Two-node explants from Sweet Orange cv. St Ives Valencia orangeshoots produced prolific callus and formed secondary abscissionzones within internodes when cultured in vitro with abscisicacid (ABA, 5 µM) or -naphthaleneacetic acid (NAA, 5 µM).Benzyladenine (BA, 1 µm) induced callus but had littleeffect on abscission. Secondary abscission zone formation wasassociated with ABA-induced and auxin-induced ethylene formation.Treatment of explants with inhibitors of ethylene synthesis[aminoethoxyvinyl glycine (AVG), Co2+, PO43–] preventedformation of secondary abscission zones but had variable effectson callus formation. Newly made explants contained high concentrationsof endogenous ABA (up to 6000 ng g–1 f.wt), as measuredby GC/MS/SIM. Long-term subculture of explants (two years) inmedia containing BA (1 µm) led to a reduction in endogenousABA level (40 ng g–1 f. wt) and to loss of capacity toform extensive callus and secondary abscission zones. Elongation of coleoptile segments, having or not having a tip,excised from rice (Oryza sativa L. cv. Sasanishiki) seedlingswas promoted by exogenous ethylene above 0.3 µl l–1as well as by IAA above 0.1 µM. Ethylene production ofdecapitated segments was stimulated by IAA above 1.0µM,and this was strongly inhibited by 1.0 µM AVG. AVG inhibitedthe IAA-stimulated elongation of the decapitated segment witha 4 h lag period, and this was completely recovered by ethyleneapplied at the concentration of 0.03 µl l–1, whichhad no effect on elongation without exogenous IAA. The effectsof IAA and ethylene on elongation were additive. These factsshow that ethylene produced in response to IAA promotes ricecoleoptile elongation in concert with IAA, probably by prolongingthe possible duration of the IAA-stimulated elongation, butthat they act independently of each other. Moreover, AVG stronglyinhibited the endogenous growth of coleoptile segments withtips and this effect was nullified by the exogenous applicationof 0.03 µl l–1 ethylene. These data imply that theelongation of intact rice coleoptiles may be regulated cooperativelyby endogenous ethylene and auxin in the same manner as foundin the IAA-stimulated elongation of the decapitated coleoptilesegments. The ethylene production rate of cut sweet pea flower buds increased37-fold during the first 48 h of their vase life. This increasein ethylene production was accompanied by petal wilting at 72h and abscission of the buds 24 h later. This paper presents an analysis of abscission reponses of cottonexplants to (a) 1-naphthaneneacetic acid; (b) photodecompositionproducts of 1-naphthaleneacetic acid: 1-methylnaphthalene, 1-naphthaldehyde,1-naphthoic acid, naphthalene, and phthalic acid; and (c) arelated compound: naphthaleneacetyl aspartate. Abscission wasaccelerated by low amounts and retarded by high amounts of 1-naphthaleneaceticacid and 1-naphthoic acid. No significant effect on abscissionwas observed from 1- methylnaphthalene, 1-naphthaldehyde, orphthalic acid applied in amounts from 10–8 to 10.0 µgper petiole; or with naphthalene from 10–3 to 10.0µgper petiole. Naphthaleneacetyl aspartate had no effect at 5?104to 5?10–3 µg per petiole, but completely inhibitedabscission at 5 ? 10–1 and 5.0 µg per petiole. Thedata are analyzed on part by a previously described mechanicalmethod for the determination of abscission indexes, and in partby a new method described herein, using a digital computer forthe analysis of the abscission time-course data. A survey of avocado (Persea americana Mill.) fruit drop andethylene production was carried out during fruit development.Natural drop of avocado fruitlets started soon after set (May)and continued at a gradually decreasing rate until September,except for a temporarily increasing rate in late July. Fruitletsweighing up to 0?2 g dropped at a rate of over 30 percent perweek. With larger fruits, the rate was under 1 percent per week.Fruit drop ceased after September, when fruit growth declinedand the seed coat began to shrivel. A positive correlation was found between the rate of fruitletand fruit drop and ethylene production. Fruitlets with defectiveseeds produced ethylene at a very high rate of 7–10 timesmore than apparently normal fruits. The high incidence of defectiveseeds might be the cause of the very high levels of ethyleneproduction by young avocado fruitlets. The seed was found to be the main site of ethylene productionin fruitlets. Abscisic acid (ABA) levels in young abscissing fruits were 7times as high as those in non-abscissing fruits.  相似文献

