为建立基于绿色荧光蛋白(GFP)的药物筛选模型,并用此模型从包括中药提取物在内的化合物中筛选新型蛋白酶体抑制剂,本研究构建了pGC-E1-ZU1-GFP融合蛋白慢病毒表达载体并感染A549细胞,筛选稳定表达细胞株,用已知蛋白酶体抑制剂PS-341处理细胞,荧光显微镜检测处理前后细胞GFP水平变化。结果获得了稳定表达pGC-E1-ZU1-GFP的A549细胞,这些细胞用PS-341处理24h后用荧光显微镜检测,发现细胞绿色荧光强度相对于对照组明显增强。利用这一模型对一些化合物进行筛查,发现了一些新的蛋白酶体抑制剂。  相似文献

探讨蛋白酶体抑制剂MG132 在诱导人白血病K562细胞凋亡过程中作用.分别以不同浓度的蛋白酶体抑制剂MG132 处理人白血病细胞K562,通过MTT法检测K562细胞活力,应用Annexin Ⅴ和PI 双染的细胞流式法检测K562细胞凋亡率和细胞内活性氧(ROS) 水平,应用酶标仪法检测K562细胞内Caspase- 3活性变化的情况.结果表明,随着MG132浓度的增加,各个指标与对照组比较差异均有显著性(P<0.05):K562细胞增殖明显受到抑制;细胞凋亡率明显增加,且当MG132浓度为900 nmol/L时,细胞凋亡率达36.5 %;同时,ROS 水平和caspase- 3活性明显升高.因次,蛋白酶体抑制剂MG132可显著抑制人白血病细胞K562增殖并促进其凋亡.  相似文献

目的建立一种动态检测活细胞内泛素-蛋白酶体系统活性的方法。方法将表达绿色荧光蛋白(GFP)或红色荧光蛋白(DsRed2)的质粒分别改建为表达带有内泛素-蛋白酶体系统降解信号CL1的GFP或DsRed2的pGFP^u或pDsRed2质粒,然后转染HEK293细胞,通过G418筛选得到稳定表达GFP^u或DsRed2^u的细胞系。在蛋白酶体抑制N—Acetyl—Leu-Leu—Norleu—al(ALLN)处理GFP^u或DsRed2^u细胞后,应用免疫印记技术检测细胞内GFP或DsRed,含量的变化,应用荧光显微镜和激光扫描共聚焦显微镜技术观察GFP或DsRed,荧光强度的变化。结果ALLN处理能使GFP“和DsRed2^u细胞内GFP和DsRed。含量明显增加,荧光强度显著增强,并呈现明显的剂量/时间-效应关系。结论本文成功地建立了检测内泛素-蛋白酶体系统活性的方法,该方法能有效地对活细胞的内泛素-蛋白酶体系统活性进行实时动态检测。  相似文献

【目的】水通道蛋白(aquaporin,AQP)是一种广泛存在于细胞生物膜系统中的跨膜转运蛋白,它在水分渗透压平衡过程中发挥着重要的作用。本研究旨在前期克隆茶尺蠖Ectropis obliqua Prout水通道蛋白AQP1(EoAQP1)基因全长的基础上,进一步明确该蛋白的细胞器定位,并探析其对细胞形态和增殖的影响。【方法】采用双酶切法构建了绿色荧光蛋白(green fluorescent protein,GFP)载体以及绿色荧光蛋白与EoAQP1的真核融合(EoAQP1-GFP)表达载体。通过荧光显微镜和激光共聚焦显微镜观测了GFP与EoAQP1-GFP蛋白在黑腹果蝇Drosophila melanogaster胚胎细胞系(S2)中的表达特征。利用溶酶体和高尔基体红色荧光探针明确了EoAQP1蛋白在S2细胞内的细胞器定位。通过流式细胞仪和酶标仪检测了EoAQP1蛋白对S2细胞系细胞的颗粒度、大小和增殖的影响。【结果】成功构建了GFP和EoAQP1-GFP的真核表达载体,将其分别命名为pAc5.1-GFP和pAc5.1-EoAQP1-GFP。荧光显微镜和激光共聚焦显微镜观测结果表明,荧光蛋白GFP均匀填充于S2细胞内稳定表达。融合蛋白EoAQP1-GFP存在两种表达形式:一种以类似于溶酶体的球形和橄球形结构,围绕细胞核在细胞质中散点表达;另一种以类似于高尔基体的半球状和弓形结构表达于细胞质中。溶酶体和高尔基体红色荧光探针检测结果表明,EoAQP1蛋白在溶酶体内没有表达,但与高尔基体完全重叠。流式细胞仪和酶标仪检测结果表明,过表达EoAQP1蛋白可使S2细胞膨胀增大,细胞颗粒度增强,但对细胞增殖无明显影响。【结论】EoAQP1定位于高尔基体中行使功能,该蛋白能改变细胞形态,但不能促进细胞的生长。  相似文献

