Two seed lots of Calluna vulgaris were obtainedfrom English Nature (seed of Cornish provenance) (EN) and John ChambersWildflower Seeds (JCWS). In laboratory tests, under continuous light untreatedseeds of both seed lots were partially dormant at temperatures between14–35 °C, but JCWS seeds were more deeply dormant thanENseeds. The optimum temperature for germination for both lots was ca 18°C. Germination of EN seeds was much lower in the dark than inthe light at all temperatures; JCWS seeds did not germinate in the dark. In thelight at 22 °C, dormancy of both seed lots was broken whenseeds were incubated in GA4/7 solution(2×10–4 M). Dormancy ofJCWSseeds at 22 °C in the light was broken when seeds wereincubated in four different smoke solutions but more so when used incombinationwith GA4/7. Soaking seeds for 4h insmoke/GA4/7solutions before sowing improved both the speed andpercentage germination in pot experiments on a mist bench in the glasshouse byat least 10-fold. Soaking with GA4/7 alone produced a 5-fold increasein germination but seedlings were more etiolated than with thesmoke/GA4/7 mixtures. A seed advancement treatment modified from thatused commercially on sugar beet seeds also promoted germination in bothlaboratory and glasshouse tests. This entailed soaking seeds in 0.2% thiramsuspension for 4h followed by incubation in excess solution at 22°C for 4 days. This treatment was not as effective as thesmoke/GA4/7 seed soaks.  相似文献

Flixweed is one of the most abundant weeds in North America and China, and causes a reduction in crop yields. Dormancy of flixweed seeds is deep at maturity and is maintained in soil for several months. To identify regulators of seed dormancy and germination of flixweed, the effect of environmental and hormonal signals were examined using dormant and non-dormant seeds. The level of dormancy was decreased during after-ripening and stratification, but long imbibition (over 5 days) at 4 °C in the dark resulted in the introduction of secondary dormancy. The strict requirement of duration of cold treatment for the break of dormancy may play a role in the seasonal regulation of germination. The germination of non-dormant flixweed seeds was critically regulated by red (R) and far-red (FR) light in a photoreversible manner. Sodium nitroprusside, a donor of nitric oxide (NO), promoted germination of half-dormant seeds, suggesting that NO reduced the level of seed dormancy. As has been shown in other related species, light elevated sensitivity to GA4 in dark-imbibied flixweed seeds, but cold treatment did not affect GA4-sensitivity unlike in Arabidopsis. Taken together, our results indicate that seed germination in flixweed and its close relative Arabidopsis is controlled by similar as well as distinct mechanisms in response to various endogenous and environmental signals.  相似文献

Nondormant A. caudatus seeds germinated in the darkat temperatures between 20 and 35° but not at 45 °C.Incubation at this temperature for at least 10 h inhibited seedgermination over the temperature range 20 to 35 °C,temperatures previously suitable for germination. Thus incubation at 45°C induced secondary dormancy. Mechanical or chemicalscarification or exposure to pure oxygen caused complete or almost completegermination of dormant seeds although more slowly in comparison to nondormantseeds. Secondary dormant scarified seeds required a lower concentration of ABAthan nondormant seeds to inhibit germination. The high temperature, whichinduced dormancy, 45 °C, caused the seed coat to be partiallyresponsible for secondary dormancy. Involvement of ABA (synthesis orsensitivity) in the induction and/or maintenance of this dormancy should beconsidered.  相似文献

The effect of the application of gibberellins to Tagetes minuta L. achenes (seeds) was determined at both 25°C, the optimal germination temperature, and 35°C, at which temperature the achenes are thermoinhibited. Both GA3 and GA4+7 accelerated germination at 25°C. Seed germination at 25°C was inhibited by paclobutrazol, but on subsequent application of GA4+7 rapid germination was induced. Following application of GA3 or GA4+7 to thermoinhibited seeds, a significantly higher final germination percentage was observed than in the distilled water control. However, endogenous gibberellin levels in germinating and thermoinhibited seeds did not differ significantly.  相似文献

