The musculus masseter has been studied in 12 bioptates of 19-24-year-old men after they have been operated on for correction of true progeny which results in a decreasing functional loading on the masticatory musculature. SDG activity, ratio of myons of various types, cross section area of muscle fibres, content ratio of the connective tissue layers, number of blood capillaries around the myons, relative volume of the submicroscopic structures of the muscle fiber have been estimated. Pronounced pathological disorders in the muscle are absent, it is connected with a gradual change in conditions of functioning and a prolonged time for adaptation. The first type myons ratio increases, comparing to that in the control, the number of the capillaries around the myons decreases, while the amount of the connective tissue grows large. Marked quantitative ultrastructural changes of the energy and contractile-myofibrillar apparatus take place.  相似文献

As demonstrates estimation of myosin ATPase and SDG activity, the guinea pig is already born with differentiated muscle fibers (MF), and the first histochemical differences between them take place in the uterine 10 days before birth. Tonic oxidative fibers of the first type, arranging hexagonally, develop especially quickly at early stages of postnatal ontogenesis. Their relative contents up to the end of the observations (185 days) do not change, and area of their transversal section increases but slightly in comparison to the phasic fibers. The main age changes of the muscle tissue are connected with formation and rearrangement of the phasic fibers. The most intensive reconstructions of the phasic fibers coincide with the period of game activity and sex maturation. In mixed muscles the part of the glycolytic fibers increase during the postnatal ontogenesis. In the process of ontogenesis the soleus muscle fully consists of oxidative fibers. The definitive level of the MF development is established after the guinea pigs have reached their sex maturation. Comparing the results of the given investigation with the previous data on development of MF in rats, it is possible to conclude that term and premature animals have various rates in development of the muscle system, however, main stages of myogenesis coincide, though they are connected with various phases of ontogenesis.  相似文献

In 105 male rats of Wistar strain distal parts of one of the thoracic extremities are amputated with keeping intact the places where the brachial muscle is fixed. This does not restrict the volume of the brachium movements but essentially decreases their dynamic component (power loading). For 45 days dynamics of succinate-dehydrogenase (SDG) activity is being revealed in muscle fibers (MF) of functionally different muscles: m. brachialis, m. serratus ventralis and m. triceps brachii (the medial head). Average tendency of the process, changes in the distribution margins, asymmetry and kurtosis are taken into account. Under hypodynamia reconstruction of the MF has a wavy character with a gradually longer period of fluctuations. In all the muscles appear MF with a greater than in the control SDG activity. In the medial head of the m. triceps brachii the fibers with the lowest SDG activity disappear. The amount of MF with the lowest activity decreases, while those with the higher--increases, the process being more pronounced in the m. triceps brachii. The amount of MF with middle activity of the enzyme remains nearly unchanged. MF with different initial enzymatic activity do not change simultaneously. The degree of the changes in the fibers and the power leading are connected with each other, the fibers with the low initial SDG activity including into the reaction at a sharper decrease of the latter. The changes in quantitative ratio of MF with different SDG activity are not the same in every muscle studied.  相似文献

In 140 male rats of Wistar strain the distal part of one thoracic extremity has been amputated in such a way that places of the brachial muscle fixation remained intact. This resulted in decrease of the force component of muscle contractions (more intensive in the musculus triceps brachii) without essential swing and speed of movements. The operation was performed during one day at three series: at 7 a. m. 3 p. m. and 11 p. m. During following 45 days the muscles developing various contraction force were studied: the medial head of the musculus triceps brachii and brachial muscle. Succinate dehydrogenase (SDG) activity in muscle fibers (MF) was estimated photometrically. In MF initially characterized with various SDG activity, attention was payed to its dynamics and also relation of the fibers number, possessing various level of the enzymatic activity was taken into consideration. SDG activity in MF and relation of various MF change wavy-like, fluctuating from the control meaning and returning to them again. MF with different SDG activity do not similarly react to a sharp decrease of the force loading. Fibers with high enzymatic activity react to changes of the force loading immediately, intensity of the reaction at this stage slightly depends on degree of biomechanical changes; at later stages the response is differentiated: if the loading is decreased considerably (musculus triceps brachii), the process continues developing and increasing uninterruptedly, if it decreases moderately (brachial muscle)--it stabilizes and even has tendency to adaptation and returning towards the control state.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The purpose of the present study was to verify the smooth muscle cell distribution and elastic system fibers volumetric density (Vv) in the corpus spongiosum and corpus cavernosum of the wild boar penis. Adult wild boars (n=13) were used. The penile mid shaft fragments were fixed with 4% phosphate buffered formalin solution and/or Bouin's liquid during 24-48 h, and processed using standard histological techniques. The sections were stained with Weigert's Resorcin-Fucsin with previous oxidation. The elastic system fibers Vv was determined in 25 random fields of each fragment using M42 test system. For immunohistochemical analysis, monoclonal anti-alpha actin smooth muscle was used. The histochemical methods detected elastic system fibers in both corpus spongiosum and corpus cavernosum of all animals. The elastic fibers Vv average was 36.6%+/-0.9 for corpus spongiosum and 11.7%+/-0.5 for corpus cavernosum. Through immunocitochemistry, a small quantity of smooth muscle cells was visualized in intimate relationship with blood vessels wall. The great amount of elastic fibers and the smooth muscle cell distribution beneath the endothelium suggest that these fibers may have an important role in penile erection process in the penis of wild boars.  相似文献