为了探讨长裂苦苣菜水提取物对肺癌细胞A549的影响,本研究用不同浓度的长裂苦苣菜水提取物作用于体外培养的A549细胞,从细胞形态(显微镜观察)、细胞增殖活力(CCK-8法)、细胞凋亡(Annexin V-FITC/PI染色)、线粒体膜电位(JC-1染色)和细胞内活性氧水平(荧光探针DCFH-DA染色)几个方面检测长裂苦苣菜水提取物对体外培养的A549细胞的影响。当处理浓度达到1 mg/mL作用细胞48 h后,细胞出现明显皱缩的凋亡形态学特征,与对照组相比处理组细胞增殖活力明显下降(P0.05)。进一步用流式细胞仪检测发现≥1mg/mL的处理浓度作用细胞48 h后细胞凋亡率显著提高(P0.05),同时检测到处理组细胞的膜电位都明显下降,细胞内活性氧水平明显上升,都呈剂量依赖关系。这些结果表明长裂苦苣菜水提取物可以诱导A549细胞凋亡,并抑制其生长和增殖,是一种潜在的预防和抑制肿瘤生长的药食两用植物。  相似文献

目的探讨维甲酸对A549细胞增殖和凋亡及相关基因表达的影响。方法MTT法观察ATRA对A549细胞增殖的抑制作用;流式细胞仪、AO/EB荧光双染法检测细胞凋亡;免疫细胞化学检测ATRA处理前后A549细胞Skp2、p27^kip1蛋白表达的情况。结果ATRA处理后①MTT法结果显示ATRA对A549细胞具有增殖抑制作用,在一定范围内呈时间-剂量依赖性。②AO/EB荧光双染色法观察到ATRA 25μmol/L作用A549细胞48h后,即可发现典型的凋亡形态学改变。③流式细胞仪结果出现凋亡峰,与对照组细胞相比,实验组细胞周期延长,主要表现为G0/G1期细胞比例增加,同时S期细胞比例减少。④免疫细胞化学结果显示,ATRA 25μmol/L处理细胞48h后,维甲酸处理组Skp2有明显下调,p27^kip1则明显上调。结论ATRA具有抑制肺腺癌A549细胞增殖,诱导细胞凋亡的作用,其机制可能与下调Skp2,上调p27^kip1蛋白的表达水平有关。  相似文献

目的:探讨组蛋白去乙酰化酶抑制剂曲古霉素A(trichostatin A,TSA)对人非小细胞肺癌(NSCLC)A549细胞增殖抑制作用及机制.方法:以不同剂量TSA(0.1μM,0.5pM和1μM)处理A549细胞.MTT法检测细胞增殖情况,碘化丙啶(PI)染色结合流式细胞仪检测细胞周期,Westem blot法检测P21蛋白表达,流式细胞仪检测细胞线粒体膜电位和细胞凋亡.结果:TSA剂量依赖性抑制肺癌A549细胞增殖,表现为细胞周期阻滞于G2/M期,同时P21蛋白表达增高;此外,TSA还可以剂量依赖性的促进A549细胞凋亡,伴有线粒体膜电位下降.结论:TSA促进NSCLCA549细胞周期阻滞和凋亡,从而抑制其增殖.  相似文献

该研究旨在探讨柯里拉京对人肺癌A549细胞凋亡的影响及其潜在作用机制。采用CCK-8细胞活性检测试剂盒检测柯里拉京对A549细胞活性的影响;通过流式细胞术检测细胞凋亡;JC-1线粒体膜电位检测试剂盒检测线粒体膜电位;免疫印迹法检测凋亡相关蛋白(bax、bcl-2、cleaved-caspase-3、cleaved-PARP)的表达量;通过DCFH-DA探针标记检测细胞内ROS水平。研究结果显示,柯里拉京处理能够剂量依赖性地抑制A549细胞的活性,并通过上调bax的表达、下调bcl-2的表达,破坏线粒体膜电位,促进有活性的cleaved-caspase-3以及cleaved-PARP的形成,诱导A549细胞凋亡。活性氧清除剂NAC能够明显逆转柯里拉京诱导的细胞凋亡。因此,柯里拉京可能通过调节胞内ROS水平诱导人肺癌细胞A549发生凋亡。  相似文献