Abscisic acid (ABA) levels in seeds from three cultivars of apple (Malus domestica Borkh.) which have substantially different chilling requirements were investigated by gas chromatography mass-spectrometry selected ion monitoring (GCMS-SIM) during stratification. The ABA content of dormant unchilled seeds was similar in the three cultivars, suggesting no relationship between the chilling requirement of those seeds and their ABA status. That chilling is not related to ABA changes during stratification was confirmed by warm (20°C) and cold (5°C) stratification experiments. ABA content dropped rapidly and nearly identically under both temperature regimes, but only cold stratification promoted germination. The decline in ABA during stratification was due in large part to leaching from the seed coat and nucellar membrane; the ABA content of the embryo remained nearly constant. The radicle in intact seeds stratified at 5°C began growing 20–30 days after the ABA in the seed coat and nucellar membrane had nearly disappeared. Radicle growth did not occur in unchilled seeds, even though ABA had leached from them as well. It is possible that the leaching of ABA from the seed allows certain promotive forces to develop, but if so, these can develop only at chilling temperatures. Dormant Amaranthus retroflexus seeds do not germinate in the dark at temperatures below 35°C. Fully dormant seeds germinate only at 35–40°C whereas non-dormant ones germinate within a wider range of temperatures (15 to 40°C). Germination of non-dormant seeds requires at least 10% oxygen, but the sensitivity of seeds to oxygen deprivation increases with increasing depth of dormancy. 10–6 to 10–4 M ethephon, 10–3 M 1-aminocyclopropane 1-carboxylic acid (ACC) and 10–3 M gibberellic acid (GA3) break this dormancy. In the presence of 10–3 M GA3 dormant seeds are able to germinate in the same range of temperatures as non-dormant seeds. The stimulatory effect of GA3 is less dependent on temperature than that of ethephon, while ACC stimulates germination only at relatively high temperatures (25–30°C). Celery (Apium graveolens L.) plants cv. Jason overwintered in a polythene tunnel flowered earlier and grew taller than similar plants given a 10-week cold-treatment at 5°C prior to transplanting in the same tunnel in mid-February. However, there was no significant difference in the yield of seeds obtained from both treatments, plants grown at a density of 4m-2 yielded less seeds than those at 2m-2, though the yield per unit area was slightly higher from the high density treatment. Treatment with 100 mgl-1 GA3 applied twice just prior to flowering and during anthesis increased flower stalk, flower pedicel and stamen length but delayed flower opening and seed ripening and decreased seed set and seed yield. Treatment with a mixture of 1000 mgl-1 GA4 and GA7 plus 1000 mgl-1 ethephon on three occasions during seed ripening decreased seed yield and reduced seed germination though those seeds capable of germinating were less dormant than seeds from untreated plants. The size distribution of seeds was unaffected by any treatment other than the preseeding spray with GA3 which reduced the percentage of medium-size seeds.  相似文献

The effect of temperature on the level of dormancy of primary and secondary dormant Carex pendula and Carex remota seeds was investigated. Primary dormant and secondary dormant seeds were stratified for 4 weeks at 5, 11, 13, and 15 °C, respectively, and tested for germination at 15/5 °C in light. To obtain secondary dormant seeds, primary dormant seeds were stratified at 5 °C and afterwards at 25 °C for 4 weeks. Germination tests were carried out in water and in 25 μmol KNO3-solution to examine differences in sensitivity to nitrate between seeds relieved from primary and secondary dormancy. In both species, seeds with primary and with induced secondary dormancy showed no significant differences in germination. The two sedges showed significant differences in the effect of stratification temperatures between primary and secondary dormant seeds. Primary dormant seeds of C. pendula showed high germination (>80%) in nitrate-solution after stratification at all temperatures, while only temperatures of 5, 11, and 13 °C led to higher germination in nitrate-solution in secondary dormant seeds. Germination percentages of primary and of secondary dormant C. pendula seeds in water increased to a higher extent only after stratification at 5 and 11 °C; stratification of 11 °C was more effective in secondary than in primary dormant seeds. The only temperature that relieved primary dormancy in C. remota seeds was 5 °C where germination in water and nitrate-solution was >90%. Germination of secondary dormant seeds was increased by stratification at 11 °C independent of the test solution but higher germination after stratification at 13 °C occurred only in nitrate-solution. The results support the existence of physiological differences in the regulation of primary and secondary dormancy by temperature, and in the reaction of nitrate, at least in C. remota.  相似文献