Studies on the effect of fruit size on endogenous ABA and isopentenyladenine(iP) in developing avocado (Persea americana Mill. cv. Hass)fruit revealed that ABA content was negatively correlated withfruit size whilst the iP/ABA ratio showed a linear relationshipwith increasing size of fruit harvested 226 d after full bloom.The effect of this change in hormone balance on the relationshipbetween symplastic solute transport and appearance of the smallfruit variant was examined following manipulation of the endogenouscytokinin (CK)/ABA ratio. Application of ABA caused seed coatsenescence and retarded fruit growth but these effects wereabsent in fruit treated with equal amounts of ABA plus iP. Thus,the underlying physiological mechanisms associated with ABA-inducedretardation of Hass avocado fruit growth appeared to be inextricablylinked to a decline in CK content and included: diminution ofmesocarp and seed coat plasmodesmatal branching, gating of mesocarpand seed coat plasmodesmata by deposition of electron densematerial in the neck region, abolishment of the electrochemicalgradient between mesocarp and seed coat parenchyma, and arrestof cell-to-cell chemical communication. A single-gene recessive mutant (Abs-) of Lupinus angustifoliusL. ‘Danja’ that does not abscise any organs wascompared with its parent during continuous exposure of explantsfrom 14 d old seedlings to 10 µl l-1ethylene. Both endo-(1,4)-ß- D -glucanase (cellulase) and polygalacturonase(PGA) activities increased significantly and progressively inpetiole-stem abscission zones of the parent before the onsetof abscission, and were reflected in a rapid decline in breakstrengthfrom 300 to 70 g within 32 h. In the mutant there was negligibleincrease in hydrolytic enzyme activity, breakstrength declinedslowly (to 180–200 g by 72 h) and there was no abscission.Isoelectric focusing showed two cellulase isoforms (pI 5.0 andpI 8.5) expressed in abscission zones of the parent; these wereexpressed at much lower levels in the mutant. These data areinterpreted to indicate that expression of at least two formsof cellulase activity is enhanced by ethylene in normal petioleabscission zones of lupin. PGA activity also increased in theabscission zone tissue of the parent but to a lesser extentin that of the mutant. We attribute the Abs-phenotype to mutationof a gene regulating ethylene-responsive expression of abscission-specifichydrolytic enzymes. Experiments were conducted on developing fruitlet explants of two mango (Mangifera indica L.) cultivars to establish the source and dynamics of ethylene production prior to and during fruitlet abscission. Abscission of all fruits in the samples occurred at approximately 86 and 74 hours postharvest in `Keitt' and `Tommy Atkins,' respectively. Increased abscission began 26 hours from harvest and was preceded by enhanced ethylene synthesis. Enhanced ethylene production initiated approximately 48 hours prior to abscission and increased to a maximum near the time of fruitlet abscission. The seed produced the highest amount of ethylene on a per gram fresh weight basis. The pericarp, however, was the main source of ethylene on an absolute basis, since it represented more than 85% of total fruitlet weight. Pedicels containing the abscission zone produced no detectable ethylene prior to or at the moment of abscission. Fumigation of `Tommy Atkins' fruitlets with 1, 15, or 100 microliters per liter ethylene accelerated abscission by 24 to 36 hours in comparison with unfumigated controls. Diffusion of ethylene from distal fruitlet tissues to the abscission zone triggers the events leading to separation of the fruit from the tree.  相似文献

Imbibition of seeds of oil seed rape (Brassica napus cv Jetneuf)in 10–3 M aminoethoxyvinylglycine (AVG) or 10–2silver thiosulphate (Ag+) had no effect on germination nor onthe emergence of seedlings from uncompacted or lightly compressedsoil, but significantly reduced emergence from moderately compressedsoil of 68.4 or 143.3 N cm–2 impedance. Exertion of force by emerging control seedlings against a staticcantilever bar fitted with strain gauges reached a maximum (Fmax)of 6 g over 10 h. Higher axial forces were achieved when theseedlings were emerging from compressed soil, without any changein the time required to reach Fmax, so that the build-up offorce was considerably (1.8 fold) faster than in