The xylem exudation of detopped 7-d-old seedlings of Zea maysL. doubled when KCI was present in the root medium comparedto seedlings maintained on water. It was further enhanced whenKCI was replaced by nitrogen compounds such as nitrate, ammoniumand glutamine. The role of the nitrate assimilation pathwayon the enhancement of xylem exudation rate was investigatedusing tungstate, an inhibitor of nitrate reductase (NR) activity,and phosphinothricin or methionine sulphoximine, inhibitorsof glutamine synthetase (GS) activity. The sap levels of NO3,NH4+, glutamine, and asparagine was used to ascertain the invivo inhibition of both enzymes. The tungstate effects werealso checked by measuring leaf in vitro NA activity and NR proteincontent. Xylem exudation rate of detopped seedlings fed withKNO3 decreased when the nitrate assimilation pathway was blockedeither at the NR or at GS sites. This decrease was preventedwhen urea (acting as NH4+ supply) was given simultaneously withtungstate. KNO3 does not act directly on exudation, but throughthe involvement of NH4+. The involvement of glutamine was alsoshown since GS inhibition resulted in a cancellation of theenhancing effect of KNO3 on exudation. As change of exudationrate was not linked to change in sap osmolarity, it is assumedthat the assimilation chain could modify root water conductance.The role of glutamine was discussed. An experimentally-based modelling technique was developed todescribe quantitatively the uptake, flow, storage and utilizationof NO3-N over a 9 d period in mid-vegetative growth of sandcultured castor bean (Ricinus communis L.) fed 12 mol m–3nitrate and exposed to a mean salinity stress of 128 mol m–3NaCl. Model construction used information on increments or lossesof NO3-N or total reduced N in plant parts over the study periodand concentration data for NO3-N and reduced (amino acid) Nin phloem sap and pressure-induced xylem exudates obtained fromstem, petiole and leaf lamina tissue at various levels up ashoot. The resulting models indicated that the bulk (87%) of incomingnitrate was reduced, 51% of this in the root, the remainderprincipally in the laminae of leaves. The shoot was 60% autotrophicfor N through its own nitrate assimilation, but was oversuppliedwith surplus reduced N generated by the root and fed to theshoot through the xylem. The equivalent of over half (53%) ofthis N returned to the root as phloem translocate and, mostly,then cycled back to the shoot via xylem. Nitrate comprised almosthalf of the N of most xylem samples, but less than 1% of phloemsap N. Laminae of leaves of different age varied greatly inN balance. The fully grown lower three leaves generated a surplusof reduced N by nitrate assimilation and this, accompanied byreduced N cycling by xylem to phloem exchange, was exportedfrom the leaf. Leaf 4 was gauged to be just self-sufficientin terms of nitrate reduction, while also cycling reduced N.The three upper leaves (5–7) met their N balance to varyingextents by xylem import, phloem import (leaves 6 and 7 only)and assimilation of nitrate. Petioles and stem tissue generallyshowed low reductase activities, but obtained most of theirN by abstraction from xylem and phloem streams. The models predictedthat nodal tissue of lower parts of the stem abstracted reducedN from the departing leaf traces and transferred this, but notnitrate, to xylem streams passing further up the shoot. As aresult, xylem sap was predicted to become more concentratedin N as it passed up the shoot, and to decrease the ratio ofNO3-N to reduced N from 0·45 to 0·21 from thebase to the top of the shoot. These changes were reflected inthe measured N values for pressure-induced xylem exudates fromdifferent sites on the shoot. Transfer cells, observed in thexylem of leaf traces exiting from nodal tissue, were suggestedto be involved in the abstraction process. Low temperature (6 C) growth was examined in two cultivarsof Vicia faba L. supplied with 4 and 20 mol m–3 N as nitrateor urea. Both cultivars showed similar growth responses to increasedapplied-N concentration regardless of N-form. Total leaf areaincreased, as did root, stem and leaf dry weight, total carboncontent and total nitrogen content. In contrast to findingsat higher