The content of saccharides and phenolic compounds (including flavonoids and lignin) and the activity of L-phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) were determined in leaves and tillering nodes of winter wheat Triticum aestivum L.) cultivars Inna and Moskovskaya 39, differing in the level of frost resistance. These parameters were determined in three groups of plants—control, hardened, and treated with synthetic antioxidants (ambiol and amerol 2000). Cold hardening increased accumulation of primary and secondary metabolites in tissues but decreased the enzyme activity. Treatment with antioxidants also increased the content of saccharides and phenolic compounds (primarily flavonoids) and PAL activity. These changes were more pronounced in cultivar Inna, which is less frost resistant than Moskovskaya 39.  相似文献

Phenylalanin ammonia-lyase (PAL) plays a pivotal role in the production of phenolic compounds, which are responsible for the success of the defense strategies in harsh environments in response to different stimuli. Measurements of the PAL activity, total phenolics, total flavonoids and anthocyanin contents were performed in flowers, leaves and fruits of three pistachio cultivars “Ahmadaghaii”, “Ohadi” and “Kallehghuchi”. The results showed that PAL activity was different in cultivars and in plant organs of pistachio trees (flowers, leaves and fruits). The highest activity rate of their compounds was observed in Ahmadaghaii cultivar. A positive correlation was observed between PAL activity, total phenolics and total flavonoids in leaves, and a negative correlation between PAL activity and anthocyanin contents in leaves and flowers of Ahmadaghaii cultivar. PAL activity and total phenolics in fruits of pistachio suffered a decrease when the maturation processes began. It is suggested that the hulls of the pistachio fruits, containing high level of phenolic compounds (especially in Ahmadaghaii cultivar), may function as a protective layer of defense chemicals against ultraviolet radiation and pathogens. The final concentration of phenolic compounds, flavonoids and antocyanins in the kernel depend on PAL activity in the kernel’s cultivar. The results led to the conclusion that increase in PAL activity, phenolic compounds and flavonoids in Ahmadaghaii can help the plant to cope with the stresses better than the other cultivars. Since phenolic compounds are antioxidant and scavenge free oxygen, it is postulated that Ahmadaghaii is the most resistant cultivar to the environmental stresses.  相似文献

Defence reactions occurring in resistant (cv. Gankezaomi) and susceptible (cv. Ganmibao) muskmelon leaves were investigated after inoculating with Colletotrichum lagenarium. Lesion restriction in resistant cultivars was associated with the accumulation of hydrogen peroxide (H2O2). The activity of antioxidants catalase (CAT) and peroxidase (POD) significantly increased in both cultivars after inoculation, while levels of both CAT and POD activity were significantly higher in the resistant cultivar. Ascorbate peroxidase (APX) increased in both cultivars after inoculation, and level of APX activity was significantly higher in the resistant cultivar. Glutathione reductase (GR) activity significantly increased in both cultivars following inoculation, but was higher in the resistant cultivar, resulting in higher levels of ascorbic acid (AsA) and reduced glutathione (GSH). Phenylalanine ammonia lyase (PAL) significantly increased in inoculated leaves of both cultivars, resulting in higher levels of total phenolic compounds and flavonoids. The pathogenesis‐related proteins chitinase (CHT) and β‐1, 3‐glucanase (GLU) significantly increased following inoculation with higher activity in the resistant cultivar. These findings show that resistance of muskmelon plants against C. lagenarium is associated with the rapid accumulation of H2O2, resulting in altered cellular redox status, accumulation of pathogenesis‐related proteins, activation of phenylpropanoid pathway to accumulation of phenolic compounds and flavonoids.  相似文献

Investigations have been made to study the production of phenolic compounds (total phenolics, flavonoids and phenylpropanoids) and total antioxidant capacity in 27 Macedonian traditional medicinal plants to improve its potential as a source of natural antioxidants. Antioxidant potential of plant extracts was analyzed by five different assays: cupric reducing antioxidant capacity (CUPRAC), phosphomolybdenum method (PM), reducing power (RP), 2,2-diphenyl-1-picrylhydrazyl (DPPH·) and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS·+) radical scavenging activity. Origanum vulgare extract consistently exhibited the highest content of phenolic compounds and the strongest antioxidant capacity based on the tests performed, and can be proposed as a promising source of natural antioxidants. Melissa officinalis and Salvia ringens were also identified as valuable sources of antioxidant compounds. A positive linear correlation between antioxidant activity and total phenolics, flavonoids and phenylpropanoids indicates that these compounds are likely to be the main antioxidants contributing to the observed activities of evaluated plants. These findings suggest that the medicinal plants studied in this paper are good sources of bioactive compounds for the food and pharmaceutical industries.  相似文献