应用酶联免疫吸附测定法(ELISA)研究了梾木种子低温层积过程中内源激素含量的动态变化,分析了内源激素与种子休眠与发芽的关系。结果表明:(1)梾木种子中IAA含量在层积处理初期剧烈降低,持续一段时间后又显著升高,但后期下降,且IAA/ABA也出现同样的变化;种子中ABA含量在层积处理前期较高,但随着处理时间的延长趋于下降;种子内GA1/3含量以及GA1/3/ABA均随层积处理时间的延长逐渐增大;种子内ZRs和iPAs含量的变化相对较为平稳,尽管有一定的波动,但整体呈渐趋增高趋势。(2)梾木种子发芽率及发芽势在未经层积处理以及处理时间少于90d的条件下均为0,但随着层积处理时间的延长二者明显上升,层积处理的时间长短对梾木种子萌发有显著影响。(3)相关分析表明,梾木种子内GA1/3含量与种子的发芽率、发芽势均呈显著正相关关系,相关系数分别为0.688、0.662;种子内GA1/3/ABA增大有利于种子休眠解除和萌发。  相似文献

The activities of several gibberellins in stimulating germination of wild-type and GA-deficient gal seeds of Arabidopsis thaliana were compared. Of the six compounds tested GA4 and GA7-isolactone had the highest activity and GA7 and GA9 the lowest; activities of GA1 and GA3 were intermediate. Combined application of pure GAs presented no indications that more than one GA receptor is involved. Four GAs were identified in extracts from wild-type and GA-insensitive gai seeds by combined gas chromatography mass spectrometry: GA1, GA3, GA4 and GA9. Effects of light and chilling on levels of GA1, GA4 and GA9 were studied using deuterated standards. Light increased both GA levels and germination in unchilled wild-type and gai seeds. As a result of irradiation GA levels in gai seeds were 7–10 times as high as in wild-type seeds. In the dark germination was 0%, in the light 14% of gai seeds and 95% of wild-type seeds germinated. A chilling pre-treatment of 7 days at 2°C was required to enhance further the germination of gai seeds in the light. Light did not increase GA levels of chilled seeds of either genotype; levels of GA4 and GA9 of chilled gai seeds, in the light were respectively 7 and 12 times lower than in non-chilled seeds, whereas the latter seeds germinated better. Slightly elevated levels of GA4 were detected in darkness after chilling, but germination capacity was still 0%. These results strengthened the conclusion that GAs are required for germination of A. thaliana seeds, whereby GA4 has intrinsic biological activity. However, it is unlikely that light and chilling stimulate germination primarily by increasing levels of GA. Instead GA sensitivity is a possible alternative.  相似文献