Six fiber types have been described in the ambiens muscle of red-eared turtles. These include one slow oxidative type, two fast oxidative types, two fast oxidative and glycolytic types, and one fast glycolytic type. Fiber types are non-randomly distributed throughout cross sections of the muscle. There is a decreasing gradient of oxidative staining and an increasing gradient of glycolytic staining along an axis from the superficial to deep regions of the muscle. The slow oxidative fibers are predominantly located within one or two fascicles of the superficial surface of the muscle. The fast glycolytic fibers are predominant in deep fascicles. In contrast to previous reports of histochemically monotypic intrafusal fibers in turtle muscle, ambiens muscle spindles have been observed containing one to eleven intrafusal fibers, including two fiber types. Fiber diameter and area are consistently smaller than observed in most extrafusal fibers. Spindles are predominantly located in superficial and cranial fascicles of the ambiens muscle and are located in regions characterized by extrafusal fibers with high oxidative activity.  相似文献

Reconstruction from thick serial transverse slices of frog skeletal muscle fibers stained with peroxidase and examined by high-voltage electron microscopy has revealed that the T system networks at successive sarcomeres are connected together in a helicoidal fashion. From zero to eight helicoids have been found in each of a group of 21 fibers reconstructed in cross section. Helicoids can have either right- or left-handed screw senses, and both senses can be found in one fiber cross section. Because the T system maintains a relatively precise alignment with the myofibrillar striations, it follows that the striations must also have a helicoidal arrangement. This has been found before, but has not been widely accepted in recent times. The presence of helicoids in the bands and membrane networks is not thought per se to alter very much our thinking about excitation and contraction mechanisms in skeletal muscle fibers.  相似文献

TheM. biceps brachii ofMacaca fascicularis was examined, noting the total number of muscle fibers, the number of muscle fibers per square millimeter, the cross sectional area of Venter musculi and the thickness of individual fibers in the cross sectional area of the right brachial biceps specimens of 10 adult crab-eating macaques, five males and five females. The results of this investigation are compared with a previous study of a similar type made on the brachial biceps of the adult human. Comparative analysis shows that the mean ventral cross sectional area of the macaque biceps muscle is 1/7 to 1/3 that of the cross sectional area in the human muscle. Macaque brachial biceps muscle shows approximately one half the total number of muscle fibers of the human specimens, though the number of fibers per square millimeter is one to two times greater than in human specimens. The macaque muscle fibers were 1/2 to 5/6 as thick as those in the human specimens. The relations between the ventral cross sectional area and thickness of individual muscle fibers is discussed. Comparisons are made between the macaque and human specimens. It is suggested that such factors as age, sex, and the nutritional history of the specimen donors may have influenced the myofibrous organization in both human and macaque specimens. It is suggested also, that differences in myofibrous organization may be related to more continuous or sustained muscular activity in the macaque and more forceful muscular contraction in humans.  相似文献