该研究以幼果期、白果期、转色期的离体‘北陆’蓝莓果实为试材,设置0(CK)、5、10、15min紫外光辐照处理,24h后取样分析蓝莓果实中可溶性糖、总酚、类黄酮和花青苷含量,以及苯丙氨酸裂解酶(PAL)和查尔酮异构酶(CHI)活性的变化,探究UV-B紫外照射处理对不同发育时期蓝莓主要果实品质及相关酶活的影响。结果显示:(1)对于幼果期蓝莓,5min UV-B处理可显著增加果实内可溶性糖含量;10min UV-B处理果实PAL活性增加效果最为显著;15min UV-B处理对果实总酚和花青苷积累的促进作用最大,但显著降低了类黄酮含量和CHI活性。(2)对于白果期蓝莓,5min UV-B处理显著增加了果实类黄酮含量和CHI活性,10min处理使果实可溶性糖和总酚含量较对照分别增加25%和18%;15min处理对果实花青苷含量和PAL活性影响作用最大。(3)对于转色期蓝莓,各处理除果实可溶性糖及类黄酮含量降低外,其余物质含量均显著增加。(4)UV-B处理并未改变果实发育过程中可溶性糖、总酚、类黄酮和花青苷含量及PAL、CHI酶活性的积累规律。(5)蓝莓果内PAL活性与其可溶性糖、总酚和类黄酮的积累呈极显著正相关关系,而CHI活性仅与其可溶性糖呈极显著正相关。研究表明,UV-B辐照处理促进了幼果期和白果期可溶性糖的积累,也能促进不同发育时期蓝莓果实总酚和花青苷及白果期类黄酮的积累,对蓝莓果实主要品质能够产生积极的影响。  相似文献

Callus from Opuntia streptacantha (cv. Tuna loca), Opuntia megacantha (cv. Rubí reina), and Opuntia ficus-indica (cv. Rojo vigor) were exposed to jasmonic acid (JA) and abiotic stress (drought and UV light) to improve the metabolite production. The callus growth curves, phenolic acids and flavonoids content, antioxidant activity and phenylalanine ammonia lyase (PAL) activity were analyzed under normal and stress conditions. In O. streptacantha callus, the phenolics concentration increased 1.6 to 3 times times in presence of 5% PEG or after irradiation with UV light for 240 min, respectively, while flavonoids triplicate with UV light. A significant increase in antioxidant activity was observed in calli from the three Opuntia species in media with 50 µM JA. The relationships between metabolites/PAL activity, and metabolites/antioxidant activity were analyzed using a surface response methodology. Results showed that PAL activity, induced with PEG and UV, correlated with flavonoids content in O. megacantha and O. ficus-indica calli; PAL activity was related to both flavonoids and phenolics compounds in O. ficus-indica and O. megacantha calli exposed to JA, but only to flavonoids in O. streptacantha callus. In general, the JA stimulated simultaneously the metabolic pathways for phenolics and flavonoids synthesis, while abiotic stress induced mainly flavonoids route. As the stressed Opuntia calli exhibited as high antioxidant activity as cladodes, they are a promising system for research on antioxidant biosynthesis and/or to identify new compounds with antioxidant properties.  相似文献

While the health benefits of antioxidant compounds from terrestrial plants are widely accepted in Western counties, there is less recognition of the health benefits of marine algal antioxidant compounds. Oceans are an abundant source of biomaterials, with many natural antioxidants derived from marine algae being investigated as potential anti-aging, anti-inflammatory, anti-bacterial, anti-fungal, cytotoxic, anti-malarial, anti-proliferative, and anti-cancer agents. The aim of this work was to quantify and compare polyphenolic content and free radical scavenging activity of algal extracts using normal phase and reverse phase thin layer chromatography. Post-chromatographic derivatization with neutral ferric chloride (FeCl3) solution and with 2,2-diphenyl-1-picrylhydrazyl (DPPH·) free radical were used to assess total polyphenolic content and free radical scavenging activities in algal samples. Total phenolic content quantified on normal phase plates was correlated to phenolic content established on reverse phase plates. Similarly, free radical scavenging activity established on normal phase and reverse phase plates were in good agreement. However, although free radical scavenging activities determined on normal phase plates were highly correlated with polyphenolic content, this correlation was low for reverse phase plates. Lipophilic reversed phase TLC plates do not effectively separate mixtures of highly polar compounds like flavonoids, phenolic compounds and their glucosides. Thus, although reversed phase plates are recommended for assessment of free radical scavengers, as they do not influence the free radical-antioxidant reaction, they may not provide the best separation of polar phenolic compounds, especially flavonoids, and therefore may not accurately quantify polyphenolic content and free radical scavenging potential.  相似文献