Mature seeds of the Cape Verde Islands (Cvi) ecotype of Arabidopsis thaliana (L.) Heynh. show a very marked dormancy. Dormant (D) seeds completely fail to germinate in conditions that are favourable for germination whereas non-dormant (ND) seeds germinate easily. Cvi seed dormancy is alleviated by after-ripening, stratification, and also by nitrate or fluridone treatment. Addition of gibberellins to D seeds does not suppress dormancy efficiently, suggesting that gibberellins are not directly involved in the breaking of dormancy. Dormancy expression of Cvi seeds is strongly dependent on temperature: D seeds do not germinate at warm temperatures (20–27°C) but do so easily at a low temperature (13°C) or when a fluridone treatment is given to D seeds sown at high temperature. To investigate the role of abscisic acid (ABA) in dormancy release and maintenance, we measured the ABA content in both ND and D seeds imbibed using various dormancy-breaking conditions. It was found that dry D seeds contained higher amounts of ABA than dry ND after-ripened seeds. During early imbibition in standard conditions, there was a decrease in ABA content in both seeds, the rate of which was slower in D seeds. Three days after sowing, the ABA content in D seeds increased specifically and then remained at a high level. When imbibed with fluridone, nitrate or stratified, the ABA content of D seeds decreased and reached a level very near to that of ND seeds. In contrast, gibberellic acid (GA3) treatment caused a transient increase in ABA content. When D seeds were sown at low optimal temperature their ABA content also decreased to the level observed in ND seeds. The present study indicates that Cvi D and ND seeds can be easily distinguished by their ability to synthesize ABA following imbibition. The effects of water temperature and bottom sediment type were studied on seed dormancy and germination of Zostera japonica Ascherson & Graebner in mesocosm. To test whether the germination rate is affected by cold stratification, seeds were divided into two groups: those exposed to cold (7 °C) and those left untreated (23–15 °C). Additionally, to mimic tidal variation, we used five tidal depth treatments for germination experiments in mesocosm. In mesocosm tanks, there was a wide range of daily fluctuating temperature at datum line +40 cm (17–25 °C), D.L. +20 cm (15 °C), and D.L. +0 cm (4–7 °C). In contrast, the maximum temperature range at D.L. −20 cm and −40 cm was narrow (5–6 °C). In the no cold stratification group, the maximum germination rates on sandy, muddy sand, and muddy bottom sediment were 3%, 11%, and 3%, respectively. In the cold stratification group (7 °C), the maximum germination rates were 40%, 53%, and 54%, respectively. First germination was observed 36 ± 0 days and 43 ± 6 days after the start of the germination experiment in the cold stratification group and the no cold stratification group, respectively. Bottom sediment type and tidal level did not affect seed germination in the both stratification group. Cold stratification strongly increases germination in all sediment types tested and under varying temperature regimes and at different tidal levels. We also tested whether seed germination is affected by daily fluctuations in temperature (10 °C constant, 15 °C/10 °C, and 20 °C/10 °C were compared) in an indoor incubator. Forty-two days after being sown, the maximum seedling emergence rates in the three groups were 3 ± 5%, 21 ± 7%, and 42 ± 26%, respectively. At 20 °C/10 °C, first germination was observed 11 days after the start of incubation, the germination rate rose sharply after 18 day of incubation, and then it leveled off after 32–42 days of incubation. In the no cold stratification group, seed germination was not observed in any of the three treatments. This finding suggests that the breaking of seed dormancy and germination of Z. japonica seeds are determined strongly by cold temperature and daily fluctuations of temperature, respectively.  相似文献

Freshly harvested, dormant seeds of Amaranthus retroflexus were unable to germinate at 25 and 35 °C. To release their dormancy at the above temperatures, the seeds were stratified at a constant temperature (4 °C) under laboratory conditions or at fluctuating temperatures in soil or by outdoor burial in soil. Fully dormant, or seeds stratified or buried (2006/2007 and 2007/2008) for various periods were treated with exogenous gibberellic acid (GA3), ethephon and abscisic acid (ABA). Likewise, the effects of these regulators, applied during stratification, on seed germination were determined. The results indicate that A. retroflexus seed dormancy can be released either by stratification or by autumn–winter burial. The effect of GA3 and ethylene, liberated from ethephon, applied after various periods of stratification or during stratification, depends on dormancy level. GA3 did not affect or only slightly stimulated the germination of non-stratified, fully dormant seeds at 25 and 35 °C respectively. Ethylene increased germination at both temperatures. Seed response to GA3 and ethylene at 25 °C was increased when dormancy was partially removed by stratification at constant or fluctuating temperatures or autumn–winter burial. The response to GA3 and ethylene increased with increasing time of stratification. The presence of GA3 and ethephon during stratification may stimulate germination at 35 °C. Thus, both GA3 and ethylene can partially substitute the requirement for stratification or autumn–winter burial. Both hormones may also stimulate germination of secondary dormant seeds, exhumed in September. The response to ABA decreased in parallel with an increasing time of stratification and burial up to May 2007 or March 2008. Endogenous GAn, ethylene and ABA may be involved in the control of dormancy state and germination of A. retroflexus. It is possible that releasing dormancy by stratification or partial burial is associated with changes in ABA/GA and ethylene balance and/or sensitivity to these hormones.  相似文献