growth temperatures, 20 mol m–3 urea-N gavesubstantially greater growth (all parameters measured) than20 mol m–3 nitrate-N. The increased carbon content per plant associated with increasedapplied nitrate or urea concentration, or with urea in comparisonto nitrate, was due to a greater leaf area per plant for CO2uptake and not an increased CO2, uptake per unit area, carbon,chlorophyll or dry weight, all of which either remained constantor decreased. Nitrate reductase activity was substantial inplants given nitrate but negligible in plants given urea. Neitherfree nitrate nor free urea contributed greatly to nitrogen levelsin plant tissues. It is concluded that there is no evidence for a restrictionin nitrate reduction at 6 C, and it is likely that urea givesgreater growth than nitrate because of greater rates of uptake. In hydroponically grown Lycopersicon esculentum (L.) Mill. cv.F144 the site of NO3 reduction and assimilation withinthe plant was shifted from the shoot to the root by salinity.Uptake of NO3 from the root solution was strongly inhibitedby salinization. Consequently, NO3 concentrations inthe leaf, stem and root tissues as well as the nitrate reductaseactivities of the leaves were lower in salinized than in controlplants. Lower NO3, but higher reduced-N, concentrationswere observed in the xylem sap as a result of the enhanced participationof the root in NO3 reduction in salinized plants. Lowerstem K+ concentrations and leaf malate concentrations were foundin salinized compared to control plants which indicates reducedfunctioning of the K+–shuttle in the salinized plants. Incorporation of inorganic carbon by the root was determinedby supplying a pulse of NaH14CO3 followed by extraction andseparation of the labelled products on ion exchange resins.The rate of H14CO3 incorporation was c. 2-fold higherin control than in salinized plants. In salinized plants theproducts of H14CO3 incorporation within the roots werediverted into amino acids, while the control plants divertedrelatively more 14C into organic acids. Products of inorganiccarbon incorporation in the roots of salinized plants providean anaplerotic source of carbon for assimilation of reducedNO3 into amino acids, while in control plants the productswere predominantly organic acids as part of mechanisms to maintainionic balance in the cells and in the xylem sap. The effects of NO-3 and NH+4 nutrition on hydroponically grownwheat (Triticum aestivum L.) and maize (Zea mays L.) were assessedfrom measurements of growth, gas exchange and xylem sap nitrogencontents. Biomass accumulation and shoot moisture contents ofwheat and maize were lower with NH+4 than with NO-3 nutrition.The shoot:root ratios of wheat plants were increased with NH+4compared to NO-3 nutrition, while those of maize were unaffectedby the nitrogen source. Differences between NO-3 and NH+4-fedplant biomasses were apparent soon after introduction of thenitrogen into the root medium of both wheat and maize, and thesedifferences were compounded during growth. Photosynthetic rates of 4 mM N-fed wheat were unaffected bythe form of nitrogen supplied whereas those of 12 mM NH+4-fedwheat plants were reduced to 85% of those 12 mM NO-3-fed wheatplants. In maize supplied with 4 and 12 mM NH+4 the photosyntheticrates were 87 and 82% respectively of those of NO-3-fed plants.Reduced photosynthetic rates of NH+4 compared to NO-3-fed wheatand maize plants may thus partially explain reduced biomassaccumulation in plants supplied with NH+4 compared to NO-3 nutrition.Differences in the partitioning of biomass between the shootsand roots of NO-3-and NH+4-fed plants may also, however, arisefrom xylem translocation of carbon from the root to the shootin the form of amino compounds. The organic nitrogen contentof xylem sap was found to be considerably higher in NH+4- thanin NO-3-fed plants. This may result in depletion of root carbohydrateresources through translocation of amino compounds to the shootin NH+4-fed wheat plants. The concentration of carbon associatedwith organic nitrogen in the xylem sap of maize was considerablyhigher than that in wheat. Arabidopsis halleri is a Cd hyperaccumulator; however, the mechanismsinvolved in the root to shoot translocation of Cd are