colorant is deposited. The solution is sealed into the well with a small piece of stretched parafilm through which the thrips can feed on the solution. Thrips mortality is assessed after 24 h with the help of a dissecting stereoscope. In 2001, onion thrips populations were collected from 16 different sites and resistance ratios were >1,000 in five populations. Percent mortality at 100 ppm, a recommended field rate, varied from 9 to 100%, indicating high levels of variation in susceptibility. Particular instances of resistance appeared to be the result of practices within an individual field rather than a regional phenomenon. In 2002, we also observed large differences in onion thrips susceptibility, not only between individual fields but also between thrips collected in a single field at mid season and late season, again suggesting that insecticide-use practices within an individual field caused differences in susceptibility. Additional tests indicated no differences in susceptibility between adult and larval onion thrips populations and only relatively minor differences between populations collected from different parts of the same field. Using TIBS, several populations of onion thrips with different susceptibilities to lambda-cyhalothrin were identified and then subjected to lambda-cyhalothrin-treated onion plants. There was a highly significant positive relationship between percent mortality of thrips from TIBS and percent mortality from the treated onion plants, indicating that results from TIBS could be used to predict spray performance. These data suggest that use of TIBS for evaluating susceptibility to particular insecticides could be instrumental for developing a resistance management strategy for onion thrips.  相似文献

Plant disease resistance loci have been used successfully in breeding programs to transfer traits from resistant germplasm to susceptible plant cultivars. The molecular cloning of plant disease resistance genes now permits the transfer of such traits across species boundaries by genetic transformation of recipient hosts. The tomato disease resistance gene Pto confers resistance to strains of the bacterial pathogen Pseudomonas syringae pv tomato expressing the avirulence gene avrPto. Transformation of Nicotiana benthamiana with Pto results in specific resistance to P. s. pv tabaci strains carrying avrPto. The resistant phenotype is manifested by a strong inhibition of bacterial growth and the ability to exhibit a hypersensitive response. Resistance cosegregates with the Pto gene in transgene selfings and testcrosses. Our results demonstrate the conservation of disease resistance functions across genus boundaries and suggest that the utility of host-specific resistance genes can be extended by intergeneric transfer.  相似文献

To develop an insecticide resistance management program for onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), on onions (Allium spp.), we surveyed populations in commercial onion fields in New York and evaluated their susceptibility to the two most widely used classes of insecticides plus two new insecticides during 2003-2005. All insecticide evaluations were conducted using the Thrips Insecticide Bioassay System (TIBS). As in our surveys conducted during 2002-2003, there were large temporal and spatial variations in susceptibility to the pyrethroid lambda-cyhalothrin (Warrior) across onion-growing regions in 2003. New data indicate that the field rate of methomyl (Lannate LV) still provides control but that the genes for resistance to methomyl are present in some populations. Tests with the two new insecticides, acetamiprid (Assail 70 WP) and spinosad (SpinTor 2CS), indicated they provided > 85% mortality at the field rate. To determine the spatial variation in insecticide susceptibility within a region, a series of systematic assays were conducted with lambda-cyhalothrin and methomyl. In 2004 and 2005, our data indicated that the within-region spatial variation in susceptibility to lambda-cyhalothrin was not large at the field rate or for the 100 ppm rate of methomyl. In 2005, a year in which T. tabaci densities in most fields were much higher than in 2004, growers were unable to control T. tabaci in particular fields and attributed this lack of control to resistance. Yet, we found similar levels of high susceptibility in all fields when using TIBS. This finding suggests that resistance had not developed and that variation in control may have been due to other factors, such as localized higher populations, poor spray coverage, too much time between spray applications, or different onion varieties.  相似文献

Onion thrips is a major threat to onion crop throughout the world. It is a potential vector of Iris yellow spot virus and causes significant economic damage to bulb production. Phenotypic and biochemical traits of onion cultivars were assessed against Thrips tabaci. Onion Gawran LR-241 (OG) cultivar was tolerant against the infestation of T. tabaci whereas Onion White (OW) was susceptible. Number and size of stomata, cuticle thickness, cell wall thickness and surface wax of onion leaves were studied with the help of scanning electron microscope and quantitative and qualitative analysis was carried out to estimate epicuticular wax and other bio-chemical components through GC/MS. Onion Gawran has thick cell wall, sharp and dense wax crystals, wider central angle and small sized stomata compared to other cultivars. Epicuticular wax components of OG cultivar were heptacosane (5.2%), octacosanol-1 (9.2%), 2-methyl octacosane (4.2%), heptadecanol-1 (5.2%), hexacosanol-1 (4.2%), azulene, 1,4-dimethyl-7-(1-methylethyl) (36.9%), hexadecanoic acid (1.95%), heptadecane (4.2%), triacontanol-1 (5.8%) and hentriacontanone-16 (23.40%). Azulene, 1, 4-dimethy-l-7-(1-methy-l-ethyl) was only found 36.9% in OG but absent in other three cultivars. 2-methyl octacosane was absent in Poona Red Desi and OW cultivars. Hentriacontanone-16, 2-methyl octacosane, fatty alcohols (Octacosanol-1 and Triacontanol-1) and azulene, 1, 4-dimethy-l-7-(1-methy-l-ethyl) were effective in the formation of epicuticular wax in onion cultivars. It implies that phenotypic and biochemical characteristics of OG cultivar proved as resisting features to T. tabaci.  相似文献