The influence of spaceflight on the distribution of succinate dehydrogenase (SDH) activity throughout the cross section of fibers in the soleus was studied in five male rats and in five rats maintained under ground-based simulated flight conditions (control). The flight (COSMOS 1887) was 12.5 days in duration, and the animals were killed approximately 2 days after return to 1 G. Fibers were classified as slow-twitch oxidative or fast-twitch oxidative-glycolytic in histochemically prepared tissue sections. The distribution of SDH activity throughout the cross section of 20-30 fibers (each type) was determined using quantitative histochemical and computer-assisted image analysis techniques. In all the fibers, the distribution of SDH activity was significantly higher in the subsarcolemmal than in intermyofibrillar region. After spaceflight the entire regional distribution of SDH activity was significantly altered in the slow-twitch oxidative fibers. The fast-twitch oxidative-glycolytic fibers of the spaceflight muscles exhibited a significantly lower SDH activity only in their subsarcolemmal region. These data suggest that when determining the influence of spaceflight on muscle fiber oxidative metabolism enzymes, it is important to consider the location of the enzyme throughout the cross section of a fiber. Furthermore the functional properties of the soleus that depend on the metabolic support of mitochondria in the subsarcolemmal region may be primarily affected by exposure to microgravity.  相似文献

Enzymatic heterogeneity of the capillary bed of rat skeletal muscles   总被引:4,自引:0,他引:4  
This study of the capillaries in rat skeletal muscle involved the use of a histochemical method that allows one to distinguish between arterial and venous portions of capillaries. Under controlled staining conditions, the arterial portion of the capillary bed reacts positively for alkaline phosphatase (AP) activity, and the venous portion is positive for dipeptidyl peptidase IV (DPP IV) activity. A short transitional capillary segment is positive for the activity of both enzymes. Capillaries of the normal soleus muscle and the red and white portions of the sternomastoid muscle have been quantitatively analyzed. Quantitative data demonstrated differences in capillary dimensions among the muscles studied. Capillaries of the white part of the sternomastoid were the longest, and they had the shortest DPP IV-positive segment (8% of the total capillary length). Capillaries of the soleus muscle were the shortest, and they also had short DPP IV-positive segments (16%). In contrast, the DPP IV-positive segments of the red part of the sternomastoid occupied 60% of the total capillary length. Survey cross sections reveal a mosaic distribution of patches of capillaries stained for AP and DPP IV activity. This study reveals that within given bundles of muscle fibers, the capillaries that run parallel to the muscle fibers are aligned relative to one another in such a manner that their arterial and venous segments are in register.  相似文献

By means of morphometrical and histochemical methods for revealing myosin ATPase and SDG activity development of various types of muscle fibers (MF) has been studied in the postmortem material, using m. biceps and m. triceps brachii in human ontogenesis. The flexors and extensors have features in common in the dynamics of the MF maturation, and some distinctive peculiarities. The appearance of histochemical distinctions between the MF takes place on the 5th-6th months of the intrauterine development. Morphofunctional specialization begins with formation of tonic fibers. During the 1st-2nd years phasic fibers form. A relative amount of fast MF in both muscles increases at the age of 11-12 years. The dynamics of final specialization of the MF is connected with stages of sexual maturation. The first stage of the sexual maturation (about 14 years of age) is connected with decrease in the relative amount of the MF of glycolytic type of energy supply and corresponding increase in the number of oxidative type structures. From 15-17 years of age a final differentiation begins, it is connected with an intensive transversal growth of all the MF and distinguish of thick glycolytic MF. The m. biceps brachii has a relatively greater amount of oxidative fibers, and the m. triceps brachii, glycolytic ones. The transversal section area of the MF in the m. triceps brachii exceeds that of the m. biceps brachii, beginning from the 7th month of the intrauterine development up to 14 years of age. The investigation performed does not reveal any anticipating development either in the flexors or in the extensors. The differentiating processes in the m. biceps and m. triceps brachii occur nearly simultaneously.  相似文献

The muscle fibers of the feline biceps femoris have tapered ends, across which tension is transmitted to the endomysium. The angle of taper of 11 ends, measured on scanning electron micrographs, varied between 0.16 degrees and 1.18 degrees. The muscle fibers are highly variable in cross-sectional shape. The shape of the fibers has been quantified as the ratio (form factor [FF]) of the measured perimeter to the calculated circumference of a circle having an area equal to that contained by the fiber perimeter. The FF for 173 terminal portions of fibers varied between 1.06 and 1.85 and was found to have a highly significant negative correlation with sarcomere length. The slope of the regression line suggests that the fibers maintain both volume and surface area as they change length. These studies suggest that isovolumic muscle fibers maintain a constant surface area by changing shape as they change length.  相似文献

