Chloroplast DNA sequencing and genomic in situ hybridization(GISH) were used to investigate the genomic origin and organizationof the alpine grass Poa jemtlandica. Using genomic probes ofP. alpina and P. flexuosa, GISH clearly distinguished betweenthese two putative parental genomes and thus confirmed the hybridnature of P. jemtlandica. The chloroplast trn L intron and trnL–trn F intergenic spacer (IGS) sequence genotypes ofP. flexuosa and P. jemtlandica were 100% identical but differedfrom those of P. alpina by a total of ten or 11 nucleotide substitutionsand six indels over 866 aligned positions, identifying P. flexuosaas the maternal parent of the P. jemtlandica population studiedhere and supporting a relatively recent origin of the hybrid.GISH revealed the presence of intergenomic translocations inthe hybrid genome, indicating that the two parental genomeshave undergone some rearrangements following hybridization.It is likely that some of these chromosome changes took placesoon after hybridization in order to overcome the adverse interactionsbetween the nuclear and the cytoplasmic genomes and to facilitatethe successful establishment of the newly formed hybrid. Thepresence of intergenomic chromosome changes may play an importantrole in the evolution of natural hybrids and the establishmentof new evolutionary lineages. In Situ Localization of Parental Genomes in a Wide Hybrid   总被引:24,自引:0,他引:24  
In situ hybridization enabled DNA originating from the two parentalgenomes to be distinguished in plant hybrids. A probe of biotinylatedtotal genomic DNA from Secale africanum labelled the chromosomesof S. africanum origin but not those from Hordeum chilense inroot-tip chromosome spreads of the sexual hybrid between thetwo species. Hybridization of total genomic DNA from S. africanumto DNA on filters (dot blots) confirmed the distinction betweenDNA from Hordeum and Secale. The total genomic probe hybridizedto the whole length of the chromosomes from S. africanum remarkablyuniformly, labelling both euchromatin and heterochromatin, exceptat the centromeric region. The probe binding was visualizedas a yellow colour by the fluorescein-coupled detection systemwhich contrasted with the red fluorescing counterstain of theunlabelled chromatin. The chromosomes originating from bothparents could be seen and distinguished as red and yellow fluorescenceat all stages of the cell cycle. At interphase and prophase,the chromatin originating from the two parental genomes didnot mix. Chromosomes or groups of chromosomes occupied distinctdomains and also tended to be arranged in a Rabl configurationwith the centromeres clustered at one end of the nucleus. 基因组原位杂交(GISH)技术可以鉴定植物多倍体物种起源、杂种亲本染色体来源和组成,分析栽培种与其近缘野生种的亲缘关系,研究减数分裂染色体行为等。基因组原位杂交包括多色基因组原位杂交、比较基因组原位杂交和自身基因组原位杂交等。基因组原位杂交技术的关键步骤是染色体制片、探针制备及长度优化、探针与封阻的浓度比例和杂交后洗脱强度。该文对近年来国内外有关基因组原位杂交技术的发展及其在园艺植物基因组研究中的应用现状进行了综述,并指出随着多种园艺植物全基因组的测定,未来应从基因组信息中寻找更多的染色体特异性标记,结合荧光显带及荧光原位杂交技术,为深入研究园艺植物的起源以及遗传关系鉴定等提供技术支持。  相似文献

Background and Aims: Earlier studies have suggested that the tetraploid Primula egaliksensis(2n = 40) originated from hybridization between the diploidsP. mistassinica (2n = 18) and P. nutans (2n = 22), which werehypothesized to be the maternal and paternal parent, respectively.The present paper is aimed at verifying the hybrid nature ofP. egaliksensis using cytogenetic tools, and to investigatethe extent to which the parental genomes have undergone genomicreorganization. Methods: Genomic in situ hybridization (GISH) and fluorescent in situhybridization (FISH) with ribosomal DNA (rDNA) probes, togetherwith sequencing of the internal transcribed spacer (ITS) regionof the rDNA, were used to identify the origin of P. egaliksensisand to explore its genomic organization, particularly at rDNAloci. Key Results: GISH showed that P. egaliksensis inherited all chromosomes fromP. mistassinica and P. nutans and did not reveal major intergenomicrearrangements between the parental genomes (e.g. interchromosomaltranslocations). However, karyological comparisons and FISHexperiments suggested small-scale rearrangements, particularlyat rDNA sites. Primula egaliksensis lacked the ITS-bearing heterochromaticknobs characteristic of the maternal parent P. mistassinicaand maintained only the rDNA loci of P. nutans. These resultscorroborated sequence data indicating that most ITS sequencesof P. egaliksensis were of the paternal repeat type. Conclusions: The lack of major rearrangements may be a consequence of theconsiderable genetic divergence between the putative parents,while the rapid elimination of the ITS repeats from the maternalprogenitor may be explained by the subterminal location of ITSloci or a potential role of nucleolar dominance in chromosomestabilization. These small-scale rearrangements may be indicativeof genome diploidization, but further investigations are neededto confirm this assumption.  相似文献