not wellunderstood. In this study, we characterized Cd transfer fromthe root medium to xylem in this species. Arabidopsis halleriaccumulated 1,500 mg kg–1 Cd in the shoot without growthinhibition. A time-course experiment showed that the releaseof Cd into the xylem was very rapid; by 2 h exposure to Cd,Cd concentration in the xylem sap was 5-fold higher than thatin the external solution. The concentration of Cd in the xylemsap increased linearly with increasing Cd concentration in theexternal solution. Cd transfer to the xylem was completely inhibitedby the metabolic inhibitor carbonyl cyanide 3-chlorophenylhydrazone(CCCP). Cd concentration in the xylem sap was decreased by increasingthe concentration of external Zn, but enhanced by Fe deficiencytreatment. Analysis with 113Cd-nuclear magnetic resonance (NMR)showed that the chemical shift of 113Cd in the xylem sap wasthe same as that of Cd(NO3)2. Metal speciation with Geochem-PCalso showed that Cd occurred mainly in the free ionic form inthe xylem sap. These results suggest that Cd transfer from theroot medium to the xylem in A. halleri is an energy-dependentprocess that is partly shared with Zn and/or Fe transport. Abstract The partitioning of nitrate assimilation between root and shoot of higher plant species is indicated by the relative proportions of total plant nitrate reductase activity (NRA) in the two plant parts and the relative concentrations of nitrate and reduced N in the xylem sap. These have been collated here from the literature and temperate and tropical species compared. Both the distribution of NRA and xylem sap nitrate: reduced N indicate that the following four generalizations can be made.
  • 1 Temperate, perennial species growing in low external nitrate concentrations (about 1 mol m?3) carry out most of their nitrate assimilation in the root. As external nitrate concentration increases (in the range found in agricultural soils, 1–20 mol m?3), shoot nitrate assimilation becomes increasingly important.
  • 2 Temperate, annual legume species growing in low external nitrate concentrations carry out most of their nitrate assimilation in the root. Shoot nitrate assimilation increases in importance as external nitrate concentration is increased.
  • 3 Temperate, annual non-legume species vary greatly in their partitioning of nitrate assimilation between root and shoot when growing in low external nitrate concentrations. Regardless of the proportion carried out in the root at low external nitrate concentrations, nitrate assimilation in the shoot becomes increasingly important as external nitrate concentration is increased.
  • 4 Tropical and subtropical species, annual and perennial, carry out a substantial proportion of their nitrate assimilation in the shoot when growing in low external nitrate concentrations. The partitioning of nitrate assimilation between root and shoot remains constant as external nitrate concentration increases.
It is proposed that a greater proportion of nitrate assimilation occurs in the shoot when an increase in the rate of nitrate uptake does not induce an increase in NR level in the root. Thus, a greater proportion of the nitrate taken up remains unassimilated and is passed into the xylem. A constant partitioning of nitrate assimilation between root and shoot is achieved by balancing NR levels in the root with rates of nitrate uptake. The advantages and disadvantages of assimilating nitrate in either the root or shoot are discussed in relation to temperate and tropical habitats.  相似文献

Kouchi, H. and Higuchi, T. 1988. Carbon flow from nodulatedroots to the shoots of soybean {Glycine max L. Merr.) plants:An estimation of the contribution of current photosynthate toureides in the xylem stream.