Muscle fiber deformation is related to its cellular structure, as well as its architectural arrangement within the musculoskeletal system. While playing an important role in aponeurosis displacement, and efficiency of force transmission to the tendon, such deformation also provides important clues about the underlying mechanical structure of the muscle. We hypothesized that muscle fiber cross section would deform asymmetrically to satisfy the observed constant volume of muscle during a contraction. Velocity-encoded, phase-contrast, and morphological magnetic resonance imaging techniques were used to measure changes in fascicle length, pinnation angle, and aponeurosis separation of the human gastrocnemius muscle during passive and active eccentric ankle joint movements. These parameters were then used to subsequently calculate the in-plane muscle area subtended by the two aponeuroses and fascicles and to calculate the in-plane (dividing area by fascicle length), and through-plane (dividing muscle volume by area) thicknesses. Constant-volume considerations of the whole-muscle geometry require that, as fascicle length increases, the muscle fiber cross-sectional area must decrease in proportion to the length change. Our empirical findings confirm the definition of a constant-volume rule that dictates that changes in the dimension perpendicular to the plane, i.e., through-plane thickness, (-6.0% for passive, -3.3% for eccentric) equate to the reciprocal of the changes in area (6.8% for passive, 3.7% for eccentric) for both exercise paradigms. The asymmetry in fascicle cross-section deformation for both passive and active muscle fibers is established in this study with a ~22% in-plane and ~6% through-plane fascicle thickness change. These fiber deformations have functional relevance, not only because they affect the force production of the muscle itself, but also because they affect the characteristics of adjacent muscles by deflecting their line of pull.  相似文献

鱼类快肌和慢肌分别占据骨骼肌的不同位置,表现不同的生长发育特征。为了解鳜(Sinipercachuatsi)慢肌纤维的胚后发育特征,本研究通过制作孵化后1~33日龄鳜个体的石蜡切片,采用慢肌特异抗体的免疫组织化学染色,观察了背鳍起点处躯干横切面慢肌的发育变化特征,并利用图像分析软件统计慢肌纤维的数目和面积。结果表明,孵化后鳜仔鱼慢肌位于水平肌隔附近,呈楔形,向背、腹两侧生长。孵化后1~9日龄为单层肌纤维,11日龄发育为多层肌纤维,19日龄覆盖侧线附近,33日龄延伸至背侧第2背肌节、腹侧腹部肌肉2/3处,并在水平肌隔和侧线处分别形成两个肌群。位于骨骼肌最外层的扁平状表层细胞,可能为慢肌增生生长的主要来源。躯干单侧慢肌肌纤维数目由孵化后6个增加至315个,总面积从13.18μm2增加到7 839.58μm2,孵化后13日龄的增生生长占优势,其他发育阶段,肥大生长一直占主导优势。  相似文献

The adaptation of a slow (soleus, Sol) and a fast (medial gastrocnemius, MG) skeletal muscle to spaceflight was studied in five young male rats. The flight period was 12.5 days and the rats were killed approximately 48 h after returning to 1 g. Five other rats that were housed in cages similar to those used by the flight rats were maintained at 1 g for the same period of time to serve as ground-based controls. Fibers were classified as dark or light staining for myosin adenosine triphosphatase (ATPase). On the average, the fibers in the Sol of the flight rats atrophied twice as much as those in the MG. Further, the fibers located in the deep (close to the bone and having the highest percentage of light ATPase and high oxidative fibers in the muscle cross section) region of the MG atrophied more than the fibers located in the superficial (away from the bone and having the lowest percentage of light ATPase and high oxidative fibers in the muscle cross-section) region of the muscle. Based on quantitative histochemical assays of single muscle fibers, succinate dehydrogenase (SDH) activity per unit volume was unchanged in fibers of the Sol and MG. However, in the Sol, but not the MG, the total amount of SDH activity in a 10-microns-thick section of a fiber decreased significantly in response to spaceflight. Based on population distributions, it appears that the alpha-glycerophosphate dehydrogenase (GPD) activities were elevated in the dark ATPase fibers in the Sol, whereas the light fibers in the Sol and both fiber types in the MG did not appear to change. The ratio of GPD to SDH activities increased in the dark (but not light) fibers of the Sol and was unaffected in the MG. Immunohistochemical analyses indicate that approximately 40% of the fibers in the Sol of flight rats expressed a fast myosin heavy chain compared with 22% in control rats. Further, 31% of the fibers in the Sol of flight rats expressed both fast and slow myosin heavy chains compared with 8% in control rats. Immunohistochemical changes in the MG were minimal. These data suggest that the magnitude and direction of enzymatic activity and cell volume changes are dependent on the muscle, the region of the muscle, and the type of myosin expressed in the fibers. Further, the ability of fibers to maintain normal or even elevated activities per unit volume of some metabolic enzymes is remarkable considering the marked and rapid decrease in fiber volume.  相似文献