The origin of the two common cultivars of Crocus, C. 'Stellaris'(2n = 2x = 10) and C. 'Golden Yellow' (2n = 3x = 14) was investigatedby fluorescent in situ hybridization using both total genomicDNA and cloned DNA sequences as probes. The clear differentiationbetween the chromosomes after genomic in situ hybridizationsupports the proposals of a hybrid origin of the cultivars andshows that they have the same parental genomes originating fromC. flavus (2n = 8) and C. angustifolius (2n = 12). C. 'Stellaris'has four chromosomes of C. flavus origin and six chromosomesof C. angustifolius origin. C. 'Golden Yellow' has eight chromosomesof C. flavus origin and six chromosomes of C. angustifoliusorigin. The number and location of 18S-5·8S-26S rRNAgenes on the chromosomes of the hybrids and of the parentalspecies agree with the results from the genomic probings. Karyological and genomic in situ hybridization (GISH) approaches provided evidence of the parentage and origin of the hybrid species Narcissus obsoletus. Here, we demonstrate that the putative parental species, N. serotinus L. and N. elegans (Haw.) Spach, recently proposed because of their intermediate morphological traits, have participated in the hybridization process forming this taxon. Karyotype characterization of parental genomes in populations from S Spain and N Morocco has revealed differences in chromosome length and karyotype asymmetry, highlighting their diploid nature. Multicolour GISH on metaphase plates of N. obsoletus, with N. serotinus and N. elegans DNA used as probes, showed differential fluorescent staining of 10 and 20 chromosomes from parental genomes, respectively. Both parental genomes were detected in the allopolyploid, albeit in a duplicated manner. Secondary hybridization between N. obsoletus and N. serotinus was also detected karyologically. Allium fistulosum harbours a number of desirable agronomical traits for the breeding of onions. However exploitation of A. fistulosum for onion breeding via direct sexual hybridization is problematic. Therefore, we examined if a bridge cross, using A. roylei as a bridging species, might provide an alternative. By means of genomic in situ hybridization (GISH) we showed that each of the three parental genomes can be distinguished from the others in interspecific hybrids, suggesting that these genomes contain sufficiently different repetitive DNA families. We succeeded in carrying out multi-colour GISH to metaphase spreads of a first-generation bridge-cross individual [A. cepa× (A. fistulosum×A. roylei], which is composed of three parental genomes. Recombination between the genomes of A. fistulosum and A. roylei took place to a large extent: 7 recombined chromosomes were observed, and it could be shown that the proximal regions of the recombined A. fistulosum/A. roylei chromosomes belonged to the former, whereas the distal parts belonged to the latter. The high percentage of bound bivalent arms in metaphase I of pollen mother cells of a fertile bridge-cross individual suggests the introgression of A. fistulosum genes, mediated by A. roylei, into the genome of A. cepa. However, the presence of univalents reflects decreased pairing and recombination between the three genomes. Pollen fertility and pollen-tube growth of the first-generation bridge-cross individual seem to be sufficient to produce a second generation bridge-cross (A. cepa×first-generation bridge cross) progeny. Somatic hybrids between the wild incongruent species Solanum bulbocastanum (2n = 2x = 24) and S. tuberosum haploids (2n = 2x = 24) have been characterized for their nuclear and cytoplasmic genome composition. Cytologic observations revealed the recovery of 8 (near-)tetraploid and 3 hexaploid somatic hybrids. Multicolor genomic in situ hybridization (GISH) analysis was carried out to study the genomic dosage of the parental species in 5 somatic hybrids with different ploidy. The GISH procedure used was effective in discriminating parental genomes in the hybrids; most chromosomes were unambiguously colored. Two (near-)tetraploid somatic hybrids showed the expected 2:2 cultivated-to-wild genomic dosage; 2 hexaploids revealed a 4:2 cultivated-to-wild genomic dosage, and 1 hexaploid had a 2:4 cultivated-to-wild genomic dosage. Characterization of hybrid cytoplasmic genomes was performed using gene-specific primers that detected polymorphisms between the fusion parents in the intergenic regions. The analysis showed that most of the somatic hybrids inherited the plastidial and mitochondrial DNA of the cultivated parent. A few hybrids, with a rearranged mitochondrial genome (showing fragments derived from both parents), were also identified. These results confirmed the potential of somatic hybridization in producing new variability for genetic studies and breeding.  相似文献