–J. exp. Bot. 39: 1015–1023. Well-nodulated, water-cultured soybean plants were allowed toassimilate 13CO2 at a constant specific activity for 10 h andthe 13C-labelling of total carbon and ureides in xylem sap wasinvestigated. Labelled carbon appeared very rapidly in the xylem stream. Percentageof labelled carbon (relative specific activity, RSA) in xylemsap was 18% at 2 h after the start of 13CO2 assimilation andreached 53% at the end of the 10 h assimilation. The amountof labelled carbon exported from nodulated roots to the shootsvia the xylem during the 10 h labelling period accounted for33% of total labelled carbon imported into the nodulated roots.Ureides (allantoin and allantoic acid) in xylem sap were stronglydependent on currently assimilated carbon. The RSA of ureidesin xylem sap had reached 83% at the end of the assimilationperiod. Labelled carbon in ureides accounted for 51% of totallabelled carbon returned from nodulated roots to the shootsvia the xylem during the 10 h assimilation period. A treatmentwith 20 mol m–3 nitrate in the culture medium for 2 ddecreased the ureide concentration in the xylem sap slightly,but greatly decreased the RSA of ureides. By comparing the data with the results of analysis of the xylemsap of nodule-detached plants, it was concluded that the majorityof labelled carbon exported to the xylem stream from noduleswas in ureide form. A considerable amount of carbon was alsoreturned from roots to shoots via the xylem stream but it wasmore dependent on (non-labelled) carbon reserved in the roottissues. Seedlings of Ricinus communis L. cultivated in quartz sand weresupplied with a nutrient solution containing either 1 mol m–3NO3 or 1 mol m–3 NH+4 as the nitrogen source. Duringthe period between 41 and 51 d after sowing, the flows of N,C and inorganic ions between root and shoot were modelled andexpressed on a fresh weight basis. Plant growth was clearlyinhibited in the presence of NH+4. In the xylem sap the majornitrogenous solutes were nitrate (74%) or glutamine (78%) innitrate or ammonium-fed plants, respectively. The pattern ofamino acids was not markedly influenced by nitrogen nutrition;glutamine was the dominant compound in both cases. NH+4 wasnot transported in significant amounts in both treatments. Inthe phloem, nitrogen was transported almost exclusively in organicform, glutamine being the dominant nitrogenous solute, but theN-source affected the amino acids transported. Uptake of nitrogenand carbon per unit fresh weight was only slightly decreasedby ammonium. The partitioning of nitrogen was independent ofthe form of N-nutrition, although the flow of nitrogen and carbonin the phloem was enhanced in ammonium-fed plants. Cation uptakerates were halved in the presence of ammonium and lower quantitiesof K+, Na+ and Ca2+ but not of Mg2+ were transported to theshoot. As NH+4 was balanced by a 30-fold increase in chloride in thesolution, chloride uptake was increased 6-fold under ammoniumnutrition. We concluded that ammonium was predominantly assimilated inthe root. Nitrate reduction and assimilation occurred in bothshoot and root. The assimilation of ammonium in roots of ammonium-fedplants was associated with a higher respiration rate. Treatments were applied to vary C and N availability in Alnusglutinosa L. and plant growth, nodule activity (including acetylenereduction) and amino acid composition of the xylem sap weremeasured. Removing the buds, a sink for N, caused a decreasein nodule activity. Flushing root systems daily with 100% O2destroyed nitrogenase activity and substantially decreased theamount of citrulline in the xylem sap. The amino acid compositionof xylem saps also altered according to the mode of N nutrition.In plants fed , xylem sap composition was similar to N2-fixing plants, however, when plants were fed, citrulline content increased. The assimilation and subsequent distribution of nitrate wasfollowed in an experiment in which labelled 15 was added to the base of plant pots. After 12 h7% of root N was from applied 15 and this increased to 75% at 7 d; substantial enrichment ofN from 15 also occurred in stems, buds and leaves. After 7 d, 3.5% of nodule N was from15, consistent with some N being supplied by recycling of shoot N. Xylem saps were alsocollected and after 12 h, glutamate and aspartate were enrichedwith 15N to 53% and 37% increasing after 7 d to 80% and 49%,respectively. Citrulline content of the xylem sap increasedfrom 3 to 9 µmol cm–3 following addition of 15 and at 7 d 80% of the N in the citrullinehad been derived from 15N. It is hypothesized that the growthand activity of A. glutinosa root nodules is sensitive to theN status of the plant and that the level of citrulline (or otheramino acids) returning to