目的:探讨青蒿素诱导人白血病细胞K562凋亡的线粒体机制.方法:用青蒿素处理K562细胞.通过MTT比色法检别细胞增殖抑制的效果;荧光显微镜观察细胞的凋亡;流式细胞术(flow cytometry,FCM)进行细胞周期分析;Western-blotting测定药物作用前后线粒体、细胞浆细胞色素C的表达.结果:青蒿素抑制K562细胞的增殖,IC5D为1.5× 10-5mol·L-1;Hoechst33342/PI双荧光染色可观察到明显的核浓缩、凝集等细胞凋亡表现;流式细胞仪检测G2期细胞比例增高,S期减少;Western-blotting检测药物处理细胞后线粒体细胞色素C表达水平下调,细胞浆出现明显细胞色素C蛋白条带.结论:青蒿素可能通过线粒体细胞色素C途径诱导K562细胞凋亡.  相似文献

本研究的目的是观察奥利司他(Orlistat)逆转肺腺癌耐顺铂细胞株A549/DDP的耐药作用并考察其可能的分子机制。应用CCK-8法检测并计算肺腺癌耐药细胞株A549/DDP对顺铂(cisplatin, DDP)的耐药指数(resistance index, RI),筛选奥利司他的最佳实验浓度并观察其对A549/DDP细胞的耐药逆转效果;倒置荧光显微镜下观察各实验组细胞的形态学变化及Hoechst 33258荧光染色后细胞凋亡形态学改变;采用流式细胞术检测不同药物处理对细胞凋亡率的影响;Western blotting检测各实验组细胞P-gp、FASN、PI3K、Akt、p-Akt、NF-κB、Bcl-2和cleved-caspase-3的表达情况。结果显示,奥利司他抑制了A549/DDP耐药细胞株的增殖,且具有浓度依赖性。奥利司他与DDP联用增加了耐药株A549/DDP细胞对DDP的敏感性,耐药逆转倍数为5.02。倒置荧光显微镜观察到联合用药组细胞出现了明显的凋亡形态学改变。流式细胞术结果表明,与对照组和单药组比较,两种药物联用后细胞的凋亡率显著提高(p0.05)。Western blotting检测结果显示,相较于其它3组,联合用药组中耐药相关蛋白P-gp表达下调(p0.01),PI3K、p-Akt、NF-κB表达水平均显著降低(p0.01),抗凋亡蛋白Bcl-2表达下降(p0.01),凋亡相关蛋白cleved-caspase-3表达上调(p0.01);虽然FASN的表达在单用奥利司他组及联合用药组中均降低,但这两组间的FASN表达水平并无统计学差异。以上结果表明,奥利司他能够提高A549/DDP耐药细胞株对化疗药物DDP的敏感性,具有逆转肺腺癌细胞耐药性的作用,其机制与降低耐药蛋白P-gp的表达、抑制PI3K/Akt/NF-κB信号通路以及其下游凋亡相关蛋白有关。  相似文献

Heme-mediated regulation, presented in many biological processes, is achieved in part with proteins containing heme regulatory motif. In this study, we demonstrate that FLAG-tagged PpsR isolated from Rhodobacter sphaeroides cells contains bound heme. In vitro heme binding studies with tagless apo-PpsR show that PpsR binds heme at a near one-to-one ratio with a micromolar binding constant. Mutational and spectral assays suggest that both the second Per-Arnt-Sim (PAS) and DNA binding domains of PpsR are involved in the heme binding. Furthermore, we show that heme changes the DNA binding patterns of PpsR and induces different responses of photosystem genes expression. Thus, PpsR functions as both a redox and heme sensor to coordinate the amount of heme, bacteriochlorophyll, and photosystem apoprotein synthesis thereby providing fine tune control to avoid excess free tetrapyrrole accumulation.  相似文献

To know the status of sparganum (plerocercoid of Spirometra erinacei) infection in the Korean wild life, several species of wild animals were captured in Gangwon-do and examined for their status of infection with spargana. From February to December 2011, a total of 62 wild boars, 5 badgers, 1 weasel, 1 Siberian chipmunk, and 53 wild rodents were captured, and their whole muscles were examined with naked eyes for the presence of spargana worms. From the weasel and 1 wild boar, a total of 5 spargana specimens were extracted. The weasel was for the first time recorded as an intermediate or paratenic/transport host of S. erinacei in Korea, and both the weasel (Mustela sibirica manchurica) and wild boar (Sus scrofa) were added to the list of wild animals carrying spargana.  相似文献