Investigations on seeds of Scrophularia marilandica L. were undertaken to determine their germination requirements. Seeds were collected from three naturally occurring sites and one greenhouse-grown population in London, Ontario in September and October of 1997. Some were set to germinate immediately after collection; others were stored in or on soil outside and/or under controlled laboratory conditions before testing. Germination was assessed under two light/temperature regimes (35°C 14 h light, 20°C 10 h dark and 25°C 14 h light, 10°C 10 h dark), in continuous darkness, and in the presence of two germination-promoting chemicals (GA3 and KNO3). Fresh seeds germinated best at 35/20°C, while stored seeds germinated best at 25/10°C. No differences in percent germination were found among three seed-maturity stages. All chemical treatments, except 0.01 M KNO3, increased percent germination. Significant differences were found both among and within sites for most chemical treatments, but exposure to 3 × 10−4 M GA3 caused almost every seed to germinate. When compared to the control, both the gibberellic acid and the soil-storage treatments contributed to faster germination. Exposure of seeds to naturally prevailing conditions on the soil surface followed by testing under the 25/10°C regime produced the highest percent germination. No seeds germinated in the dark. In summary, seeds of S. marilandica exhibit physiological dormancy, which can be alleviated by exposure to light, after-ripening and/or cold stratification. It is probable that the differences in germination response among sites can be attributed to differences in environmental conditions during seed production. These experiments indicate that the seeds of S. marilandica must be buried shortly after dispersal in order to form a persistent seed bank.  相似文献

Methyl jasmonate (JA-Me) inhibited or retarded germination of Amaranthus caudatus seeds in darkness at 24°C, Ethephon, ACC and gibberellins (GA3 or GA4+7) partially or completely reversed this inhibition depending on the concentration of JA-Me applied. Both ethephon and the gibberellins were more effective than ACC. The percentage germination of seeds of parsley cv. Imperial Curled was higher in the light than in the dark, the high temperature limits for germination being 30 and 28°C for light and dark respectively. At the higher temperatures, the germination rate was slower in the dark. At 30°C, treatment with a gibberellin A4/7 mixture at 2 × 10–4 M partially alleviated the inhibiting effect of darkness on the germination percentage. Pre-incubation of parsley seeds at 35°C in the dark for 30 h increased the rate, but decreased the percentage, of germination of seeds incubated at 15°C in the light. Germination and seedling emergence studies were made on seed harvested from four different umbel positions. Although heavier seeds were produced from primary umbels than from other umbel orders, they were less viable as measured by seedling emergence in the glasshouse. The rate of emergence was decreased with increasing umbel order i.e. with later seed development: this was reflected in subsequent seedling weights, with seedlings from quarternary umbel seeds being about half the weight of those from primary umbel seeds. BA at 10–5 M, GA3 at 3×10–4 M or GA4+7 at 3×10–5 M partially or largely reversed the inhibition of Amaranthus caudatus seed germination due to JA-Me. BA or GA3 did not affect ethylene production and ACC oxidase activity in vivo in the presence of JA-Me before radicle protrusion. However, both increased ethylene production after 72 h of incubation, when the reversal of the JA-Me inhibition of seed germination was observed. AVG at 3×10–4 M decreased ethylene production when it was applied simultaneously with BA and JA-Me or GA3 and JA-Me, but it had no effect on seed germination. NBD almost completely reversed the stimulatory effect of BA, GA3 or GA4+7 on the germination of seeds in the presence of JA-Me. Exogenous ethylene reversed the inhibitory effect of NBD. The results indicate that action of endogenous ethylene is involved in the response of JA-Me inhibited seeds to BA or GAs.  相似文献