the nodules may feed back to regulatenodule growth and activity. Experiments were conducted during the 1974–75 and 1975–76winter season with the barley (Hordeum vulgare L.) cultivarJyoti. From amongst the various plant parts, the flag leaf bladehad higher in vivo nitrate reductase (NR) activity than thelower two leaf blades, glumes, and grains. However, the potentialof a plant part to reduce NO3 is a function of its freshweight and the NR per unit fresh weight. On this basis, thesecond and third leaf blades could reduce more NO3 thanthe flag leaf blade. N fertilizer application resulted in enhancementof the activity of the leaf blades alone. N fertilizer appliedduring the reproductive phase was taken up and assimilated bythe various plant parts. The studies suggest that, even whenthe fertilizer is applied at optimum levels for obtaining maximumyields, the upper leaf blades have sub-optimal NR activity andthat there is a likelihood of either a preferential flow ofNO3 to the leaf blades or transnational barriers to NO3movement to the ear.  相似文献

Orthophosphate (Pi) accumulation by barley (Hordeum vulgareL.) roots was specifically inhibited by NO3 as comparedto Cl and SO42 –, and Pi secretion into the xylemwas stimulated. The inhibition of Pi accumulation by NO3was also observed in roots of intact photosynthesizing horsebean(Vicia faba L.), rice (Oryza sativa L.) and soybean (Glycinemax L.) plants. NO3 effects on Pi transport by rootswere more thoroughly investigated with corn (Zea mays L.). Theywere due to intracellular NO3. Pi secretion was stillstimulated by NO3 after Pi withdrawal from the absorptionsolution. 32Pi influx decreased during Pi accumulation, supportingthe hypothesis that this ion allosterically regulated its owntransport system by feedback control. This control was modulatedby other anions: the decrease was more pronounced in the presenceof nitrate. Chronologically, the depressive effect of NO3on 32Pi influx appeared after the inhibition of Pi accumulation.Furthermore, under conditions where Pi accumulation was notaffected by NO3, 32Pi influx and Pi secretion into thexylem became insensitive to the presence of nitrate. Our hypothesisis that the stimulative effect of NO3 on Pi secretionand the depressive one on 32Pi influx are the repercussionsof an increase in the Pi cytosolic concentration due to an NO3-induced decrease in Pi uptake by the vacuoles. The uptake of ammonium sulphate by 14-month-old potted tea plantsgrown in a glasshouse was studied over the 11-week period followingapplication. Concurrent changes in the starch of root-wood,amino acids of xylem sap, and total nitrogen of leaves, stems,root-wood, and feeder roots were determined. Depletion of nitrogenfrom the soil at different depths and transformation of NH4+to NO3 was also followed. The results show that the uptake of nitrogen commences within2 days of application as indicated by a marked increase in theamino-acid content of the xylem sap. Glutamine and, to a lesserextent, theanine were quantitatively the most important aminoacids in the sap. The amino-acid content of the sap was a maximumat about the time rapid depletion of the ammonium of the soiltook place. An interesting feature of the work is the reciprocalrelationship between the changes in the starch of root-woodand amino acids in the sap a few days after fertilizer application.Studies on the ammonium and nitrate levels of the soil at differentdepths showed that transformation of NH4+ to NO3 occurredin the soil. The response of the various tissues to applied fertilizer nitrogenand increase in the fresh weight of the shoot system showedsimilar trends and may be correlated with the depletion of ammonium-nitrogenfrom the soil.  相似文献

The Occurrence of Nitrate Reduction in the Leaves of Woody Plants   总被引:13,自引:1,他引:12  