Since 1960, landings of Atlantic herring have been the greatest of any marine species in Canada, surpassing Atlantic cod and accounting for 24% of the total seafood harvested in Atlantic Canada. The Scotian Shelf‐Bay of Fundy herring fisheries (NAFO Division 4VWX) is among Canada''s oldest and drives this productivity, accounting for up to 75% of the total herring catch in some years. The stocks’ productivity and overall health have declined since 1965. Despite management measures to promote recovery implemented since 2003, biomass remains low and is declining. The factors that drive the productivity of 4VWX herring are primarily unresolved, likely impeding the effectiveness of management actions on this stock. We evaluated potential drivers of herring variability by analyzing 52 time‐series that describe the temporal and spatial evolution of the 4VWX herring population and the physical, ecological, and anthropogenic factors that could affect them using structural equation models. Variation in herring biomass was best accounted for by the exploitation rate''s negative effect and the geographic distribution of fishing and recruitment. Thermal phenology and temperature adversely and egg predation positively impacted the early life stage mortality rate and, ultimately, adult biomass. These findings are broadly relevant to fisheries management, but particularly for 4VWX herring, where the current management approach does not consider their early life stage dynamics or assess them within the ecosystem or climate change contexts.  相似文献

报道了产自云南麻栗坡的兰科兰属一新杂种——麻栗坡长叶兰(Cymbidium×malipoense L.J.Chen,H.B.Zhou&Z.J.Liu),对该种的形态特征进行了描述。该种可能是长叶兰(Cymbidium erythraeum Lindl.)与龙州兰(C.eburneum var.longzhouense Z.J.Liu et S.C.Chen)的天然杂交种。本种与长叶兰的区别是:本种的花葶自叶腋发出;萼片和花瓣浅黄绿色,具浅紫红色的纵条纹和斑点;唇盘具黄色褶片;蕊柱浅紫红色;中裂片卵状肾形。长叶兰的萼片和花瓣绿色,有红褐色脉纹和不规则斑点;中裂片心形到剑形;唇盘密被毛。本种与龙州兰的区别是:本种萼片和花瓣浅黄绿色,具浅紫红色的纵条纹和斑点;侧裂片有紫红色条纹;蕊柱浅紫红色。龙州兰萼片和花瓣白色,外表面略带粉红色;唇瓣白色,侧裂片和中裂片具有紫红色斑点。  相似文献

Heparan sulfate (HS) proteoglycans play critical roles in a wide variety of biological processes such as growth factor signaling, cell adhesion, wound healing, and tumor metastasis. Functionally important interactions between HS and a variety of proteins depend on specific structural features within the HS chains. The fruit fly (Drosophila melanogaster) is frequently applied as a model organism to study HS function in development. Previous structural studies of Drosophila HS have been restricted to disaccharide composition, without regard to the arrangement of saccharide domains typically found in vertebrate HS. Here, we biochemically characterized Drosophila HS by selective depolymerization with nitrous acid. Analysis of the generated saccharide products revealed a novel HS design, involving a peripheral, extended, presumably single, N-sulfated domain linked to an N-acetylated sequence contiguous with the linkage to core protein. The N-sulfated domain may be envisaged as a heparin structure of unusually low O-sulfate content.  相似文献

A stilbene fluorescent brightener, Tinopal LPW, was used as an ultraviolet (UV) protectant for the entomopathogenic nematode Steinernema carpocapsae (All strain). Irradiation of an aqueous suspension of nematodes produced a LC₅₀ in 15.7 minutes under a sunlamp and in 31.7 minutes in direct sunlight. Irradiation by both sunlamp and sunlight of a suspension of nematodes in Tinopal LPW did not reduce their biological activity as measured by their ability to parasitize wax moth larvae after exposure of 8 hours and 4 hours, respectively. Tinopal LPW appeared promising as a radiation protectant.  相似文献

The structure-based design, synthesis, and screening of a glucuronic acid scaffold library of affinity ligands directed toward the catalytic cleft on porcine pancreas alpha-amylase are presented. The design was based on the simulated docking to the enzyme active site of 53 aryl glycosides from the Available Chemicals Directory (ACD) selected by in silico screening. Twenty-three compounds were selected for synthesis and screened in solution for binding toward alpha-amylase using nuclear magnetic resonance techniques. The designed molecules include a handle outside of the binding site to allow their attachment to various surfaces with minimal loss of binding activity. After initial screening in solution, one affinity ligand was selected, immobilized to Sepharose (Amersham Biosciences), and evaluated as a chromatographic probe. A column packed with ligand-coupled Sepharose specifically retained the enzyme, which could be eluted by a known inhibitor.  相似文献

Two species of melitid amphipod were collected from the Gulf of Thailand. Dulichiella pattaniensis is new to science, and Melitalati latiflagella Ren & Andres, 2012 has not been previously reported from Thai Waters. Dulichiella pattaniensis is characterized by male gnathopod 2 distolateral crown with 4 spines; pleonite/urosomite formula 7-7-7-5-6-2; pereopod 5-7 dactylus with 2 accessory spines. This combination of characters has not been recorded previously in the Dulichiella. The characters of the specimens are described and illustrated. All specimens are deposited in the Princess Maha Chakri Sirindhorn Natural History Museum, Prince of Songkla University, Thailand.  相似文献