An electrical network is presented as a model of the electrical connections between cells in a cross section of visceral smooth muscle. Methods of operational calculus are used to analyze the network and to determine the effect of parallel paths on measured resistances between the nodes. The analysis of the model suggests that actual resistances between two adjacent cells if they are isolated from the other cells in the cross section could be three times the values measured when the cells are interconnected with the parallel fibers. It is felt that the analysis presented makes possible a more accurate interpretation of electrical measurements of intercellular resistance by providing an estimate of the shunting effect of the current paths through parallel cells.  相似文献

Tetrapeptide tuftsin action on the activity of succinate dehydrogenase (SDG), malate dehydrogenase (MDG) and monoamine-oxidase (MAO) in microstructures of the neocortex, hippocampus and hypothalamus (supraoptic and paraventricular nuclei, retrochiasmic zone) has been investigated by means of histochemical methods. Simultaneously, pyruvate-, malate-, glutamate-, alfaketoglutamate-, succinate- and lactate-dehydrogenase activity in the neocortex and in the structures of the limbic system has been studied biochemically. SDG and MDG activity increases in neurons and glycocytes of all the hypothalamic formations mentioned. Changes in the activity of dehydrogenases in the hippocampus and neocortex under the same stimulation are less pronounced. MAO activity also increases in the nerve terminals converging on the bodies and dendrites of hypothalamic neurons and in the preterminal fibers of the neocortex, Tuftsin increases oxidative-reducing processes in various structures of the brain, but at the same time it possesses a predominant influence on the limbic system structures.  相似文献

Oxidative capacity and capillary density of diaphragm motor units   总被引:2,自引:0,他引:2  
Motor units in the cat diaphragm (DIA) were isolated in situ by microdissection and stimulation of C5 ventral root filaments. Motor units were classified based on their isometric contractile force responses and fatigue indexes (FI). The muscle fibers belonging to individual units (i.e., the muscle unit) were identified using the glycogen-depletion method. Fibers were classified as type I or II based on histochemical staining for myofibrillar adenosine triphosphatase (ATPase) after alkaline preincubation. The rate of succinate dehydrogenase (SDH) activity of each fiber was determined using a microphotometric procedure. The location of capillaries was determined from muscle cross sections stained for ATPase after acid (pH = 4.2) preincubation. The capillarity of muscle unit fibers was determined by counting the number of capillaries surrounding fibers and by calculating the number of capillaries per fiber area. A significant correlation was found between the fatigue resistance of DIA units and the mean SDH activity of muscle unit fibers. A significant correlation was also observed between DIA unit fatigue resistance and both indexes of muscle unit fiber capillarity. The mean SDH activity and mean capillary density of muscle unit fibers were also correlated. We conclude that DIA motor unit fatigue resistance depends, at least in part, on the oxidative capacity and capillary density of muscle unit fibers.  相似文献

Summary Tissue capillarity and diffusion distances were determined for red and white skeletal muscles of adult birds ranging in mass from 10.8 to 6200 g. In addition, literature values for capillarity and diffusion distances in skeletal muscles of mammals were incorporated into the data set. Muscle mass was closely coupled to body mass. However, no significant allometric relations were found for any of the other variables measured. Number of capillaries per fiber was not correlated with cross sectional area of individual muscle fibers. Thus, capillary density decreased in a hyperbolic manner against fiber area and diffusion distance decreased in a hyperbolic manner against the number of capillaries per muscle fiber. Red muscles had significantly higher numbers of capillaries per fiber and significantly shorter diffusion distances than did white muscles. The patterns for tissue capillarity and diffusion distances in avian muscle reported here are similar to values reported previously for mammalian muscles. In both taxanomic groups capillarity and diffusion distances are independent of body mass.