Unimbibed Amaranthus caudatus seeds were found to contain stachyose, raffinose, verbascose, sucrose, galactinol, myo-inositol, glucose and fructose, while no galactose, maltose and maltotriose was detected. During imbibition, seed concentrations of verbascose, stachyose, raffinose, galactinol, myo-inositol (temporary) and fructose (transient) were observed to decrease; concentrations of galactose and maltose remained fairly constant, while those of sucrose, glucose and maltotriose increased, the increase in sucrose concentration was only temporary. Effects of gibberellin A3 (GA3) at 3 × 10−4 M and ethephon at 3 × 10−4 M alone or in the presence of methyl jasmonate (Me-JA) at 10−3 M on concentrations of soluble sugars during germination of A. caudatus seeds were examined. Me-JA was found to inhibit seed germination and fresh weight of the seeds, but did not affect sucrose, myo-inositol, galactose and maltose concentrations during imbibition for up to 20 h. The exogenously applied GA3 was observed to enhance germination, stachyose breakdown and glucose concentration after 20 h of incubation. Ethephon stimulated seed germination as well as utilisation of stachyose, galactinol (both after 14 and 20 h) and raffinose (after 14 h of incubation). Although the stimulatory effect of either GA3 or ethephon on seed germination was blocked by Me-JA; these stimulators increased mobilisation of raffinose and stachyose, but only ethephon enhanced both glucose and fructose after 14 and/or 20 h of incubation in the presence of Me-JA. The maltose concentration was increased by both GA3 and ethephon alone and in the presence of Me-JA. Of the growth regulators studied, ethephon alone and/or in combination with Me-JA significantly increased the concentrations of glucose, fructose, galactose, maltose and maltotriose. The differences in sugar metabolism appear to be linked to ethylene or GA3 applied simultaneously with Me-JA.  相似文献

We tested the hypothesis that seeds of the monocarpic perennial Ferula gummosa from the Mediterranean area and central Asia have deep complex morphophysiological dormancy. We determined the water permeability of seeds, embryo morphology, temperature requirements for embryo growth and seed germination and responses of seeds to warm and cold stratification and to different concentrations of GA3. The embryo has differentiated organs, but it is small (underdeveloped) and must grow inside the seed, reaching a critical embryo length, seed length ratio of 0.65–0.7, before the seed can germinate. Seeds required 9 weeks of cold stratification at <10°C for embryo growth, dormancy break and germination to occur. Thus, seeds have morphophysiological dormancy (MPD). Furthermore, GA3 improved the germination percentage and rate at 5°C and promoted 20 and 5% germination of seeds incubated at 15 and 20°C, respectively. Thus, about 20% of the seeds had intermediate complex MPD. For the other seeds in the seed lot, cold stratification (5°C) was the only requirement for dormancy break and germination and GA3 could not substitute for cold stratification. The temperature-dependent primary dormancy of cv Florida 683 celery seeds in darkness was broken by GA4/7 (2 × 10-4 M) alone but other growth regulators such as BA, ethephon or daminozide were necessary to break dormancy of cv Lathom Blanching seeds in the presence of GA4/7 at this concentration. Although AgNO3 partially inhibited both the ethephon- and BA- induced germination of cv Lathom Blanching seeds in the presence of GA4/7 in the dark it did not affect the promotive action of daminozide. Ethephon did not overcome the inhibitory action of high concentrations of AgNO3 in the light. The ethylene synthesis inhibitor aminoethoxyvinylglycine (AVG) did not inhibit the germination of cv Lathom Blanching seeds induced by growth regulators in the dark or in the absence of growth regulators in the light. Fusicoccin (FC) did not break celery seed dormancy unless applied in the presence of GA4/7. Germination of cv Lathom Blanching celery seeds treated with GA4/7 at 16°C in the dark was inhibited by the K+ ionophore benzo-18-crown-C-6 (18-C-6) and in the presence of Ca2+ by the Ca2+ ionophore A23187; the 18-C-6 inhibition was reversed by BA.It is concluded that the involvement of gibberellin in celery seed dormancy is not dependent on endogenous ethylene and is directly or indirectly controlled through the action of other hormones on transmembrane ion fluxes.  相似文献