Nymphs and alates of aphid Nasonovia ribisnigri (Mosley) (Homoptera: Aphididae) were tested on 10 lettuce cultivars with N. ribisnigri resistance gene Nr and 18 cultivars without the resistance gene in various bioassays. Bioassays used whole plants, leaf discs, or leaf cages to determine susceptibility of commercial lettuce cultivars to N. ribisnigri infestation and to evaluate screening methods for breeding lettuce resistance to N. ribisnigri. Resistant and susceptible plants were separated in 3 d when using whole plant bioassays. Long-term (> or =7 d) no-choice tests using leaf cages or whole plants resulted in no survival of N. ribisnigri on resistant plants, indicating great promise of the Nr gene for management of N. ribisnigri. Effective screening was achieved in both no-choice tests where resistant or susceptible intact plants were tested separately in groups or individually and in choice tests where susceptible and resistant plants were intermixed. Leaf discs bioassays were not suitable for resistance screening. All lettuce cultivars without the resistance gene were suitable hosts for N. ribisnigri, indicating the great importance of this pest to lettuce production and the urgency in developing resistant lettuce cultivars to manage N. ribisnigri.  相似文献

The silverleaf whitefly Bemisia tabaci (Genn.) biotype B has become a serious problem for soybean cultivation because it can significantly reduce soybean productivity. The use of soybean cultivars resistant to whitefly attack is an important strategy in an integrated pest management (IPM) program. This study evaluated the preference for oviposition and colonization by B. tabaci biotype B on different soybean genotypes. In the free-choice test, the genotypes studied were 'IAC 17' and 'IAC 19' as the standards for resistance and 'IAC Holambra Stwart' as the standard for susceptibility, as well as BABR01-0492, BABR01-0173, BABR01-1259, BABR01-1576, BABR99-4021HC, BABR99-4021HP, 'Barreiras', 'Conquista', 'Corisco', 'BRS Gralha', PI274454, PI227687, and PI171451. In the no-choice test, the four best genotypes selected in the free-choice test, in addition to the susceptible and resistant standards were evaluated. Our data indicated 'Barreiras' as the most resistant genotype against B. tabaci biotype B. 'BRS Gralha', which was the least attractive to whitefly adults in the free-choice test, did not show resistance to insect attack when they were confined in cages in the no-choice test. Despite the high number of eggs observed, BABR01-1576 and BABR99-4021HC showed a reduced number of nymphs, indicating antibiosis. The genotypes with a high level of resistance can be used as a tool against B. tabaci in IPM or as a source of resistance in plant-breeding programs.  相似文献

Jasmonate is an important endogenous chemical signal that plays a role in modulation of plant defense responses. To understand its mechanisms in regulation of rice resistance against the fungal pathogen Magnaporthe oryzae, comparative phenotype and proteomic analyses were undertaken using two near‐isogenic cultivars with different levels of disease resistance. Methyl‐jasmonate (MeJA) treatment significantly enhanced the resistance against M. oryzae in both cultivars but the treated resistant cultivar maintained a higher level of resistance than the same treated susceptible cultivars. Proteomic analysis revealed 26 and 16 MeJA‐modulated proteins in resistant and susceptible cultivars, respectively, and both cultivars shared a common set of 13 proteins. Cumulatively, a total of 29 unique MeJA‐influenced proteins were identified with many of them known to be associated with plant defense response and ROS accumulation. Consistent with the findings of proteomic analysis, MeJA treatment increased ROS accumulation in both cultivars with the resistant cultivar showing higher levels of ROS production and cell membrane damage than the susceptible cultivar. Taken together, our data add a new insight into the mechanisms of overall MeJA‐induced rice defense response and provide a molecular basis of using MeJA to enhance fungal disease resistance in resistant and susceptible rice cultivars.  相似文献