uncompressedsoil. This adaptive response to soil impedance was modified by theseed pretreatments employed. Seedlings from AVG or Ag+pretreatedseeds produced lower axial forces than controls, and neitherFmax nor the rate at which force developed showed any responseto soil compression. After pretreatment in 10–3 ethephon or 10–4 naphthaleneacetic acid (NAA) the seedlings achieved similar Fmax to controlseedlings, but responded more rapidly to soil compression sothat the rate of build up of emergence force was 2.3 fold (NAA)or 2.8 fold (ethephon) faster in compressed than in uncompressedsoil. The results suggest that the exertion of force by a seedlingagainst a barrier involves a dynamic response to impedance onthe part of the seedling. This response can be either enhancedor suppressed by substances which affect ethylene productionor ethylene action. Such compounds may have promise for modifyingseedling emergence from impeding soils. To elucidate the mode of action of chromosaponin I (CSI) instimulating the growth of lettuce roots (Lactuca sativa L. cv.Grand Rapids), the possible involvement of ethylene was examined.Lettuce seedlings evolved ethylene at a rate of 0.7 nl 10 seeds–1h–1. The growth of lettuce roots was stimulated by 2-aminoethoxyvinyl-glycine(AVG), an inhibitor of ethylene synthesis, and 2,5-norbornadiene(NBD), an inhibitor of ethylene action, as well as by CSI. Incontrast to ethylene, treatments with CSI, AVG and NBD promotedlongitudinal elongation of cortical cells of roots and inhibitedtheir lateral expansion. Application of CSI slightly reducedethylene production from lettuce, but this reduction was notsufficient to account for the CSI-in-duced stimulation of growth.The maximal promotive effects of AVG and NBD were obtained at3 µM and 150 µl liter–1, respectively. Thegrowth promotion by CSI disappeared in the presence of the optimumlevels of AVG or NBD; a further addition of ethylene causedthe stimulatory effects of CSI to increase, depending on theconcentration of ethylene. Thus, CSI reduced both the sensitivityof the roots to ethylene and the maximal effects of ethylene.The CSI-induced stimulation of growth was ascribed to the reductionof the response to ethylene in the lettuce roots. Endogenous cytokinin and gibberellin-like activity were measuredby bioassay in developing fruit of the orchid Epidendrum ibaguense.Cytokinins decline during the first 30 d after pollination,then begin to accumulate, with very high levels (1–13µ g zeatin eq. g–1 dry wt. ) in the mature fruitand seed. The major structural change in developing fruit duringthe first 30 d is the ongoing cell division in the fruit wall.By day 30 most ovules have been fertilized and embryo developmentbegins. The increase in cytokinin activity thus coincides withthe onset of embryo development. Gibberellin levels declinein the fruit throughout development, although high activity(0.9 µ g GA3 eq. g–1 dry wt. ) is observed in themature seed. The mature embryo shows no obvious structural differentiationinto embryonic axis and cotyledon and no endosperm develops.  相似文献

In heterotrophic cell suspensions of sunflower (Helianthus annuusL. cv. Spanners Allzweck) the effect of Pmg elicitor, a fungalelicitor preparation from Phytophthora megasperma f. sp. glycinea,on the induction of chitinase and ß-1,3-glucanaseactivity was studied in relation to changes in ethylene biosynthesis.Dose-response experiments with Pmg elicitor showed that theonset of the induction of intracellular chitinase and ß-1,3-glucanaseactivity coincided or followed a transient rise in ethyleneand particularly endogenous 1-aminocyclopropane-1-carboxylicacid (ACC) levels within 5 h of application. Treatment with5 µg ml–1 elicitor stimulated ethylene and ACC levels1.6-fold and 4-fold, relative to control, respectively. Themolar ratio of ACC to ethylene changed from approximately 3:1in controls to 9:1 in treated cells. During further incubation,ethylene formation and, to a lesser degree, ACC levels declinedand the ACC/ethylene ratio increased to 56:1 in elicitor-treatedcells. On a protein basis, the activities of ß-1,3-glucanaseand chitinase increased approximately 5-fold and 8-fold, respectively,48 h after elicitor application. Additional treatment with theACC synthesis inhibitor aminoethoxyvinyiglycine (AVG) decreasedelicitor-induced enzyme activities and the levels of both ethyleneand ACC. Elicitor effects on chitinase and ß-1,3-glucanaseactivities could be fully