Cancer prevention and treatment using traditional Chinese medicines have attracted increasing interest. This study characterizes antioxidant activity and phenolic compounds of traditional Chinese medicinal plants associated with anticancer, comprising 112 species from 50 plant families. The improved ABTS(*+) method was used to systematically assess the total antioxidant capacity (Trolox equivalent antioxidant capacity, TEAC) of the medicinal extracts. The TEAC values and total phenolic content for methanolic extracts of herbs ranged from 46.7 to 17,323 micromol Trolox equivalent/100 g dry weight (DW), and from 0.22 to 50.3 g of gallic acid equivalent/100 g DW, respectively. A positive, significant linear relationship between antioxidant activity and total phenolic content (all R(2) values>/=0.95) showed that phenolic compounds were the dominant antioxidant components in the tested medicinal herbs. Major types of phenolic compounds from most of the tested herbs were preliminarily identified and analyzed, and mainly included phenolic acids, flavonoids, tannins, coumarins, lignans, quinones, stilbenes, and curcuminoids. These medicinal herbs exhibited far stronger antioxidant activity and contained significantly higher levels of phenolics than common vegetables and fruits. Traditional Chinese medicinal plants associated with anticancer might be potential sources of potent natural antioxidants and beneficial chemopreventive agents.  相似文献

Foliar spray and micro-injection of plant growth-promoting rhizobacterial species, viz. Pseudomonas fluorescens and P. aeruginosa on chickpea induced synthesis of phenylalanine ammonia-lyase (PAL) when tested against Sclerotinia sclerotiorum. Induction of PAL was also associated with increased synthesis of phenolic compounds such as tannic, gallic, caffeic, chlorogenic and cinnamic acids. Treatment with P. fluorescens was found to be more effective in inducing phenolic compounds as compared to P. aeruginosa. However, persistence of PAL activity was observed more with P. aeruginosa. Although both the inoculation methods were effective, foliar application was found to be superior to micro-injection in terms of rapid PAL activity leading to the synthesis of phenolic compounds.  相似文献

The study aimed at comparing the changes in the content of free amino acids, phenolic compounds and the activity of PAL and TAL caused by two piercing-sucking arthropods: the grape mealybug (Pseudococcus maritimus Ehrh.) and the two-spotted spider mite (Tetranychus urticae Koch) in the leaves of orchid and strawberry, respectively. The obtained results show that the amino acid content and the ratio of amino acids to phenolic compounds increased in both plant species infested by the mealybug and the mite. However, such response was weakly dependent on changes in activity of the analysed enzymes. The pest feeding affected accumulation of the phenolic compounds, since the induction of the PAL activity in mealybug-infested orchid leaves during the first 5 h of the experiment preceded the increase in phenolic compounds during the first week of insect feeding. Instead, the increased activity of TAL was accompanied by elevated levels of phenolic compounds in the leaves of strawberry infested by mites. Mechanisms of biochemical plant responses induced by infestation of the studied herbivorous arthropods are discussed.  相似文献

L-Phenylalanine-ammonia lyase (PAL) activity and phenolic content were investigated during growth and maturation of sweet cherry fruit on the tree. Phenolic acid, flavonoid and tannin contents, were studied in relation to PAL activity over two years. Concomitant variations in PAL activity and in flavonoid or phenolic acid concentrations were found during growth, suggesting that PAL is the major element controlling the synthesis of phenolic compounds in sweet cherry. During ripening, the increase in PAL activity did not appear to be directly related to anthocyanin synthesis. The concentrations of neochlorogenic acid and narcissin measured over the same period were also related to PAL activity.  相似文献