Identifying locations where onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), overwinter and subsequently disperse is important for designing control strategies. In upstate New York from 2003 through 2006, potential overwintering sites in the commercial onion, Allium cepa L., cropping system were investigated early in the spring before onion seedling emergence and again late in the season after onions were harvested. Onion thrips adults were sampled directly from the soil and indirectly from the soil by using emergence cages. Sampling locations included onion field interiors and edges and areas outside of these fields, including woods. Host material sampled included onion culls; volunteer onions, which sprout from cull onions left behind after harvest; and weeds. Onion thrips adults were found in all sections of onion fields and in locations outside of onion fields, with the fewest emerging from woods. Emergence began in early May and extended into June. Peak emergence occurred during the last half of May, at which time 50-75% of the population had emerged. Adults colonized volunteer onions as early as late March and as late as mid-November. No adults were found overwintering in onion cull piles. Adults also colonized several weed species, especially pigweed, Amaranthus hybridis L., and lambsquarters, Chenopodium album L., late in the fall. Our results indicate that onion thrips adults overwinter in the soil within and near onion fields and that they probably colonize volunteer onion plants before subsequent generations infest the onion crop in the spring. Volunteer onions and weeds also provide onion thrips with a host after onions are harvested. Consequently, onion thrips management strategies should include tactics that reduce volunteer onion and weed abundance.  相似文献

Components of early blight resistance were quantified in leaves of different ages in four potato cultivars. The components of resistance: incubation period (IP), lesion number (LN), early blight severity, lesion expansion rate (LER), latent period (LP) and spore production by lesion area (SPLA), were evaluated separately in the lower, middle and upper leaves of four potato cultivars. Plants of cultivar Aracy (resistant), Delta (moderately resistant), Desirée (susceptible) and Bintje (susceptible) were inoculated with an Alternaria solani isolate at the beginning of the flowering stage. Disease severity varied in different plant parts. In all cultivars, regardless of resistance, the smallest values of LN, and severity were recorded on the upper leaves, suggesting that young tissues are less susceptible. In cultivar Aracy, the IP was long, with small values of LN and LER and consequently, low values of early blight severity in all leaf positions were recorded. Although IP was long in cultivar Aracy, no differences between the moderately resistant cultivar Delta and the susceptible cultivars Bintje and Desirée could be detected for this component. The IP was only influenced by leaf position in cultivar Aracy. Clear differences in resistance levels among cultivars could be detected regarding LN, severity and LER. However, neither LP nor SPLA were associated with resistance level of cultivars or with leaf position. Analyses according to plant part suggest that evaluations on leaves of the middle third part are most suitable for screening for early blight resistance in potato.

We compared smooth-leaf okra- and normal-leaf upland cotton (Gossypium hirsutum L.) strains and cultivars for susceptibility to colonization by Bemisia tabaci (Gennadius) biotype B. Experiments were conducted at seven field sites, five at Holtville, CA, and two at Maricopa, AZ, during 1996-2000. Okra-leaf strains and cultivars, as a group, had lower numbers of adults, eggs, and nymphs compared with normal-leaf strains and cultivars indicating the potential of okra-leaf genetic traits for reducing colonization by B. tabaci. Results also suggest that okra-leaf shape may provide less favorable micro-environmental conditions for the habitat of B. tabaci because of more open canopy as evidenced by higher leaf perimeter to leaf area ratio. The okra-leaf cultivar 'Siokra L-23' appears to have genetic traits that should be examined further as a source of B. tabaci resistance.  相似文献

Wheat (Triticum aestivum L.) is the largest cereal crop grown in Western Canada where drought during late vegetative and seed filling stages affects plant development and yield. To identify new physiochemical markers associated with drought tolerance, epidermal characteristics of the flag leaf of two wheat cultivars with contrasting drought tolerance were investigated. The drought resistant ‘Stettler’ had a lower drought susceptibility index, greater harvest index and water‐use efficiency than the susceptible ‘Superb’. Furthermore, flag leaf width, relative water content and leaf roll were significantly greater in Stettler than in Superb at moderate drought stress (MdS). Visible differences in epicuticular wax density on the adaxial flag leaf surfaces and larger bulliform cells were identified in Stettler as opposed to Superb. Mid‐infrared attenuated total internal reflectance spectra revealed that Stettler flag leaves had increased asymmetric and symmetric CH2 but reduced carbonyl esters on its adaxial leaf surface compared to Superb under MdS. X‐ray fluorescence spectra revealed a significant increase in total flag leaf Zn concentrations in Stettler in response to MdS. Such information on the microstructural and chemical features of flag leaf may have potential as markers for drought tolerance and thereby accelerate the selection and release of more drought‐resistant cultivars.  相似文献