restored when ACC was additionallyapplied. Concomitantly, the ACC/ ethylene ratio increased. Neithertreatments with ACC alone, which simultaneously increased internalACC and ethylene levels, nor treatments with AVG alone, whichsimultaneously reduced ACC and ethylene levels, could generallystimulate chitinase or ß-1,3-glucanase activitiesin the cells. It is suggested that ACC functions as a promotingfactor in the induction of chitinase and ß-1,3-glucanaseactivity triggered by Pmg elicitor and appears to reverse aninhibiting influence of ethylene. Differences in premature leaf abscission and in visible steminjury in genetic lines of poplar followed continuous fumigationswith air pollutant levels of SO2 (90–100 nl l–1)and O3 (70–80 nl l–1) either separately or together.The clones used were: Populus deltoides var. missiouriensisMarsh., P. nigra cv. ‘italicd’ L., and the hybridsP. nigra cv. ‘italica’ xP. deltoides (He-X/3) andP. nigra cv.‘italica’ x P. nigra cv. ‘Serres’(He-K/7). While most leaf abscission occurred within 20 d fromthe start of fumigation, stem lesions (intumescences), appearedonly after 72 d. Their anatomical characteristics include theformation of lysigenous aerenchyma in the lower parts of theintumescence, the sloughing of superficial cells from the injuredarea, and the development of crystalline formations on the surfaceof the lesions. P. deltoides exhibited the least morphologicalresponse to the gases. Ethylene released from fumigated leaves was determined at thesame gas concentration of SO2 (100 nl l–1), O3 (75 nll–1) and their mixture. Leaves of P. deltoides consistentlyshowed the lowest ethylene production after the gas treatments.P. ‘italica’ production was higher but was littlealtered by the treatments. The two hybrids He-X/3 and He-K/7showed the greatest increases in ethylene evolution with time.With He-K/7 exposed to the gas mixture the production of ethylenedecreased after the initial sharp rise during days 1–2,and reflected the considerable leaf damage observed after day3. The results suggest that sensitivity to air pollution, (as shownby leaf abscission and the formation of stem intumescences)can be correlated with the level of pollutant-induced ethyleneevolution from leaves. Initially high levels could induce abscission,whilst prolonged production could be responsible for intumescenceinitiation. The discussion proposes a series of events fromSO2 and/or O3 entry into the leaf and the physiological reasonsfor the clonal differences. The anatomy of abscissed avocado fruitlets of the Hass varietywas compared with that of actively growing control fruitletsto investigate the reason for abscission. Control fruitletshad a small embryo (4–14 cells) and numerous endospermcells by seven days after pollination. Cotyledon developmenthad commenced by 21 days and a root/shoot axis was present at28 days. Over 90 per cent of the fruitlets which abscissed during thefirst week after the end of flowering were unfertilized and18 per cent were abnormal. By the fourth week after the endof flowering all abscissed fruitlets were fertilized and noneabnormal. Abnormalities included ovaries with more than oneembryosac or ovule, with an immature embryo-sac, with an ovulein an abnormal position or with a deficiency in ovule structure.In a few fertilized abscissed fruitlets either the embryo orendosperm had ceased development first. In the majority of casesembryo and endosperm were anatomically normal. The majorityof fertilized abscissed fruitlets had reached a stage correspondingto 14 days after pollination of the control tree. Degenerationwas often observed in both unfertilized and fertilized abscissedfruitlets. It is suggested that this degeneration occurred inthe period between the cessation of development and abscissionof the fruit. This period was approximately one week. The proportion of abnormal ovaries was too low to significantlyaffect yields, and sufficient flowers had been fertilized togive an adequate crop. 1-Aminocyclopropane-1-carboxylic acid (ACC) synthase activityincreased rapidly after wounding of mesocarp tissue of wintersquash fruit (Cucurbita maxima Duch.) and reached a peak at16 h after excision and then declined sharply. The rise in ACCsynthase activity was followed by increases in the endogenousACC content and the rate of ethylene production. The activityof ethylene forming enzyme (EFE) also increased rapidly in theexcised discs of mesocarp of winter squash