Nitrate reductase activities greater than 02 µmol h–1g–1 f. wt, measured by an in vivo assay, occurred in 41per cent of a large sample (555 species) of woody plants. Ifseveral taxonomic groups (Gymnosperms, Ericaceae and Proteaceae)with consistently low activities were discounted activitiesgreater than 02 µmol h–1 g–1 f. wt occurredin 73 per cent of the species. This compares with 93 per centin herbaceous species, suggesting that leaf nitrate reductionis of common occurrence in woody plants. In a small sample ofspecies leaf nitrate reductase activity correlated with nitrateconcentration in the xylem sap. Low activities occurred consistentlyin the Gymnosperms, Ericaceae and Proteaceae. Feeding cut shootsof representatives of these groups with nitrate caused inductionof leaf nitrate reductase activity in the Gymnosperms and Proteaceae,but only limited induction in the Ericaceae. The Ericaceae,with the exception of two species, had low activities and lownitrate reductase inducibility. Root assimilation may predominatein the Gymnosperms and Proteaceae. It is suggested that nitratereduction generally occurs in the leaves of trees from a varietyof plant communities and that this may be related to the lowerenergy cost of leaf, as opposed to root, nitrate assimilation. The responses of seedlings of four species of southwestern Australiansandplain Epacridaceae to added phosphate (P), added NH4N03(N) or Complete nutrients were studied in glasshouse pot cultureusing cores of habitat soil as rooting substrate. Positive responsesto N and Complete nutrients were evident for three species interms of shoot height and shoot dry weight in comparison withControl plants supplied only with deionized water, but no speciesresponded significantly in its shoot growth to P. Root dry weightwas generally less in Complete and N treatments compared toP and Control, leading to considerably higher shoot: root ratiosin the former two treatments. There was no effect of treatmenton infection intensity of hair roots. Root xylem sap compositionshowed greatly elevated levels of nitrogen in the Complete andN treatments and of phosphate in the P treatment. Ammonium comprisedthe major nitrogenous solute of xylem in Control and P treatmentswhile nitrate levels exceeded ammonium in Complete and N treatments.Glutamine levels were particularly high in the P treatments.Labelling of the Complete or N treatments with 15NH3 or 15NO3(supplied as single labelled ammonium nitrate) indicated thatboth forms of N were taken up and incorporated into plant insolubleN. Salinity has been shown to be a major factor contributing to low nitrogen availability in plants. To verify the changes in nitrogen metabolism activity as affected by the exogenous application of proline under salt stress and its relation to salt tolerance, in vitro rice shoot apices were used as a model to study the growth performance and changes in nitrogen assimilation activities in two Malaysian rice cultivars MR 220 and MR 253. Results revealed that salt stress greatly reduced the plant height, shoot nitrate (NO3 ?) content, shoot glutamine synthetase (GS), and root nitrate reductase (NR) activities in both cultivars. Supplementation of proline significantly increased the plant height, number of roots, root NO3 ? content, root NR, and root GS activities under salt stress in both cultivars with greater enhancement in MR 253 than MR 220. The results also indicated that MR 253 possessed higher nitrite reductase (NiR) and glutamate synthase (NADH–GOGAT) activities as compared with MR 220 in all tested treatments. It was suggested that the NO3 ? content, NR, and GS activities played important roles in regulating nitrogen metabolism under salt stress. Taken together, it was concluded that the ability of proline in mitigating salt stress-induced damages was correlated with the changes in nitrogen assimilation activities.  相似文献

Relationships between nitrate (NO-3) supply, uptake and assimilation,water uptake and the rate of mobilization of seed reserves wereexamined for the five main temperate cereals prior to emergencefrom the substrate. For all species, 21 d after sowing (DAS),residual seed dry weight (d.wt) decreased while shoot plus rootd.wt increased (15–30%) with increased applied NO-3concentrationfrom 0 to 5–20 mM . Nitrogen (N) uptake and assimilationwere as great with addition of 5 mM ammonium (NH+4) or 5 mMNO-3but NH+4did not affect the rate of mobilization of seedreserves. Chloride (Cl-) was similar to NO-3in its effect onmobilization of seed reserves of barley (Hordeum vulgare L.).Increased rate of mobilization of seed reserves with additionalNO-3or Cl-was associated with increases in