The genusAvena L. (Poaceae) consists of diploid, tetraploid,and hexaploid species, with the B genome known only in tetraploidspecies and the D genome in the hexaploid species. DNA:DNAinsitu hybridization, using total genomic DNA from diploidAvenastrigosa Schreb. (Asgenome) as a probe, labelled all 28 chromosomesof the AB tetraploidAvena vaviloviana (Malz.) Mordv. stronglyand uniformly, revealing the close relationship between thesetwo genomes. Comparison of patterns of size-separated DNA restrictionfragments between the diploidA. strigosa and the tetraploidA.vaviloviana , using 32 different restriction enzymes, revealedno differences. Southern hybridization using total AB genomicDNA as a probe also gave no differences in banding patternsbetween the two genomes, even when a large excess of A genomicDNA was used as a block. From anA. vaviloviana genomic library,1800 colonies were blotted and probed sequentially with A andAB genomic DNA, but no colony was identified to be B genomespecific. DNA digests of AB genome tetraploids with restrictionenzymeHae III gave a strong band at 4.2 kb. Clone pAbKB3, derivedfrom the 4.2 kb band, was found to be part of aTy1-copia -likeretrotransposon present in A and B genome chromosomes. ClonedrRNA genes were used forin situ hybridization and showed thatdiploidA. strigosa has four major sites for 18S-25S rDNA andtwo pairs of sites for 5S rDNA (pairs on the same satellitedchromosome, on different chromosome arms), while 4xA. vavilovianahas eight major sites for 18S-25S rDNA and four pairs of sitesfor 5S rDNA (pairs on the same satellited chromosome, on differentchromosome arms). A repetitive sequence from rye pSc119.2, showeddispersed hybridization, while the telomeric sequence in clonepLT11 hybridized to telomeres. Again no discrimination was possiblebetween A and B genome chromosomes. The molecular similaritiesbetween the diploidA. strigosa and thebarbata group tetraploidsclearly indicate that thebarbata group of tetraploids arosefrom Asdiploids through autotetraploidization. The successful production of interspecific hybrids between membersof the dysploid chromosome series inDahlia offers a unique opportunityto investigate chromosome evolution. Analysis of meiotic metaphaseI in these hybrids using genomic in situ hybridization (GISH)has shown that pairing occurs both between and within parentalgenomes. The Peruvian scallop (Argopecten purpuratus) has been introduced to China and has successfully been hybridized with the bay scallop (A. irradians irradians). The F1 hybrids of these two scallops exhibited a large increase in production traits and some other interesting new characteristics. To understand the genetic basis of this heterosis, nuclear gene and partial mtDNA sequences, and genomic in situ hybridization (GISH) were employed to analyze the genomic organization of the hybrids. Amplification of the ribosomal DNA internal transcribed spacer (ITS) showed that the parental ITS sequences were present in all the hybrid individuals, illustrating that the hybrid offspring inherited nuclear DNA from both parents. Sequence analyses of the ITS region further confirmed that the hybrids harbored alleles from their parents; some recombinant variants were also detected, which revealed some alterations in the nuclear genetic material of the hybrids. The analysis of mitochondrial 16S rDNA showed that the hybrids possessed sequences that were identical to the 16S rDNA of the female parents, proving a matrilineal inheritance of mitochondrial genes in scallops. In addition, GISH clearly discriminated between the parental chromosomes and indicated a combination of haploid genomes of duplex parents in the hybrids. The genetic analyses in our study illustrated that the F1 hybrids inherited nuclear material from both parents and cytoplasmic genetic material maternally, and some variations occurred in the genome, which might contribute to a further understanding of crossbreeding and heterosis in scallop species.  相似文献

Repetitive sequences have been widely used for examining genomeand species relationships by in situ and Southern hybridization.In the present study, double-stranded DNA sequences, from denaturedDNA reannealed to Cot = 1, from Avena strigosa(2 n = 2x = 14;A genome; referred to as CotA) and Avena sativa(2n = 6 x =42; ACD genome; referred to as CotACD) were isolated with ahydroxyapatite column, and were used for in situ hybridizationon hexaploid A. sativa chromosomes. Probe CotACD labelled allchromosomes evenly throughout their length at the same intensity.Probe CotA labelled the 28 A and D genome chromosomes stronglyand the 14 C genome chromosomes weakly. Three cloned repetitivesequences, pAvKB9 (126 bp), pAvKB26 (223 bp) and pAvKB32 (721bp) were characterized in the A, B, C and D Avena genomes andthe genus Arrhenatherum using molecular and cytological methods.Clones pAvKB9 and pAvKB26 were absent from the Avena C genome,while both could identify the presence of the D genome by Southernhybridization. In situ hybridization to diploid and tetraploidAvena species revealed that the probes showed a dispersed genomicorganization and that they are present on both arms of all