fruit. ACC synthase activity was strongly inhibited by aminoethoxyvinylglycinewith a Ki value of 2.1 µM. Michaelis-Menten constant ofACC synthase for S-adenosylmethionine was 13.3 µM. Ethylene suppressed the induction of ACC synthase in the woundedmesocarp tissue. The suppression by ethylene increased withthe increasing concentrations of applied ethylene and the maximumeffect was obtained at about 100 µl 1–1 ethylene,at which point the induction was suppressed by 54%. Modifications to the gaseous envelope by callus during culturein Petri dishes were shown to reduce growth and promote necrosisof several rice (Oryza sativa L.) cultivars. Incubatingcallusunder a continuous flow of gas mixtures of known compositionsuggested that the inhibition of growth was caused by the accumulationof ethylene, the depletion of oxygen and, to a lesser extent,the accumulation of carbon dioxide. In order to evaluate theimportance of ethylene accumulation aminoethoxyvinylglycine(AVG), 1-aminocyclopropane-l-carboxylic acid (ACC and silvernitrate (AgNO3), were added to the nutrient medium and ethylenemeasurements performed during callus culture. Ethylene restrictedcallus growth particularly under high (35 °C) as comparedto moderate (25 °C) temperatures and under illuminated ascompared to darkened incubation. Under illuminated incubationat 25 °C AVG (5 mmol m–3) and AgNO°(50 mmol m–3)significantly improvedcallus growth (100 and 60% respectively)while ACC (200 mmol m–3) significantly decreased growth(40%). AVG and AgNO3 were less effective under dark incubationat 25 °C where ethylene production was lower. Furthermore,callus growth was significantly better in large as comparedto small culture vessels since the ethylene concentration wasdiluted and more oxygen was available for respiration. Bettercontrol of ethylene and increased oxygen availability couldbe a way ofproducing healthy callus for the formation of embryogenictissues of otherwise recalcitrant cultivars of rice (e.g. IndicaIR42) and may be a way of improving manipulation of other cerealspecies. Plantlets were produced by induction of nucellar embryony (apomixis)in isolated unfertilized ovules of a non-apomictic plant—thegrapevine (Vitis vinifera L. cv. Cabernet-Sauvignon). Ovulesgrown in liquid culture with benzyladenine (5–10 µM)and ß-naphthoxyacetic acid (5–25 µM) formeda nucellar callus which gave rise to somatic embryos. For growthof plantlets the embryos were transferred to an agar mediumcontaining gibberellic acid (1 µM) and 2-isopentenyladenine(5 µM). A cDNA clone (TAB7) encoding a putative woundinduced (Win) proteinhas been isolated from a tomato (Lycopersicon esculentum Mill.cv. Ailsa Craig) leaf abscission zone cDNA library using a differentialscreening strategy. The clone has a high degree of homologyat the amino acid level to both the potato win1 and 2 genes,Hevea brasiliensis hevein and Nicotiana tabacum PR-4a and PR-4bproteins. The mRNA encoded by TAB7 is up-regulated within 12h of exposure to ethylene (10µl l–1) and its expressionincreases steadily within the cells comprising the leaf abscissionzone and to a lesser extent in the adjacent non-zone tissue.This rise precedes the onset of cell separation. Southern analysisindicates that the mRNA is encoded by either a single gene ora small gene family. The role of the protein during abscissionis discussed. Changes in carbon fixation rate and the levels of photosyntheticproteins were measured in fourth leaves of Lolium temulentumgrown until full expansion at 360 µmol quanta m–2s–1 and subsequently at the same irradiance or shadedto 90 µmol m–2 s–1. Ribulose-1,5-bisphosphatecarboxylase/oxygenase (Rubisco), light-harvesting chlorophylla/b protein of photosystem II (LHCII), 65 kDa protein of photosystemI (PSI), cytochrome f (Cytf) and coupling factor 1 (CF1) declinedsteadily in amount throughout senescence in unshaded leaves.In shaded leaves, however, the decrease in LHCII and the 65kDa protein was delayed until later in senescence whereas theamount of Cyt f protein decreased rapidly following transferto shade and was lower than that of unshaded leaves at the earlyand middle stages of senescence. Decreases in the Rubisco andCF1 of shaded leaves occurred at slightly reduced rates comparedwith unshaded leaves. These results indicate that chloroplastproteins in fully-expanded leaves are controlled individually,in a direction appropriate to acclimate photosynthesis to agiven irradiance during senescence.