The effects of 2-aminoindane-2-phosphonic acid (AIP), a potent phenylalanine ammonia-lyase (PAL) inhibitor, on the accumulation of cadmium and nickel in chamomile (Matricaria chamomilla) were examined in this study. In vitro assay of AIP effect showed a 90% reduction in PAL activity. In plants cultured for 7 days in Cd or Ni solutions with AIP, PAL activity was higher in both shoots and roots (in comparison with metals without AIP), and was correlated with changes in free phenylalanine content. Individual amino acids were both positively and negatively affected by AIP, with the accumulation of tyrosine and proline showing increases in some variants. Contents of soluble phenols and flavonoids were not considerably affected, while amounts of coumarin-related compounds, cell wall-bound phenols and phenolic acids were substantially reduced in AIP-treated variants. Lignin accumulation decreased in controls and increased in Cd variants in response to AIP. Shoot Cd content was depleted, but shoot Ni was elevated by AIP. Total root content of Cd and Ni decreased in +AIP variants. AIP also caused more expressive changes in hydrogen peroxide and superoxide content in Cd than in Ni variants. Our results indicate that phenols have important roles in the uptake of Cd and Ni. The present findings are discussed in the context of available data regarding AIP's effect on phenols.  相似文献

The impact of melatonin (50 µM L?1) applied to Vigna radiata seeds by hydro-priming on phenolic content, L-phenylalanine ammonialyase (PAL) activity, MEL level, antioxidant properties of ethanol extracts as well as electrolyte leakage from chilled and re-warmed Vigna radiata roots of seedlings were examined. Seedlings obtained from non-primed seeds, hydro-primed and hydro-primed with MEL were investigated after 2 days of chilling and 2 days of re-warming. At 25°C, the level of MEL in roots derived from seeds hydro-primed with MEL was 7-fold higher than in roots derived from non-primed seeds. However, the content of MEL significantly decreased in all variants investigated after re-warming, in contrast to PAL activity and phenolic levels, which reached the highest values. The antioxidant capacity of ethanol extracts from chilled and re-warmed roots, determined by ABTS+· assay, was correlated with phenolic content while the reducing ability of these extracts, determined by the FRAP method, correlated with PAL activity. However, both were the highest in rewarmed roots with applied MEL, which was accompanied by a significant decline in electrolyte leakage. Taken together, results may indicate that MEL can play a positive role in plant acclimation to stressful conditions and activation of phenolic pathway by this molecule.  相似文献

Promising plant species with high antioxidant activity and maximum content of phenols have been found. The present study demonstrated the combined effect of 24-epibrassinolide and salicylic acid against lead (Pb, 0.25, 0.50, and 0.75 mM) toxicity in Brassica juncea seedlings. Various parameters including water status, metal uptake, total water- and lipid-soluble antioxidants, metal chelator content (total thiols, protein-bound thiols, and non-protein-bound thiols), phenolic compounds (flavonoids, anthocyanins, and polyphenols), and organic acids were studied in 10-day-old seedlings. Dry matter content and the heavy metal tolerance index were reduced by 42.24 and 52.3%, respectively, in response to Pb treatment. Metal uptake, metal-chelating compounds, phenolic compounds, and organic acids were increased in Pb-treated seedlings as compared to control plants. The treatment of Pb-stressed seedlings with combination of EBL and SA resulted in enhancement of heavy metal tolerance index by 40.07%, water content by 1.84%, and relative water content by 23.45%. The total water- and lipid-soluble antioxidants were enhanced by 21.01 and 2.21%, respectively. In contrast, a significant decline in dry weight, metal uptake, thiol, and polyphenol contents was observed following the application of 24-epibrassinolide and salicylic acid. These observations indicate that Pb treatment has an adverse effect on B. juncea seedlings. However, co-application of 24-epibrassinolide and salicylic acid mitigates the negative effects of Pb, by lowering Pb metal uptake and enhancing the heavy metal tolerance index, water content, relative water content, antioxidative capacities, phenolic content, and organic acid levels.  相似文献

The content of total phenolic compounds and flavonoids was determined in methanol extracts of root, stem, leaves, and inflorescences from wild growing and greenhouse cultivated plants of Castilleja tenuiflora. The antioxidant activity in each extract was evaluated using three in vitro models: scavenging of free radicals with 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS), and reducing power by the phosphomolybdenum assay. Both, antioxidant activity and phytochemicals content were influenced significantly (P < 0.05) by the source of the plant material and the organ. Cultivated plants had the highest content of phenolic compounds (37.95 mg gallic acid equiv. g?1 dry weight, P < 0.05) and the strongest antioxidant activity. Total phenolic compounds content correlated significantly with the antioxidant activity for all studied plant material and organs (P < 0.05). TLC profile using DPPH as a detection reagent indicated that the phenylethanoids verbascoside and isoverbascoside are the main contributors to the free-radical scavenging of C. tenuiflora. Cultivated plants of C. tenuiflora are an alternative source of natural antioxidants to wild growing plants. The antioxidant properties of C. tenuiflora may be associated with its traditional use to treat conditions consistent with radical-related diseases (e.g. inflammation, tumors).  相似文献