chromosomes.These sequences were excluded from areas where tandem repeats,such as rRNA genes and telomeres, are present. These resultsindicate the close relationship between A and D genomes andthe presence of common DNA sequences between A and C Avena genomes.All three clones hybridized to Southern blots containingArrhenatherumdigested genomic DNA, indicating Arrhenatherum’s closeaffinity to A, B and D Avena genomes. Phylogenetic relationships of five taxa of Clivia, one probablenew species plus four recognized species, and three outgroupspecies were studied using sequences of the nuclear ribosomal5S non-transcribed spacer and the internal transcribed spacer(ITS) of 45S rDNA. Analysis of the data sets separately generatedsome well-supported groupings and congruent phylogenies. Cliviaminiata and C. gardenii are closely related. ‘Robust Gardenii’,the putative new species, is a sister clade of this group. Clivianobilis is distantly related to these three taxa and C. caulescensoccupies an intermediate position between the two groups. Chromosomelocations and distribution patterns of the 5S nuclear ribosomalgene in the species of Clivia were investigated using fluorescenceinsitu hybridization (FISH). In all species, only one pair of5S rDNA signals was observed. Different combinations of chromosome N- or C-banding with in situ hybridization (ISH) or genomic in situ hybridization (GISH) were sequentially performed on metaphase chromosomes of wheat. A modified N-banding-ISH/GISH sequential procedure gave best results. Similarly, a modified C-banding - ISH/GISH procedure also gave satisfactory results. The variation of the hot acid treatment in the standard chromosome N- or C-banding procedures was the major factor affecting the resolution of the subsequent ISH and GISH. By the sequential chromosome banding - ISH/GISH analysis, multicopy DNA sequences and the breakpoints of wheat-alien translocations were directly allocated to specific chromosomes of wheat. The sequential chromosome banding- ISH/GISH technique should be widely applicable in genome mapping, especially in cytogenetic and molecular mapping of heterochromatic and euchromatic regions of plant and animal chromosomes.  相似文献

Differences in premature leaf abscission and in visible steminjury in genetic lines of poplar followed continuous fumigationswith air pollutant levels of SO2 (90–100 nl l–1)and O3 (70–80 nl l–1) either separately or together.The clones used were: Populus deltoides var. missiouriensisMarsh., P. nigra cv. ‘italicd’ L., and the hybridsP. nigra cv. ‘italica’ xP. deltoides (He-X/3) andP. nigra cv.‘italica’ x P. nigra cv. ‘Serres’(He-K/7). While most leaf abscission occurred within 20 d fromthe start of fumigation, stem lesions (intumescences), appearedonly after 72 d. Their anatomical characteristics include theformation of lysigenous aerenchyma in the lower parts of theintumescence, the sloughing of superficial cells from the injuredarea, and the development of crystalline formations on the surfaceof the lesions. P. deltoides exhibited the least morphologicalresponse to the gases. Ethylene released from fumigated leaves was determined at thesame gas concentration of SO2 (100 nl l–1), O3 (75 nll–1) and their mixture. Leaves of P. deltoides consistentlyshowed the lowest ethylene production after the gas treatments.P. ‘italica’ production was higher but was littlealtered by the treatments. The two hybrids He-X/3 and He-K/7showed the greatest increases in ethylene evolution with time.With He-K/7 exposed to the gas mixture the production of ethylenedecreased after the initial sharp rise during days 1–2,and reflected the considerable leaf damage observed after day3. The results suggest that sensitivity to air pollution, (as shownby leaf abscission and the formation of stem intumescences)can be correlated with the level of pollutant-induced ethyleneevolution from leaves. Initially high levels could induce abscission,whilst prolonged production could be responsible for intumescenceinitiation. The discussion proposes a series of events fromSO2 and/or O3 entry into the leaf and the physiological reasonsfor the clonal differences. The effects of exposure of up to 2 h with sulphur dioxide ona range of plant species was observed by measuring changes inthe rate of net photosynthesis under closely controlled environmentalconditions. Ryegrass, Lolium perenne ‘S23’ was thespecies most sensitive to SO2; significant inhibition was detectedat 200 nl l–1. Fumigations at 300 nl l–1 also inhibitedphotosynthesis in field bean (Vicia faba cv. ‘Three FoldWhite’ and ‘Blaze’) and in barley (Hordeumvulgare cv. ‘Sonja’). No effect was detected inwheat (Triticum aestivum cv. ‘Virtue’) at concentrationsup to 600 nl l–1 SO2, or in oil-seed rape (Brassica napuscv. ‘Rafal’) except at 800 nl l–1 SO2). Recoverycommenced immediately after the fumigation was terminated andwas complete within 2 h when inhibition had not exceeded 20%during the SO2 treatment.