shoot, root and residualseed anion content, total seedling water and residual seed watercontent (% water) 21 DAS. Addition of NH+4did not affect totalseedling water or residual seed water content. For barley suppliedwith different concentrations of NO-3or mannitol, the rate ofmobilization of seed reserves was positively correlated (r >0.95)with total seedling water and residual seed water content. Therate of mobilization of seed reserves of barley was greaterfor high N content seed than for low N content seed. Seed watercontent was greater for high N seed than for low N seed, 2 DAS.Additional NO-3did not affect total seedling water or residualseed water content until 10–14 DAS. The effects of seedN and NO-3on mobilization of seed reserves were detected 10and 14 DAS, respectively. It is proposed that the increasedrate of mobilization of seed reserves of temperate cereals withadditional NO-3is due to increased water uptake by the seedlingwhile the seed N effect is due to increased water uptake bythe seed directly. Fumigation of Aleppo pines with episodes of O3 (up to 110 nll) causes immediate depressions of in vivo nitrate reductase(NaR) activities, slightly delayed reductions in the rates ofethene emissions (typical of O3 plants), steady accumulationsof total polyamines (although putrescine declines), and increasesin pool sizes of reduced glutathione (GSH) and ascorbate incurrent year needles. Severe droughting produces smaller plantswith reduced stomatal conductance and CO2 assimilation ratesas well as lower protein contents. Their roots have low ratesof nitrate uptake but virtually no root NaR activities, whilelevels of shoot activities and NaR-associated proteins are unaffectedalthough they have no substrate. Less severe droughting allowsa restricted uptake of nitrate which is still reflected in reducedNaR activities, protein and total N contents, but the additionalpresence of O3 (up to 120 nl l–1) has no interactive effecton N cycling. Drought and O3 together, however, depress CO2assimilation still further, which can not be accounted for byadditional stomatal closure, but the interactive effects ofdrought and air pollution reduce levels of total phenols, GSHand ascorbate which, combined with a 12-fold reduction in glutathionereductase-(GR)-associated proteins, point to an increased susceptibilityof Aleppo pines to photoinhibition as a reason for their currentdecline in Mediterranean areas. The growth of four heathland species, two grasses (D. flexuosa,M. caerulea) and two dwarf shrubs (C. vulgaris, E. tetralix),was tested in solution culture at pH 4.0 with 2 mol m–3N, varying the N03/NH4+ ratio up to 40% nitrate. In addition,measurements of NRA, plant chemical composition, and biomassallocation were carried out on a complete N03/NH4+ replacementseries up to 100% nitrate. With the exception of M. caerulea, the partial replacement ofNH4+ by NO3 tended to enhance the plant's growth ratewhen compared to NH4+ only. In contrast to the other species,D. flexuosa showed a very flexible response in biomass allocation:a gradual increase in the root weight ratio (RWR) with NO3increasing from 0 to 100%. In the presence of NH4+, grassesreduced nitrate in the shoot only; roots did not become involvedin the reduction of nitrate until zero ambient NH4+. The dwarfshrubs, being species that assimilate N exclusively in theirroots, displayed an enhanced root NRA in the presence of nitrate;in contrast to the steady increase with increasing NO3in Calluna roots, enzyme activity in Erica roots followed arather irregular pattern. Free nitrate accumulated in the tissuesof grasses only, and particularly in D. flexuosa. The relative uptake ratio for NO3 [(proportion of nitratein N uptake)/(proportion of nitrate in N supply)] was lowestin M. caerulea and highest in D. flexuosa. Whereas M. caeruleaand the dwarf shrubs always absorbed ammonium highly preferentially(relative uptake ratio for NO3 <0.20), D. flexuosashowed a strong preference for NO3 at low external nitrate(the relative uptake ratio for N03 reaching a value of2.0 at 10% NO3). The ecological significance of thisprominent high preference for NO3 at low NO3/NH4+ratio by D. flexuosa and its consequences for soil acidificationare briefly discussed.
